应用cDNA微阵列技术筛选大鼠脊髓损伤修复相关基因   总被引:2,自引:0,他引:2

脊髓损伤是一类常见的、高致残率的中枢神经系统疾病,由于多种复杂因素影响其损伤后的修复过程,损伤脊髓的再生能力非常有限。本研究采用cDNA微阵列技术筛选大鼠脊髓损伤后出现的差异表达基因。实验组动物在T8-T9进行脊髓全横断手术,对照组动物只打开椎板;4.5d后取脊髓进行RNA提取并在反转录过程中进行Cy3/Cy5标记,然后与预制的、带有4041条特异性探针的芯片进行杂交。Cy5/Cy3信号比值≥2.0视为脊髓损伤后出现差异表达的基因。通过筛选,我们得到了65个上调表达基因(21个已知基因,30个已知EST和14个未知基因)和79个下调基因(20个已知基因,42个已知EST和17个未知基因)。进一步通过半定量RT-PCR对其中的5个上调已知基因(Timpl,Tagln,Vim,Fc gamma receptor,Ctss)和三个下调已知基因(stearyl-CoA desaturase,F2,Ensa)的表达情况进行了验证,结果显示与芯片结果一致。这些基因可能在脊髓损伤后的修复过程中起一定的作用,对其深入研究将有助于揭示脊髓损伤修复的分子机制。  相似文献

以易出胚大白菜‘09C3’为材料,克隆到7个小孢子胚胎发生相关候选基因的cDNA核心片段,并采用半定量RT-PCR方法研究了这些基因在游离小孢子热激处理和培养期间的表达。结果表明,获得的7个基因的核心片段长度在213bp和417bp之间,克隆号分别为C31、C33、99H6、99H8、EV14、EX01和C35。NCBI进行Blastn和Blastx分析结果显示,这些片段与源基因HSP70、BcHSP、Lec2、Fus3、PAP14、HSP17.6和HSP18.2的同源性均超过80%。半定量RT-PCR结果表明,HSP70和Lec2在热激24h后上调表达,而常温培养下24h后没有变化;BcHSP、HSP18.2、Fus3和HSP17.6在热激24h后和常温下24h都上调表达;PAP14在热激24h后和常温下24h表达下调。  相似文献

Abstract

Reactive oxygen species (ROS) and the NADPH oxidase (NOX) enzyme are both up-regulated after spinal cord injury (SCI) and play significant roles in promoting post-injury inflammation. However, the cellular and temporal expression profile of NOX isotypes, including NOX2, 3, and 4, after SCI is currently unclear. The purpose of this study was to resolve this expression profile and examine the effect of inhibition of NOX on inflammation after SCI. Briefly, adult male rats were subjected to moderate contusion SCI. Double immunofluorescence for NOX isotypes and CNS cellular types was performed at 24 h, 7 days, and 28 days post-injury. NOX isotypes were found to be expressed in neurons, astrocytes, and microglia, and this expression was dependent on injury status. NOX2 and 4 were found in all cell types assessed, while NOX3 was positively identified in neurons only. NOX2 was the most responsive to injury, increasing in both microglia and astrocytes. The biggest increases in expression were observed at 7 days post-injury and increased expression was maintained through 28 days. NOX2 inhibition by systemic administration of gp91ds-tat at 15 min, 6 h or 7 days after injury reduced both pro-inflammatory cytokine expression and evidence of oxidative stress in the injured spinal cord. This study therefore illustrates the regional and temporal influence on NOX isotype expression and the importance of NOX activation in SCI. This information will be useful in future studies of understanding ROS production after injury and therapeutic potentials.  相似文献

Angiopoeitin-2 (Ang-2) antagonizes Angiopeitin-1 (Ang-1)-mediated Tie-2 signaling. Ang-1 is reported to up-regulate anti-apoptotic Survivin expression. Here, we investigated the interplay between Ang-2 and Survivin in response to oxidized low density lipoprotein (OxLDL)-induced apoptosis. We demonstrate that treatment of human aortic endothelial cells (HAEC) with 100 μg/ml of OxLDL down-regulated Ang-2 expression as early as 4h after treatment and persisted up to 24h (p<0.05, n=3), but did not down-regulate Survivin until the 24h point. Further, treatment of HAEC with recombinant Ang-2 up-regulated Survivin expression (at Ang-2 ≥200 ng/ml, p<0.05, n=3) and attenuated the OxLDL-mediated down-regulation of Survivin (p<0.05, n=3). Knockdown of Ang-2 further down-regulated Survivin expression, whereas over-expression of Survivin attenuated OxLDL-induced HAEC apoptosis (p<0.05, n=3). Hence, Ang-2 mediated Survivin expression in response to OxLDL-induced endothelial apoptosis.  相似文献
