Genetic and environmental influences on the worker honey bee retinue response to queen mandibular gland pheromone (QMP) were investigated. Worker progeny were reared from queens originating from four sources: Australia, New Zealand, and two locations in British Columbia, Canada (Simon Fraser University and Vancouver Island). Progeny from New Zealand queens responded significantly higher (P < 0.05) than progeny from Australia in a QMP retinue bioassay. Retinue response was not related to queen production of pheromone or colony environment, and the strain-dependent differences in retinue bioassay responses were maintained over a wide range of dosages. Selected high- and low-responding colonies were bioassayed over the course of 1 year. High-responding colonies contacted QMP lures more frequently than low-responding colonies (P < 0.05) throughout the year except in late summer. We conclude that there is a strong genetic component to QMP response by worker honey bees, as well as a seasonal effect on response.  相似文献

Honey bee colonies consist of tens of thousands of workers and a single reproductive queen that produces a pheromone blend which maintains colony organization. Previous studies indicated that the insemination quantity and volume alter queen mandibular pheromone profiles. In our 11-month long field study we show that workers are more attracted to high-volume versus low-volume inseminated queens, however, there were no significant differences between treatments in the number of queen cells built by workers in preparation for supersedure. Workers exposed to low-volume inseminated queens initiated production of queen-like esters in their Dufour's glands, but there were no significant difference in the amount of methyl farnesoate and juvenile hormone in worker hemolymph. Lastly, queen overwintering survival was unexpectedly lower in high-volume inseminated queens. Our results suggest that the queen insemination volume could ultimately affect colony health and productivity.  相似文献

The mode of intranest transfer of the honey bee queen mandibular gland pheromone complex (QMP) was investigated in unpopulous and populous, slightly congested colonies, using synthetic QMP containing tritiated 9-keto-2(E)-decenoic acid, one of the QMP components. Radiolabel was rapidly transported from the center to the peripheral regions of the nest, and in a manner consistent with worker to worker transport. Population size and congestion had no effect on the relative rates of movement from the center to the periphery of the nest or on the mean amounts of radiolabel on individual bees. However, a significantly smaller proportion of the workers in the populous colonies received detectable amounts of radiolabel than in the uncongested colonies, and workers carrying especially large amounts of radiolabel were less numerous in the crowded colonies. It is suggested that, at the stage of colony development that the colonies were in, population size has more of an effect on intranest pheromone transmission than does crowding. Interference with pheromone transfer may occur only at higher levels of congestion than were created, and nearer to the reproductive phase of colony development. An alternative hypothesis is that colony crowding does not significantly affect QMP transport and that the onset of reproductive queen rearing may be associated more with changes in worker thresholds of response to QMP.  相似文献

Reproduction in species of eusocial insects is monopolized by one or a few individuals, while the remaining colony tasks are performed by the worker caste. This reproductive division of labor is exemplified by honey bees (Apis mellifera L.), in which a single, polyandrous queen is the sole colony member that lays fertilized eggs. Previous work has revealed that the developmental fate of honey bee queens is highly plastic, with queens raised from younger worker larvae exhibiting higher measures in several aspects of reproductive potential compared to queens raised from older worker larvae. Here, we investigated the effects of queen reproductive potential (“quality”) on the growth and winter survival of newly established honey bee colonies. We did so by comparing the growth of colonies headed by “high-quality” queens (i.e., those raised from young worker larvae, which are more queen-like morphologically) to those headed by “low-quality” queens (i.e., those raised from older worker larvae, which are more worker-like morphologically). We confirmed that queens reared from young worker larvae were significantly larger in size than queens reared from old worker larvae. We also found a significant positive effect of queen grafting age on a colony’s production of worker comb, drone comb, and stored food (honey and pollen), although we did not find a statistically significant difference in the production of worker and drone brood, worker population, and colony weight. Our results provide evidence that in honey bees, queen developmental plasticity influences several important measures of colony fitness. Thus, the present study supports the idea that a honey bee colony can be viewed (at least in part) as the expanded phenotype of its queen, and thus selection acting predominantly at the colony level can be congruent with that at the individual level.  相似文献

SYNTHETIC QUEEN MANDIBULAR GLAND PHEROMONE (QMP) WAS APPLIED TO HONEY BEE COLONIES TO TEST TWO HYPOTHESES: (i) QMP acts like a primer pheromone in the regulation of age-related division of labor, and (ii) this primer effect, if present, varies in three strains of workers that show genetically-based differences in their retinue attraction response to QMP (a pheromone releaser effect). Strains of workers that were high, or low in their response to QMP in a laboratory bioassay, as well as unselected 'wild-type' workers, were fostered in queenright colonies with or without supplemental QMP. Effects of QMP on foraging ontogeny and juvenile hormone III (JH) blood titers in worker honey bees were measured. Bees in QMP-supplemented colonies showed significant delays in foraging ontogeny, and foraging activity was reduced. They also had significantly lower JH titers, although the titer curves were somewhat atypical. There were no differences in foraging ontogeny or JH titers among the three strains. We conclude that (i) QMP can delay the ontogeny of foraging by some mechanism that suppresses JH production, (ii) this QMP primer response is independent of the retinue releaser response, and (iii) QMP can play an important role in regulating division of labour.  相似文献

Summary Queen attendance behavior of workers from selected honey bee colonies with high and low worker retinue response to synthetic queen mandibular gland pheromone (QMP) was investigated. Antennating, licking, grooming, and feeding of the queen by workers from high and low responding colonies were examined. High and low QMP responding workers did not attend the queen differently. However, workers originating from different colonies antennated and licked the queen more frequently than others, suggesting there may be a genetic basis for queen attendance behavior not necessarily associated with response to QMP. In the honey bee colony queen rearing is usually suppressed by releaser effects of the queen's pheromone. This is part of the dominance hierarchy maintaining the monogynous homeostasis. Under queenless conditions, the queen's control over the construction of emergency queen cells by the workers can be substitued by exposure to only one component of the mandibular pheromone secretion of a queen, the main compound (E)-9-oxo-2-decenoic acid. Many beekeepers feed their western honey bee (Apis mellifera) colonies artificial pollen substitutes to provide colonies with adequate nutrition during times of limited pollen quantity or quality. We provided caged worker bees commercially available pollen substitutes (AP23, MegaBee, UltraBee) and wildflower pollen in a choice-test to determine their relative attraction to/preference for the diets. We measured diet consumption by honey bees and observed honey bee behaviour to evaluate bee preferences for certain diets. The bees interacted with and consumed more wildflower pollen than they did any of the commercially available pollen substitutes. Our data suggest that bees have a strong preference for wildflower pollen over commercially available pollen substitutes.  相似文献

The roles of honey bee queen mandibular pheromone and colony congestion in the inhibition of swarming were investigated. Two colony siz.es were used: small, congested colonies and large, uncongested colonies. Both groups of colonies were treated with various dosages of the five-component, synthetic queen mandibular pheromone in the spring, and the extent and timing of swarming were followed. Most treatment groups received pheromone or a solvent blank (control) on a stationary slide; one group of the congested colonies received a pheromone treatment via an aerosol spray. The pheromone was not effective at delaying swarming in the congested colonies at any dosage applied on slides, but the aerosol spray-treated colonies swarmed significantly later in the season than the control colonies. The uncongested, pheromone-treated colonies exhibited a dose-dependent delay in swarming, with the highest dosage colonies swarming almost four weeks later than the control colonies. These results indicate an interaction between congestion and pheromone in the control of honey bee reproduction. While congestion may in itself be a factor stimulating swarming, these results are consistent with the interpretation that colony congestion reduces the transmission of queen pheromone within the nest, thereby removing the queen 's pheromone-based inhibition of queen rearing and subsequent swarming by workers.  相似文献

【目的】本实验旨在研究王台中残留啶虫脒对西方蜜蜂Apis mellifera蜂王培育质量的影响。【方法】通过融化蜂蜡并添加啶虫脒药液制作王台,使各王台中分别含4个不同剂量的啶虫脒(0, 10, 100和1 000 μg/kg蜂蜡)。同时,控制蜂王产卵6 h, 3 d后,将孵化为1日龄的幼虫分别移入各组王台中,并放入蜂群哺育。移虫后第3和6天分别统计各组王台中幼虫的接受率和封盖率,待蜂王出房时,计算其出房率,测定蜂王个体初生重、胸重和胸宽指标;采用实时荧光定量PCR(qPCR)技术测定蜂王卵巢中卵黄原蛋白基因(Vg)、储存蛋白基因(hex110和hex70b)的相对表达量。【结果】100 μg/kg蜂蜡和1 000μg/kg蜂蜡这两个啶虫脒剂量组西方蜜蜂蜂王的出房率都显著低于0 μg/kg蜂蜡和10 μg/kg蜂蜡剂量组,而0 μg/kg蜂蜡与10 μg/kg蜂蜡剂量组之间及100 μg/kg蜂蜡与1 000 μg/kg蜂蜡剂量组之间出房率 均差异不显著;这4个剂量组的王台幼虫接受率和封盖率以及蜂王的初生重、胸重和胸宽均无显著差异。qPCR结果显示,Vg基因的相对表达量随啶虫脒浓度的增加而下降,其中,1 000 μg/kg蜂蜡剂量组Vg基因的相对表达量显著低于10 μg/kg蜂蜡剂量组和0 μg/kg蜂蜡剂量组,其余各剂量组之间差异不显著;这4个剂量组之间hex110和hex70b基因的表达量差异不显著。【结论】西方蜜蜂王台中啶虫脒残留超过100 μg/kg蜂蜡剂量时,会对蜂王培育产生不利影响。  相似文献

Summary The number and frequencies of subfamilies in a honey bee colony were determined by DNA fingerprinting. Queen and brood samples were taken from three colonies with artificially inseminated queens and from one colony with a naturally mated queen. UsingHae III restriction enzyme and (GATA)4 oligonucleotide, the number of subfamilies in the colonies with artificially inseminated queens corresponded with the number of drones used for insemination. In the colony with the naturally mated queen, 12 subfamilies were found in a random sample of 104 workers. Considering that subfamily frequencies range from 1 to 26%, introcolonial worker relationship was estimated to be 0.328, corresponding to a genetical effective number of 6.4 matings.  相似文献

The vitellogenin egg yolk precursor protein represents a well-studied case of social pleiotropy in the model organism Apis mellifera. Vitellogenin is associated with fecundity in queens and plays a major role in controlling division of labour in workers, thereby affecting both individual and colony-level fitness. We studied the molecular evolution of vitellogenin and seven other genes sequenced in a large population panel of Apis mellifera and several closely related species to investigate the role of social pleiotropy on adaptive protein evolution. We found a significant excess of nonsynonymous fixed differences between A. mellifera, A. cerana and A. florea relative to synonymous sites indicating high rates of adaptive evolution at vitellogenin. Indeed, 88% of amino acid changes were fixed by selection in some portions of the gene. Further, vitellogenin exhibited hallmark signatures of selective sweeps in A. mellifera, including a significant skew in the allele frequency spectrum, extreme levels of genetic differentiation and linkage disequilibrium. Finally, replacement polymorphisms in vitellogenin were significantly enriched in parts of the protein involved in binding lipid, establishing a link between the gene's structure, function and effects on fitness. Our case study provides unequivocal evidence of historical and ongoing bouts of adaptive evolution acting on a key socially pleiotropic gene in the honey bee.  相似文献

Each mandibular gland of the queen bee, Apis mellifera L. (Hymenoptera: Apidae) consists of an axial cavity lined with a thin cuticular intima, secreted by a flat epithelium, and numerous glandular units (type 3), each unit with one duct cell and a large polyploid glandular cell. Mitochondria and endoplasmic reticulum are preponderant organelles. During the ontogeny of the queen bee, the ultrastructure of the glandular cells evolves as her level of pheromonal activity. Variations mainly concern the mitochondrial system. Hence, at the beginning of the imaginal life, the homogeneous population of small mitochondria increases. Towards the 3rd–5th day of the queen's imaginal life, the pheromonal activity increases and the mitochondrial differentiation results in the appearance of giant forms. During the highest activity phase (6 to 18-month-old queens), giant mitochondria, associated with endoplasmic reticulum, invade cytoplasmic areas. In queens aged from 18 to 24 months, the reduction of pheromonal and secretory activities is associated with the reduction of mitochondrial population as well as with the accumulation of lipid droplets and various lytic structures.  相似文献

Antennal movements of the honey bee can be conditioned operantly under laboratory conditions. Using this behavioural paradigm we have developed a preparation in which the activity of a single antennal muscle has been operantly conditioned. This muscle, the fast flagellum flexor muscle, is innervated by an identified motoneuron whose action potentials correlate 1:1 with the muscle potentials. The activity of the fast flagellum flexor muscle was recorded extracellularly from the scapus of the antenna. The animal was rewarded with a drop of sucrose solution whenever the muscle activity exceeded a defined reward threshold. The reward threshold was one standard deviation above the mean spontaneous frequency prior to conditioning. After ten conditioning trials, the frequency of the muscle potentials had increased significantly compared to the spontaneous frequency. The conditioned changes of frequency were observed for 30 min after conditioning. No significant changes of the frequency were found in the yoke control group. The firing pattern of the muscle potentials did not change significantly after conditioning or feeding. Fixing the antennal joints reduces or abolishes associative operant conditioning. The conditioned changes of the frequency of muscle potentials in the freely moving antenna are directly comparable to the behavioural changes during operant conditioning. Neuronal activity of the antennal lobes, mushroom bodies, and cervical connective in wild-type honey bees and snowlaranija mutants was recorded at different stages of the ontogeny (on the 1st, 3rd, 7th, and 25th days). The mutation snowlaranija affects the structural gene of tryptophane oxygenase, the first key exzyme in the kynurenine pathway of tryptophane metabolism, and leads to a deficit of kynurenines. Changes in neuronal activity in nutant bees were most pronounced in the cervical connective. A significant decrease in the pulse rate was revealed only in homozygous but not in heterozygous individuals. This finding is in accordance with previously reported inhibitory effect of the mutation at the behavioral level. Less pronounced effects were obtained when the neuronal activity was recorded in the antennal lobes or mushroom bodies. This may be related to a complex character of biochemical changes in different parts of mutants brain.  相似文献
