Scanning electron microscopic study of Opisthorchis viverrini tegument and its alterations induced by amoscanate. International Journal for Parasitology16: 19–26. When examined by scanning electron microscopy, the surface of adult Opisthorchis viverrini is covered with short microvilli that are closely packed together. Microvilli are more numerous and are taller on the ventral surface. Distributed among microvilli are two types of papillae, each one with a dome-shaped base (approx. 3 μm in diameter) with a projecting cilium in one case but not the other. Papillae are scattered in groups over the surface but are especially numerous around the suckers and laterally.When the flukes were treated with a potent schistosomicidal agent, amoscanate (C 9333-Go/CGP 4540), the tegument was damaged. Lesions that occurred on the flukes recovered from infected hamsters 1, 9, 30 and 90 days after treatment were compared with those which occurred when the flukes were exposed to the agent in vitro. Total disruption of the basic structure of the tegument was noted within 2 h of in vitro incubation with 1% amoscanate; the damage was more severe after 24 h of treatment. Flukes obtained from hamsters 24 h after treatment for 4 consecutive days with a total of 40 mg also showed tegumental lesions, including pronounced swellings into large bulbs that eventually ruptured and sloughed. However, complete regeneration of the tegument was noted within 30 days after treatment.  相似文献

Mycobacterium ulcerans infection causes a neglected tropical disease known as Buruli ulcer that is now found in poor rural areas of West Africa in numbers that sometimes exceed those reported for another significant mycobacterial disease, leprosy, caused by M. leprae. Unique among mycobacterial diseases, M. ulcerans produces a plasmid-encoded toxin called mycolactone (ML), which is the principal virulence factor and destroys fat cells in subcutaneous tissue. Disease is typically first manifested by the appearance of a nodule that eventually ulcerates and the lesions may continue to spread over limbs or occasionally the trunk. The current standard treatment is 8 weeks of daily rifampin and injections of streptomycin (RS). The treatment kills bacilli and wounds gradually heal. Whether RS treatment actually stops mycolactone production before killing bacilli has been suggested by histopathological analyses of patient lesions. Using a mouse footpad model of M. ulcerans infection where the time of infection and development of lesions can be followed in a controlled manner before and after antibiotic treatment, we have evaluated the progress of infection by assessing bacterial numbers, mycolactone production, the immune response, and lesion histopathology at regular intervals after infection and after antibiotic therapy. We found that RS treatment rapidly reduced gross lesions, bacterial numbers, and ML production as assessed by cytotoxicity assays and mass spectrometric analysis. Histopathological analysis revealed that RS treatment maintained the association of the bacilli with (or within) host cells where they were destroyed whereas lack of treatment resulted in extracellular infection, destruction of host cells, and ultimately lesion ulceration. We propose that RS treatment promotes healing in the host by blocking mycolactone production, which favors the survival of host cells, and by killing M. ulcerans bacilli.  相似文献

The mermithid parasite Heleidomermis magnapapula was maintained in larvae of the midge Culicoides variipennis for 20 months in enamel pans containing nutrient-rich water and polyester pads as a substrate. Inseminated female mermithids were introduced to the pad surface when the host was in the late second or early third-instar. Host larvae were harvested from the pans 9 days after exposure and held in tap water for nematode emergence. Preparasite yield was positively correlated with female nematode size and averaged 1,267 preparasites/female. Male and female nematodes emerged an average of 12.2 and 13.4 days after host exposure, respectively. Supplemental host food (Panagrellus) during the final days of parasitism did not alter time of emergence. Parasites emerging singly were 64% females, whereas superparasitized hosts yielded males (up to nine/host). Nematode carryover into the adult midge normally occurred at a level of 0.5-2.5%. Parasite load (nematodes/ parasitized individual) in midge adults was lower than that of larvae from the same cohort, and adult midges were more likely to harbor female parasites. Exposure of fourth-instar host larvae resulted in higher levels of adult parasitism (up to 17%).  相似文献

In order to avoid excess feed consumption during the force-feeding period in foie gras production, a dose-response experiment with seven feed consumption levels (450, 540, 630, 720, 810, 900, 990 g/day per bird) was conducted to evaluate the effects of feed consumption levels on growth performance and carcass composition of male Mule ducks from 91 to 102 days of age. One-day-old Mule ducklings (sterile and artificial hybrid of male Albatre Muscovy duck and female Pekin duck were fed a two-phase commercial diets for ad libitum intake from hatching to 91 days of age, followed by graded feeding levels of a corn diet by force-feeding from 91 to 102 days of age. Fifty-six 91-day-old male Mule ducks with similar BW were randomly assigned to seven treatments, with eight birds per treatment. Birds were housed in individual pens. At 102 days of age, final BW was measured and BW gain and feed conversion ratio of ducks from each treatment were calculated from day 91 to 102, and then all ducks were slaughtered to evaluate the yields of skin with subcutaneous fat, abdominal fat, breast meat (including pectoralis major and pectoralis minor), leg meat (including thigh and drum stick), and liver. Significant differences in BW gain, total liver weight and liver relative weight were observed among the treatments (P<0.001). According to the broken-line regression analysis, the optimal feed consumption levels of male Mule ducks from 91 to 102 days of age for maximum BW gain, total liver weight and liver relative weight were 217, 227 and 216 g feed/kg BW0.75·per day, respectively.  相似文献

An initial natural infection of Saurocytozoon tupinambi in a juvenile Tupinambis teguixin from Venezuela was studied for 131 days following capture of the host. Intralymphocytic parasites appeared in this sequence: small uninucleate and binucleate stages (days 1–31 and again on day 41); schizonts with 3–102 nuclei (days 8–14 and 29–35); immature gametocytes (days 29–35) and apparently mature gametocytes of Saurocytozoon tupinambi from day 41. Maximum parasitemia of trophozoites and binucleate schizonts occurred on day 4 when 11% of lymphocytes were infected. Maximum parasitemia by larger schizonts occurred on day 8 at 0.13% of lymphocytes, while maximum gametocytemia was found on day 49 with 16.4% of lymphocytes parasitized. Two types of schizonts were observed: intralymphocytic and the same type free of host cells, and fragments of varying size which may have been torn from capillary endothelium.Due to presence of concurrent infection by a small Plasmodium species, identity of intralymphocytic asexual stages with S. tupinambi cannot be established. Presence of asexual and sexual stages in the same type of host cells (lymphocytes and close derivatives), sequential appearance of trophozoites, schizonts and gametocytes over a period of 40 days, and correlated fluctuations in lymphocyte density suggest they are conspecific, and that Saurocytozoon, which has a plasmodiid type of sporogony may prove to further differ from leucocytozoids by presence of an asexual cycle in circulating blood cells.  相似文献

Wildlife reservoirs of Mycobacterium bovis represent serious obstacles to the eradication of tuberculosis from livestock, particularly cattle. In Michigan, USA tuberculous white-tailed deer transmit M. bovis to other deer and cattle. One approach in dealing with this wildlife reservoir is to vaccinate deer, thus interfering with the intraspecies and interspecies transmission cycles. Thirty-three white-tailed deer were assigned to one of two groups; oral vaccination with 1×108 colony-forming units of M. bovis BCG Danish (n = 17); and non-vaccinated (n = 16). One hundred eleven days after vaccination deer were infected intratonsilarly with 300 colony-forming units of virulent M. bovis. At examination, 150 days after challenge, BCG vaccinated deer had fewer gross and microscopic lesions, fewer tissues from which M. bovis could be isolated, and fewer late stage granulomas with extensive liquefactive necrosis. Fewer lesions, especially those of a highly necrotic nature should decrease the potential for dissemination of M. bovis within the host and transmission to other susceptible hosts.  相似文献

We studied the role of surface GlcNAc residues on the surface of invasive (mouse-blood and insect-derived trypomastigotes) and non-invasive amastigote forms of Trypanosoma cruzi on parasite association with (i.e., surface binding plus internalization) macrophages and heart myoblasts. Removal of GlcNAc from the three forms of the parasite with β-N-acetylglucosaminidase markedly increased the number of organisms per 100 cells and caused the organisms to associate with a greater percentage of host cells. N-Acetylglucosaminidase did not produce this effect after heat-inactivation and a substrate of the enzyme, N,N′-diacetylchitobiose, reduced it when it was present during the enzymatic treatment. The N-acetylglucosaminidase effect on T. cruzi was reversible after 2.5 h. When macrophages or myoblasts were treated with N-acetylglucosaminidase, their capacities to associate with blood or insect-derived trypomastigotes was reduced. Since removal of GlcNAc residues from the parasite surface increased their association with the host cells, GlcNAc would appear to interfere with the association process. On the other hand, GlcNAc residues on the host cell appear to favor the association.  相似文献

Systematic dose- and time-response trials were first conducted to determine a treatment regimen with cambdendazole (CBZ, 2-[4′-thiazolyl]-5-isopropoxycarbonylaminobenzimidazole) that would be therapeutically effective against adult Trichinella spiralis in experimentally-infected mice. CBZ proved to be highly effective against adult Trichinella in a 3-day treatment course during the enterai phase of experimental trichinellosis. When treatment began 72 h after the mice were inoculated with parasites, the number of adult worms recovered from the host intestine was greatly reduced by twice-daily oral doses of CBZ at 25 mg kg?1 body wt. The efficacy of the divided daily oral dosage treatment regimen that was effective against adult T. spiralis during the enteral phase of infection was then tested during the parenteral phases of trichinellosis. Gavage administration of CBZ at 25 mg kg?1bis m die for 3 consecutive days during the invasive (days 14–16) and encystment (days 28–30) phases of infection significantly reduced (29 and 90 %, respectively) the number of larvae subsequently recovered from the host musculature on day 44 postinoculation.  相似文献

Helicosporidium sp. is a pathogenic alga that replicates in the hemolymph of various invertebrate hosts. Morphogenesis of the infectious life stage, the cyst, occurs in the infected host, but to date cannot be induced in vitro. Using larvae of the heterologous host Helicoverpa zea, we examined potential factors influencing pathogenicity and in vivo cyst production of the alga and the impact of infection on host survival. Factors tested were cyst dosage administered per os (ranging from 102 to 105 cysts per larva) and host age at exposure (third, fourth, and fifth larval instar). Cyst production occurred between 7 and 13 days after treatment, regardless of host age at treatment. Increasing dosage increased both percent infection and mortality, but cyst production did not track the total infection response. Increasing host age at exposure mitigated dosage effects on infection and mortality and also elevated cyst production in later-treated larvae. Only the highest dosage produced a significant decrease in the overall time to death. Moderate cyst dosages and later host ages were most effective at regenerating Helicosporidium cysts.  相似文献

Higher populations of Meloidogyne incognita larvae and Pratylenchus penetrans were recovered from soil treated with carbofuran 10 and 15 days after treatment, respectively, than were recovered from untreated control soil. The number of P. penetrans, however, was lower 50 days after treatment, and symptoms developed only occasionally on the root systems of host plants. Populations of Tylenchorhynchus claytoni inoculated at different distances from the base of corn seedlings growing in carbofuran-treated soil did not move toward the plant, whereas they were attracted in untreated soil from a distance of 12 cm. P. penetrans moved at random in treated agar medium when inoculations occurred 4 cm away from the root tips of tomato seedlings under aseptic conditions. Those nematodes that reached the roots were never observed feeding during a 20-day observation period. Specimens of P. penetrans placed on the developing roots moved at random and never penetrated. In contrast, numerous P. penetrans penetrated roots of seedlings growing in untreated medium.  相似文献

Background

The neglected tropical disease Buruli ulcer (BU) caused by Mycobacterium ulcerans is an infection of the subcutaneous tissue leading to chronic ulcerative skin lesions. Histopathological features are progressive tissue necrosis, extracellular clusters of acid fast bacilli (AFB) and poor inflammatory responses at the site of infection. After the recommended eight weeks standard treatment with rifampicin and streptomycin, a reversal of the local immunosuppression caused by the macrolide toxin mycolactone of M. ulcerans is observed.

Methodology/Principal Findings

We have conducted a detailed histopathological and immunohistochemical analysis of tissue specimens from two patients developing multiple new skin lesions 12 to 409 days after completion of antibiotic treatment. Lesions exhibited characteristic histopathological hallmarks of Buruli ulcer and AFB with degenerated appearance were found in several of them. However, other than in active disease, lesions contained massive leukocyte infiltrates including large B-cell clusters, as typically found in cured lesions.

Conclusion/Significance

Our histopathological findings demonstrate that the skin lesions emerging several months after completion of antibiotic treatment were associated with M. ulcerans infection. During antibiotic therapy of Buruli ulcer development of new skin lesions may be caused by immune response-mediated paradoxical reactions. These seem to be triggered by mycobacterial antigens and immunostimulators released from clinically unrecognized bacterial foci. However, in particular the lesions that appeared more than one year after completion of antibiotic treatment may have been associated with new infection foci resolved by immune responses primed by the successful treatment of the initial lesion.  相似文献

A Russian knapweed (Acroptilon repens) shoot culture system, initiated from shoot tip culture, was used to generate a source of host plant tissue for the rearing of the nematode Subanguina picridis, a biocontrol agent for Russian knapweed. Young shoots growing on solid B5G medium in petri dishes developed galls on leaves, petioles, and shoot tips 7 days after release of 50 nematodes onto the surface of the medium. After 3 months of culturing, each petri dish yielded 7,000-10,000 nematodes. In vitro cultured Subanguina picridis were virulent on greenhouse-grown Russian knapweed plants. Galls were first found on seedlings 12 days after infestation; after 2 months, 90% of seedlings were galled on leaves, petioles, and shoot tips, with 1-6 galls per seedling. Three months after shoot emergence, 64% of vegetative shoots originating from root segments were also galled by the cultured nematodes. Similarly, vegetatively regenerated shoots of Russian knapweed were also susceptible to infestation by cultured nematodes.  相似文献

The calyx fluid in the lateral oviduct of a gregarious parasitoid, Apanteles glomeratus contained ellipsoid particles of ca. 130 × 200 nm. These calyx fluid particles did not appear to be embedded in a fibrous outer layer on the surface of eggs in the lateral oviduct. They were not observed on the surfaces of the eggs 3 to 4 hr after being deposited into the host haemocoele. Oviposition experiments indicated that the occurrence of haemocytic defence reactions of the late 2nd instar larvae of the Pieris rapae crucivora against 1 st instar larvae of the parasitoid increased with a decreasing number of the parasitoid eggs introduced into a host, and that more than 5 to 9 parasitoid eggs were needed for suppressing the ability of the host to encapsulate its parasitoid larvae immediately after hatching. When eggs with calyx fluid obtained from egg reservoir were injected into the host, they were found to be encapsulated 1 to 2 days after the injection. They could not start their embryonic development. When calyx fluid-free 3-hr-old eggs were injected in a number of more than 5 eggs into a 5th instar larva of Pieris, 58% of 31 eggs injected had normally hatched without evoking encapsulation reactions by the host. Both electron microscopic observations of parasitoid eggs in the host haemocoele and the experimental results suggested that calyx fluid or calyx fluid particles of the parasitoid might not be involved in the encapsulation-inhibiting activity of the parasitoid eggs. Rather it was anticipated that a substance (or substances) might be secreted by the parasitoid eggs into the haemocoele of the host, which suppressed defence reactions of the host.  相似文献

When immune NIH mice were killed 10 days after a challenge infection with Nematospiroides dubius, approximately 10% of the inoculated larvae were recovered from the intestinal lumen, irrespective of the dose administered. When such mice were treated with cortisone from Day 10 for a period of 8 to 14 days and were subsequently killed for worm counts, it was found that they had significantly more worms than the immune control mice killed on Day 10. During the week following the beginning of treatment with cortisone there was little change in the low worm burdens in immune mice. However, 9 to 11 days after this treatment worm counts indicated that worms were accumulating in the intestinal lumen, and concurrently eggs were recorded in the feces of the mice. These observations indicated that a period of 9 to 11 days was required after the initiation of cortisone treatment on Day 10 for the worms in immune mice to complete their development to the adult lumen-dwelling stage. It is suggested that the larvae in the challenge infection became arrested early in their development in the intestinal wall and that growth resumed only after cortisone treatment. When treatment with cortisone was initiated later after challenge, it was still effective in reactivating arrested worms, but the lower worm recoveries in these mice indicated that the arrested larvae were being slowly rejected by the host. In subsequent experiments it was established that the arrested larvae of N. dubius were insusceptible to the activity of pyrantel embonate, an anthelmintic which is 99% effective against adult worms in the intestinal lumen. The mechanism whereby the larvae of N. dubius became arrested in immune mice and subsequently resumed their development after cortisone treatment is discussed.  相似文献

The rice blast fungus Magnaporthe oryzae is a global food security threat due to its destruction of cultivated rice. Of the world's rice harvest, 10–30 % is lost each year to this pathogen, and changing climates are likely to favor its spread into new areas. Insights into how the fungus might be contained could come from the wealth of molecular and cellular studies that have been undertaken in order to shed light on the biological underpinnings of blast disease, aspects of which we review herein. Infection begins when a three-celled spore lands on the surface of a leaf, germinates, and develops the specialized infection structure called the appressorium. The mature appressorium develops a high internal turgor that acts on a thin penetration peg, forcing it through the rice cuticle and into the underlying epidermal cells. Primary then invasive hyphae (IH) elaborate from the peg and grow asymptomatically from one living rice cell to another for the first few days of infection before host cells begin to die and characteristic necrotic lesions form on the surface of the leaf, from which spores are produced to continue the life cycle. To gain new insights into the biology of rice blast disease, we argue that, conceptually, the infection process can be viewed as two discrete phases occurring in markedly different environments and requiring distinct biochemical pathways and morphogenetic regulation: outside the host cell, where the appressorium develops in a nutrient-free environment, and inside the host cell, where filamentous growth occurs in a glucose-rich, nitrogen-poor environment, at least from the perspective of the fungus. Here, we review the physiological and metabolic changes that occur in M. oryzae as it transitions from the surface to the interior of the host, thus enabling us to draw lessons about the strategies that allow M. oryzae cells to thrive in rice cells.  相似文献

Rats were passively protected against challenge infection with Taenia taeniaeformis by the administration of immune serum. Macroscopic liver lesions were rarely seen following challenge. The fate of oncospheres was determined by histological examination of rat livers at various intervals after infection, and also by in vitro experimentation.Oncospheres were killed soon after exposure to immune serum in vitro, but numbers reaching the liver were not significantly different in both passively protected and control rats. Oncospheral reorganisation did not take place in passively protected rats. In control rats, only 20 per cent of oncospheres reaching the liver were able to undergo reorganisation. Developing larvae also appeared to be susceptible to host rejection mechanisms, especially between days 5 and 9 after infection.  相似文献

An investigation of the chemotherapeutic effectiveness of a new broad-spectrum anthelmintic, oxfendazole (OXF, methyl 5[6]-phenylsulfinyl-2-benzimidazolecarbamate), against Trichinella spiralis is reported. OXF was highly effective against adult T. spiralis in mice subjected to a 4-day course of treatment during the enterai phase of experimental trichinellosis. When treatment began 72 h after the mice were inoculated with parasites, the number of adult worms recovered from the host intestine was greatly reduced by twice-daily oral doses of OXF at 12.5 mg/kg body wt. The same dosage regimen (12.5 mg/kg bis in die) was also highly effective against the muscle-dwelling larvae of T. spiralis when given on 4 successive days during the invasive (days 14–17) and encystment phases (days 28–31) of infection.  相似文献

The heterotrich ciliate Mirofolliculina limnoriae occurs in an obligate association with wood-boring species of the isopod Limnoria. It attaches to the dorsal surface of the limnoriid pleotelson. Folliculinid-bearing populations of Limnoria tripunctata from the Ria Formosa, Portugal, and of Limnoria quadripunctata from Portsmouth, UK, were sampled. Up to four folliculinids were found on one individual of L. quadripunctata and up to twelve on L. tripunctata. There was no evidence that folliculinid load increased with host size. The nature of the epibiont/basibiont relationship was examined by observing animals with and without epibionts, kept individually with small sticks of wood in cell culture wells over periods of up to 15 days. Faecal pellet production and moulting were recorded. In both host species, feeding rate as measured by faecal pellet production was significantly lower in animals bearing folliculinids. Feeding rate also diminished with the onset of moulting, but returned to the original rate within a week after moulting. The estimated intermoult period for the population of L. tripunctata examined was about 25 days and for L. quadripunctata was about 32 days. However, the proportion of individuals moulting over the period of observation declined with increasing size in L. tripunctata, suggesting increasing length of intermoult period with age. As the host faces toward the blind end of its tunnel and ventilates the burrow with its pleopods, the folliculinids are well placed to intercept particulate food in the incoming water stream. There are no clear advantages for the host in the association, but the suppression of feeding and the potential of the folliculinids to hinder swimming during dispersal migrations indicate that the association should be viewed as ectoparasitism rather than mutualism or commensalism.  相似文献

We determined the physiological effects of joint and separate nucleopolyhedrovirus (NPV) infection and parasitism by the endoparasitoid Microplitis pallidipes Szepligeti on biochemical events in the noctuid Spodoptera exigua (Hübner). The results indicated that in parasitized larvae, compared to healthy larvae, total protein concentration in host hemolymph began to decline and total sugar concentration significantly increased by the first day, while lipid content in the host body significantly increased by the second day after parasitization. Meanwhile, in jointly infected and parasitized hosts, compared to parasitized larvae, total protein concentration was consistently higher, total sugar concentration was consistently lower, and lipid content became higher by the second day after treatment. In virus-infected larvae, compared to healthy larvae, total protein concentration sharply declined during the first two days but increased by the third, while total sugar concentration increased on the second and third days after virus infection but decreased at other observation times, and lipid content began to increase by the second day after virus infection. Finally, in larvae that were both parasitized and virus-infected, compared to just virus-infected larvae, total protein concentration increased during the first two days but decreased by the third, total sugar concentration increased only on the first and fourth days, and lipid content decreased significantly on the first day but began to increase by the second day after treatment. These findings led us to conclude that parasitization inhibited protein mobilization but stimulated sugar mobilization in host hemolymph, and promoted lipid mobilization in the host body, while Spodoptera exigua NPV infection stimulated protein mobilization induced by parasitization but inhibited sugar mobilization induced by parasitization.  相似文献

Isolates R1 and R2 of Puccinia recondita f. sp. tritici of low or high virulence on leaves of a series of cultivars of wheat behaved very similarly up to the formation of appressoria above stomata and sub-stomatal vesicles. Differences in growth of hyphae and in responses of host cells became clearly apparent only 24 h after infection; they then increased so that 10 days after inoculation high virulence isolates had produced freely sporulating sori (infection type 4) whereas low virulence isolates produced small colonies which did not erupt and sporulate and were associated with large numbers of dead host cells. The cultivar Timmo was selected for experiments on induced susceptibility and induced resistance. It was highly susceptible (infection type 4) to isolate Rl and to a mutant RlM, similar in pathogenicity on many cultivars but with uredospores different in colour from those of R1 and those of isolate R2 to which Timmo was highly resistant (infection type 0). This difference facilitated interpretation of results of experiments in which leaves were inoculated with more than one isolate. Inoculation with R1 (or with RUM which always behaved similarly) made leaves susceptible to R2 inoculated 4 days later. R2 sori were about 70 % the area of and about 70 % as frequent as Rl sori when inoculated on the same area on the same surface or on an area on the lower surface below a similar area on the upper surface inoculated with Rl. There also developed in the same area small, non-erumpent colonies associated with large numbers of dead host cells; these necrotic lesions were very similar to those produced in leaves inoculated only with R2. There were significantly fewer sori and more necrotic lesions when Rl was inoculated 1 or 2 days before R2. Totals of sori and necrotic lesions were about the same when the interval between inoculations was 1, 2 or 4 days; they were also similar with an interval of 3 days when numbers of sori were substantially less than after 4 days. Inoculation with Rl had some but much less effect in making tissues susceptible to R2 when again it was inoculated later but about 2 mm away. Although inoculation with R1 sufficiently in advance had the striking effect of making tissue almost as susceptible to normally low virulence R2 as it was to R1, colonies and sori of R1 which developed in association with those of R2 were substantially smaller than those which developed when R1 was inoculated alone. But there were similar differences in sizes of colonies and sori when R1 and RIM were inoculated together or separately. They probably depended on competition for nutrients. Possible mechanisms of induced susceptibility and induced resistance and implications for disease in the field are discussed briefly.  相似文献
