Beet necrotic yellow vein virus (BNYVV) is the most devastating pathogen of sugar beet worldwide. This virus has been reported in the majority of sugar beet growing regions of Iran as well. For the present study, we collected samples from different sugar beet varieties with suspected symptoms of BNYVV from the main important sugar beet growing regions in eight provinces of Iran. Infection of collected samples to BNYVV was tested by ELISA and RT-PCR. Upon testing of 167 collected samples of BNYVV suspected through ELISA and RT-PCR, 115 (68.9%) were infected. Different incidences of BNYVV through surveyed provinces may represent the presence of diverse infective viral sources or resistance genes in tested sugar beet varieties which need further attempts to develop control strategies. Results also showed that BNYVV has been recently distributed throughout some surveyed regions. Otherwise, trace infection or resistance to BNYVV infection in some varieties of distinct regions may represent proper sources of resistance to BNYVV. 相似文献

Analysis of gene inheritance and expression in hybrids between transgenic sugar beet and wild beets 总被引:1,自引:0,他引:1
Reciprocal gene exchange between cultivated sugar beet and wild beets in seed production areas is probably the reason for the occurence of weed beets in sugar beet production fields. Therefore, when releasing transgenic sugar beet plants into the environment, gene transfer to wild beets ( Beta vulgaris ssp. maritima ) has to be considered. In this study the transfer of BNYVV- (beet necrotic yellow vein virus) resistance and herbicide-tolerance genes from two transgenic sugar beet lines that were released in field experiments in 1993 and 1994 in Germany to different wild beet accessions was investigated. In order to evaluate the consequences of outcrossing, manual pollinations of emasculated wild beet plants with homozygous transgenic sugar beet plants were performed. In the resulting hybrids the transgenes were stably inherited according to Mendelian law. Gene expression in leaves and roots of the hybrids was in the same range as in the original transgenic sugar beet plants. Moreover, it was found that in one of the wild beet accessions, transfer and expression of the BNYVV resistance gene did considerably increase the level of virus resistance. 相似文献

Rhizomania caused by Beet necrotic yellow vein virus (BNYVV) is one of the most devastating sugar beet diseases. Sugar beet plants engineered to express a 0.4 kb inverted repeat
construct based on the BNYVV replicase gene accumulated the transgene mRNA to similar levels in leaves and roots, whereas
accumulation of the transgene-homologous siRNA was more pronounced in roots. The roots expressed high levels of resistance
to BNYVV transmitted by the vector, Polymyxa betae. Resistance to BNYVV was not decreased following co-infection of the plants with Beet soil borne virus and Beet virus Q that share the same vector with BNYVV. Similarly, co-infection with the aphid-transmitted Beet mild yellowing virus, Beet yellows virus (BYV), or with all of the aforementioned viruses did not affect the resistance to BNYVV, while they accumulated in roots.
These viruses are common in most of the sugar beet growing areas in Europe and world wide. However, there was a competitive
interaction between BYV and BMYV in sugar beet leaves, as infection with BYV decreased the titres of BMYV. Other interactions
between the viruses studied were not observed. The results suggest that the engineered resistance to BNYVV expressed in the
sugar beets of this study is efficient in roots and not readily compromised following infection of the plants with heterologous

Monitoring gene flow from transgenic sugar beet using cytoplasmic male-sterile bait plants 总被引:10,自引:0,他引:10
One of the most discussed environmental effects associated with the use of transgenic plants is the flow of genes to plants in the environment. The flow of genes may occur through pollen since it is the reproductive system that is designed for gene movement. Pollen-mediated gene escape is hard to control in mating plants. Pollen from a wind pollinator can move over distances of more than 1000 m. To investigate the efficiency of transgenic pollen movement under realistic environmental conditions, the use of bait plants might be an effective tool. In this study, cytoplasmic male-sterile (CMS) sugar beets were tested with regard to their potential for monitoring transgene flow. As the pollen source, transgenic sugar beets were used that express recombinant DNA encoding viral (beet necrotic yellow vein virus) resistance, and antibiotic (kanamycin) and herbicide (glufosinate) tolerance genes. In a field trial, the effectiveness of a hemp (Cannabis sativa) stripe containment strategy was tested by measuring the frequency of pollinated CMS bait plants placed at different distances and directions from a transgenic pollen source. The results demonstrated the ineffectiveness of the containment strategy. Physiological and molecular tests confirmed the escape and production of transgenic offspring more than 200 m behind the hemp containment. Since absolute containment is unlikely to be effective, the CMS-bait plant detection system is a useful tool for other monitoring purposes. 相似文献

Point mutations were introduced into the genes encoding the triple gene bock movement proteins P13 and P15 of beet necrotic yellow vein virus (BNYVV). Mutations which disabled viral cell-to-cell movement in Chenopodium quinoa were then tested for their ability to act as dominant negative inhibiters of movement of wild-type BNYVV when expressed from a co-inoculated BNYVV RNA 3-based replicon. For P13, three types of mutation inhibited the movement function: non-synomynous mutations in the N- and C-terminal hydrophobic domains, a mutation at the boundary between the N-terminal hydrophobic domain and the central hydrophilic domain (mutant P13-A12), and mutations in the conserved sequence motif in the central hydrophilic domain. However, only the boundary mutant P13-A12 strongly inhibited movement of wild-type virus when expressed from the co-inoculated replicon. Similar experiments with P15 detected four movement-defective mutants which strongly inhibited cell-to-cell movement of wild-type BNYVV when the mutants were expressed from a co-inoculated replicon. Beta vulgaris transformed with two of these P15 mutants were highly resistant to fungus-mediated infection with BNYVV. 相似文献

Under greenhouse conditions, a pot experiment was conducted to clarify the potential of using some legumes as intercropped plants for reducing the root-knot nematode Meloidogyne incognita infecting sugar beet (Beta vulgaris L.) cv. DS-9004 compared to non-legume plant, garlic and non-intercropped plants. The obtained results revealed that all legumes including chickpea, Egyptian clover, faba bean, fenugreek, lentil and lupin significantly (p ≤ 0.05) reduced nematode criteria on the roots of sugar beet at different degrees. Chickpea and Egyptian clover reduced the number of galls on the roots of sugar beet as the percentage of reductions were 54 and 50%, respectively, followed by lupin and fenugreek, while garlic achieved 72% reduction compared to non-intercropped plants. Lupin reduced the number of egg masses by 59% followed by Egyptian clover and fenugreek (32%), three months after the treatment. On the other hand, six months after the treatment, chickpea reduced the number of galls by 55.7% followed by lupin (53.4%) and Egyptian clover (52.3%) and the percentage of reduction of egg masses behaved the same trend. Also, the treatments improved plant growth criteria of sugar beet, weight of roots (tubers) and the percentage of total soluble solids (TSS). 相似文献

Summary Infection of sugar beet roots by beet necrotic yellow vein virus (BNYVV) was investigated with transmission electron microscopy, immunogold labelling and enzyme linked immuno sorbent assay (ELISA). Here we show that infection of sugar beet roots is very fast, occurring during germination. Seedlings grown directly in infected soil showed higher BNYVV infection than plants transplanted into infected soil after seven days of initial growth in sterilized soil. The earlier the initial infection, the faster was its spread. The study showed that a few differentiated cells of the cortex and of the xylem parenchyma were the preferred sites of viral multiplication. The spread of viral infection was slow through differentiated tissues. Intact virions were frequently found in undifferentiated and mature vessel elements and xylem parenchyma, whereas they were rare in sieve elements. Virus particle number in the differentiating tracheary elements was high, suggesting that infection of the vessel elements preceded their differentiation. This would explain increased infection after early inoculation. Even the xylem tissue of the primary root was highly infected, the seedlings lacked virus particles in their hypocotyls and leaves. 相似文献

Soil moisture and temperature, sowing depth and penetration resistance affect the time and percentage of seedling emergence, which are crucial for the simulation of drought‐limited crop production. The aim of this research was to measure the effect of soil water potential on germination and emergence, shoot and root elongation rates (SER and RER) of two different seed/crop types. Sugar beet and durum wheat seeds were sown into two soils (clay and loam), submitted to five matric potentials (?0.01, ?0.1, ?0.2, ?0.4 and ?0.8 MPa) and incubated at constant temperature (25°C) and humidity. Cumulative count analysis was used to estimate parameters of the distribution of germination or emergence times for each box of beet or wheat seeds and to derive estimates for base potentials (ψb), hydrothermal times (H) and numbers of viable units. In a second experiment, NaCl solution was used to mimic the soil matric potentials to estimate potential RER and SER. Germination of sugar beet was slightly more sensitive to matric potential than durum wheat (ψb of ?1.13 and ?1.23 MPa, respectively). H(g) was longer for sugar beet than for durum wheat (67 vs 47 MPa °Cd). For emergence ψb was similar for both seed types and soils but hydrothermal times (H(e)) were 40 MPa °Cd higher for sugar beet than for wheat. Emergence was about 20 MPa °Cd earlier in loam than in clay. SER measured in soils were similar for both crops and for durum wheat it agreed with those determined in NaCl solution. RER and SER fell with decreasing osmotic potential to approximately 20% of their maximum values (1.03 mm h?1 and 0.57 mm h?1, respectively). Seedling viability decreased with decreasing matric potential and more in clay than in loam soil and more for sugar beet than durum wheat. Seed and soil aggregate size are discussed with respect to the effects of water diffusion and soil–seed contact on germination and emergence modelling. 相似文献

转基因作物的商品化生产和大规模环境释放在带来巨大利益的同时,也引起了全球对其生物安全问题的广泛关注和争议,其中转基因通过花粉介导的基因漂移逃逸到非转基因作物及其野生近缘种,进而导致的潜在环境和生态风险就是备受争议的生物安全问题之一。转基因植物的环境生物安全涉及两方面关键问题:如何科学评价转基因植物商品化种植以后带来的环境和生态影响;如何利用环境生物安全的研究成果来制定科学有效的风险监测和管理措施。对转基因逃逸及其潜在生态风险的科学评价应包括三个重要环节:(1)检测转基因的逃逸的频率;(2)检测转基因逃逸后的表达和遗传规律;(3)确定逃逸后的转基因对野生近缘种群体适合度的影响及其进化潜力,本文将围绕对转基因逃逸及其潜在环境风险的科学评价,以转基因水稻为案例来对转基因逃逸带来生态影响的研究好评价的进展进行简要介绍,并对目前依据风险评价研究成果制定的各种管理策略进行了讨论。只有提高对转基因生物环境安全研究和评价的水平,并制定有效的风险监测和管理措施,才能为我国转基因技术的发展和转基因产品的商品化应用保驾护航。 相似文献

Survival of Myzus persicae confined in clip-cages on mature leaves of sugar beet declined as the plants aged. Death of aphids was often preceded by the appearance of a black deposit in the aphids' stomachs, which may have been the cause of death. Both the rate of death and the proportion of aphids dying with black deposits was significantly less when plants were infected with beet yellows virus or beet mild yellowing virus, by comparison with healthy plants. The implication of these phenomena on the onset of mature plant resistance is discussed. 相似文献

This paper studies the influence of previous infestation on the host quality of sugar beet (Beta vulgaris L.) for aphids and the influence of previous infestation on sugar beet yellowing virus epidemiology. Sugar beet previously infested with Myzus persicae (Sulzer) or Aphis fabae Scopoli (Homoptera: Aphididae) had an improved host quality for subsequently infesting aphids of the same species. There was a significant negative relationship between the number of M. persicae infesting a plant and the proportion of those that died with a dark deposit in their stomachs, and a significant positive relationship between the number that settled on a plant and the number that infested it previously. Nymphs feeding on previously infested plants grew more rapidly than those on control plants. The beneficial effect of previous infestation persisted for at least 2 weeks and prolongation of the infestation beyond 2 weeks was of no further benefit to the aphids. Field grown sugar beet, previously colonised by M. persicae, was more susceptible to natural infestation by M. persicae up to 5 days after exposure. Previously infested plants were also more susceptible to infection with beet mild yellowing virus (BMYV) but not beet yellows virus (BYV), suggesting that the aphids on the previously infested sugar beet settled more readily and were more inclined to feed (and thus transmit BMYV) than aphids on the previously uninfested plants. The consequences for the control of sugar beet yellowing virus vectors are discussed. 相似文献

In three field experiments in 1985 and 1986, we studied the effect of the date of primary infection on the spread of beet yellows closterovirus (BYV) and beet mild yellowing luteovirus (BMW) from artificially inoculated sugar beet plants. Laboratory-reared vector aphids, Myzus persicae, were placed on these sources of virus. There was no substantial natural immigration of vectors or viruses. In two experiments, one with BMYV in 1985 and the other in BYV in 1986, populations of vector aphids remained low and there was little virus spread, i.e. c. 50 infected plants from one primarily infected source. The cause of this small amount of spread was the low number of vector aphids. In the third experiment, with BYV in 1986, large populations of M. persicae developed and there was substantial virus spread: c. 2000 infected plants in the plots which were inoculated before canopy closure. In later-inoculated plots in the same experiment, there was much less spread: c. 100 infected plants per virus source plant. Differences between fields in predator impact are implicated as the most probable factor causing differences in vector establishment and virus spread between these three experiments. Virus spread decreased with later inoculation in all three experiments. A mathematical model of virus spread incorporating results from our work has been used to calculate how the initial proportion of infected plants in a crop affects the final virus incidence. This model takes into account the effect of predation on the development of the aphid populations. The processes underlying the spread and its timing are discussed. 相似文献

The coding sequences for the variable regions of heavy and light chains of monoclonal antibodies (mAbs) to beet necrotic yellow vein virus (BNYVV) coat protein (cp) or the 25 kDa nonstructural protein (P25) were cloned into the pCOCK vector and expressed as single-chain antibody fragments (scFv) in Escherichia coli. For expression in higher plants the scFv were targeted either to the secretory pathway by including the sequences encoding the pectate lyase B (PelB) or the phytohemagglutinin (PHA) signal peptides in the vector constructs or they were targeted to the cytoplasm by omitting a signal peptide-encoding sequence from the constructs. The scFv were detected mainly in plants in which the PHA signal peptide had been used for targeting demonstrating for the first time the usefulness of this peptide for enabling scFv expression in plants. The scFv were not secreted into the culture fluids of suspension cultures, but were retained in the cells. The amount of expression of scFv in the best expressing plants was at least as high as in bacterial culture supernatants. In a dot blot immunoassay, 0.4 ng BNYVV cp or 0.8 ng P25 were detected by the respective scFv either from E. coli or from plants. The majority of the 21 plants expressing cp-specific scFv had near-normal growth whereas the three plants expressing P25-specific scFv grew poorly and did not form roots. 相似文献

Sugar beet cells grown in batch suspension culture have been used to study the regulation of polyamine levels during the transition from a quiescent to a proliferating state. The quiescent state was achieved by maintenance of the phytohormone autonomous cells in the stationary phase of the batch culture cycle. After subculture into fresh medium there was an increase in DNA synthesis which was accompanied by a dramatic increase in cellular polyamine levels. The levels of both free and bound cellular putrescine and spermidine within the cells reached a peak before the onset of the first synchronous division. The levels of putrescine, spermidine and to some extent spermine in the culture medium also increased dramatically shortly after subculture. The increase in polyamines was preceded by a rapid but transient increase in omithine decarboxylase (EC 4.1.1.17) and S -adenosylmethionine decarboxylase (EC 4.1.1.50). Arginine decarboxylase (EC 4.1.1.19) and S -adenosylmethionine synthetase (EC 2.5.1.6) activity did not show the same pattern of cell division-related variation. Inhibition of S -adenosylmethionine biosynthesis with methylglyoxal bis-(guanylhydra-zone) (MGBG) reduced cell division in the suspension culture. Inhibitors of ornithine decarboxylase and arginine decarboxylase individually had little effect on cell division, but in combination led to a reduction in cell division. Molecular markers linked to resistance genes are useful to facilitate the introgression of one or more of these genes in
breeding materials. Following the approach of bulked segregant analysis, RAPD markers linked to resistance genes against beet
necrotic yellow vein virus were identified in the four Beta accessions Holly-1-4, R104, R128 and WB42. Two primers were found which generate RAPD markers tightly linked to resistance
in segregating families of Holly-1-4, R104 and R128, indicating that the resistance genes in these accessions might be situated
at the same locus. Other, specific, primers were identified which generate RAPD markers linked to resistance in each of these
accessions. Short-range maps were established around the resistance locus in these accessions. For WB42, RAPD markers were
only identified at a relatively large distance from the resistance gene. Conversion of three RAPD primers of Holly-1-4, R104
and R128 into STS primers resulted in STS markers which can be readily used for marker-assisted selection in breeding programmes.
