The interaction of trizine aldehydes with the aldehyde binding site of bacterial luciferases was investigated using a series of triazine aldehydes with different aldehyde chain length, and substituents on the s-triazine ring. Substrate activity was determined using luciferase from Photobacterium fischeri and Vibrio harveyi in a dithionite-based luciferases assay. The chain length optimum was determined for two triazine aldehyde classes to be C-10 and C-11, respectively. Only the substrate activity of 10-(4-chloro-6-methyithio-s-triazine-2-yl)aminodecanal (5) was as high as n-decanal, the reference aldehyde. All other triazine derivatives reduced light emission, probably by hindered binding of the substrates. The degree of activity reduction correlated with the volume of the triazine ring moiety. The triazine moiety volume of compound 5 was estimated to be 200 × 10?30 m3. Triazine aldehydes which showed reduced light emission had an estimated volume of 228 × 10?30 m3 or greater. All triazine aldehydes showed approximately 10-fold lower activities for Vibrio harveyi than for Photobacterium fischeri luciferase. Substrate specificity was the same for both luciferases. A schematic superposition of quinone aldehydes and triazine aldehydes which showed substrate activities equivalent to n-decanal, indicated potential interaction sites of aldehyde substrates with the aldehyde binding site of bacterial luciferases. The in vivo relevance of the results is discussed.  相似文献

Aldehydes, a group of volatile organic compounds (VOCs) often detected in the atmosphere, play a key role in atmospheric chemistry and plant resistance to stresses. We used gas chromatography/mass spectrometry to examine the volatiles of saturated aldehydes C6–C10 that were emitted from cuttings of ashleaf maple (Acer negundo L.) under varying levels of light intensity (80, 400, and 800 μmol m−2 s−1), O2 (2% and 50%), and CO2 (600, 1,000, and 1,200 ppm). An apparent, positive correlation was found between light intensity and emissions, and their release also was significantly enhanced by higher O2 concentrations. In contrast, emissions clearly were negatively correlated with CO2 levels. We speculate that the reactive oxygen species (ROS) generated during photosynthesis contribute to these elevated emissions. However, the mechanism for this ROS trigger is unknown.  相似文献

Lipid profiles of three strains (Mexico, Australia, Japan) of Chattonella marina (Subrahmanyan) Hara et Chihara were studied under defined growth (phosphate, light, and growth phase) and harvest (intact and ruptured cells) conditions. Triacylglycerol levels were always <2%, sterols <7%, free fatty acids varied between 2 and 33%, and polar lipids were the most abundant lipid class (>51% of total lipids). The major fatty acids in C. marina were palmitic (16:0), eicosapentaenoic (EPA, 20:5ω3), octadecatetraenoic (18:4ω3), myristic (14:0), and palmitoleic (16:1ω7c) acids. Higher levels of EPA were found in ruptured cells (21.4–29.4%) compared to intact cells (8.5–25.3%). In general, Japanese N‐118 C. marina was the highest producer of EPA (14.3–29.4%), and Mexican CMCV‐1 the lowest producer (7.9–27.1%). Algal cultures, free fatty acids from C. marina, and the two aldehydes 2E,4E‐decadienal and 2E,4E‐heptadienal (suspected fatty acid‐derived products) were tested against the rainbow trout fish gill cell line RTgill‐W1. The configuration of fatty acids plays an important role in ichthyotoxicity. Free fatty acid fractions, obtained by base saponification of total lipids from C. marina showed a potent toxicity toward gill cells (median lethal concentration, LC50 (at 1 h) of 0.44 μg · mL?1 in light conditions, with a complete loss of viability at >3.2 μg · mL?1). Live cultures of Mexican C. marina were less toxic than Japanese and Australian strains. This difference could be related to differing EPA content, superoxide anion production, and cell fragility. The aldehydes 2E,4E‐decadienal and 2E,4E‐heptadienal also showed high impact on gill cell viability, with LC50 (at 1 h) of 0.34 and 0.36 μg · mL?1, respectively. Superoxide anion production was highest in Australian strain CMPL01, followed by Japanese N‐118 and Mexican CMCV‐1 strains. Ruptured cells showed higher production of superoxide anion compared to intact cells (e.g., 19 vs. 9.5 pmol · cell?1 · hr?1 for CMPL01, respectively). Our results indicate that C. marina is more ichthyotoxic after cell disruption and when switching from dark to light conditions, possibly associated with a higher production of superoxide anion and EPA, which may be quickly oxidized to produce more toxic derivates, such as aldehydes.  相似文献

The enzyme responsible for formaldehyde removal in industrial wastewaters by cells of Rhodococcus erythropolis UPV-1 was identified as a broad-specific aldehyde dehydrogenase (EC 1.2.1.3). The enzyme was purified to electrophoretic homogeneity from ethanol-grown cells with a specific activity of 19.5 U mg−1 protein and an activity recovery of 56%. The enzyme showed an isoelectric point (pI) of 5.3 and was a trimer of 162 kDa consisting of three identical 54-kDa subunits. It was specific for NAD+ and showed hyperbolic kinetics for this coenzyme (K m=90 μM), but sigmoidal kinetics for the aliphatic aldehydes used as substrates. The enzyme affinity for aldehydes increased with their hydrocarbon chain length, ranging from 333 μM for formaldehyde to 85 nM for n-octanal. The corresponding calculated Hill coefficients were in the 1.55–2.77 range. With n-propanal as substrate, the optimum pH and temperature for activity were 9.5–10.0 and 47.5°C, respectively, with an E a for catalysis of 28.6 kJ mol−1. NAD+ protected the enzyme against thermal inactivation, but aldehydes were ineffective. The activity was severely inhibited by p-hydroxymercuribenzoate, indicating that a thiol was essential for catalysis. The 1,524-bp aldhR gene encoding a 507-amino-acid protein was expressed in cells of Escherichia coli M15 as a hexahistidine-tagged protein.  相似文献

Procedures for the purification of an aldehyde dehydrogenase from extracts of the obligate methylotroph, Methylomonas methylovora are described. The purified enzyme is homogeneous as judged from polyacrylamide gel electrophoresis. In the presence of an artificial electron acceptor (phenazine methosulfate), the purified enzyme catalyzes the oxidation of straight chain aldehydes (C1-C10 tested), aromatic aldehydes (benzaldehyde, salicylaldehyde), glyoxylate, and glyceraldehyde. Biological electron acceptors such as NAD+, NADP+, FAD, FMN, pyridoxal phosphate, and cytochrome c cannot act as electron carriers. The activity of the enzyme is inhibited by sulfhydryl agents [p-chloromercuribenzoate, N-ethylmaleimide and 5,5-dithiobis (2-nitrobenzoic acid)], cuprous chloride, and ferrour nitrate. The molecular weight of the enzyme as estimated by gel filtration is approximately 45000 and the subunit size determined by sodium dodecyl sulfate-gel electrophoresis is approximately 23000. The purified enzyme is light brown and has an absorption peak at 410 nm. Reduction of enzyme with sodium dithionite or aldehyde substrate resulted in the appearance of peaks at 523 nm and 552 nm. These results suggest that the enzyme is a hemoprotein. There was no evidence that flavins were present as prosthetic group. 3-Hydroxypropionic acid (3-HP), a versatile and valuable platform chemical, has diverse industrial applications; but its biological production from glycerol is often limited by the capability of the enzyme aldehyde dehydrogenase (ALDH) to convert an intermediary compound, 3-hydroxypropionaldehyde (3-HPA), to 3-HP. In this study, we report a new ALDH, PuuC, from Klebsiella pneumoniae DSM 2026, that efficiently converts 3-HPA to 3-HP. The identified gene puuC was cloned, expressed in Escherichia coli, purified, and characterized for its properties. The recombinant enzyme with a molecular weight of 53.8 kDa exhibited broad substrate specificity for various aliphatic aldehydes, especially C2–C5 aldehydes. NAD+ was the preferred coenzyme for the oxidation of most aliphatic and aromatic aldehydes tested. The optimum pH and temperature for PuuC activity were pH 8.0 and 45°C. The K m values for 3-HPA and NAD+ were 0.48 and 0.09 mM, respectively. The activity of PuuC was enhanced in the presence of reducing agents such as 2-mercaptoethanol or dithiothreitol, while several metal ions, particularly Hg2+, Ag+, and Cu2+ inhibited its activity. The predicted structure of PuuC indicated the presence of K191 and E194 in close proximity to the glycine motif, suggesting that PuuC belongs to class 2 ALDHs.  相似文献

TiOx (x < 2) nanoparticles with tunable colors from white to gray to blue–gray to black are synthesized by magnesium (Mg) reduction of white P25 TiO2 nanocrystals followed by removal of excess Mg with aqueous HCl and distilled water. Increasing amounts of Mg smoothly decrease the oxygen content in TiOx which is responsible for the gradual increase in light absorption and concomitant darkening of its color from white to black with decreasing values of x. The as‐synthesized TiOx nanoparticles are spin‐coated onto the surface of a stainless steel mesh followed by surface superhydrophobization in order to test their performance as a solar water evaporator. Results from the tests show that the black TiOx efficiently generates water vapor with a solar thermal conversion efficiency as high as 50% under solar‐simulated light irradiance at an intensity of 1000 W m–2 (1 Sun). Moreover, TiOx nanoparticles have inherent advantages over other materials used for solar water desalination, such as their tunable light absorption, low‐cost, low‐toxicity, superhydrophobicity, and chemical stability.  相似文献

The effect of blue and red light on the adaptation to low CO2 conditions was studied in high-CO2 grown cultures of Chlorella Pyrenoidosa (82T) and Chlamydomonas reinhardtii(137+) by measuring O2 exchange under various inorganic carbon (Ci) concentrations. At equal photosynthetic photon flux density (PPFD), blue light was more favourable for adaptation in both species, compared to red light. The difference in photosynthetic oxygen evolution between cells adapted to low Ciunder blue and red light was more pronounced when oxygen evolution was measured under low Ci compared to high Ci conditions. The effect of light quality on adaptation remained for several hours. The different effects caused by blue and red light was observed in C. pyrenoidosa over a wide range of PPFD with increasing differences at increasing PPFD. The maximal difference was obtained at a PPFD above 1 500 μmol m?2s?1. We found no difference in the extracellular carbonic anhydrase activity between blue- and red light adapted cells. The light quality effect recorded under Ci-limiting conditions in C. reinhardtii cells adapted to air, was only 37% less when instead of pure blue light red light containing 12.5% of blue light (similar PPFD as blue light) was used during adaptation to low carbon. This indicates that in addition to affecting photosynthesis, blue light affected a sensory system involved in algal adaptation to low Ci conditions. Since the affinity for Ci of C. Pyrenoidosa and C. reinhardtii cells adapted to air under blue light was higher than that of cells adapted under red light, we suggest that induction of some component(s) of the Ci accumulating mechanism is regulated by the light quality.  相似文献

The photosynthesis‐irradiance response (PE) curve, in which mass‐specific photosynthetic rates are plotted versus irradiance, is commonly used to characterize photoacclimation. The interpretation of PE curves depends critically on the currency in which mass is expressed. Normalizing the light‐limited rate to chl a yields the chl a‐specific initial slope (αchl). This is proportional to the light absorption coefficient (achl), the proportionality factor being the photon efficiency of photosynthesis (φm). Thus, αchl is the product of achl and φm. In microalgae αchl typically shows little (<20%) phenotypic variability because declines of φm under conditions of high‐light stress are accompanied by increases of achl. The variation of αchl among species is dominated by changes in achl due to differences in pigment complement and pigment packaging. In contrast to the microalgae, αchl declines as irradiance increases in the cyanobacteria where phycobiliproteins dominate light absorption because of plasticity in the phycobiliprotein:chl a ratio. By definition, light‐saturated photosynthesis (Pm) is limited by a factor other than the rate of light absorption. Normalizing Pm to organic carbon concentration to obtain PmC allows a direct comparison with growth rates. Within species, PmC is independent of growth irradiance. Among species, PmC covaries with the resource‐saturated growth rate. The chl a:C ratio is a key physiological variable because the appropriate currencies for normalizing light‐limited and light‐saturated photosynthetic rates are, respectively, chl a and carbon. Typically, chl a:C is reduced to about 40% of its maximum value at an irradiance that supports 50% of the species‐specific maximum growth rate and light‐harvesting accessory pigments show similar or greater declines. In the steady state, this down‐regulation of pigment content prevents microalgae and cyanobacteria from maximizing photosynthetic rates throughout the light‐limited region for growth. The reason for down‐regulation of light harvesting, and therefore loss of potential photosynthetic gain at moderately limiting irradiances, is unknown. However, it is clear that maximizing the rate of photosynthetic carbon assimilation is not the only criterion governing photoacclimation.  相似文献

The sensory physiology and behaviour of many fish species are strongly affected by light. This short study demonstrates that in the Australasian snapper (Chrysophrys auratus) absolute light intensity governs visual acuity and also guides preference behaviour, with fish choosing to ‘rest where they see best’. Use of the optomotor response to test visual acuity at four light intensities (0.01, 0.05, 1 and 3 μmol s?1 m2), showed that visual acuity (measured as directional bias) was best at a light intensity of 0.05 μmol s?1 m2 (84.9% directional bias), but weakened at the highest and lowest light intensity (41.1 and 35.3%). When provided with a choice of the same four light environments fish also spent most time in the 0.05 μmol s?1 m2 light environment, while the highest and lowest intensity light environments were usually avoided. Acclimated light intensity (that is daytime light intensity of the home aquarium) was also 0.05 μmol s?1 m2. By selecting an environment where visual function is optimised, C. auratus are also optimising the ability to search for prey and to detect predators. The results are discussed with reference to habitat utilisation and thoughts for trawl fishing practices.  相似文献

High ratios of dissolved organic nitrogen (DON) to dissolved inorganic nitrogen (DIN) have been suggested to favor the growth of the brown tide alga Aureococcus anophagefferens. DON could provide a particular advantage in low light levels, as occur when blooms induce self-shading. We examined the effects of varying DON:DIN ratios on the photosynthetic abilities of cultured Aureococcus at two light intensities, 93 and 17 μmol photons m−2 s−1. Glutamic acid and urea were used as DON sources, and the remainder of the nitrogen was added as nitrate.In experiments examining Aureococcus growth with varying ratios of DONGlu:DINNitrate at two light intensities in batch culture, higher growth rates and biomass were observed in treatments containing DIN than in those with DON only, which contrasts with the results of previous studies. In semi-continuous growth experiments with varying DONUrea:DINNitrate ratios, low light cultures with urea had higher growth rates than those without urea. Also, the effective target area for light absorption per cell and photosystem II efficiency were greater for the low light cultures of each nutrient treatment, particularly when DON:DIN mixtures (33 and 67% NUrea) were used. The same pattern was seen in the maximum photosynthetic rates per cell in the light-saturated (Pmcell) and in the initial slope (αcell) of the PE (photosynthesis versus irradiance) curve, and in PON, POC and chlorophyll a cell−1. This indicates that the ability of Aureococcus to acclimate to low light conditions may be enhanced by the presence of both organic and inorganic nitrogen sources. These results suggest that Aureococcus physiology and photosynthesis are different during growth on a mixture of urea-N and nitrate than when either nitrogen source is present alone. Results of this study suggest that Aureococcus may not respond to all DON substrates in the same way, and that mixtures of DON and DIN may provide for higher photosynthetic rates, especially at low light. Our results did not, however, support earlier suggestions that growth on DON alone provides the brown tide alga with a large advantage at low light levels.  相似文献

Effects of growth light intensity on the temperature dependence of CO2 assimilation rate were studied in tobacco (Nicotiana tabacum) because growth light intensity alters nitrogen allocation between photosynthetic components. Leaf nitrogen, ribulose 1·5‐bisphosphate carboxylase/oxygenase (Rubisco) and cytochrome f (cyt f) contents increased with increasing growth light intensity, but the cyt f/Rubisco ratio was unaltered. Mesophyll conductance to CO2 diffusion (gm) measured with carbon isotope discrimination increased with growth light intensity but not with measuring light intensity. The responses of CO2 assimilation rate to chloroplast CO2 concentration (Cc) at different light intensities and temperatures were used to estimate the maximum carboxylation rate of Rubisco (Vcmax) and the chloroplast electron transport rate (J). Maximum electron transport rates were linearly related to cyt f content at any given temperature (e.g. 115 and 179 µmol electrons mol?1 cyt f s?1 at 25 and 40 °C, respectively). The chloroplast CO2 concentration (Ctrans) at which the transition from RuBP carboxylation to RuBP regeneration limitation occurred increased with leaf temperature and was independent of growth light intensity, consistent with the constant ratio of cyt f/Rubisco. In tobacco, CO2 assimilation rate at 380 µmol mol?1 CO2 concentration and high light was limited by RuBP carboxylation above 32 °C and by RuBP regeneration below 32 °C.  相似文献

A study has been made on the possibility of replacing leucofuchsin by colored basic fuchsin for the histochemical demonstration of aldehydes. Several tissues from mammals and various pertinent fixatives were used. Aldehydes were freed from carbohydrates by oxidation and from thymonucleic acid by hydrolysis.

It was found that the colored form and not necessarily the leucoform of basic fuchsin can be used histochemically in demonstrating aldehydes. The technic used is as follows: (1) Treat with 1.0–0.5% H5IO6 (or in 1% KIO4 in M/1 H2SO4) for 5 to 10 min. and wash thoroughly. For thymonucleic acid hydrolize with N HCl 5 min. at room temperature, 10 min. at 60°C. and 5 min. at room temperature. (2) Stain for 2–3 min. with 0.05% basic fuchsin in 5% ethanol, 3% phenol. (3). Transfer immediately to 1 or 2 changes of 1% sodium bisulphite or potassium metabisulphite in 0.1–0.2 N H2SO4 for a total of 5 min. (4) Rinse with water and treat with M H2SO4 in 95% ethanol for 3–5 min. 6. Wash thoroughly in water and dehydrate, clear, and mount. For glycogen and mucin the following counterstaining solution is recommended: orange G, 0.25 g.; light green SFY, 0.10 g.; phosphotungstic acid 0.50 g.; 50% ethanol, 100 ml.; glacial acetic acid, 0.25 ml.  相似文献

Two populations of a diatom alga Thallassiosira weisflogii were grown at photon flux densities (PFD) of 0.8 and 8 μmol/(m2 s). For both diatom populations, the recovery of chlorophyll fluorescence parameters (F 0, F m, F v/F m, and NPQ) was monitored after nondestructive irradiation by visible light at PFD of 40 μmol/(m2 s) and after high-intensity irradiation by visible light (1000–4000 μmol/(m2 s)). The exposure of diatoms to PFD of 40 μmol/(m2 s)—higher than PFD used for algal growth but still nondamaging to photosynthetic apparatus—induced nonphotochemical quenching (NPQ), which was stronger in algae grown at higher PFD (8 μmol/(m2 s)) than in algae grown at low light. After irradiation with high-intensity light, the recovery of chlorophyll fluorescence parameters was more pronounced in algae grown at elevated PFD level. During short-term irradiation of diatoms with high-intensity visible light (1000 μmol/(m2 s)), a stronger NPQ was observed in the culture adapted to high irradiance. After the treatment of algae with dithiothreitol (an inhibitor of carotenoid deepoxidase in the diadinoxanthin cycle) or NH4Cl (an agent abolishing the proton gradient at thylakoid membranes), a short exposure of algae to PFD of 40 μmol/(m2 s) induced hardly any nonphotochemical quenching. The results indicate the dominant contribution of xanthophyll cycle carotenoids to energy-dependent quenching.  相似文献

If one wishes to distinguish chromatic effects from irradiance effects on metabolism. it is technically invalid to expose cells lo irradiances of equal photosynthetically available radiation (PAR) unless PAR is absorbed in totality. Therefore, the effects of blue light on growth and cellular concentrations of carbon, protein and carbohydrate of the diatom Chaetoceros protuberans Lauder, (in semi-continuous cultures), were studied by exposing the cells to irradiances of equal photosynthetically usable radiation (PUR), of white (PURw) and blue (PURb) light. Three average levels: PURw=PURb=56, 125 and 13 μ m?2.s?1 were used in the stated sequence. With increasing PUR the relative concentration of carbohydrate increased in both types of light. The efficiency of this increase was not modified by blue light. The relative concentration of protein remained constant in white light out the efficiency of net protein production improved at the lowest blue light irradiance. The saturation threshold of this chromatic effect was higher in C. proluberans than in the Chlorophyceae. The chromatic change did not affect the average cell doubling rate, calculated over (5 or 6 days) an observation which does not agree with recently published work.  相似文献

The first successful enantioseparation of representative O,O‐diphenyl‐N‐arylthioureidoalkylphosphonates, (±)‐Ptc‐ValP(OPh)2 & (±)‐Ptc‐LeuP(OPh)2 and thiourylenedi(isobutyl phosphonate), Tcm[ValP(OPh)2]2 on analytical and semipreparative scale was achieved by high‐performance liquid chromatography using polysaccharide‐based chiral stationary phases (CPs). Atc‐AAP(OPh)2 was obtained using modified tricomponent condensations of the corresponding aldehydes, N‐arylthiourea and triphenyl phosphite whereas Tcm[ValP(OPh)2]2 by the condensations of aldehydes, thiourea, and triphenyl phosphite. The prepared, racemic (±)‐Atc‐AAP(OPh)2 [(±)‐Ptc‐ValP(OPh)2, (±)‐Ptc‐LeuP(OPh)2, (±)‐Ptc‐PglyP(OPh)2 and (±)‐Ntc‐PglyP(OPh)2] and racemic (±)‐Tcm[AAP(OPh)2]2 [(±)‐Tcm[NvaP(OPh)2]2 & (±)‐Tcm[ValP(OPh)2]2] were adequately characterized and used for chromatographic separations on high‐performance liquid chromatography–chiral stationary phases. The best results were obtained for (±)‐Ptc‐ValP(OPh)2, (±)‐Ptc‐LeuP(OPh)2 and (±)‐Tcm[ValP(OPh)2]2.  相似文献

Oxidative deamination of putrescine, the precursor of polyamines, gives rise to γ-aminobutyraldehyde (ABAL). In this study an aldehyde dehydrogenase, active on ABAL, has been purified to electrophoretic homogeneity from rat liver cytoplasm and its kinetic behaviour investigated. The enzyme is a dimer with a subunit molecular weight of 51,000. It is NAD+-dependent, active only in the presence of sulphhydryl compounds and has a pH optimum in the range 7.3–8.4. Temperatures higher than 28°C promote slow activation and the process is favoured by the presence of at least one substrate. Km for aliphatic aldehydes decreases from 110 μM for ABAL and acetaldehyde to 2–3 μM for capronaldehyde. The highest relative V-values have been observed with ABAL (100) and isobutyraldehyde (64), and the lowest with acetaldehyde (14). Affinity for NAD+ is affected by the aldehyde present at the active site: Km for NAD+ is 70 μM with ABAL, 200 μM with isobutyraldehyde and capronaldehyde, and>800 μM with acetaldehyde. The kinetic behaviour at 37°C is quite complex; according to enzymatic models, NAD+ activates the enzyme (Kact 500 μM) while NADH competes for the regulatory site (Kin 70 μM). In the presence of high NAD+ concentrations (4 mM), ABAL promotes further activation by binding to a low-affinity regulatory site (Kact 10 mM). The data show that the enzyme is probably an E3 aldehyde dehydrogenase, and suggest that it can effectively metabolize aldehydes arising from biogenic amines.  相似文献

Abstract Field gas exchange measurements on intact peach (Prunus persica (L.) Batsch) leaves indicate that leaf nitrogen content (NL) and leaf weight per unit leaf area (Wa) are highly correlated with CO2 assimilation rate (A) and mesophyll conductance (gm). Therefore, NL and Wa were used to study seasonal relationships between leaf carboxylation capacity and natural light exposure in tree canopies. From mid-season onwards, NL and Wa were linearly correlated with light exposure expressed as the amount of time during a clear day that a leaf was exposed to a photosynthetic photon flux density (Q) of ≥ 100 μmol m?2 s?1. The data support the hypothesis that whole-tree photosynthesis is optimized by partitioning of photosynthetic capacity among leaves in deciduous tree canopies with respect to natural light exposure.  相似文献

The αβ subunits of the tungsten-containing reversible aldehyde oxidoreductase of Clostridium thermoaceticum were shown to contain a pterin cofactor in the form of a mononucleotide. The substrate specificity of the enzyme for aliphatic and aromatic aldehydes and for carboxylates was broad. The K m values for ethanal, propanal and butanal were 0.010–0.006 mM, but the value for methanal was 1.6 mM. Benzaldehyde derivatives with a hydroxy group in the 4-position showed millimolar K m values that were 2–3 orders of magnitude higher than those of other aromatic and aliphatic aldehydes. The ratio of k cat /K m for aldehydes and the corresponding acids is 104–105. For carboxylate reduction, 4-hydroxy benzoate again showed the highest K m value of all substrates tested. When the 4-hydroxy groups of the aldehyde and the acid were methylated, the K m values were decreased drastically. From the temperature dependence of carboxylate reduction at the expense of viologens, activation energies that depended on the substrate and on the applied viologen were calculated. The pH optima of the carboxylate reductions depended on the pK values of the acids and shifted to lower pH values with lower pK values of the acids. The ternary complex α3β3γ of the aldehyde oxidoreductase was able to dehydrogenate aldehydes to acylates with NADP+. Surprisingly the reverse reaction was observed too, although at very low rates. When exposed to air, the aldehyde oxidoreductase showed markedly enhanced lability in its reduced state compared to its oxidized state. To test the hypothesis that in temperate deciduous trees acclimation to potentially damaging high irradiances occurs via long-term adjustments in foliar photosynthetic capacity, and short-term changes in xanthophyll cycle pool size in response to weather fluctuations, nitrogen concentration and pigment composition were examined along a canopy light gradient in three species –Betula pendula, Populus tremula and Tilia cordata (from most shade intolerant to tolerant), and foliage photosynthetic potentials in P. tremula and T. cordata. Integrated quantum flux density (Qi) incident on leaves was estimated with a method combining hemispherical photography and light measurements with quantum sensors made over the growing season. Long- and short-term light indices – average total seasonal daily integrated quantum flux density (Ts, mol m–2 d–1) and that of the 3 d preceding foliage sampling (T3d) – were calculated for each sampled leaf. In addition to total integrated quantum flux density, the part of Qi attributable to direct flux was also computed. Strong linear relationships between the capacity for photosynthetic electron transport per area (Jamax), estimated from in situ measurements of effective quantum yield of photosystem II (PS II), and Qi averaged over the season and over the preceding 3 d were found for all studied species. However, the major determinant of Jamax, the product of electron transport capacity per leaf dry mass (Jmmax) and leaf dry mass per area (MA), was MA rather than Jmmax, which was relatively constant along the light gradient. There was evidence that Jamax is more tightly related to Ts, which characterizes the light climate during foliar development, than to short-term integrated light, possibly because there is little flexibility in adjustments in MA after the completion of foliar growth. Leaf chlorophyll concentrations and the investment of leaf nitrogen in chlorophyll (Chl/N) were negatively related to Qi– an investment pattern which improves light harvesting in low light. Xanthophyll cycle pool size (VAZ, violaxanthin + antheraxanthin + zeaxanthin) either expressed per unit chlorophyll (VAZ/Chl) or as a fraction of total carotenoids (VAZ/Car) increased with increasing Qi in all species. However, contrary to Jamax, it tended to correlate more strongly with short-term than with long-term average integrated light. There were few interspecific differences in Jamax, Chl/N, VAZ/Chl and VAZ/Car when the variability in light level incident to the leaves was accounted for, indicating that the foliage of both shade-intolerant and -tolerant temperate tree species possesses considerable phenotypic flexibility. Collectively these results support the view that rapid adjustment of the xanthophyll cycle pool size provides an important means for acclimation to light fluctuations in a time scale of days, during which the potential for photosynthetic quenching of excitation energy is not likely to change appreciably.  相似文献
