In response to a balance disturbance, older individuals often require multiple steps to prevent a fall. Reliance on multiple steps to recover balance is predictive of a future fall, so studies should determine the mechanisms underlying differences between older adults who can and cannot recover balance with a single step. This study compared neural activation parameters of the major leg muscles during balance recovery from a sudden forward loss of balance in older individuals capable of recovering with a single step and those who required multiple steps to regain balance. Eighty-one healthy, community dwelling adults aged 70 ± 3 participated. Loss of balance was induced by releasing participants from a static forward lean. Participants performed four trials at three initial lean magnitudes and were subsequently classified as single or multiple steppers. Although step length was shorter in multiple compared to single steppers (F = 9.64; p = 0.02), no significant differences were found between groups in EMG onset time in the step limb muscles (F = 0.033–0.769; p = 0.478–0.967). However, peak EMG normalised to values obtained during maximal voluntary contraction was significantly higher in single steppers in 6 of the 7 stepping limb muscles (F = 1.054–4.167; p = 0.045–0.024). These data suggest that compared to multiple steppers, single steppers recruit a larger proportion of the available motor unit pool during balance recovery. Thus, modulation of EMG amplitude plays a larger role in balance recovery than EMG timing in this context.  相似文献

In this study we investigated balancing responses to lateral perturbations during slow walking (0.85 m/s). A group of seven healthy individuals walked on an instrumented treadmill while being perturbed at the level of waist at left heel strike in outward and inward lateral directions. Centre of mass (COM) and centre of pressure (COP), rotation of pelvis around vertical axis, step lengths, step widths and step times were assessed. The results have shown that beside control of COP in lateral direction, facilitated by adequate step widths, control of COP in sagittal direction, slowing down movement of COM was present after commencement of lateral perturbations. Sagittal component of COM was significantly retarded as compared to unperturbed walking for both inward (4.32 ± 1.29 cm) and outward (9.75 ± 2.17 cm) perturbations. This was necessary since after an inward perturbation first step length (0.29 ± 0.04 m compared to 0.52 ± 0.02 m in unperturbed walking) and step time (0.45 ± 0.05 s compared to 0.61 ± 0.04 s in unperturbed walking) were shortened while after an outward perturbation first two step lengths (0.36 ± 0.05 m and 0.32 ± 0.11 m compared to 0.52 ± 0.03 m in unperturbed walking) were shortened that needed to be accommodated by the described modulation of COP in sagittal plane. In addition pronounced pelvis rotation assisted in bringing swing leg to new location. The results of this study show that counteracting lateral perturbations at slow walking requires adequate response in all three planes of motion.  相似文献

Restoring functional gait speed is an important goal for rehabilitation post-stroke. During walking, transferring of one’s body weight between the limbs and maintaining balance stability are necessary for independent functional gait. Although it is documented that individuals post-stroke commonly have difficulties with performing weight transfer onto their paretic limbs, it remains to be determined if these deficits contributed to slower walking speeds. The primary purpose of this study was to compare the weight transfer characteristics between slow and fast post-stroke ambulators. Participants (N = 36) with chronic post-stroke hemiparesis walked at their comfortable and maximal walking speeds on a treadmill. Participants were stratified into 2 groups based on their comfortable walking speeds (≥0.8 m/s or <0.8 m/s). Minimum body center of mass (COM) to center of pressure (COP) distance, weight transfer timing, step width, lateral foot placement relative to the COM, hip moment, peak vertical and anterior ground reaction forces, and changes in walking speed were analyzed. Results showed that slow walkers walked with a delayed and deficient weight transfer to the paretic limb, lower hip abductor moment, and more lateral paretic limb foot placement relative to the COM compared to fast walkers. In addition, propulsive force and walking speed capacity was related to lateral weight transfer ability. These findings demonstrated that deficits in lateral weight transfer and stability could potentially be one of the limiting factors underlying comfortable walking speeds and a determinant of chronic stroke survivors’ ability to increase walking speed.  相似文献

The central nervous system (CNS) utilizes anticipatory (APAs) and compensatory (CPAs) postural adjustments to maintain equilibrium while standing. It is known that these postural adjustments involve displacements of the center of mass (COM) and center of pressure (COP). The purpose of the study was to investigate the relationship between APAs and CPAs from a kinetic and kinematic perspective. Eight subjects were exposed to external predictable and unpredictable perturbations induced at the shoulder level while standing. Kinematic and kinetic data were recorded and analyzed during the time duration typical for anticipatory and compensatory postural adjustments. When the perturbations were unpredictable, the COM and COP displacements were larger compared to predictable conditions with APAs. Thus, the peak of COM displacement, after the pendulum impact, in the posterior direction reached 28 ± 9.6 mm in the unpredictable conditions with no APAs whereas it was 1.6 times smaller, reaching 17 ± 5.5 mm during predictable perturbations. Similarly, after the impact, the peak of COP displacement in the posterior direction was 60 ± 14 mm for unpredictable conditions and 28 ± 3.6 mm for predictable conditions. Finally, the times of the peak COM and COP displacements were similar in the predictable and unpredictable conditions. This outcome provides additional knowledge about how body balance is controlled in presence and in absence of information about the forthcoming perturbation. Moreover, it suggests that control of posture could be enhanced by better utilization of APAs and such an approach could be considered as a valuable modality in the rehabilitation of individuals with balance impairment.  相似文献

The present study compared neuromuscular adaptations to 12 weeks of plyometric (PLY) or pneumatic (PNE) power training and their effects on dynamic balance control. Twenty-two older adults aged 60–70 (PLY n = 9, PNE n = 11) participated in the study. Measurements were conducted at Pre, 4, 8 and 12 weeks. Dynamic balance was assessed as anterior–posterior center of pressure (COP) displacement in response to sudden perturbations. Explosive isometric knee extension and plantar flexion maximal voluntary contractions (MVCs) were performed. Maximal drop jump performance from optimal dropping height was measured in a sledge ergometer. Increases in knee extensor and ankle plantar flexor torque and muscle activity were higher and occurred sooner in PNE, whereas in drop jumping, PLY showed a clearer increase in optimal drop height (24%, p < 0.01) after 8 weeks of training and soleus muscle activity after 12 weeks of training. In spite of these training mode specific adaptations, both groups showed similar improvements in dynamic balance control after 4 weeks of training (PLY 38%, p < 0.001; PNE 31%, p < 0.001) and no change thereafter. These results show that although power and plyometric training may involve different neural adaptation mechanisms, both training modes can produce similar improvements in dynamic balance control in older individuals. As COP displacement was negatively correlated with rapid knee extension torque in both groups (PLY r = −0.775, p < 0.05; PNE r = −0.734, p < 0.05) after training, the results also highlight the importance of targeting rapid force production when training older adults to improve dynamic balance.  相似文献

ObjectiveTo provide normative postural stability data in young subjects.MethodsNinety-six healthy participants (58 W, 28 ± 6y) stood on a force plate during 60 s. We measured effects of support width (feet apart, FA; feet together, FT), vision (eyes open, EO; closed, EC), and cognitive load (single task, ST; dual tasking, DT) on anteroposterior (AP) and medio-lateral (ML) ranges, area and planar velocity of center of pressure (COP) trajectory.ResultsAll variables increased with FT (AP range, +15%; ML, +185%; area, +242%; velocity, +50%, p < 0.0002 for all, MANOVA). Visual deprivation increased COP ranges with added constraints (FT or DT, p = 0.002) and increased velocity in all conditions (FA/ST, +16%; DT, +18%; FT/ST, +29%; DT, +23%, p < 0.0002 for all). Dual tasking reduced COP displacements with FT (AP range, EO, −15%; EC, −11%; ML range, EO, −19%; EC, −13%; area, EO, −40%; EC, −28%, p < 0.0002 for all) and increased velocity in most conditions (FA/EO, +15%; FA/EC, +16%; FT/EO, +7%, p < 0.0002 for all).ConclusionIn young healthy adults, base of support reduction increases COP displacements. Vision particularly affects postural stability with feet together or dual tasking. Dual tasking increases velocity but decreases COP displacements in challenging postural tasks, potentially by enhanced lower limb stiffness.  相似文献

Most hip fractures are thought to occur after falling during everyday activities. We speculated that hip fractures might also occur because of excessive loading of the hip joint during an unexpected misstep consequently leading to a fall. The aims of this study were to explore the kinematics and kinetics of the lower extremity joints during missteps as compared with regular stepping, as well as to compare the magnitude of forces acting upon the hip joint with the threshold forces expected to fracture the hip. Fourteen healthy adults performed two forward steps on a 17.8 cm high platform under the following four conditions: forward with and without vision, as well as a misstep (the box for the final step unexpectedly removed without participant awareness), and regular stepping down with eyes open. The results revealed no differences between stepping forward with and without vision. When compared with both stepping forward and regular stepping down, the misstep revealed altered joint positions accompanied by increased forces and moments acting upon the hip joint. For example, the peak vertical proximal thigh segment force was 3.05±0.55 BW vs. 1.23±0.14 BW and 0.91±0.09 BW (p<.001; misstep vs. regular stepping down and stepping forward, respectively), while the proximal thigh segment moment in frontal plane was 1.39±0.70 Nm/kg vs. 0.18±0.32 Nm/kg of adduction and 0.16±0.19 Nm/kg of abduction (p<.001). When compared with the literature data, the forces during misstep were within the range of those forces that could result in hip fractures in the elderly. Therefore, it may be possible for the elderly to experience hip/proximal femur fractures during missteps prior to falling.  相似文献

The aim of the current study was to investigate the response of healthy older and young adults to a change in step direction from a forward to a lateral target in response to a sensory perturbation. Nine healthy older (75.1 ± 6.7 years; age range, 65–81 years) and nine young adults (27 ± 3.6 years; age range, 23–31 years) participated in the study. The sensory perturbation was a visual cue presented at random times while subjects stepped over an obstacle from quiet stance. For both young and elderly subjects there was an abrupt change in the slope of both shear ground reaction forces (GRFs) of the stance limb following the perturbation. The slope and peak of the change in GRFs was greater for the young subjects and the onset significantly earlier (205 ms compared to 271 ms). Changes in the GRFs were accompanied by an increase in bilateral gluteus medius and stance limb soleus activity. A late visual cue resulted in a delayed response for elderly subjects. These data show that a stepping response to a sensory perturbation was both delayed and of less magnitude for older adults which has implications for fall risk.  相似文献

The process of corn cobs alkaline hydrolysis to produce solutions with high hydroxy-cinnamic acids content was investigated. In particular the attention was focused on the solubilisation of ferulic acid (FA) and related compounds, mainly p-coumaric acid (p-CA). Although these compounds have applications as antioxidants, the purpose of this work was to obtain FA solutions that can be used as feedstock for the biotechnological production of vanillin in future studies. The effects of different concentrations of NaOH (0.2  Ca  2.0N) and solid/liquid ratios (0.028  S/L  0.168 g/g) on the solubilisation of FA versus time have been investigated at room temperature. Optimal hydrolysis conditions (Ca = 0.5N, S/L = 0.084 g/g after 6 h) ensured the production of hydrolysates with relatively high contents of both FA (1171 ± 34 mg/L) and p-coumaric acid (2156 ± 64 mg/L), which can be used in future studies for the microbial transformation into vanillin.  相似文献

The kinetics of a stomatal response to sudden increases or decreases of CO2 concentrations ([CO2]) was studied in 13 plant species growing in the field. Plants were well supplied with water. In each plant, gas exchange measurements were made on a fully developed leaf that was first left to achieve steady-state stomatal conductance (gs) at 400 μmol (CO2) mol−1) and then exposed to a step change of [CO2] (to 700 μmol mol−1 in one experiment; and to 700 and back to 400 μmol mol−1 in a second experiment). Porometric data were captured in intervals of 3 s until a new steady state was reached.A comparison of t1/2, the half-time needed to achieve new gs, indicates similar responses of stomata in grasses when compared to herbs. The stomata of C4 plants responded in approximately 5 min, the highest closure rate was detected in Echinochloa crus-galli and Digitaria sanguinalis. Opening rates were similar to closing rates and the response as a whole was rather symmetric. In C3 plants, the full response of stomata was much slower. Analysis revealed differences in absolute rates of gs change between C3 and C4 plants. These differences can be related to the specificities of the type of photosynthetic metabolism. C4 photosynthesis enables plants to reduce gs, which can hasten further changes of diffusivity in response to the environmental signals. A possible coupling of C4 metabolism to the regulation of guard cells also has to be taken into account when explaining the observed results.  相似文献

Technical advancements in instrumentation and analytical methods have improved the ability of assessing balance control. This study investigated the effects of early stages of aging on postural sway using traditional and contemporary postural indices from different domains. Eleven healthy young adults and fourteen healthy non-faller older adults performed two postural tasks: (a) functional limits of stability and (b) unperturbed bipedal stance for 120 s. Postural indices from spatial, temporal, frequency, and structural domains were extracted from the body’s center of pressure (COP) signals and its Rambling and Trembling components. Results revealed a preservation of functional limits of upright stability in older adults accompanied by larger, faster, and shakier body sway in both anterior-posterior and medio-lateral directions; increased medio-lateral sway frequency; increased irregularity of body sway pattern in time in both directions; and increased area, variability, velocity, and jerkiness of both rambling and trembling components of the COP displacement in the anterior-posterior direction (p < 0.02). Such changes might be interpreted as compensatory adjustments to the age-related decline of sensory, neural, and motor functions. In conclusion, balance assessment using postural indices from different domains extracted from the COP displacement was able to capture subtle effects of the natural process of aging on the mechanisms of postural control. Our findings suggest the use of such indices as potential markers for postural instability and fall risk in older adults.  相似文献

The study investigated relations between effects of repeated ankle plantar-flexion movements exercise on the soleus Hoffmann (H) reflex and on postural body sway when maintaining upright stance. Ten young volunteers performed five sets of ankle plantar-flexions of both lower limbs. Assessment of the feet centre-of-pressure (COP) displacement and H-reflex tests were carried out in quiet stance before, during and after the exercise. H-max and M-max responses were obtained in 8 subjects and reported as the peak-to-peak amplitudes of the right soleus muscle electromyographic waves. Mean dispersion of COP along the antero-posterior direction increased significantly during the exercise; whilst the overall H-reflex response indicated a reduction without a concomitant modification in the M-max response. H-reflex responses, however, varied between participants during the first sets of exercise, showing two main trends of modulation: either depression or early facilitation followed by reduction of the H-reflex amplitude. The extent of reflex modulation in standing position was correlated to the concentric work performed during the exercise (r = 0.85; p < 0.01), but not to the antero-posterior COP dispersion. These results suggest that during a repeated ankle plantar-flexions exercise, modulation of the H-reflex measured in upright stance differs across individuals and is not related to changes of postural sway.  相似文献

Composition and phase dependence of the mixing of 1,2-Dipalmitoyl-sn-glycero-3-phosphocholine (DPPC), and 1,2-Dioleoyl-sn-glycero-3-phosphocholine (DOPC), with the oxidized phospholipid, 1-palmitoyl-2-glutaryl-sn-glycero-3-phosphocholine (PGPC) were investigated by characterizing the aggregation states of DPPC/PGPC and DOPC/PGPC using a fluorescence quenching assay, dynamic light scattering, and time-resolved fluorescence quenching in the temperature range 5–60 °C. PGPC forms 3.5 nm radii micelles of aggregation number 33. In the gel phase, DPPC and PGPC fuse to form mixed vesicles for PGPC molar fraction, XPGPC  0.3 and coexisting vesicles and micelles at higher XPGPC. Data suggest that liquid phase DPPC at 50 °C forms mixed vesicles with segregated or hemi fused DPPC and PGPC for XPGPC  0.3. At 60 °C, DPPC and PGPC do not mix, but form coexisting vesicles and micelles. DOPC and PGPC do not mix in any proportion in the liquid phase. Two dissimilar aggregates of the sizes of vesicles and PGPC micelles were observed for all XPGPC for T  22 °C. DOPC–PGPC and DPPC–PGPC mixing is non-ideal for XPGPC > 0.3 in both gel and fluid phases resulting in exclusion of PGPC from the bilayer. Formation of mixed vesicles is favored in the gel phase but not in the liquid phase for XPGPC  0.3. For XPGPC  0.3, aggregation states change progressively from mixed vesicles in the gel phase to component segregated mixed vesicles in the liquid phase close to the chain melting transition temperature to separated coexisting vesicles and micelles at higher temperatures.  相似文献

A water footprint considers both the water volumes involved in production processes and the resulting waste water generated. The grey water (GW) footprint represents the volume of fresh water required to assimilate pollutants to acceptable concentrations—a concept proposed by the water footprint network—but it faces several difficulties when applied to agricultural production systems. Crop production cannot be fully controlled and it is weather-dependent, which greatly affects the year-to-year GW calculations.In this study, we examined the effect of time step on the calculation of annual GW footprints by utilizing 30 years of daily average nitrate-nitrogen (NO3-N) concentrations in drainage water (both leachate and runoff water derived from a process-based model) from corn and soybean production systems. For each crop year, the volume of water required to assimilate NO3-N to an acceptable threshold concentration (i.e. <10 mg L−1) was calculated over different time steps (daily, weekly, monthly, seasonally and yearly), and each case was summed to an annual GW value. Daily average NO3-N concentrations in the effluent water were generally below the acceptable threshold concentrations, with intermittent exceedances. Thus, the fields often provided their own ‘dilution’ water, and annual average concentrations were only 2.0 mg L−1 and 0.4 mg L−1 for corn and soybean, respectively.The GW footprint varied significantly when calculated for different time steps. The greatest annual footprint occurred when calculated daily (shortest time step). The GW footprint for corn ranged from 2.7 × 103 m3 ha−1, or 2700 mm of water, when estimated daily to zero for the yearly time step. For soybean it ranged from 0.5 × 103 m3 ha−1, or 500 mm of water, to zero. The GW footprint results are therefore highly dependent on the time step of calculation. The effect of this issue extends beyond crop production as it is exported and amplified through feed rations to affect the GW footprint from animal production. To be able to reconcile these problems, the GW calculation pathways should be reconsidered and standardized.  相似文献

An amperometric immunosensor for cardiac troponin T detection in human serum troponin T, a marker considered as “gold standard” for acute myocardial infarction diagnosis, is described. A stable carboxylic film to covalently bind antibodies against cTnT onto electrode surface was achieved with electropolymerization of the o-aminobenzoic acid. A fractional factorial study was performed to optimize the electropolymerization parameters. Cyclic voltammetry assays were carried out for characterize steps of the modified electrode surface. The obtained calibration curve at −0.05 V by amperometry presented a good linear response range from 0.05 to 5.0 ng mL−1 cTnT with a correlation coefficient of 0.992 (n = 6) and 0.016 ng mL−1 detection limit. The electrodes showed a good stability upon the analytical responses retaining 91.6% of its initial response after 18 days. This sensor showed outgoing results regarding sensitivity allowing reliable measurements of the cTnT at levels of clinical significance for acute myocardial infarctions diagnosis.  相似文献

A psychrothermotolerant alkaline protease isolated from Bacillus pumilus MP27 with a molecular mass ∼53 kDa was isolated from Southern ocean water samples. It was partially purified by single step TPP with purity fold of 16.65. The enzyme was found to be widely stable within a range of temperature and pH, maintaining 52.25% of its activity at 50 °C and 92% at pH 12. The enzyme exhibited an exceptional activity along with tested detergents, showing 98% stability with SDS (10 mg/ml) and ̴ 99% stability with Tide detergent (7 mg/ml). Further, the alkaline protease gene of 1152 bp was successfully cloned in pGEM-T Easy vector in E. coli DH5α. The gene sequence was further translated, modeled and molecular dynamic simulation was performed. The modeled protein was highly unstable during the first 5 ns and therefore could not able to form bonds with the ligand after 1 ns of simulation.  相似文献

N,N-dimethyldodecylamine-N-oxide (C12NO) is a surfactant that may exist either in a neutral or cationic protonated form depending on the pH of aqueous solutions. Using small angle X-ray diffraction (SAXD) we observe the rich structural polymorphism of pH responsive complexes prepared due to DNA interaction with C12NO/dioleoylphosphatidylethanolamine (DOPE) vesicles and discuss it in view of utilizing the surfactant for the gene delivery vector of a pH sensitive system. In neutral solutions, the DNA uptake is low, and a lamellar Lα phase formed by C12NO/DOPE is prevailing in the complexes at 0.2  C12NO/DOPE < 0.6 mol/mol. A maximum of ~ 30% of the total DNA volume in the sample is bound in a condensed lamellar phase LαC at C12NO/DOPE = 1 mol/mol and pH 7.2. In acidic conditions, a condensed inverted hexagonal phase HIIC was observed at C12NO/DOPE = 0.2 mol/mol. Commensurate lattice parameters, aHC  dLC, were detected at 0.3  C12NO/DOPE  0.4 mol/mol and pH = 4.9–6.4 suggesting that LαC and HIIC phases were epitaxially related. While at the same composition but pH ~ 7, the mixture forms a cubic phase (Pn3m) when the complexes were heated to 80 °C and cooled down to 20 °C. Finally, a large portion of the surfactant (C12NO/DOPE > 0.5) stabilizes the LαC phase in C12NO/DOPE/DNA complexes and the distance between DNA strands (dDNA) is modulated by the pH value. Both the composition and pH affect the DNA binding in the complexes reaching up to ~ 95% of the DNA total amount at acidic conditions.  相似文献

Mycobacteria have been modified to transform sterols to produce valuable steroids. Here, we demonstrated that the oxidation of sterols to sterones is a rate-limiting step in the catabolic pathway of sterols in Mycobacterium neoaurum. Two cholesterol oxidases ChoM1 and ChoM2 involved in the step were identified in M. neoaurum and the ChoM2 shared up to 45% identity with other cholesterol oxidases. We demonstrated that the combination of ChoM1 and ChoM2 plays a significant role in this step. Accordingly, we developed a strategy to overcome this rate-limiting step by augmenting the activity of cholesterol oxidases in M. neoaurum strains to enhance their transformation productivity of sterols to valuable steroids. Our results indicated that the augmentation of ChoM2 achieved 5.57 g/l androst-1,4-diene-3,17-dione in M. neoaurum NwIB-01MS and 6.85 g/l androst-4-ene-3,17-dione in M. neoaurum NwIB-R10, greatly higher than the original yield, 3.87 g/l androst-1,4-diene-3,17-dione and 4.53 g/l androst-4-ene-3,17-dione, respectively.  相似文献

Tetranychus urticae populations were collected from ten commercial apple orchards and their susceptibilities to 12 acaricides were tested using a leaf disc bioassay. The resistance of each T. urticae population was reported as the LC50 value, the resistance ratio (RR) and the slope of the probit–concentration regression. Cross resistances of T. urticae populations were estimated using the Spearman's rank correlation coefficient. Most local populations showed low resistance levels (RR  10). Development of resistance to METI and pyrethroid acaricides differed among local populations. The highest RR value (154.6) was found in the Uiseong population to tebufenpyrad. The Geochang population was highly resistant, especially to METI and pyrethroid acaricides. T. urticae populations collected from Suwon, Chungju, Yeongju and Geochang showed moderate resistance (10 < RR  40) to more than two acaricides. Resistance ratios to abamectin, chlorfenapyr, fenbutatin-oxide and milbemectin were low (RR  10) in all populations. The LC50 values of abamectin, chlorfenapyr, fenbutatin-oxide and milbemectin ranged from 0.06 to 0.2 mg/l, from 0.67 to 3.38 mg/l, from 10.12 to 40.85 mg/l and from 0.47 to 3.01 mg/l, respectively. We discuss possible cross-resistance to acaricides using Spearman's rank correlation coefficient.  相似文献

This prospective study examined normalized stability differences based on dominance side and visual feedback. Subjects with low back pain (LBP) (n = 26; 9 men, 17 women) and without LBP (n = 28; 11 men, 17 women) participated in this study. All subjects were asked to maintain single leg standing balance with the contralateral hip flexed 90° for 25 s. The outcome measures included normalized holding duration and stability. The combined rotation (Rxyz) was also calculated to compare the upper and lower thorax and lumbar axes relative to the core spine axis. The holding duration was significantly different between groups (T = ?2.21, p = 0.03). The subjects without recurrent LBP (control group) demonstrated longer hold duration times (24.60 ± 4.2 s) than the subjects with recurrent LBP (21.2 ± 7.1 s). For the normalized hold duration, there was a significant difference between groups based on visual input (F = 7.13, p = 0.009). There was also a significant difference in standing stability based on visual input (F = 93.93, p = 0.0001) and trunk area (F = 101.51, p = 0.0001). In addition, the normalized stability was significantly different based on dominance and visual input (F = 11.28, p = 0.002). Therefore, trunk stability could prompt an uncoordinated bracing effect with poor proprioception from injury to passive structures or due to interference of pain during central processing of information in subjects with recurrent LBP.  相似文献
