Oxamyl applied to field soil prevented Longidorus elongatus from acquiring and transmitting tomato black ring virus for at least 6 wk, although numbers of nematodes were not greatly decreased compared with the untreated control. Glasshouse and laboratory tests examined the effects of oxamyl on viruliferous L. elongatus and Xiphinema diversicaudatum. In these tests oxamyl (1.0 ppm) in the soil water largely prevented L. elongatus transmitting virus to bait plants over a period of 1 month. X. diversicaudatum was equally affected by smaller concentrations, 0.1 ppm being sufficient to inhibit virus transmission in one test. Inhibition of virus transmission was associated with a decrease in the number of root tip galls produced by nematode feeding, especially that of X. diversicaudatum. Few nematodes were seriously affected by oxamyl, except at the greatest concentration tested (100 ppm), when numbers of L. elongatus, but not of X. diversicaudatum, were decreased. In vitro-treated viruliferous nematodes protracted their stylets, but their subsequent ability to transmit virus was unimpaired. 相似文献

The geographical distribution of Xiphinema and Longidorus nematodes in the British Isles and Ireland 总被引:3,自引:0,他引:3
The distribution of Xiphinema and Longidorus species in the British Isles and Ireland were mapped by means of a computer programme from 5527 records. Longidorus caespiticola, L. elongatus, L. goodeyi and L. leptocephalus were found to be widespread among a wide range of vegetation types, particularly in grassland and arable crops; L. macrosoma and L. profundorum were recorded frequently in southern and central England, and L. attenuatus in East Anglia; L. vineacola was recorded once only in Eire. Paralongidorus maximus was recorded from two localities in eastern Scotland and one in southern England, on each occasion from intensively cultivated ground. Four species of Xiphinema were recorded but only X. diversicaudatum is widespread although apparently with a northerly limit in central Scotland; X. diversicaudatum was equally prevalent in arable crops, grassland and deciduous woodland; X. coxi and X. vuittenezi were recorded from a few scattered localities in England, and the few records for X. mediterraneum were all from Kent, south east England. All species were found in a wide range of soil types but mostly in light soils with a large sand fraction. Many of the species occurred together in mixed populations; L. caespiticola was most frequently associated with X. diversicaudatum and with L. leptocephalus and L. elongatus. X. diversicaudatum also was commonly found with L. goodeyi. Longidorus attenuatus produces galls at the tips of roots of field crops, including sugar beet, growing in alkaline, sandy soils in eastern England. L. elongatus produces similar, but often larger, galls on the roots of sugar beet and other crops in sandy soils in the W. Midlands and in Fen peats. Trichodorus spp. cause ‘stubby root’ of sugar beet and can feed on many field crops. Seven species of Trichodorus were found in sandy soils in eastern England. L. attenuatus, L. elongatus and Trichodorus spp. aggregate around roots and stunt sugar beet and other crop plants. L. attenuatus is commoner below plough depth than in the topsoil, whereas T. cylindricus, T. pachy-dermus and T. anemones are more abundant in the topsoil. The ability of 10 nematode species to transmit three strains of cherry leaf roll virus (CLRV) was tested by three methods: (1) virus-infected source plants and virus-free bait plants were grown concurrently in nematode-infested soil, (2) as for (1) but virus source plants were removed before bait plants were planted, and (3) nematodes were extracted from soil after access to virus source plants, and were added to pots containing bait plants. The occurrence of galls on roots showed that nematodes fed both on source and on bait plants in all experiments and, in some experiments, CLRV was detected by direct assays (slash tests) of Longidorus elongatus, L. leptocephalus and Paralongidorus maximus. Although the nematodes readily transmitted control viruses, for which they are known to be vectors, CLRV was detected by root assays in only a few bait plants exposed to L. elongatus, L. macrosoma, Xiphinema diversicaudatum or L. leptocephalus + X. vuittenezi in tests by method 1. The recovery of CLRV in these tests is interpreted as being due to contamination. These results add to the increasing circumstantial evidence against the involvement of nematodes in the transmission of CLRV. Other possible mechanisms of spread are discussed. 相似文献

The frequency with which the four virus-vector species of longidoroid nematodes occurring in Britain transmitted their associated plant viruses were compared in a series of experiments using a standard procedure. In these tests Xiphinema diversicaudatum proved an effective vector of British isolates of arabis mosaic virus and strawberry latent ringspot virus and Longidorus attenuatus of an isolate of tomato black ring virus from England. In comparison, isolates of raspberry ringspot virus and tomato blackring virus from Scotland and of raspberry ringspot virus from England were transmitted much less readily by their respective vectors, L. elongatus and L. macrosoma. These differences in ability to transmit virus were not related to differences in feeding access on the virus source- or bait-plants, in the extent to which virus was retained within the nematode feeding apparatus or in the frequency with which virus was recovered from Longidorus in concurrent slash tests. Three Scottish isolates of raspberry ringspot and tomato black ring viruses were transmitted equally infrequently by two populations of L. elongatus and the frequency with which virus was transmitted was not greatly increased when the species of source or bait plants was changed. 相似文献

Oogenesis and fertilization in longidorid nematodes has been examined for the first time at electron microscope level in Xiphinema diversicaudatum. Oogonia in the germinative zone of the ovary are irregularly shaped and lie adjacent to each other or separated by processes of the epithelial cells of the ovary. Developing oocytes pass in single file up to the growth zone and fibrogranular formation occurs around their nucleus. The perinuclear deposits remain until the oocyte is fully grown. Oocytes increase rapidly in volume because of the production of secretory granules. Three types of granules are recognizable. Type 1 granules are spherical, amorphous in structure and delimited by a lighter area, probably consisting of lipoprotein. Type 2 granules, electron lucent, arranged in groups, are lipid inclusions. Type 3 are dense spheres and may represent yolk bodies. The two last are then utilized by the developing embryo. Mature oocytes assume a smooth, cylindrical configuration as they traverse the oviduct. A cone of fertilization seems to be formed at the distal pole of the oocyte, where the sperm penetrates. The sperm totally penetrates the oocyte, through an invagination formed at the oocyte surface. The oocyte continues to undergo two unequal cytoplasmic divisions, resulting in the formation of a female pronucleus and two polar bodies. Long-term trends in populations of Xiphinema diversicaudatum were measured in two microplot experiments in which annual and perennial crops of seven plant families were grown in monoculture. Numbers of X. diversicaudatum increased under perennial ryegrass, raspberry and strawberry but decreased under hop, spring barley, potato, sugar beet, cabbage, winter wheat and winter beans; these effects probably reflected differences in the inherent host status of the crops. There were fewer X. diversicaudatum at 40–60 cm below annual crops than below perennial crops, and there were differences between perennial crops in nematode vertical distribution. Soil pore space did not limit nematode population density; total pore space was uniformly large under all crops and there was no detectable instability to water slaking. Under ryegrass a greater proportion of soil volume was occupied by pores too small to accommodate X. diversicaudatum, but since ryegrass supported the second largest nematode population, it seems unlikely that this was seriously limiting. 相似文献

In vitro feeding of Xiphinema brevicolle, X. index and Longidorus africanus on roots of host seedlings is described. Both Xiphinema spp. fed mainly along roots rather than at tips and up to several days at a single site. Feeding of L. africanus was confined to root tips and lasted up to 15 min. No visible short term reaction of roots parasitized by the Xiphinema spp. could be discerned, but both swelling and cessation of growth of root tips were observed within 20 hr after feeding by L. africanus. Long-term (12-month) symptoms on roots of several host plants caused by cultured populations of X. brevicolle, X. index, X. italiae, L. africanus and L. brevicaudatus are described. All the Xiphinema spp. caused a thinning and distinct darkening of root systems and, at some sites, a breakdown of the cortex. Both species of Longidorus caused stubby and swollen root tips. Radopholus similis and Meloidogyne spp. are the main nematode parasites of banana plants grown under plastic shelters in Morocco. A test was made in pots to evaluate the resistance of four genotypes of banana to these nematodes. Infection by Meloidogyne spp. brought about an increase in root weight in all banana plants tested because of gall formation. The inoculation of R. similis produced a reduction in length and diameter of the pseudo-trunk as well as in root and aerial mass in all genotypes. Pisang jari buaya showed the significantly lowest number of Meloidogyne nematodes per 10 g of roots, whereas for R. similis, the significantly smallest numbers were obtained in Pisang berlin and Pisang jari buaya. Therefore, Pisang jari buaya was the only banana genotype studied to show some degree of resistance to both nematodes. 相似文献

About half the nurseries and glasshouses in the Lea Valley of Hertfordshire were found to be infested with either or both Pratylenchus vulnus and Xiphinema diversicaudatum. The latter nematode probably occurred naturally in the soil on which the glasshouses were built but P. vulnus is thought to have been imported with rose rootstocks and is not known to occur outdoors in Britain. Both nematode species can cause decline of roses and even small numbers seem harmful. Numbers of P. vulnus can increase greatly, especially on rootstocks of Rosa canina, although numbers were often smaller when roses were severely damaged than on healthier crops. Numbers of X. diversicaudatum increased more slowly and R. canina was a better host for it than R. chinensis. X. diversicaudatum seemed to respond less quickly than P. vulnus to reduced host-plant vigour. The two rootstocks exhibited different host-status for the two species of nematode and cuttings of various rose cultivars showed different host-status to P. thornei, another species of lesion nematode which is not known to be pathogenic to roses. Two applications of dibromochloropropane liquid at the rate of 70 l/ha in a large volume of water maintained nematode densities at an acceptably low level, and growers who adopted this treatment as a supplement to pre-planting steam sterilisation and/or DD (dichloro-propane: dichloropropene) prolonged the productive life of their crops for several years. 相似文献

Enterococcus spp. are utilized worldwide as faecal indicator bacteria, but certain strains exhibit extended survival in environmental habitats and the factors influencing their persistence are poorly understood. We used flowing freshwater mesocosms to explore the effect of submerged aquatic vegetation (SAV) on the persistence of natural enterococci populations from a subtropical lake. The highest mean densities of culturable enterococci over 2 weeks occurred in SAV [8.6 × 102 colony‐forming units (cfu) per 100 g wet weight], followed by sediments (1.3 × 102 cfu per 100 g) and water (18 cfu per 100 ml). However, due to relative differences in the total mass of each substrate in the entire system (water > sediments > SAV), SAV‐associated enterococci represented only a minor proportion of the total population. Vegetated mesocosms harboured significantly higher mean cfu per mesocosm and cfu densities in sediments compared with their unvegetated counterparts, suggesting that SAV indirectly facilitates persistence in aquatic habitats. Populations were dominated (> 96%) by a single Enterococcus casseliflavus strain according to BOX‐PCR genotyping, which did not change over the 10‐month study and strongly suggests bacterial replication in the lake. The presence of such strains in the environment may represent highly competitive, naturalized and reproducing indicator bacteria populations that are not directly related to pollution events. 相似文献

An outbreak of strawberry latent ringspot virus (SLRV) in a plantation of Mailing Jewel raspberry coincided with the greatest abundance of the nematode vector, Xiphinema diversicaudatum. Arabis mosaic virus (AMV) was not detected in the crop but was, together with SLRV, in many weed species present. AMV was transmitted through the seed of Poa annua, Capsella bursa-pastoris and Senecio vulgaris and SLRV through the seed of Mentha arvensis. X. diversicaudatum were more numerous within the rows than between them and vertical sampling showed that most occurred between 4 and 12 in depth in both locations. Monthly sampling showed that egg laying occurred from April to July; populations increased to a peak in late autumn but declined during the winter, resulting in about a twofold annual increase in numbers. Females, males and juveniles transmitted AMV and SLRV to cucumber seedlings, and in the absence of plants the nematode retained AMV for 112 days and SLRV for 84 days. 相似文献

Short- and long-term persistence of pathogenic (i.e., tumor forming) agrobacteria in soil was investigated in six nursery plots with a history of high crown gall incidence. No pathogenic Agrobacterium strains were isolated in soil samples taken in fall and winter in any plots, but such strains were isolated from both bulk soils and weed rhizospheres (over 0.5 x 10(5) pathogenic CFU/g of bulk soil or rhizosphere) in three out of six plots in spring and summer. PCR amplifications of a vir sequence from DNA extracted from soil confirmed the presence of Ti plasmids in summer and their absence in fall and winter. The results indicate that strains that harbor a Ti plasmid had an unforeseen positive fitness versus Ti plasmid-free strains in soil and rhizosphere in spring and summer in spite of the apparent absence of tumor, and hence of opines. The gain of fitness occurred during a bloom of all cultivable agrobacteria observed only in conducive soils. An evolution of the pathogenic population was recorded during a 4-year period in one particularly conducive soil. In 1990, the pathogenic population in this soil consisted of only biovar 1 strains harboring both octopine- and nopaline-type Ti plasmids. In 1994, it consisted of only nopaline-type Ti plasmids equally distributed among biovar 1 and 2 strains. These results suggest that nopaline-type Ti plasmids conferred a better survival ability than octopine-type Ti plasmids to biovar 2 agrobacteria under the present field conditions. 相似文献

Superoxide dismutase (SoDase) polymorphism in reference populations of potato cyst nematodes (PCN) was studied by isoelectric focusing and compared with the banding pattern obtained from 26 English field populations of PCN. Qualitative and quantitative differences were found between the reference populations: a band detected only in the G. rostochiensis Ro4 and Ro5 reference populations was found in four field populations of varying pathotype composition and two field populations of G. rostochiensis Rol possessed extra bands that were not detected in the other field populations. Analysis of the band differences showed little correlation between pathotype and banding pattern in the field populations. 相似文献
