Control of the alary muscles of locust dorsal diaphragm   总被引:1,自引:0,他引:1  
ABSTRACT. The heartbeat in whole, intact, adult Locusta migratoria R.F. was characterized by a regular rate but apparently irregular amplitude. Cutting segmental nerves often eliminated apparent amplitude fluctuations, and electrically shocking a segmental nerve in the whole animal evoked apparent amplitude changes corresponding to the shocks. Saline-perfused tissue preparations showed that the apparent amplitude fluctuations could be duplicated by segmental nerve stimulation, and that the fluctuations were due largely to contractions of the alary muscles of the dorsal diaphragm which shifted the position of the heart chamber without a change in volume. The alary muscles are each multi-terminally innervated by one motor axon. Neurally-evoked postsynaptic potentials facilitated and summated, and the diaphragm muscles began visibly contracting at stimulation rates as low as 2 Hz. Stimulation at higher frequencies caused greater depolarization of the muscle fibres with no indication of electrically-excited responses. The alary muscles were insensitive to perfusion with acetylcholine, l -glutamate, l -aspartate, dopamine, octopamine, noradrenaline, proctolin, 5-hydroxytryptamine, or gamma aminobutyric-acid in saline at concentrations up to 10-3M. Larval or adult brain extracts of Locusta at 10 μg/μl and diluted 1:5 in saline caused uniform contractions of the alary muscle preparation, while perfusion of skeletal muscle extracts produced no response.  相似文献

  • 1.1. The peripheral axon distribution from two bursting neurons was investigated using electrophysiological methods. Both of these cells send out a bundle of axon branches which goes via the visceral nerve to the heart and in the uterine nerve.
  • 2.2. The relative number of axon branches in the two nerves was determined through double-shock experiments.
  • 3.3. The axon bundle takes the visceral nerve and its uterine branch, supplying the effector systems in parallel.
  • 4.4. Slight differences in conduction velocity between the axon branches produce an enlargement of the efferent volley.
  • 5.5. The number of active axon branches conveying orthodromic or antidromic spikes is controlled by inhibitory potentials converging onto the initial part of the bundle, so that the two bursting cells amount to a pool of 35 to 40 interconnected nerve cells.
  • 6.6. The atypical axonal distribution of the two bursting cells might be related to their neurosecretory properties.

Summary The alary muscles of Locusta migratoria adults make up the major tissue of the dorsal diaphragm which separates pericardial and perivisceral sinuses in the abdomen. The alary muscles are striated with a sarcomere at rest measuring about 9 m. The Z-line has a staggered-beaded arrangement with A-bands and I-bands readily discernable. Thick myofilaments are surrounded by 10 or more thin filaments. The sarcoplasm has few mitochondria near the area of the Z-line, dyads are present and sarcoplasmic reticulum is poorly developed. Axons which innervate the alary muscle are either contained within invaginated folds of the sarcolemma of the muscle cells or the muscle cells send finger-like projections to envelop the axons. The synaptic terminals contain synaptic vesicles between 40 and 45 nm in diameter and a few electron-dense granules near or less than 170 nm in diameter. Away from synaptic terminals the axon profiles show few or no granules. The axons are accompanied everywhere by well-developed glial cells. This then is not typical neurosecretomotor innervation, however, the presence of electron-dense granules suggests the possibility of peptidergic neurotransmission.  相似文献

Previously described salines for lepidoptera did not maintain a constant heart rate for a very long. We have been successful in maintaining a normal heartbeat for many hours in a newly designed saline. This saline was also suitable for maintaining normal neuromuscular junctional potentials. The cardiac reflexes studied in larvae of Bombyx and Agrius were five types of cardiac responses induced by mechanical stimuli to sensillar setae. The cardiac responses were caused by electrical stimulation of nerves in the reflex pathways. The antidromic heartbeat was triggered even in larvae before the 5th instar by stimulation of axons in the visceral nerve arising from the frontal ganglion and terminating at the aorta, while spontaneous heartbeat reversal started to occur in wandering larvae. Other axons in the visceral nerve terminate at the rear end of the heart. Electrical stimuli to the nerves caused cardiac inhibition of the orthodromic heartbeat. Nerves extending from the visceral nerve to the alary muscles of the 2nd abdominal segment contain axons to increase the tone of the muscles. Nerves extending from the 7th abdominal ganglion to the most posterior alary muscles also contain axons to increase the tone of the muscles, and were responsible for acceleration of the antidromic and orthodromic heartbeat, respectively.  相似文献

Dry weight, protein, lipid, and glycogen were determined at various times during cultivation of the Chytridiomycetes, Rhizophydium sphaerotheca and Monoblepharella elongata. M. elongata had relatively stable levels of glycogen, but, in R. sphaerotheca, glycogen levels showed significant changes, particularly in older cultures in which a depletion of glycogen was accompanied by a marked thickening of the cell walls. Glycogen was a significant cellular constitutent in both chytridiomycete species. In R. sphaerotheca and M. elongata, respectively, glycogen accounted for as much as 6% and 8.1% of the dry weight. In purified glycogens of both species, only -1,4-and -1,6-linked glucosyl residues were detected and the absorbance spectra of I2-complexes were similar to those of other well characterized glycogens. Purified Rhizophydium glycogen had a -amylolysis limit of 43%, and a of approximately 12. For the Monoblepharella polysaccharide, the respective values were 45% and 11. In extracts of the Oomycetes, Pythium debaryanum, Mindeniella spinospora, and Apodachlya sp., only -1,3- and -1,6-linked glucosyl residues were detected. These glucans were not iodophilic nor were they sensitive to -amylase and -amylase. The properties of the oomycete polysaccharides suggested that they were similar to the mycolaminarans of Phytophthora spp. Although both investigated chytridiomycete species produced glycogen with typical properties, glycogen was apparently absent in the investigated Oomycetes.  相似文献

Summary Crustacean cardioactive peptide-immunoreactive neurons occur in the entire central nervous system of Locusta migratoria. The present paper focuses on mapping studies in the ventral nerve cord and on peripheral projection sites. Two types of contralaterally projecting neurons occur in all neuromers from the subesophageal to the seventh abdominal ganglia. One type forms terminals at the surface of the thoracic nerves 6 and 1, the distal perisympathetic organs, the lateral heart nerves, and on ventral and dorsal diaphragm muscles. Two large neurons in the anterior part and several neurons of a different type in the posterior part of the terminal ganglion project into the last tergal nerves. In the abdominal neuromers 1–7, two types of ipsilaterally projecting neurons occur, one of which gives rise to neurosecretory terminals in the distal perisympathetic organs, in peripheral areas of the transverse, stigmata and lateral heart nerves. Four subesophageal neurons have putative terminals in the neurilemma of the nervus corporis allati II, and in the corpora allata and cardiaca. In addition, several immunoreactive putative interneurons and other neurons were mapped in the ventral nerve cord. Summary Cephalopod chromatophores are made of a central pigment cell surrounded by 10 to 20 radially arranged muscle fibres under direct nervous control. Innervation of these muscle fibres was studied with anterograde cobalt fills of peripheral nerve bundles and light and electron microscopy. Individual axons branch repeatedly to innervate the muscles of chromatophores scattered over several millimeters. Axons contained in several dermal nerves converge to innervate the same chromatophores. Among the chromaophores, axons were found running either singly or in small bundles, often accompanied by sheath cells. Single chromatophore muscles were innervated by at least one axon running across or along its length. Since nerves terminating on chromatophore muscles are very rare, neuromuscular contact seems to be made en passant. Varicosities of the axons apposed to the muscles are thought to be presynaptic sites. However, morphological differentiations of the pre-or post-synaptic membranes were not visible. Summary Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) were employed to study the innervation and vascular supply of crayfish skeletal muscle.Blood vessels and nerve terminals identified by TEM were often closely associated. Synaptic regions of the nerve terminals were always located under sarcolemma and contained both dense-cored and agranular synaptic vesicles. Axo-axonal synapses of several different types were observed. Blood vessels consisted of several vessel cells or supporting cells enclosing a lumen, which was connected to the exterior by fine channels between the supporting cells.SEM of whole freeze-dried muscles revealed two types of ramifying structure, which often ran in parallel over the muscle surface. One, identified as nerve, was more cylindrical and had a smoother surface than the other, which was identified as blood vessel. Fine nerve branches disappeared under the sarcolemma, probably near synaptic regions, but synapses could not be seen. Blood vessels also had fine terminations which merged into the sarcolemma.Supported by grants from the National Research Council of Canada and The Muscular Dystrophy Association of Canada. Cells of Nostoc commune UTEX 584 from liquid cultures expressed an upshift in nitrogenase activity when immobilised on inert supports and exposed to matric water potentials between -1.10 and -99.5 MPa. Cells incubated at 0.10 MPa (aw=c 1.0) maintained increased activity for at least 48 h following immobilization. At water potentials below -23.1 MPa (aw=0.85), the upshift was transitory. Nitrogenase activity decreased rapidly when immobilised cells were incubated at lower values of m.Desiccated cells stored at -99.5 MPa (aw=0.50) underwent an upshift in nitrogenase activity, and in the size of the intracellular ATP pool, when rewetted with either distilled water or liquid MBo medium (o =-0.18 MPa). The upshift in nitrogenase activity was chloramphenicol-sensitive and was preceeded by a lag. The duration of the lag depended on the time taken to equilibrate cells to-99.5 MPa, the time desiccated, and the conditions of storage and rewetting. Cells that had no, or very low, nitrogenase activity when rewetted in air, showed a marked stimulation of nitrogenase activity in the presence of 5% v/v CO2 under both aerobic and anerobic conditions.When rewetted in the presence of 1% w/v glucose (o =-0.14 MPa), vegetative cells remained intact, but heterocysts underwent autolysis and nitrogenase activity was not detected, even in the presence of 5% v/v CO2.Abbreviations TTC 2,3,5-triphenyl-2-tetrazolium chloride - m matric water potential - o osmotic water potential - aw water activity  相似文献

Summary Intraepidermal nerve fibers of human glabrous digital skin were investigated using a new silver impregnation method. Nerves were observed to enter the epidermis without regional preference, and to extend into the stratum granulosum. They are non-varicose (smooth) or varicose and range from less than 0.2 m to approximately 2 m in diameter, with varicosities up to 3 m in diameter. Some axons branch profusely within the epithelium, giving off fine branches of differing diameters, while others appear to remain unbranched. At least some intraepidermal axons are fine branches of larger axons taking a horizontal course below the epithelial layer. Others are, at least topically, closely associated with Meissner's corpuscles. At 57 nerves per mm2 surface area, the density of intraepidermal nerves found in this investigation is much greater than that reported in recent publications, and agrees closely with values given in several older studies.Dedicated to Prof. Dr. A. Summary Following observation of conical groups of stiff, but motile cilia on the tentacles of the branchial crown of Sabella pavonina, these were examined with the electron microscope. The bundles consist of about 40 unenclosed standard cilia supported by one or two primary sense cells with centrally directed axons of 0.1–0.2 diameter. Axons in the distal portions of the branchial crown occur in small bundles surrounded by a basement membrane. More centrally, glial elements appear and the nerves are surrounded by a collagenous sheath. The branchial nerve trunk shows similarities in organisation to other previously investigated annelid central nervous tissue in that the whole nerve is surrounded by a fibrous sheath central to which there is a layer of glial cells with processes penetrating a central neuropile. The 0.1–0.2 axons commonly occur in glial-enveloped groups of < 40 whilst other axons of larger and mixed diameter are found together.Each tentacle has two branchial nerves on the oral side, and each nerve gives rise to two small 75-axon branches running to each pinnule. The branchial nerves fuse to form the branchial nerve trunk running to the supra-oesophageal ganglia.Sections of the branchial nerves of the branchial crown at progressively more central levels show that the branchial nerve trunk contains enough axons of 0.1–0.2 diameter to account for all the sensory cells on the tentacles. This is taken as evidence for the sensory cells having axons terminating within the central nervous system and that there is no peripheral confluence or fusion of these afferent axons.  相似文献

We have followed the normal development of the different cell types associated with the Drosophila dorsal vessel, i.e. cardioblasts, pericardial cells, alary muscles, lymph gland and ring gland, by using several tissue-specific markers and transmission electron microscopy. Precursors of pericardial cells and cardioblasts split as two longitudinal rows of cells from the lateral mesoderm of segments T2-A7 (cardiogenic region) during stage 12. The lymph gland and dorsal part of the ring gland (corpus allatum) originate from clusters of lateral mesodermal cells located in T3 and T1/dorsal ridge, respectively. Cardioblast precursors are strictly segmentally organized; each of T2-A6 gives rise to six cardioblasts. While moving dorsally during the stages leading up to dorsal closure, cardioblast precursors become flattened, polarized cells aligned in a regular longitudinal row. At dorsal closure, the leading edges of the cardioblast precursors meet their contralateral counterparts. The lumen of the dorsal vessel is formed when the trailing edges of the cardioblast precursors of either side bend around and contact each other. The amnioserosa invaginates during dorsal closure and is transiently attached to the cardioblasts; however, it does not contribute to the cells associated with the dorsal vessel and degenerates during late embryogenesis. We describe ultrastructural characteristics of cardioblast differentiation and discuss similarities between cardioblast development and capillary differentiation in vertebrates. Correspondence to: V. Zusammenfassung In einer Muskelspindel der Ratte, die an einer Serie alternierender Semidünn- und Ultradünnquerschnitte untersucht wurde, wurden zwei benachbart an nuclear chain-Fasern gelegene Nervenendformationen unterschiedlicher Bauweise festgestellt. Die beiden Endformationen sind mit ein und derselben Nervenfaser der Gruppe II verbunden und daher als Bestandteile einer sekundären sensorischen Endigung zu betrachten.Die Nervenendformation an einer der beiden nuclear chain-Fasern hat anulo-spirale Form.Die Nervenendformation an der anderen nuclear chain-Faser weist am Querschnittsbild eine Anzahl von Axonen auf, die zwischen Plasmalemm und Basalmembran der intrafusalen Muskelfaser eng aneinanderliegen. Nicht alle Axonquerschnitte stehen in direktem Kontakt mit der intrafusalen Faser. Das terminale Axon scheint sieh nach Eintritt unter die Basalmembran der intrafusalen Faser mehrfach in relativ dünne Äste unregelmäßigen Verlaufs zu teilen. Diese Form der Endigung könnte ein Korrelat der sog. flower spray-Endigung im Sinne Ruffinis (1898) darstellen.Die beiden morphologisch unterschiedlichen Endformationen innerhalb der sekundären Endigung gleichen einander und den Endformationen der primären Endigung bezüglich folgender Ultrastrukturmerkmale: 1. Es besteht synaptischer Kontakt zwischen Axon und intrafusaler Muskelfaser (synaptischer Spalt durchschnittlich 200 Å) ohne Zwischenlagerung von Basalmembranmaterial; 2. die terminalen Axonabschnitte liegen direkt unter der Basalmembran der intrafusalen Muskelfaser und sind nicht von Schwannschen Zellen bedeckt; 3. Mangel an synaptischen Bläschen; 4. desmosomenartige Verhaftungen zwischen Zellmembranen von Axon und intrafusaler Faser; 5. dyadenartige Anlagerungen des sarkoplasmatischen Retikulums an die Zellmembran der intrafusalen Faser im Bereich des synaptischen Spaltes.Nach unseren derzeitigen Vorstellungen sprechen diese Ultrastrukturmerkmale für eine rezeptorische Natur der beschriebenen Nervenendigungen.
Two electron-microscopically different forms of secondary sensory endings in a muscle spindle of the rat
Summary In a rat muscle spindle transversally cut into a series of alternating semithin and ultrathin sections, two different forms of nerve terminations were found on two neighbouring nuclear chain fibres. The two nerve terminations were connected to the same group II nerve fibre and are consequently constituents of one particular secondary sensory ending.The nerve termination on one of the two nuclear chain fibres is of the anulo-spiral type.The nerve termination on the second nuclear chain fibre shows a number of axons lying closely together between plasma membrane and basement membrane of the intrafusal muscle fibre. Not all of these axons are in direct contact with the intrafusal fibre. The terminating nerve fibre seems to be divided into several branches of rather small diameters and irregular courses. It is suggested that this kind of termination could be a correlate of the so-called flower spray type of sensory endings in muscle spindles.The two morphologically different nerve terminations in the secondary ending have the following ultrastructural characteristics in common with those of the primary ending: 1) Synaptic contact between axon and intrafusal muscle fibre (synaptic gap about 200 Å) without interposition of basement membrane material; 2) terminal axons located beneath the basement membrane layer of intrafusal muscle fibres without covering by Schwann cells; 3) lack of synaptic vesicles; 4) desmosome-like structures between plasma membranes of axon and intrafusal muscle fibre, and 5) dyads of the sarcoplasmic reticulum adjacent to the synaptic cleft. According to present knowledge these features indicate that all of these endings are sensory ones.

Summary Endings of four skeletofusimotor axons in a spindle of the cat tenuissimus muscle were examined in semithin (1-m thick) and ultrathin transverse serial sections. Two (dynamic) axons terminated on the nuclear bag1 intrafusal muscle fiber and on extrafusal fibers of the dark type. Two (static) axons terminated on the nuclear chain intrafusal fibers and extrafusal fibers of the intermediate type. The degree of indentation of axon terminals into the muscle surface, thickness of the sole plate and extent of folding of subjunctional membranes differed among intrafusal and extrafusal terminations of the same axon. Endings of axons on the bag1 and chain fibers were also morphologically dissimilar. Motor axons may not determine ending morphology. Rather the form and structure of a bag1 or chain ending may be determined by the type of intrafusal fiber on which the ending lies and the ending's distance from the primary sensory axon.  相似文献

Summary An antiserum to the sequence Arg-Phe-amide (RFamide) was used to stain the nervous systems of various physonectid siphonophores. In the stem of Nanomia bijuga, this antiserum stained an ectodermal nerve net, which was interrupted, at regular intervals, by transverse collars of neurons. Injection of Lucifer yellow into the giant axon of the stem showed that this axon was dye-coupled to an ectodermal nerve net that resembled the RFamide-positive network. Ectodermal nets of neurons were also found in the pneumatophore, gastrozooids, tentacles and tentilla. At the junctions of the pneumatophore, the gastrozooids, the dactylozooids and the gonozooids with the stem, and at the junctions of tentacles and tentilla, collars or rings of neurons occurred. The stem was connected to the phyllozooids and nectophores by muscular lamellae, which were bordered by chains of neurons. At the margin of the nectophores, an immunoreactive nerve ring was found. Connected to this ring and located in theseitliche Zapfen (sidely-located patche), were two agglomerations of nerve cells. On the upper side of the bell margin, positioned at 90° relative to the seitliche Zapfen, a delta-shaped neuronal structure was found. This structure was connected to the nerve ring and was associated with a muscle, which ran a short distance along the exumbrellar surface.The nervous systems of Agalma elegans, Forskalia edwardsi, Forskalia leuckarti and Halistemma rubrum resembled that of Nanomia bijuga in all major respects.  相似文献

Summary Silver impregnation of nerves, histochemical reactions for acetylcholinesterase, and electron microscopy reveal an efferent innervation of the notochord in amphioxus. Extensions of the notochordal lamellae end in groups (the notochordal horns) just below the ventro-lateral surface of the spinal cord where they are opposed to large nerve terminals originating as short collaterals of axons running longitudinally in the nerve cord. This neurochordal junction resembles an ordinary neuro-muscular junction in several respects and is interpreted as a part of the muscular system found in the notochord itself. Acknowledgement. Summary Steroid producing cells in human tissues can be demonstrated histochemically by the use of isopropanol as substrate. The Leydig cells of the testis, the theca cells of the ovary and the zona reticularis of the adrenal are stained by this method. A possible relation of this 'secondary alcohol dehydrogenase with the enzyme that catalyses the oxidative cleavage of the cholesterol side-chain is discussed.  相似文献

A mixture of galactoglucomannan-derived oligosaccharides (GGMOs), degree of polymerization 4–8, (1.2 M and 12 M) stimulated the viability of spruce [Picea abies (L.) Karst] embryos predominately on media supplemented with indole-3-acetic acid: zeatin (0.011, 10.01 mg · 1-1), at pH 5.O. Their effects on the development and morphogenesis of embryos were dependent on the culture conditions used. These GGMOs also improved the viability of spruce protoplasts when applied at the same concentrations in combination with 1-naphthaleneacetic acid, and to a lesser extent with 2,4-dichlorophenoxyacetic acid at pH 3.8. We analyzed the anatomy of two diffuse neurohemal systems for serotonin in the head of the Colorado potato beetle Leptinotarsa decemlineata by means of immunohistochemistry. One system is formed by axons from two bilateral pairs of neurons in the frontal margin of the suboesophageal ganglion that enter the ipsilateral mandibular nerve, emerge from this nerve at some distance from the suboesophageal ganglion, and cover all branches of the mandibular nerve with a dense plexus of immunoreactive axon swellings. The other system is formed by axons from two large neurons in the frontal ganglion that enter the ipsilateral frontal connectives, emerge from these connectives, and form a network of axon swellings on the labroforntal, pharyngeal, and antennal nerves and on the surface of the frontal ganglion. Immunohistochemical electron microscopy demonstrated that the axon swellings are located outside the neural sheaths of the nerves and hence in close contact with the hemolymph. We therefore suggest that these plexuses represent extensive neurohemal systems for serotonin. Most immunoreactive terminals are in direct contact with the hemolymph, and other terminals are closely associated with the muscles of the mandibles, labrum, and anterior pharynx, as well as with the salivary glands, indicating that these organs are under serotoninergic control.  相似文献

Summary Nerve endings in the extraocular muscles of the rat were submitted to histochemical tests for formalin-induced fluorescence and carboxylic esterases. Acetylthiocholine, butyrylthiocholine and -naphthyl acetate were used as substrates and iso-OMPA, 284C51, eserine and E-600 as inhibitors. The ultrastructure of the endings was studied with the electron microscope.Both single and multiple nerve terminals were observed in all six extraocular muscles. The single terminals of myelinated axons were comparable in their light and electron microscopic structure with the typical motor end plates of other striated muscles, and like these they exhibit acetylcholinesterase (AChE), non-specific cholinesterase (ns. ChE) and non-specific esterase (ns. E) activity. These endings were apposed to twitch-type muscle fibres.The multiple terminals were classified with the light microscope into two types. The larger type was 1/3 of the size of the motor end plate; 2–5 endings innervated the same muscle fibre; subneural infoldings were weakly developed and possessed only slight AChE and ns. ChE and probably no ns. E activity. No subneural lamellae were visible under the light microscope in the smaller type, which also possessed AChE and ns. ChE and was composed of 10–20 small dots dispersed along a single muscle fibre. The Schwann cells along nerve fibres leading to these two types of multiple endings exhibited ns. ChE but not AChE and ns. E activity.The ultrastructure of the two types of multiple endings was principally similar. The main difference, compared with the motor end plate, was that these endings were derived from unmyelinated axons which either make synaptic contacts along their course with the muscle fibre at variable distances (smaller-type) or these terminals were grouped closely together (larger-type).A few dense-core vesicles were observed in these unmyelinated nerves and in their terminals which were considerably smaller than those in the motor end plate. They were not always separated from each other by sarcoplasm and teloglia (larger-type) and contained also empty vesicles. The secondary synaptic clefts were often sparse and irregular or even absent, but the typical myoneural postsynaptic electron density was always observed. These multiple endings, in contrast to the motor end plate, were apposed only to muscle fibres with slow contraction.No catecholamine containing nerve endings were observed in the extraocular muscles.
