The binding of the stereoisomers of [{Ru(Me2bpy)2}2(μ-bpm)]4+, [{Ru(phen)2}2(μ-bpm)]4+ and [{Ru(Me2phen)2}2(μ-bpm)]4+ (Me2bpy = 4,4′-dimethyl-2,2′-bipyridine; bpm = 2,2′-bipyrimidine; phen = 1,10-phenanthroline; Me2phen = 4,7-dimethyl-1,10-phenanthroline) to a tridecanucleotide d(CCGAGAATTCCGG)2 which contains a single adenine bulge site, and four control dodecanucleotides, have been studied using a fluorescence intercalator displacement (FID) assay. The meso isomer of [{Ru(phen)2}2(μ-bpm)]4+ showed the strongest binding to the bulge-containing tridecanucleotide. In order to gain a greater understanding of the basis of the higher affinity exhibited by the meso isomer towards the bulge sequence, a 1H NMR study of the binding of the two enantiomers (ΔΔ and ΛΛ) of rac-[{Ru(phen)2}2(μ-bpm)]4+, and the, meso (ΔΛ) diastereoisomer, to the tridecanucleotide d(CCGAGAATTCCGG)2 was carried out. The NMR results suggest that the meso isomer binds selectively at the bulge site in the tridecanucleotide minor groove, but closer to the 3′-direction and with less structural perturbations of the groove than the ΔΔ and ΛΛ isomers. The results of this study confirm that dinuclear ruthenium complexes have excellent potential as DNA bulge probes, and meso-[{Ru(phen)2}2(μ-bpm)]4+ in particular has a high affinity (1 × 106 M−1) and selectivity for a single adenine bulge site.  相似文献

A potentially 18 donor macrocyclic ligand bearing four acetohydrazide pendant arms has been used to synthesize nine new dinuclear metal complexes with Co(II), Ni(II), Cu(II), Zn(II) and Cd(II). All compounds have been characterized by elemental analysis, MALDI-TOF-MS and FAB spectrometry, and by IR, UV-Vis and NMR spectroscopy. The system is a very good candidate for in situ recognition effects by MALDI-TOF-MS spectrometry and absorption spectroscopy. The presence of four acetohydrazide groups in L allows the synthesis of new three-dimensional self-assembling ditopic supramolecular compounds.  相似文献

The dinuclear complexes [Pd2(L)2(bipy)2] (1), [Pd2(L)2(phen)2] (2), [Pt2(L)2(bipy)2] (3) and [Pt2(L)2(phen)2] (4), where bipy = 2,2′-bipyridine, phen = 1,10-phenanthroline and L = 2,2′-azanediyldibenzoic dianion) dibridged by H2L ligands have been synthesized and characterized. The binding of the complexes with fish sperm DNA (FS-DNA) were investigated by fluorescence spectroscopy. The results indicate that the four complexes bound to DNA with different binding affinity, in the order complex 4 > complex 3 > complex 2 > complex 1, and the complex 3 binds to DNA in both coordination and intercalative mode. Gel electrophoresis assay demonstrates the ability of the complexes to cleave the pBR 322 plasmid DNA. The cytotoxic activity of the complexes was tested against four different cancer cell lines. The four complexes exhibited cytotoxic specificity and significant cancer cell inhibitory rate.  相似文献

The formation constants of equimolar and bis-chelate copper(II) and zinc(II) complexes with three aliphatic and four aromatic-substituted β-aminophosphonates have been determined in water solution by potentiometric studies. Spectroscopic parameters clearly indicate involvement of {NH3, PO3 2−} in both metal ions coordination. The comparison of the stability constants reveals slightly higher coordination power of the aliphatic-substituted β-aminophosphonic acids, which may be due to the higher basicity of their amino groups. All studied ligands are more effective in Cu2+ and Zn2+ coordination than phosphonic analogue of simple β-amino acid.  相似文献

A new series of dinuclear squarato-bridged nickel(II) and copper(II) complexes [Ni2(2,3,2-tet)21,3-C4O4)(H2O)2](ClO4)2 (1), [Ni2(aepn)21,3-C4O4)(H2O)2](ClO4)2 (2), [Cu2(pmedien)21,3-C4O4)(H2O)2](ClO4)2.4H2O (3) and [Cu2(DPA)21,2-C4O4)(H2O)2](ClO4)2 (4) where is the dianion of 3,4-dihydroxycyclobut-3-en-1,2-dione (squaric acid), 2,3,2-tet = 1,4,8,11-tetraazaundecane, aepn = N-(2-aminoethyl)-1,3-propanediamine, pmedien = N,N,N′,N″,N″-pentamethyldiethylenetriamine and DPA = di(2-pyridylmethyl)amine were synthesized and structurally characterized by X-ray crystallography. The spectral and structural characterization as well as the magnetic behaviour of these complexes is reported. In this series, structures consist of the groups as counter ions and the bridging the two M(II) centers in a μ-1,3- (1-3) and in a μ-1,2-bis(monodentate) (4) bonding fashions. The coordination geometry around the Ni(II) ions in 1 and 2 is six-coordinate with distorted octahedral environment achieved by N atoms of the amines and by one or two oxygen atoms from coordinated water molecules, respectively. In the Cu(II) complexes 3 and 4, a distorted square pyramidal geometry is achieved by the three N-atoms of the aepn or DPA and by an oxygen atom from a coordinated water molecule. The electronic spectra of the complexes in aqueous solutions are in complete agreement with the assigned X-ray geometry around the M(II) centers. The complexes show weak antiferromagnetic coupling with ∣J∣ = 1.8-4.2 cm−1 in the μ-1,3- bridged squarato compounds 1-3, and J = −16.1 cm−1 in the corresponding μ-1,2- bridged squarato complex 4. The magnetic properties are discussed in relation to the structural data.  相似文献

New copper(II) complexes [CuL2]2+ (L2=7,7,9-trimethyl-1,3,6,10,13-pentaazabicyclo[11,2,11.13]hexadec-9-ene) and [Cu2(L3)(H2O)2]4+ have been prepared by the reaction of [CuL1]2+ (L1=5,5,7-trimethyl-1,4,8,11,14-pentaazatetradce-7-ene) and formaldehyde. The mononuclear complex [CuL2]2+ has a square-planar coordination geometry with a 5-6-5-6 chelate ring sequence and is relatively stable even in low pH at room temperature. The dinuclear complex [Cu2(L3)(H2O)2]4+ consists of two unsaturated 15-membered pentaaza macrocyclic units (7,7,9-trimethyl-1,3,6,10,13-pentaazacyclopentadec-9-ene) that are linked together by a methylene group in a tilted face-to-face arrangement [Cu?Cu distance: 7.413(2) Å ]. Each macrocyclic unit of [Cu2(L3)(H2O)2]4+ contains one four-membered chelate ring and has a severely distorted octahedral coordination polyhedron. The dinuclear complex is quite stable in aqueous solutions containing an excess of formaldehyde or in dry acetonitrile but is decomposed to [CuL1]2+ and [CuL2]2+ in pure water.  相似文献

Two novel Cd(II) and Zn(II) complexes with the condensation product of 2-formylpyridine and selenosemicarbazide were synthesized. The structure of Cd(II) complex was determined by X-ray crystallography. The ligand is coordinated in a neutral form via pyridine and azomethine nitrogen atoms and the selenium donor. The cadmium ion completes its five-coordination by two chloride ligands, forming a square-pyramidal geometry. The structure of Zn(II) complex was established by analysis of spectroscopic data, which indicated coordination of the ligand as a bidentate via the selenium and the azomethine nitrogen atoms. The cytotoxic activity of the newly synthesized complexes, as well as if five structurally related complexes and the ligand evaluated against eight tumor cell lines. The new Cd(II) complex showed the highest activity similar to cisplatin with IC50 less than 10 μM for all cell lines. Cell cycle distribution and apoptosis study showed that Cd(II) complex and cisplatin might have some similarity in anticancer activity, which was not the case for cisplatin and other studied complexes. Effects of the complexes on matrix metalloproteinases (MMPs) MMP-9 and MMP-2 was also studied. Cd(II) and Zn(II) complexes and cisplatin increased MMP-2 activity in supernatants of tested cells, while Ni(II) complex with the same ligand decreased the activity, implying a possible activity in preventing tumor invasion and metastasis processes.  相似文献

Aminophosphonates were found to be effective metal ion chelators. Ethylenediamine (EN) itself binds strongly to a series of metal ions forming efficient five-membered chelate ring using two nitrogen donors. Combination of EN with phosphonate function results in the family of low molecular chelating agents. Introduction of a pyridine moiety into EN-phosphonate structure results in the very effective ligands involving a four-nitrogen donor set to coordinate the metal ion. In this work, the potentiometric and spectroscopic data for two analogues 2,2′-(ethylenedi-imino)bis(3-pyridylphosphonic-acid) - L2 and 2,2′-(ethylenedi-imino)bis(2-pyridyl-phosphonic-acid) - L3, comprising EN, two phosphonates and two pyridines with Cu(II), Ni(II) and Zn(II) ions are presented showing very high efficacy of one of the ligands studied.  相似文献

Pyrazole-3,5-dicarboxylate-bridged dinuclear ruthenium(II) and osmium(II) complexes of 2,2-bipyridine of composition [(bpy)2Ru(pzdc)Ru(bpy)2](ClO4) · H2O (1) and [(bpy)2Os(pzdc)Os(bpy)2](ClO4) · H2O (2) have been obtained in high yield and have been separated to their homochiral (ΛΛ/ΔΔ) rac (1a, 2a) and heterochiral (ΛΔ/ΔΛ) meso (1b, 2b) diastereoisomers. The distinctive structural features of these diastereoisomers have been characterized by 1-D and 2-D 1H NMR spectroscopy. The X-ray crystal structure of rac-[(bpy)2Os(pzdc)Os(bpy)2](ClO4) · H2O (2a) has been determined. The electrochemical and electronic spectral studies have established that there remain difference in properties and hence difference in intermetallic communication between the diastereoisomeric forms in each case.  相似文献

In this study, the effects of four novel mononuclear palladium(II) and platinum(II) complexes on the activity of human serum paraoxanase1 were examined. First, four novel mononuclear palladium(II) and platinum(II) complexes were synthesized with a nitrogen donor ligand 1‐(2‐aminoethyl)pyrrolidine and nonsteroidal anti‐inflammatory drugs diclofenac, mefenamic acid. These complexes were characterized by spectroscopic, thermal, and elemental analyses. The crystal structures of complex [Pd(2‐amepyr)2](dicl)2 1 and [Pd(2‐amepyr)2](mef)2 3 were determined by X‐ray crystallography. Then, paraoxonase1 enzyme was purified from human serum. The effects of these complexes on enzyme were evaluated in vitro. The complexes consist of the cationic unit and the counterions. The diclofenac and mefenamic acid acted as a counterion in the complexes. It was observed that all the complexes were stable up to high temperatures. These complexes, even at low doses, inhibited the activity of the enzyme with different inhibition mechanisms.  相似文献

Herein, we report the syntheses, spectral and structural characterization, and magnetic behavior of four new dinuclear terephthalato-bridged copper(II) complexes with formulae [Cu2(trpn)2(μ-tp)](ClO4)2 · 2H2O (1), [Cu2(aepn)2(μ-tp)(ClO4)2] (2), [Cu2(Medpt)2(μ-tp)(H2O)2](ClO4)2 (3) and [Cu2(Et2dien)2(μ-tp)(H2O)](ClO4)2 (4) where tp = terephthalate dianion, trpn = tris(3-aminopropyl)-amin, aepn = N-(2-aminoethyl)-1,3-propanediamine, Medpt = 3,3′-diamino-N-methyldipropylmine and Et2dien = N,N-diethyldiethylenetriamine. The structures of these complexes consist of two μ-tp bridging Cu(II) centers in a bis(monodentate) bonding fashion. The coordination geometry of the Cu(II) ions in these compounds may be described as close to square-based pyramid (SP) with severe significant distortion towards trigonal bipyramid (TBP) stereochemistry in 1. The visible spectra of the complexes in aqueous solutions are in complete agreement with the assigned X-ray geometry around the Cu(II) centers. Also, the solid infrared spectral data for the stretching frequencies of the tp-carboxalato groups, the ν(COO) reveals the existence of bis(monodentate) coordination mode for the bridged terephthalate ligand. The susceptibility measurements at variable temperature over the range 2-300 K are reported. Despite the same bonding mode of the tp bridging ligand, there has been observed slight antiferromagnetic coupling for the compounds 1 and 4 with J values of −0.5 and −2.9 cm3 K mol−1, respectively, and very weak ferromagnetic coupling for 2 and 3 with J values of 0.8 and 10.1 cm3 K mol−1, respectively. The magnetic results are discussed in relation to other related μ-terephthalato dinuclear Cu(II) published compounds.  相似文献

Six dinuclear platinum(II) complexes with a chiral tetradentate ligand, (1R,1′R,2R,2′R)-N1,N1′-(1,4-phenylenebis(methylene))dicyclohexane-1,2-diamine, have been designed, synthesized and characterized. In vitro cytotoxicity evaluation of these metal complexes against human A549, HCT-116, MCF-7 and HepG-2 cell lines have been carried out. All compounds showed antitumor activity to HepG-2, HCT-116 and A549. Particularly, compounds A1 and A2 exhibited significant better activity than other four compounds and A2 even showed comparable cytotoxicity to cisplatin against HepG-2 cell line.  相似文献

The dinuclear terephthalato-bridged nickel(II) complexes [Ni2(cyclen)2(μ-tp)](ClO4)2 (1) [Ni2(trpn)2(μ-tp)(H2O)2](ClO4)2 (2) and [Ni2(3,3,3-tet)2(μ-tp)(H2O)2](ClO4)2 · 2H2O (3), where tp = terephthalate dianion, cyclen = 1,4,7,10-tetraazacyclododecane, trpn = tris(3-aminopropyl)amine and 3,3,3-tet = 1,5,9,13-tetraazatridecane, were synthesized and structurally characterized by X-ray crystallography. Their magnetic susceptibilities were also determined at variable temperatures over the range 2-300 K. The structures of these complexes consist of μ-tp bridging two Ni(II) centers in a bis(bidentate) bonding fashion in 1 and in bis(monodentate) bonding fashion in 2 and 3. The coordination geometry around the Ni(II) ions in these compounds has a distorted octahedral geometry with four nitrogen atoms from the amine ligand (cyclen, trpn or 3,3,3-tet) and two coordinated oxygen atoms supplied by the chelated carboxylate group of the bridged terephthalate ligand in 1, and by one tp-carboxylate-oxygen in 2 and 3. The sixth coordination site in the last two complexes 2 and 3 is achieved via an oxygen atom from a coordinated water molecule. The intradimer Ni…Ni distances in these complexes are 10.740, 11.428 and 11.537 Å for 1, 2 and 3, respectively. The electronic spectra of the complexes in aqueous solutions are in complete agreement with the assigned X-ray geometry around the Ni(II) centers. Also, the analysis of the infrared spectral data for the ν(COO) stretching frequencies of the tp-carboxalato groups reveals the existence of the bis(bidentate) and bis(monodentate) coordination modes for the bridged terephthalate ligand in 1, 2 and 3, respectively. Despite the different coordination modes of the tp bridging ligand in these complexes, they all exhibit very weak antiferromagnetic coupling. The coupling constants J were found to be −2.2, −0.6 and −1.5 cm3 K mol−1 for the complexes 1, 2 and 3, respectively. The structural and magnetic results of 1-3 are discussed in relation to the other related published μ-terephthalato dinuclear Ni(II) compounds.  相似文献

Schiff base ligand (L) was obtained by condensation reaction between 4-aminopyrimidin-2(1H)-one (cytosine) with 2-hydroxybenzaldehyde. The synthesized Schiff base was used for complexation with Cu(II) and Fe(II) ions used by a molar (2 : 1 mmol ration) in methanol solvent. The structural features of ligand, Cu(II), and Fe(II) metal complexes were determined by standard spectroscopic methods (FT-IR, elemental analysis, proton and carbon NMR spectra, UV/VIS, and mass spectroscopy, magnetic susceptibility, thermal analysis, and powder X-ray diffraction). The synthesized compounds (Schiff base and its metal complexes) were screened in terms of their anti-proliferative activities in U118 and T98G human glioblastoma cell lines alone or in combination with electroporation (EP). Moreover, the human HDF (human dermal fibroblast) cell lines was used to check the bio-compatibility of the compounds. Anti-proliferative activities of all compounds were ascertained using an MTT assay. The complexes exhibited a good anti-proliferative effect on U118 and T98G glioblastoma cell lines. In addition, these compounds had a negligible cytotoxic effect on the fibroblast HDF cell lines. The use of compounds in combination with EP significantly decreased the IC50 values compared to the use of compounds alone (p<0.05). These results show that newly synthesized Cu(II) and Fe(II) complexes can be developed for use in the treatment of chemotherapy-resistant U118 and T98G glioblastoma cells and that treatment with lower doses can be provided when used in combination with EP.  相似文献

The synthesis and crystal structures of two new copper complexes with chelating dicarboxylic acids are described. Reaction of copper(II) acetate with diacid H2L2 (HO2CC(Me)2OArOC(Me)2CO2H, Ar=1,3-substituted phenyl) gave a bischelate complex (L2)2Cu2 · 2MeOH with the normal paddlewheel structure and tilted, trans-oriented chelate rings with skewed conformations. The overall structure was reasonably well reproduced by density functional calculations on (L2)2Cu2. Treatment of the product from reaction of Cu2(OAc)4 and diacid H2L3 (Ar=1,3-substituted 2,4-dibromophenyl) with pyridine gave a six-coordinate mononuclear chelate (L3)Py2Cu · H2O in which one chelate carboxylate is monodentate, the other is unsymmetrically bidentate, and the pyridines are cis-coordinated.  相似文献

Controlled hydrolysis of donor-substituted titanium-salan complexes led to the formation of well-defined dinuclear complexes. Structure determination by means of X-ray and NMR-studies revealed the presence of a single μ-oxo bridge and one labile alkoxide ligand per titanium center. Concomitant cytotoxicity assays of the isolated dinuclear complexes showed cytotoxicities in the low micro-molar region, surpassing in this respect even their monomeric ancestors, thus making them possible highly active metabolites of titanium-salan anti-cancer drugs.  相似文献

The dichlorobis(2-phenylazopyridine)ruthenium(II) complexes, [Ru(azpy)(2)Cl(2)], are under renewed investigation due to their potential anticancer activity. The three most common isomers alpha-, beta- and gamma-[RuL(2)Cl(2)] with L= o-tolylazopyridine (tazpy) and 4-methyl-2-phenylazopyridine (mazpy) (alpha indicating the coordinating Cl, N(pyridine) and Nazo atoms in mutual cis, trans, cis positions, beta indicating the coordinating Cl, N(pyridine) and Nazo atoms in mutual cis, cis, cis positions, and gamma indicating the coordinating Cl, N(pyridine) and Nazo atoms in mutual trans, cis, cis positions) are synthesized and characterized by NMR spectroscopy. The molecular structures of gamma-[Ru(tazpy)(2)Cl(2)] and alpha-[Ru(mazpy)(2)Cl(2)] are determined by X-ray diffraction analysis. The IC(50) values of the geometrically isomeric [Ru(tazpy)(2)Cl(2)] and [Ru(mazpy)(2)Cl(2)] complexes compared with those of the parent [Ru(azpy)(2)Cl(2)] complexes are determined in a series of human tumour cell lines (MCF-7, EVSA-T, WIDR, IGROV, M19, A498 and H266). These data unambiguously show for all complexes the following trend: the alpha isomer shows a very high cytotoxicity, whereas the beta isomer is a factor 10 less cytotoxic. The gamma isomers of [Ru(tazpy)(2)Cl(2)] and [Ru(mazpy)(2)Cl(2)] display a very high cytotoxicity comparable to that of the gamma isomer of the parent compound [Ru(azpy)(2)Cl(2)] and to that of the alpha isomer. These biological data are of the utmost importance for a better understanding of the structure-activity relationships for the isomeric [RuL(2)Cl(2)] complexes.  相似文献

Abstract

The side effects and resistance of metal-based anticancer drugs prompted us to synthesis a novel series of five Pd(II) complexes of the type [Pd(8-QO)(AA)]; where 8-QO?=?anion of 8-hydroxyquinoline and AA?=?anions of amino acids having nonpolar aliphatic side chain such as glycine (–H), alanine (–CH3), valine (–CH(CH3)2), leucine (–CH2–CH(CH3)2) and isoleucine (–CH(CH3)CH2–CH3). The complexes have been characterized with the help of FT-IR, UV–Vis, one and two-dimensional 1H-NMR, elemental analysis and conductivity measurements. On the basis of these characterization data, a four coordinated square planar geometry for all of these complexes have been proposed. The compounds were screened for their in vitro activities against human cancer cell line, MOLT-4 and their 50% inhibition concentration were ascertained by means of MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. Since four out of the five newly synthesized compounds were found to be more active than the standard anticancer drug, cisplatin, their detailed interaction with calf thymus DNA (as a target) and bovine serum albumin (BSA) (as a carrier) were also carried out by utilizing absorption spectra, fluorescence spectra and ethidium bromide displacement studies. In these experiments, several binding and thermodynamic parameters were also calculated. These results suggested that hydrogen binding and van der Waals forces play a major role in the interaction between metal complexes with CT-DNA and BSA.

The dinuclear dicarboxylato-bridged copper(II) complexes [Cu2(TPA)2(μ-tp)](ClO4)2 · H2O (1), [Cu2(TPA)2(μ-fum)](ClO4)2 · 2H2O (2) and [Cu2(pmedien)2(μ-fum)(H2O)2](ClO4)2 (3) (tp = terephthalate dianion, fum = fumarate dianion, TPA = tris(2-pyridylmethyl)amine and pmedien = N,N,N′,N″,N″-pentamethyldiethylenetriamine) were synthesized and structurally characterized by X-ray crystallography. The structures of the TPA complexes 1 and 2 consist of μ-tp or μ-fum bridging two Cu(II) centers in a bis(monodentate) bonding fashion. The coordination geometry around the Cu(II) ions in these compounds has a distorted trigonal bipyamidal geometry, TBP with four nitrogen atoms from the TPA ligand and a coordinated oxygen atom supplied by the carboxylate group of the bridged dicarboxylato ligand. Complex 3 has a distorted square pyramidal geometry achieved by the three N-atoms of the pmedien, one fum-carboxylate-oxygen and by an oxygen atom from a coordinated water molecule. The intradimer Cu…Cu distances in these complexes are 11.078(3), 8.663(4) and 9.520(3) Å for 1, 2 and 3, respectively. The electronic spectra of the complexes in aqueous solutions are in complete agreement with the assigned X-ray geometry around the Cu(II) centers. Also, analysis of the infrared spectral data for the ν(COO) stretching frequencies of the tp-carboxalato groups reveals the existence of the bis(mondentate) coordination mode for the bridged dicarboxylato ligands in compounds 1 and 2. The susceptibility measurements at variable temperature over the 2-300 K range are reported. For 1-3, it has been observed slight antiferromagnetic coupling with J values of −0.8, −3.0 and −2.9 cm−1, respectively.  相似文献
