Corticosteroid (C) release by adrenals of male Mongolian gerbils (Meriones unguiculatus) has been studied during continuous and discontinuous (flow stop) superfusion. Flow stops of superfusion for 1, 5, 10 or 20 min resulted in a significant accumulation of C within adrenal tissue and superfusion flask. Amounts of C in the first 2-min samples after re-start of superfusion were positively correlated with the amounts secreted during continuous superfusion (5 min: r = 0.97, 10 min: r = 0.97, 15 min: r = 0.74, 20 min: r = 0.84, all p less than 0.001) and with the length of flow stops (1-20 min: r = 0.92, p less than 0.001). However, C concentrations in superfusates were significantly lower than values calculated from secretion during continuous superfusion and the length of flow stops (0 min = 100%, 1 min: 92%, 5 min: 65%, 10 min: 49%, 15 min: 39%, 20 min: 35%). As is evident from the very similar C amounts secreted by adrenals incubated for 15 min without or with 95%O2/5%CO2 (234 vs 256% of basal secretion), flow stop-induced inhibition of corticosteroidogenesis was not due to a lack of oxygen during flow stops. The results demonstrate that superfusion of sliced adrenal tissue gives insights into regulatory mechanisms, including the rapid changes of corticosteroidogenesis during short-lasting flow stops, which cannot be studied in static incubation of either tissue slices or isolated cells. The possibility that the observed decline in steroidogenesis during flow stops may be due to a local feedback inhibition resulting from C accumulating in the microenvironment of adrenal cells is discussed.  相似文献

Two opioid peptides, beta-endorphin and dynorphin1-17 were bioassayed with isolated rat adrenocortical cells. beta-Endorphin increases basal production of corticosterone as well as the adrenal responsiveness to low doses of ACTH, these effects being partially reversed by naloxone. Dynorphin1-17, without affecting basal corticosterone synthesis, increases adrenocortical responsiveness to ACTH; naloxone does not influence this effect. It is suggested that peripheral opioid peptides may participate in the maintenance of the homeostatic balance by modulating adrenal corticosteroidogenesis.  相似文献

The cardiovascular teratogenicity of nimustine hydrochloride (ACNU) was studied in rat fetuses. This drug is a nitrosourea derivative anticancer agent and produces alkylation of DNA. Pregnant Donryu rats were treated with single doses of 10, 11 or 13 mg/kg of the teratogen at various stages during gestation. Examination of the hearts was performed by microdissection after sacrificing the animals on the 20th day of gestation. The highest frequency of cardiovascular anomalies was found in the groups treated on the 8th day of gestation, but there was no difference in the rates induced by the three dosages of ACNU administered. The most common cardiovascular anomalies observed were ventricular septal defect (76.8%) and double outlet right ventricle (10.3%). A considerable number of affected fetuses (37/263) showed complex cardiac anomalies with atrioventricular (AV) malalignment and other AV valve anomalies. These anomalies include: double inlet left ventricle, straddling AV valve, atresia or stenosis of the AV valve, and dysplastic AV valve. ACNU appears to be a useful teratogenic agent for inducing complexes of cardiac anomalies which include AV malalignment.  相似文献

Cytosol from the adrenal gland of male and female rats contains a specific binding protein for oestradiol-17β. This protein has all the characteristics of a cytoplasmic oestrogen receptor. It is excluded by Sephadex G-200 gel filtration, has a sedimentation coefficient of 8–9 S by sucrose density gradient centrifugation in low salt and dissociates into a 4 S form by centrifugation in high salt (0.5 M KCl). The binding protein is heat sensitive and oestradiol-17β binding is eliminated by protease and by sulphydryl blocking reagents (2mM p-chloromercuriphenylsulphonate). The bound oestradiol dissociates very slowly at 0°C. The adrenal oestrogen receptors have a very high affinity for oestradiol-17β, but lower affinity for oestradiol-17α and do not bind testosterone, androstene-3,17-dione or corticosterone. Scatchard analysis of the saturation data for oestradiol revealed one class of high affinity binding sites with an apparent equilibrium constant of dissociation KD at 0°C of 5.8 × 10−10M. The number of binding sites was calculated to be 70 fmol/mg cytosol protein. Cytosol fractions from androgen insensitive (tfm) male rats contain oestrogen receptors in amounts very similar to that of the normal littermates.  相似文献

The constituents of the intermediate filament network of adrenal gland cells have not been deeply investigated in vivo. Adrenocortical cells have been reported to express cytokeratins and vimentin, but the intermediate filament components of the adrenomedullary cells are still unknown. Nestin is an intermediate filament protein that is mainly expressed in the developing nervous and muscle systems. It has been reported to be unable to form filaments by itself and it co-assembles with vimentin. Using immunocytochemical and biochemical approaches, the present study demonstrates that nestin is expressed in situ either in the cortex or in the medulla of adult rat adrenal glands. Nestin-negative cells prevalently form the zona glomerulosa whereas the zona fasciculata and the zona reticularis are mainly nestin-immunoreactive. Nestin-positive cells always express vimentin-like immunoreactivity but several cells apparently expressing only vimentin are detectable too. Nestin is also expressed by adrenomedullary cells that also display a faint vimentin-like immunoreactivity. We hypothesise that the inconstant detection of nestin in adrenocortical cells depends on their different functional moments. Moreover, even though our data do not allow to confirm vimentin in adrenomedullary cells, in situ detection of nestin in the adrenal medulla indirectly supports in vivo expression of vimentin in chromaffin cells.  相似文献

Rat adrenal gland preparations were incubated with radioactive cholesterol and 23,24-dinor-5-cholen-3 beta-ol. Steroids were isolated, purified by thin-layer and high performance liquid chromatography and crystallised to constant specific activity. It was found that the rat adrenal gland can utilise 23,24-dinor-5-cholen-3 beta-ol to produce corticosterone. Also, in contrast to the conversion of cholesterol to corticosterone which occurs in the mitochondrial fraction, the conversion of 23,24-dinor-5-cholen-3 beta-ol to corticosterone occurs in the microsomal fraction. It was concluded that the sesterterpene pathway for steroid biosynthesis can function in the rat adrenal gland and that the intermediates are converted to steroid hormones in the microsomal fraction.  相似文献

A series of betamethasone 17alpha-carbamates were designed, synthesized, and evaluated for their ability to dissociate the two main functions of the glucocorticoid receptor, that is, transactivation and transrepression, in rat cell lines. A number of alkyl substituted betamethasone 17alpha-carbamates were identified with excellent affinity for the glucocorticoid receptor (e.g., 7, GR IC(50) 5.1 nM) and indicated dissociated profiles in functional assays of transactivation (rat tyrosine aminotransferase, TAT, and rat glutamine synthetase, GS) and transrepression (human A549 cells, MMP-1 assay). Gratifyingly, the in-vivo profile of these compounds, for example, 7, also indicated potent anti-inflammatory activity with impaired effects on glucose, insulin, triglycerides, and body weight. Taken together, these results indicate that dissociated glucocorticoid receptor modulators can be identified in rodents.  相似文献

 The detailed distribution and heterogeneity of various immunocompetent cells were characterized in the normal adrenal gland of the rat, with special emphasis on major histocompatibility complex (MHC) class II-expressing cells and macrophages. All adrenals contained at least two different populations of cells reactive with the dendritic cell or the macrophage antibodies. These cells were clearly distinguished from adrenal parenchymal cells by their morphology and location. The majority of dendritic cells were immunoreactive for the MHC class II (Ia) antigen (MRC OX6) and/or the dendritic cell antibodies (MRC OX62), and negative for the macrophage antibodies (ED1, ED2, and/or MRC OX42), whereas the main population of macrophages was immunonegative for the former antibodies and positive for the latter. The OX62-positive cells and the OX42-labeled cells occurred exclusively throughout the medulla. The cellular density of dendritic cells in the adrenal cortex was significantly higher than that of macrophages. Double-immunoperoxidase staining for ED1 and OX6 revealed that positively stained cells could be classified into the following categories: ED1+OX6+, ED1+OX6, and ED1OX6+. More then 40% of OX6+ cells were immunoreactive for ED1 in the zona glomerulosa, while approximately 15%, 20%, and 30% of OX6+ cells were positive for ED1 in the zona fasciculata, zona reticularis and medulla, respectively. ED1+ED2 cells were more frequently detected in the zona glomerulosa than in other adrenal zones. Only a few ED1ED2+ cells were located in the zona glomerulosa, whereas a large number of them were found in the zona fasciculata. In the zona reticularis and medulla, ED1+ED2+, ED1+ED2, and ED1ED2+ cells were detected in the ratio 2:1:3. Our rsults suggest that dendritic cells and macrophages mature during their migration within the adrenal gland. These immunocompetent cells may contribute to a paracrine regulation of adrenal function under physiological conditions. Accepted: 3 November 1997  相似文献
