Germination responses of Diplotaxis harra to temperature and salinity   总被引:1,自引:1,他引:0  
Diplotaxis harra (Forssk.) Boiss, an annual herb in the family of Brassicaceae, is widely distributed in many sandy and gypseous areas in southern Tunisia. Laboratory experiments were carried out to assess the effects of temperature and salinity on seed germination and recovery responses after seed transfer to distilled water. The germination responses of the seeds in complete darkness were determined over a wide range of temperatures (5, 10, 15, 20, 25 and 30 °C) and salinities (0, 50, 100, 150 and 200 mM NaCl). Germination was inhibited by either an increase or decrease in temperature from the optimal temperature (15 °C). Highest germination percentages were obtained under non-saline conditions and an increase in NaCl concentrations progressively inhibited seed germination. Rate of germination decreased with an increase in salinity at all temperatures but comparatively higher rates were obtained at 15 °C. Salt stress decreased both the percentage and the rate of germination. An interaction between salinity and temperature yielded no germination at 200 mM NaCl. Seeds were transferred from salt solution to distilled water after 20 days, and those from low salinities recovered at all temperatures. At NaCl concentration of 200 mM, the recovery of germination was completely inhibited.  相似文献

The effect of different types and concentrations of dormancy regulating chemicals (DRCs) on innate and induced dormancy was evaluated under optimal germination conditions in the invasive Prosopis juliflora shrub. Lower concentrations of gibberellic acid (0.3 mM) and kinetin (0.05 mM) were more effective in enhancing germination % and rate at higher concentrations of NaCl, but the reverse was true for thiourea. None of the DRCs alleviated innate dormancy of P. juliflora. Germination % and rate decreased as salinity increased. Percent final germination of non-treated seeds was significantly reduced at 500 mM NaCl and virtually inhibited in 600 mM NaCl. Germination reduction in 500 mM NaCl was not alleviated by any of the DRCs, but inhibition induced at 600 mM NaCl was partially alleviated by all the DRCs. Gibberellic acid had a significantly greater effect than kinetin in alleviating germination inhibition. For restoration of saline soil through the use of P. juliflora, results suggest using DRCs, particularly gibberellic acid and thiourea, as a preseeding treatment can overcome the problem of reduced germination.  相似文献

Triglochin maritimum (Juncaginaceae). Germination tests were carried out at three salinity levels (0,200,400 mM NaCl in which seeds were exposed to increasing- or decreasing temperatures. Effects of moist-chilling pretreatment (stratification) in 0,200 and 400 mM NaCl on seed germination at 0,200 and 400 mM NaCl, respectively, were also examined. Under the highest salinity condition (400 mM NaCl), no germination was observed. The seeds germinated very well (88%) in fresh-water after 5-month moist-chilling pretreatment. Longer moist-chilling pretreatment resulted in higher germination percentages. Moist chilling pretreatment in 0,200 and 400 mM NaCl for 5 month enhanced germination percentages to 10% (in 400 mM NaCl) –88% (in 0 mM NaCl). Seed germination and seedling growth of the annual halophyte species Suaeda japonica Makino were investigated in response to variable salinity of sediment pore water. The germination percentage of S. japonica’s soft brown seeds, which are dominant among dimorphic seeds, decreased with an increase in salinity, although germination was still observed at 1200‐mM NaCl concentration. The germination percentage and germination speed observed in April were higher than those observed in December when treated with sediment water with 400–1200 mM of NaCl concentrations. These data suggest that S. japonica seedlings could be established on sediments that experience high temperatures. Germination recovery of S. japonica seeds transferred from 600‐mM NaCl containing sediment (seawater equivalent) was lowest among 0–1200‐mM NaCl treatments, implying the low tolerance of seawater conditions of S. japonica seeds. Seeds germinated in 900‐ to 1200‐mM NaCl medium showed poor growth, but survived, in hypersaline conditions, and exhibited improvement in growth upon transfer to lower salinity.  相似文献

All seeds of Aeluropus lagopoides and Urochondra setulosa germinated under non-saline conditions except for Sporobolus ioclados which showed only 40 % germination. Increase in salinity substantially inhibited germination and few seeds germinated at 400 mM NaCl. Germination at 200 mM NaCl was alleviated in U. setulosa by the application of gibberellic acid and fusicoccin, in A. lagopoides by thiourea, betaine, kinetin, fusicoccin and ethephon, and in S. ioclados by gibberellin and ethephon. High salinity (400 mM NaCl) induced germination inhibition was alleviated by proline, kinetin, fusicoccin and ethephon only in A. lagopoides.  相似文献

Salsola ferganica L. (Chenopodianceae) is an annual halophytic species. Experiments were carried out in laboratory to determine the effects of temperature, perianths and various types of salinity on seed germination and germination recovery. Seeds were germinated at 6 levels of temperature with perianths, plus perianths and removed perianths in complete darkness for 9 days. The germination responses of the seeds without perianths at 25 °C were determined over a wide range of NaCl, NaHCO3 or NaCl–NaHCO3 mixed stress for 13 days. Perianths seriously affected germination as a barrier for seed germination and the optimal temperature was at 25 °C. Highest germination percentage was obtained under control and seed germination was progressively inhibited with the increase of salinity concentration. The negative effect of NaHCO3 at the same concentration on germination was stronger than that of NaCl and NaCl–NaHCO3 mixed. When substrate salinity was removed, seeds exposed to a high NaCl concentration (400–800 mM), NaHCO3 (50–200 mM) and NaCl–NaHCO3 mixed (100–400 mM) germinated well. Final germination of Salsola ferganica seeds was significantly affected by types of salt at the low salinity (?200 mM) and with increased salinity it was influenced mainly by salinity concentration for various proportion of salt–alkali mixed stress.  相似文献

Salvia aegyptiaca is a xerophytic perennial herb belongs to the Lamiaceae family commonly used for medicinal purposes. Laboratory experiments were carried out to assess the effects of temperature and salinity on seed germination and recovery responses after transferring to distilled water. Temperatures between 10 and 40 °C seem to be favourable for the germination of this species. Germination was inhibited by either an increase or decrease in temperature from the optimum (30 °C). The highest germination percentages were obtained at 0 mM NaCl; however, the increase of solution osmolalities progressively inhibited seed germination. The germination rate decreased with an increase in salinity for most of tested temperatures, but comparatively higher rates were obtained at 30 °C. Salt stress decreased both the percentage and the rate of germination. An interaction between salinity and temperature yielded no germination at 300 mM NaCl. By experimental transfer to distilled water, S. aegyptiaca seeds that were exposed to moderately saline conditions recovered and keep their ability to germinate mostly at low temperatures. At 300 mM NaCl, germination recovery decreased with increasing temperature and it was completely inhibited at 40 °C.  相似文献

Algal extracts provide a safe regime for enhancing crop productivity under stressful conditions. The present study evaluated the efficiency of aqueous and ethanolic extracts of the brown alga Dictyota dichotoma in alleviation of salt stress on germination of rice seeds. Firstly, seeds were germinated using the aqueous extract of D. dichotoma at concentrations of 0, 5, 10, 20, and 50 g L−1, prepared either at room temperature (RTE) or by boiling (BLE). The % germination of rice increased from 84% in non-treated seeds to 100% when treated with 20 g L−1 BLE, although this treatment caused reduced uniformity of germination. Embryo growth was maximum at 20 g L−1 of both extracts with superiority of BLE over RTE. In the second experiment, the effect of 20 g L−1 aqueous and ethanolic extracts relative to a balanced nutrient supply on germination of seeds treated with 0, 40, 90, and 170 mM NaCl was investigated. Salinity reduced % germination with delayed onset but high uniformity of germination, whereas algal amendments counterbalanced the effect of salinity to a greater extent relative to the nutrient supply. Upon withdrawal of salt stress, seeds promptly recovered, with more efficient recovery of seeds exposed to 170 mM than from 90 mM NaCl. The lower recovery of salt-treated seeds compared with the control seed germination suggests that rice suffered from the toxic ion effect of salinity on embryo rather than from the osmotic effect. Extracts of D. dichotoma can enhance and also alleviate salinity stress on rice seed germination.

A study was conducted to determine germination response to temperature and salinity and seedling response to salinity by three height forms of the salt marsh grass Spartina alterniflora Loisel. Germination tests showed that seeds cannot withstand drying at moderate temperature, as viability was lost within 40 days in seeds stored dry at 72 F. Cold storage at 43 F is adequate to prevent desiccation up to 40 days, but after 8 months viability is lost. Viability is retained at least 8 months when seeds are stored in sea water at 43 F. Germination response was good in a 65–95 F alternating diurnal thermoperiod but was poor in a 72 F constant thermoperiod. Germination response to salinity was an inverse curvilinear relationship with germination inhibition at high salinities apparently due to osmotic effects. The maximum tolerance limit for germination lies between 6 and 8 % NaCl. Seeds from short, medium, and tall plants responded similarly in storage and temperature studies. However, in salinity studies, seeds of the Ocracoke Island short form and the Oak Island tall form performed best. A logarithmic curve best described seedling growth response to various NaCl levels. Growth response as measured by seedling dry weight was best in 0.5 % NaCl solution. Seedlings grew taller in both 0.5 and 1.0 % NaCl than in 0 % NaCl. No significant difference in seedling growth response due to height form of the parent plant was detected. Thus, on the basis of germination and seedling responses, the height forms of S. alterniflora in North Carolina salt marshes are best described as ecophenes.  相似文献

The action of phytohormone producing bacteria and plant growth regulators on germination and seedling growth of wheat under saline conditions were studied. Seed dormancy enforced by salinity (100 mM NaCl) was substantially alleviated and the germination was promoted by gibberellin, auxin, zeatin, and ethephon from 54 to 97%. The IAA producing bacterial strains Pseudomonas aureantiaca TSAU22, Pseudomonas extremorientalis TSAU6 and Pseudomonas extremorientalis TSAU20 significantly increased seedling root growth up to 25% in non-salinated conditions and up to 52% at 100 mM NaCl, compared to control plants. It is concluded that growth regulators considerably alleviated salinity-induced dormancy of wheat seeds. The facts mentioned above make it possible to recommend root colonizing bacteria that produce phytohormone to alleviate salt stress of wheat grown under conditions of soil salinity.  相似文献

The present study demonstrates the regeneration of plantlets of guava (Psidium guajava L.) from somatic embryos developed under salt-stress conditions. With increasing concentrations of NaCl in induction medium (MS + 4.52 μM 2,4-d + 5% sucrose) from 0 to 200 mM, the number of somatic embryos per responsive explant decreased. Somatic embryos induced on 0–100 mM NaCl containing medium developed into torpedo stages, whereas, the development of somatic embryos that differentiated on 150 and 200 mM NaCl-supplemented medium was arrested prior to torpedo stage and did not undergo maturation phase. Somatic embryos that developed on NaCl-containing medium, showed better germination in the presence of NaCl as compared with those developed on medium without NaCl. The effect of increasing salt-stress was also investigated on plant growth, chlorophyll and carotenoids, Na+ and K+, and proline and glycine betaine accumulation in in vitro grown plantlets. The level of Na+ in leaves increased with increasing concentrations of NaCl in the medium. Accumulation of free proline and glycine betaine in leaves significantly increased with increasing salinity. The results suggest that accumulation of proline and glycine betaine may be important for osmotic adjustment in guava under salinity stress.  相似文献

Nitraria retusa and Atriplex halimus (xero-halophytes) plants were grown in the range 0–800 mM NaCl while Medicago arborea (glycophyte) in 0–300 mM NaCl. Plants were harvested after 120 days of salt-treatment. The present study was designed to study the effect of salinity on root, stem and leaf anatomy, water relationship, and plant growth in greenhouse conditions. Salinity induced anatomical changes in the roots, stems and leaves. The cuticle and epidermis of N. retusa and A. halimus stems were unaffected by salinity. However, root anatomical parameters (root cross section area, cortex thickness and stele to root area ratio), and stem anatomical parameters (stem cross section area and cortex area) were promoted at 100–200 mM NaCl. Indicating that low to moderate salinity had a stimulating effect on root and stem growth of these xero-halophytic species. At higher salinities, root and stem structures were altered significantly, and their percentages of reduction were higher in A. halimus than in N. retusa whereas, in M. arborea, they were strongly altered as salinity rose. NaCl (100–300 mM) reduced leaf water content by 21.2–56.2% and specific leaf area by 51–88.1%, while increased leaf anatomical parameters in M. arborea (e.g. increased thickness of upper and lower epidermis, palisade and spongy mesophyll, entire lamina, and increased palisade to spongy mesophyll ratio). Similar results were evidenced in A. halimus leaves with salinity exceeding 100 mM NaCl. Leaves of N. retusa were thinner in salt-stressed plants while epidermis thickness and water content was unaffected by salinity. The size of xylem vessel was unchanged under salinity in the leaf’s main vein of the three species while we have increased number in M. arborea leaf main vein in the range of 200–300 mM NaCl. A longer distance between leaf vascular bundle, a reduced size and increased number of xylem vessel especially in stem than in root vascular system was evidenced in M. arborea treated plants and only at (400–800 mM) in the xero-halophytic species. The effects of NaCl toxicity on leaf, stem and root ultrastructure are discussed in relation to the degree of salt resistance of these three species. Our results suggest that both N. retusa and A. halimus show high tolerance to salinity while M. arborea was considered as a salt tolerant species.  相似文献

Seeds of four lettuce (Lactuca sativa L.) varieties (Romaine, Augusta, Vista and Verte) differing in their salt sensitivity were sown at 0 (Control), 50, 100 and 150 mM NaCl. The final germination percentage decreased with the increasing salinity and was annulated at the highest salt concentration in Vista and Verte, the most sensitive varieties. However, in the less sensitive ones, Romaine and Augusta, it was slightly modified at 50 and 100 mM NaCl and then decreased by 50% compared with the control, at 150 mM. The effects of NaCl 100 mM on seedling growth, phytase activities, phytate and inorganic phosphorus contents were studied in Romaine and Vista showing different behaviours towards salinity. Radicle and hypocotyl length and fresh and dry weights were reduced by salt treatment in both varieties. In addition, radicle phytase activity exhibited an increase in Romaine (less sensitive) and a decrease in Vista (more sensitive). In hypocotyl, this activity showed no difference with the control in the two varieties. However, in cotyledons, and during early hours after germination, salinity decreased phytase activity in both varieties whereas in the later hours (72–96 h) this activity reached the value of the control in Romaine. The enhancement of phytase activity was concomitant with an increase in orthophosphate content and a decrease in phytate reserve. These results suggest that salt presence in the medium delays Pi remobilization from phytate stock, but stimulates assimilation of phosphorus more than its accumulation in the organs of the two lettuce varieties.  相似文献

Effects of salinity, temperature and their interactions on the rate and final percentage of germination were evaluated for two populations (Msarref, Oued dkouk) of the invasive glycophyte Lotus creticus Linné, grown under arid environmental conditions of the Tunisia. Seeds that were not treated with NaCl germinated well in a wide range of temperatures. For both populations, maximum germination occurred in distilled water at 25°C and lowest germination for all salinities was at 35°C. Germination was substantially delayed and significantly reduced with an increase in NaCl to levels above 300 mm . Compared to the Oued dkouk population, final germination and germination rate of the Msarref population was completely inhibited at 300 mm NaCl. The interactive effect of temperature and NaCl concentration on final germination and germination rate was significant (P < 0.01), indicating that the germination response to salinity depended on temperature. The inhibition of Oued dkouk population seed germination at high salt concentration was mostly due to osmotic effects while ionic effects were noted at Msarref population. The germination behaviour of the Oued dkouk population would therefore imply adaptive mechanisms to saline environments, while in the Msarref population such mechanisms seem to be absent. Since seed germination is more sensitive to salinity stress than the growth of established plants, the greater tolerance to salinity of Oued dkouk population would be an adaptive feature of this population to saline environment.  相似文献

The stem succulent perennial halophyteHaloxylon recurvum Bungeex. Boiss. grows and produces seed under highly saline conditionsand seeds are deposited in saline soils. Experiments were conductedto determine the effect of salinity and temperature on the germinationof seeds. Results indicate that seeds can germinate at veryhigh salt concentrations (500 mM). However, highest germinationpercentages were obtained in distilled water. Physiological and molecular mechanisms of adaptation to abiotic stresses of grass pea (Lathyrus sativus L.) are still poorly understood. Responses of four genotypes of grass pea to salinity stress in tissue culture conditions were investigated at early seedling growth stages. Salinity stress was induced in the agar media by adding 0, 50, 100 and 200 mM of NaCl. Germination and seedling emergence percentage was not significantly affected by 50 and 100 mM of NaCl. However, NaCl in 200 mM concentration lowered level of these parameters. Generally, exposure to NaCl stress significantly reduced length of grass pea seedling organs (root and shoot) but did not influence the content of dry weight in shoots and increased it in the roots in two cases. Increasing salt concentration decreased integrity of cellular membranes both in root and shoot tissues. Higher accumulation of phenolic compounds and significant changes in activity of antioxidant enzymes (peroxidase and catalase) were observed in the roots but not in the shoots. Similarly, the content of proline increased mostly in the roots from moderate (100 mM) salinity conditions. Adverse conditions did not resulted in alterations in photosynthetic pigments content of any tested genotypes. The better performance of shoots than roots may result from in vitro conditions in which experiments were conducted.

The present work addresses the importance of antioxidant, redox and energetic parameters in regulating salt-tolerance in Sesuvium portulacastrum. Experiments were conducted on 45 days old plants subjected to 250 and 1,000 mM NaCl stress for 2–8 days. Plants showed no significant change in growth parameters (shoot length, dry weight, and water content) at 250 mM NaCl as compared to control. However, growth of plants was significantly affected at 1,000 mM NaCl. The differential growth behaviour could be attributed to a greater decline in the energetic parameters (in terms of ratios of NADP/NADPH and ATP/ADP) at 1,000 mM NaCl than at 250 mM NaCl. The osmotic stress imposed to plants at 250 mM NaCl was presumably balanced by the accumulation of sodium ions (Na+), an energetically favorable process, and did not require an increased synthesis of proline. In contrast, to counter osmotic stress at 1,000 mM NaCl, plants accumulated Na+ as well as proline and were, therefore, energetically stressed. Further, the response of enzymatic and molecular antioxidants at 1,000 mM was either close to or even lower than that at 250 mM, which resulted in oxidative damage at 1,000 mM, particularly on longer durations. In conclusion, it is suggested that altered redox and energetic status of the plants could play a key role in mediating the tolerance of Sesuvium under salinity stress.  相似文献

The antioxidative defense mechanism to salinity was assessed by monitoring the activities of some antioxidative enzymes and levels of antioxidants in an obligate halophyte, Salicornia brachiata, subjected to varying levels of NaCl (0, 200, 400, and 600 mM) under hydroponic culture. In the shoots of S. brachiata, salt treatment preferentially enhanced the activities of ascorbate peroxidase (APX), guaiacol peroxidase (POX), glutathione reductase (GR), and superoxide dismutase (SOD), whereas it induced the decrease of catalase (CAT) activity. Similarly, salinity caused an increase in total glutathione content (GSH + GSSG) and a decrease in total ascorbate content. Growth of S. brachiata was optimum at 200 mM NaCl and decreased with further increase in salinity. Salinity caused an increase in Na+ content and a decrease in K+ content of shoots. Proline levels did not change at low (0-200 mM NaCl) or moderate (400 mM NaCl) salinities, whereas a significant increase in proline level was observed at high salinity (600 mM NaCl). Accumulation of Na+ may have a certain role in osmotic homeostasis under low and moderate salinities in S. brachiata. Parameters of oxidative stress such as malondialdehyde (MDA), a product of lipid peroxidation, and H2O2 concentrations decreased at low salinity (200 mM NaCl) and increased at moderate (400 mM NaCl) and high salinities (600 mM NaCl). As a whole, our results suggest that the capacity to limit ionic and oxidative damage by the elevated levels of certain antioxidative enzymes and antioxidant molecules is important for salt tolerance of S. brachiata.  相似文献

Plants respond differently to salt stress depending on their genetic structure and the severity of the stress. Salinity reduces seed germination, delays plant emergence, and inhibits seedling growth. The selection of the tolerant genotypes, however, plays a vital role in increasing agricultural output since various genotypes greatly vary for their tolerance to salinity. Therefore, this study determined the impact of five different NaCl levels (i.e., 0, 50, 100, 150 and 200 mM) on seed germination and growth attributes of 10 flax (Linum usitatissimum L.) genotypes. The germination and growth characteristics of the genotypes under study were examined using the biplot approach at varied salt levels. The results indicated that individual and interactive effects of genotypes and salinity levels significantly (p ≤ 0.01 or p ≤ 0.05) affected several seed germination traits. The relations of genotype × germination traits indicated that ‘G4′ and ‘G6′ were the most stable genotypes with the highest performance regarding seed germination characteristics. The genotype ‘G2′ was associated with shoot length, while ‘G7′ was linked with salinity tolerance index. The biplot divided the germination characteristics into five different groups according to sector analysis. Most of the germination parameters had higher values under 100 mM, while some of the parameters had better values under 0, 50 and 200 mM NaCl levels. The tested genotypes varied for their seed germination and growth response depending on the NaCl levels. The genotypes ‘G4′, ‘G5′ and ‘G6′ proved more tolerant to high NaCl levels. Therefore, these genotypes can be used to improve flax productivity under saline soils.  相似文献

Sarcobatus vermiculatus (Hook) Torrey is a leaf succulent, sodium-accumulating shrub usually found in saline substrates of the Great Basin desert, Utah, USA. Laboratory experiments were conducted to determine the effect of salinity (0, 200, 400, 600, 800 and 1000 mM NaCl) and temperature (day/night: 5/15, 10/20, 15/25, 20/30, and 25/35°C) on seed germination. S. vermiculatus showed 100% germination in non-saline controls, at all thermoperiods. Percentage and rate of germination decreased with increases in salinity and few seeds germinated at even 1000 mM NaCl. High salinity exposure caused the loss of viability at higher temperature regimes, while some recovery was recorded in low salinity treatments.  相似文献
