Patterned spontaneous activity in the developing retina is necessary to drive synaptic refinement in the lateral geniculate nucleus (LGN). Using perforated patch recordings from neurons in LGN slices during the period of eye segregation, we examine how such burst-based activity can instruct this refinement. Retinogeniculate synapses have a novel learning rule that depends on the latencies between pre- and postsynaptic bursts on the order of one second: coincident bursts produce long-lasting synaptic enhancement, whereas non-overlapping bursts produce mild synaptic weakening. It is consistent with “Hebbian” development thought to exist at this synapse, and we demonstrate computationally that such a rule can robustly use retinal waves to drive eye segregation and retinotopic refinement. Thus, by measuring plasticity induced by natural activity patterns, synaptic learning rules can be linked directly to their larger role in instructing the patterning of neural connectivity.  相似文献

Dephosphorylation of the natriuretic peptide receptor-A (NPR-A) is hypothesized to mediate its desensitization in response to atrial natriuretic peptide (ANP) binding. Recently, we identified six phosphorylation sites within the kinase homology domain of NPR-A and determined that the conversion of these residues to alanine abolished the ability of the receptor to be phosphorylated or to be activated by ANP and ATP. In an attempt to generate a form of NPR-A that mimics a fully phosphorylated receptor but that is resistant to dephosphorylation, we engineered a receptor variant (NPR-A-6E) containing glutamate substitutions at all six phosphorylation sites. Consistent with the known ability of negatively charged glutamate residues to substitute functionally, in some cases, for phosphorylated residues, we found that NPR-A-6E was activated 10-fold by ANP and ATP. As determined by guanylyl cyclase assays, the hormone-stimulated activity of the wild-type receptor declined over time in membrane preparations in vitro, and this loss was blocked by the serine/threonine protein phosphatase inhibitor microcystin. In contrast, the activity of NPR-A-6E was more linear with time and was unaffected by microcystin. The nonhydrolyzable ATP analogue adenosine 5'-(beta,gamma-imino)-triphosphate was half as effective as ATP in stimulating the wild-type receptor but was equally as potent in stimulating NPR-A-6E, suggesting that ATP is required to keep the wild-type but not 6E variant phosphorylated. Finally, the desensitization of NPR-A-6E in whole cells was markedly blunted compared with that of the wild-type receptor, consistent with its inability to shed the negative charge from its kinase homology domain via dephosphorylation. These data provide the first direct test of the requirement for dephosphorylation in guanylyl cyclase desensitization and they indicate that it is an essential component of this process.  相似文献

BackgroundCommunity health workers (CHWs) can supplement professional medical providers, especially in rural settings where resources are particularly scarce. Yet, outcomes of studies evaluating CHWs effectiveness have been highly variable and lack impact when scaled nationally. This study examines if child and maternal outcomes are better when existing government CHWs, who are perinatal home visitors, receive ongoing enhanced supervision and monitoring, compared to standard care.Methods and findingsA cluster randomized controlled effectiveness trial was conducted comparing outcomes over 2 years when different supervision and support are provided. Primary health clinics were randomized by clinic to receive monitoring and supervision from either (1) existing supervisors (Standard Care (SC); n = 4 clinics, 23 CHWs, 392 mothers); or (2) supervisors from a nongovernmental organization that provided enhanced monitoring and supervision (Accountable Care [AC]; n = 4 clinic areas, 20 CHWs, 423 mothers). Assessments were conducted during pregnancy and at 3, 6, 15, and 24 months post-birth with high retention rates (76% to 86%). The primary outcome was the number of statistically significant intervention effects among 13 outcomes of interest; this approach allowed us to evaluate the intervention holistically while accounting for correlation among the 13 outcomes and considering multiple comparisons.The observed benefits were not statistically significant and did not show the AC’s efficacy over the SC. Only the antiretroviral (ARV) adherence effect met the significance threshold established a priori (SC mean 2.3, AC mean 2.9, p < 0.025; 95% CI = [0.157, 1.576]). However, for 11 of the 13 outcomes, we observed an improvement in the AC compared to the SC. While the observed outcomes were not statistically significant, benefits were observed for 4 outcomes: increasing breastfeeding for 6 months, reducing malnutrition, increasing ARV adherence, and improving developmental milestones. The major study limitation was utilizing existing CHWs and being limited to a sample of 8 clinics. There were no major study-related adverse events.ConclusionsSupervision and monitoring were insufficient to improve CHWs’ impact on maternal and child outcomes. Alternative strategies for staff recruitment and narrowing the intervention outcomes to the specific local community problems are needed for consistently high impact.Trial registrationClinicaltrials.gov, NCT02957799.

Muscles are required for animal movement, feeding, heartbeat, and reproduction. Disruption of muscle function can lead to mobility impairments and diseases like muscular dystrophy and cardiac myopathy; therefore, research in this area has significant implications for public health. Recent work by Vaziri and colleagues has taken genetic, cell biological, and biochemical approaches to identify Protein kinase C-d (Pkcδ) as a novel regulator of the essential myosin light chain 2 (MLC2) by phosphorylation. The authors determine which residues of MLC2 are modified by Pkcδ and show that phosphorylation by Pkcδ is required for proper sarcomere assembly and function. This study underscores the importance of Drosophila melanogaster as a model system for muscle function and highlights how protein phosphorylation is a vital part of post-translational gene regulation.  相似文献

The 20th IUPAB Congress took place online, together with the annual meetings of the Brazilian Biophysical Society and the Brazilian Society for Biochemistry and Molecular Biology, from the 4th to the 8th of October, 2021. The ten keynote lectures, 24 symposia, two poster sessions, and a series of technical seminars covered the full diversity of current biophysical research and its interfaces with other fields. The event had over 1000 attendees, with an excellent gender balance. Although the Americas dominated, there were also significant numbers of participants from Europe, Asia, and Africa.

The International Union of Pure and Applied Biophysics (IUPAB) came into existence in Stockholm in 1961 and has been a member of the International Science Council since 1966 (Solomon 1968). Its overall objectives aim to foster international collaboration in all aspects of biophysics and related areas and to catalyze the advancement of basic biophysical research as well as its many applications. Although IUPAB is active on many fronts, undeniably one of its showcase events is the IUPAB Congress, traditionally organized every three years in different locations worldwide. In 2021, the event was organized and run from Brazil, albeit for the very first time in a virtual format due to travel restrictions imposed by the COVID-19 pandemic. On this occasion, the Congress was organized in conjunction with the annual meetings of both the Brazilian Biophysical Society (SBBf, in its 45th edition) and the Brazilian Society for Biochemistry and Molecular Biology (SBBq, in its 50th edition). Even with the united forces of these well-established local societies, it turned out to be a bumpy ride to bring the event to fruition.Plans for the 20th Congress began in 2016, almost immediately after the decision to hold the event in Brazil, a cause championed by the then-president of the Brazilian Biophysical Society, Marcelo Morales. The original plans had the meeting to be held in the Cidade Maravilhosa (The Wonderful City) of Rio de Janeiro in October 2020. However, it soon became apparent that the political and economic difficulties that the State of Rio was facing at the time meant that it would be wise to search for an alternative venue. The previous experience of SBBq in organizing similar events in the city of Foz do Iguaçu, on the borders with Argentina and Paraguay, made this an obvious choice. Furthermore, the natural attraction of the spectacular Iguaçu waterfalls seemed to be an ideal compensation for Sugar Loaf Mountain, Copacabana beach, and the statue of Christ the Redeemer on Corcovado Mountain.Then came the pandemic. By mid-2020, it had become apparent that there were too many unknowns to make it possible to proceed with an in-person event in October of that year. It was decided to postpone the congress to 2021 but with a firm belief that things would be “back to normal.” Sweet delusion! As 2020 turned into 2021 and the severity and longevity of the pandemic became clearer and clearer (not to mention the abysmal performance of the Brazilian government in failing to rise to the challenge), the inevitable decision was taken to transform the event into an “on-line” congress. This was a first for both the local organizers and the IUPAB.The move to an online format immediately had an impact on the organization of the Young Scientist Program. This was initially envisaged to be a combination of formal and informal activities aimed at uniting about 40 early carrier scientists and post-docs for a couple of days prior to the main event in a stimulating atmosphere conducive to networking. Skillfully conceived, organized, and executed by Eneida de Paula (Campinas) and Eduardo Reis (São Paulo), this too had to be adapted to a “virtual reality.” The successful solution turned out to be a series of fortnightly thematic webinars, including a talk from a recognized authority in the field followed by three or four short presentations from the participants themselves (Table (Table1).1). The standard was extremely high and the YSP ended up being a highly effective warm-up to the congress itself. Furthermore, there was excellent geographical diversity among the participants with Europe, Africa, Asia, the Middle East, and both North and South America represented.Table 1Young Scientist Webinar Program

Although the Americas dominated, there were also significant numbers of participants from Europe, Asia, and Africa (Fig. 1). Table Table22 gives an excellent idea of the diverse subject matter covered during the 5 days of the congress itself. As to be expected, the way in which biophysics naturally interfaces with biochemistry, molecular biology, cell biology, chemistry (including medicinal chemistry), physics, engineering, etc. was more than apparent. Nevertheless, several themes appeared to be particularly recurrent throughout the event. Notwithstanding the plethora of other topics, several main threads permeated the proceedings, and these included (1) lipids, membranes, their assembly, and dynamics; (2) bioimaging at all levels; (3) drug targets and drug development/delivery; and (4) molecular recognition including membrane/protein interactions. This special issue aims to cover the main topics of the event as comprehensively as possible in similar vein to previous efforts (Hall and dos Remedios, 2017). In over 50 articles, including reviews, commentaries, letters, and editorials, we aim to convey the full flavor of the congress. It is hoped that this will serve simultaneously as both a useful source of reference and a historical record. The short, focused review articles are all up-to-date and expected to be of particular value to a broad readership. The Nobel laureate Richard Henderson set the ball rolling with a beautifully clear historical overview of how cryo-EM got to be where it is now and what we might expect for the near future. Tony Watts (the new president-elect of IUPAB) closed the event with the Avanti/IUPAB award lecture and a clear message that biophysics is not all about proteins—lipids are important (also)! Midweek, a second Nobel prize winner, Michael Levitt, gave his take on the COVID-19 pandemic by applying his talent for mathematical modeling in much the same way as he so successfully applied it to macromolecular systems in the past. At the very least, his talk gave plenty of food for thought to those who were present.Table 3Keynote speakers

Open in a separate windowOverall, the sessions were very well attended with typically over 200 participants. The ease of moving from one session to another under the virtual format proved to be a notable advantage. Furthermore, since many of the talks were pre-recorded, most of the sessions kept to time rather better than is often the case at traditional events. The two poster sessions were also very well frequented, and the pre-recorded videos were generally of high quality. Approximately 10% of all poster presenters were awarded prizes during the closing ceremony, and six special prizes were generously provided by the Royal Society of Chemistry.Several special activities were held throughout the week. These included technical seminars by some of the sponsors, including Cytiva, Thermo-Fisher, and Sartorius as well as sessions devoted to Brazil-German exchange programs and one on “Gender in Science.” The latter was particularly motivational for the congress participants, whose demographic was heavily biased towards early-career scientists, post-docs, and students (Fig. 2). Biophysical Reviews organized two early-morning sessions, one of which was an editorial board meeting whilst the other was open to all interested parties and represented an opportunity to promote the journal within the community. The IUPAB held its general assembly on the 6th of October. Manuel Prieto formally took over as President with Marcelo Morales stepping down but continuing as a council member in the role of immediate Past President. BackgroundThe epidemic of Zika virus (ZIKV) was associated with a sudden and unprecedented increase in infants born with microcephaly. Colombia was the second most affected country by the epidemic in the Americas. Primary caregivers of children with ZIKV-associated microcephaly, their mothers mainly, were at higher risk of suffering anxiety and depression. Often, these women were stigmatized and abandoned by their partners, relatives, and communities.Methodology/Principal findingsThis study aimed to understand the perceptions about ZIKV infection among mothers of children born with microcephaly during the ZIKV epidemic in Caribbean Colombia, and the barriers and facilitators affecting child health follow-up. An exploratory qualitative study, based on Phenomenology and Grounded Theory, was conducted in Caribbean Colombia. Data were collected through In-Depth Interviews (IDI) from women who delivered a baby with microcephaly during the ZIKV epidemic at Clínica Salud Social, Sincelejo, Sucre District (N = 11). The themes that emerged during the interviews included experiences from their lives before pregnancy; knowledge about ZIKV; experiences and perceptions when diagnosed; considering a possible termination of pregnancy, and children’s clinical follow-up. In some cases, women reported having been told they were having a baby with microcephaly but decided not to terminate the pregnancy; while in other cases, women found out about their newborn’s microcephaly condition only at birth. The main barriers encountered by participants during children’s follow-up included the lack of psychosocial and economic support, the stigmatization and abandonment by some partners and relatives, and the frustration of seeing the impaired development of their children.ConclusionsThis study contributed to identifying the social, medical, psychological, and economic needs of families with children affected by the ZIKV epidemic. Commitment and action by local and national governments, and international bodies, is required to ensure sustained and quality health services by affected children and their families.  相似文献

BackgroundAlthough vector-borne zoonotic diseases are a major public health threat globally, they are usually neglected, especially among resource-constrained countries, including those in sub-Saharan Africa. This scoping review examined the current knowledge and identified research gaps of vector-borne zoonotic pathogens in Zambia.Methods and findingsMajor scientific databases (Web of Science, PubMed, Scopus, Google Scholar, CABI, Scientific Information Database (SID)) were searched for articles describing vector-borne (mosquitoes, ticks, fleas and tsetse flies) zoonotic pathogens in Zambia. Several mosquito-borne arboviruses have been reported including Yellow fever, Ntaya, Mayaro, Dengue, Zika, West Nile, Chikungunya, Sindbis, and Rift Valley fever viruses. Flea-borne zoonotic pathogens reported include Yersinia pestis and Rickettsia felis. Trypanosoma sp. was the only tsetse fly-borne pathogen identified. Further, tick-borne zoonotic pathogens reported included Crimean-Congo Haemorrhagic fever virus, Rickettsia sp., Anaplasma sp., Ehrlichia sp., Borrelia sp., and Coxiella burnetii.ConclusionsThis study revealed the presence of many vector-borne zoonotic pathogens circulating in vectors and animals in Zambia. Though reports of human clinical cases were limited, several serological studies provided considerable evidence of zoonotic transmission of vector-borne pathogens in humans. However, the disease burden in humans attributable to vector-borne zoonotic infections could not be ascertained from the available reports and this precludes the formulation of national policies that could help in the control and mitigation of the impact of these diseases in Zambia. Therefore, there is an urgent need to scale-up “One Health” research in emerging and re-emerging infectious diseases to enable the country to prepare for future epidemics, including pandemics.  相似文献
