The adaptation to extreme concentrations of Ca2+ and its consequence on the properties of the 45Ca2+ transport were studied in submerged mycelia of Trichoderma viride. The adaptation to low [Ca2+]o did not cause changes in kinetic parameters of the 45Ca2+ influx but the adaptation to high [Ca2+]o increased the KM(Ca2+). The Vmax of the 45Ca2+ influx decreased with the age of (non-adapted) mycelia with concomitant decrease of the KM(Ca2+) these changes were prevented in mycelia adapted to high Ca2+. High [Ca2+]o decreased the stimulation by the uncoupler, 3, 3′, 4′, 5-tetrachloro salicylanilide (TCS) (30 μM), as compared to the control, whereas the Ca2+ chelator, EGTA, stimulated it. In the aged mycelia, the stimulation by TCS of the 45Ca2+ influx faded away, in parallel with the activity of the H+-ATPase. The 45Ca2+ efflux from mycelia was affected by TCS in a similar way as the 45Ca2+ influx. The results demonstrate the adaptive responses of transport processes participating in the mycelial Ca2+ homeostasis and ageing are in agreement with a notion that both Ca2+-influx and-efflux are coupled by the H+-homeostasis at the plasma membrane.  相似文献

Cyanobacteria are known to endure various stress conditions due to the inbuilt potential for oxidative stress alleviation owing to the presence of an array of antioxidants. The present study shows that Antarctic cyanobacterium Nostoc commune possesses two antioxidative enzymes viz., superoxide dismutase (SOD) and catalase that jointly cope with environmental stresses prevailing at its natural habitat. Native-PAGE analysis illustrates the presence of a single prominent isoform recognized as Fe-SOD and three distinct isoforms of catalase. The protein sequence of Fe-SOD in N. commune retrieved from NCBI protein sequence database was used for in silico analysis. 3D structure of N. commune was predicted by comparative modeling using MODELLER 9v11. Further, this model was validated for its quality by Ramachandran plot, ERRAT, Verify 3D and ProSA-web which revealed good structure quality of the model. Multiple sequence alignment showed high conservation in N and C-terminal domain regions along with all metal binding positions in Fe-SOD which were also found to be highly conserved in all 28 cyanobacterial species under study, including N. commune. In silico prediction of isoelectric point and molecular weight of Fe-SOD was found to be 5.48 and 22,342.98 Da respectively. The phylogenetic tree revealed that among 28 cyanobacterial species, Fe-SOD in N. commune was the closest evolutionary homolog of Fe-SOD in Nostoc punctiforme as evident by strong bootstrap value. Thus, N. commune may serve as a good biological model for studies related to survival of life under extreme conditions prevailing at the Antarctic region. Moreover cyanobacteria may be exploited for biochemical and biotechnological applications of enzymatic antioxidants.  相似文献

A slow in vivo uptake of cobalt from a growth medium resulted in an increase in density of mitochondria of Schizophyllum commune. Differential labeling of donor and resident mycelia, and subsequent analysis of resident mycelia surrounding donor implants, detected cobalt-dense mitochondria and demonstrated exchange of mitochondria after hyphal fusion. Transfer of mitochondria occurred in fully compatible, common-A, and common-AB matings, but was not detected in common-B matings of the tetrapolar Basidiomycete S. commune.  相似文献

In ecosystems, plant and bacterial volatile organic compounds (VOCs) are known to influence plant growth but less is known about the physiological effects of fungal VOCs. We have used Arabidopsis thaliana as a model to test the effects of VOCs from the soil fungus Trichoderma viride. Mature colonies of T. viride cultured on Petri plates were placed in a growth chamber in a shared atmosphere with A. thaliana without direct physical contact. Compared to controls, plants grown in the presence of T. viride volatiles were taller, bigger, flowered earlier, and had more lateral roots. They also had increased total biomass (45 %) and chlorophyll concentration (58 %). GC–MS analysis of T. viride VOCs revealed 51 compounds of which isobutyl alcohol, isopentyl alcohol, and 3-methylbutanal were most abundant. We conclude that VOCs emitted by T. viride have growth promoting effects on A. thaliana in the absence of direct physical contact.  相似文献

The gastropod mollusc, Oncomelania hupensis is a unique intermediate host for the human parasite Schistosoma japonicum. It is a primary factor for the epidemic of schistosomiasis and its distribution is consistent with the epidemic area of schistosomiasis. Here we report the functional properties of hemocyanin of O. hupensis (OhH), a copper-containing respiratory protein which was isolated from its hemolymph and purified by ammonium sulfate fractionation and ultracentrifugation. We identified the protein characters including UV absorption at 340 nm, copper content and quaternary structure. Furthermore, by induction of phenoloxidase and enzyme-linked immunosorbent assay we show that OhH exhibited o-diphenoloxidase activity after limited proteolysis, and shared carbohydrate epitopes with glycoconjugates of S. japonicum.  相似文献

A wild strain of Schizophyllum commune (MTCC 9670) isolated from Achanakmar-Amarkantak Biosphere Reserve of Central India was evaluated for the production of bioactive compounds. The chemical constituents of wild and in vitro grown cultures were compared. Under optimized conditions, different organic and aqueous extracts from mycelia and fruiting bodies were used to extract chemical components from the cultures grown in vitro. The gas chromatography combined wih mass spectrometry analysis of extracts identified two phenolic compounds, namely Phenyl benzoate (C13H10O2) and 4-(phenyl methoxy) phenol (C13H12O2) in the ethanolic extract of in vitro grown fruiting bodies and one antibacterial compound Pyrrolo (1, 2-a) piperazine-3, 6-dione (C7H10O2N2) in the methanolic extract of mycelia. High-performance liquid chromatography analysis revealed that the gallic acid and l-ascorbic acid were identifiable antioxidant components in the extracts possessing high free radical scavenging activity. The findings suggest that the wild strain of S. commune may serve as the source of novel bioactive compounds with effective antimicrobial and antioxidant activities.  相似文献

Vacuole-mediated proteolysis is important to sustained growth of filamentous wood-decaying fungi such as Schizophyllum commune. Demonstrating that specific proteases are vacuole associated has been difficult in these organisms due to the lack of specific markers for vacuolar compartments. We used 5-(and 6-)-carboxy-2′, 7′-dichlorofluorescein diacetate (carboxy-DCFDA) and a proprietary vacuolar membrane marker for yeast (MDY-64; Molecular Probes) for in situ fluorescent labeling of the vacuoles of S. commune mycelia grown on microscope slides. MDY-64 labels numerous small vesicles in S. commune mycelia in addition to larger vacuolar structures. In contrast, carboxy-DCFDA apparently is taken up by a subset of the MDY-64-labeled vesicles, accumulating primarily in larger vacuoles. Staining of mycelia with carboxy-DCFDA shows a transition from mostly cytoplasmic fluorescence in apical cells with little vacuolar fluorescence to nearly complete sequestration of the stain in vacuoles of older cells. In penultimate cells, both cytoplasm and vacuolar structures fluoresce. Vacuoles stained with carboxy-DCFDA typically were spherical and ranged in size from 0.4 μm to 3.2 μm in diameter with a mean of 1.8 um. Occasionally, in penultimate cells, tubular structures which stained with carboxy-DCFDA were found. ScPrB, a principal enzyme of nitrogen-limitation induced autolysis in S. commune, copurified in sucrose density gradients with carboxy-DCFDA and acid phosphatase, demonstrating its vacuolar localization. Optimization experiments with response surface statistical analysis were performed with Schizophyllum commune to obtain high β-glucosidase yields. The factors in the optimization experiment were the concentrations of cellulose, peptone, and KH2PO4. Their optimal values were 3.2, 3.0, and 0.2 g/100 ml, respectively. Enzyme assays revealed very high β-glucosidase (22.2 U/ml) and cellobiase (68.9 U/ml) yields. The avicelase yield was low as compared with that from Trichoderma reesei. Mixtures of S. commune and T. reesei culture filtrates caused faster and more extensive saccharification of Avicel than could be achieved by either filtrate alone. A β-glucosidase was isolated and purified from the optimized culture filtrate of S. commune. The electrophoretic mobility of the purified β-glucosidase indicated a molecular weight of 97,000. The amino acid composition was similar to that of β-glucosidase from T. reesei. The acidic (aspartate and glutamate) residues or their amides or both made up approximately 20% of the protein. The NH2-terminal amino acid of the enzyme was histidine.  相似文献

Extracts of the heads of the stingless bees, Trigona mexicana and T. pectoralis, contain mixtures of compounds that are identifiable by gas chromatography and mass spectrometry. These compounds form homologous series of aliphatic alcohols and ketones with an odd number of carbon atoms and functional groups at the 2-position. The alcohols and the ketones range from 7 to 17 carbon atoms. Benzaldehyde and a nitrogen containing compound are also present in the mixtures. The series of compounds from the two species are nearly identical qualitatively. They differ in the absence of 2-undecanol and 2-pentadecanol from the extracts of T. mexicana and T. pectoralis, respectively. The highest concentration of material is found in the 7-carbon fraction in T. mexicana and in the 13 to 15 carbon range in T. pectoralis. There is a major difference in the relative concentration of 2-heptanol and 2-heptanone in the two species with the concentration of the alcohol being one-fourth that of 2-heptanone in T. mexicana and ten times greater than the ketones in T. pectoralis.Both the alcohols and ketones are alarm pheromones. The alcohols are more active in inducing attack by the bees than are the ketones, but a mixture of the ketones and benzaldehyde was very active.  相似文献

A study of the temporal relationship between the appearance of phenoloxidase activity and the formation of fruiting bodies in Schizophyllum commune was undertaken. The results indicate that phenoloxidase activity increases in colonies subsequent to the formation of aggregate masses of hyphae, continues to increase until mature, sporulating fruiting structures are produced, and then declines significantly.  相似文献

A plate clearing assay was devised to screen for high-producing cellulase mutants of Trichoderma viride. The method employs (i) the use of either rose bengal or oxgall to limit colony size and (ii) Phosfon D (tributyl-2, 4-dichloroben-zylphosphonium chloride) to enhance cellulase detection, in combination with acid-swollen cellulose on agar plates. The method was used to isolate constitutive cellulase mutants of T. viride and should prove useful for isolating high-producing mutants from a range of organisms. This technique has been also used to determine the concentration at which glucose and glycerol inhibit cellulase synthesis by catabolite repression in the wild-type strains.  相似文献

Trichoderma (Ascomycetes, Hypocreales) strains that have warted conidia are traditionally identified as T. viride, the type species of Trichoderma. However, two morphologically distinct types of conidial warts (I and II) have been found. Because each type corresponds to a unique mitochondrial DNA pattern, it has been questioned whether T. viride comprises more than one species. Combined molecular data (sequences of the internal transcribed spacer 1 [ITS-1] and ITS-2 regions and of part of the 28S rRNA gene along with results of restriction fragment length polymorphism analysis of the endochitinase gene and PCR fingerprinting), morphology, physiology, and colony characteristics distinguish type I and type II as different species. Type I corresponds to “true” T. viride, the anamorph of Hypocrea rufa. Type II represents a new species, T. asperellum, which is, in terms of molecular characteristics, close to the neotype of T. hamatum.  相似文献

Eight saponins were isolated from alfalfa roots (Medicago sativa). The sensitivity of Trichoderma viride to the saponin varied with the individual saponin isolate. Seven isolates appeared to contain the aglycone, medicagenic acid, and while the other did not, it inhibited the growth of the fungus at higher concentrations than the other isolates. One pair and a triplet of saponins with divergent Rfs evoked near identical biological responses suggesting structural similarity toxic to T. viride.  相似文献

Transport of Ca2+ in membrane vesicles of the cyanobacterium Anabaena variabilis has been investigated. The light membranes previously shown to carry a Mg2+-dependent, Ca2+-stimulated ATPase (Lockau, W. and Pfeffer, S. (1982) Z. Naturforsch. 37C, 658–664) accumulate Ca2+ upon addition of ATP, whereas the (heavier) thylakoids do not. A stoichiometry of 0.3 Ca2+ taken up per ATP hydrolyzed has been determined from initial rates, which is considered to be an underestimation of the true stoichiometry of the pump. Calcium transport and Ca2+-stimulated ATPase activity are both sensitive to Na3VO4 (an inhibitor of ATPases forming a phosphorylated intermediate), show the same pH optimum and a comparable dependence on ATP concentration. Calcium transport is also supported by nucleoside triphosphates other than ATP, although at lower rates. Accumulation of calcium is abolished by an ionophore of divalent cations, ionophore A23187, but is resistant to ionophores of monovalent cations and to the inhibitor of F1-F0-type ATPases, N,N′-dicyclohexylcarbodiimide. It is concluded that the ATPase is a primary calcium pump.  相似文献

A total of ten flavonoids, all flavonols, were isolated from leaves of eleven species of Solanum sect. Solanum. Most species had relatively few compounds, primarily quercetin 3-O-glycosides. Little intraspecific variation in flavonoids was observed, except in S. americanum were it correlated with previously recognized races. Flavonoid data are of little help in determining ancestries of polyploid species, but do rule out S. sarrachoides as a progenitor of S. villosum and S. nigrum. Solanum sarrachoides is unique among species examined in having free flavonoid aglycones as well as extensive 3-O-methoxylation.  相似文献

This article presents a review of the enzyme mechanisms involved in degradation of cellulose by the white-rot fungus Sporotrichum poulverulentum. The hydrolytic enzymes involved include: (1) five endo-1,4-β-glucanases; (2) one exo-1,4-β-glucanase, and (3) one or several 1,4-β-glucosidases. A recently discovered oxidative enzyme of importance in in vitro cellulose degradation seems to be a cellobiose oxidase. An oxidoreductase, cellobiose:quinone oxidoreductase, is of importance both in cellulose and in lignin degradation. Regulatory mechanisms of the extracellular enzyme activities, such as monosugar levels causing catabolite repression of the endoglucanases, have also been investigated. The enzymes used by S. pulverulentum in cellulose hydrolysis are compared to those used by Trichoderma viride. Very similar types of enzymes are used in both cases. However, no oxidative enzyme has so far been found to be involved in extracellular cellulose degradation in the case of T. viride. Recommendations for further research are given.  相似文献

Vigorous vegetative growth of various Streptomyces species (S. auroefaciens, S. collinus and S. granaticolor) was achieved in a new semisynthetic liquid medium. Unlike the media commonly used for the cultivation of the submerged mycelia of different streptomycetes, this one does not contain insoluble material which enables direct and reliable measurement of net production of biomass. The medium was formulated to meet the nutritional requirements of all the three species. Is also supported production of antibiotic in each of the strains. A method for bulk preparation of Streptomyces aerial spores, involving cultivation on agar plates covered with cellophane, was developed. Advantage of this method lies in higher yields of spores, their higher purity and easier harvesting. The spores were activated by amild treatment with an Ultra-Turrax homogenizer resulting in the breakage of fibrous sheath, suspended in 20% glycerol, and stored at ?60°C. Thus, treated spores germinated synchronously even after several months of the storage. Hence, such spore material may be used for precise inoculation in a large series of experiments implying synchronous germination, and the inoculations can be carried out from the same batch over a long period.  相似文献
