Persistence is a central issue in population ecology with important implications for population management. Most theoretical studies have focused on continually interacting populations, even though many systems are subject to ecological disturbances which confound analysis of persistence. In this paper, we use a combination of a simple parasite–hyperparasite model with disturbances and field data to investigate the factors contributing to the observed persistence of the parasite population. The field data are taken from a two-year experiment (including five growing seasons) investigating the use of the mycoparasite Sporidesmium sclerotivorum as a persistent biological control agent of Sclerotinia minor, an economically important fungal parasite of lettuce. We show that the standard assumption of homogeneous mixing fails to predict the observed persistence of the parasite population. We demonstrate that allowing for heterogeneous mixing prevents the fade-out predicted in the homogeneous mixing case. The implications of the results for broad classes of host–parasite systems are discussed.  相似文献

A key step in the analysis of microarray data is the selection of genes that are differentially expressed. Ideally, such experiments should be properly replicated in order to infer both technical and biological variability, and the data should be subjected to rigorous hypothesis tests to identify the differentially expressed genes. However, in microarray experiments involving the analysis of very large numbers of biological samples, replication is not always practical. Therefore, there is a need for a method to select differentially expressed genes in a rational way from insufficiently replicated data. In this paper, we describe a simple method that uses bootstrapping to generate an error model from a replicated pilot study that can be used to identify differentially expressed genes in subsequent large-scale studies on the same platform, but in which there may be no replicated arrays. The method builds a stratified error model that includes array-to-array variability, feature-to-feature variability and the dependence of error on signal intensity. We apply this model to the characterization of the host response in a model of bacterial infection of human intestinal epithelial cells. We demonstrate the effectiveness of error model based microarray experiments and propose this as a general strategy for a microarray-based screening of large collections of biological samples.  相似文献

We model the genealogies of coupled haploid host-virus populations. Hosts reproduce and replace other hosts as in the Moran model. The virus can be transmitted between individuals of the same and succeeding generations. The epidemic model allows a selective advantage for susceptible over infected hosts. The coupled host-virus ancestry of a sample of hosts is embedded in a branching and coalescing structure that we call the Ancestral Infection and Selection Graph, a direct analogue to the Ancestral Selection Graph of Krone and Neuhauser [1997. Theoret. Population Biol. 51, 210-237]. We prove this and discuss various special cases. We show that the inter-host viral genealogy is a scaled coalescent. Using simulations, we compare the viral genealogy under this model to earlier published models and investigate the estimatability of the selection and infectious contact rates. We use simulations to compare the persistence of the disease with the time to the ultimate ancestor.  相似文献

Oral administration of soluble protein Ags typically induces Ag-specific systemic nonresponsiveness. However, we have found that feeding a model food protein, OVA, to helminth-infected mice primes for a systemic OVA-specific Th2 response. In this report we show that, in addition to creating a Th2-priming cytokine environment, helminth infection up-regulates costimulatory molecule expression on mucosal, but not peripheral, APCs. To examine the consequences of mucosal infection for the T cell response to orally administered Ag, we adoptively transferred transgenic, OVA-specific, T cells into normal mice. We found that helminth infection enhances the expansion and survival of transgenic T cells induced by Ag feeding. Transfer of 5,6-carboxyfluorescein diacetate succinimidyl ester-labeled donor cells showed that T cell proliferation in response to Ag feeding takes place primarily in the mesenteric lymph nodes. Upon subsequent peripheral exposure to Ag in adjuvant, the proliferative capacity of the transferred transgenic T cells was reduced in noninfected mice that had been fed OVA. Helminth infection abrogated this reduction in proliferative capacity. Our data suggests that enteric infection can act as an adjuvant for the response to dietary Ags and has implications for allergic responses to food and the efficacy of oral vaccination.  相似文献

The trade‐off between within‐host infection rate and transmission to new hosts is predicted to constrain pathogen evolution, and to maintain polymorphism in pathogen populations. Pathogen life‐history stages and their correlations that underpin infection development may change under coinfection with other parasites as they compete for the same limited host resources. Cross‐kingdom interactions are common among pathogens in both natural and cultivated systems, yet their impacts on disease ecology and evolution are rarely studied. The host plant Plantago lanceolata is naturally infected by both Phomopsis subordinaria, a seed killing fungus, as well as Plantago lanceolata latent virus (PlLV) in the Åland Islands, SW Finland. We performed an inoculation assay to test whether coinfection with PlLV affects performance of two P. subordinaria strains, and the correlation between within‐host infection rate and transmission potential. The strains differed in the measured life‐history traits and their correlations. Moreover, we found that under virus coinfection, within‐host infection rate of P. subordinaria was smaller but transmission potential was higher compared to strains under single infection. The negative correlation between within‐host infection rate and transmission potential detected under single infection became positive under coinfection with PlLV. To understand whether within‐host and between‐host dynamics are correlated in wild populations, we surveyed 260 natural populations of P. lanceolata for P. subordinaria infection occurrence. When infections were found, we estimated between‐hosts dynamics by determining pathogen population size as the proportion of infected individuals, and within‐host dynamics by counting the proportion of infected flower stalks in 10 infected plants. In wild populations, the proportion of infected flower stalks was positively associated with pathogen population size. Jointly, our results suggest that the trade‐off between within‐host infection load and transmission may be strain specific, and that the pathogen life‐history that underpin epidemics may change depending on the diversity of infection, generating variation in disease dynamics.  相似文献

Abstract. 1. Hosts experiencing frequent variation in density are thought to benefit from allocating more resources to parasite defence when density is high (‘density‐dependent prophylaxis’). However, high density conditions can increase intra‐specific competition and induce physiological stress, hence increasing host susceptibility to infection (‘crowding‐stress hypothesis’). 2. We studied monarch butterflies (Danaus plexippus) and quantified the effects of larval rearing density on susceptibility to the protozoan parasite Ophryocystis elektroscirrha. Larvae were inoculated with parasite spores and reared at three density treatments: low, moderate, and high. We examined the effects of larval density on parasite loads, host survival, development rates, body size, and wing melanism. 3. Results showed an increase in infection probability with greater larval density. Monarchs in the moderate and high density treatments also suffered the greatest negative effects of parasite infection on body size, development rate, and adult longevity. 4. We observed greater body sizes and shorter development times for monarchs reared at moderate densities, and this was true for both unparasitised and parasite‐treated monarchs. We hypothesise that this effect could result from greater larval feeding rates at moderate densities, combined with greater physiological stress at the highest densities. 5. Although monarch larvae are assumed to occur at very low densities in the wild, an analysis of continent‐wide monarch larval abundance data showed that larval densities can reach high levels in year‐round resident populations and during the late phase of the breeding season. Treatment levels used in our experiment captured ecologically‐relevant variation in larval density observed in the wild.  相似文献

1. ‘Ecological fitting’ is the process whereby the suites of traits an organism carries from previous evolutionary relationships are used to enable colonisation of novel environments or resources. 2. The concept has much explanatory power in studies of novel host associations, particularly when data suggest a deviation from optimal foraging theory, but is often overlooked in studies of herbivore host selection behaviour in favour of evolutionary hypotheses. 3. In the present study, the concept was used to explain the unusual host selection behaviour of the New Zealand endemic oligophagous butterfly Lycaena salustius Fabricius, the larvae of which feed on endemic Polygonaceae species and the introduced and closely related Fagopyrum esculentum Moench. 4. In field cage oviposition choice assays involving only endemic plants, females preferred to oviposit on the rare Muehlenbeckia astonii Petrie. However, the novel host F. esculentum was overwhelmingly preferred in an additional greenhouse assay. In larval no‐choice performance assays, fitness indicators were variable for the novel host. 5. This imperfect relationship between oviposition preference and larval performance is discussed as a possible example of ecological fitting and highlights the potential use of the concept as an explanatory tool in novel host selection behaviour studies.  相似文献

ABSTRACT

Vector-transmitted diseases of plants have had devastating effects on agricultural production worldwide, resulting in drastic reductions in yield for crops such as cotton, soybean, tomato, and cassava. Plant-vector-virus models with continuous replanting are investigated in terms of the effects of selection of cuttings, roguing, and insecticide use on disease prevalence in plants. Previous models are extended to include two replanting strategies: frequencyreplanting and abundance-replanting. In frequency-replanting, replanting of infected cuttings depends on the selection frequency parameter ε, whereas in abundance-replanting, replanting depends on plant abundance via a selection rate parameter also denoted as ε. The two models are analysed and new thresholds for disease elimination are defined for each model. Parameter values for cassava, whiteflies, and African cassava mosaic virus serve as a case study. A numerical sensitivity analysis illustrates how the equilibrium densities of healthy and infected plants vary with parameter values. Optimal control theory is used to investigate the effects of roguing and insecticide use with a goal of maximizing the healthy plants that are harvested. Differences in the control strategies in the two models are seen for large values of ε. Also, the combined strategy of roguing and insecticide use performs better than a single control.  相似文献

In this paper, an epidemiological model with age of infection and disease relapse is investigated. The basic reproduction number for the model is identified, and it is shown to be a sharp threshold to completely determine the global dynamics of the model. By analysing the corresponding characteristic equations, the local stability of a disease-free steady state and an endemic steady state of the model is established. By means of suitable Lyapunov functionals and LaSalle's invariance principle, it is verified that if the basic reproduction number is less than unity, the disease-free steady state is globally asymptotically stable, and hence the disease dies out; if the basic reproduction number is greater than unity, the endemic steady state is globally asymptotically stable and the disease becomes endemic.  相似文献

Using a servosphere (locomotion compensator), locomotory behavior of Colorado potato beetle Leptinotarsa decemlineata Say (Coleoptera: Chrysomelidae) larvae was measured in detail in response to pulsed and non-pulsed odors of hostplant and conspecific pheromone. Second instars showed decreased straightness of movement, and all larvae showed decreased speed, in response to potato odor. Change in straightness by 2nd instars was also significantly affected by the interaction of pheromone and pulsing treatments. Fourth instars showed increased positive anemotaxis in response to the combined hostplant and pheromone odors. To our knowledge, this is the first demonstration of larval behavioral responses to adult pheromone in a holometabolous insect.  相似文献

The cytotoxic T lymphocyte (CTL) response to the infection of CD4+ T cells by human T cell leukemia virus type I (HTLV-I) has previously been modelled using standard response functions, with relatively simple dynamical outcomes. In this paper, we investigate the consequences of a more general CTL response and show that a sigmoidal response function gives rise to complex behaviours previously unobserved. Multiple equilibria are shown to exist and none of the equilibria is a global attractor during the chronic infection phase. Coexistence of local attractors with their own basin of attractions is the norm. In addition, both stable and unstable periodic oscillations can be created through Hopf bifurcations. We show that transient periodic oscillations occur when a saddle-type periodic solution exists. As a consequence, transient periodic oscillations can be robust and observable. Implications of our findings to the dynamics of CTL response to HTLV-I infections in vivo and pathogenesis of HAM/TSP are discussed.  相似文献

 A model for macroparasitic infection with variable aggregation is considered. The starting point is an immigration-and-death process for parasites within a host, as in [3]; it is assumed however that infections will normally occur with several larvae at the same time. Starting from here, a four-dimensional, where free-living larvae are explicitly considered, and a three-dimensional model are obtained with same methods used in [26]. The equilibria of these models are found, their stability is discussed, as well as some qualitative features. It has been found that the assumption of “clumped” infections may have dramatic effects on the aggregation exhibited by these models. Infections with several larvae at the same time also increases the stability of the endemic equilibria of these models, and makes the occurrence of subcritical bifurcations (and consequently multiple equilibria) slightly more likely. The results of the low-dimensional model have also been compared to numerical simulations of the infinite system that describes the immigration-and-death process. It appears that the results of the systems are, by and large, in close correspondence, except for a parameter region where the four-dimensional model exhibits unusual properties, such as the occurrence of multiple disease-free equilibria, that do not appear to be shared by the infinite system. Abstract.  1. Most empirical and theoretical studies treat egg load as a major determinant of insect oviposition behaviour, and predict a positive link between egg load and readiness to oviposit.
2. In the present study, the correlation between egg load and readiness to lay eggs on a low-ranked host was examined in wild-caught moths, Scotopteryx chenopodiata L. (Lepidoptera: Geometridae).
3. No overall effect of egg load on oviposition could be detected. However, there was a significant interaction between egg load and wing wear: in only one wing-wear class, out of four, high egg load tended to promote oviposition, while in the three other classes the effect was insignificant.
4. The results suggest that the effect of egg load on oviposition may depend on the age of the ovipositing insect.  相似文献

Microfilariae of Brugia malayi were obtained from the peritoneal cavities of infected gerbils and were then injected intravenously into mice. A sub-periodic, nocturnal microfilaraemia was produced. The level of microfilaraemia was proportional to the number of parasites injected, with approximately 1-3% of microfilariae being found in the peripheral circulation. The duration of microfilaraemia was proportional to the number of parasites injected; it subsided by 30 days after injection of 104 microfilariae but was still present at a low level 120 days after injection of 2 x 105 microfilariae. A transient splenomegaly developed after injection of microfilariae. Histopathological examination revealed large numbers of microfilariae free in the lumens of pulmonary small blood vessels and without any accompanying inflammatory reaction. Lesser numbers of microfilariae were seen in the cardiac blood and hepatic and renal blood vessels for the first few days after injection. There was cellular proliferation in the splenic white pulp and vascular congestion of the red pulp. Microfilariae labelled with 51Cr were injected intravenously; 57% of radioactivity was found in the lungs, 8.5% in the liver and 2.9% in the spleen. Mice developed immediate hypersensitivity reactions to B. malayi antigen by 4 weeks after injection, but Arthus and delayed hypersensitivity reactions were not seen at any time. when mice which had been injected 5 months previously were challenged with a 2nd injection of microfilariae, there was an accelerated clearance of parasites over 2 weeks and a marked peripheral blood eosinophilia developed. In contrast with natural infections, in which the continuous production of microfilariae complicates assessment, this model provides a system in which factors controlling the circulation of microfilariae in the bloodstream can be studied independently.  相似文献
