We studied the effects of nutrient availability and protistan grazing on bacterial dynamics and community composition (BCC) in different parts of the canyon-shaped Rímov reservoir (Czech Republic). The effects of protistan grazing on BCC were examined using a size fractionation approach. Water from the dam area with only bacteria (<0.8 microm), bacteria and heterotrophic nanoflagellates (<5 microm), or whole water were incubated in situ inside dialysis bags. Top-down or predator manipulations (size fractionation) were also combined with bottom-up or resource manipulations, i.e., transplantation of samples to the middle and upper inflow parts of the reservoir with increased phosphorus availability. Significant genotypic shifts in BCC occurred with transplantation as indicated by denaturing gradient gel electrophoresis. Using different probes for fluorescence in situ hybridization, we found that 10 to 50% of total bacteria were members of the phylogenetically small cluster of beta-proteobacteria (targeted with the probe R-BT065). These rod-shaped cells of very uniform size were vulnerable to predation but very fast growing and responded markedly to the different experimental manipulations. In all the grazer-free treatments, the members of the R-BT065 cluster showed the highest net growth rates of all studied bacterial groups. Moreover, their relative abundance was highly correlated with bacterial bulk parameters and proportions of bacteria with high nucleic acid (HNA) content. In contrast, increasing protistan bacterivory yielded lower proportions of R-BT065-positive and HNA bacteria substituted by increasing proportions of the class Actinobacteria, which profited from the enhanced protistan bacterivory.  相似文献

Bacterivorous protists are known to induce changes in bacterial community composition (BCC). We hypothesized that changes in BCC could be related quantitatively to a measure of grazing: the ratio of bacterial mortality to growth rate. To test this hypothesis, we analyzed time-course changes in BCC, protistan grazing rate, and bacterial production from 3 in situ studies conducted in a freshwater reservoir and three laboratory studies. In the field experiments, samples were manipulated to yield different levels of protistan bacterivory and incubated in dialysis bags. Laboratory investigations were continuous cultivation studies in which different bacterivorous protists were added to bacterial communities. BCC was assessed using 4–6 different rRNA-targeted oligonucleotide probes for community analysis. Change in BCC (Δ BCC) was estimated as the sum of changes in the proportions of the two phylogenetic groups that showed the largest shifts. Analysis of a set of 22 estimates of shifts in the ratio of grazing to production rate over periods of 48–72 h and Δ BCC showed that Δ BCC was positively and tightly correlated (r 2 = 0.784) with shifts in the ratio of grazing mortality to cell production. While the nature of a shift in BCC is unpredictable, the magnitude of the change can be related to changes in the balance between bacterial production and protistan grazing. Changes in the pelagic community structure and activity along the longitudinal axis of the eutrophic Sau Reservoir (Catalonia, NE Spain) were studied between 1996 and 1999. Samples were taken from several transects from river to dam, measuring dissolved organic carbon (DOC), bacterial abundance and production, chlorophyll a concentration, heterotrophic nanoflagelate (HNF) and ciliate abundances and their grazing rates, and zooplankton density. The role of microbial and classical food chains (i.e., based directly on phytoplankon) were compared in the Sau Reservoir by analysing river-to-dam gradients in biomass and carbon and their temporal changes. The detritic metabolic pathway was more important near to the inflow, due to high allochthonous organic matter loads allowing the rapid development of the microbial food web. Protozoans (HNF and ciliates) consumed most of the bacterial production (i.e., >50%) in the reservoir. As opposed to the systems of lower trophic status ciliate carbon biomass and bacterivory contributions were larger than those of the HNF. We estimated species-specific ciliate growing rates on bacteria and distinguished several periods with high importance of distinct ciliate communities.  相似文献

Bacterioplankton from a meso-eutrophic dam reservoir was size fractionated to reduce (<0.8-microm treatment) or enhance (<5-microm treatment) protistan grazing and then incubated in situ for 96 h in dialysis bags. Time course samples were taken from the bags and the reservoir to estimate bacterial abundance, mean cell volume, production, protistan grazing, viral abundance, and frequency of visibly infected cells. Shifts in bacterial community composition (BCC) were examined by denaturing gradient gel electrophoresis (DGGE), cloning and sequencing of 16S rDNA genes from the different treatments, and fluorescence in situ hybridization (FISH) with previously employed and newly designed oligonucleotide probes. Changes in bacterioplankton characteristics were clearly linked to changes in mortality rates. In the reservoir, where bacterial production about equaled protist grazing and viral mortality, community characteristics were nearly invariant. In the "grazer-free" (0.8-microm-filtered) treatment, subject only to a relatively low mortality rate (approximately 17% day(-1)) from viral lysis, bacteria increased markedly in concentration. While the mean bacterial cell volume was invariant, DGGE indicated a shift in BCC and FISH revealed an increase in the proportion of one lineage within the beta proteobacteria. In the grazing-enhanced treatment (5-microm filtrate), grazing mortality was approximately 200% and viral lysis resulted in mortality of 30% of daily production. Cell concentrations declined, and grazing-resistant flocs and filaments eventually dominated the biomass, together accounting for >80% of the total bacteria by the end of the experiment. Once again, BCC changed strongly and a significant fraction of the large filaments was detected using a FISH probe targeted to members of the Flectobacillus lineage. Shifts of BCC were also reflected in DGGE patterns and in the increases in the relative importance of both beta proteobacteria and members of the Cytophaga-Flavobacterium cluster, which consistently formed different parts of the bacterial flocs. Phage abundance and infection of bacterioplankton were studied from March to November 2003 in the Sep Reservoir (Massif Central, France), together with temperature, chlorophyll, bacteria (abundance and production), and heterotrophic nanoflagellates (abundance and potential bacterivory). Virus abundance (VA) ranged from 0.6 to 13 × 1010 viruses l−1, exceeding bacterial abundance (BA) approximately sixfold on average. In terms of carbon, viruses corresponded to up to 25% of bacterial biomass. A multiple regression model indicated that BA was the best predictor for VA (R2 = 0.75). The frequency of infected bacteria (estimated from the percentage of visibly infected cells) varied from 1% to 32% and was best explained by a combination of temperature (R2 = 0.20) and bacterial production (R2 = 0.25). Viruses and flagellates contributed about equally to bacterial mortality. Both factors destroyed 55% of bacterial production, with a shift from phage bacteriolysis in early spring to protistan bacterivory in late summer. The vertical differences in most of the biological variables were not significant, contrasting with the seasonal differences (i.e., spring vs. summer-autumn). All biological variables under study were indeed significantly coupled to temperature. We regarded this to be the consequence of the enhanced discharge of the reservoir in 2003 (compared to previous years). This substantially weakened the stability and the thermal inertia of the water column, thereby establishing temperature as a stronger forcing factor in setting the conditions for optimal metabolic activity of microbial communities.  相似文献

The abundance and biomass of the main components of the microbial plankton food web (“microbial loop”)—heterotrophic bacteria, phototrophic picoplankton and nanoplankton, heterotrophic nanoflagellates, ciliates and viruses, production of phytoplankton and bacterioplankton, bacterivory of nanoflagellates, bacterial lysis by viruses, and the species composition of protists—have been determined in summer time in the Sheksna Reservoir (the Upper Volga basin). A total of 34 species of heterotrophic nanoflagellates from 15 taxa and 15 species of ciliates from 4 classes are identified. In different parts of the reservoir, the biomass of the microbial community varies from 26.2 to 64.3% (on average 45.5%) of the total plankton biomass. Heterotrophic bacteria are the main component of the microbial community, averaging 63.9% of the total microbial biomass. They are the second (after the phytoplankton) component of the plankton and contribute on average 28.6% to the plankton biomass. The high ratio of the production of heterotrophic bacteria to the production of phytoplankton indicates the important role of bacteria, which transfer carbon of allochthonous dissolved organic substances to a food web of the reservoir.  相似文献

A modified fluorescence in situ hybridization (FISH) method was used to analyze bacterial prey composition in protistan food vacuoles in both laboratory and natural populations. Under laboratory conditions, we exposed two bacterial strains (affiliated with beta- and gamma-Proteobacteria -- Aeromonas hydrophila and Pseudomonas fluorescens, respectively) to grazing by three protists: the flagellates Bodo saltans and Goniomonas sp., and the ciliate Cyclidium glaucoma. Both flagellate species preferably ingested A. hydrophila over P. fluorescens, while C. glaucoma showed no clear preferences. Differences were found in the digestion of bacterial prey with B. saltans digesting significantly faster P. fluorescens compared to two other protists. The field study was conducted in a reservoir as part of a larger experiment. We monitored changes in the bacterial prey composition available compared to the bacteria ingested in flagellate food vacuoles. Bacteria detected by probe HGC69a (Actinobacteria) and R-BT065 were negatively selected by flagellates. Bacteria detected by probe CF319a were initially positively selected but along with a temporal shift in bacterial cell size, this trend changed to negative selection during the experiment. Overall, our analysis of protistan food vacuole content indicated marked effects of flagellate prey selectivity on bacterioplankton community composition.  相似文献

Hypolimnetic oxygen content in lentic ecosystems has traditionally been modeled as a function of variables measured at the epilimnion, or that are supposed to drive epilimnetic processes, like total phosphorus load. However, in man-made reservoirs the river inflow can plunge into deep layers, directly linking the hypolimnion with the surrounding watershed. In these circumstances, organic matter carried by the river can influence the hypolimnetic oxygen content without important intervention of epilimnetic processes. Taking long-term data from two reservoirs in Spain, we applied an empirical regression approach to show that the dissolved organic matter carried by the river is the main driver shaping the hypolimnetic oxygen content. By contrast, typical variables commonly included in the modeling of the oxygen content in the hypolimnion (nutrient concentrations, chlorophyll a, and dissolved organic carbon measured in the water column) did not show any significant correlation. Interpretations from this regression approach were supported by a comparison between the monthly oxygen consumption in the hypolimnion and the monthly dissolved organic carbon load from the river inflow. We also revisited the prediction of the year-to-year variability of the Nürnberg’s anoxic factor in four reservoirs from Spain and the USA, explicitly including the allochthonous sources in the equations. These sources were significant predictors of the anoxic factor, especially in those systems subject to relatively high human impact. Thus, effects of allochthonous dissolved organic carbon should always be considered in empirical modeling and management of reservoir hypolimnetic processes related to oxygen content (for example, anoxia, nutrient internal loading, or phosphorus cycle resilience).  相似文献

Ecological quality assessment of non-natural water bodies is, in contrast to natural systems, less developed and requires determining biological indicators that reliably reflect environmental conditions and anthropogenic pressures. This study was motivated to propose fish indicators appropriate for assessment of reservoir ecosystems in central Europe. We analysed changes in water quality, total biomass and the taxonomic, trophic and size composition of fish communities along the longitudinal axes of four elongated, deep-valley reservoirs. Due to high nutrient inputs from their catchments, the reservoirs exhibited pronounced within-system gradients in primary productivity and water transparency. Although fish communities were similar among the reservoirs and dominated by few native species, the community structure and biomass systematically changed along the longitudinal axes of the reservoirs. The biomass and proportion of planktivores/benthivores in the fish community were highest at eutrophic sites near the river inflow and declined substantially towards deep, more oligotrophic sites close to the dam. The biomass and proportion of piscivores significantly increased downstream within the reservoirs alongside improving water quality. At species level, perch Perca fluviatilis and bream Abramis brama responded most sensitively, although in opposite directions, to the longitudinal environmental gradient. The major longitudinal changes in fish community characteristics were found to be consistent between pelagic and benthic habitats. The results of this study suggest that fish communities are appropriate indicators of eutrophication and can be used for ecological quality assessment of non-natural lentic water bodies, such as reservoirs. Moreover, our results underline the necessity to consider within-system gradients in water quality and the fish community when planning sampling programmes for deep-valley reservoirs.  相似文献

Seasonal and depth variations of the abundance, biomass, and bacterivory of protozoa (heterotrophic and mixotrophic flagellates and ciliates) were determined during thermal stratification in an oligomesotrophic lake (Lake Pavin, France). Maximal densities of heterotrophic flagellates (1.9 × 103 cells ml–1) and ciliates (6.1 cells ml–1) were found in the metalimnion. Pigmented flagellates dominated the flagellate biomass in the euphotic zone. Community composition of ciliated protists varied greatly with depth, and both the abundance and biomass of ciliates was dominated by oligotrichs. Heterotrophic flagellates dominated grazing, accounting for 84% of total protistan bacterivory. Maximal grazing impact of heterotrophic flagellates was 18.9 × 106 bacteria 1–1h–1. On average, 62% of nonpigmented flagellates were found to ingest particles. Ciliates and mixotrophic flagellates averaged 13% and 3% of protistan bacterivory, respectively. Attached protozoa (ciliates and flagellates) were found to colonize the diatom Asterionella formosa. Attached bacterivores had higher ingestion rates than free bacterivorous protozoa and may account for 66% of total protozoa bacterivory. This study was undertaken to determine the influence of a deep-storage reservoir and a downstream underground reservoir on the physicochemical limnology of a 98-km segment of a permanent river in central Texas. Seventeen parameters were studied. In general, impoundment in the deep-storage reservoir improved whereas inflow from the underground reservoir deteriorated water quality of the parent river. Overall combined effect of impoundment and inflow showed an improved water quality downstream from the study area from that found upstream of the study area.  相似文献

To determine longitudinal changes in phytoplankton composition and biomass in the Warnow River (Germany), single water parcels were followed during their downstream transport in August and October 1996 and April 1997. In summer, the phytoplankton assemblage was dominated by centric diatom and cyanobacteria species. Stephanodiscus hantzschii, Pseudanabaena limnetica, Planktothrix agardhii and Aulacoseira granulata var. angustissima were the most frequent species. In autumn, small centric diatoms dominated the whole river course. Irrespective of the season, in the fluvial lakes of the upper river, a substantial increase of phytoplankton biomass was observed. Shallow upstream river stretches were associated with large biomass losses. In the deep, slow flowing lower regions, total biomass remained constant. Longitudinal changes in biomass reflected downstream variations in flow velocity and river morphology. Cyanobacteria, cryptophytes and diatom species were subjected to large biomass losses along fast flowing, shallow river sections, whereas chlorophytes were favoured. Diatoms and cryptophytes benefited from low flow velocity and increased water depth in the downstream river. Changes in water depth and flow velocity have been found as key factors that cause the longitudinal differences in phytoplankton composition and biomass in small rivers.  相似文献

The Yolo Bypass, a large, managed floodplain that discharges to the headwaters of the San Francisco Estuary, was studied before, during, and after a single, month-long inundation by the Sacramento River in winter and spring 2000. The primary objective was to identify hydrologic conditions and other factors that enhance production of phytoplankton biomass in the floodplain waters. Recent reductions in phytoplankton have limited secondary production in the river and estuary, and increased phytoplankton biomass is a restoration objective for this system. Chlorophyll a was used as a measure of phytoplankton biomass in this study. Chlorophyll a concentrations were low (<4 μg l?1) during inundation by the river when flow through the floodplain was high, but concentrations rapidly increased as river inflow decreased and the floodplain drained. Therefore, hydrologic conditions in the weeks following inundation by river inflow appeared most important for producing phytoplankton biomass in the floodplain. Discharges from local streams were important sources of water to the floodplain before and after inundation by the river, and they supplied dissolved inorganic nutrients while chlorophyll a was increasing. Discharge from the floodplain was enriched in chlorophyll a relative to downstream locations in the river and estuary during the initial draining and later when local stream inflows produced brief discharge pulses. Based on the observation that phytoplankton biomass peaks during drainage events, we suggest that phytoplankton production in the floodplain and biomass transport to downstream locations would be higher in years with multiple inundation and draining sequences.  相似文献

A dam on a river course induces numerous changes in the aquatic environment both in the newly formed reservoir and in the river downstream. These changes modify the food resources available to fishes. As a consequence, fish communities undergo rapid transformations particularly in terms of trophic organization. Tucuruí Dam, closed on the Tocantins River, Brazil, in September 1984, formed a large reservoir of approximately 2200km2. Analyses of fish stomach contents were performed before and after the completion of the dam in the downstream section of the river as well as in the reservoir. Resource availability was seen through the relative contribution of food items in supporting the biomass. Main changes caused by the dam consisted of an increase in fishes as a food resource and of a parallel decrease of sediment both in the reservoir and in the downstream part of the river. In addition, in the downstream section, the relative contribution of plankton as a food resource diminished after dam closure. We identified 8 feeding regimes before dam closure. From them the trophic structure of fish communities were established and compared. Most of the community biomass was from specialist feeders. Contribution of piscivores increased after closure; planktivores became unimportant after closure downstream. Some species were shown to change their diet in the transformed environments either downstream or in the reservoir. However, these changes in individual species diet did not seem to play a major role in the transformation of trophic structure of the fish communities.  相似文献

We conducted a transplant experiment to elucidate the effects of different levels of grazing pressure, nutrient availability, especially phosphorus, and the impact of viruses on the changes in the structure of bacterioplankton assemblage in a meso-eutrophic reservoir. A sample taken from the nutrient-rich inflow part of the reservoir was size-fractionated and incubated in dialysis bags in both inflow and dam area. The structure of bacterial assemblage was examined by fluorescence in situ hybridization using oligonucleotide probes with different levels of specificity. In terms of the relative proportions of different bacterial groups, we found very few significant changes in the bacterioplankton composition after transplanting the treatments to the nutrient-poor dam area. However, we observed marked shifts in morphology and biomass towards the development of filaments, flocs and "vibrio-like" morphotypes of selected probe-defined groups of bacteria induced by increased grazing pressure. Despite the very high abundances of viruses in all the treatments, their effects on bacterioplankton were rather negligible.  相似文献

1. River food webs rely on two major food sources: autochthonous primary production within the river and allochthonous organic matter transferred to the river. We characterised the consumer communities and assessed the food sources of dominant consumers along a subtropical mountainous river (the Lanyang River of north‐eastern Taiwan) at the catchment scale from the headwater to the estuary using natural abundances of stable carbon and nitrogen isotopes. 2. The downstream transport of fine particulate organic matter (FPOM) was two orders of magnitude greater than that of coarse particulate organic matter (CPOM). Transport of both materials increased from the headwater and reached a maximum in the midstream reach. CPOM composition exhibited a gradual shift from leaves and branches in the headwater, an area characterised by high canopy cover, to algae in the midstream reaches and marsh plants in the downstream reaches. 3. Consumer communities can be classified into two regional categories: the upland category in the headwater and upstream and midstream reaches and the lowland category comprised of samples from the downstream reach and estuary. The upland category revealed a clear and gradual seasonal shift in community composition, but a seasonal shift was not apparent for the lowland category. Nutrient concentrations and water temperature were the main factors explaining longitudinal and seasonal variations. 4. The use of sources of organic matter by dominant consumers along the Lanyang River was primarily determined by their availability. Riparian C3 plants were the major food sources in the headwater, upstream reach and estuary, but the contribution of periphyton increased in the upper midstream reach where the river flows through an agricultural area. In the lower midstream and downstream reaches, the contribution of riparian C4 plants became dominant. 5. The trophic transfer of organic materials in the Lanyang River may be influenced by the fast current velocity and by sewage nutrient loading in the river, both of which have important implications for predicting how the functioning of subtropical river food webs will respond to human‐related changes in land use.  相似文献

Chronic bacterial lung infections in cystic fibrosis (CF) are the leading cause of morbidity and mortality. While a range of bacteria are known to be capable of establishing residence in the CF lung, only a small number have a clearly established link to deteriorating clinical status. The two bacteria with the clearest roles in CF lung disease are Pseudomonas aeruginosa and bacteria belonging to the Burkholderia cepacia complex (BCC). A number of common adaptations by P. aeruginosa strains to chronic lung infection in CF have been well described. Typically, initial isolates of P. aeruginosa are nonmucoid and display a range of putative virulence determinants. Upon establishment of chronic infection, subsequent isolates ultimately show a reduction in putative virulence determinants, including swimming motility, along with an acquisition of the mucoid phenotype and increased levels of antimicrobial resistance. Infections by BCC are marked by an unpredictable, but typically worse, clinical outcome. However, in contrast to P. aeruginosa infections in CF, studies describing adaptive changes in BCC bacterial phenotype during chronic lung infections are far more limited. To further enhance our understanding of chronic lung infections by BCC bacteria in CF, we assessed the swimming motility phenotype in 551 isolates of BCC bacteria from cystic fibrosis (CF) lung infections between 1981 and 2007. These data suggest that swimming motility is not typically lost by BCC during chronic infection, unlike as seen in P. aeruginosa infections. Furthermore, while we observed a statistically significant link between mucoidy and motility, we did not detect any link between motility phenotype and clinical outcome. These studies highlight the need for further work to understand the adaptive changes of BCC bacteria during chronic infection in the CF lung.  相似文献

Within an extensive programme on the Seine river, including the large reservoirs in the upstream basin, zooplankton populations were investigated at the reservoir-river interface to find out their fate when released into the river. Zooplankton structure, abundance and biomass were analysed in the river upstream and downstream from the reservoir, in the release canal and in the reservoir. Whereas the river zooplankton are dominated by rotifers, microcrustaceans represent a large part of the population in the reservoir, because of the longer residence time (6 months). We have shown that the reservoir is a source of zooplankton; we calculated a biomass export of 30-60 tons C year-1, which is, however, rapidly lost in the river. One of the main loss factors was found to be selective fish predation on larger zooplankton species developed in the reservoir.   相似文献

The assembly of bacterial communities in the rhizosphere is well-documented and plays a crucial role in supporting plant performance. However, we have limited knowledge of how plant rhizosphere determines the assembly of protistan predators and whether the potential associations between protistan predators and bacterial communities shift due to rhizosphere selection. To address this, we examined bacterial and protistan taxa from 443 agricultural soil samples including bulk and rhizosphere soils. Our results presented distinct patterns of bacteria and protistan predators in rhizosphere microbiome assembly. Community assembly of protistan predators was determined by a stochastic process in the rhizosphere and the diversity of protistan predators was reduced in the rhizosphere compared to bulk soils, these may be attributed to the indirect impacts from the altered bacterial communities that showed deterministic process assembly in the rhizosphere. Interestingly, we observed that the plant rhizosphere facilitates more close interrelationships between protistan predators and bacterial communities, which might promote a healthy rhizosphere microbial community for plant growth. Overall, our findings indicate that the potential predator–prey relationships within the microbiome, mediated by plant rhizosphere, might contribute to plant performance in agricultural ecosystems.  相似文献
