Leafminer flies, especially, Liriomyza huidobrensis, Liriomyza sativae and Liriomyza trifolii, are quarantine species in many countries. Their morphological similarity makes identification difficult. To develop a rapid, reliable, sensitive and simple molecular identification method using multiplex PCR, we newly sequenced the mitochondrial cytochrome oxidase I (COI) genes of Liriomyza bryoniae, Liriomyza chinensis, L. huidobrensis, L. sativae, L. trifolii, Chromatomyia horticola and four parasitoid species. We aligned them with all the COI sequences of the leafminer flies found in the international DNA nucleotide sequence databases (DDBJ/EMBL/GenBank). We then designed species‐specific primers to allow us to differentiate between L. bryoniae, L. chinensis, L. huidobrensis, L. sativae, and L. trifolii.  相似文献

Host plant species can affect the behavior and attributes of parasitoids, such as host searching, oviposition, and offspring fitness. In this study, parasitism, host feeding, and sex ratios of Diglyphus isaea (Walker) (Hymenoptera: Eulophidae) on Liriomyza huidobrensis (Blanchard), Liriomyza sativae Blanchard, and Liriomyza trifolii (Burgess) (Diptera: Agromyzidae) larvae reared on Phaseolus vulgaris L., Pisum sativum L., Solanum lycopersicum L., and Vicia faba L. were determined. In no-choice tests, L. huidobrensis had the highest rate of parasitism when reared on P. vulgaris (46%), L. sativae when reared on V. faba (59%) and P. vulgaris (59%), and L. trifolii when reared on S. lycopersicum (68%). Host feeding in no-choice tests ranged between 2% and 36% and was highest on L. trifolii reared on V. faba. Results of choice tests showed a significant interaction effect for host plant and Liriomyza species on parasitism and host feeding. Within plant mixtures, L. sativae reared on P. vulgaris had the highest rate of parasitism (31%), followed by L. trifolii on S. lycopersicum (29%) and L. huidobrensis on V. faba (28%). Host feeding was highest on L. trifolii reared on S. lycopersicum (14%) and lowest on L. huidobrensis reared on P. sativum and S. lycopersicum (1%). In some instances, plant mixtures resulted in a higher proportion of females of D. isaea than single plant species. The highest proportion of females was obtained in plant mixtures on L. huidobrensis and L. trifolii on V. faba (71 and 72%, respectively). This study suggests that planting crop mixtures can potentially lead to higher proportions of females, thus improving parasitism and host feeding, depending on Liriomyza and host plant species.  相似文献

The Braconidae, a family of parasitic wasps, constitute a major taxonomic challenge with an estimated diversity of 40,000 to 120,000 species worldwide, only 18,000 of which have been described to date. The skeletal morphology of braconids is still not adequately understood and the terminology is partly idiosyncratic, despite the fact that anatomical features form the basis for most taxonomic work on the group. To help address this problem, we describe the external skeletal morphology of Opius dissitus Muesebeck 1963 and Biosteres carbonarius Nees 1834, two diverse representatives of one of the least known and most diverse braconid subfamilies, the Opiinae. We review the terminology used to describe skeletal features in the Ichneumonoidea in general and the Opiinae in particular, and identify a list of recommend terms, which are linked to the online Hymenoptera Anatomy Ontology. The morphology of the studied species is illustrated with SEM-micrographs, photos and line drawings. Based on the examined species, we discuss intraspecific and interspecific morphological variation in the Opiinae and point out character complexes that merit further study.  相似文献

Rearing experiments at different temperatures (10, 15, 20, 25 and 30°C) were made onOpius dissitus Muesebeck, a parasitoid ofLiriomyza trifolii (Burgess). The effects of Bt transgenic cottons (Bt-I expressing cry1Ac and Bt-II expressing cry1Ab and cry2Ab or cry1Ab and cry1Fa) and non-Bt cottons on feeding, oviposition and longevity of adults, and development and survival of Liriomyza trifolii larvae were studied under laboratory conditions; and infestation on four Bt and two non-Bt cotton traits were investigated under field conditions. Laboratory choice and no-choice tests showed that L. trifolii adults were capable of distinguishing between Bt cottons and non-Bt cottons. In a choice test on younger plants (4-5 leaves), the adults were found more often and made more feeding punctures (FP) on non-Bt cottons than on Bt cottons. On older plants (8-9 leaves), adults made the most FP on non-Bt cotton followed by those on Bt-II cottons and the least on Bt-I cotton. The females oviposited more eggs (6.7 eggs per leaf) on non-Bt cotton than on Bt-I (1.7 eggs per leaf) and Bt-II (0.8 eggs per leaf) cottons on younger plants and oviposited similar numbers of eggs (0.7-1.3 eggs per leaf) on non-Bt and Bt cottons on older plants. In a no-choice test, the females also fed more FP on non-Bt cottons than on Bt cottons on both younger and older plants. The females oviposited more eggs (15.6 eggs per leaf) on non-Bt cotton than on Bt-I (8.2 eggs per leaf) and Bt-II (6.5 eggs per leaf) cottons on younger plants and similar numbers of eggs (2.5-3.3 eggs per leaf) on non-Bt and Bt cottons on older plants. Larval and puparial survivals were not different among Bt and non-Bt cottons. The occurrence and damage of leafminers on cottons in the field showed that L. trifolii infested more plants and leaves and had more mines on non-Bt cotton than on Bt cottons.  相似文献

The effect of fungal infection by Metarhizium anisopliae on feeding and oviposition of adult Liriomyza huidobrensis was examined on three host plants, faba bean (Vicia faba), French bean (Phaseolus vuklgaris) and snow pea (Pisum sativum) in the laboratory. Flies were contaminated with dry conidia and allowed to feed and oviposit on the different host plants. Mortality in L. huidobrensis varied between 14% and 20% in the controls and between 77% and 100% in fungal treatments 120 h post-infection for the three host plants. L. huidobrensis made more punctures (47.3–52.6 cm?2) in the control than in the fungal treatments (23.1–26.9 cm?2) for the three host plants. The cumulative average number of punctures cm?2/female by L. huidobrensis was higher in the controls than in fungal treatments from 72 h post-treatment in faba bean (12.2 vs. 8.2) and French bean (14.8 vs. 8.9), and from 48 h post-inoculation in snow pea (8.5 vs. 5.7). Female L. huidobrensis laid more eggs in the control (0.6–6.1) than in fungal treatments (0.2–1.5) across the host plants tested. The cumulative mean number of eggs cm?2/female was significantly higher in the controls than in fungal treatments from 48 h post-treatment in faba bean (0.4 vs. 0.2) and French bean (0.1 vs. 0), and 96 h post-inoculation in snow pea (0.2 vs. 0.1). The host plant did not affect the average total number of punctures but had a significant effect on egg laying, with faba bean harboring greater number of eggs in both control and fungal treatments. A proper timeline application of the fungus before onset of feeding and oviposition peaks will be crucial in field suppression of the pest using M. anisopliae. In addition, a great consideration must be given to the target host plants prior to application of the fungus.  相似文献

The parasitic incidence of Opius concolor (Hymenoptera, Braconidae) on a replacement host, Ceratitis capitata (Diptera, Trypetidae) was studied using polyacrylamide gel electrophoresis and immunoelectrophoresis methods. A noticeable modification of C. capitata proteins was observed when parasited by O. concolor. But the most important phenomenon is the finding of common antigens between the host and the parasite. These results are discussed with regard to trophic and parasitic behaviour of the parasite.  相似文献

Liriomyza leafminer flies represent a serious threat to horticultural production in East Africa. Total field parasitism rates recorded in Kenya are below 5%, with the indigenous ectoparasitoid Diglyphus isaea Walker being one of the key parasitoid species. The International Centre of Insect Physiology and Ecology (icipe), in collaboration with the International Potato Centre (CIP), imported into Kenya the endoparasitoid Phaedrotoma scabriventris Nixon to improve natural control of leafminers. The objective of this study was to investigate the interactions between D. isaea and P. scabriventris when used together for the biological control of Liriomyza species. These interactions were studied under laboratory conditions, using treatments that involved single, simultaneous and sequential releases of the different parasitoid species onto plants infested by L. huidobrensis larvae. While used separately, parasitism rates of D. isaea and P. scabriventris were 30.4 ± 10.9% and 63.6 ± 7.7% respectively. However, when used simultaneously, the total parasitism rate increased to 77.0 ± 5.3%. Although P. scabriventris had no effect on D. isaea, the presence of D. isaea reduced the specific parasitism rate of P. scabriventris. In addition, both parasitoids induced leafminer mortality through larval-feeding and stinging. However, feeding and stinging mortality induced by D. isaea (41.9 ± 9.1%) was significantly higher compared to P. scabriventris (11.9 ± 8.7). Similarly, pupal mortality due to feeding and stinging activity was 49.1 ± 6.5% and 21.6 ± 1.9% when exposed to D. isaea and P. scabriventris respectively. The implication for simultaneous use of both parasitoids in East Africa is discussed.  相似文献

Liriomyza bryoniae, L. huidobrensis, L. sativae and L. trifolii are leafminers of great economic importance; morphologically they are difficult to separate, many being inseparable at the pre-adult stages. Horizontal starch gel electrophoresis of larvae, pupae and adults was carried out in order to search for diagnostic allozymic characters. Analysis of the variation patterns at 15 genetic loci reveals that each species can be identified at all three stages of the life cycle. A biochemical key is presented.  相似文献

Host acceptability and suitability Psyttalia concolor (Szépligeti) is a koinobiont, larval parasitoid of tephritid fruit flies. Individuals of P. concolor were field-collected from coffee in the central highlands of Kenya, and cultured initially on Mediterranean fruit fly (medfly), Ceratitis capitata (Wiedemann). They were then examined for their ability to oviposit in and develop on five other tephritid species that are pests in Kenya. In addition to the medfly, acceptability for oviposition and suitability for development were tested against the mango fruit fly, Ceratitis cosyra (Walker), the Natal fruit fly, Ceratitis rosa Karsch, Ceratitis fasciventris (Bezzi), Ceratitis anonae Graham and the melon fruit fly, Bactrocera cucurbitae (Coquillett). Ceratitis capitata and C. cosyra were accepted as hosts significantly more often than the other species. Superparasitism was recorded only from C. capitata and C. cosyra. Two days after oviposition, parasitoid eggs in C. fasciventris and B. cucurbitae were encapsulated, whereas those in C. rosa and C. anonae were encapsulated, and often melanized. Ceratitis capitata was the most suitable host for Kenyan populations of Psyttalia concolor in terms of progeny production, and proportion of female progeny.  相似文献

Behavioral interactions among Cotesia urabaeAustin and Allen, Dolichogenidea eucalyptiAustin and Allen (Hymenoptera: Braconidae), and their host Uraba lugensWalker, the gum leaf skeletonizer (Lepidoptera: Noctuidae), were observed at three host sizes over a 20-min period. These sizes were first instar (small, gregarious), fourth-fifth instar (mid, gregarious), sixth-seventh instar (large, solitary) larvae. Unlike C. urabae, D. eucalyptiused its legs to hold small larvae before ovipositor insertion. D. eucalyptialso visited patches of small larvae more frequently, proceeded less often through patches of mid larvae, and made significantly fewer ovipositions in mid and large larvae. Small larvae responded to both parasitoids by dispersing outward, while mid larvae responded to parasitoids by moving inward to form a denser group. Larvae reared or thrashed after each parasitoid visit, especially mid larvae, and some continued to do so for up to 2h after parasitoid departure. Mid and large larvae occasionally injured parasitoids by biting their appendages. By rearing or thrashing immediately prior to an encounter with a parasitoid, mid and large larvae decreased the likelihood of being parasitized by up to 50%.  相似文献

Liriomyza trifolii is a serious pest of chrysanthemum greenhouses and other crops around the world. The larvae feed within the leaves of the host plants and create serpentine mines. The Sterile Insect Technique (SIT) has been successfully used against some Dipteran species in the field, and it is especially efficient against isolated populations like those on islands. Therefore, SIT against pest populations in confined environments such as greenhouses should have high potential for success. The objectives of this study were to determine the optimum gamma‐irradiation dosage required for the sterilisation of L. trifolii adults, and to determine the quality (emergence percentage, flight ability, longevity, copulatory success, and sperm transfer) of the irradiated males compared with that of non‐irradiated (normal) males. We found that sterility (< 0.7 mine per female) was achieved with the dose of 170 ± 5% Gy. The copulatory success and sperm transfer during copulation of the sterile males were not significantly different from those of normal males. Moreover, the longevity, emergence percentage and flight ability of irradiated males were also not significantly different from normal males. The SIT experiments suggest that the release of sterile L. trifolii can significantly reduce the reproductive capacity of a wild leafminer population. Our studies indicate that sterilisation of L. trifolii flies is feasible and that sterile males are of high quality and competitive with normal males. Based on these data, research on the use of SIT against L. trifolii populations in greenhouses is ongoing.  相似文献

Phylogenetic relationships among populations of the polyphagous pea leafminer, Liriomyza huidobrensis (Blanchard), were investigated using DNA sequence data. Maximum parsimony analysis of 941 bp of mitochondrial cytochrome oxidase I and II genes showed that L. huidobrensis contains two well-defined monophyletic groups, one composed of specimens from California and Hawaii and one composed of specimens from South and Central America together with populations that have been recently introduced into other parts of the world. The differentiation between the two clades within L. huidobrensis is equivalent to that seen between other agromyzid species, suggesting that L. huidobrensis as currently defined contains two cryptic species. This finding is consistent with field observations of differences in pest status and insecticide resistance between L. huidobrensis populations. Until additional studies are complete, no changes in L. huidobrensis taxonomy are proposed. However, researchers and quarantine officials may wish to consider the findings of the current study in designing research, pest management, and quarantine programs for L. huidobrensis.  相似文献

In the laboratory, the effect of host fruit species Citrus paradisi, C. aurantium, Prunus persica, and Psidium guajava on A. fraterculus parasitism by Diachasmimorpha longicaudata was studied. The number of ovipositor-probing events and the probing-time were documented to evaluate the role of fruit chemistry, and epicarp and mesocarp thickness, respectively. The relationship between the parasitization rate and fruit size in particular plant species was analyzed by applying a simple regression. Results showed that guava and peach yielded significantly more parasitoids than both Citrus spp. Probably, the parasitization rate of D. longicaudata on A. fraterculus would be influenced in part by chemical and physical factors from fruit species.  相似文献

The stone leek leafminer Liriomyza chinensis Kato is one of the most important pests of Allium spp. In recent years, a new genotype of L. chinensis, genotype B, has been shown to cause more severe damage to Allium spp. compared with the native genotype A in Japan. Thus, identification of the genotypes is important for establishing L. chinensis control strategies. In this study, we developed a simple and rapid method of genotype identification using PCR-restriction fragment length polymorphism (RFLP). Primers were designed to mitochondria cytochrome oxidase subunit I gene (mtCOI) nucleotide sequences from both genotypes. Genotypes A and B were successfully identified using the restriction enzymes PvuI and DdeI, respectively. Using PCR-RFLP, we identified the genotype of L. chinensis samples (n = 108) collected in seven Japanese bunching onion fields in Ibaraki Prefecture, Japan. All individuals were shown to be genotype B, as expected.  相似文献

This study was conducted to develop sequential sampling plans to estimate leafmine density by Liriomyza sativae (Blanchard) at two fixed-precision levels in a cucumber greenhouse. The within-greenhouse spatial patterns of leafmines were aggregated. The slopes and intercepts of Taylor’s power law did not differ between years. A fixed-precision level sampling plan was developed using the parameters of Taylor’s power law generated from total number of leafmines in a cucumber leaf at two precision levels (D) of 0.1 and 0.25. The resulting sampling plans were tested with sequential bootstrap simulations (n = 500) using 10 independent data sets for validation. Bootstrap simulation within a wide range of densities demonstrated that actual D′ values at desired D = 0.25 averaged less than or equal to 0.25 in all cases. Even at the lowest density of leafmine (0.27 mine per leaf), the actual mean D′ was 0.24 at D = 0.25. This result shows that the sampling plan developed in this study is effective and reliable for estimating the mine densities in cucumber greenhouses.  相似文献

Fopius (= Biosteres) arisanus (Sonan) (= Opius oophilus Fullaway) is an egg-pupal parasitoid of tephritid fruit flies. Our breakthrough in the development of a laboratory-adapted strain of F. arisanus facilitated insectary rearing of parasitoids in large numbers. First colonized in captivity in 1989, F. arisanus has been reared routinely on the oriental fruit fly, Bactrocera (= Dacus) dorsalis (Hendel), its natural host. Parasitization by F. arisanus results in the latent death of developing hosts. Host mortality data are presented in a life table to estimate cost of parasitization on the economics of F. arisanus mass rearing. Percent kills of 10, 32, and 76% exerted by F. arisanus on the egg, larval, and pupal stages of the rearing hosts, respectively, resulted in a mean parasitoid recovery of 23.6%. The production cost was estimated by integrating life table data with the costs of rearing supplies and materials and personnel-hours requirements. Production of 1 million parasitoid adults requires the exposure of 4.2 million B. dorsalis eggs (= propagation hosts). We estimated the cost of producing 1 million parasitoids to be $2,363.30 and identified the most expensive aspects of F. arisanus rearing. Recommendations for streamlining the rearing process to reduce costs are provided.  相似文献
