Cholinergic synaptic vesicles isolated from the electric organ of Torpedo californica are confirmed to exhibit energy-linked uptake of [3H] acetylcholine. [3H]Acetylcholine is concentrated in the vesicles by a factor of 10–14 in the presence of MgATP and bicarbonate. This active uptake can be completely inhibited by the mitochondrial uncouplers 3-t-butyl-5-Cl-2′-Cl-4′-nitro-salicylanilide (S-13) and p-nitrophenol. The vesicle-associated ATPase is stimulated by S-13 in the same concentration range which inhibits [3H]acetylcholine active uptake. The ATPase also is stimulated by valinomycin. Both S-13 and valinomycin effects are independent of exogenous Ca2+. Thus, a proton gradient generated by the vesicle-associated ATPase appears to be coupled to active [3H]acetylcholine uptake.  相似文献

It is demonstrated that two classes of binding site for acetylcholine are present on Torpedocalifornica acetylcholine receptor. One class is the well documented site on each of the two subunits of 40,000 daltons, which can be covalently modified by bromocetylcholine. Both in the absence and in the presence of bromoacetylcholine another binding site is shown to exist by virtue of acetylcholine dependent fluorescence changes in the receptor covalently modified by 4-[N-(iodoacetoxy)ethyl-N-methyl]-amino-7-Nitrobenz-2-oxa-1,3 diazole (IANBD). This site has a low affinity for acetylcholine (Kd ~ 80 μM) that corresponds closely with the known concentration dependence of acetylcholine mediated activation of this receptor and we conclude that it may represent a site of association that participates in channel opening in this system.  相似文献

The effects of 1-methyladenine on oxygen consumption and adenine nucleotide levels were examined in oocytes of Pisaster ochraceus and Patiria miniata. Oocytes of both genera to which 1-methyladenine was added consumed more oxygen than control oocytes beginning 1 to 112h after 1-methyladenine addition. The increase in oxygen consumption was correlated with maturation changes in the oocytes and particularly with germinal vesicle breakdown. Pre-fertilization oxygen consumption of eggs did not differ significantly from post-fertilization oxygen consumption of eggs in either genus for 212h after fertilization. ATP and AMP concentrations within the oocytes decreased during 1-methyladenine induced maturation, while the ADP concentration increased. It was suggested that increases in ADP concentration and decreases in ATP concentration within maturing starfish oocytes occurred in response to greater energy demands. The simultaneous increase in oocyte oxygen consumption was interpreted as an indicator of increased oxidative phosphorylation acting to restore initial nucleotide concentrations.  相似文献

Biochemical and electrophysiological studies were conducted on the electric organ of the electric fish of the Nile, Malapteruruselectricus, in order to determine if transmission was chemically mediated. There was no binding of [3H] acetylcholine, [3H] quinuclidinyl benzilate or [3H]-perhydrohistrionicotoxin; but low acetylcholinesterase activity was observed, as was binding of [125I] α-bungarotoxin. The latter binding was detectable at 0.85 ± 0.07 pmol/g tissue, and was totally inhibited by 1 μM α-bungarotoxin or 100 μM d-tubocurarine. A tetrodotoxin-sensitive action potential was measured which was Na+- dependent. Depolarization (30–40 mV) was caused by carbamylcholine, and this was blocked by d-tubocurarine or α-bungarotoxin. The data suggest that this electric organ which may be a rich source for electrically excitable channels, is innervated by nicotonic cholinergic motoneurons, but the concentrations of acetylcholine receptors and acetylcholinesterase are very low.  相似文献

The nicotinic acetylcholine receptor from electrogenic tissue of Torpedo californica was solubilized by tryptic digestion of membrane fragments obtained from autolysed tissue, without use of detergent. The water-soluble acetylcholine receptor was purified by affinity chromatography on a cobra-toxin-Sepharose resin. The purified receptor bound 4000–6000 pmol per mg protein of α-[125]bungarotoxin, and toxin-binding was specifically inhibited by cholinergic ligands. Gel filtration revealed a single molecular species of Stokes radius 125 ± 10 Å and on sucrose gradient centrifugation one major peak was observed of 20–22 S. Polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate and β-mercaptoethanol revealed two major polypeptides of mol. wt. 30 000 and 48 000.  相似文献

Piribedil, (1–2″-pyrimidyl)-4-piperonyl piperazine), an agent proposed for the treatment of Parkinson's disease, was found to increase acetylcholine levels in the rat striatum and diencephalon but not in the mesencephalon, cerebellum or hemispheres. The effect was most marked in the striatum (greater than 100%) and long-lasting (at least 8 hours after a single administration of 60 mg/kg i.p.). Striatal choline levels were also increased by piribedil but did not parallel at all times and doses the effect on acetylcholine. Furthermore, choline levels were increased in all brain regions except the hemispheres. Striatal choline acetyltransferase and acetylcholinesterase were not affected by in vitro or in vivo treatment with even high doses of piribedil. α-Methyl-p-tyrosine was ineffective in blocking piribedil while pimozide, a blocker of dopamine receptors, completely antagonized the action of piribedil on striatal acetylcholine. It is concluded that piribedil produced the increase in striatal acetylcholine by directly stimulating dopamine receptors.  相似文献

γ-Aminobutyric acid (GABA), a known inhibitory neurotransmitter in mammals, can elicit two different types of excitatory response in the nervous system of the marine mollusc, Aplysia. These responses are depolarizing when GABA is applied ionophoretically, and result from either an increase in membrane conductance to Na+ or a decrease in conductance to K+. In addition, GABA on other neurons causes an inhibitory response similar to that commonly found in other preparations. Although not all neurons have GABA receptors, identified single cells consistently have the same type of response. These observations suggest the possibility that GABA may function in at least some preparations as an excitatory neurotransmitter in addition to its documented inhibitory function.  相似文献

Membrane preparations containing essentially only the four polypeptides considered to constitute the acetylcholine receptor are purified from Torpedocalifornica electroplax. Treatment of these membranes with 2% (wv aqueous sodium cholate followed by removal of all insoluble matter results in a solubilized purified receptor preparation that can be reassociated with phospholipids during dialysis to remove the detergent. Such reconstituted receptor is shown to retain the capability of translocating 22Na+ across the membrane in response to carbamylcholine binding in a highly reproducible manner. The dose response for this effect is similar to that observed for the original electroplax membrane preparation and the carbamylcholine induced signal is completely blocked by α-bungarotoxin.  相似文献

Oocytes of Platynereis dumerilii in early vitellogenesis were injected into female worms with oocytes of similar diameter. The donor oocytes were labeled by the or gene controlling eye pigmentation and, after some weeks of growth, were spawned together with the host oocytes. In most cases, a few donor progeny could be found among the offspring produced by the hosts. Donor progeny were examined with respect to an or gene-dependent maternal effect which normally causes wild-type eye color in homozygous (oror) larvae originating from the crossings of heterozygous (or+or) females and homozygous (oror) males. This maternal effect was absent from homozygous (oror) larvae derived from homozygous (oror) donor oocytes which had developed in heterozygous females. Conversely, this maternal effect was observed in homozygous (oror) larvae derived from heterozygous (or+or) donor oocytes which had developed in homozygous (oror) host females. It is concluded that the oocyte genome is active at the or+ locus during oogenesis and that the oocyte is autonomous with respect to the product of synthesis of the or+ locus. In the present case, the “maternal effect” is therefore caused by synthetic activity in the growing oocyte. The results are discussed with respect to current information on gene products from animal genomes.  相似文献

Closed membrane vesicles derived from the innervated face of Torpedocalifornica electroplax respond to the cholinergic agonist carbamylcholine by a rapid efflux of cations. This response is detected by release of 22Na enclosed within the vesicles and is considerably faster than previously reported for this invitro system. It is considered likely that the rapid response is analogous to physiological phenomena since it has the appropriate pharmacological characteristics; it desensitizes upon prolonged contact with the agonist and it has a dose-response curve in a physiological range. It is further shown that a dimeric form of the acetylcholine receptor, stabilized by chemical modification methods, is fully active in terms of the carbamylcholine elicited response.  相似文献

Rapid efflux of 22Na from within closed vesicles derived from Torpedocalifornica electroplax membranes has been studied as an invitro assay of acetylcholine receptor functionality. The most highly purified membrane preparations contained major polypeptides of M.W. 43 and 90 × 103 daltons in addition to the four peptides characteristic of the acetylcholine receptor (40, 50, 60, 65 × 103 daltons). Removal of these extra peptides by base extraction did not significantly alter the characteristics of carbamylcholine induced 22Na efflux: the agonist dose response curve was similar, preequilibration with agonist caused desensitization, the irreversible antagonist α-Bungarotoxin blocked the efflux and the reversible blockade by the neurotoxin perhydrohistrionicotoxin was also retained. The dose response curve for perhydrohistrionicotoxin corresponded closely to its known binding characteristics for base extracted membranes.  相似文献

Gaps in daughter-strand deoxyribonucleic acid (DNA) synthesized after exposure of wild-type Escherichiacoli to ultraviolet light are filled during reincubation. In this study the dnaG, dnaC, and dnaA gene products have been examined for their role in postreplication repair. These gene products are unique in their specific control of certain types of DNA synthesis: initiation of rounds of replication and chain propagation. Initiation of rounds of replication is not essential to gap filling; however, chain propagation by short DNA piece initiation appears to be essential for gap filling.  相似文献

Acetylcholine receptor-rich membranes from Torpedocalifornica contain a binding site for [14C] pentobarbital which has a dissociation constant of 210 ± 24 μM and 1.4 ± 0.18 sites per acetylcholine site. (+) pentobarbital competes for this site three times more effectively than (?) pentobarbital. Cholinergic ligands decrease [14C] pentobarbital binding and this effect is blocked by pre-incubation with α-bungarotoxin. Pentobarbital decreases [3H] acetylcholine binding non-competitively with an apparent dissociation constant similar to the dissociation constant for [14C] pentobarbital binding. Thus, the pentobarbital and acetylcholine binding sites appear to interact with each other allosterically.  相似文献

The antinociceptive effects of intraventricularly administered acetylcholine (ACh) and its congeners have been demonstrated by previous investigators. The opiate receptor binding concept was used in this study to investigate possible correlations between ACh antinociception and its effects on opiate stereospecific binding. ACh in vitro decreased the stereospecific binding of 3H-dihydromorphine in mouse brain homogenates. Such decrease was also observed in the brain homogenates of mice which had been treated with ACh intracerebroventricularly (i.v.t.). The decrease in the stereospecific binding of 3H-dihydromorphine induced by (i.v.t.) acetylcholine was inhibited by naloxone, atropine, cyclazocine and pentazocine. The d-isomers of cyclazocine and pentazocine were more potent than the l-isomers in antagonizing the inhibitory effects of i.v.t. acetylcholine upon the stereospecific binding of 3H-dihydromorphine to mouse brain homogenates. The same stereospecificity of these two narcotic analgesics in blocking acetylcholine had been previously observed in the tail-flick test. It is suggested that the antinociceptive effects of acetylcholine are related to the inhibition of opiate stereospecific binding, the mechanism of which is yet to be understood.  相似文献

The factors influencing the overall mobility of the major proteins of the acetylcholine receptor-rich membranes from Torpedo marmorata have been investigated by saturation transfer ESR spectroscopy and the lateral distribution of these proteins has been studied by electron microscopy. A spin-labelled derivative of maleimide, 3-maleimido-2,2,5,5-tetramethyl-1-pyrrolidinyloxyl (MSL), was used under various conditions of incubation, enabling us to attach it mainly to either an extrinsic protein of 43 kdaltons, or an intrinsic protein (40 kdaltons) bearing the α-toxin-binding site. (1) The direct reaction of MSL with the membrane fragments resulted in almost exclusive labelling of the 43 kdalton protein, an extrinsic protein located on the inner face of the receptor-rich membranes. (2) After the free SH groups were blocked with N-ethylmaleimide and the disulfide bridges opened with the reducing agent dithiothreitol, MSL reacted with both the 40 and 43 kdalton proteins (6.0±0.6MSL molecules per α-toxin-binding site). (3) After the latter labelling procedure membranes were exposed to pH 11, resulting in extraction of the 43 kdalton protein and leaving 2.2 ± 0.4MSL molecules per α-toxin-binding site; sodium dodecyl sulfate polyacrylamide gel electrophoresis performed with N-[14C]ethylmaleimide suggested that MSL was bound mainly to the 40 kdalton polypeptide chain of the acetylcholine receptor. The following conclusions were made with the native and alkaline-treated membranes: In the native membranes, saturation transfer ESR does not reveal any significant protein rotational diffusion (itrotational correlation time τc > 1 ms). Temperature variations and/or lipid modifications obtained by fusion of exogenous lipids and/or cholesterol exchange have little influence on the saturation transfer ESR spectra. Electron microscopy reveals that upon lipid addition, proteins remain in the form of clusters while areas depleted of proteins appear. On the other hand, alkaline treatment strikingly enhances the motion of the MSL-labelled proteins in the membrane (100 ? τc ? 120 μs). Furthermore, the rotational diffusion of the MSL-labelled proteins (mainly the 40 kdalton protein) becomes sensitive to temperature, lipid composition and the lipid-to-protein ratio. Electron microscopy shows that alkaline extraction does not cause large reorganization of the acetylcholine receptor in the plane of the membrane. However, when phospholipids are added to pH 11 treated membranes, a dispersion of the receptor rosettes is observed. In contrast, cholesterol enrichment of the latter membranes induces clustering of the receptor and immobilization as judged by saturation transfer ESR. Upon reassociation of the pH 11 soluble proteins with the alkaline-treated membranes, the restriction of the acetylcholine receptor rotational mobility is also restored (τc ? 1 ms).  相似文献

Thermotropic properties of purified cytochrome c1 and cytochrome c have been studied by differential scanning calorimetry under various conditions. Both cytochromes exhibit a single endothermodenaturation peak in the differential scanning calorimetric thermogram. Thermodenaturation temperatures are ionic strength, pH, and redox state dependent. The ferrocytochromes are more stable toward thermodenaturation than the ferricytochromes. The enthalpy changes of thermodenaturation of ferro- and ferricytochrome c1 are markedly dependent on the ionic strength of the solution. The effect of the ionic strength of solution on the enthalpy change of thermodenaturation of cytochrome c is rather insignificant. The formation of a complex between cytochromes c and c1 at lower ionic strength causes a significant destabilization of the former and a slight stabilization of the latter. The destabilization of cytochrome c upon mixing with cytochrome c1 was also observed at high ionic strength, under which conditions no stable complex was detected by physical separation. This suggests formation of a transient complex between these two cytochromes. When cytochrome c was complexed with phospholipids, no change in the thermodenaturation temperature was observed, but a great increase in the enthalpy change of thermodenaturation resulted.  相似文献

The amino terminal amino acid sequence of the 41,000 dalton subunit of Electrophoruselectricus acetylcholine receptor has been determined for 35 cycles by automated sequencing procedures. Comparison of the unique polypeptide sequence obtained for this molecule with that of the major subunit of Torpedocalifornica acetylcholine receptor reveals extensive primary structural homology between the two proteins.  相似文献

Isolated porcine Graafian follicles which were explanted in vitro and maintained in organ culture were used as a test-system for the meiosis-inducing action of PMSG and hCG. The addition of either PMSG or hCG alone (10 or 20 IU/ml, respectively) to the culture medium was not effective, whereas the simultaneous administration of these hormones (1515IU/ml) induced resumption of meiosis in 90.3% (3741). The same hormone concentrations were used in a second series of experiments in which the inhibition and induction of meiosis of isolated oocytes were tested by transferring them into host follicles. In host follicles containing up to 12 foreign eggs, which were cultured in control media, meiosis was prevented in 86.0% of all oocytes (104121). By adding PMSG (15 IU/ml) simultaneously with hCG (15 IU/ml) to the medium, meiosis was induced in 95.0% of all oocytes (133140).The assumption is made that the signal initiating resumption of meiosis of the isolated and transferred oocytes is mediated by the follicular fluid, since intimate contact with the membrana granulosa of the host follicle was prevented by using a roller technique.  相似文献

Adult male guinea pigs were pretreated with estrogen, progesterone, or estrogen plus progesterone and the in vitro contractile response of the gallbladder to cholinergic stimulation examined. The data were compared with results obtained from control animals. Progesterone pretreatment was associated with a significant decrease in the maximal contractile response of the tissues and with a significant increase in the dose of acetylcholine needed to produce a threshold response. Estrogen pretreatment significantly decreased the threshold dose requirements but had no effect on maximum tension development. In addition, estrogen pretreatment antagonized the inhibitory effect of progesterone pretreatment. The data support the hypothesis that the ovarian steroid hormones can affect gastrointestinal smooth muscle. Furthermore, the hormones appear to exert independent and opposite effects on gallbladder motility. Additional studies will be required to determine the physiological significance of these observations.  相似文献
