The ichnospecies Cladichnus parallelum consists of upper components characterized by downward branching, decreasingly inclined tubes, and lower components having horizontal, long, parallel tubes formed in the laminated part of a muddy, marly turbidite. The tubes of both components were successively produced, and the lower ones display an annulated fill. The producers of Cladichnus exhibit thigmotactic and phobotactic behavior in that the tubes are regularly spaced apart, do not cross over- or cross-cut, and their orientation is related to the fabric of the turbidite. The tube fill of both components is enriched in terrigenous material and organic matter suggesting selective feeding on the sediment surface, and the tubes do not show any grain sorting around them. The strong compression of the horizontal tubes suggests that the fill initially had high porosity. The producer of Cladichnus parallelum probably took advantage of burrowing in an anoxic setting; microbes likely played a role in converting organic material stored within the tubes or in processing dissolved compounds drained into the highly porous tubes.  相似文献

Nicotiana tabacum was used as a pistillate parent and crossed with three self-compatible species, N. rustica, N. repanda and N. trigonophylla, which were previously reported to have pollen tubes unilaterally inhibited by N. tabacum pistil. Temporal and morphological observations revealed distinct differences of pollen tube behavior among these incongruous crosses. Pollen tubes of N. repanda were arrested in stigma and those of N. rustica in the middle of the style. On the other hand, pollen tubes of N. trigonophylla continued growing at a slow rate. Tubes of N. repanda and N. rustica showed morphological abnormalities such as swelling, thick wall, and irregular callose deposition. In addition, tubes of N. rustica often elongated in reverse direction and wound about in the middle of the style. Although the tubes of N. trigonophylla were apparently normal in morphology, they were distributed throughout the transmitting tissue, differing from the self-pollination of N. tabacum in which they were confined to the peripheral region of it. The diversity of pollen tube behavior indicates that physiological causes of incongruity are different among the three crosses. Bud pollination enabled pollen tubes to reach the ovary in all crosses, indicating that the N. tabacum pistil acquired its ability to inhibit foreign pollen tube elongation with its development. When interspecific hybrids between N. tabacum and the other three species were pollinated by parental species, tubes reached the ovary in all crosses, but the elongation rate of tubes slowed down and morphology was abnormal.  相似文献

Atypical isolates of Candida albicans which failed to produce germ tubes in routine diagnostic procedures were examined for their ability to produce germ tubes in various media. Bovine serum was more effective than defined media for induction of germ tubes in the majority of isolates. A few strains formed appreciable germ tubes only in bovine serum with added thioglycollate or cysteine. One strain did not produce germ tubes in any medium. Germ-tube maturation appeared to be dependent upon mitochondrial RNA polymerase activity. The failure by an isolate to produce germ tubes, particularly in tests without strictly controlled conditions, does not preclude the possibility that the organism is C. albicans.  相似文献

A laboratory culture of an Israeli benthic midge, Chironomus luridus, was exposed to two chemicals: copper and monochloramine. The objective of this study was to determine the protective nature of Chironomus' larval tube. Three experimental conditions were tested: larva with sand tubes, with silt tubes and without tubes. Larvae without tubes were significantly more sensitive to copper and chloramine than larvae that had sand or silt as tube building substrate. The tubes protected the insects against chemicals throughout 14 days of exposure time. Silt tubes had higher protective value than sand tubes, especially when exposed to copper for a short period of time (LC50/ 24 h, with silt, sand, or none: 80.0, 7.0 and 3.4 mg l–1 copper, respectively). C. luridus seemed to be better protected against copper than against chloramine (LC50/ 24 h, with silt, sand, or none: 12.2, 6.4 and 3.7 mg l–1 chloramine, respectively). The acute toxicity of copper to chironomid larvae was investigated using a cytochemical method. Larva in silt tubes had significantly higher non-specific esterase activity in midgut cells than larvae without tubes. We conclude that, in addition to its role in feeding, respiration and anti-predation shelter, the C. luridus tube protects its inhabitant from toxic substances.  相似文献

  • Pollen‐pistil interactions are a fundamental process in the reproductive biology of angiosperms and play a particularly important role in maintaining incipient species that exist in sympatry. However, the majority of previous studies have focused on species with syncarpous gynoecia (fused carpels) and not those with apocarpous gynoecia (unfused carpels).
  • In the present study, we investigated the growth of conspecific pollen tubes compared to heterospecific pollen tubes in Sagittaria species, which have apocarpous gynoecia. We conducted controlled pollinations between S. pygmaea and S. trifolia and observed the growth of conspecific and heterospecific pollen tubes under a fluorescence microscope.
  • Heterospecific and conspecific pollen tubes arrived at locules within the ovaries near simultaneously. However, conspecific pollen tubes entered into the ovules directly, whereas heterospecific tubes passed through the carpel base and adjacent receptacle tissue, to ultimately fertilize other unfertilized ovules. This longer route taken by heterospecific pollen tubes therefore caused a delay in the time required to enter into the ovules. Furthermore, heterospecific pollen tubes displayed similar growth patterns at early and peak pollination. The growth pattern of heterospecific pollen tubes at late pollination was similar to that of conspecific pollen tubes at peak pollination.
  • Heterospecific and conspecific pollen tubes took different routes to fertilize ovules. A delayed entry of heterospecific pollen into ovules may be a novel mechanism of conspecific pollen advantage (CPA) for apocarpous species.

The kinorhynch fauna in two Arctic fjords at Spitsbergen was explored and eight species of Echinoderes were recorded, of which three are new to science and described herein. Echinoderes daenerysae sp. nov. is recognized by middorsal spines on segments 6 and 8 only, lateroventral spines on segments 6–9, laterodorsal and ventrolateral tubes on segment 2, lateroventral tubes on segment 5, lateral accessory tubes on segment 8, laterodorsal tubes on segment 9 and small laterodorsal tubes on segment 10. Echinoderes rhaegali sp. nov. has middorsal spines on segments 4, 6 and 8, lateroventral spines on segments 6–9, sublateral and ventrolateral tubes on segment 2, lateroventral tubes on segment 5, lateral accessory tubes on segment 8 and laterodorsal tubes on segment 10 in males. Echinoderes drogoni sp. nov. has middorsal spines on segments 4–8, lateroventral spines on segments 6–9, lateral accessory tubes on segment 5, glandular cell outlets type 2 in subdorsal, laterodorsal, sublateral and ventromedial positions on segments 2, in midlateral positions on segment 5, in sublateral positions on segment 8 and in subdorsal positions on segment 10; segment 11 has divided tergal plates. Moreover, types of other Arctic Echinoderes, including E. angustus, E. aquilonius, E. eximus, E. peterseni, E. svetlanae and E. tubilak, were re-examined and new information on glandular cell outlets type 2 is provided. Our results suggest that Arctic Echinoderes species have a circum-Artic distribution. Morphological analysis indicates that the occurrence of tubes may show intraspecific variation.

Endocytotic uptake of fluorescent dextrans by pollen tubes grown in vitro   总被引:5,自引:0,他引:5  
Summary Pollen tubes grow by tip growth, with high levels of exocytosis at the apex. The commercial availability of FITC labelled -linked dextrans provides a source of biologically inert tracers for endocytotic activity in pollen tubes. Growing tubes ofNicotiana andTradescantia were transferred to media containing 1% FD-4 for varying period of time before washing in control media and observation in a fluorescence microscope. Fluorescent material appeared to enter the pollen tubes only at the tip region, and to accumulate in vacuoles, starting with smaller vacuoles near the tip and spreading to the main vacuolated part of the tube. Mature tubes, with callose plugs, were only labelled up to the first complete plug from the tip, younger tubes without plugs were labelled into the pollen grain vacuole. The fluorescent material within the pollen tubes was shown to represent uptake of intact high molecular weight dextran by the following criteria: (i) free FITC and low molecular weight dextrans could not be detected in any of the media or pollen tubes using thin layer chromatography and (ii) pollen tube growth rates were unaffected by the fluorescent dextran, but were severely inhibited by low levels of free FITC.  Microtubules in pollen tubes are evident within the vegetative and generative cell cytoplasm. This observation led to the formulation of several hypotheses regarding the role of microtubules in cytoplasmic movement and the migration of the vegetative nucleus/generative cell along the pollen tube. The study of microtubular motor proteins in pollen tubes followed the discovery and characterization of an immunoreactive homolog of mammalian kinesin in tobacco pollen tubes. Recent identification of dynein-related polypeptides in pollen tubes of Nicotiana tabacum and pollen of Ginkgo biloba is a significant step in the definition of the role of microtubule function within pollen and pollen tubes. In flowering plants, the interaction of pollen tubes with female tissues is important for the accomplishment of double fertilization. Little information is known about the mechanisms that underlie signalling between pollen tubes and female tissues. In this study, two Arabidopsis pollen tube‐expressed CrRLK1L protein kinases, Buddha's Paper Seal 1 (BUPS1) and BUPS2, were identified as being required for normal tip growth of pollen tubes in the pistil. They are expressed prolifically in pollen and pollen tubes and are localized on the plasma membrane of the pollen tube tip region. Mutations in BUPS1 drastically reduced seed set. Most of the bups1 mutant pollen tubes growing in the pistil exhibited a swollen pollen tube tip, leading to failure of fertilization. The bups2 pollen tubes had a slightly abnormal morphology but could still accomplish double fertilization. The bups1 bups2 double mutant exhibited a slightly enhanced phenotype compared to the single bups1 mutants. The BUPS1 proteins could form homomers and heteromers with BUPS2, whereas BUPS2 could only form heteromers with BUPS1. The BUPS proteins could interact with the Arabidopsis pollen‐expressed RopGEFs in the yeast two‐hybrid (Y2H) and bimolecular fluorescence complementation (BiFC) assays. The results indicated that the BUPSs may mediate normal polar growth of pollen tubes in the pistil.  相似文献

 A biolistic particle delivery system was used to genetically transform pollen tubes of three species of white pine (Pinus aristata, P. griffithii and P. monticola). The introduced plasmid DNA contained the GUS coding sequence flanked by the 35S CaMV promoter and NOS terminator sequences. Successful gene delivery was demonstrated by transient GUS expression as evaluated by standard histochemical assay. Distance of target specimens significantly influenced transient GUS expression in all three species of white pine. A target distance of 6 cm resulted in a significant number of transformed pollen tubes in P. aristata and P. griffithii, while distances of 6 and 9 cm resulted in a significant number of transformed pollen tubes in P. monticola. Generally, the number of pollen tubes expressing GUS activity was higher in P. aristata than in P. griffithii and P. monticola. The possibility of using GUS-transformed pollen tubes in conjunction with in vitro fertilization in conifers was examined. Gene expression in pollen tubes was also examined under electron microscopy where the X-glu reaction product occurred as large crystalline electron-dense precipitates in the cytoplasm. Nuclear behavior in the developmental process of appressoria inAlternaria alternata was investigated. In pregerminated conidia, approximately 94% of the conidial cells were uninucleate. The migration of a nucleus into an elongating germ tube from a germinating conidium was confirmed after 2h of incubation at 24±1°C in PDB. Peak frequencies of binucleate and trinucleate germ tubes were detected 1 and 2h after the peak frequency of uninucleate germ tubes, respectively. Four-and five-nucleate germ tubes did not show marked peak frrequencies. A marked peak frequency of the six-nucleate germ tubes occurred about 1 h after the peak frequency of the trinucleate germ tubes, suggesting that the nuclei in the trinucleate germ tubes each divided once within 1 h. The significance of early establishment of multinucleate appressorial cells in the colonization of host plants by pathogenicA. alternata was discussed.  相似文献

The growth of pollen tubes of a sampling of Lilium species in styles of L. longiflorum incubated at 24 C for 48 hr after pollination indicated two types of interspecific incompatibility. Pollen tubes of two self-compatible species of section Leucolirion, the section including L. longiflorum, stopped growth abruptly upon reaching the stylar canal, were of abnormal morphology, and were incapable of continued growth with longer incubation. Pollen tubes of self-compatible or self-incompatible species of sections Sinomartagon, Pseudolirium, Liriotypus, and Daurolirion approached but did not exceed the length of intraspecific incompatible pollen tubes in styles of L. longiflorum. Pollen tube morphology was normal and tubes were capable of continued growth with additional incubation. Unilateral interspecific incompatibility occurred in reciprocal crosses between self-incompatible L. longiflorum and self-compatible L. regale and L. formosanum, but exceptions occurred in Aurelian hybrids. Incubation of interspecifically pollinated L. longiflorum styles at 39 C, which removes the self-incompatibility reaction, had no effect on interspecific incompatibility.  相似文献

The specificity of S-RNase-based self-incompatibility (SI) is controlled by two S-locus genes, the pistil S-RNase gene and the pollen S-locus-F-box gene. S-RNase is synthesized in the transmitting cell; its signal peptide is cleaved off during secretion into the transmitting tract; and the mature “S-RNase”, the subject of this study, is taken up by growing pollen tubes via an as-yet unknown mechanism. Upon uptake, S-RNase is sequestered in a vacuolar compartment in both non-self (compatible) and self (incompatible) pollen tubes, and the subsequent disruption of this compartment in incompatible pollen tubes correlates with the onset of the SI response. How the S-RNase-containing compartment is specifically disrupted in incompatible pollen tubes, however, is unknown. Here, we circumvented the uptake step of S-RNase by directly expressing S2-RNase, S3-RNase and non-glycosylated S3-RNase of Petunia inflata, with green fluorescent protein (GFP) fused at the C-terminus of each protein, in self (incompatible) and non-self (compatible) pollen of transgenic plants. We found that none of these ectopically expressed S-RNases affected the viability or the SI behavior of their self or non-self-pollen/pollen tubes. Based on GFP fluorescence of in vitro-germinated pollen tubes, all were sequestered in both self and non-self-pollen tubes. Moreover, the S-RNase-containing compartment was dynamic in living pollen tubes, with movement dependent on the actin–myosin-based molecular motor system. All these results suggest that glycosylation is not required for sequestration of S-RNase expressed in pollen tubes, and that the cytosol of pollen is the site of the cytotoxic action of S-RNase in SI.  相似文献

Despite the increasing importance of airlift fermentors, very little published information is available on how the geometric configurations of the draft tubes and the air-sparging system affect the mixing and oxygen transfer characteristics of the fermentor. A 14-L air-lift fermentor was designed and build with a fixed liquid height to diameter ratio of 1.5 utilizing four equally spaced air jets at the bottom. Two jet orifice sizes were used, 1.27 and 3.81 mm i.d., and for each jet size the following four geometric configurations were used: Single inner concentric draft tube, single outer concentric draft tube, two concentric draft tubes, and no draft tubes where the fermentor was operated as a shallow bubble column. It was found that the presence of draft tubes stabilized liquid circulation patterns and gave systemically higher mixing times than those obtained in the absence of draft tubes. In addition, the double draft tube geometry resulted in higher mixing times than the single draft tubes. For the power unit volume range 20 to about 250 W/m3 the larger 3.81-mm orifices gave systemically higher kL a values than the smaller 1.27-mm i.d. orifices. At 200 W/m3 the use of a single outer draft tube with the 3.81-mm orifices resulted in 94% increase in kL a values over that obtained with no draft tubes. However, the effect of draft tube geometry on kL a values when the 1.27-mm orifices were used was not significant. The air bubble formation characteristics at the jet orifices were found to be different, which reflected the differences observed in mass transfer and mixing characteristics. The power economy for oxygen transfer was found to be depend strongly on the orifice size and less on the geometric configuration of draft tubes.  相似文献

Tube‐nesting bees and wasps were collected with bamboo tube nest traps in a cherry blossom forest and an old secondary forest in Tama Forest Science Garden in the western suburbs of Tokyo. Curtain type traps, in which bamboo tubes were vertically arranged, attracted more bees and wasps to nests than bundle type traps, in which bamboo tubes were bundled haphazardly. The attractiveness of small tubes was similar between small tube traps, which were only composed of small tubes (about 4 mm in diameter), and size mixture traps, which set small tubes together with larger tubes, indicating that the presence of the larger species did not affect the nesting of small species. In the cherry forest, which was characterized by the sparse distribution of deciduous cherry trees with a thin canopy layer and rich growth of weeds, Megachilidae, Colletidae, Sphecidae and Eumenidae were common. In the shady secondary forest, where evergreen trees (Abies firma Sieb. et Zucc., Quercus glauca Thunb.) and shrubs (Aucuba japonica Thunb., Eurya japonica Thunb., Camellia japonica L.) were prevalent, Pompilidae was abundant. Among five species of Crabronidae recorded in this study, four were collected mostly in the cherry forest, whereas the most abundant, Trypoxylon malaisei Gussakovskij, was also common in the secondary forest. Likewise, Auplopus carbonarius (Scopoli) (Pompilidae) was common in the secondary forest and not rare in the cherry forest. These results suggest that the tube‐nesting bees and wasps can be a good bioindicator for monitoring environment.  相似文献

以异叶苦竹为材料,采用扫描电镜、荧光显微镜技术及传统的石蜡制片技术,解剖观察其花粉管生长途径及双受精过程。结果表明:(1)授粉后,花粉在柱头上吸水膨胀,约30 min即可萌发。(2)授粉1~2 h后花粉管可达到花粉长度的5~10倍,花粉管在柱头分支中进一步伸长,并开始伸入花柱中生长。(3)授粉后5 h,大量花粉管沿引导组织进入花柱基部与子房顶部之间的子房壁,有少量花粉管在子房壁与外珠被之间的缝隙中生长。(4)授粉后8 h,少量花粉管到达珠孔端。(5)授粉后15~18 h,精核与极核融合,形成初生胚乳核;精、卵核融合,形成合子。(6)授粉后20~30 h,仍可在花柱中见到大量呈束状的花粉管。(7)授粉后48 h,子房内的大部分花粉管出现解体,大多数花粉死亡。研究认为,精细胞到达胚珠的时间为8 h。  相似文献

The movement of organelles and vesicles in pollen tubes depends on F-actin. However, the molecular mechanism through which plant myosin XI drives the movement of organelles is still controversial, and the relationship between myosin XI and vesicle movement in pollen tubes is also unclear. In this study, we found that the siliques of the myosin xi-b/e mutant were obviously shorter than those of the wild-type (WT) and that the seed set of the mutant was severely deficient. The pollen tube growth of myosin xi-b/e was significantly inhibited both in vitro and in vivo. Fluorescence recovery after photobleaching showed that the velocity of vesicle movement in the pollen tube tip of the myosin xi-b/e mutant was lower than that of the WT. It was also found that peroxisome movement was significantly inhibited in the pollen tubes of the myosin xi-b/e mutant, while the velocities of the Golgi stack and mitochondrial movement decreased relatively less in the pollen tubes of the mutant. The endoplasmic reticulum streaming in the pollen tube shanks was not significantly different between the WT and the myosin xi-b/e mutant. In addition, we found that myosin XI-B-GFP colocalized obviously with vesicles and peroxisomes in the pollen tubes of Arabidopsis. Taken together, these results indicate that myosin XI-B may bind mainly to vesicles and peroxisomes, and drive their movement in pollen tubes. These results also suggest that the mechanism by which myosin XI drives organelle movement in plant cells may be evolutionarily conserved compared with other eukaryotic cells.  相似文献

Pollen tubes were grown under controlled temperatures in stigmas and styles which had been cut from the flowers at the point of juncture of the style with the ovary. At constant temperatures the tubes compatible with the styles grew rapidly at uniform rates. They showed accelerated growth with increase in temperature within the range 10°–30°C. Growth of incompatible tubes was irregular and slow and was neither measurably accelerated nor depressed within this range of temperatures. Routine tests for compatibility were conducted at 27°C; at this temperature compatible tubes grew through the entire length of the styles (ca. 58 mm) in 9 hours. Much of the incompatible pollen failed to germinate; that which did, grew to a maximum length ofca. 2 mm inca. 4 hours. The incompatible tubes did not even enter the styles but remained confined to the stigmas. ThreeS alleles were involved and conformed in their behavior to the gametophytic system of incompatibility. Colchicine-induced tetraploid derivatives of these plants showed no increase in compatibility.  相似文献

The study of winter-active small mammals beneath the snowpack has proved challenging for researchers because of the relative inaccessibility. We present a technique using hair tubes that permits the detection of small mammals active in the subnivean space. Hair tubes are cylindrical or funnel-shaped structures containing suitable bait and an adhesive surface that harvests hairs from small mammals as they attempt to reach the bait. Hair tubes eliminate many of the difficulties often associated with live trapping and permit the expansion of systematic sampling to larger scales than allowed by conventional live-trapping methods. The technique was used successfully to detect five small mammal species in the subnivean space in Kosciuszko National Park (KNP) in southeastern Australia. These included the common bush-rat, Rattus fuscipes; the dusky and agile antechinus, Antechinus swainsonii and A. agilis; the broad-toothed rat, Mastacomys fuscus; and the mountain pygmy possum, Burramys parvus. Although hair tubes have a number of limitations, such as not providing a measure of abundance or allowing the identification of individual animals, we believe that these limitations are balanced by the fact that the technique can be used at any spatial scale. Hair tubes are particularly suited to studies of animal distribution at the landscape-scale, because many hair tubes can be deployed and dispersed over large areas, and monitored on a regular basis by a small team of researchers. The technique also makes use of readily available, low-cost materials and could be easily adapted to a range of conditions and different target species.  相似文献
