Three unique classes of carbohydrates were isolated from the hyphal cell walls of Phytophthora megasperma f.sp. glycinea (Pmg) and compared with other substances for their activity as elicitors of the phytoalexin glyceollin in soybean tissues. Glucomannans extracted from cell walls with soybean β-1,3-endoglucanase were purified and proved to be the most active elicitors yet reported. They were approximately 10 times more active in soybean cotyledons than the heterogeneous β-glucan elicitor fraction extracted from Pmg walls. In addition, the glucomannan fraction gave race-specific elicitor activity in soybean hypocotyls. Pronase was found to be a suitable reagent for the mild extraction of glycopeptides from Pmg cell walls. All of the carbohydrates isolated from Pmg cell walls possessed significant elicitor activity, but other glucans, a glucomannan and mannan from other sources, were much less active. Chitin and chitosan, reported to function as elicitors in other plants, had low activity in soybean cotyledons. Arachidonic acid was inactive, despite its previously observed elicitor activity in potato tubers. The results indicated that, for Pmg, the carbohydrate elicitor most probably involved in the initiation of phytoalexinmediated defense during fungus infection of soybean plants is the glucomannan fraction liberated by endoglucanase.  相似文献

At the apices of growing hyphae of Phytophthora megasperma f. sp. glycinea (Pmg) distinctly more fluorescein-isothiocyanate labelled Concanavalin A was bound than else-where in the mycelium. In combination with some other analytical evidence this observation conclusively indicates that the α-mannan in the cell wall of the fungus is predominantly concentrated or exposed at the hyphal tips. The possible relevance of this fact to the host-pathogene interaction between Pmg and soybean is discussed.  相似文献

Two new steroids, (22R,23S)‐3β‐hydroxy‐23‐methyl‐17,20‐epoxyergost‐5‐en‐22‐yl acetate and (22R,23S)‐5‐hydroperoxy‐23‐methyl‐5α‐17,20‐epoxyergost‐6‐ene‐3β,22‐diol, were isolated from the South China Sea soft coral Lobophytum sp., together with two related known ones. The structures of all compounds were elucidated by extensive spectroscopic analysis and by comparing their spectral data with those previously reported. The structure of (22R,23S)‐3β‐hydroxy‐23‐methyl‐17,20‐epoxyergost‐5‐en‐22‐yl acetate was further confirmed through chemical correlation. All the isolates were evaluated for the in vitro inhibitory activity against NF‐κB, a potential target for the treatment of cancer, and (22R,23S)‐5‐hydroperoxy‐23‐methyl‐5α‐17,20‐epoxyergost‐6‐ene‐3β,22‐diol exhibited moderate inhibition activity with IC50 value of 8.96 μg/mL.  相似文献

The antagonism between Gliodadium roseum, Trichoderma harzianum, or Trichothecium roseum and Phytophthora megasperam f. sp. glycinea (Pmg), cause of Phytophthora rot of soybeans (Glycine max), was studied. G. roseum, T. harzianum, and 17 isolates of T. roseum were grown separately on modified Czapek-Dox medium (MCD) in the dark for 25 days at 25 °C. Culture filtrates of T. roseum at 0.5% and 20.0% concentrations inhibited mycelial growth of Pmg at 3.1% and 90.4%; and those of G. roseum at –0.7% and 44.0%; and of T. harzianum at 0.7% and 46.0%, respectively. Culture filtrates of T. roseum at 0.5% concentration inhibited zoosporangenesis of Pmg at 98.0% and at 1.0% concentration prevented it. Culture filtrates of G. roseum and T. harzianum at 5% concentration significantly (P=0.05) inhibited zoosporangenesis of Pmg, but differences were not significant from that on MCD. Culture filtrates of 17 isolates of T. roseum inhibited mycelial growth and zoosporangenesis of Pmg at different percentages with different concentrations with those of isolate 9 showing the greatest inhibition of both. Mycelial growth of most of 16 races of Pmg was prevented at 10% concentration of the culture filtrates of isolate SS-2 of T. roseum, and all 16 races, except race 6, was prevented at 20% concentration. Zoosporangenesis of all races of Pmg was prevented at 2% the culture filtrate of SS-2. Culture filtrates of SS-2 inhibited zoosporangenesis of Pmg in soil.  相似文献

Four different PNA fragments containing units of either the R- or S- isomer of N-(2-pyrrolidine-methyl)-N-(thymine-1-acetyl)-glycine (Pmg) were synthesized on a solid support. UV thermal melting experiments with complementary RNAs were performed and it was found that R-Pmg containing PNAs bind better to RNA than those containing the S-Pmg units.  相似文献

With the large extent and great amount of soil carbon (C) storage, drylands play an important role in terrestrial C balance and feedbacks to climate change. Yet, how dryland soils respond to gradual and concomitant changes in multiple global change drivers [e.g., temperature (Ts), precipitation (Ppt), and atmospheric [CO2] (CO2)] has rarely been studied. We used a process‐based ecosystem model patch arid land simulator to simulate dryland soil respiration (Rs) and C pool size (Cs) changes to abrupt vs. gradual and single vs. combined alterations in Ts, Ppt and CO2 at multiple treatment levels. Results showed that abrupt perturbations generally resulted in larger Rs and had longer differentiated impacts than did gradual perturbations. Rs was stimulated by increases in Ts, Ppt, and CO2 in a nonlinear fashion (e.g., parabolically or asymptotically) but suppressed by Ppt reduction. Warming mainly stimulated heterotrophic Rs (i.e., Rh) whereas Ppt and CO2 influenced autotrophic Rs (i.e., Ra). The combined effects of warming, Ppt, and CO2 were nonadditive of primary single‐factor effects as a result of substantial interactions among these factors. Warming amplified the effects of both Ppt addition and CO2 elevation whereas Ppt addition and CO2 elevation counteracted with each other. Precipitation reduction either magnified or suppressed warming and CO2 effects, depending on the magnitude of factor's alteration and the components of Rs (Ra or Rh) being examined. Overall, Ppt had dominant influence on dryland Rs and Cs over Ts and CO2. Increasing Ppt individually or in combination with Ts and CO2 benefited soil C sequestration. We therefore suggested that global change experimental studies for dryland ecosystems should focus more on the effects of precipitation regime changes and the combined effects of Ppt with other global change factors (e.g., Ts, CO2, and N deposition).  相似文献

Two new abietane diterpenoids, (3S,5R,10S)‐3‐hydroxy‐12‐O‐demethyl‐11‐deoxy‐19(4→3)‐abeo‐cryptojaponol, 12,19‐dihydroxyabieta‐8,11,13‐trien‐7‐one, were isolated from Selaginella moellendorffii Hieron., together with one known abietane diterpenoid and four known tetracyclic triterpenoids. Their structures were characterized by their 1D‐ and 2D‐NMR, ECD and mass spectral studies. All compounds were tested for their inhibitory effects on proliferation of three human cancer cells (human non‐small‐cell lung carcinoma cell lines A549 and human breast adenocarcinoma cell lines MDA‐MB‐231 and MCF‐7) in vitro. Among them, three compounds displayed modest cytotoxic activities against the above three human cancer cell lines with IC50 values ranging from 16.28 to 40.67 μM.  相似文献

Ethidium bromide treatment (15 mg/l, 26°C, 18 h) of a sorghum Zheltozernoe 10 callus culture yielded line Zh10-br1 displaying multiple genetic instability. The line was characterized by a broad variety of mutations, which were identified in consecutive generations obtained from one initial regenerant via self-pollination. The mutations caused male sterility (male sterility, generation R1), a low plant height (dwarfness, R2), a reduced awn length (awnless, R3), yellow leaves in seedlings (xantha, R6), leaf variegation (leaf variegation, R6), leaf bleaching (virescence, R6), etc. In some cases, segregation in families suggested a monogenic recessive inheritance for the induced mutations. Male sterility was due to a range of defects that affected microsporogenesis and microgametogenesis and were probably caused by mutations of several genes. Leaf variegation was due to the appearance of green sectors in originally albino seedling leaves; the reversion occurred only in somatic tissues without affecting male and female gametes. In male-sterile and variegated mutants, sequence-specific amplified polymorphism analysis with primers to the Isaak transposon revealed new DNA fragments, which were absent from the original line. The results supported the hypothesis that the mutations isolated in line Zh10-br1 result from transposon mobilization induced by ethidium bromide and/or in vitro culture conditions.  相似文献

Summary Late blight in potato is caused by the fungusPhytophthora infestans and can inflict severe damage on the potato crop. Resistance toP. infestans is either based on major dominantR genes conferring vertical, race-specific resistance or on minor genes inducing horizontal, unspecific resistance. A dihaploid potato line was identified which carried theR1 gene, conferring vertical resistance to allP. infestans races, with the exception of those homozygous for the recessive virulence allele of the locusV1. The F1 progeny of a cross between this resistant parent P(R1) and P(r), a line susceptible to all races, was analysed for segregation ofR1 and of restriction fragment length polymorphism (RFLP) markers distributed on the potato RFLP map comprising more than 300 loci. TheR1 locus was mapped to chromosome V in the interval between RFLP markers GP21 and GP179. The map position ofR1 was found to be very similar to the one ofRx2, a dominant locus inducing extreme resistance to potato virus X.  相似文献

Immersion of roots of 2-day-old soybean seedlings (Glycine max cv. Harosoy 63) into solutions of several glucan elicitors caused the accumulation to various degrees of the soybean phytoalexin glyceollin. Laminarin and polytran proved to be more effective elicitors in this system than the glucan elicitor from Phytophthora megasperma f.sp. glycinea (Pmg). Digitonin and tomatin caused, in addition to glyceollin accumulation, the deposition of callose in the rhizodermis. Pretreatment of the soybean roots with laminarin effected an increase in resistance of the seedlings against a compatible race of Pmg.  相似文献

Withanolide-type steroids, withametelin Q (1) and 12α-hydroxydaturametelin B (2) along with three known withanolides, were isolated from leaves of Datura metel L. (Solanaceae). The respective structures, characterized mainly by NMR spectroscopy, were identified as (20R,22R,24R)-21,24-epoxy-1α,3β-dihydroxywitha-5,25(27)-dienolide-3-O-β-d-glucopyranoside (1) and (20R,22R,24R)-12α,21,27-trihydroxy-1-oxowitha-2,5,24-trienolide-27-O-β-d-glucopyranoside (2). The cytotoxicity of isolated compounds was evaluated against human lung carcinoma cells (A549) and human colorectal adenocarcinoma cells (DLD-1), respectively. Compound 2 exhibited cytotoxicity against A549 and DLD-1 cell lines, with IC50 values of 7 and 2.0 μM, respectively. However, for compounds 6 and 7, cytotoxicities were higher against DLD-1 cells with IC50 values of 0.6 and 0.7 μM. Both compounds blocked the cell cycle in the S-phase and induced apoptosis.  相似文献

Paris tetraphylla has a greater diversity of pollen exine sculpture than was previously thought. On the basis of 357 plants from 52 localities of Japan, we distinguished seven sculpture types that are tentatively referred to here as G1, G2, G3, R1, R2, R3 and S. G1, G2 and G3 have gemmate sculpture in which the gemmae are characteristically large, medium and small in size, respectively; likewise, R1, R2 and R3, have coarsely to finely ruglate sculpture; and S, scrobiculate sculpture. In general, any one of the seven types always or predominantly occurs in each plant, thus allowing us to make comparisons among plants and further among local populations. We thus found that G2 and G3, respectively, are the most and the second most widespread and occur in plants of 41 and 22 respectively, of the localities examined, although their frequencies differ with locality. Generally R3 occurs in plants distributed at higher latitudes, while R2 is rather widespread and common to plants growing on high mountains. G1 and R1 are restricted to plants occurring south of Ibaraki Pref. and north of Gunma Pref., respectively. Such localized distributional patterns of some types may reflect the reproductive nature and the history of geographical isolation inParis tetraphylla over the Japanese Islands.  相似文献

The epimeric diterpenes (+)‐(1S,3E,7E,11S,12S)‐verticilla‐3,7‐dien‐12‐ol ( 1 ), isolated from Bursera suntui, and (+)‐(1S,3E,7E,11S,12R)‐verticilla‐3,7‐dien‐12‐ol ( 2 ), isolated from Bursera kerberi, gave the same Wagner‐Meerwein rearrangement product (?)‐(1E,4Z,8Z,11S,12R)‐phomacta‐1,(15)4,8‐triene ( 3 ). The Et2O:BF3‐induced transformations evidence that verticillenes and phomactanes, both containing the bicyclo[9.3.1]pentadecane skeleton, are biogenetically related through the verticillen‐12‐yl cation ( A + ), which also is a key intermediate in the biosynthetic pathways to generate antitumor taxanes. Molecular modeling using the Monte Carlo protocol, followed by density functional theory (DFT) geometry optimization employing the hybrid functionals B3LYP and B3PW91, both with the DGDZVP basis set, secured the configuration of 3 as followed from the good agreement between the calculated and experimental vibrational circular dichroism spectra. Similar DFT calculations allowed determining the absolute configuration of (+)‐(1R,4R,5R,8S,9S,11S,12R,15R)‐1,15:4,5:8,9‐triepoxyphomactane ( 9 ), which surprisingly derives from epoxidation of the second minimum energy conformer of 3 .  相似文献

The contemporary races of Puccinia striiformis f. sp. tritici (Pst) in Egypt during 2016–2018 were differentiated based on virulence and molecular patterns. Virulence patterns based on the reaction of the 17 World/European differential sets carrying stripe rust resistance genes (Yr genes) resulted in ten races including four new (first recorded in Egypt) and six old (previously recorded in Egypt). The new races were identified as 64E0 (virulence [V] Yr4, Su), 0E16 (V Yr8, 19), 66E0 (V Yr4, 7, 22, 23, Su) and 4E130 (V Yr2, 6, 7, 25, HVII), while the old were 0E0 (avirulence), 2E0 (V Yr7, 22, 23), 2E16 (V Yr7, 8, 19, 22, 23), 4E0 (V Yr2, 6), 6E4 (V Yr2, 6, 7, 22, 23, 25) and 70E4 (V Yr2, 4, 6, 7, 22, 23, 25, Su). Cluster analysis differentiated Pst races based on virulence frequency to Yr genes. Simple sequence repeat (SSR) markers were used to detect the molecular polymorphism of the Pst races. Clustering separated the old and new races into two groups, indicating their common ancestry since the new races were very distinct from the old races. Although clustering based on virulence revealed some evolutionary patterns, where the new races 64E0 and 66E0 may have probably evolved from the old races (2E16, 2E0, 6E4, 70E4) and the new race 4E130 may be evolved from the joint race 4E0. However, clustering based on molecular patterns indicated that the new races appear to be genetically distinct and may represent an exotic introduction rather than a mutation in isolates of the old races. A weak association between virulence and molecular patterns revealed that they are independent of each other. The SSR markers did not correspond to the virulences in the pathogen. Further studies on the potential virulence genes of the detected Pst virulences are needed.  相似文献

Summary Resistance to race 3 of Fusarium wilt in the wild tomato Lycopersicon pennellii (LA 716) was previously found to be controlled by one major locus, I-3, tightly linked to Got-2 on chromosome 7. This accession was also found to carry resistance to races 1 and 2; a genetic analysis of these resistances is reported in this paper. This analysis proceeded in two steps. First, allelism tests demonstrated that race 1 and 2 resistances carried by L. pennellii were not allelic to the I and I-2 genes originally incorporated into L. esculentum from L. pimpinellifolium. Second, an interspecific backcross with L. pennellii (BC1) was used to determine the mode of inheritance of these new resistances and their chromosomal location by segregation and linkage analysis. BC1 responses to each of the races were determined using progeny tests (BC1S1). BC1S1 plants were inoculated with race 1 or 2 and evaluated after 1 month using a visual disease rating system; mean disease ratings were calculated for each BC1 individual for each race based on the progeny scores. A bimodal frequency distribution of the BC1 mean disease ratings was observed for both races, indicating that one major locus controlled resistance in each case. Statistical comparisons of the mean disease ratings of homozygous versus heterozygous individuals at each of 17 segregating enzyme loci were used to map the resistances to races 1 and 2. Tight linkage was detected between the enzyme locus Got-2 and resistances to both races, as was previously reported for the I-3 locus. Therefore, the Got-2 locus can be used as a selectable marker for resistances to all three races. The relationship of these resistances is discussed in the paper. In addition, as previously reported for race 3, significance was also detected for the chromosome segment marked by Aps-2 on chromosome 8 for both races. Currently many cultivars carry I and I-2 resistances to races 1 and 2. Strawberry fungi were isolated from fresh fruits and juice on the two types of media (Sabouraud dextrose agar, SDA and potato-dextrose agar, PDA) at 28 °C. Nineteen fungal species belong to 12 genera were isolated from fruits and juice on both isolation media. The most common fungal genera and species were Aspergillus flavus, A. niger, Mucor racemosus, Neurospora crassa, Penicillium chrysogenum, Rhizopus stolonifer and Trichoderma harzianum. Twenty A. flavus and A. parasitics isolates were assayed for their abilities to produce aflatoxins. The concentration of aflatoxins ranged between 25.8–75.2 and 23.6–71.1 ng/ml at 350 and 365 nm, respectively. Among A. flavus and A. parasiticus strains tested, aflatoxin B contributed 30–60% of total isolates. However, G type contributed 85–90%. The Rf values of B1, B2, G1 and G2 were 0.79, 0.61, 0.44 and 0.32, respectively. High-performance liquid chromatography analysis of extracts revealed the presence of aflatoxins with variable levels.  相似文献

Conjugal transferability of drug resistance was examined, in eleven Pseudomonas aeruginosa strains which were isolated in Frankfurt. Four R factors were demonstrated from three strains using P. aeruginosa as recipients but they were nontransferable to Escherichia coli K12. Two R factors, i.e., Rms146 and Rms147, mediated resistances to tetracycline (TC), streptomycin (SM), sulfanilamide (SA), kanamycin (KM), lividomycin (LV), gentamicin C complex (GM) and 3′,4′-dideoxykanamycin B (DKB). They mediated the formation of aminoglycoside-inactivating enzymes, i.e., SM phosphotransferase, SM adenylyltransferase, KM and LV phosphotransferase 1, and GM and DKB 6′-N-acetyltransferase. TC resistance conferred by these R factors was due to impermeability of the drug. P. aeruginosa Ps 142 carried two kinds of R factor in one cell, Rms148 (SM) and Rms149 (SM·SA·GM·CPC) (CPC, carbenicillin). Rms148 (SM) was transferable at a high frequency of 10–1 and mediated the formation of SM phosphotransferase. Rms149 mediated the formation of drug-inactivating enzymes, i.e., GM 3-N-acetyltransferase and β-lactamase, but did not inactivate SM. SM resistance was probably due to impermeability of the drug.  相似文献

Fusarium oxysporum f. sp. ciceri, the causal agent of chickpea wilt, is an important fungal pathogen in India. Thirteen oligonucleotide probes complementary to microsatellite loci, in combination with 11 restriction enzymes, were used to assess the potential of such markers to study genetic variability in four Indian races of the pathogen. Hybridisation patterns, which were dependent upon both the restriction enzyme and oligonucleotide probe used, revealed the presence of different repeat motifs in the F. oxysporum f. sp. ciceri genome. Among the restriction enzymes used, hexa-cutting enzymes were more informative than tetra- and penta-cutting enzymes, whereas tetranucleotide and trinucleotide repeats yielded better hybridisation patterns than dinucleotide repeats. Dependent upon the levels of polymorphism detected, we have identified (AGT)5, (ATC)5 and (GATA)4 as the best fingerprinting probes for the F. oxysporum f. sp. ciceri races. The distribution of microsatellite repeats in the genome revealed races 1 and 4 to be closely related at a similarity index value of 76.6%, as compared to race 2 at a similarity value of 67.3%; race 3 was very distinct at a similarity value of 26.7%. Our study demonstrates the potential of oligonucleotide probes for fingerprinting and studying variability in the F. oxysporum f. sp. ciceri races and represents a step towards the identification of potential race diagnostic markers. Two new sterols 1 and 2 and five known ones 3 – 7 were isolated for the first time from the fruiting bodies of Cortinarius glaucopus. Their structures were established by 1‐ and 2D‐NMR spectra and HR‐FABS‐MS. The relative configuration of 1 was firmly determined by comparison of the observed 1H–1H couplings and NOESY correlations, with those predicted for the computed geometries of the conformers. Calculations were performed by means of DFT with the B3LYP functional at 6‐31 + G(d,p) level of theory, in CHCl3 as the solvent. The structures of the new ergosterol derivatives, called glaucoposterol A ( 1 ) and B ( 2 ), were thus established as (3S,5R,7R,10R,13R,17R,20S,22R,23R,24R)‐5,6‐epoxy‐3,7,23‐trihydroxystrophast‐8‐en‐14‐one and (22E,3S,5S,9S,10R,13R,17R,20R,24R)‐3,5‐dihydroxyergosta‐6,8(14),22‐trien‐15‐one, respectively. Moreover, the configuration of known strophasterol C ( 3 ) was determined as (3S,5R,6S,7R,10R,13R,17R,20S,22S,24R). Glaucoposterol A ( 1 ) and strophasterol C ( 3 ) represent the second finding in nature of steroids with the rare strophastane skeleton.  相似文献
