Benzene, toluene, ethylbenzene and xylene (BTEX) substrate interactions for a mesophilic (25°C) and thermophilic (50°C) toluene-acclimatized composted pine bark biofilter were investigated. Toluene, benzene, ethylbenzene, o-xylene, m-xylene and p-xylene removal efficiencies, both individually and in paired mixtures with toluene (1:1 ratio), were determined at a total loading rate of 18.1 g m–3 h–1 and retention time ranges of 0.5–3.0 min and 0.6–3.8 min for mesophilic and thermophilic biofilters, respectively. Overall, toluene degradation rates under mesophilic conditions were superior to degradation rates of individual BEX compounds. With the exception of p-xylene, higher removal efficiencies were achieved for individual BEX compounds compared to toluene under thermophilic conditions. Overall BEX compound degradation under mesophilic conditions was ranked as ethylbenzene >benzene >o-xylene >m-xylene >p-xylene. Under thermophilic conditions overall BEX compound degradation was ranked as benzene >o-xylene >ethylbenzene >m-xylene >p-xylene. With the exception of o-xylene, the presence of toluene in paired mixtures with BEX compounds resulted in enhanced removal efficiencies of BEX compounds, under both mesophilic and thermophilic conditions. A substrate interaction index was calculated to compare removal efficiencies at a retention time of 0.8 min (50 s). A reduction in toluene removal efficiencies (negative interaction) in the presence of individual BEX compounds was observed under mesophilic conditions, while enhanced toluene removal efficiency was achieved in the presence of other BEX compounds, with the exception of p-xylene under thermophilic conditions.  相似文献

A laboratory-scale biofilter unit packed with a mixture of compost, sugarcane bagasse, and granulated activated carbon (GAC) in the ratio of 55:30:15 by weight was used for a biofiltration study of air stream containing benzene, toluene, ethylbenzene, and o-xylene (BTEX). The effect of superficial velocity on mass transfer coefficient for the packing was studied by maintaining gas flow rates of 3, 4, 5, 6, and 8 L min?1 for inlet concentrations of 0.1, 0.4, and 0.8 g m?3 for each of benzene, toluene, ethylbenzene, and o-xylene. The maximum elimination capacity was found to be 20.92, 22.72, 20.73, and 18.94 g m?3 h?1 for BTEX, respectively, for stated flow rates. Removal efficiency of BTEX decreased from 99% to 71% for increasing inlet concentration from 0.1 to 0.8 g m?3. Gas film mass transfer coefficient predicted by modified Onda's equation was within ±10% of the experimental values.  相似文献

Thermophilic biodegradation of BTEX by two consortia of anaerobic bacteria   总被引:1,自引:0,他引:1  
Two thermophilic anaerobic bacterial consortia (ALK-1 and LLNL-1), capable of degrading the aromatic fuel hydrocarbons, benzene, toluene, ethylbenzene, and the xylenes (BTEX compounds), were developed at 60 °C from the produced water of ARCO'S Kuparuk oil field at Alaska and the subsurface water at the Lawrence Livermore National Laboratory gasoline-spill site, respectively. Both consortia were found to grow at 45–75 °C on BTEX compounds as their sole carbon and energy sources with 50 °C being the optimal temperature. With 3.5 mg total BTEX added to sealed 50-ml serum bottles, which contained 30 ml mineral salts medium and the consortium, benzene, toluene, ethylbenze, m-xylene, and an unresolved mixture of o- and p-xylenes were biodegraded by 22%, 38%, 42%, 40%, and 38%, respectively, by ALK-1 after 14 days of incubation at 50 °C. Somewhat lower, but significant, percentages of the BTEX compounds also were biodegraded at 60 °C and 70 °C. The extent of biodegradation of these BTEX compounds by LLNL-1 at each of these three temperatures was slightly less than that achieved by ALK-1. Use of [ring-14C]toluene in the BTEX mixture incubated at 50 °C verified that 41% and 31% of the biodegraded toluene was metabolized within 14 days to water-soluble products by ALK-1 and LLNL-1, respectively. A small fraction of it was mineralized to 14CO2. The use of [U-14C]benzene revealed that 2.6%–4.3% of the biodegraded benzene was metabolized at 50 °C to water-soluble products by the two consortia; however, no mineralization of the degraded [U-14C]benzene to 14CO2 was observed. The biodegradation of BTEX at all three temperatures by both consortia was tightly coupled to sulfate reduction as well as H2S generation. None was observed when sulfate was omitted from the serum bottles. This suggests that sulfate-reducing bacteria are most likely responsible for the observed thermophilic biodegradation of BTEX in both consortial cultures. BTEX catabolism interactions in a toluene-acclimatized biofilter   总被引:1,自引:0,他引:1  
BTEX substrate interactions for a toluene-acclimatized biofilter consortium were investigated. Benzene, ethylbenzene, o-xylene, m-xylene and p-xylene removal efficiencies were determined at a loading rate of 18.07 g m−3 h−1 and retention times of 0.5–3.0 min. This was also repeated for toluene in a 1:1 (m/m) ratio mixture (toluene: benzene, ethylbenzene, or xylene ) with each of the other compounds individually to obtain a final total loading of 18.07 g m−3 h−1. The results obtained were modelled using Michaelis–Menten kinetics and an explicit finite difference scheme to generate v max and K m parameters. The v max/K m ratio (a measure of the catalytic efficiency, or biodegradation capacity, of the reactor) was used to quantify substrate interactions occurring within the biofilter reactor without the need for free-cell suspended and monoculture experimentation. Toluene was found to enhance the catalytic efficiency of the reactor for p-xylene, while catabolism of all the other compounds was inhibited competitively by the presence of toluene. The toluene-acclimatized biofilter was also able to degrade all of the other BTEX compounds, even in the absence of toluene. The catalytic efficiency of the reactor for compounds other than toluene was in the order: ethylbenzene>benzene>o-xylene>m-xylene>p-xylene. The catalytic efficiency for toluene was reduced by the presence of all other tested BTEX compounds, with the greatest inhibitory effect being caused by the presence of benzene, while o-xylene and p-xylene caused the least inhibitory effect. This work illustrated that substrate interactions can be determined directly from biofilter reactor results without the need for free-cell and monoculture experimentation. The primary objective of this study was to evaluate the impact of substrate interactions on the biotransformation rates and mineralization potentials of gasoline monoaromatics and methyl tert-butyl ether (MTBE), compounds that commonly co-exist in groundwater contaminant plumes. A mixed culture was derived from gasoline-contaminated aquifer material using toluene as the enrichment substrate. Two pure cultures, Rhodococcus sp. RR1 and RR2, were isolated from the mixed culture. The three toluene-grown cultures were shown to biotransform all of the six BTEX compounds (benzene, toluene, ethylbenzene, o-xylene, m-xylene, and p-xylene), both individually and in mixtures, over a broad range of concentrations. The mixed culture was shown to degrade all of the BTEX compounds to 14CO2, while the two isolates mineralized BTE(m-/p-)X, but biotransformed o-xylene without production of carbon dioxide. Studies to evaluate substrate interactions caused by the concurrent presence of multiple BTEX compounds during their biodegradation revealed a number of patterns,including competitive inhibition and cometabolism. Ethylbenzene was shown to significantly inhibit BTX degradation in mixtures. MTBE was not biodegraded by any of the three toluene-grown cultures over a range of MTBE concentrations. Furthermore, the presence of MTBE at concentrations of 2 to 100?mg/L had no effect on BTEX biotransformation rates.  相似文献

High-temperature biotrickling filtration of hydrogen sulphide   总被引:1,自引:0,他引:1  
Biofiltration of malodorous reduced sulphur compounds such as hydrogen sulphide has been confined to emissions that are at temperatures below 40°C despite the fact that there are many industrial emissions (e.g. in the pulp and paper industry) at temperatures well above 40°C. This paper describes our study on the successful treatment of hydrogen sulphide gas at temperatures of 40, 50, 60 and 70°C using a microbial community obtained from a hot spring. Three biotrickling filter (BTF) systems were set up in parallel for a continuous run of 9 months to operate at three different temperatures, one of which was always at 40°C as a mesophilic control and the other two were for exploring high-temperature operation up to 70°C. The continuous experiment and a series of batch experiments in glass bottles (250 ml) showed that addition of glucose and monosodium glutamate enhanced thermophilic biofiltration of hydrogen sulphide gas and a removal rate of 40 g m−3 h−1 was achieved at 70°C. We suggest that the glucose is acting as a carbon source for the existing microbial community in the BTFs, whereas glutamate is acting as a compatible solute. The use of such organic compounds to enhance biodegradation of hydrogen sulphide, particularly at high temperatures, has not been demonstrated to our knowledge and, hence, has opened up a range of possibilities for applying biofiltration to hot gas effluent.  相似文献

Methanogenesis was studied using stirred, bench-top fermentors of 3-1 working volume fed on a semi-continuous basis with waste obtained from cattle fed a high grain, finishing diet. Digestion was carried out at 40 and 60°C. CH4 production was 11.8, 18.3, 61.9 and 84.5% higher in the thermophilic than the mesophilic digestor at the 3, 6, 9 and 12 g volatile solids (VS) l–1 reactor volume loading rates, respectively. When compared on an energetic basis CH4 production was 7.4, 18.3, 72.9 and 107.3 kJ day higher in the thermophilic than the mesophilic digestor. CH4 production decreased more rapidly with each increase in VS loading rate and decrease in retention time (RT) in the mesophilic than the thermophilic digestor. When expressed as l g–1 VS fed or as kJ kJ–1 fed, the amount of CH4 was 49% less at the highest compared to the lowest loading rate in the mesophilic digestor. In the thermophilic digestor the decrease was only 16%. Propionate accumulated in the mesophilic digestor at the two highest loading rates, reaching concentrations of about 50 mM, but were only about 13 mM in the thermophilic digestor. Isobutyrate, isovalerate plus 2-methylbutyrate, and valerate also accumulated at the higher loading rates.  相似文献

Benzene, toluene, ethylbenzene, and xylene are collectively known as BTEX which contributes to volatile environmental contaminants. This present study investigates the microbial degradation of BTEX in batch and continuous soil column experiments and its effects on soil matric potential. Batch degradation experiments were performed with different initial concentrations of BTEX using the BTEX tolerant culture isolated from petroleum-contaminated soil. In batch study, the degradation pattern for single substrate showed that xylene was degraded much faster than other compounds followed by ethylbenzene, toluene, and benzene with the highest μmax = 0.140 h?1 during initial substrate concentration of 100 mg L?1. Continuous degradation experiments were performed in a soil column with an inlet concentration of BTEX of about 2000 mg L?1 under unsaturated flow in anaerobic condition. BTEX degradation pattern was studied with time and the matric potential of the soil at different parts along the length of the column were determined at the end of the experiment. In continuous degradation study, BTEX compounds were degraded with different degradation pattern and an increase in soil matric potential was observed with an increase in depth from top to bottom in the column with applied suction head. It was found that column biodegradation contributed to 69.5% of BTEX reduction and the bacterial growth increased the soil matric potential of about 34% on an average along the column height. Therefore, this study proves that it is significant to consider soil matric potential in modeling fate and transport of BTEX in unsaturated soils.  相似文献

Mesophilic and thermophilic biotreatment of BTEX-polluted air in reactors   总被引:4,自引:0,他引:4  
This study compares the removal of a mixture of benzene, toluene, ethylbenzene, and all three xylene isomers (BTEX) in mesophilic and thermophilic (50 degrees C) bioreactors. In the mesophilic reactor fungi became dominant after long-term operation, while bacteria dominated in the thermophilic unit. Microbial acclimation was achieved by exposing the biofilters to initial BTEX loads of 2-15 g m(-3) h(-1), at an empty bed residence time of 96 s. After adaptation, the elimination capacities ranged from 3 to 188 g m(-3) h(-1), depending on the inlet load, for the mesophilic biofilter with removal efficiencies reaching 96%. On the other hand, in the thermophilic reactor the average removal efficiency was 83% with a maximum elimination capacity of 218 g m(-3) h(-1). There was a clear positive relationship between temperature gradients as well as CO(2) production and elimination capacities across the biofilters. The gas phase was sampled at different depths along the reactors observing that the percentage pollutant removal in each section was strongly dependant on the load applied. The fate of individual alkylbenzene compounds was checked, showing the unusually high biodegradation rate of benzene at high loads under thermophilic conditions (100%) compared to its very low removal in the mesophilic reactor at such load (<10%). Such difference was less pronounced for the other pollutants. After 210 days of operation, the dry biomass content for the mesophilic and thermophilic reactors were 0.300 and 0.114 g g(-1) (support), respectively, reaching higher removals under thermophilic conditions with a lower biomass accumulation, that is, lower pressure drop.  相似文献

The dynamics of mesophilic and thermophilic bacterial population of compost was studied. The bacteria population in the compost ranged from 109 to 105 CFU g?1 and was found to be maximum during mesophilic phase, and then decreased during the thermophilic, the cooling and maturation phases. Assessment of culturable bacteria by 16S rDNA revealed phylogenetic lineage of different polymorphic class bacilli, γ, β-proteobacteria and actinobacteria. Bacterial isolates produced extracellular enzymes: proteases, cellulase, xylanase, pectinase, tannase and amylase. Among them, mesophilic bacteria exhibited xylanolytic (81.25 %) and cellulolytic (63 %) activity. Thermophilic bacteria showed cellulolytic (75 %) and xylanolytic (66.6 %) activity, but a few isolates also produced tannase and pectinase. All bacterial isolates were observed to cause inhibition of three isolates of Bacillus pumilus and one isolate each of Staphylococcus sciuri and Kocuria sp. The physiological effect of compost on shoot length, leaf size and fruit maturation of tomato have been evaluated; the compost (75 g/pot) improved these parameters as compared to known compost (SOM). The efficacy of compost and SOM on photochemistry of tomato leaves was studied, based on imaging-PAM of the chlorophyll fluorescence parameters. Fv/Fm and electron transport rate (ETR) were increased significantly in compost (75 g) amended pot within 30 days of growth. Likewise, highest Y (II) of photosystem II (PS II) yield was found in compost (75 g) pot in 15 days. The findings of this study proved that the compost comprising of various bacteria involved in degradation of substrates was found to be beneficial for enhancement of tomato growth and development.  相似文献

We present a sensitive and quick way to determine benzene, toluene and dimethylbenzene (BTEX) in air, applying a cataluminescence (CTL) sensor based on a nano‐sized composite material, γ‐Al2O3/PtO2. The factors that affect the sensor's performance were studied, including the sensing material, temperature, rate of air carrier and wavelength. It was shown that when Pt accounted for 0.2% of the sensing material, the rate of the air carrier that carries target gas was 450 mL/min, the determination wavelength was 400 nm and temperature was 236°C, this sensor showed the best CTL intensity to BTEX. In addition, the CTL intensity had a high linear relation with the concentration of BTEX, with a linear range from 0.5 to 100 mL/m3, and a detection limit 0.22 mL/m3. This nano‐sized material had a quick response within 1.5 s, short recovery time within 1 min and a long lifetime, showing good potential for a variety of applications. Biodegradation of BTEX by a microbial consortium isolated from a closed municipal landfill was studied using respirometric techniques. The kinetics of biodegradation were estimated from experimental oxygen uptake data using a nonlinear parameter estimation technique. All of the six compounds were rapidly degraded by the microbial culture and no substrate inhibition was observed at the concentration levels examined (200 mg L−1 as COD). Microbial growth and contaminant degradation were adequately described by the Monod equation. Considerable differences were observed in the rates of BTEX biodegradation as seen from the estimates of the kinetic parameters. A three-fold variation was seen in the values of the maximum specific growth rate, μmax. The highest value of μmax was 0.389 h−1 for p-xylene while o-xylene was characterized by a μmax value of 0.14 h−1, the lowest observed in this study. The half saturation coefficient, K s, and the yield coefficient, Y, varied between 1.288–4.681 mg L−1 and 0.272–0.645 mg mg−1, respectively. Benzene and o-xylene exhibited higher resistance to biodegradation while toluene and p-xylene were rapidly degraded. Ethylbenzene and m-xylene were degraded at intermediate rates. In biodegradation experiments with a multiple substrate matrix, substrate depletion was slower than in single substrate experiments, suggesting an inhibitory nature of substrate interaction. In the current studies, we characterized the degradation of a hot mixture of benzene and toluene (BT) gases by a thermophilic biofilter using polyurethane as packing material and high-temperature compost as a microbial source. We also examined the effect of supplementing the biofilter with yeast extract (YE). We found that YE substantially enhanced microbial activity in the thermophilic biofilter. The degrading activity of the biofilter supplied with YE was stable during long-term operation (approximately 100 d) without accumulating excess biomass. The maximum elimination capacity (1,650 g x m(-3) h(-1)) in the biofilter supplemented with YE was 3.5 times higher than that in the biofilter without YE (470 g g x m(-3) h(-1)). At similar retention times, the capacity to eliminate BT for the YE-supplemented biofilter was higher than for previously reported mesophilic biofilters. Thus, thermophilic biofiltration can be used to degrade hydrophobic compounds such as a BT mixture. Finally, 16S rDNA polymerase chain reaction-DGGE (PCR-DGGE) fingerprinting revealed that the thermophilic bacteria in the biofilter included Rubrobacter sp. and Mycobacterium sp.  相似文献

Biofiltration is a technique which is frequently applied for the odour abatement of waste gases. This technique is based on the ability of microorganisms (generally bacteria, and to a small extent moulds and yeasts) to degrade several organic as well as inorganic compounds to mineral end-products, like water and carbon dioxide. In the case of biofiltration, microorganisms are attached to suited packing materials in the filter, which contain the inorganic nutrients necessary for microbial growth. In order to make biofiltration applicable on a larger scale in process industry, it is necessary to find microorganisms able to eliminate compounds which are strange to life, the so-called xenobiotics. At the Eindhoven University of Technology microorganisms were isolated for the elimination of a number of xenobiotics, e.g. aromatic compounds and chlorinated hydrocarbons.Both the microkinetics of the biodegradation in aqueous batch systems and the macrokinetics of biofilters were studied. Special attention was paid to the influence of the superficial gas velocity and the organic load on the filter bed's elimination capacity. The effect of temperature and oxygen on diazotrophic growth of the thermophilic cyanobacterium HTF (High Temperature Form) Chlorogloeopsis was investigated using cells grown in light-limited continuous culture at a dilution rate of 0.02 h-1. Diazotrophy was more sensitive to elevated temperatures than growth with combined nitrogen. The maximum temperature for growth of cultures gassed with CO2-enriched air was more than 55 °C but less than 60 °C with N2 as the sole nitrogen source, but between 60°C and 65°C when nitrate was present in the medium. The effect of temperature on nitrogenase activity, photosynthesis and respiration in the dark was determined using cells grown at 55°C. Maximal rates of all three processes were observed at 55°C and rates at 60°C during shortterm incubations were not less than 75% of the maximum. However, nitrogenase activity at 60°C was unstable and decayed at a rate of 2.2 h-1 under air and at 0.3 h-1 under argon. Photosynthesis and respiration were more stable at 60°C than anoxic nitrogen fixation. A biofiltration system inoculated with the mold Paecilomyces variotii CBS115145 showed a toluene elimination capacity (EC) of around 250 g/m3 of biofilter/h, which was higher than the values usually reported for bacteria. P. variotii assimilated m- and p-cresols but not the o isomer. Initial toluene hydroxylation occurred both on the methyl group and through the p-cresol pathway. These results were corroborated by detecting benzyl alcohol, benzaldehyde, and p-cresol as volatile intermediates. In liquid cultures with toluene as a substrate, the activity of toluene oxygenase (TO) was 5.6 nmol of O2/min/mg of biomass, and that of benzyl alcohol dehydrogenase was 16.2 nmol of NADH/min/mg of protein. Toluene biodegradation determined from the TO activity in the biofilter depended on the biomass distribution and the substrate concentration. The specific enzymatic activity decreased from 6.3 to 1.9 nmol of O2/min/mg of biomass along the reactor. Good agreement was found between the EC calculated from the TO activity and the EC measured on the biofilter. The results were confirmed by short-time biofiltration experiments. Average EC measured in different biofiltration experiments and EC calculated from the TO activity showed a linear relation, suggesting that in the biofilters, EC was limited by biological reaction. As the enzymatic activities of P. variotii were similar to those reported for bacteria, the high performance of the fungal biofilters can possibly be explained by the increased transfer of the hydrophobic compounds, including oxygen, from the gas phase to the mycelia, overcoming the transfer problems associated with the flat bacterial biofilms.  相似文献

A thermophilic, autotrophic methanogen (strain CB12, DSM 3664) was isolated from a mesophilic biogas digestor. This bacterium used H2-CO2 or formate as a substrate and grew as short rods, sometimes in pairs and in crooked filaments. Motility was not observed. Its optimum temperature (56°C) was lower than that of other thermophilic members of the genus Methanobacterium. The maximum observed specific growth rate was 0.564 h−1 (74-min doubling time).  相似文献

The biotreatment of complex mixtures of volatile organic compounds (VOCs) such as benzene, toluene, ethylbenzene, and xylene isomers (BTEX) has been investigated by many workers. However, the majority of the work has dealt with the treatment of aqueous or soil phase contamination. The biological treatment of gas and vapor phase sources of VOC wastes has recently received attention with increased usage of biofilters and bioscrubbers. Although these systems are relatively inexpensive, performance problems associated with biomass plugging, gas channeling, and support media acidification have limited their adoption. In this report we describe the development and evaluation of an alternative biotreatment system that allows rapid diffusion of both BTEX and oxygen through a silicone membrane to an active biofilm. The bioreactor system has a rapid liquid recycle, which facilitates nutrient medium mixing over the biofilm and allows for removal of sloughing cell mass. The system removed BTEX at rates up to 30 μg h−1 cm−2 of membrane area. BTEX removal efficiencies ranged from 75% to 99% depending on the BTEX concentration and vapor flowrate. Consequently, the system can be used for continuous removal and destruction of BTEX and other potential target VOCs in vapor phase streams. Interactions of toluene and p-xylene in air treatment biofilters packed with an inert filter media were studied. The effect of the inlet load of toluene, p-xylene and mixtures of both compounds on the biodegradation rate was analyzed in three lab-scale biofilters. A maximum elimination capacity (EC) of 26.5 and 40.3 g C m−3 h−1 for an inlet load (IL) of 65.6 and 57.8 g C m−3 h−1 was obtained for p-xylene and toluene biofilters, respectively. Inhibition of p-xylene biodegradation by the presence of toluene took place when the mixture was treated, whereas the presence of p-xylene had an enhancing effect on the toluene removal efficiency. Specific growth rates (μ) from 0.019 to 0.068 h−1 were calculated in the mixed biofilter, where the highest values were similar to mixtures with lower p-xylene levels (ILp-Xyl 8.84 ± 0.29 g C m−3 h−1). Two biofilters fed toluene-polluted air were inoculated with new fungal isolates of either Exophiala oligosperma or Paecilomyces variotii, while a third bioreactor was inoculated with a defined consortium composed of both fungi and a co-culture of a Pseudomonas strain and a Bacillus strain. Elimination capacities of 77 g m–3 h–1 and 55 g m–3 h–1 were reached in the fungal biofilters (with removal efficiencies exceeding 99%) in the case of, respectively, E. oligosperma and Paecilomyces variotii when feeding air with a relative humidity (RH) of 85%. The inoculated fungal strains remained the single dominant populations throughout the experiment. Conversely, in the biofilter inoculated with the bacterial–fungal consortium, the bacteria were gradually overgrown by the fungi, reaching a maximum elimination capacity around 77 g m–3 h–1. Determination of carbon dioxide concentrations both in batch assays and in biofiltration studies suggested the near complete mineralization of toluene. The non-linear toluene removal along the height of the biofilters resulted in local elimination capacities of up to 170 g m–3 h–1 and 94 g m–3 h–1 in the reactors inoculated, respectively, with E. oligosperma and P. variotii. Further studies with the most efficient strain, E. oligosperma, showed that the performance was highly dependent on the RH of the air and the pH of the nutrient solution. At a constant 85% RH, the maximum elimination capacity either dropped to 48.7 g m–3 h–1 or increased to 95.6 g m–3 h–1, respectively, when modifying the pH of the nutrient solution from 5.9 to either 4.5 or 7.5. The optimal conditions were 100% RH and pH 7.5, which allowed a maximum elimination capacity of 164.4 g m–3 h–1 under steady-state conditions, with near-complete toluene degradation.  相似文献
