We previously showed (Science 198:1178, 1977) that fenfluramine or fluoxetine, drugs thought to enhance serotoninergic transmission, selectively decrease the rat's consumption of carbohydrates, without affecting protein intake, when animals are given simultaneous access to diets differing in protein (5% vs 45%) and carbohydrate contents; in contrast, d-amphetamine lacks this selective effect. Present studies affirm this relationship using another serotoninergic drug, MK-212, and show that the suppression of carbohydrate intake occurs whether the test diets contain variable amounts of protein, or carbohydrates, or of both nutrients. Evidence is presented that rats given diet mixtures containing various proportions of carbohydrates have the ability to regulate their carbohydrate intakes (i.e., to choose to eat amounts of each diet that, taken together, will give them a desired proportion of carbohydrate), and that this ability is independent of whether or not the carbohydrate consumed has a sweet taste. It is proposed that serotoninergic neurons comprise part of a behavioral feedback loop, whereby the consumption of carbohydrate (which, by altering plasma amino acid patterns, accelerates serotonin synthesis in brain neurons) diminishes the rat's subsequent tendency to consume additional carbohydrates. Drugs that enhance central serotoninergic transmission can probably be substituted for dietry carbohydrate to activate this behavioral loop.  相似文献

The effects of four different diets (control diet: 19.5% protein, 60.5% carbohydrate, 10% fat; diet I: 65% protein, 10% carbohydrate, 10% fat; diet II: 5% protein, 76% carbohydrate, 10% fat; diet III: 20% protein, 69% carbohydrate, 1% fat; diet IV: 69% protein, 15% carbohydrate, 1% fat) and supplementation with 3 amino acids (tryptophan: 150 mg/kg/d; arginine: 400 mg/kg/d; taurine: 380 mg/kg/d) on the voluntary consumption of ethanol were investigated in rats using the 2 bottle method. First, rats received the control diet and diets I, II, III and IV for 20 days with a choice of ethanol for the last 6 days only. Ethanol consumption was similar in all dietary groups. Second, rats received the control diet for 8 days followed by diets I, II and IV for another 8 days. Ethanol was offered throughout both periods. The switch to the special diets did not affect ethanol consumption. Third, rats received a control diet with arginine, tryptophan or taurine added to the drinking fluids for 16 days with a choice of ethanol for the last 5 days; thereafter supplementation stopped but the ethanol choice remained. No difference in the voluntary intake of ethanol was noted but ethanol consumption fell after cessation of arginine supplementation. In conclusion, diets differing greatly in their composition or supplementation with these 3 amino acids did not affect the voluntary choice of ethanol by rats in a significant manner.  相似文献

The effects of five anorexic agents on food consumption were tested in rats offered single, isocaloric, isonitrogenous diets differing in carbohydrate content. Three of the test agents, damphetamine, benzphetamine and chlorphentermine, are sympathomimetic and cause CNS stimulation; the others, MK-212 and d-fenfluramine, are thought to facilitate serotonin-mediated neurotransmission. At ED50 doses, the sympathomimetic drugs reduced food consumption whether the test diet was rich (75% dextrin) or poor (25% dextrin) in carbohydrate. In contrast, MK-212 and d-fenfluramine failed to reduce consumption of the 25% dextrin test diet. These observations suggest that anorexic drugs like d-amphetamine and d-fenfluramine do not act via a common “amphetamine receptor,” and are compatible with earlier observations, made on rats given diet pairs simultaneously, that enhanced serotoninergic neurotransmission selectively suppresses appetite for carbohydrates.  相似文献

Norepinephrine (NE) injected into the paraventricular nucleus (PVN) of the hypothalamus of rats is a potent stimulant of food intake, more specifically ingestion of the carbohydrate nutrient. In 2 experiments of the present study, this effect was found to be dose-dependent, and the effectiveness of NE in potentiating total food consumption was greatly reduced when the carbohydrate diet was removed. In addition, experiments using a computer-automated data acquisition apparatus were performed to characterize, in detail, the impact of PVN injection of NE and peripheral administration of the α2-nor-adrenergic agonist clonidine (CLON) on the macrostructure of feeding behavior in animals given 3 pure macronutrient diets. These 2 compounds, injected at the onset of the nocturnal feeding cycle, had very similar effects on meal patterns, with both affecting nutrient intake by increasing meal size and duration rather than by increasing meal frequency. They both affected primarily the first meal of the dark cycle, selectively enhancing carbohydrate ingestion by increasing Kcal intake, percent composition in the total diet and feeding time, and also by decreasing the satiating impact of this macronutrient. These stimulatory effects of NE and CLON on carbohydrate ingestion during the first meal were followed by complete recovery over the next 1 to 2 hours after injection. In addition to these predominant effects on carbohydrate intake, PVN NE at the highest doses tested (10 and 20 nmoles) produced a small increase in fat intake, whereas peripheral CLON actually decreased intake of fat and protein over the 12-hour cycle. The similarities in the impact of NE and CLON on carbohydrate feeding patterns support the hypothesis that both agonists may be acting via the same PVN α2-noradrenergic system controlling ingestion of the carbohydrate-rich meals which predominate at dark onset.  相似文献

Objective: The Protein‐Leverage Hypothesis proposes that humans regulate their intake of macronutrients and that protein intake is prioritized over fat and carbohydrate intake, causing excess energy ingestion when diets contain low %protein. Here we test in a model animal, the mouse: (i) the extent to which intakes of protein and carbohydrate are regulated; (ii) if protein intake has priority over carbohydrates so that unbalanced foods low in %protein leads to increased energy intake; and (iii) how such variations in energy intake are converted into growth and storage. Methods and Procedures: We fed mice one of five isocaloric foods having different protein to carbohydrate composition, or a combination of two of these foods (N = 15). Nutrient intake and corresponding growth in lean body mass and lipid mass were measured. Data were analyzed using a geometric approach for analyzing intake of multiple nutrients. Results: (i) Mice fed different combinations of complementary foods regulated their intake of protein and carbohydrate toward a relatively well‐defined intake target. (ii) When mice were offered diets with fixed protein to carbohydrate ratio, they regulated the intake of protein more strongly than carbohydrate. This protein‐leverage resulted in higher energy consumption when diets had lower %protein and led to increased lipid storage in mice fed the diet containing the lowest %protein. Discussion: Although the protein‐leverage in mice was less than what has been proposed for humans, energy intakes were clearly higher on diets containing low %protein. This result indicates that tight protein regulation can be responsible for excess energy ingestion and higher fat deposition when the diet contains low %protein.  相似文献

The short-term effects of diet on jejunal growth, alanine transport rate, and leucine aminopeptidase activity (LAP) were compared in the domestic and wild turkey poult. One-day-old poults of each strain were fed diets of high vs., low protein, with carbohydrate varied to maintain isocaloric conditions. Prior to feeding, relative jejunal mass and alanine transport rates were not significantly different in the two turkey strains, whereas LAP activity was 270% higher in wild poults. After feeding for 72 h, relative jejunal mass doubled in both turkey strains. In domestic turkeys, alanine transport rate and LAP activity were reduced by approximately 42% and 25%, respectively, in poults fed a 24% protein-69% carbohydrate diet vs. a 49% protein-35% carbohydrate diet. Analysis of the combined data from feeding experiments revealed that alanine transport rate was not correlated with total food, protein or lipid intake, but was negatively correlated with carbohydrate consumption (P<0.05). In wild turkeys, neither alanine transport rate nor LAP activity were altered by diet. These results reveal that domestic turkey hatchlings can modulate protein digestive and absorptive functions as protein/carbohydrate composition of the diet changes and suggest that high dietary carbohydrate down-regulates the intestinal alanine transporter.  相似文献

Although it has been known for more than 50 years that zinc (Zn) deficiency regularly and consistently causes anorexia in many animal species, the basic mechanism(s) that cause this phenomenon still remain(s) an enigma. The following studies describe feeding behavior in the early stages of zinc deficiency in the rat model. In one experiment, we used computerized feeding monitors that measured the intake of individual rats at 10-min intervals over 24-hr periods. Male rats were acclimated to the cages and fed a Zn-adequate egg-white-based diet, or a similar diet with <1.0 mg Zn/kg. Food intake was monitored for seven, consecutive 24-hr periods. The 24-hr food intake pattern of the Zn-deprived rats did not differ from the controls; they simply ate less food, mainly during the night hours, with no differences between groups during the day. Although Zn-deprived rats ate less food than controls, the percentage of total diet consumed during night and day did not differ between groups. In another experiment, we simultaneously offered male rats three isocaloric diets with different macronutrient compositions and with or without adequate Zn, and measured the amount of each diet selected during seven, 24-hr periods. The three diets contained either 57% protein from egg white, 30% fat from soybean oil, or 80% carbohydrate from a combination of starch, hydrolyzed starch, and sucrose. For the first four days on experiment, rats selected similar amounts of each diet. Then the Zn-deprived rats began to select only 50% as much of the protein diet as the controls. Similar results were obtained when the data were expressed on the basis of each macronutrient as a percentage of the total diet selected. Zn-deprived rats selected a diet that contained 8% protein, 73% carbohydrate, and 6% fat while the Zn-adequate rats selected 12% protein, 69% carbohydrate, and 6% fat. Fat intake was not affected by Zn-deprivation. The results confirm our previous findings, and are discussed in terms of Zn-deprivation blunting the pathways of signal transduction that involve the peptide hormones known to affect food intake regulation.  相似文献

Individual grass carp, Ctenopharyngodon idellu , were maintained in a respirometer for a month and fed pelleted diets containing various proportions of carbohydrate, fat and protein at different ration levels. Oxygen consumption was measured continuously, allowing the effects of consecutive daily feeding on respiration to be studied. The relationships established between daily food intake and oxygen consumption showed that, on average, 23.3% (high protein diet), 15.3% (high carbohydrate diet), 20.7% (high lipid diet) and 7.0% ( Lemnu diet ) of the absorbed energy was partitioned into specific dynamic action (SDA). (Here the term SDA is used to describe the oxygen consumption of a feeding fish in excess of the routine metabolic rate.) In terms of the overall energy budgets of growing fish, SDA represented between 12 and 58% of the total heat lost over the experimental period and was equivalent to between 14 and 33% of the consumed energy. Ration was positively correlated with heat loss due to total respiration ( r = 0.881) and with heat loss due to SDA ( r = 0.762). As ration increased, the size of SDA relative to total respiration increased. Significant positive correlations were found between oxygen consumption (total or due to SDA) and specific growth rate, and between oxygen consumption and the deposition of protein and energy. However, growth rate had a minimal influence on daily oxygen consumption when compared with food intake.  相似文献

The effects of acute (2 day) and chronic (15 day) administration of desipramine (DMI) on clonidine-induced suppression of exploratory behavior was investigated in the rat. In accordance with previous results, low doses (25–100 μg/kg) of clonidine produced a dose-dependent decrease in explaratory activity in control animals. This effect of clonidine was markedly attenuated in the chronic DMI groups. Acute treatment with DMI was without effect. The results provide behavioral support for recent biochemical and neurophysiological data which have suggested that chronic DMI administration results in subsensitivity of presynaptic receptors on central noradrenergic neurons.  相似文献

Agouti-related peptide (AgRP) is a receptor antagonist of central nervous system (CNS) melanocortin receptors and appears to have an important role in the control of food intake since exogenous CNS administration in rats and overexpression in mice result in profound hyperphagia and weight gain. Given that AgRP is heavily colocalized with neuropeptide Y (NPY) and that orexigenic effects of NPY depend on activity at opioid receptors, we hypothesized that AgRP's food-intake effects are also mediated by opioid receptors. Subthreshold doses of the opioid receptor antagonist naloxone blocked AgRP-induced intake when given simultaneously but not 24 h after AgRP injection. Opioids not only influence food intake but food selection as well. Hence, we tested AgRP's effect to alter food choice between matched diets with differing dietary fat content. AgRP selectively enhanced intake of the high-fat but not the low-fat diet. Additionally, AgRP selectively increased chow intake in rats given ad libitum access to a 20% sucrose solution and standard rat chow. The current results indicate that AgRP influences not only caloric intake but food selection as well and that the early effects of AgRP depend critically on an interaction with opioid receptors.  相似文献

We examined dietary self‐selection and rules of compromise for protein (P) and digestible carbohydrate (C) intake by fifth‐instar Vanessa cardui L. (Lepidoptera: Nymphalidae: Nymphalini). We presented six fat‐free diet pairs to larvae in a choice trial to determine the ‘intake target’. In addition, we fed larvae seven fat‐free single diets differing in dietary nutrient ratio in no‐choice trials to determine the rules of compromise they exhibit when constrained to a singular, sub‐optimal dietary source. In choice trials, caterpillars regulated nutrient intake to a ratio of 1 protein to 1.09 carbohydrate (1P:1.09C), exhibiting tighter regulation of protein than of carbohydrate. Furthermore, larvae from different diet pair treatments did not differ in pupal mass or stadium duration. In no‐choice experiments, larvae reduced consumption on increasingly protein‐biased diets and increased consumption on increasingly carbohydrate‐biased diets, relative to a 1P:1C ratio diet. Differences in carbohydrate consumption were much greater between no‐choice treatments than differences in protein consumption. Dietary nutrient ratio affected pupal mass when accounting for initial larval mass. Pupal mass decreased as nutrient ratio was shifted off of 1P:1C, but to a greater extent when the ratio was skewed toward carbohydrate. Stadium duration increased as nutrient ratio diverged from 1P:1C, being more pronounced when shifted toward carbohydrate than toward protein. Regulation to near 1P:1C is consistent with results found for other Lepidoptera, and the rule of compromise exhibited by V. cardui is consistent with that expected for a generalist herbivore.  相似文献

Relationships among feeding paradigm (single diet vs food selection) and arterio-venous differences (δAV) of glucose, insulin and tryptophan were studied by measuring the temporal patterns of food intake and plasma parameters during 8 hr feeding cycles in rats. Adult male Sprague-Dawley rats were offered a single diet of fixed composition (20% casein) or a choice between two isocaloric diets (0% and 60% casein) for 2 weeks under 8-hr daily feeding conditions, food being offered during the dark cycle. Groups of animals were then killed at the beginning and at 2-hourly intervals throughout the feeding period. With both feeding paradigms, rats showed temporal patterns of energy, carbohydrate and protein intakes with a peak at the beginning and a trough at the end of the feeding period. However, in rats offered a dietary choice the intake of carbohydrate was significantly lower, and the intakes of energy and protein significantly higher than those found in rats offered a single diet. Throughout the feeding period, these differences between single and choice diets became less accentuated in the case of carbohydrate intake, but more accentuated for energy and protein intakes. Paradoxically, rats fed a choice of diets had a significantly lower weight gain than rats fed a single diet. The temporal variation of insulin secretion and tryptophan absorption varied inversely with the two diet paradigms. Moreover, in rats offered a choice of diets, macronutrient intake was significantly correlated with insulin secretion and venous glucose concentration. The opposed physiologic and metabolic responses to the feeding paradigms suggest the need for future studies to examine the possibility that such can function as synchronizers of biological rhythms.  相似文献

The purpose of this study was to explore the effect of acute mild stress (12–48 hour food and water deprivation) and acute severe stress (12 hour food and water deprivation followed by 10 min swim in water at 4°) on the intake of different isocaloric dietary regimes. Each group of experimental animals was given only one particular diet. Rats subjected to mild stress showed very little preference of dietary regimes. When the food intake was measured during 3 hour period, following 48 hours of fasting, animals showed 2 to 3 fold increase in the food and water intake but no particular dietary preference. However, when rats were subjected to severe stress, there was an increase in the food intake of 154% (control diet); 174% (high-carbohydrate diet); 310% (high protein diet) and 423% (high fat diet) compared to animals subjected to mild stress. In terms of the absolute quantity of food, the animals subjected to severe stress ate more high-fat diet than any other diet; the consumption of high fat diet was 142% more than high-protein diet, 180% more than control diet and 258% more than high carbohydrate diet. Animals subjected to severe stress and given high-carbohydrate and high fat diet also showed 80% increase in the water intake. Prior administration of naloxone (1 mg/kg body weight, i.p.) reduced the stress induced increase in the intake of food and water. Naloxone inhibited the intake of high-fat diet more than any other diet. The ability of naloxone to block the increase in the intake of high-fat diet, and the reported increase in the concentration of β-endorphin in the different regions of brain of the animals subjected to the cold swim, suggest that endogenous opioid system in body is activated during stress. An activation of the endogenous opioid system leads to a preferential increase in the intake of palatable foods.  相似文献

Diets with restricted energy or protein during lactation programs body weight in the adult offspring. We have investigated the hypothesis that protein or energy-restricted diets during lactation alter the feeding response to peripheral leptin treatment of the adult offspring. Five Wistar rats were randomly assigned to one of the following groups on the day that the offspring were born: C, control diet with 23% protein; PR, protein restricted diet with 8% protein; and ER, energy-restricted, receiving the control diet in restricted quantities, which were calculated according to the mean ingestion of the PR group. After weaning (day 21), two animals from each litter (10 pups in each group) were randomly selected and placed together in the cage with free access to water and standard diet until 150 days of age, when they were tested for its response to either leptin (0.5 mg/kg body wt ip) for groups Clep, PRlep and ERlep or saline vehicle for groups Csal, PRsal and ERsal on food intake. In the control groups, food intake was reduced two hours (36%), four hours (41%) and six hours (25%) after leptin treatment. In contrast, no response was observed to leptin treatment in the PRlep and ERlep groups, suggesting leptin resistance. We demonstrated the development of resistance to the anorectic leptin effect and its program in a critical life period associated to nutritional and hormonal factors.  相似文献

SINGER, LORI K, DAVID A YORK, GEORGE A BRAY. Feeding response to mercaptoacetate in Osborne-Mendel and S5B/PL Rats. The purpose of this experiment was to determine if Osborne-Mendel (OM) rats, which are susceptible to dietary-induced obesity, and S5B/PL (S5B) rats, which are resistant to dietary-induced obesity, differ in their feeding responses to mercaptoacetate (MA), which blocks fatty acid oxidation, or 2-deoxy-D-glucose (2DG), which blocks glucose utilization. 2DG (100 mg/kg or 200 mg/ kg) increased food intake in both strains of rats on a high-fat diet (56% energy from fat). Mercaptoacetate (600 umol/kg) increased food intake in OM but not S5B rats on a high-fat diet. When maintained on a low-fat diet (10% energy from fat), MA (400 umol/kg or 600 umol/kg) stimulated food intake in OM rats, whereas S5B rats increased food intake only after the highest dose of MA (600 umol/kg). MA stimulated carbohydrate and protein intake in OM rats maintained on a macro-nutrient selection diet, whereas S5B rats maintained on this diet did not significantly increase intake of any mac-ronutrient after MA. These results demonstrate that OM and S5B rats have a similar food intake response to 2DG but a dissimilar response to MA. The variable response to MA in these strains may be due to a difference in peripheral or central signaling systems related to fatty acid oxidation or a difference in metabolic environments between the strains, which in turn affects the feeding response to MA. These studies suggest that a difference in control of fatty acid oxidation may account for the difference in susceptibility to obesity when eating a high-fat diet.  相似文献

In a 14-day experiment, weaned and adult rats were given ad libitum isocaloric diets with a mounting casein content (5, 10, 15, 25 and 40% by weight) and growth parameters of protein biological value, PER and NPR, and the utilization parameters NPU (body protein) and LPU (liver protein) were determined together with phosphoenolpyruvate carboxykinase (gluconeogenetic enzyme) and pyruvate kinase (glycolytic enzyme) activity in the animals' liver. The decrease in all the biological value parameters in weaned rats on 25% and 40% casein diets and in adult rats on 15%, 25% and 40% casein diets shows that these concentrations are too high for the organism. The decrease in PER and diminished weight and body and liver nitrogen increments in both age groups in animals with a low protein intake is evidence that 5% casein is an inadequate concentration. The optimum diet for weaned rats is thus a 15% casein diet and for adult rats a 10% casein diet, as confirmed by the linear correlation between weight increments, body and liver nitrogen and protein intake and also by gluconeogenetic enzyme activity. Under the given experimental conditions the study is a contribution to the determination of optimum physiological doses of proteins.  相似文献

Hypoglycaemia of varying duration was induced with graduated doses of long-acting insulin (10, 40 and 160 U/kg b.w.) in young, growing (8- to 10-week-old) male Wistar rats fed ad libitum on two different diets. In all, 6 doses of insulin were administered, at 48-hour intervals. The blood glucose level and food consumption were determined 3, 6, 12, 24 and 48 hours after injecting the insulin. Both the hypoglycaemic effect and elevated food intake during hypoglycaemia depended on the dose of insulin and not on the type of diet. Total food consumption during the experiment (12 days) was markedly raised only in the groups given the maximum dose of insulin (160 U/kg b.w.). The weight gain in all the experimental groups was lower than in the control groups and food consumption per weight gain unit was higher.  相似文献

Animals balance their intake of specific nutrients, but little is known about how they do so when foraging in an environment with toxic resources and whether toxic foods promote adaptations that affect life history traits. In German cockroach (Blattella germanica) populations, glucose aversion has evolved in response to glucose‐containing insecticidal baits. We restricted newly eclosed glucose‐averse (GA) and wild‐type (WT) female cockroaches to nutritionally defined diets varying in protein‐to‐carbohydrate (P : C) ratio (3 : 1, 1 : 1, or 1 : 3) or gave them free choice of the 3 : 1 and 1 : 3 diets, with either glucose or fructose as the sole carbohydrate source. We measured consumption of each diet over 6 days and then dissected the females to measure the length of basal oocytes in their ovaries. Our results showed significantly lower consumption by GA compared to WT cockroaches when restricted to glucose‐containing diets, but also lower fructose intake by GA compared to WT cockroaches when restricted to high fructose diets or given choice of fructose‐containing diets. Protein intake was regulated tightly regardless of carbohydrate intake, except by GA cockroaches restricted to glucose‐containing diets. Oocyte growth was completely suppressed in GA females restricted to glucose‐containing diets, but also significantly slower in GA than in WT females restricted to fructose‐containing diets. Our findings suggest that GA cockroaches have adapted to reduced diet breadth through endocrine adjustments which reduce requirements for energetic fuels. Our study illustrates how an evolutionary change in the chemosensory system may affect the evolution of other traits that govern animal life histories.  相似文献

Previous work from our laboratory indicates that when rats are given a choice between a high-fat and a high-sucrose diet, opioid blockade with naltrexone (NTX) in a reward-related site (central amygdala) inhibits intake of the preferred diet only, whereas NTX injected into a homeostasis-related site, such as the hypothalamic paraventricular nucleus (PVN), inhibits intake of both diets. However, other work suggests that opioids increase intake of fat specifically. The present study further investigates the role of PVN opioids in food choices made by calorically-replete animals. We used a binge model with chow-maintained rats given 3-h access to a choice of a high-fat or high-sucrose diet 3 days a week. We hypothesized that intra-PVN injection of the mu-opioid agonist, DAMGO (0, 0.025, 0.25, and 2.5 nmol) would enhance, and NTX (0, 10, 30, and 100 nmol) would inhibit intake of both diets to an equal extent. We found that when animals were divided into groups according to sucrose or fat preference, DAMGO increased fat intake in fat-consuming animals, while having no effect on intake of either diet in sucrose-consuming animals. NTX, however, inhibited fat intake in both groups. Intra-PVN NTX did not inhibit intake of sucrose when presented in the absence of a fat choice, but did so when injected peripherally. Furthermore, intra-PVN and systemic NTX inhibited intake of chow by 24-h-food-deprived animals. These results indicate a complex role for PVN opioids in food intake with preference, nutrient type, and energy state affecting the ability of these compounds to change behavior.  相似文献
