反义药物以其合理设计药物的可能性和精确的特异性广泛吸引了人们的注意,但反义药物的研究并非如人们最初预想的那样简单。本文从其特异性,稳定性,透过靶细胞的能力,作用强度,活性判定,给药途径,安全性和毒性,生产成本等诸方面对反义药物的研究现状,现存问题进行了综述。相信伴随这些问题的解决,反义药物很可能成为药典的一部分,给疾病的治疗带来益处。 相似文献

Pulsed field gradient diffusion constant measurements were used to resolve the ambiguity in determining the conformational states of single-stranded DNA dodecanucleotides (d1s, d4s and d5s). For d1s and d5s, because of the spectral symmetry conventional NMR analyses cannot differentiate whether they are hairpins or homo-duplexes. However, the diffusion constants of these sequences at 300 K are 1.4 times greater than those of the comparison complementary duplexes. This result agrees well with what is expected for Dhairpin>/Dduplex based on classic liquid-phase translational diffusion models and the Einstein–Stokes equation, confirming that d1s and d5s form hairpins. d4s did not show a structured spectral pattern, but its diffusion constant measurement suggests that this sequence may not be a random coil. The DNA sequences studied contain chemically modified backbone linkages and are potential antisense agents for gene regulation. The knowledge of their diffusion constants, in combination with conventional NMR analysis and other biophysical spectroscopic measurements, provides new insights into the relationships of chemical structure and conformational preference of antisense oligonucleotides and their analogs. 相似文献

Approximately 48.2% of couples of 15 to 49 years of age practice family planning methods in India. Female sterilization accounts
for 34.2%, with male sterilization declining from 3·4% in 1992–93 to 1·9% in 1998–99. Use of the condom increased to 3·1%
from 2·4%. There is an urgent need for research to develop new contraceptive modalities especially for men and also for women
and to make existing methods more safe, affordable and acceptable. Current efforts in India to develop a male contraceptive
are mainly directed towards (i) development of antispermatogenic agents to suppress sperm production, (ii) prevention of sperm
maturation, (iii) prevention of sperm transport through vas deferens or rendering these sperm infertile and (iv) prevention
of sperm deposition. Research work in the field of prevention of sperm transport through vas deferens has made significant
advances. Styrene maleic anhydride (SMA) disturbed the electrical charge of spermatozoa leading to acrosome rupture and consequent
loss in fertilizing ability of sperm. A multicentre phase-III clinical trial using SMA is continuing and it is hoped that
the SMA approach would be available in the near future as an indigenously developed injectable intra-vasal male contraceptive.
The safety and efficacy of available oral contraceptives were evaluated. An indigenously developed oral contraceptive ‘Centchorman’,
which is a nonsteroidal, weakly estrogenic but potently antiestrogenic, was found to be safe and effective and is now being
marketed in India since 1991 as a ‘once a week’ pill. Cyclofem and Mesigyna have been recommended as injectable contraceptives
with proper counselling and service delivery by Indian studies. It has been recommended that these injectable contraceptives
be added to the existing range of contraceptive methods available in the National Family Planning Programme. Based on the
Indian studies CuT 200 was also recommended. Studies have indicated the advantage of intrauterine devices (IUD); they are
long acting, relatively easily removed and fertility returns rapidly after their removal. Recent studies have recommended
CuT 200 for use up to 5 years. The combination of some plant products i.e.Embelia ribes, Borax andPiper longum has been found to be safe and effective as a female contraceptive and the results of phase-I clinical trials are encouraging.
Research work is going on in the country in various areas with special reference to hormonal contraceptive — a three monthly

Genome-wide association studies in cancer have already identified over 150 regions associated with two dozen specific cancers.
Already, a handful of multi-cancer susceptibility regions have been uncovered, providing new insights into perhaps common
mechanisms of carcinogenesis. For each new susceptibility allele, investigators now face the arduous task of interrogating
each region beginning with fine mapping prior to pursuing the biological basis for the direct association of one or more variants.
It appears that there may be a significant number of common alleles that contribute to the heritability of a specific cancer.
Since each region confers a small contribution to the risk for cancer, it is daunting to consider any single nucleotide polymorphism
(SNP) as a clinical test. Since the complex genomic architecture of each cancer differs, additional genotyping and sequence
analysis will be required to comprehensively catalog susceptibility alleles followed by the formidable task of understanding
the interactions between genetic regions as well as the environment. It will be critical to assess the applicability of genetic
tests in specific clinical settings, such as when to perform screening tests with calculable risks (e.g., biopsies or chemoprevention),
before incorporating SNPs into clinical practice. To advance the current genomic observations to the clinical venue, new studies
will need to be designed to validate the utility of known genetic variants in assessing risk for cancer as well as its outcomes. 相似文献

复杂疾病目前正在全球范围流行, 极大地影响人类的健康。研究发现, 复杂疾病的性状受到多个位点的相互作用影响。目前的全基因组关联分析(Genome-wide association study, GWAS)仅仅解析单个SNP位点对疾病易感性的贡献, 单纯依靠这一种策略并不能在寻找复杂疾病的病因上得到根本性的突破。基因-基因相互作用可能是复杂疾病致病的主要因素之一。针对这一点, 科学家已经提出了一些检验基因相互作用的算法, 包括惩罚logistic回归模型、多因子降维(Multifactor dimensional reduction)、集合关联法(Set-association approach)、贝叶斯网络(Bayesian networks)、随机森林法等。文章首先对目前这些方法做了综述, 并指出了其中的不足, 包括计算复杂度太高、假设驱动、数据会过度拟合、对低维数据不敏感等, 进而简述了一种由笔者所在实验室开发的基于GPU的研究基因相互作用的算法, 该算法复杂度低, 不需要任何假设, 没有边际效应, 有很好的稳定性, 速度快, 适用于进行全基因组范围内的基因-基因相互作用计算。 相似文献

Essential genes are indispensable for the survival of an organism in optimal conditions. Recently, study on essential gene is becoming a hot topic of microbiology, genomics, and bioinformatics. This paper described the experiments that determined essential genes in some microbes and the theoretical researches on essential genes were reviewed. The major content contained comparison of essential genes and non-essential genes based on information on evolutionary conservation and sequence composition, and in silico prediction of essential genes, and analysis of the chromosomal distributions of essential genes. Knowledge about dose levels in radiation protection is an important step for risk assessment. However, in most cases of real or suspected accidental exposures to ionizing radiation (IR), physical dosimetry cannot be performed for retrospective estimates. In such situations, biological dosimetry has been proposed as an alternative for investigation. Briefly, biodosimetry can be defined as individual dose evaluation based on biological endpoints induced by IR (so-called biomarkers). The relationship between biological endpoints and absorbed dose is not always straightforward: nausea, vomiting and diarrhoea, for example, are the most well-known biological effects of individual irradiation, but a precise correlation between those symptoms and absorbed dose is hardly achieved. The scoring of unstable chromosomal-type aberrations (such as dicentrics and rings) and micronuclei in mitogen-stimulated peripheral blood, up till today, has been the most extensively biodosimetry assay employed for such purposes. Dicentric assay is the gold standard in biodosimetry, since its presence is generally considered to be specific to radiation exposure; scoring of micronuclei (a kind of by-product of chromosomal damages) is easier and faster than that of dicentrics for dose assessment. In this context, the aim of this work is to present an overview on biodosimetry based on standard cytogenetic methods, highlighting its advantages and limitations as tool in monitoring of radiation workers’ doses or investigation into accidental exposures.
