Forty-eight 2-year-old Liaoning Cashmere goats (body weight = 38.0 ± 2.94 kg) were used to investigate the effects of dietary iodine (I) and selenium (Se) supplementation on nutrient digestibility, serum thyroid hormones, and antioxidant status during the cashmere telogen period to learn more about the effects of dietary I and Se on nutrition or health status of Cashmere goats. The goats were equally divided into six groups of eight animals each that were treated with 0, 2, or 4 mg of supplemental I/kg dry matter (DM) and 0 or 1 mg of supplemental Se/kg DM in a 2 × 3 factorial arrangement of treatments. The six treatments were I0Se0, I2Se0, I4Se0, I0Se1, I2Se1, and I4Se1. The concentrations of I and Se in the basal diet were 0.67 and 0.09 mg/kg DM, respectively. The study started in March and proceeded for 45 days. Supplemental I or Se alone had no effect on nutrient digestibility and nitrogen metabolism. However, the interaction between I and Se was significant regarding the digestibility of acid detergent fiber (ADF; P < 0.05), and compared with group I4Se1, the digestibility of ADF was significantly increased in group I4Se0 (P < 0.05). Selenium supplementation did not affect serum triiodothyronine (T3) or thyroxine (T4) concentrations. However, the concentration of serum T4 but not that of T3 was significantly increased with I supplementation (P < 0.05). In addition, serum superoxide dismutase (SOD) activity was not affected (P > 0.05), but serum glutathione peroxidase (GSH-Px) activity was significantly decreased by I supplementation (P < 0.05). The antioxidant status was improved by Se supplementation, and the activities of SOD and GSH-Px were significantly increased (P < 0.05).  相似文献

Keshan disease is a cardiomyopathy restricted to the endemic areas of China and seen in residents having an extremely low selenium (Se) status. Prophylactic administration of sodium selenite has been shown to decrease significantly the incidence of acute and subacute cases. The aim of the study was to assess the relative bioavailability of selenite versus organic Se-yeast in a Se-deficient area in China with a randomized double-blind double-dummy design. Healthy children (n=30) between 14 and 16 yr of age were randomized into three equal groups receiving either 200 μg/d selenite Se or 200 μg/d Se-yeast or placebo for 12 wk. Blood was drawn at baseline, 4, 8, and 12 wk and 4 wk postsupplementation. The plasma Se concentration (mean ± SD) was 0.16±0.03 μmol/L at baseline. Selenite and Se-yeast supplementation increased plasma Se to plateau values, 1.0±0.2 and 1.3±0.2 μmol/L, respectively. In red cells, Se-yeast increased the selenium level sixfold and selenite threefold compared to placebo. The relative bioavailability of Se-yeast versus selenite measured as glutathione peroxidase (GSHPx) activity was similar in plasma, red blood cells, and platelets. GSHPx activity reached maximal levels in plasma and platelets of 300% and 200%, respectively, after 8 wk compared to the placebo group, but continued to increase in red cells for 16 wk. Our study showed that although both forms of Se were equally effective in raising GSHPx activity, Se-yeast provided a longer lasting body pool of Se. Se-yeast may be a better alternative to selenite in the prophylaxis of Keshan disease with respect to building up of body stores.  相似文献

Ulcerative colitis increases oxidative damage accompanied by production of free oxygen radicals. Selenium (Se) and vitamin E are two natural antioxidants. The present study was undertaken to investigate the possible protective role of Se and vitamin E combination in experimental colitis induced by acetic acid (AA) in rats. This study was carried out on three groups, namely the first (control), the second (experimental colitis group, 2 ml 5% acetic acid), and the third groups (2 ml 5% acetic acid, vitamin E (100 mg/kg body weight (bw)) plus Se (0.2 mg/kg bw)). The activities of catalase (CAT), prolidase (PRS), myeloperoxidase (MPO), total antioxidant capacity (TAC), total oxidant status (TOS), oxidative stress index (OSI), total thiol (T-SH) were determined in plasma and colon samples. Macroscopic and microscopic damages in colon were increased by AA treatment (p < 0.01 and p < 0.01, respectively), whereas they were decreased by selenium and vitamin E treatment (p < 0.05 and p < 0.01, respectively). The activities of CAT and PRS in the plasma and colon were significantly affected (p < 0.05 and p < 0.01) by treatment of AA, Se, and vitamin E. MPO activity in colon was increased (p < 0.01) by AA treatment and decreased (p < 0.05) by Se and vitamin E administration. The values of TOS and OSI in plasma were increased (p < 0.5) by AA. The TAC and T-SH in colon were decreased (p < 0.05) by AA and increased (p < 0.05) by Se and vitamin E. Based upon these results, Se and vitamin E may play an important role in preventive indication of the oxidative damage associated by acetic acid caused inflammation.  相似文献

Ten pregnant female camels divided into two groups received, after a 2-week adaptation period, an oral selenium (Se) supplementation (0 and 2 mg, respectively) under sodium selenite form for 6 months from the three last months of gestation up to the three first months of lactation. Feed intake was assessed daily. Blood samples and body weight were taken on a biweekly basis, both in dams and their camel calves after parturition. Feces and urine samples were collected monthly and milk on a biweekly basis. The Se concentration in serum increased significantly in the supplemented group and was threefold higher than the concentration compared to the control group, respectively, 305.9 ± 103.3 and 109.3 ± 33.1 ng/mL. The selenium concentration increased in similar proportion in milk (86.4 ± 39.1 ng/mL in the control group vs 167.1 ± 97.3 ng/mL in treated group), in urine, and feces. The gluthathione peroxidase (GSH-Px) activity varied between 18.1 ± 8.7 IU/g hemoglobin (Hb) in control group and 47.5 ± 25.6 IU/g Hb in treated group but decreased after parturition in both groups. Vitamin E did not change significantly and was, on average, 1.17 ± 0.72 and 1.14 ± 0.89 ng/mL in the control and treated groups, respectively. Significant correlations were reported between serum Se, milk Se, GSH-Px, and fecal and urinary excretion or concentration. Blood values in camel calves were similar to those of the dams. The results seemed to confirm the sensitivity of camel to Se supplementation with an important increase of selenium in serum and milk.  相似文献

Selenium is an essential trace element possessing immune-stimulatory properties. The purpose of this 42-day study was to investigate the effects of excess dietary sodium selenite on immune function by determining morphological changes and apoptosis of bursa of Fabricius. Three hundred 1-day-old Avian broilers were fed on a basic diet (0.2 ppm selenium) or the same diet amended to contain 1, 5, 10, and 15 ppm selenium supplied as sodium selenite (n = 60/group). Relative weight of bursa was significantly decreased in the 1, 5, 10, and 15 ppm groups at 28 days of age, when compared with that of 0.2 ppm group. Pathological lesions were progressed with the dietary Se level increased. The gross lesions of bursa involved obvious atrophy with decreased volume and pale color. Histopathologically, decreased number of lymphocytes and loosely packed lymphocytes appeared in the medulla and cortex in the follicles. Ultrastructurally, mitochondria injury and increased apoptotic cells with condensed nuclei were observed. In comparison to that of control group, excess Se (5, 10, and 15 ppm) intake increased the percentage of Annexin V positive cells, as measured by flow cytometry. Terminal deoxynucleotidyl transferase 2′-deoxyuridine 5′-triphosphate nick end-labeling assay showed that there were increased frequencies of apoptotic cells in 10 and 15 ppm selenium groups. These data suggest that Se supplementation with sodium selenite should be carefully evaluated as excess selenium (more than 5 ppm) intake could cause profound immunologic inhibition.  相似文献

Eight young female camels shared in four groups of two 2 years received a basal diet enriched respectively with 0, 2, 4, and 8 mg selenium under sodium selenite form for 64 days. Feed intake was assessed daily; blood samples were taken on weekly basis. One camel from each group was killed at the end of the experiment. Se concentration in serum was increased significantly in the supplemented groups with an average of 176.3 ± 18.0 ng/mL in the control group, 382.7 ± 107.6 in the group receiving 2 mg Se, 519.8 ± 168.4 in the group receiving 4 mg Se, and 533.4 ± 158.6 in the group receiving 8 mg Se daily. For glutathione peroxidase (GSH-Px) activity, the control group (51.0 IU/g Hb) and the group receiving 2 mg (50.5 IU/g Hb) were significantly different than groups receiving 4 and 8 mg (respectively, 65.9 and 76.1 IU/g Hb). No significant variation occurred for vitamin E (mean, 0.56 ± 0.23 ng/mL). Significant correlation between serum Se and GSH-Px was reported. Kidney was the richest organ in selenium followed by lung, spleen, and liver, but the increase in supplemented groups was more marked in liver and kidney. The hair seemed to be the best indicator of selenium intake in camel.  相似文献

The goal of the experiment was to monitor the changes in the selenium concentration in goat milk during short-term oral supplementation of three different forms of selenium. The experiment involved 24 lactating goats of white shorthaired breed. Group C was the control; group S received selenium in the form of selenium-enriched yeast, group L in the form of lactate, and group B in the form of proteinate. Individual selenium preparations were administered individually orally in 250 μg Se dose per animal for 20 days. After the beginning of selenium supplementation, the selenium concentration in milk during the first 5 days grew gradually in group S. Between days 7 and 20 of Se supplementation, the mean Se concentrations in milk in groups were 12.53 ± 3.69 μg l−1 (C), 25.90 ± 6.30 μg l−1 (S), 13.14 ± 3.54 μg l−1 (L), 11.70 ± 3.69 μg l−1 (B). Differences between group S and other groups (C, B, L) were highly significant (p < 0.0001). Based on our results, selenium in the form of lactate and proteinate was excreted into the milk similarly, but selenium in the form of yeast, which contains high amount of selenomethionine, was excreted by milk in the highest amounts.  相似文献

Deletion of selenoprotein P alters distribution of selenium in the mouse   总被引:15,自引:0,他引:15  
Selenoprotein P (Se-P) contains most of the selenium in plasma. Its function is not known. Mice with the Se-P gene deleted (Sepp(-/-)) were generated. Two phenotypes were observed: 1) Sepp(-/-) mice lost weight and developed poor motor coordination when fed diets with selenium below 0.1 mg/kg, and 2) male Sepp(-/-) mice had sharply reduced fertility. Weanling male Sepp(+/+), Sepp(+/-), and Sepp(-/-) mice were fed diets for 8 weeks containing <0.02-2 mg selenium/kg. Sepp(+/+) and Sepp(+/-) mice had similar selenium concentrations in all tissues except plasma where a gene-dose effect on Se-P was observed. Liver selenium was unaffected by Se-P deletion except that it increased when dietary selenium was below 0.1 mg/kg. Selenium in other tissues exhibited a continuum of responses to Se-P deletion. Testis selenium was depressed to 19% in mice fed an 0.1 mg selenium/kg diet and did not rise to Sepp(+/+) levels even with a dietary selenium of 2 mg/kg. Brain selenium was depressed to 43%, but feeding 2 mg selenium/kg diet raised it to Sepp(+/+) levels. Kidney was depressed to 76% and reached Sepp(+/+) levels on an 0.25 mg selenium/kg diet. Heart selenium was not affected. These results suggest that the Sepp(-/-) phenotypes were caused by low selenium in testis and brain. They strongly suggest that Se-P from liver provides selenium to several tissues, especially testis and brain. Further, they indicate that transport forms of selenium other than Se-P exist because selenium levels of all tissues except testis responded to increases of dietary selenium in Sepp(-/-) mice.  相似文献

The current study aimed to investigate if different sources of supplemental zinc (Zn), manganese (Mn), copper (Cu), and selenium (Se) fed to dry and lactating dairy cows affect reproductive performance, lameness status, and colostrum production. The experiment was carried out on 60 multiparous non-lactating Holstein cows in a commercial dairy herd. The cows received randomly mineral mixtures in three treatment groups containing inorganic, 25% organic–75% inorganic, or 50% organic–50% inorganic forms of Zn, Mn, Cu, and Se starting from approximately 3 weeks prior to calving up to 90 days postpartum. Supplements were included in rations and fed twice a day. Reproductive parameters including days to first service, days to first estrus, service per conception, and conception rate were investigated. After parturition, colostrum production, composition, and immunoglobulin G1 concentration were determined. Lameness, as an indicator of trace minerals deficiency, was evaluated in a five-score scale. The source of mineral supplement only numerically improved the assessed parameters excluding colostrum fat, protein, and ash percent. The organic form of supplements did not significantly affect reproductive performance, lameness score, or colostrum production.  相似文献

Footrot (FR) is a common, contagious bacterial disease of sheep that results in lameness and significant economic losses for producers. We previously reported that sheep affected with FR have lower whole-blood (WB) selenium (Se) concentrations and that Se supplementation in conjunction with routine control practices accelerates recovery from FR. To determine whether oral Se-yeast administered at supranutritional levels (>4.9 mg Se/week) alters the ability of sheep to resist or recover from FR infection, 60 ewes with and 60 ewes without FR were drenched once weekly for 62.5 weeks with 0, 4.9, 14.7, or 24.5 mg organic Se-yeast (30 ewes per treatment group). Footrot prevalence and severity were measured at 0, 20, 28, 40, and 60 weeks of Se supplementation. Genomic expression of eight WB-neutrophil genes for selenoproteins and seven WB-neutrophil genes for proteins involved in innate immunity was determined at the end of the treatment period using SYBR Green and quantitative polymerase chain reaction methodology. Supranutritional Se-yeast supplementation successfully increased Se status in sheep but did not prevent FR. Supranutritional Se-yeast supplementation increased WB-neutrophil expression of genes involved in innate immunity: l-selectin, interleukin-8 receptor, and toll-like receptor 4, which were or tended to be lower in ewes affected with FR. Furthermore, supranutritional Se-yeast supplementation altered the expression of selenoprotein genes involved in innate immunity, increasing selenoprotein S and glutathione peroxidase 4 and decreasing iodothyronine deiodinases 2 and 3. In conclusion, supranutritional Se-yeast supplementation does not prevent FR, but does alter WB-neutrophil gene expression profiles associated with innate immunity, including reversing those impacted by FR.  相似文献

The present study was conducted to investigate the effects of chromium histidinate (CrHis) against experimentally induced type II diabetes and on chromium (Cr), zinc (Zn), selenium (Se), manganese (Mn), iron (Fe), and copper (Cu) in serum, liver, and kidney of diabetic rats. The male Wistar rats (n = 60, 8 weeks old) were divided into four groups. Group I received a standard diet (12% of calories as fat); group II were fed standard diet and received CrHis (110 mcg CrHis/kg body weight per day); group III received a high-fat diet (HFD; 40% of calories as fat) for 2 weeks and then were injected with streptozotocin (STZ) on day 14 (STZ, 40 mg/kg i.p.; HFD/STZ); group IV were treated as group III (HFD/STZ) but supplemented with 110 mcg CrHis/kg body weight per day. The mineral concentrations in the serum and tissue were determined by atomic absorption spectrometry. Compared to the HFD/STZ group, CrHis significantly increased body weight and reduced blood glucose in diabetic rats (p < 0.001). Concentrations of Cr, Zn, Se, and Mn in serum, liver, and kidney of the diabetic rats were significantly lower than in the control rats (p < 0.0001). In contrast, higher Fe and Cu levels were found in serum and tissues from diabetic versus the non-diabetic rats (p < 0.001). Chromium histidinate supplementation increased serum, liver, and kidney concentrations of Cr and Zn both in diabetic and non-diabetic rats (p < 0.001). Chromium supplementation increased Mn and Se levels in diabetic rats (p < 0.001); however, it decreased Cu levels in STZ-treated group (p < 0.001). Chromium histidinate supplementation did not affect Fe levels in both groups (p > 0.05). The results of the present study conclude that supplementing Cr to the diet of diabetic rats influences serum and tissue Cr, Zn, Se, Mn, and Cu concentrations.  相似文献

The objective of this study was to investigate the effects of different forms of Se supplementation on the antioxidant defense and glucose homeostasis in experimental diabetes. Sodium selenate (SS) or selenomethionine (SM) were administered (2 μmol Se kg−1 day−1) via orogastric route to streptozotocine (STZ)-induced diabetic rats in addition to basal diet for 12 weeks. Glucose levels in whole blood, glutathione peroxidase (GSH-Px) activity in erythrocytes, Se and fructosamine levels in plasma were evaluated monthly. Plasma Se levels increased significantly in all diabetic groups compared to basal measurements, being more prominent in SM group [p(SM3/SM0) = 0.018]. The increase in GSH-Px activities was significant at the end of the second month in SS [p(SS2/SS0) = 0.028], whereas at the end of the third month in SM the value was lower [p(SM3/SM0) = 0.018] and the unsupplemented diabetic control (DC) groups, p(DC3/DC0) = 0.012. Glucose increased significantly only in DC group. Fructosamine increased gradually in all diabetic groups, being significant in DC and SS groups. At the end of the third month, highest fructosamine levels were observed in SS group, which were significantly higher than the SM group [p(SM/SS) = 0.010]. In conclusion, Se augmented the antioxidant defense by increasing GSH-Px activity and this effect was more prominent when Se was supplemented as SM, which exerted positive effects also on glucose homeostasis.  相似文献

The trace element selenium is an essential micronutrient for human health and its low levels in serum are implicated in the pathogenesis of several chronic diseases. Therefore, the determination of total selenium in serum may contribute to the assessment of the health and nutritional status of certain populations. The objective of the present work was to determine total selenium in the serum of 506 healthy volunteers that participated in the ATTICA study. Selenium was determined in serum by using the technique of inductively coupled plasma mass spectrometry. The mean serum selenium concentration was determined to be 91.8 ± 33.7 μg/L (N = 506); 87.6% of women and 88.5% of men had serum selenium concentration below 125 μg/L, the cutoff considered to be required for optimal glutathione peroxidase activity. No association was found between serum selenium levels and the gender of the participants while a significant decline of selenium with age (p < 0.0001) was observed. According to our results, no anthropometric, lifestyle, nutritional, or biochemical indices were able to affect the association between serum selenium and age. This result may indicate that other factors such as selenium distribution as well as retention may be affecting the relationship between serum selenium and age.  相似文献

Since there are no data about the protective role of selenium (Se) against cadmium (Cd)-induced oxidative damage in early life, we studied the effect of Se supplementation on antioxidative enzyme activity and lipid peroxidation (through thiobarbituric acid reactive substances; TBARS) in suckling Wistar rats exposed to Cd. Treated animals received either Se alone for 9 days (8 μmol, i.e., 0.6 mg Se as Na2SeO3 kg−1 b.w., daily, orally; Se group), Cd alone for 5 days (8 μmol, i.e., 0.9 mg Cd as CdCl2 kg−1 b.w., daily, orally; Cd group), or pre-treatment with Se for 4 days and then co-treatment with Cd for the following 5 days (Se + Cd group). Our results showed that selenium supplementation, with and without Cd, increased SOD activity in the brain and kidney, but not in the liver and GSH-Px activity across all tissues compared to control rats receiving distilled water. Relative to the Cd group, Se + Cd group had higher kidney and brain SOD and GSH-Px activity (but not the liver), while in the liver caused increased and in the brain decreased TBARS level. These results suggest that Se stimulates antioxidative enzymes in immature kidney and brain of Cd-exposed rats and could protect against oxidative damage.  相似文献

In clinical practice, selenium deficiency may arise under conditions of chronic malnutrition and especially after long-term total parenteral nutrition (TPN). In infants receiving long-term TPN, we observed plasma selenium levels as low as those previously reported in Chinese children with Keshan disease. Low plasma selenium levels were also usually associated with very low activities of glutathione peroxidase. Although clinical symptoms of selenium deficiency did not occur in our patients, several cases have been described in the literature, indicating the need for supplementation in TPN. In order to derive at the appropriate dosage, it is proposed to correlate it with the total protein supply. According to our present knowledge, .5–1.0 μg selenium/g of protein appears to be adequate to keep patients in Se balance. For Se repletion of body stores, this dosage has been increased up to 3 μg of Se/g of protein. Advantages and disadvantages of selenite and of selenomethionine as possible supplemental forms of Se for TPN solutions are discussed.  相似文献

More than 50% of Nepal’s population lives in the hill regions, usually with small land holdings, and over 60% are considered below the poverty line. Livestock, and particularly goats, provide these small-scale farmers with about 55% of their on-farm income. We studied goat production in Katteldanda, a mid-hill village of 78 households, mainly Brahmins, in Ghorka District. Subsistence farmers raise six or seven goats and one or two buffalo, and cultivate mainly maize, millet and rice. Tree fodder constituted about 70% of dry matter intake of goats for large parts of the year and was collected by lopping branches from trees on upland, rain-watered, private terraces (bari) near the household compound. Local farmers ranked the fodder trees they considered best and we measured the actual fodder they offered to goats. In addition, we evaluated and ranked 23 fodder species on the basis of laboratory in vitro nutritional and metabolizable energy yields. Using a Mantel test, a significant correlation was found between what the farmers thought was best fodder and fodder offered to goats (Mantel r = 0.398; P = 0.037) but non-significant correlations were found between either what the farmers thought was best fodder or fodder offered to goats and laboratory rankings (Mantel r = −0.027; P = 0.49 and Mantel r = 0.187; P = 0.18, respectively). We concluded that biomass produced and availability throughout the year, in addition to nutritional and energy yield, are important criteria for selecting fodder trees for goat production.  相似文献

The aim of this study was to determine the serum concentrations of selenium, vitamin E, and total- and lipid-bound sialic acid (LBSA) in lambs with white muscle disease (WMD) before and after treatment with a commercial preparation containing selenite and vitamin E. Fifteen lambs with WMD and ten control animals were used as research materials. Blood samples were collected from both groups before- and 1 month after treatment for Se analysis by fluorimetry, whereas vitamin E and sialic acid were measured by HPLC and spectrophotometry, respectively. Compared to controls, in the diseased animals, there was a significant increase of serum total sialic acid (TSA) and LBSA, together with significant decreases of serum Se and vitamin E concentrations (p < 0.001). One month after treatment, a reversal of trend was observed with decreases of TSA and LBSA and increases of Se and vitamin E concentrations. The TSA and LBSA levels, however, remained significantly higher than those of the controls, p < 0.05 and 0.001, respectively. The Se and vitamin E concentrations of the treated animals were the same as those of controls. This is the first study on total and LBSA concentrations in lambs with WMD, showing that these markers can be used in the prognosis of the disease.  相似文献
