The mixture of tertiary and quaternary alkaloids isolated from Argemone ochroleuca was separately assessed against spore germination of some plant pathogenic fungi, e.g. Alternaria alternata, Alternaria brassicae, Alternaria cajani, Bipolaris sp., Curvularia lunata, Curvularia sp., Colletotrichum musae, Fusarium udum, Helminthosporium sp., Helminthosporium pennisetti and Helminthosporium speciferum. Spore germination of Fusarium udum and Helminthosporium sp. was completely inhibited at very low concentration (200 ppm). A similar effect was observed on A. alternata, C. musae and H. pennisetti at 600, 800 and 1000 ppm. With quaternary alkaloids, Curvularia sp. and Colletotrichum musae were most sensitive as complete inhibition of spore germination was observed at 400, 600, 800 and 1000 ppm and a similar effect was observed with A. brassicae and A. cajani at 600, 800 and 1000 ppm. The remaining fungi were also highly sensitive to the mixture at different concentrations.

Berberine and (±)-bicuculline were isolated from roots and leaves, respectively, ofCorydalis chaerophylla. Both were effectivein vitro against spore germination of some plant pathogenic fungi (Alternaria brassicicola, A. brassicae, A. cheiranthi, A. melongenae, A. solani, Colletotrichum musae, C. falcatum, Curvularia penniseti, C. lunata, C. maculans, C. pallescens, Curvularia sp.,Erysiphe pisi, E. cichoracearum, Erysiphe sp.,Fusarium udum, Helminthosporium spiciferum, H. penniseti, H. frumentacei, Heterosporium sp.,Oidium erysiphoides andUstilago cynodontis). Berberine and (±)-bicuculline significantly inhibited spore germination of all the fungi at concentrations of 100–1000 ppm. Berberine was effective against all the fungi at all concentrations; most of the fungi did not germinate at 1000 ppm.H. penniseti conidia did not germinate at any cencentration of (±)-bicuculline.U. cynodontis was the least sensitive fungus at lower concentrations but 800 ppm dose was highly effective.

The isoquinoline alkaloids hunnemanine and norsanguinarine have been isolated from methanolic extract of the whole plant of Eschscholtzia californica. These two alkaloids were checked for their antifungal activity against phytopathogenic fungi Alternaria melongenae, A. brassicola, A. brassicae, Curvularia lunata, C. maculans, Helminthosporium pennisetti, H. oryzae, H. turcicum, Fusarium undum and F. lini. Hunnemanine exhibited 100 % inhibition of spore germination of A. brassicae, H. pennisetti and F. lini at 1000 ppm whereas norsanguinarine exhibited 100 % inhibition of A. brassicicola and C. maculans at this concentration.

(+/-)-alpha-Hydrastine and (+/-)-beta-hydrastine were isolated from Corydalis longipes; both exhibited considerable efficacy against spore germination of some saprophytic and phytopathogenic fungi. While (+/-)-alpha-hydrastine was effective against most of the fungi, Helminthosporium echinoclova was least affected even at the highest dose (150 ppm). (+/-)-beta-Hydrastine was equally effective against several fungi. Mixture of both compounds was more effective than each one individually. Helminthosporium species were again the most resistant toward the mixture. The effect of both alkaloids independently on germination and development of E. pisi conidia on excised pea leaves was also shown. After pre-inoculation with (+/-)-alpha-hydrastine, the effect was more pronounced than the addition post-inoculation; maximum inhibition occurred at 200 ppm. (+/-)-beta-Hydrastine also reduced germination of conidia but was less effective than (+/-)-alpha-hydrastine. The number of primary and secondary branches of conidia and number of appressoria were not affected significantly by either compound.

Berberine and santonin were isolated from rhizomes ofBerberis aristata and unexpanded flower buds ofArtemisia maritima, respectively. Efficacy of these two chemicals individually as well as of their mixtures, was tested against spore germination of some saprophytic and obligate fungi. While berberine individually was effective against most of the fungi,Helminthosporium spp. were least affected even at the highest dose (1500 ppm). Santonin was equally effective against several fungi. Mixture of both alkaloids found to be more effective than individual ones. Keeping the dose of berberine constant and santonin at two different concentrations (viz. 250 and 500 ppm) the spore germination ofHelminthosporium, oryzae was stimulated. Increasing concentration of santonin inhibited the spore germination of all other fungi tested,Colletotrichum capsici being affected only by 20 and 5% (at berberine concentration of 250 and 500 ppm, respectively). On the other hand, santonin being constant and berberine at different concentrations, the mixture was effective against all the fungi.

Summary Effect of cumin seed exudates on spore germination of Curvularia lunata, Alternaria solani and Colletotrichum capsici was studied in the laboratory. Three-hour exudate obtained for first 3-hour favoured initial protrusion but inhibited subsequent growth of the germ tubes. Two-hour exudate obtained between 5 and 7 hours after soaking was suggested to have no effect on initial protrusion but was found to have promotive effect on subsequent growth of germ tube. Short treatment with inhibitor for 45 minutes followed by a treatment with promotor ensured high germination and uniform germ tube growth. Modifying effects on the spermatosphere and adaptive significance of sequential exudation are discussed. re]19730125  相似文献

Four saprophytic and pathogenic fungi were isolated from the leaf surface of Calliandra haematocephala, a tropical legume known to contain large amounts of rare nonprotein imino acids in its leaves and seeds. The fungi Aspergillus niger, Aspergillus sp., Curvularia sp., and Penicillium sp. were cultured in the laboratory and tested for susceptibility to leaf extracts of the host plant and to proline, pipecolic acid, cis-5-hydroxypipecolic acid, and 2,4-trans-4,5-cis-4,5-dihydroxypipecolic acid. Fungal spore germination and germ tube growth were measured. Aspergillus sp. was inhibited by plant extracts and by pipecolic acid and cis-5-hydroxypipecolic acid. Curvularia sp. growth was stimulated by plant extracts and by pipecolic acid. The other two fungi were unaffected by any of the treatments. The data indicate that imino acids may play a role in the specific resistance of Calliandra spp. to Aspergillus sp.

A natural red dye which is produced by the tiny insects Kerria lacca while feeding on host trees is popularly known as lac dye. Lac dye is a mixture of at least five closely related pure compounds all being anthraquinone derivatives designated as laccaic acid A, B, C, D and E. Anthraquinones isolated from different natural sources and reported to have potent antimicrobial activity. The lac dye, which is also a mixture of anthraquinone derivatives, is expected to exhibit antifungal and antibacterial activity. Lac dye cannot be used as antibacterial and antifungal agent due to its low water solubility and high polarity. Therefore, it is modified into its methyl derivative to enhance its bio-efficacy. Methylated lac dye is characterized with the help of TLC, UV–Vis spectroscopy and FT-IR, NMR analysis. An in vitro spore germination assay was carried out to evaluate the antifungal efficacy of methylated lac dye against some phytopathogenic fungi which commonly caused a various foliar diseases in crop plants viz., Alternaria solani, Curvularia lunata, Erysiphe pisi, Helminthosporium oryzae and Verticillium sp. Among the tested fungi, Verticillum sp. showed highest sensitivity, which showed 100% inhibition at 750 and 1000 µg/ml as compared to control. However, E. pisi an obligate parasite also showed varied sensitivity but at 1000 µg/ml showed 100% spore germination as compared to control. Methylated lac dye also showed strong antibacterial properties against Ralstonia solanacearum at very low concentration (40 and 50 µg/ml). Hence, lac dye may serve as potent antifungal and antibacterial agent in plant disease management.

We report on the elucidation of two separate pathways of spore germination in a plant pathogenic fungus Colletotrichum gloeosporioides f. sp. aeschynomene. Conidia of the fungus can germinate either from one side or from both sides, depending on external conditions. In shake culture that includes an extract made up from fresh peas, the unicellular conidium divides and one of the two cells develops a germ tube. On a solid surface this germ tube differentiates an appressorium. In rich medium without pea extract, germination is highly similar to Aspergillus spore germination: the conidium swells, forms a single germ tube and then divides and forms a second germ tube. Conidia that germinate in a rich medium do not form appressoria even on a solid surface and are non-pathogenic. In rich medium, cAMP stimulates germination in rich liquid cultures and induces appressoria formation on a hard surface. In pea extract cAMP induces swelling and formation of irregular germ tubes and appressoria. Our results suggest that plant surface signals induce pathogenic-specific spore germination in a cAMP-independent manner. cAMP is required for saprophytic germination and for appressorium formation.

The aim of the present study was to evaluate the antifungal activity of fatty acids against phytopathogenic fungi. Two pot experiments were conducted by mixing palmitic and oleic acids in the soil in which poor plant growth was observed. In addition, the antifungal activities of nine fatty acids (butyric acid, caproic acid, caprylic acid, capric acid, lauric acid, myristic acid, palmitic acid, oleic acid, and linoleic acid) against four phytopathogenic fungi: Alternaria solani, Colletotrichum lagenarium, Fusarium oxysporum f. sp. Cucumerinum, and Fusarium oxysporum f. sp. lycopersici, were assessed by measuring mycelial growth and spore germination via Petri dish assay. The results of the pot experiments showed that the mixture of palmitic and oleic acids enhanced the growth of the seedlings of continuous-tomato and continuous-cucumber. Except for oleic acid, in the Petri dish assay, the fatty acids tested were observed to inhibit the mycelial growth of one or more tested fungi. In addition to the suppression of mycelial growth, butyric acid, caproic acid, caprylic acid, capric acid, lauric acid, and palmitic acid showed an inhibitory effect against spore germination and the extent of inhibition varied with both the type of fatty acids, and the fungi. In particular, capric acid displayed strong inhibitory effect against C. lagenarium on the mycelial growth and spore germination. The saturated fatty acids, i.e. palmitic acids, showed stronger antifungal activity than the unsaturated fatty acids, i.e. oleic acid. It suggests that fatty acids might be applicable to exploring for alternative approaches to integrated control of phytopathogens.

Summary Evans' tube and cellophane agar-disc techniques were used to evaluate the colonization capacity of the soil microbes and fungistasis, respectively. Spore germination ofAlternaria tenuis, Curvularia geniculata, Helminthosporium rostratum andPestalotia sp. was studied on a natural soil. Aspergillus fumigatus was the pioneer colonizer, followed in the order of decreasing colonizing capacity by,A. flavus, A. niger, A. terreus andPenicillium nigricans. Sensitivity of test spores to soil fungistasis declined in the order,C. geniculata → Pestalotia sp. →A. tenuis → H. rostratum. The fungistatic level of the soil samples decreased gradually from side arms 1 to 7 of the Evans tube, inoculated with unsterilized soil from one end. Studies conducted with an inoculum of four soil fungi (A. fumigatus, A. flavus, A. niger, andP. nigricans) as well as those involving fumigation of Evans tube by CS2 also proved the greater colonizing capacity ofA. fumigatus. The level of fungistasis followed closely the colonization pattern of this fungus. When added individually to the soil, all fungi were able to inhibit spore germination of test fungi to some degree or other.

The anti-fungal activity of two alkaloids isolated from bulbs of Zephyranthes citrina was observed against 10 fungi, viz, Aternaria solani, A. triticina, Curvularia lunata, C. maculuns, Cercospora malvacearum, Erysiphe sp., Fusarium udum, Helminthosporium pisi, H. speciferum and Ustilago cynodontis. Different concentrations (200, 400, 600, 800, 1000 µg/ml of alkaloids (A and B) were used. Spore germination was inhibited at 600, 800, 1000 µg/ml. B alkaloid was used against Erysiphe cichoracearum causing powdery mildew in balsam (Impatiens balsamina) in the field as pre- and post-inoculation treatments at 1000, 1500, 2000 µg/ml doses. The extract was effective in both pre- and post-inoculation treatments. Foliar application of this alkaloid resulted in inducing synthesis of phenolic acid in the leaves of balsam. Maximum phenolics were detected in the leaves treated with 1500 µg/ml in both pre- and post-inoculation treatments. The increase in the production of phenolics in treated leaves of balsam can be correlated with the induction of resistance in treated plants against powdery mildew. The significant efficacy of the alkaloid under field conditions opens the possibility of its use by farmers for also controlling other diseases.

Antibacterial and antifungal properties of wax and hexane extracts of Citrus spp. peels were tested using bioautographic and microdilution techniques against three plant pathogenic fungi (Penicillium digitatum, Curvularia sp., and Colletotrichum sp.), two human pathogens (Trichophyton mentagrophytes and Microsporum canis), and two opportunistic bacteria (Escherichia coli and Staphylococcus aureus). Two polymethoxylated flavonoids and a coumarin derivative, were isolated and identified from peel extracts, which presented antimicrobial activity especially against M. canis and T. mentagrophytes: 4',5,6,7,8-pentamethoxyflavone (tangeritin) and 3',4',5,6,7,8-hexamethoxyflavone (nobiletin) from C. reticulata; and 6,7-dimethoxycoumarin (also known as escoparone, scoparone or scoparin) from C. limon.

Aerobiological studies at three different indoor sitesviz., food grain godowns, library building and bakerywere carried out. The main objective of the study wasto find out the fungal flora at these places and itsimpact on the organic materials which are stored /processed there. The study reveals that over 40fungal types prevail in such organic matter richenvirons. The incidence of fungi was found to befluctuating according to months and seasons. Studieswere carried out by Andersen two stage sampler. Maximum spore concentration was observed in thelibrary followed by bakery and food grain godowns. Common spore types belonged to the species of Aspergillus. Alternaria, Cladosporium,Helminthosporium, Curvularia, Rhizopus etc. Suchfungi are mainly responsible for the deterioration ofpaper materials in library and food grains in godowns. Bakery products also get contaminated with a goodnumber of fungi types. The people working at suchplaces also get allergic due to these fungalcomponents.

Infection of bean pods with Colletotrichum lindemuthianum leads to the appearance in diffusates of a range of fluorescent and phenolic compounds and of at least two inhibitory compounds. These compounds were found to be absent or in reduced concentration in control diffusates, although the inhibitors were frequently observed to appear at low concentrations without infection. Although sugars and amino acids were found to be released into diffusates, variation in the concentration of these stimulants was found to have little effect on spore germination. Evidence from solvent partition, spectrophotometry and chromatography suggests that the two inhibitors can be tentatively identified with inhibitors previously described from French bean, and both appear to be phenolic substances. It is suggested that inhibitor production may be regarded as part of a general change in aromatic biosynthesis following infection.
