为筛选适宜的四倍体诱导条件, 研究了细胞松弛素B(CB)浓度(0.3、0.5、0.7和0.9 mg/L)、起始诱导时间(5、10和15min)、诱导持续时间(10、15、20和25min)及三者的交互作用对长牡蛎“海大2号”四倍体诱导率(四倍体率)和D形幼虫率的影响, 并对诱导处理组获得的幼贝倍性进行检测分析, 进一步验证了研究诱导方法获得可存活长牡蛎“海大2号”四倍体幼贝的可行性。结果表明, CB浓度和起始诱导时间对长牡蛎“海大2号”四倍体率有显著影响(P<0.05), 而诱导持续时间对四倍体率影响不显著(P>0.05), 但是三者的交互作用对四倍体率有显著影响(P<0.05)。在CB浓度为0.5 mg/L、起始诱导时间为15min及诱导持续时间为20min时, 四倍体率有最大值为37.82%。CB浓度、起始诱导时间和诱导持续时间均对D形幼虫率有显著影响(P<0.05), 但是三者的交互作用对D形幼虫率没有显著影响(P>0.05)。增加CB浓度或延长诱导持续时间, D形幼虫率均呈降低的趋势; 起始诱导时间为5min时, D形幼虫率显著高于其他起始诱导时间。获得幼贝的两个诱导处理组的倍性检测结果显示, 长牡蛎“海大2号”四倍体率分别为2.22%和3.03 %, 与幼虫期相比四倍体率显著下降。通过对CB浓度、起始诱导时间及诱导持续时间进行优化, 可以筛选出适宜的长牡蛎“海大2号”四倍体诱导条件。研究结果为长牡蛎“海大2号”四倍体培育提供了重要的参考资料。  相似文献

为获得高效的熊本牡蛎的三倍体诱导方法, 分别比较了6-DMAP、高盐和低盐3种诱导方法在不同的诱导浓度(盐度)、起始诱导时间和持续诱导时间下的卵裂率、孵化率和三倍体率, 同时比较了3种方法获得的幼虫的生长、存活和三倍体率变化情况。结果表明, 在6-DMAP诱导组中, 三倍体率和诱导效率分别可达37.97%—58.01%和34.30%—42.50%, 培育期间幼虫的平均存活率27.19%, 生长率13.03 μm/d, 三倍体率降低了24.94%; 在低盐诱导组中, 三倍体率和诱导效率分别可达7.32%—42.25%和2.17%—31.41%, 培育期间幼虫的平均存活率33.92%, 生长率12.71 μm/d, 三倍体率降低了20.64%; 在高盐诱导组中, 三倍体率和诱导效率分别可达7.47%—63.03%和6.58%—49.41%, 培育期间幼虫的平均存活率31.66%, 生长率13.08 μm/d, 三倍体率降低了17.64%。综合来看, 高盐诱导是诱导三倍体熊本牡蛎的最优方法, 其诱导条件的最佳组合为盐度55, 起始诱导时间受精后15min, 持续诱导时间20min。研究为熊本牡蛎的三倍体诱导提供了技术支持, 对熊本牡蛎的多倍体育种具有重要的理论指导意义。  相似文献

研究以乳糖代替IPTG作为诱导剂诱导重组人胸腺肽α1表达的可行性,对乳糖诱导的时机、乳糖浓度、诱导持续时间以及其它诱导条件进行研究,确定了乳糖诱导的最佳条件。结果表明,乳糖能有效地诱导重组人胸腺肽α1的表达,并且目的蛋白的表达量略高于IPTG的诱导量。  相似文献

This paper reveals that a discontinuous in vitro induction, namely, the periodic presence and absence of foreign induction factors, might be, under a certain condition, more effective to stimulate stem cells' differentiation than a continuous induction. Bone marrow stem cells (BMSCs) derived from Sprague Dawley rats were employed to examine the effects of discontinuous additions of osteogenic supplements with a series of alternate frequency in contrast to those with continuous induction or no induction. The results demonstrated that a suitable discontinuous induction was more able to achieve osteogenesis than not only no induction but also the associated continuous induction. Additionally, the osteogenic supplements were confirmed to enhance cell differentiation but suppress cell proliferation. So, the combination of differentiation extent per cell and cell number accounts for the "unexpected" good osteogenic effect of the discontinuous induction. The induction effect was found to be dependent upon alternate frequency, and the optimum alternate period in our experimental systems was determined to be around 4 days. Since it is very common to change culture medium every 2-4 days, such a strategy of discontinuous induction does not bring any extra manual work but reduces the consumption of foreign induction factors and significantly enhances the global differentiation efficacy. Our work thus affords a convenient and practical approach to achieve differentiation of BMSCs, which might be useful for potential large-scale culture and differentiation of stem cells. Meanwhile, the existence of optimum frequency implies some unknown inherent rhythms of cell proliferation and differentiation.  相似文献

Color induction in the honeybee is investigated in color discrimination experiments. An individual bee walks in a dark arena and is trained to a self-luminant stimulus presented from below. In the dual-choice tests the dark background is replaced by a colored induction stimulus. Choice behavior is recorded by TV camera and analyzed by computer. Successive color induction is separated from simultaneous induction by analysis of the walking paths. Only successive color induction occurs. Simultaneous effects are not observed. That is a stimulus acts as a color inducing stimulus only when the bee crosses this stimulus. Thus, the color perceived by a given eye region is found to be dependent on the viewing history, but not on the stimuli presented simultaneously on neighboring parts of the retina. Color induction in the honeybee described in terms of selective sensitivity decrease (adaptation) does not explain all behavioral effects induced by the stimulus. The time course of successive color induction is calculated from the exposure times to the induction stimulus and from the choice behavior. The data suggest that color induction is complete after a few seconds. Photoreceptor adaptation is sufficient to explain the observed time course.  相似文献

Cost-effective induction of bacteriophage 5Qm in Escherichia coli on reporter-protein production is presented. Long-duration temperature induction increases the mortality of bacterial hosts and decreases the productivity efficiency; however, sufficient time span of induction is essential to overcome the induction threshold. Thus, the optimal duration for cost-effective induction is approximately 30 min, since the benefit of induction longer than 30 min is unlikely to pay off the expense of significant host viability loss. Global optimization of economically feasible induction occurs at a critical optical density of ca. 1.0 for 30 min duration at 38v°C.  相似文献

以侧芽为外植体材料,对其组织培养的各个阶段进行了研究。结果表明:品种对外植体的污染率、褐变率、芽诱导率及丛生芽增殖倍数均有显著影响;外源激素的种类和浓度在芽诱导分化、增殖、生根等各个阶段都是主要的影响因素;外植体在1/2MS+NAA0.2mg·L~(-1)+6-BA2mg·L~(-1)上,48d后芽开始分化,诱导率为40%,平均芽分化数为6个,诱导分化效果最好;芽增殖培养阶段,若培养基中仅含6-BA,且浓度在0~4mg·L~(-1)之间,芽增殖倍数随其浓度增大而增大;若培养基中仅含NAA,且其浓度为0.2mg·L-1时,增殖倍数最大;培养基中同时添加6-BA和NAA,比单独添加6-BA或NAA时,芽的增殖效果好,且在6-BA3mg·L~(-1)+NAA0.5mg·L~(-1)的MS培养基上,G.dissitisflora和G.‘Claret’增殖倍数最大,分别为5.24和3.84;在任何相同的培养基上,G.dissitisflora的增殖倍数显著高于G.‘Claret’;小苗在含有NAA0.5mg·L~(-1)+IBA0·5mg·L~(-1)的生根培养基上生长,生根率可达91.03%,平均根数为5.3条/株,生根效果较好。  相似文献

The effects of nutrients and hormones on the mRNA levels of acetyl-CoA carboxylase, fatty acid synthase, malic enzyme, and glucose 6-phosphate dehydrogenase were examined in primary cultures of rat hepatocytes during the process of induction. The addition of both glucose and insulin to the culture medium markedly enhanced the lipogenic enzyme mRNA induction due to either of them, in 16 h. Fructose or glycerol proved to be an effective substitute for glucose, suggesting that glycolytic metabolites were involved in the mRNA induction. It is remarkable that mRNA induction of acetyl-CoA carboxylase was the most sensitive to glucose and also to insulin among the lipogenic enzymes. Polyunsaturated fatty acids markedly reduced the mRNA induction of lipogenic enzymes. Dexamethasone enhanced all the lipogenic enzyme mRNA induction by insulin. On the other hand, triiodothyronine addition greatly increased the mRNA concentrations of lipogenic enzymes, but dexamethasone decreased rather than increased the mRNA induction by triiodothyronine. The effects of insulin on the induction of the lipogenic enzyme mRNAs were similar, but those of triiodothyronine were not. Triiodothyronine markedly enhanced malic enzyme mRNA induction by insulin with dexamethasone, and tended to enhance the induction of the acetyl-CoA carboxylase and fatty acid synthase mRNAs, but not that of glucose 6-phosphate dehydrogenase mRNA. It appeared that insulin and triiodothyronine synergistically enhanced lipogenic enzyme mRNA induction by glucose, but the mechanisms were different.  相似文献

The kinetics of induction of cytosolic DT-diaphorase (NAD(P)H dehydrogenase-quinone, EC 1.6.99.2) by benzo(a)pyrene (BP) in the liver of the 8-day-old rat has been studied. After a lag phase of 8 h, DT-diaphorase reaches its maximum activity in three waves, with plateau levels of activity between 15–18, 26–36, and 40 h after administration of BP, at 4, 15, and 26 times the basal activity, respectively. A lower degree of induction of DT-diaphorase could be observed in the kidney cortex of the young rat and in the liver of the adult rat. No induction was observed in the fetal liver and in the adult kidney cortex. Lead acetate treatment of the adult rat resulted in induction of DT-diaphorase by BP in the liver and in the kidney cortex. Induction could not be observed in the regenerating liver of the adult rat. Experiments with picolinic acid (PA)—as a G1 inhibitor—administered simultaneously or at different time intervals after BP administration resulted in an inhibition of induction, depending on the time of administration of picolinic acid. It is concluded that a mitotic cell cycle is necessary for DT-diaphorase induction by BP. Evidence is presented that BP acts in late G1. The kinetics of induction of aryl hydrocarbon hydroxylase (AHH) by BP in liver microsomes of the 8-day-old rat has been compared with the induction of cytosolic DT-diaphorase. The effect of PA on the induction of AHH has also been studied. In view of the differences in kinetics of induction and in the effects of PA, it is concluded that the induction of AHH and that of DT-diaphorase are dissociated. AHH induction may take place in all hepatocytes, in contrast to DT-diaphorase induction.  相似文献

Abstract. Winged and wingless individuals of a pink clone of the pea aphid, Acyrthosiphon pisum (Harris), showed differences in the response curves for photoperiodic induction of both males and sexual females (oviparae). The critical night length (CNL) for ovipara induction in winged aphids was 0.75 h shorter than in wingless aphids, whereas the CNL for male induction in winged aphids was 1.0h longer than in wingless aphids. This means that in winged aphids the CNL for male induction in winged aphids was 0.5 h longer than that for ovipara induction, while in wingless aphids the CNL for male induction was 1.0–1.5 h shorter than that for ovipara induction, and also the shapes of the curves differed.
Winged aphids were produced by wingless mothers which were crowded as young adults. However, when young adults were crowded in long nights, winged aphids were not produced, and the CNL for wing inhibition was between 9.5 and 10h. This effect of photoperiod on wing induction was maternal.  相似文献

以三种柚子(Citrus grandis L.)品种叶片为外植体诱导愈伤组织,以幼嫩茎段诱导不定芽和不定根。结果表明:1.0 mg/L2,4-D与6%蔗糖处理的叶片愈伤组织诱导率最高,最高诱导率达82.5%;6-BA0.5 mg/L和NAA0~0.5 mg/L激素配比诱导丛生芽效果最高,最高可在每个茎段上诱导4个不定芽;1/4 MS+IBA 2.0 mg/L诱导生根效果最好,最高达40%,虽然根系很弱,难以成苗,但可以用于试管微嫁接。所有的培养中,?溪蜜柚和沙田柚的诱导率接近,都比江津柚高大约30%~50%。  相似文献

In the pineapple sector of Benin, poor fruit quality prevents pineapple producers to enter the European market. We investigated effects of common cultural practices, flowering and maturity synchronisation, (1) to quantify the trade-offs of flowering and maturity synchronisation for pineapple quality and the proportion of fruits exportable to European markets, and (2) to determine the effect of harvesting practice on quality attributes. Four on-farm experiments were conducted during three years using cultivars Sugarloaf and Smooth Cayenne. A split-split plot design was used in each experiment, with flowering induction practice as main factor (artificial or natural flowering induction), maturity induction practice as split factor (artificial or natural maturity induction) and harvesting practice as the split-split factor (farmers’ harvest practice or individual fruit harvesting at optimum maturity). Artificial flowering induction gave fruits with lower infructescence weight, higher ratio crown: infructescence length, and a lower proportion of fruits exportable to European markets than natural flowering induction. The costs of the improvements by natural flowering induction were huge: the longer durations from planting to flowering induction and harvesting, the higher number of harvestings of the fruits increasing the labour cost and the lower proportion of plants producing fruits compared with crops from artificially flowering-induced plants. Artificial maturity induction decreased the total soluble solids concentration in the fruits compared with natural maturity induction thus decreasing the proportion of fruits exportable to European markets, at a benefit of only a slightly shorter time from flowering induction to harvesting. Harvesting individual fruits at optimum maturity gave fruits with higher total soluble solids in naturally maturity induced fruits compared with the farmers’ harvest practice. Given the huge costs of natural flowering induction, options to use artificial flowering induction effectively for obtaining high fruit quality are discussed.  相似文献

In the early development of the gastropod mollusc Patella vulgata, two inductive processes take place between the animal and vegetal hemispheres. One is the induction of the vegetally-located stem cell of the mesoderm (the 3D macromere) by the first-quartet micromeres (which are located in the animal hemisphere). The other is the induction of dorsoventral symmetry in the first-quartet micromeres by the 3D macromere. As a consequence of the latter induction process, a dorsoventrally-organised prototroch is formed. In this paper, the moment of 3D induction is determined by deleting the inducing first-quartet micromeres at successive stages of development. Furthermore, the role of gap junctions in the two above-mentioned induction processes is investigated. This was done by studying lucifer yellow dye transfer between the 3D macromere and the first-quartet micromeres, in relation to these two induction processes. Analysis of the pattern of dye-transfer in vivo, in sections and with confocal laser scanning microscopy demonstrated that the moment of dye transfer coincides with the moment of 3D induction and with the moment of induction of the dorsoventral pattern of the prototroch. This indicates that gap-junctional communication may be involved in these two induction processes.  相似文献

Transforming growth factor beta1 (TGFbeta) simultaneously induces the expression of fibronectin, fibronectin receptor, laminin, and laminin receptor (alpha6beta1 integrin) in the human colon cancer cell line Moser (Int J Cancer, 57:742, 1994). Induction of fibronectin and induction of fibronectin receptor by TGFB are tightly coupled, and disrupting fibronectin induction disrupts the induction of fibronectin receptor and cellular adhesion to fibronectin (J Cellular Physiol, 170:138, 1997). We recently demonstrated the efficacy of using antisense chain-specific laminin RNA expression vectors to disrupt the induction by TGFP of the multichain laminin molecule (J Cellular Physiol, 178:296, 1999). We now show in this report that Moser cells used alpha6 and beta1 integrins to adhere to laminin, and, as is the fibronectin and fibronectin receptor system, disrupting the induction by TGFbeta of the ligand laminin by the expression of antisense laminin A chain RNA disrupted the induction of 125I-laminin binding and cellular adhesion to laminin. Disrupting laminin induction also blocked the induction of alpha6 and beta1 integrin laminin receptor by TGFbeta. We conclude that disrupting the induction of the ligand laminin by TGFbeta disrupts TGFbeta-regulated laminin receptor function by suppressing the induction of alpha6 and beta1 integrins. Therefore, targeted disruption of the ligand laminin may be an effective means in disrupting the function of both the ligand and its receptor in cells that utilize the laminin and laminin receptor system in malignant cell behavior.  相似文献

The activities of the two beta-galactosidases of Neurospora increased in response to the presence of lactose or galactose. d-Xylose preferentially induced higher levels of activity in one of these enzymes. The initial period of the inductive response was examined in cultures transferred from noninducing to inducing media. Variations in the ratio of activities of these enzymes during this induction period indicate that their induction was not coordinate. The induction of the beta-galactosidases of Neurospora was less sensitive to low concentrations of inducers than was the induction of the beta-galactosidase of bacteria; the time lag of induction was greater in the eucaryotic organism, and the maximal induction of active enzyme was less than that observed in bacteria.  相似文献

This review provides an overview of our current understanding of signaling mechanisms involved in lens induction, which are presented in context of the major stages of lens induction (competence, bias, inhibition and specification). Although the process of lens induction is generally well conserved, we highlight aspects of induction that vary among species. Exposure of insect larvae to sublethal concentrations of crystal toxins from the soil bacterium Bacillus thuringiensis (Bt toxins) causes the induction of immune and metabolic responses that can be transmitted to offspring by epigenetic inheritance mechanisms. Given that the elevated immune status carries significant developmental penalties, we wanted to establish the relationships between immune induction, tolerance to the toxin and developmental penalties. A laboratory culture of Helicoverpa armigera was induced by a sublethal bacterial suspension containing crystal toxin Cry1Ac in one generation and maintained in the presence of toxin, acquiring significant levels of tolerance to the toxin within 12 generations of continuous exposure. Comparing tolerant and susceptible insects, we show that the induction of larval immune response and the coincident alteration of development-related metabolic activities by elicitors in the larval gut (larval induction) differs from the elevated immune status transmitted by epigenetic mechanisms (embryonic induction). Because the damaging effects of larval induction processes are higher compared to embryonic induction, it is likely that overall developmental penalties depend on the relative contribution of the two induction processes. When insects are kept with the same amount of toxin in the diet for subsequent generations, the embryonic induction process increases its contribution compared to the larval induction, resulting in reduced overall developmental penalty, while tolerance to the toxin is maintained.  相似文献

以未诱导组作为空白对照研究比较真菌诱导、超声诱导和热诱导家蝇Musca domestica 幼虫血淋巴初提液的抗真菌肽效果,比较各种诱导方法诱导后的幼虫存活率;用凝胶层析法和高效液相分离纯化热诱导家蝇3龄幼虫抗真菌肽,检测其抗白假丝酵母菌Candida albicans和新生隐球菌Cryptococcus neoformans活性;SDS-PAGE分析抗真菌肽的蛋白分子量范围。结果表明:3种诱导方法诱导后家蝇幼虫均产生具有明显抗真菌作用的抗真菌肽,其初提液抑菌圈大小没有明显差别;真菌诱导组和热诱导组幼虫存活率低于对照组,而超声诱导组与对照组相比则无明显差别。经分离纯化后,抗真菌肽仍具有较好的抗真菌活性;SDS-PAGE分析表明该抗真菌肽有效成分的蛋白分子量在14.4 kD以下。结果提示热诱导家蝇幼虫产生抗真菌肽是一种方便、有效的诱导方式。  相似文献
