The contamination of dental unit water lines (DUWL) is an emerging concern in dentistry. The aim of this study was to use an in vitro DUWL to model microbial contamination and evaluate the decontamination efficacy of tetraacetylethylenediamine (TAED) solutions. A DUWL biofilm model used to simulate clinical conditions was used to generate a range of biofilms in DUWL. Three distinct biofilms were generated: (1) biofilm from water, (2) biofilm from a mix of water + contaminating human commensal bacteria, (3) biofilm from water with contaminating oral bacteria added after biofilm formed. The contaminating oral species used were Streptococcus oralis, Enterococcus faecalis and Staphylococcus aureus. Decontamination by simple water flushing or flushing with TAED was evaluated (2, 5 and 10 min intervals). The DUWL tubes were split and samples were plated onto a range of media, incubated and bacteria enumerated. Water flushing did not reduce the number of microorganisms detected. Bacteria were not detected from any of the TAED sampling points for any of the biofilm types tested. Interestingly, if contamination was introduced to new DUWL along with the waterborne species a biofilm was formed containing only the waterborne species. If however, an existing biofilm was present before the introduction of "contaminating" bacteria then these could be detected in the biofilm. This implies that if the DUWL are new or satisfactorily cleaned on a regular basis then the associated cross-contamination aspects are reduced. In conclusion, TAED provides effective control for DUWL biofilms.  相似文献

To improve the knowledge on the risk of transmission of the caprine arthritis-encephalitis virus (CAEV) during embryo manipulations, we conducted a double-nested polymerase chain reaction (PCR) for CAEV proviral-DNA on flushing media recovered from the oviducts 48 h after the beginning of estrus and on blood from 89 donor does. Sixty-four does had negative blood and flushing media by PCR. Among the 25 CAEV infected goats (blood PCR positive), 11 were PCR flushing media positive (P < 0.01). Cell lysate from flushing media samples that were PCR positive were serially diluted 10 times at 1:100. Starting with the second 1:100 dilution all the cell lysate samples were PCR negative. The mean number of embryos recovered was not significantly different between goats with flushing media PCR positive and goats with flushing media PCR negative (6.0 +/- 5.4 versus 7.8 +/- 4.4, respectively; mean +/- S.D.) nor between goats with blood PCR positive and goats with blood PCR negative (7.0 +/- 5.0 versus 5.9 +/- 5.3; mean +/- S.D.). The presence of CAEV infected cells in oviductal flushing media from infected donor does was indicated for the first time during this study. The absence of flushing media PCR positive for goat blood PCR negative seemed to allow the use of the blood PCR test to confidently predict the absence of CAEV provirus in the oviductal fluid.  相似文献

A commercially available water purification system was evaluated for its ability to minimize chemical and microbial contaminants. The reduction or removal of these impurities from the drinking water of experimental animals would reduce experimental variability. 3 strains of bacteria were collected from the processed water. An increase in the total number of bacteria was observed the longer the filters remained in use. Determinations of heavy metals in water samples before and after processing were made for lead, zinc, copper, nickel, manganese, iron, arsenic and mercury. Calcium and magnesium levels were also determined. The concentrations of these inorganic chemicals were reduced by the purification process except at 2 time points in which desorption of the chemical could have occurred. Bacterial colonization and desorption of these chemicals were controlled by installing new filter cartridges. Volatile halocarbon concentrations were determined for water samples before and after purification. All volatile halocarbons analyzed were less than 10 ppb before and after purification at all time points. Other organic chemicals were greatly reduced by the purification process. In a study of contaminants associated with installation of the unit, it was found that flushing the unit for 8 days reduced lead and methyl ethyl ketone concentrations to insignificant levels. The purification system was found to be effective in providing high quality drinking water as verified by a microbial and chemical testing program.  相似文献

Many mosses occur both on and off serpentine soils, but experiments designed to test whether serpentine populations of widespread species display genetic adaptations for growth on serpentines have not been reported. Toward that end, two populations of the moss, Funaria flavicans, were grown on nutrient media varying in nickel and chromium concentration and in the ratio of magnesium to calcium. Three haploid siblings from each of five sporophytic families from the two populations were grown on five experimental media. There was no evidence that serpentine plants were more tolerant of nickel, chromium, high magnesium/calcium, or high nickel combined with high magnesium/calcium. In fact, plants from the nonserpentine population produced more protonemal growth than the serpentine plants on every medium except the control, on which plants from the two populations were indistinguishable. Large differences in nickel tolerance among haploid sib families (families of meiotic progeny derived from the same sporophyte) from the nonserpentine site provided evidence of genetic polymorphism in that population.  相似文献

The eutrophication model Delwaq-Bloom-Switch is developed to be a functional tool for water management. Therefore it includes nutrients, algal biomass and composition as well as water transparency. A module describing the interaction between water and bottom gives the model the flexibility to deal with measures, such as a decrease of the external phosphorus loading and flushing with water differing in composition from the lake water. This paper focuses on the functional aspects of the model, the results of an application on Lake Veluwe, The Netherlands, and the implications for water management.With one set of coefficients DBS reproduces the most important characteristics of Lake Veluwe for a period of two years before measures (reduction of the external loading and flushing during the winter months) and eight years after the measures. The phosphorus concentration decreased and became growth limiting for algae instead of nitrogen and light. Both in measurements and modelling results, the algal composition changed from blue-green algae dominance to green algae and diatom dominance. Lake Veluwe had a relatively short transient phase after reduction of external loading, because high nitrate concentrations in the flushing water inhibited a long period with high phosphorus releases from the bottom.Model calculations were carried out to investigate the effects of fish stock management and optimization of flushing. In remote areas, wild-fires often must be controlled by applying fire-retardants and suppressants dropped from small aircraft. However, impacts of these chemicals on natural stream ecosystems are poorly known. Unintentional aerial application of fire-fighting chemicals (Phos-Chek WD-881 and ForExspan S) onto two small streams during a natural wildfire on Kangaroo Island, South Australia, provided an opportunity to study the short-term effects on water chemistry and aquatic invertebrates. Within 2 weeks of application, samples of water and macroinvertebrates were collected from sites upstream of the application point, within the zone of application, and downstream where burning had been controlled on two streams. Three sites on a reference stream in the same sub-catchment that had been burned by the same fire but without application of fire-suppressants were also sampled. All sites were resampled three months later (within two weeks of the first flushing rains). There were no marked differences in water quality among the sites on the reference stream but in one of the impacted streams where flow had ceased before the fire, dissolved and total phosphorus concentrations were elevated at the site where the fire-suppressants were applied. Phosphorus concentrations were reduced 2–3-fold at this site after brief flushing by rain. Conversely, dissolved and total N and P concentrations at the other impacted stream that flowed permanently did not differ among the sites and there was no evidence for persistent changes to water quality from the applied fire-suppressant foams. Taxon richness was higher at the application and downstream sites than at upstream sites in the two impacted streams. There were also no discernible effects of the fire-suppressants on macroinvertebrate assemblage composition or taxon richness within the two streams two weeks after the chemical application or soon after flushing rains. Assemblage composition in the temporary stream was significantly different from that in the reference and the other impacted stream but also appeared unaffected by the fire-suppressants. The lack of impact on resident stream macroinvertebrates may result partly from their inherent high tolerance to the harsh physical and chemical conditions of these streams, many of which typically cease flow in summer.  相似文献

Molecular methods that permit the simultaneous detection and quantification of a large number of microbial species are currently employed in the evaluation of complex ecosystems. The checkerboard DNA-DNA hybridization technique enables the simultaneous identification of distinct bacterial species in a large number of dental samples. The original technique employed digoxigenin-labeled whole genomic DNA probes which were detected by chemiluminescence. In this study, we present an alternative protocol for labeling and detecting whole genomic DNA probes in the Checkerboard DNA-DNA hybridization method. Whole genomic DNA was extracted from five bacterial species and labeled with fluorescein. The fluorescein labeled whole genomic DNA probes were hybridized against whole genomic DNA or subgingival plaque samples in a checkerboard hybridization format, followed by chemiluminescent detection. Our results reveal that fluorescein is a viable and adequate alternative labeling reagent to be employed in the checkerboard DNA-DNA hybridization technique.  相似文献

To evaluate the effects of regular flushing, water from fifty emergency eyewash and shower stations was cultured for the presence of potentially pathogenic protozoa, heterotrophic bacteria, and Legionella species. This study also provided the opportunity to evaluate a commercially available molecular assay for the direct detection of Legionella sp in environmental samples. The Perkin Elmer Legionella EnviroAmp polymerase chain reaction (PCR) kit and culture on buffered charcoal yeast extract agar were used to detect Legionella species in water samples. Chemical and physical parameters of station water measured included: pH, hardness, alkalinity, turbidity, conductivity, total chlorine and assimilable organic carbon. Protozoal isolates were identified by classical identification methods, and isolates from the stations were identified as Hartmannella sp, Vexillifera sp, Vahlkampfia sp, Acanthamoeba sp, and Vanella sp. Heterotrophic plate counts ranged from 102 to 106 CFU ml−1 and acridine orange total counts ranged from 103 to 106 cells ml−1 after regular flushing. PCR and gene probe analysis showed that 89% of the stations (eyewash and shower) were positive for Legionella species by PCR, while 6% of the samples were culture positive. These results indicate that routine flushing alone is not sufficient to control microbial contamination and disinfection must also be included in a routine maintenance program. In addition, regular maintenance, disinfection, and monitoring of emergency eyewash and shower stations is important in preventing potential secondary microbial infections by either direct inoculation or aerosol transmission. A strategy that uses ultrafiltration (UF) to concentrate microorganisms from water samples has been developed and tested. This strategy was tested using 100-liter water samples with volume reduction achieved through ultrafiltration and recycling the microorganisms of interest through a retentate vessel, rather than returning them to the sample container, where they might pose an incremental hazard to sample takers or the environment. Three protocols based on this strategy were tested. The first protocol entailed sample volume reduction and collection of the final reduced sample. The second and third protocols both incorporated pretreatment of the filter and fluid lines with a solution to prevent microorganisms from adhering. In the second protocol, the filter was back flushed with a surfactant solution to recover microorganisms. The third protocol used recirculation of a surfactant solution to recover microorganisms. Tests were undertaken using 100-liter water samples spiked with approximately 100 or 1000 microorganisms (1 or 10 per liter). Test microorganisms included Bacillus anthracis Sterne strain, Bacillus atrophaeus subsp. globigii, and Cryptosporidium parvum. The first protocol had significantly lower recovery than the other two. Back flushing resulted in higher recovery than forward flushing, but the difference was not statistically significant.  相似文献

AIMS: To determine the level of aerosol formation and fallout within a toilet cubicle after flushing a toilet contaminated with indicator organisms at levels required to mimic pathogen shedding during infectious diarrhoea. METHODS AND RESULTS: A semisolid agar carrier containing either Serratia marcesens or MS2 bacteriophage was used to contaminate the sidewalls and bowl water of a domestic toilet to mimic the effects of soiling after an episode of acute diarrhoea. Viable counts were used to compare the numbers of Serratia adhering to the porcelain surfaces and those present in the bowl water before and after flushing the toilet. Air sampling and settle plates were used to determine the presence of bacteria or virus-laden aerosols within the toilet cubicle. After seeding there was a high level of contamination on the porcelain surfaces both under the rim and on the sides of the bowl. After a single flush there was a reduction of 2.0-3.0 log cycles cm(-2) for surface attached organisms. The number of micro-organisms in the bowl water was reduced by 2.0-3.0 log cycles ml(-1) after the first flush and following a second flush, a further reduction of c. 2.0 log cycles ml(-1) was achieved. Micro-organisms in the air were at the highest level immediately after the first flush (mean values, 1370 CFU m(-3) for Serratia and 2420 PFU m(-3) for MS2 page). Sequential flushing resulted in further distribution of micro-organisms into the air although the numbers declined after each flush. Serratia adhering to the sidewalls, as well as free-floating organisms in the toilet water, were responsible for the formation of bacterial aerosols. CONCLUSIONS: Although a single flush reduced the level of micro-organisms in the toilet bowl water when contaminated at concentrations reflecting pathogen shedding, large numbers of micro-organisms persisted on the toilet bowl surface and in the bowl water which were disseminated into the air by further flushes. SIGNIFICANCE AND IMPACT OF THE STUDY: Many individuals may be unaware of the risk of air-borne dissemination of microbes when flushing the toilet and the consequent surface contamination that may spread infection within the household, via direct surface-to-hand-to mouth contact. Some enteric viruses could persist in the air after toilet flushing and infection may be acquired after inhalation and swallowing.  相似文献

One of the many ecosystem services that mangrove systems provide is their ability to act as buffers between the land and sea, protecting human development from storm surges while also trapping terrestrial pollutants. In St. Thomas, United States Virgin Islands, an ecologically-important mangrove system sits between Bovoni Landfill and a marine protected area, the St. Thomas East End Reserves. To characterize the physical processes driving this mangrove system, groundwater hydraulic head, sediment cores, sediment surface temperatures, and water and sediment chemistry were analyzed. Hydraulic head data from January to November 2014 were used to determine vertical and horizontal groundwater flow directions. Water and sediment samples were tested for heavy metals potentially originating from Bovoni Landfill. Stratigraphic context was provided by the sediment cores and used to infer past environmental conditions. Subsamples were taken from these cores and analyzed for dry bulk density, organic matter content (through loss on ignition), and heavy metals using electron microscopy. Vertical groundwater velocity and sediment porosity were determined by calibrating a one-dimensional finite difference heat transport model to near surface temperature data from depths of 0, 7, 14, and 21 cm. Groundwater was found to flow from the terrestrial upland, through the mangroves, and toward the ocean for the majority of the study. Flow reversal was seen after long periods of little precipitation. In the surface and shallow groundwater samples, trace metal concentrations were measured from 23 to 105 μg/L for Cr, Ni, Sn, and Zn. Sediment samples collected near the landfill contained Bi, Cr, Sn, Ti, and Zn. Very slow flushing of sediment pore water was indicated by the vertical groundwater velocities produced from the heat transport model, which ranged from ±10–7 to ±10–9 m/s. This study revealed that the mangrove system is an important buffer system protecting the outer lagoon of the marine protected area from terrestrial contaminants via sediment trapping and slowing of water fluxes from the upland area into the lagoon. The results presented here can be used as a baseline for future studies and are relevant to local managers and to landfill closure plans.  相似文献

The magnetic graphene oxide (MGO) was successfully synthesised by the in situ chemical co-precipitation method with Fe3+, Fe2+ and graphene oxide (GO) in laboratory and, was used as an adsorbent for disinfection by-product (DBP) precursors removing from four natural surface water samples. The results indicate that various DBPs formation significantly decreased by 7–19% to 78–98% for the four samples after MGO treatment and, the treatment process was rapidly reached equilibrium within 20 minutes. The DBP precursors removal efficiency decreased with the increasing pH value from 4 to 10. Hydrophobic compounds (humic acid and fulvic acid) are more sensitive to MGO, whereas hydrophilic and nitrogenous compounds (aromatic proteins) are more insensitive. MGO could be regenerated by using 20% (v/v) ethanol and, the DBP precursors removal efficiency can stay stable after five cycles. These results indicate that MGO can be utilized as a promising adsorbent for the removal of DBP precursors from natural surface water.  相似文献

Background: Abernethy Forest, an area of native Caledonian Scots pine forest, includes areas of open or sparsely forested peatland, largely ombrotrophic, but locally influenced by water from mineral ground.

Aims: To relate vegetation species-composition to the limit of ombrotrophy inferred from water chemistry.

Methods: Species composition was listed in 300 sample quadrats; chemical analyses of water samples were available from 200 of these. The vegetation data were classified using TWINSPAN. Relationships between vegetation and chemical results were analysed statistically using contingency tables.

Results: Eleven ‘vegetation types’ were recognised, five (189 samples) judged as primarily ombrotrophic-bog vegetation. The others showed evidence of flushing, or transition to forest. A previous analysis of major cations in the 200 water samples indicated that a limiting Ca/Mg ratio of 1.0 divided 61 samples showing telluric influence from 139 essentially ombrotrophic samples. Neither division was sharp. Contingency tables showed that the division based on vegetation types was strongly associated with the division of the water samples based on cation ratios.

Conclusions: Large areas of bog south and east of Loch Garten are unequivocally ombrotrophic; the main telluric-influenced sites occupy openings in the forest west of the loch. The limited ‘fen’ flora probably reflects the small size and isolation of the calcium-rich sites and the dominating influence of the ombrotrophic peats.  相似文献

A method for evaluating the viability of mycobacterial cultures using fluorescein diacetate and ethidium bromide is described. Smears from 2-4 week old cultures of mycobacteria were stained with a 0.005% Dubos albumin broth dilution of fluorescein diacetate for thirty minutes at 37 C. The smears were counterstained with a 0.005% aqueous solution of ethidium bromide for ten minutes at room temperature and blotted dry. One drop of glycerol was placed on the smear and covered with a 22 × 40 mm coverslip. Stained smears were observed with fluorescence microscopy at 450 X. Viable organisms hydrolyzed the fluorescein diacetate and appeared yellow-green while dead organisms absorbed the ethidium bromide and appeared red-orange. Aliquots of material from which the slides were made were concurrently placed on Lowenstein-Jensen media and cultured for growth as a confirmation of viability. Investigation of 104 mycobacterial isolates using the described test showed that the viability of mycobacterial cultures can be determined by this method.  相似文献

Barren, subgrade serpentine substrates are difficult to revegetate due to N, P, and K deficiencies, low Ca:Mg molar ratios, potentially high levels of heavy metals including Ni, Cr, and Co, low organic matter, low CEC, and poor water holding capacity. Several large, bare roadcuts exist in the North Coast Ranges along the west coast of California, USA. Substrate was collected from a large roadcut that has remained barren for more than a decade despite conventional surface amendment with NPK fertilizer and seeding with a mix including nonserpentine grasses and herbs. Although serpentine plant communities have been studied globally in great detail, very little information exists on how to effectively restore these communities after drastic disturbance (removal of topsoil and loss of biological activity). This rhizotron study examined how surface layering and mixing yard waste compost into serpentine substrate affected biomass production, rooting distribution, and tolerance of native, as well as invasive grass species that grow on serpentine. Roots of less serpentine-tolerant species became necrotic upon contact with the serpentine substrate in the layered compost application treatment. The roots of highly serpentine-tolerant species, however, displayed a superior tolerance to the serpentine substrate and were able to grow through its entire depth. Mixing the compost into the serpentine substrate significantly improved the Ca content of the growing medium, allowing the less serpentine-tolerant species to root through the entire depth. Although the compost surface layer treatment promoted growth of the highly serpentine-tolerant species in the serpentine substrate, mixing the compost amendment into the substrate would also permit additional desirable revegetation species to become established.  相似文献

Tidal groundwater in a mangrove swamp can return to the mangrove creek by one of two mechanisms: (a) it can either flow through the swamp soil due to the water table difference between the creek and the groundwater in the swamp; or (b) it can flow via tidal flushing of animal burrows. This paper compares the magnitude of these two mechanisms for different regions of a mangrove swamp. Direct groundwater flow rates resulting from water stored in the sediment as a consequence of infiltration, especially during and after tidal inundation, were calculated for every square meter in the surface of a mangrove forest from piezometer data. Flow rates of water due to burrow flushing were determined based on published surveys, by estimating the burrow volume and the percentage of the burrow water that is flushed at each tidal inundation. Although direct groundwater flux was found to decrease further away from the creek compared to close to the creek, it was also found to have a similar range as burrow flushing flow. Specifically, direct groundwater flow ranged from 0.004 to 0.04 m3/m2/day, whilst burrow flushing flux ranged from 0.01 to 0.04 m3/m2/day.Considering the errors involved in the experiments and calculations, these ranges can be considered as being the same and neither of the two processes can be considered as negligible compared to the other. As a consequence, surveys of groundwater processes in mangrove areas, and more generally in swamp and tidal areas where animal burrows are present, will need to consider both mechanisms. Investigations of the influence over flushing mechanisms of different residence times of the water in burrows and in the sediment body would also be recommended in order to establish salt and nutrient budget in mangrove swamps.  相似文献

We have compared two methods for salvaging epididymal sperm from post-mortem samples from Iberian red deer. Of each pair of testicles (29 samples), one cauda epididymis was processed by means of cuts (sperm was immediately diluted with extender) and the other was detached from the corpus and flushed from the vas deferens with 1 mL of extender. Sperm was processed for cryopreservation, and analyzed just after recovery, pre-freezing and post-thawing. Total spermatozoa recovered, contamination (concentration of epididymal cells and red blood cells (RBCs)) and quality (motility by CASA, and acrosomal status, viability and mitochondrial status by flow cytometry) were used to compare both methods. The number of recovered spermatozoa was similar for both methods. Contamination was higher for the cuts method, but when considering the final dilution before freezing, only RBCs concentration was significantly higher. Motility was similar just after extraction, but higher for both pre-frozen and post-thawed flushed sperm. Pre-freezing acrosomal status (P < 0.05) and viability (P < 0.1) were better for flushing; however post-thawing results were similar for the two methods. A clustering analysis using CASA data showed that the subpopulation pattern of motile sperm was different depending on the method, being better for flushing. With regard to yield, lower contamination (especially RBCs) and, in general, better quality results, flushing seems to be a more recommendable method for post-mortem sperm recovery. The cuts method may be more practical on certain occasions, but care must be taken in order to achieve rapid extension of the sample and to avoid contamination in order to improve sample condition.  相似文献

Anesthetic induction chambers used for medical research are a substantial source of waste anesthetic gas (WAG). Ideally, any generated waste gas should be actively vented away from personnel operating the chamber by either a ventilated hood or snorkel. Unfortunately, the ideal environment for anesthetizing rodents is not always available. In an effort to create a safer environment, the authors designed a system to reduce WAG. This system is portable, can be adapted to different precision vaporizing anesthetic systems and fits in a variety of physical locations. The system flushes anesthetic gas out of an induction chamber before operators open the chamber. To ensure that the system was adequately flushing the anesthetic gas, the authors measured WAG concentration in the environment above the induction chamber and directly behind the vent of an activated charcoal filter. They also compared the efficiency of the filters in vertical and horizontal positions. Finally, they measured the recovery time for mice and rats after flushing the anesthetic gas from an induction chamber. The results show that flushing the induction chamber was an inexpensive and effective method for reducing WAG accumulation in the air surrounding the chamber.  相似文献

Subgrade serpentine substrates are exceptionally difficult to revegetate due to multiple limitations including low N, P, and K, low Ca:Mg molar ratios, high levels of heavy metals including Ni, Cr, and Co, low organic matter, low CEC, and low water holding capacity. To examine the influence of plant origin on the success of the revegetation of serpentine substrates, granite and serpentine accessions of Achillea millefolium were grown on subgrade serpentine substrate amended with yard waste compost, slow-release NPK fertilizer, and/or CaSO4 · 2H2O (gypsum). The goals of this study were to: (1) identify the substrate amendment combination that maximized establishment of A. millefolium on serpentine substrate, (2) compare seedling establishment, survival, and growth of the serpentine and granite A. millefolium accessions in order to determine if a serpentine edaphic ecotype of A. millefolium exists and if this ecotype is superior to the granite accession for the establishment of vegetation on serpentine substrate and (3) if a serpentine edaphic ecotype of A. millefolium does exist, what physiological features with respect to mineral nutrition convey a higher tolerance of serpentine for this ecotype than the nonserpentine ecotype. Seedling establishment, survival, and growth were greatest for A. millefolium when the subgrade serpentine substrate was amended with 30% (v/v) compost and 220 mg kg substrate−1 each of N, P, and K. The serpentine A. millefolium accession displayed a greater tolerance of the subgrade serpentine substrate, serpentine topsoil, and the amended subgrade serpentine substrate than the granite accession. Higher capacity of the serpentine A. millefolium accession for selective Ca translocation from roots to the shoot resulted in a significantly higher shoot Ca:Mg molar ratio than the granite accession and appeared to be the most important physiological feature conveying greater tolerance of the serpentine accession for serpentine substrates.  相似文献
