We developed 15 polymorphic microsatellite loci from the Hawaiian tree Metrosideros polymorpha. These loci were screened against two varieties from several populations and from 23 individuals from one mid-elevation population on Hawaii Island. Loci were variable with the number of alleles per locus ranging from three to 24. Polymorphic information content ranged from 0.222 to 0.941, and observed heterozygosity ranged from 0.261 to 0.955.  相似文献

The genus Plantago is particularly interesting for evolutionary studies because of its wide range of mating systems. We have developed primers for five highly polymorphic microsatellite loci isolated from P. lanceolata. All five loci amplified and were polymorphic in the two populations examined, Lowsteads Beach in the United Kingdom and Duke in the United States. These new markers will allow a comparison of population structure between the outcrossing species P. lanceolata, and the highly selfing species P. major.  相似文献

We describe polymerase chain reaction (PCR) primers and amplification conditions for seven microsatellite DNA loci isolated from the red‐spotted newt (Notophthalmus viridescens). Primers were tested on 16 individuals from two populations on the Savannah River Site in Aiken County, South Carolina. We detected six to 10 alleles per locus and an overall observed heterozygosity range of 0.31–0.81. Despite low heterozygosity at two of the seven loci, the high polymorphic information contents (from 0.54 to 0.85) of these markers render them useful for future studies of the behavioural and population ecology of this common salamander.  相似文献

We describe polymerase chain reaction primers and amplification conditions for 13 microsatellite DNA loci isolated from two bisexual species of whiptail lizards Aspidoscelis costata huico and Aspidoscelis inornata. Primers were tested on either 16 or 48 individuals of A. c. huico and/or 26 individuals of A. inornata. Ten of the 13 primers were also tested against a panel of 31 additional whiptail taxa. We detected three to nine alleles per locus in A. c. huico and four to 19 alleles per locus in A. inornata, with observed heterozygosity ranging from 0.60 to 0.87 and from 0.15 to 1.00, respectively. These primers will be an important resource for surveys of genetic variation in these lizards.  相似文献

Two multiplex PCR amplifications were performed to analyse six microsatellite loci of Schlegel's black rockfish, Sebastes schlegeli, an important commercial fish in the northern part of Japan and an important species for the stock enhancement program in this area. We analysed 67 wild samples from Yamada Bay, Iwate Prefecture, Japan. The observed genotype frequencies agreed with the Hardy–Weinberg expectations at all loci, and the observed heterozygosities ranged from 0.072 to 0.897.  相似文献

We obtained a microsatellite‐enriched genomic library isolated from the tissue of a single columbine (Aquilegia sp.) plant taken from a southwestern USA natural population. The primers developed for these microsatellite loci performed consistently in polymerase chain reactions and yielded multiallelic genotypes with relatively high observed heterozygosities. We describe polymerase chain reaction primers and conditions to amplify 16 unique, codominant di‐, tri‐ and tetra‐nucleotide microsatellite DNA loci so that other population biology researchers using columbine natural populations as a model system may benefit.  相似文献

In this paper, we report the isolation of 11 polymorphic microsatellite loci from Lithophragma maximum (Saxifragaceae), a rare species restricted to San Clemente Island, California. We found moderate levels of allelic variation (mean NA= 5.5) and high levels of expected heterozygosity (mean HE= 0.531) across sampled individuals. However, eight loci showed a significant absence of heterozygous individuals, which we attribute to small population size and demographic history of the species.  相似文献

Seventeen novel microsatellite markers were isolated and characterized from an enriched DNA library of the pest species Bactrocera dorsalis sensu stricto. The polymorphism of these loci was tested in individual fruit flies from a Thai mass‐rearing strain and three wild Asian populations: two from Thailand and one from Myanmar. Allele numbers per locus ranged from two to 15. In the natural populations, a high level of polymorphism was detected in all loci suggesting the usefulness of these markers to quantify the genetic variation in this highly invasive insect.  相似文献

Yuccas and their pollinator moths are a textbook example of mutualism, yet we lack sufficiently variable markers to properly study the population genetics of the plants. We characterized 13 polymorphic microsatellite loci for Yucca filamentosa by screening primers derived from an expressed sequence tag database. We found four to 13 alleles per locus and the observed heterozygosity ranged from 0.31 to 1. These markers will be useful in future ecological studies of Y. filamentosa.  相似文献

The extent of dispersal between populations of Caribbean reef fish remains largely unknown. Resolution of this issue is critical for managing populations and designing marine reserves. As part of a larger study of reef fish dispersal, we developed and characterized 11 polymorphic microsatellite loci for the bicolor damselfish (Stegastes partitus), an abundant reef fish found throughout the Caribbean. These loci will be useful for population genetic and relatedness studies of this species and also appear to be useful in other Caribbean members of the Pomacentridae.  相似文献

Six polymorphic microsatellite loci are isolated from the Oriental fruit fly Bactrocera dorsalis (Hendel), an agricultural pest in Asia, including Taiwan. To assess their potential utility as high‐resolution genetic markers, polymerase chain reaction (PCR) primers, amplification conditions, and an automated fluorescence detection protocol were developed. In analyses of 71 individual flies from six different areas of Taiwan, allele numbers ranged from five to 25 were detected for each locus. The observed heterozygosity ranged between 0.268 and 0.737 among these loci. No linkage disequilibrium was found. These microsatellite markers have potential utility to population structure and gene flow studies of B. dorsalis (Hendel).  相似文献

We developed six polymorphic microsatellite markers for an ectomycorrhizal Pisolithus sp., which are associated with Eucalyptus camaldulensis in Thailand using a method starting with amplified fragments of intersimple sequence repeats (ISSR). The number of alleles per locus varied between two and five and expected heterozygosities ranged from 0.123 to 0.625. These loci are available for analysis of genetic structure and gene flow in Eucalyptus‐associated Pisolithus spp. populations.  相似文献

We describe polymerase chain reaction (PCR) primers and conditions to amplify one dinucleotide and eight tetranucleotide microsatellite DNA loci isolated from the dollar sunfish (Lepomis marginatus). The PCR primers were tested on 20 or more individuals from five populations. The dollar sunfish microsatellite primers developed yielded a high number of alleles (4 to 14 per locus), and high observed heterozygosities (0.500–0.857).  相似文献

We have isolated and characterized five microsatellite loci from Rhizopogon occidentalis and six loci from Rhizopogon vulgaris (Boletales, Basidiomycota). Microsatellite variation was assessed using 32 R. occidentalis and 48 R. vulgaris individuals from four populations in California. The number of alleles across populations ranged from two to 10 for R. occidentalis and three to eight for R. vulgaris. Expected heterozygosity values within populations ranged from 0.00 to 0.85 for R. occidentalis and 0.00 to 0.75 for R. vulgaris. These are the first microsatellite loci isolated for R. occidentalis and R. vulgaris and will be useful in the examination of their population genetic structure.  相似文献

We isolated and characterized 15 microsatellite loci from the endemic Jamaican streamertail hummingbird Trochilus polytmus. Loci were screened in 12 individuals of both T. polytmus and its sister species T. scitulus, also a Jamaican endemic. The number of alleles per locus ranged from 2 to 10, observed heterozygosity ranged from 0 to 1, and the probability of identity values ranged from 0.038 to 0.663. Sixteen microsatellite loci isolated from Halesia tetraptera are described. All 16 are polymorphic, with four to 13 alleles across 15–19 members of a single half‐sib family. Heterozygosity ranged from 0.067 to 1. One locus departs significantly from Hardy–Weinberg equilibrium in our test family. The test population shows significant heterozygote deficiency at this and two other loci. Thirteen loci exhibit significant linkage disequilibrium. These loci will be utilized in paternity analyses of geographically diverse half‐sib families.  相似文献

Twenty microsatellite loci were isolated from a hybrid of two daylilies, Hemerocallis fulva and Hemerocallis citrina. We characterized individuals from two H. fulva populations and two H. citrina populations in Japan and observed three to 20 alleles per locus in H. fulva and one to 19 alleles per locus in H. citrina. Mean observed heterozygosity within populations ranged from 0.35 to 0.85 in H. fulva and from 0 to 0.95 in H. citrina. In about a half of the loci, the observed heterozygosity did not deviate from Hardy–Weinberg equilibrium. These loci are proved useful in studying gene flow and qualitative trait loci mapping using the two species.  相似文献

Ten microsatellite loci were isolated and characterized for Callicarpa subpubescens (Verbenaceae), an endemic tree species of the Bonin Islands. The observed number of alleles at each locus ranged from two to eight with an average of 4.9, and the expected heterozygosity ranged from 0.238 to 0.690 with an average of 0.483. All 10 loci were screened in cross-amplification tests for two other endemic Callicarpa species that also inhabit the Bonin Islands. All loci were successfully amplified in these species.  相似文献

There is considerable interest in the genetic structure of Kelletia kelletii because of its economic, ecological and scientific importance. To that end, we developed species-specific primers which amplify mitochondrial cytochrome c oxidase subunit I as well as 13 hypervariable nuclear microsatellite loci. Using dye-labelled primers, the microsatellite loci can be co-amplified in two multiplex polymerase chain reactions and scored simultaneously on an automated sequencer.  相似文献
