目的:分离及鉴定来自于斑马鱼肠道中的酵母菌.方法:采用马铃薯葡萄糖培养基( PDA),从斑马鱼肠道中分离到1株酵母菌,编号为ZF -2,进行形态学观察、生理特征、PCR扩增26S rDNA D1/D2区以及基因测序分析,并构建系统发育进化树.结果:ZF -2分离株为粉红色菌落、菌体呈椭圆形,芽殖;可以同化利用葡萄糖、乳糖、蔗糖等糖类;PCR扩增获得26S rDNA D1/D2区序列长度为642bp( HQ323251),序列比对分析表明与斯鲁菲亚红酵母(Rhodotorula slooffiae)相似性在99%-100%之间,构建的系统发育进化树显示菌株ZF-2与R.slooffiae( EU583485)关系最近,且位于同一分支.结论:首次从斑马鱼肠道中分离到斯鲁菲亚红酵母.  相似文献

【背景】星云湖属云南高原湖泊,对其酵母菌的多样性研究和产类胡萝卜素菌株的筛选有助于开发利用星云湖酵母菌资源。【目的】研究云南星云湖酵母菌群落结构多样性,筛选和评价产类胡萝卜素活性菌株。【方法】采用膜过滤平置培养对星云湖水样进行酵母菌分离;应用26S rRNA D1/D2区域序列分析和形态特征及生理生化指标相结合,对分离获得的酵母菌进行鉴定;运用软件BIO-DAP分析酵母菌类群的多样性;采用酸热法提取类胡萝卜素,紫外分光光度计测定类胡萝卜素含量。【结果】从星云湖湖水中分离得到797株酵母菌,鉴定为18个属37个种。胶红酵母(Rhodotorula mucilaginosa)和Naganishia albida是优势种,分别占总菌株数的36.64%和28.86%;有28株酵母菌类胡萝卜素产量可达200μg/g细胞干重以上;有4株类胡萝卜素产量可达300μg/g细胞干重以上;粘红酵母(Rhodotorula glutinis) 2株、胶红酵母(Rhodotorula mucilaginosa)和Rhodosporidiobolus fluviale各1株。【结论】云南高原湖泊星云湖的酵母菌具有丰富的多样性,湖泊的营养化程度以及地理差异影响酵母菌群落的结构;红冬孢酵母属(Rhodosporidiobolus)和红酵母属(Rhodotorula)是星云湖类胡萝卜素产量可达300μg/g细胞干重以上的主要类群。有进一步开发研究的价值。  相似文献

【目的】对分离自云南抚仙湖湖水的379株酵母菌进行产类胡萝卜素的筛选,以期获得具有开发应用价值的产类胡萝卜素酵母菌。【方法】采用酸热法提取类胡萝卜素,紫外分光光度计测定类胡萝卜素含量,SPSS软件分析产类胡萝卜素酵母的分布特征。【结果】318株酵母菌(占供试菌株的83.91%)具有产类胡萝卜素的能力,大多数菌株类胡萝卜素产量在10-300μg/g之间,最高达590.83μg/g。产类胡萝卜素酵母集中分布于红冬孢酵母属(Rhodosporidium)和红酵母属(Rhodotorula);担子菌酵母产类胡萝卜素的能力高于子囊菌酵母;筛选到9株产类胡萝卜素活性较强的菌株:双倒卵形红冬孢酵母(Rhodosporidium diobovatum)3株、沼泽生红冬孢酵母(Rhodosporidium paludigenum)2株、粘红酵母(Rhodotorula glutinis)、禾本红酵母(Rhodotorula graminis)、瑞纳锁掷孢酵母(Sporidiobolus ruineniae)及Cystofilobasidium macerans各1株。【结论】高原湖泊抚仙湖生存着大量产类胡萝卜素的酵母菌,"红色酵母"(Red yeasts)具有较强的产类胡萝卜素的能力,红冬孢酵母属(Rhodosporidium)和红酵母属(Rhodotorula)是抚仙湖产类胡萝卜素酵母菌的主要类群。  相似文献

从苹果果实上分离获得的50余种酵母菌中筛选出了能够有效地抑制苹果青霉病(Peniclium expansum Link)的丝孢酵母(Trichosporon pullulans(Lindner.)Diddens and Lodder)。罗伦隐球酵母(Cryptococcus laurentii(Kuffer.)skin-ner)和粘红酵母(Rhodotorula glutinis(Fresen.)F.C.Harrison)。其中,抑病效果最好的T.pullulans是一种用于采后果实生物防治的新型拮抗菌,研究了这三种拮抗菌不同浓度处理和外加营养物质以及与钙配合使用对苹果青霉病的抑病效果。实验结果表明;酵母菌浓度越高,抑病作用越强;外源营养物质的加入削弱了酵母菌的拮抗效果;在C.laurentii的细胞悬浮液中加入0.18mol/L的CaCl2能显著提高其抑病能力。但增加CaCl2对T.pullulans和R.glutinis的抑病效果却没有明显作用。  相似文献

从八二酒曲及酿造崇明老白酒过程中分离纯化得到1株白色酵母菌和1株红色酵母菌,采用分子生物学方法进行鉴定,并对其酿造老白酒的特性进行了分析。结果显示,八二酒曲及崇明老白酒酿造过程中的优势酵母菌为酿酒酵母(Saccaromyces cerevisiae),从酿酒过程中分离的红色酵母菌为粘红酵母(Rhodotorula mucilaginosa)。采用粘红酵母和米根霉曲酿造的酒液的酒精度为11.9%vol,残余还原糖含量为11.2 g/100 m L,总酸含量为4.59 g/L,总酯含量为4.42 g/L。纯化的酿酒酵母和米根霉曲酿成的酒液口味醇和爽口,酒曲的纯化有助于开发出口感更爽口的老白酒。混合酵母和米根霉曲酿造的酒液呈典型的崇明老白酒风味,粘红酵母的参与对崇明老白酒口味风格的形成有一定的作用。  相似文献

通过Illumina MiSeq平台, 对石河子143团两个蟠桃园所采集的叶片样本附生酵母菌26S rDNA D1/D2区进行高通量测序, 并分析了其物种的组成、Alpha多样性以及菌种群落结构。结果显示,两个桃园中共检测到1470个OTU (Operational Taxonomic Units, 操作分类单元), 桃园1中含有个1217个OTU, 桃园2中含有609个OTU, 其中有356个为共有OTU。共鉴定出酵母菌2门22属, 属于担子菌门(Basidiomycota)、子囊菌门(Ascomycota), 两个桃园共有的酵母属有线黑粉酵母属(Filobasidium), 接合酵母属(Zygosaccharomyces), 红酵母属(Rhodotorula), 隐球酵母属(Cryptococcus), Papiliotrema, 毕赤酵母属(Pichia), 假丝酵母属(Candida), Kwoniella, Cystobasidium, 两个桃园优势属均为线黑粉酵母属。研究结果表明石河子地区蟠桃叶表面附生酵母菌资源丰富, 值得进一步开发与利用。  相似文献

为得到有效水解杜仲角质层的角质酶,通过生物酶法提取杜仲中的活性成分。从病变的杜仲叶中分离出一株能高产角质酶的菌种,鉴定分析证实该菌株是胶红酵母(Rhodotorula mucilaginosa),这是首次从植物病原真菌以外发现产角质酶的酵母菌,并对该菌株发酵产角质酶的条件进行优化。这一发现意义重大,因为酵母菌比植物病原菌安全许多,更利于大规模生产。  相似文献

[目的]研究云南5个地区(晋宁、祥云、程海、泸沽湖、洱海)的戟叶酸模(Rumex hastatus)花中的酵母菌和类酵母.[方法]采用涂布平板法对5个地区的戟叶酸模花中酵母菌和类酵母进行分离,通过26S rDNA Dl/D2区域序列分析并结合形态观察对分离获得的酵母菌和类酵母进行鉴定;采用胞外酶定性筛选培养基进行产酶筛选;用苏丹黑B染色法筛选产油脂菌株.[结果]从戟叶酸模花中分离得到82株酵母菌和99株类酵母;82株酵母菌鉴定为6个属16个种和1个潜在新种,99株类酵母鉴定为短梗霉属(Aureobasidium)的普鲁兰类酵母(A.pullulans)及3个变种;戟叶酸模花中的优势属是类酵母短梗霉属,其次为红酵母属(Rhodotorula)和隐球酵母属(Cryptococcus);筛选到134株具有产胞外酶活性和83株产油脂的酵母菌和类酵母.[结论]研究结果显示5个地区的戟叶酸模花中酵母菌和类酵母种类多样性较为丰富,并具有产淀粉酶、蛋白酶、纤维素酶、脂肪酶和油脂的特点,有潜在的应用前景.  相似文献

采用过滤涂皿的方法对云南程海湖冬季湖水样品中的酵母菌进行分离,通过26S r DNA D1/D2区域序列分析并结合形态观察和生理生化测试对分离获得的菌株进行鉴定,同时采用胞外酶定性筛选培养基进行产酶活性筛选,分析冬季程海湖酵母菌的多样性及胞外酶活性。结果从程海湖中分离获得171株酵母菌,鉴定为14个属22个种和2个潜在的新分类单元;优势属是红冬孢酵母属Rhodosporidium、红酵母属Rhodotorula和隐球酵母属Cryptococcus;优势种是红冬孢酵母Rhodosporidium kratochvilovae和斯鲁菲亚红酵母Rhodotorula slooffiae;湖北半部的样点CH2的多样性指数(H′=1.5945)和丰富度指数(R=2.7576)均最高,均匀度指数较高的是湖北部的样点CH1(J=0.8531);样点之间种群差异大,相似性低。大多数酵母菌株具有产1种以上的胞外酶。  相似文献

Three pink-colored yeast strains 3-1-3, 10-3-3 and 19-3-3 were isolated from xylem of surface-sterilized twigs of Pinus tabulaeformis collected from Dongling Mountain, Beijing, in different seasons. These strains were identified as Rhodotorula minuta (Saito) F.C. Harrison by conventional taxonomic characterization. However, molecular phylogenetic analysis based on internal transcribed spacer region (including 5.8S rDNA) and large-subunit rDNA D1/D2 domain sequences indicated that they represent a novel basidiomycetous yeast species, for which Rhodotorula pinicola is proposed (type strain: AS 2.2193(T)=CBS 9130(T)). The new species was most closely related to Rhodotorula laryngis Reiers?l in the R. minuta complex.  相似文献

The phenotypic and genetic heterogeneity of the basidiomycetous yeast species Rhodotorula glutinis was investigated in a group of 109 isolates. A polyphasic taxonomic approach was followed which included PCR fingerprinting, determination of sexual compatibility, 26S and ITS rDNA sequence analysis, DNA-DNA reassociation experiments and reassessment of micromorphological and physiological attributes. The relationships with species of the teleomorphic genus Rhodosporidium were studied and isolates previously identified as Rh. glutinis were found to belong to Rhodosporidium babjevae, Rhodosporidium diobovatum and Rhodosporidium sphaerocarpum. Other isolates included in the study were found to belong to Rh. glutinis var. dairenensis, which is elevated to the species level, or to undescribed species. The concept of Rh. glutinis sensu stricto is proposed due to the close phenetic and phylogenetic proximity detected for Rh. glutinis, Rhodotorula graminis and R. babjevae.  相似文献

Rhodotorula aurantiaca (Saito) Lodder is an anamorphic basidiomycetous yeast species that belongs to the so-called "Erythrobasidium lineage" of the Urediniomycetes, according to molecular phylogenetic studies based on nucleotide sequence analyses of different ribosomal DNA regions. In the most recent editions of the yeast taxonomy treatises the species Rhodotorula colostri (Castelli) Lodder and Rhodotorula crocea Shifrine & Phaff were listed as synonyms of R. aurantiaca. Taxonomic heterogeneity within R. aurantiaca was demonstrated in a study based on whole-cell protein profiles and is also hinted at by the observed differences in physiological and biochemical characteristics among the different strains under that species name. We determined partial nucleotide sequences of the 26S rRNA gene (D1/D2 domains) of strains maintained in the CBS culture collection under R. aurantiaca, including the type strains of its synonyms. The results showed that R. colostri and R. crocea are clearly distinct from R. aurantiaca and from any other currently recognised basidiomycetous yeast species. Furthermore, phylogenetic analysis of the sequence data placed the former two species in separate lineages of the Microbotryomycetidae: R. colostri in the "ruineniae clade" (Sporidiobolus lineage or Sporidiobolales) and R. crocea loosely linked to Rhodotorula javanica (Microbotryum lineage).  相似文献

通过聚丙烯酰胺凝胶电泳(PAGE)和超氧化物歧化酶(SOD)的鉴别性定位染色分析,表明红酵母Rhodotorula sp.仅存在线粒体中的Mn-SOD。在类胡萝卜素含量较高的时期(培养5d),红酵母SOD活力下降, 过氧化氢酶(CAT)活力升高,而此时它对外界O2-. 和H2O2的抗性均明显高于类胡萝卜素含量较低的时期(培养2d)。以上结果表明红酵母胞质中的抗氧化作用已主要由类胡萝卜素来承担,类胡萝卜素含量越高,红酵母抗氧化能力越强。由此,利用产生的O2-. 试剂TBO处理红酵母,可筛选出稳定高产的类胡萝卜素菌株,其中以培养2d的红酵母作为筛选对象更佳,所筛菌落类胡萝卜素含量可高达1740.9~2039.2g/g干重,比未处理的对照增加了28.2%~50.1%。  相似文献

采用白地霉2.498-47在以蛋白胨为氮源发酵时,只在开始pH3.2左右产生胞外蛋白,NH_4~+则无此限制,还有助于胞外多肽的产生。深红酵母2.280胞外蛋白的最大产率约在pH5.8;尿素作唯一氮源效果最好,与蛋白胨合并使用,可产胞外蛋白约800μg/ml。酵母膏对生长和产胞外蛋白很必要,不能用玉米浆和麦芽汁代替。用0.05N NaOH浸洗不同pH和氮源所培养的细胞都能浸出胞内蛋白,接近400μg/ml。发酵产生胞外蛋白自一日后开始,至二日接近最大量,以后逐渐减少。多肽产生起自第二日,五日未达到最大量。两株酵母菌都不表现蛋白酶活性,因此这个现象还不能解释。  相似文献

Carotenoid-producing yeast species such as Rhodotorula glutinis and Sporobolomyces roseus efficiently accumulated selenium from the growth medium. It was observed that incorporation of selenium into yeast cells during the growth inhibited production of beta-carotenoid and other carotenoid precursors (torularhodin and torulene). The yeasts with high content of the carotenoid pigments and selenium may be used for the preparation of a new type of antioxidant formula that could be directly applied for various human and animal diets. We have demonstrated that such a formula can only be produced by separate processes of the cultivation of red yeasts and a subsequent sorption of selenium into the cells.  相似文献

The Rhodotorula glutinis epoxide hydrolase, Eph1, was produced in the heterologous host Escherichia coli BL21(DE3) in order to develop a highly effective epoxide hydrolysis system. A 138-fold increase in Eph1 activity was found in cell extracts of the recombinant E. coli when compared to cell extracts of Rhodotorula glutinis, despite the formation of Eph1 inclusion bodies. Optimization of cultivation conditions and co-expression of molecular chaperones resulted in a further increase in activity and a reduction of the inclusion bodies formation, respectively. Compared to Rhodotorula glutinis cells and cell extracts, a total increase in Eph1 activity of over 200 times was found for both Escherichia coli cells and crude enzyme preparations of these cells. The improved conditions for recombinant Eph1 production were used to demonstrate the Eph1-catalysed kinetic resolution of a new Eph1 substrate, 1-oxaspiro[2.5]octane-2-carbonitrile.  相似文献

从黄海和渤海海水中分离到15株红酵母,初步定名为深红酵母(Rhodotorula rubraLodder)对这些菌株及另外8株红酵母属对照种的形态、生理生化性状以及全细胞长链脂肪酸的组成进行了测定,并用多元统计方法对菌株间的相似性进行了计算,对菌株分群。结果表明,15株酵母之间性状存在许多差异,这些差异不能完全被鉴定性状所反映,在多元分析中15株菌不能形成紧密的聚类群。研究结果对利用个别形态和生理生化性状确定的深红酵母种的范围提出了异议。  相似文献
