The sperm tail of bug insects has 9 + 9 + 2 flagellar axonemes and two mitochondrial derivatives showing two to three crystalline inclusions in their matrix. During spermiogenesis, the axoneme is surrounded by a membrane cistern which, at sperm maturity, reduces to two short cisterns on the opposite sides of the axoneme adhering to the mitochondrial derivatives. Filamentous bridges connect the intertubular material of the axoneme to these cisterns. Such bridges, which represent a peculiar feature of bug insects, are resistant to detergent treatment, whereas part of the intertubular material and the inner content of microtubular doublets are affected by the treatment. After freeze‐fracture replicas, at the insertion of the bridges to the cisternal membrane, the P‐face of this membrane shows a characteristic ribbon consisting of four rows of 11 ± 1 nm staggered intramembrane particles, 13 ± 2 nm apart along each row. The changes in the cytoplasmic organelles of sperm tail in golden hamster eggs fertilized in vivo were observed by electron microscopy. Eggs were obtained from oviducts of hamsters that had been superovulated and inseminated by injection of cauda epididymal spermatozoa into the uteri. In the egg cytoplasm 10 hours after insemination, some of the mitochondria of the spermatozoon midpiece had begun to swell, and a number of multivesicular bodies were observed surrounding the midpiece. The fibrous sheath of the principal piece quickly disappeared prior to the first cleavage, whereas the axoneme and outer dense fibers were unaltered. During the two-cell stage, numerous multivesicular bodies gathered around the midpiece and fused with the mitochondria. The heterophagic vacuoles thus formed then gradually separated from the axial fibers. The outer dense fibers were disarranged and partially torn into small segments; then they seemed to dissociate into substructural granular components. The axonemal microtubules had begun to swell but were still present in the two blastomeres. It is indicated from these observations that at least the mitochondria of the tail constituents carried into the oocyte are digested into small molecular elements by the multivesicular bodies and are possibly distributed as nutrients for the blastomeres during the early stage of development.  相似文献

The behavior of the ATPase of axoneme (detergent-treated flagellum) and dynein from sea urchin sperm was investigated. The activation of the ATPase by divalent cations was attributed to formation of a complex of ATP and the divalent cation; the metal-ATP complex is an effective substrate. However, free ATP is a modifier of the ATPase. Free ATP markedly changes the affinity of the metal-ATP complex to the enzymes. Calcium-activated ATPase activity of axoneme decreased at high concentration of CaCl2, but that of dynein did not decrease.  相似文献

The mole-cricket spermatozoon (Gryllotalpa gryllotalpa) has a motile anterior tail region and an immotile posterior region. The posterior portion appears stiff and its microtubular doublets and central singlet microtubules are swollen, apparently due to an excess of material within them. In particular, doublet number 6 is of an unusually large size. The general organization of the axoneme is also modified by a loss of dynein arms and spokes in the posterior portion. When studied by a fixation technique that involves tannic acid to outline the protein molecules and PA-TCH-Ag method for staining polysaccharides it could be seen that the microtubular doublets and accessory microtubules contain rounded globules surrounded by polysaccharides. The arrangement of protofilaments within the microtubular walls is visible both in the anterior tail region with normal doublets and in the posterior region with degenerated doublets.  相似文献

We have used Drosophila testis-specific beta2-tubulin to determine sequence requirements for different microtubules. The beta2-tubulin C-terminal tail has unique sperm-specific functions [Dev Biol 158:267-286 (2003)] and is also important for forming stable heterodimers with alpha-tubulin, a general function common to all microtubules [Mol Biol Cell 12(7):2185-2194 (2001)]. beta-tubulins utilized in motile 9 + 2 axonemes contain a C-terminal sequence "axoneme motif" [Science 275 (1997) 70-73]. C-terminal truncated beta2-tubulin cannot form the sperm tail axoneme. Here we show that a partially truncated beta2-tubulin (beta2Delta7) containing only the proximal portion of the C-terminal tail, including the axoneme motif, can support production of functional motile sperm. We conclude that these proximal eight amino acids specify the binding site for protein(s) essential to support assembly of the motile axoneme. Males that express beta2Delta7, although they are fertile, produce fewer sperm than wild type males. Beta2Delta7 causes a slightly increased error rate in spermatogenesis attributable to loss of stabilizing properties intrinsic to the full-length C-terminal tail. Therefore, beta2Delta7 males would be at a selective disadvantage and it is likely that the full-length C-terminus would be essential in the wild and in evolution.  相似文献

Sperm viability and sperm competition in insects   总被引:14,自引:0,他引:14  
Sperm quality plays an important role in vertebrates in determining which male has the advantage when two or more males compete to fertilize a female's ova. In insects, however, the importance of sperm quality has never been considered, despite sperm competition being widespread and well studied in this group. We tested the hypothesis that sperm viability, measured as the proportion of live sperm, covaried with the intensity of sperm competition in insects. In a pairwise comparison of seven closely related species pairs, each comprising a monandrous and a polyandrous species (i.e., with and without sperm competition, respectively), we found that in all cases the polyandrous species had a higher proportion of live sperm in their sperm stores. The distribution of the percentage of live sperm showed considerable inter- and intraspecific variation, suggesting that, all else being equal, males will vary in their ability to fertilize ova on the basis of sperm viability alone. Our results suggest that sperm viability is one of a suite of male adaptations to sperm competition in insects.  相似文献

Muscle and connective tissue strands linking the prothoracic cuticle to the posterior foregut and anterior midgut are described for 17 species of orthopteroid insects: Dinocras cephalotes (Plecoptera); Forficula aurkularia (Dermaptera); Acheta domesticus, Gryllus bimaculatus, Brachytrupes sp. and Phaeophyllacris spectrum (Gryllidae); Schizodactylus monstrosus (Schizodactylidae); Hemideina crassidens (Stenopelmatidae); Ephippiger ephippiger, Tettigonia viridissima, Conocephalus dorsalis and Pholidoptera griseoaptera (Tetligoniidae); Schislocerca gregaria, Omocestus viridulus and Chorthippus brunneus (Acrididae), and Carausms morosus (Phasmida). Of the 18 species examined, only the Gryllotalpa sp. (Gryllotalpidae) lacked extrinsic strands to the post-cephalic gut. The arrangement of the strands in the various species is compared, and the possible evolution and function of the strands are briefly discussed.  相似文献

Tubulin belongs to a highly conserved multigenic family, in which several gene products usually coexist in the same tissue or the same cell. Moreover, seven classes of post-translational modifications of these gene products lead to an amazing diversity of tubulin polypeptide chains, within the same cell type, whose physiological function remains elusive. Such diversity has been found in a very stable microtubular organelle, the sea urchin sperm flagellum, where some tubulin isoforms have been directly implicated in motility, whereas others may play a more structural role. In particular, polyglutamylated tubulin has been shown to be crucial for motility (Gagnon et al., 1996: J Cell Sci 109:1545 p). Here, we show with the GT335 antibody that polyglutamylated tubulin is distributed according to a decreasing gradient along the sea urchin sperm axoneme, since a semi-quantitative measurement of immunofluorescence intensity reveals that in its proximal half, the axoneme is sixfold more labeled than in its distal half. This gradient along the length of the axoneme is confirmed by immunogold labeling procedures which, in addition, demonstrate a uniform distribution of polyglutamylated tubulin among peripheral doublets and a lesser content in the central pair within a same section. Moreover, our data obtained with B3, an antibody that recognizes both mono- and poly-glutamylated tubulin, suggest that the number of glutamate residues in the lateral poly-glutamyl chain of tubulin varies along the whole length of the axoneme. These novel results coupled with those published earlier may be important to understand the role of polyglutamylation in flagellar motility.  相似文献

The axonemal organization expressed in the sperm flagella of the cecidomyiid dipteran Asphondylia ruebsaameni is unconventional, being characterized by the presence of an exceedingly high number of microtubular doublets and by the absence of both the inner dynein arms and the central pair/radial spoke complex. Consequently, its motility, both in vivo and in vitro, is also peculiar. Using monoclonal antibodies directed against posttranslational modifications, we have analyzed the presence and distribution of glutamylated and glycylated tubulin isoforms in this aberrant axonemal structure, and compared them with those of a reference insect species (Apis mellifera), endowed with a conventional axoneme. Our results have shown that the unorthodox structure and motility of the Asphondylia axoneme are concomitant with: (1). a very low glutamylation extent in the alpha-tubulin subunit, (2). a high level of glutamylation in the beta-subunit, (3). an extremely low total extent of glycylation, with regard to both monoglycylated and polyglycylated sites, either in alpha- or in beta-tubulin, (4). the presence of a strong labeling of glutamylated tubulin isoforms at the proximal end of the axoneme, and (5). a uniform distribution of glutamylated as well as glycylated isoforms along the rest of the axoneme. Thus, our data indicate that tubulin molecular heterogeneity is much lower in the Asphondylia axoneme than in the conventional 9+2 axoneme with regard to both isoform content and isoform distribution along the axoneme.  相似文献

Sea urchin sperm flagellar ATPase (EC 3.6.1.3) has magnesium-ATP as an effective substrate and is inhibited by free ATP. The inhibition is prevented by high concentration of KCl or NaCl. 0.4 M KCl extracts 48% of ATPase activity from axoneme. The 0.4 M KCl extract and 0.4 M KCl-treated axoneme are also inhibited by free ATP and this inhibition is reversed by KCl. Dynein purified twice by sucrose density gradient centrifugation is alsof inhibited by free ATP; this inhibition is also reversed by KCl.  相似文献

The formation of the sperm giant axoneme of the gall-midge fly Asphondylia ruebsaameni is described here. The axoneme consists of a great number of microtubular doublets (up to 2,500) arranged in a double spiral wrapping around an axial cluster of mitochondria. Each microtubular doublet is provided with an outer arm only. In the early spermatid the occurrence of a large system of curved multi-layered filamentous material associated with membranous cisternae has been observed in the perinuclear region. Such a system extends throughout the cytoplasm to contact the plasma membrane. The filamentous material appears to act as a nucleating centre for the assembly of the microtubular doublets, which initially have a submembranous location and later are distributed in the interior of the cell. After their assembly, microtubular doublets are associated pairwise and are arranged in a single microtubular row with a zig-zag configuration. This configuration changes during spermiogenesis as a consequence both of a rotation of the microtubular doublet pairs and a compaction of the axonemal complex due to the elimination of the excess cytoplasm. As a result of this process, a double parallel spiral of microtubular doublets is formed.  相似文献

Orcokinin (NFDEIDRSGFGFN) and orcokinin homologues are crustacean peptides eliciting potent myotropic effects in gut tissues. Through HPLC purification of brain extract of the cockroach Blattella germanica, we isolated the first insect orcokinin (NFDEIDRSGFNS). This insect orcokinin-like peptide do not show myotropic properties in B. germanica gut tissues. Gene database search using orcokinin precursor sequences of the crustacean Procambarus clarkii led to putative homologues found in non-crustacean groups, including the mosquito Anopheles gambiae and the nematode Caenorhabditis elegans.  相似文献
