A new species, Hypocrea atroviridis, is described for the teleomorph of Trichoderma atroviride. Based on sequences of ITS-1, 5.8S, and ITS-2 regions of the rDNA complex and translation-elongation factor (EF-1α), T. atroviride and H. atroviridis form a well-supported clade within Trichoderma sect. Trichoderma. The conserved anamorphic phenotype of T. atroviride, observed for both conidial and ascospore derived cultures, was only found within that clade. In contrast, the teleomorph phenotype of H. atroviridis was morphologically indistinguishable from H. rufa, the teleomorph of T. viride. This Hypocrea phenotype may, therefore, be considered to be plesiomorphic within Trichoderma sect. Trichoderma, suggesting that genes controlling the expression of the teleomorph and anamorph evolve at different rates and that the genes controlling expression of the teleomorph are more conserved than are those controlling the expression of the anamorph.  相似文献

Three new species of Hypocrea/Trichoderma sect. Trichoderma (Hypocreaceae, Hypocreales, Ascomycota, Fungi) are described from recent collections in southern Europe and the Canary Islands. They have been characterized by morphological and molecular methods, including microscopic examination of the teleomorph in thin sections, the anamorph, growth rate experiments and phylogenetic analyses based on a part of the translation elongation factor 1-alpha encoding gene (tef1) containing the two last introns and a part of the rpb2 gene, encoding the second largest RNA polymerase subunit. Analyses involving tef1 did not unequivocally resolve the sister clade relationship of Hypocrea caerulescens relative to the Koningii and Viride clades, while analyses based on rpb2 clearly suggest a close relationship with the former, although the phenotype of H. caerulescens is similar to H. viridescens, particularly by its warted conidia and a coconut-like odor in CMD culture. Hypocrea hispanica and T. samuelsii however are clearly related to the Viride clade by both phylogenetic markers, despite their morphological similarity to H. koningii and its relatives. An apparently specific blue pigment is formed in CMD cultures by Hypocrea caerulescens but could not be obtained by extraction with organic solvents.  相似文献

The new species Hypocrea crystalligena (Hypocreales, Ascomycota, Fungi) is described as a holomorph and characterized based on an integrated phenotypic and phylogenetic approach, using teleomorph and anamorph morphologies, culture studies and analyses of phylogenetic markers including internal transcribed spacer 1 and 2 (ITS1 and 2), two last introns of the translation elongation factor 1-alpha encoding gene (tef1), and a portion of the rpb2 gene, encoding the second largest RNA polymerase subunit. Stromata of H. crystalligena show similarities with those of species from Trichoderma sect. Trichoderma but differ in several respects, including color, presence of white crystals on the surface and small ascospores. Colonies on CMD appear distinct, form colorless to white crystals on isolation, a yellowish to brown pigment and an anamorph with hyaline conidia exhibiting verticillium-like to gliocladium-like structural elements. ITS1 and 2 sequences exhibit all genus-specific features but also contain several unique hallmarks permitting development of a species-diagnostic barcode. Based on the analyses of partial rpb2 and tef1 sequences, H. crystalligena constitutes a separate evolutionary lineage with H. megalocitrina and H. psychrophila as its nearest neighbors. All these species form one phylogenetic clade with the H. pulvinata/H. citrina node.  相似文献

Five yeast strains belonging to the genus Cryptococcus Vuillemin were isolated from steppe plants and turf collected in the Prioksko-terrasny biosphere reserve (Moscow region, Russia). Sequence analyses of the D1/D2 domains of the 26S rDNA and of the internal transcribed spacer region revealed that these yeast strains and strain CBS 8016 have almost identical sequences and belong to the Holtermannia clade of the Tremellomycetidae (Basidiomycota, Hymenomycetes). A novel species named Cryptococcus festucosus (type strain VKM Y-2930) is proposed to accommodate these strains. Physiological characteristics and mycocin sensitivity patterns distinguishing Cryptococcus festucosus from the other species of this clade are presented.  相似文献

Trichoderma harzianum is a ubiquitous species in the environment and is effective in the biological control of plant-pathogenic fungi. T. harzianum has not been linked unequivocally to its sexual state nor has its phylogeny been studied in detail. It has been suggested that T. harzianum is a species complex based on the phenotypic and genotypic variability encountered. On the basis of morphological and cultural characters and DNA sequence data analysis of four genes (ITS rDNA, translation elongation factor 1-alpha, calmodulin, and alpha-actin), Hypocrea lixii was found to be the sexual state of T. harzianum. Both the asexual and sexual states of this species have wide geographic distributions. Phylogenetic analysis of four genes showed that T. harzianum/H. lixii is a cohesive group that is supported by bootstrap values higher than 95%. Principles of genealogical concordance indicated that T. harzianum/H. lixii is a complex of independent monophyletic lineages, but no diagnostic morphological distinctions were identified that justify formal taxonomic recognition for the different lineages.  相似文献

Trichoderma brevicompactum, a new species, was isolated from soil or tree bark in North, Central and South America, including the Caribbean Islands, and southwestern and southeastern Asia. Morphological and physiological characters, the internal transcribed spacer regions of the rDNA cluster (ITS1-5.8SrDNA-ITS2) and partial sequences of translation elongation factor 1-alpha (tef1) are described. Trichoderma brevicompactum is characterized by a pachybasium-type morphology, morphologically resembling other small-spored species referable to Trichoderma section Pachybasium but with essentially subglobose conidia. It is most closely related phylogenetically to Hypocrea lutea, from which it differs in morphological and physiological characters.  相似文献

A Gram-negative bacterium was previously isolated from an oil field in Shizuoka, Japan, and designated strain HD-1. Here we have performed detailed characterization of the strain, and have found that it represents a novel genus. The 16S rRNA sequence of strain HD-1 displayed highest similarity to various uncultured species (86.7-99.7%), along with 86.2-88.2% similarity to sequences from Azospirillum, Methylobacterium, Rhizobium, and Hyphomicrobium, all members of the alpha-Proteobacteria. Phylogenetic analysis revealed that HD-1 represented a deep-branched lineage among the alpha-Proteobacteria. DNA-DNA hybridization analysis with Azospirillum lipoferum and Hyphomicrobium vulgare revealed low levels of similarity among the strains. We further examined the biochemical properties of the strain under aerobic conditions. Among carbon sources, ethanol, n-propanol, n-butanol, and n-tetradecanol were the most preferred, while acetate, propionate, and pyruvate also supported high levels of growth. The strain could also grow on aromatic compounds such as toluene, benzene and phenol, and aliphatic hydrocarbons such as n-octane and n-tetradecane. In contrast, glycerol and various sugars, including glucose, fructose, maltose, and lactose, failed to support growth of HD-1. Under an anaerobic gas phase with butanol as the carbon source, little increase in cell weight was observed with the addition of several possible electron acceptors. As strain HD-1 represents a novel genus in the alpha-Proteobacteria, we designated the strain as Oleomonas sagaranensis gen. nov., sp. nov., strain HD-1.  相似文献

Seven strains of ballistoconidiogenous yeasts that contain xylose, form Q-10 ubiquinone, propagate by budding and don't produce stalk conidia were isolated from plant leaves collected in Thailand and were found to represent two new species. The taxonomic properties of the two species coincided with the genus Bullera so they are described as Bullera koratensis sp. nov. and Bullera lagerstroemiae sp. nov. In phylogenetic trees based on the nucleotide sequences of 18S ribosomal DNA and the D1/D2 domain of 26S rDNA, these two species are located in the Trichosporonales clade (Cryptococcus humicola-Trichosporon lineage).  相似文献

The genus Rhizomastix contains five species of amoeboflagellates with a single anterior flagellum, which live as intestinal symbionts of insects and amphibians. Though established in 1911, Rhizomastix has been neglected for decades and its phylogenetic position is uncertain. This paper describes the morphology of the first cultivated strain of Rhizomastix. The organism was isolated from an argentine horned frog and differs from the known Rhizomastix species by the presence of biflagellate cells. The isolate is described as Rhizomastix biflagellata sp. nov. A possible relationship of Rhizomastix to Archamoebae is discussed.  相似文献

From the shallow geothermally heated seafloor at the beach of Porto di Levante (Vulcano, Italy) 8 strains of long, tiny rods were isolated, which represent the first marine metal-mobilizing bacteria. Cells are Gram negative. They grow in a temperature range between 23 and 41°C with an optimum around 37°C at a salt concentration of up to 6.0% NaCl. The isolates are obligately chemolithotrophic, acidophilic aerobes which use sulfidic ores, elemental sulfur or ferrous iron as energy sources and procedure sulfuric acid. They show an upper pH-limit of growth at around 4.5. The G+C content of their DNA is around 64 mol%. Based on the results of the DNA-DNA hybridization they represent a new group within the genus Thiobacillus. Isolate LM3 is described as the type strain of the new species Thiobacillus prosperus.  相似文献

During a survey of yeasts associated with raw honey collected in Thailand, two strains of the Zygoascus clade were isolated from the Asian cavity-nesting honeybee Apis cerana and the stingless bee Homotrigona fimbriata. Phylogeny based on 26S rDNA D1/D2 sequences placed these yeasts as members of a clade including Candida bituminiphila, Candida patagonica and Candida polysorbophila. The strains of the two novel species, CBS 12271T and CBS 12270T, respectively, could be unquestionably distinguished from their relatives by rDNA sequences and other taxonomic characteristics. Therefore, the novel anamorphic species, Candida lundiana sp. nov. (type strain CBS 12271T = JCM 16823T) and Candida suthepensis sp. nov. (type strain CBS 12270T = JCM 16822T) are described.  相似文献

During an investigation of Hypocrea/Trichoderma species inhabiting mushroom bedlogs, we found five strains of an undescribed species from a culture collection. These were analyzed using a combined approach, including morphology of holomorph, cultural studies, and phylogenetic analyses of the rRNA gene cluster of the internal transcribed spacer region, translation elongation factor 1-α, and RNA polymerase subunit II gene sequences. Distinctive morphological characters include stromata with green ascospores produced on potato dextrose agar medium, and Gliocladium-like to irregularly Verticillium-like conidiophores. In phylogenetic analyses, this species belongs to the Semiorbis clade, but its morphological characteristics do not match the other members of this clade. Based on morphological observations and phylogenetic analyses, we describe this as a new species, Trichoderma mienum, representing its Hypocrea teleomorph and Trichoderma anamorph.  相似文献

A group of four motile facultative anaerobic marine isolates (Rd 8.15T [=CECT 7224T, =LMG 23850T], Rd 16.13, Rd 6.8 [=LMG 25696] and Rd2L5) were obtained from cultured clams (Ruditapes philippinarum and Venerupis pullastra) in Galicia, north-western Spain. They formed a tight phylogenetic group based on sequences of the 16S rRNA gene and the four housekeeping genes rpoA (encoding the α-chain of RNA polymerase), rpoD (encoding the sigma factor of RNA polymerase), recA (encoding RecA protein), and atpA (encoding the α-subunit of bacterial ATP synthase). The phylogenies based on these sequences indicated that the four isolates represented a novel species in the genus Vibrio, and more precisely in the Splendidus clade. DNA–DNA hybridizations with the type strains of species showing more than 98.6% 16S rRNA gene sequence similarity, revealed a DNA–DNA relatedness below 70%. The isolates could be differentiated from the phylogenetically related Vibrio species on the basis of several phenotypic features. In addition, strain Rd 8.15T showed potential pathogenic activity for adult clams in virulence assays. The name Vibrio celticus sp. nov. is proposed for this new taxon, with the type strain being Rd 8.15T (=CECT 7224T, =LMG 23850T).  相似文献

Large numbers of strains selectively isolated from soil, water and deteriorating vulcanised natural rubber pipe rings were provisionally assigned to the genus Nocardia. Twenty-eight representative isolates were found to have chemical and morphological properties typical of nocardiae. These organisms formed a monophyletic clade in the 16S rDNA tree together with Nocardia salmonicida. Three of the strains, isolates S1, W30 and R89, were distinguished from one another and from representatives of the validly described species of Nocardia using genotypic and phenotypic data. These organisms were considered to merit species status and were named Nocardia cummidelens sp. nov., Nocardia fluminea sp. nov. and Nocardia soli sp. nov. respectively. Additional comparative studies are needed to resolve the finer taxonomic relationships of the remaining isolates assigned to the Nocardia salmonicida clade and to further unravel the extent of nocardial diversity in artificial and natural ecosystems.  相似文献
