Thin cell layers (TCLs) were cultured from inflorescences of diploid (2n=4x=48) and haploid (2n=2x=24)Nicotiana tabacum L. "Samsun" and the subsequent flowers formed in vitro were then compared to in vivo flowers. Plants derived from TCLs possessed flowers that were typical of their seed or androgenetically-derived counterparts, whereas de novo flowers from TCLs were abnormal when compared to their counterparts. The TCLs of haploid plants produced more flower buds than diploid TCLs, and did so in a shorter period of time. In vitro flowers and anthers at both ploidy levels were considerably smaller than the in vivo flowers; in vitro flowers also had variable numbers of anthers and pistils. The embryogenic capacity of anthers taken from in vivo diploid flowers was 5 times greater than that of in vitro diploid or haploid anthers. In vivo haploid anthers produced no embryoids, whereas in vitro haploid anthers did produce embryoids. Observations of mitotic cells in root tips of plants derived from anther cultures of in vitro haploid flowers revealed a mixoploid nature. Diploid meiosis was regular and haploid meiosis was irregular regardless of the origin (in vitro or in vivo) of the flowers.Supported by state Hatch funds.  相似文献

Echinacea purpurea (L.) is one of the important medicinal plant species. To obtain the tetraploid plants of Echinacea purpurea with improved medicinal qualities, the root tips of two true leaves seedlings were imbibed in 0.25 % (w/v) colchicine solution for 24, 48, 72, 96 and 168 h. The ploidy level of plants was determined by chromosome counting of root tip cells, and confirmed by flow cytometric analysis. Tetraploid induction occurred in seedlings treated for 24, 48 and 72 h at colchicine solution. The morphological, physiological, cytological, and phytochemical characteristics of diploid and colchicine-induced tetraploid plants were compared. Results indicated that tetraploid plants had considerable larger stomata, pollen grain, seed and flower. Moreover, chloroplast number in guard cells, amount of chlorophyll (a, b, and a + b), carotenoids as well as width and thickness of leaves were increased in tetraploids. However, stomata frequency, leaf index, plant height, and quantum efficiency of photosystem II in tetraploid were lower than diploid plants. High-performance liquid chromatography analysis showed that leaves of the tetraploid plants had more cichoric acid (45 %) and chlorogenic acid (71 %) than diploid plants. It was concluded that morphological and physiological characteristics can be used as useful parameters for preliminary screening of putative tetraploids in this species.  相似文献

Summary Transgenic shoots were regenerated from eight diploid potato hairy root clones obtained by transformation with Agrobacterium rhizogenes harboring next to its wild-type Ri-plasmid a binary vector containing the neomycin phosphotransferase and the -glucuronidase genes. The plants exhibited the typical hairy root phenotype. Of the plants isolated, 58% were tetraploid and 38% were diploid. Flowering and tuberization was much better in the diploid than in the tetraploid plants. Transgenic plants formed a significantly larger root system when grown on kanamycin-containing medium as compared to growth on kanamycin-free medium. Direct evidence for genetic transformation was obtained by opine, neomycin phosphotransferase and -glucuronidase assays, and by molecular hybridization. Fourteen flowering diploid plants were reciprocally crossed with untransformed S. tuberosum plants, but only six were successful. Seedlings obtained from four crosses showed that all traits were transmitted to the offspring. Molecular analysis confirmed the presence of multiple integrations (copies) of both vector T-DNA and Ri-T-DNA. The genetic data, furthermore, suggest that the traits derived from Ri-T-DNA and binary vector T-DNA are linked, as no recombination between the different traits was observed.  相似文献

Shoot multiplication was obtained in vitro within 20–25 d from shoot tip expiants of 30 d old aseptically germinated seedlings of Coleus forskohlii Briq., using 2 mg/1 of 6 benzylaminopurine (BA). Shoot multiplication was further enhanced with the gradual decrease in the level of BA, and its final omission after 4 months. Different auxins supplemented at the level of 0.05 mg/1 with BA did not yield better results. Seven regenerated plants showed only diploid cells in their root tips, while three plants did not. Of these, two were diploid with occasional aneuploid cells. In one plant 32 chromosomes were observed. The potential of shoot culture in vitro and use of micropropagated plants for the production of forskolin has been demonstrated.  相似文献

Summary Anther culture of the Easter Lily (Lilium longiflorum; 2n=2x=24) was attempted in order to evaluate its potential in generating haploids for the production of hybrid cultivars. The effects of genotype, temperature (low temperature treatment of buds and high temperature treatment of cultures), sucrose concentration and growth regulators were tested. The most important factors for callus induction were the genotype and the presence of 2,4-dichlorophenoxyacetic acid. Pre-treatments at low or high temperature had no apparent effect, while high sucrose concentration was inhibitory. Callus was derived from 28 of the 108 genotypes tested and plants were regenerated. Phenotypic variations were observed among these regenerants. Somatic chromosome numbers were determined in 42 plants derived from 10 donor genotypes. Thirteen plants were diploid and 29 were mixoploid with chromosome numbers ranging from 11 to 26. Four of the mixoploid plants had a high proportion of cells with haploid chromosome numbers, particularly at early stages of development. Meiosis was examined in plants with flower buds. Most plants had 12 bivalents at Metaphase I, but also aneuploids were observed. Other irregularities included bridges and laggards at Anaphase I. The occurrence of high frequencies of haploid cells (up to 80%) in root tips suggests that some plants may be of gametic origin. In contrast with the well document role of proteoid root formation and carboxylate exudation in acclimation to P deficiency in white lupin (Lupinus albus L.), their role under other nutrient deficiencies and their ecological significance are still poorly understood. In the present work, differences in proteoid root formation, exudation of carboxylates by root clusters, non-proteoid and proteoid root tips by using a non-destructive method, and concentrations of organic acids in the tissues of plants grown in the absence of P, Fe or K were studied. Proton release from roots increased soon after withdrawing Fe from the medium; within three days the solution pH decreased from 6 to about 4, and this increased release in protons continued until the end of the experiment. Acidification appeared much later, on the 10th day and the 14th day after withdrawal of P and K, respectively; the extent of the acidification was also weaker than under –Fe (5.2 for –P and 5.7 for control on the 10th day; 6.0 for –K and 6.1 for control on the 14th day). Root clusters formed when plants were grown under –P and –Fe, but not under –K conditions. The root clusters developed sooner under –Fe conditions, but the number of clusters was far less than under –P. Under P deficiency, root clusters released mainly citrate, but also some malate; while the major organic acid released by root tips of both non-proteoid and proteoid roots was malate. However, under Fe deficiency, the majority of the organic acids exuded both by the root clusters and root tips was malate, whereas only a small amount of citrate was detected. The release rate of citrate by – P root clusters was greater than that by – Fe root clusters. Moreover, the release rate of malate was greater in –Fe root clusters than in –P root clusters, but the opposite was found in proteoid root tips, i.e. faster in –P than in –Fe proteoid root tips. The significances of proteoid root formation and release of organic acids in acclimation to different nutrient deficiencies for white lupin plants are discussed.  相似文献

嫁接辣椒根系特征及根际土壤酶活性与青枯病抗性的关系   总被引:1,自引:0,他引:1  
以‘卫士’辣椒为砧木,‘新丰2号’为接穂嫁接,通过人工接种青枯病菌研究嫁接和自根辣椒根系特征、根际土壤微生物及酶活性的变化,探讨嫁接辣椒的抗病机理.结果显示:接种青枯病菌前,嫁接辣椒的根系重量、总长度、总体积、表面积、根尖数和分叉数均显著高于自根苗,根系活力、根际土壤放线菌数量和比例,以及根际土壤酶(多酚氧化酶、过氧化物酶和脱氢酶)活性也明显高于自根苗.接种青枯病菌后,嫁接辣椒的根系受伤程度较自根苗轻,根系重量、总长度、总体积、表面积、根尖数和分叉数的降低幅度均显著小于自根苗,根系活力、根际土壤微生物数量、放线菌比例及土壤酶活性明显大于自根苗.研究表明,嫁接辣椒根系发达,根系活力增强,根际土壤放线菌比例增加及酶活性提高是其青枯病抗性增强的重要原因.  相似文献

Root tips of Fe-deficient and Fe-sufficient sugar beet plants grown in hydroponics have been used to study the changes in the amount and activity of the cytosolic enzyme phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31). Phosphoenolpyruvate carboxylase activity in extracts of the yellow Fe-deficient root tips was, at pH 7.3, 30-fold higher (when expressed on a FW basis) and 7.1-fold higher (when expressed on a protein basis) than that found in the extracts of Fe-sufficient root tips. The amount of phosphoenolpyruvate carboxylase protein determined by immuno-blotting was, on a protein basis, 35-fold larger in the yellow zone of Fe-deficient root tips than in the Fe-sufficient root tips. The inhibition of the phosphoenolpyruvate carboxylase activity by 500 m malate was 41 and 58% in the extracts Fe-deficient and Fe-sufficient roots. The possibility that post-translational regulation of phosphoenolpyruvate carboxylase may occur mediated through phosphorylation, was studied by immunological detection of phosphoserine residues in root tip extracts.  相似文献

为了探讨岷江干旱河谷丛枝菌根真菌(AMF)对寄主植物幼苗根系的影响,通过接种购买的AMF摩西球囊霉菌(Funneliformis mosseae)到优势乡土灌木小马鞍羊蹄甲(Bauhinia faberi var.microphylla)幼苗,在重度、中度和轻度干旱条件下培养3个月,研究不同干旱条件下AMF对幼苗根系形态特征、结构特征、功能性状的影响。方差分析结果表明:(1)3种干旱胁迫条件下,接菌均显著增加了幼苗的根总长、根表面积、根分枝数、根尖数(P0.001),在中度胁迫和轻度胁迫下,接菌显著促进根鲜重、根体积的增加(P0.001),轻度胁迫条件下接菌幼苗的根鲜重、根总长、根表面积、根体积、根尖数最高并显著高于其它处理,但接菌与未接菌的根平均直径之间没有显著差异;(2)接菌幼苗根系趋向于叉状分支结构,在重度胁迫时,叉状分支趋势更显著(P0.001);(3)接菌幼苗的根比例都显著小于未接菌的,但幼苗比根长不存在显著差异。相关分析结果表明:菌根侵染率与根鲜重、根总长、根表面积、根体积、根分枝数、根尖数呈极显著正相关(P0.001),与拓扑指数、根比例呈极显著负相关(P0.001)。研究表明,在干旱条件下,AMF虽然没有提高生长初期的根系的吸收效率,但接种AMF显著影响幼苗根系形态特征和结构特征,更利于植物适应干旱环境,并且AMF对幼苗根系的促生作用随着干旱胁迫程度减轻而提高。  相似文献

Somaclonal and in vitro mutagen-induced variability was studied in grapevine. Plants of Vitis vinifera cv. Podarok Magaracha were regenerated from leaf explants through somatic embryogenesis. Chromosome counts of root tips was used for screening of regenerated plants. Among 242 studied plants, six (2.5%) tetraploids (2n=4x=76) were identified; all others were diploid (2n=2x=38). Neither chimeral, nor aneuploid plants were observed. Gamma-irradiation (5-100 Gy) increased tetraploid plant formation frequency of primary (7%) and embryogenic calluses (7.6%) and some aneuploid plants were also found. Colchicine treatment was not effective for the production of tetraploid plants. Variability among regenerated plants was also found after field testing. The effect of direct inoculation of seeds with the plant growth promoting rhizobacteria (PGPR) Azospirillum lipoferum CRT1 was assessed on maize (Zea mays) grown for 35 days after sowing (d.a.s.) in controlled conditions (greenhouse) in a luvisol soil from south-eastern France. WhinRhizo® software was used to describe the following changes in the root system morphology for each plant: distribution and average root diameter, root surface and the number of tips. The stress at breakage and stiffness of the roots in tension were also determined. Evaluation of biochemical components of roots was achieved by direct Attenuated Total Reflectance (or reflection) (ATR)-Fourier transform infrared (FTIR) on root section. Inoculated roots exhibited significantly larger numbers of tips and extending surface to rhizosphere when compared to controls. Measured mechanical parameters of inoculated roots showed a slight increase in rupture stress up to the largest diameter (1.2 mm) when compared to controls. Stiffness (Young’s modulus) values were nearly constant for inoculated plants with higher values than for non-inoculated plants at day 26 and day 35. Using Principal Components Analysis of ATR-FTIR profiles, the polysaccharide enrichment of inoculated roots compared to controls was found at day 35. Noticeable absorbance at wavenumber specific to aromatic ether (lignin) was observed in control plants. All these data had a pattern of immature root properties, when maize was inoculated with Azospirillum lipoferum CRT1. Observed modifications of root development are possibly conducive to unseen beneficial effects, like water retention, resistance to mechanical stress, or root litter quality. Studies on more mature plants are required to assess if the differences between inoculated and control plants would persist or become accentuated with time until harvest.  相似文献

Potted maize seedlings were subjected to a single period of water stress. As the severity of water stress increased, measurements were made of leaf and root solute and water potentials, leaf diffusive conductance and leaf and root growth. After day four of the drying cycle, the rate of leaf extension and the development of leaf area were reduced. This reduction correlated well with a reduction in leaf turgor which occurred at this time. A significant accumulation of solutes in the root tips of the unwatered plants resulted in the maintenance of root turgor for the duration of the water stress treatment. Root growth of the unwatered plants was also maintained as the severity of water stress increased. A mild degree of water stress resulted in a net increase in root growth compared to the situation in well-watered plants. Climate change (elevated atmospheric CO2, and altered air temperatures, precipitation amounts and seasonal patterns) may affect ecosystem processes by altering carbon allocation in plants, and carbon flux from plants to soil. Mycorrhizal fungi, as carbon sinks, are among the first soil biota to receive carbon from plants, and thereby influence carbon release from plants to soil. One step in this carbon release is via fine root and mycorrhizal turnover. It is necessary to know the lifetime and temporal occurrence of roots and mycorrhizae to determine the capacity of the soil ecosystem to sequester carbon assimilated aboveground. In this study, ponderosa pine (Pinus ponderosa Laws) seedlings were grown under three levels of atmospheric CO2 (ambient, 525 and 700 mol CO2 mol-1) and three levels of annual nitrogen additions (0,100 and 200 kg N ha-1) in open-top chambers. At a two-month frequency during 18 months, we observed ectomycorrhizal root tips observed using minirhizotron tubes and camera. The numbers of new mycorrhizal root tips, the numbers of tips that disappeared between two consecutive recording events, and the standing crop of tips at each event were determined. There were more mycorrhizal tips of all three types seen during the summer compared with other times of the year. When only the standing crop of mycorrhizal tips was considered, effects of the CO2 and N addition treatments on carbon allocation to mycorrhizal tips was weakly evident. However, when the three types of tips were considered collectively, tips numbers flux of carbon through mycorrhizae was greatest in the: (1) high CO2 treatment compared with the other CO2 treatments, and (2) intermediate N addition treatment compared with the other N addition treatments. A survival analysis on the entire 18 month cohort of tips was done to calculate the median lifetime of the mycorrhizal root tips. Average median lifetime of the mycorrhizal tips was 139 days and was not affected by nitrogen and CO2 treatments.  相似文献

The impact of asparagus virus I (AV-I), a potyvirus, and asparagus virus II (AV-II), an ilarvirus, on micropropagation of field-grown asparagus was studied. Apical shoot tips excised from singly or doubly-infected plants were slow to develop roots and had a 15 to 75% reduction in survival in culture, respectively, compared to those excised from virus-free plants. The four virus infection groups were ranked: virus-free >AV-II>AV-I>AV-I & II for capacity of explants to both root and survivein vitro. Micropropagated plants infected with AV-II exhibited slight reductions in fresh and dry weights, with greater reductions associated with infection with AV-I and double infection, compared to the virus-free controls. Eighty-one virus-infected apical shoot tips yielded 7 (8.6%) virus-free clones, as determined by rub inoculation on indicator plants.  相似文献

Cytological and cytophotometric analysis of root tips of regenerated plants, derived from rhizome expiant ofKniphofia nelsonii andKniphofia uvaria, revealed marked difference in behaviour of chromosomes and level of 4C nuclear DNA content. Karyotypic stability could be retained in all 52 regenerants of K. nelsonii whereas inK. uvaria out of 75 regenerants analysed, 12 plants were exclusively diploid and the rest 63 plants were predominantly diploid comprising variable amount of aneuploid and tetraploid cells. Cytological data was further confirmed by nuclear DNA content estimation. Alteration in the structure of chromosomes could also be noted in 57 regenerants ofK. uvaria giving rise to two new karyotypes. The use of polysomatic tissue for securing variantsin vitro inK. uvaria has been suggested.  相似文献

The interaction of boron (B) and aluminium (Al) was investigated in 5-day-old seedlings of soybean cv. Maple Arrow. Al treatment inhibited root elongation and callose formation in root tips particularly after 4-h Al treatment. After 10 and 24 h, both parameters indicated increasing recovery from Al stress. B deficiency aggravated Al toxicity compared with B sufficiency. B deficiency did lead to an increase in unmethylated pectin in the first 3 mm of the root tip. This increase in potential binding sites is reflected in generally higher Al contents in root tips of B-deficient plants. A fractionated extraction of Al from the root tips showed that citrate-exchangeable and non-exchangeable Al steeply increased up to 4 h, but then decreased after 10- and 24-h Al treatment faster in B-sufficient than in B-deficient plants. This decrease of Al contents can be explained by an Al-enhanced release of citrate from the root tips after 10-h Al treatment. However, the citrate exudation rate was the same (after 10 h) or even lower (after 24 h) in B-sufficient plants and thus cannot explain the faster decrease in Al contents of the root tips compared with the B-deficient plants. We, therefore, propose that under B deficiency, Al is more strongly bound by the pectic network of the cell wall of the root tips, which delays or prevents the recovery from initial Al stress through exudation of citrate, and thus explains the greater Al sensitivity of B-deficient common bean roots.  相似文献

Zeatin O-glycosides have been reported as inactive and stable storage forms of cytokinins whose concentrations increase in cold stressed plants. Zeatin O-glycosides accumulation in developing bean seeds has been correlated with an increase of zeatin O-glycosyltransferase , which is specific to trans-zeatin, and catalyzes the conjugation of zeatin O-glycosides. When Phaseolus vulgaris and Zea mays seedlings were grown for 3 days at 25 and then incubated at 4 or 10 for 6 days no further growth was observed in roots. Hypertrophy was observed in the root tips of both species. In shoot-hypocotyl complexes, in contrast, growth occurred when seedlings were incubated at 10 . Western analysis, with Mabs specific to zeatin O-glycosyltransferase, detected antigenically related proteins in roots, shoot tips and cotyledons after seedlings were cold stressed for 1–6 days at 4 or 10 . Immunolocalization, of both maize and bean root sections grown at 25 revealed antigenically related proteins that were detected at low levels in cortical cells. The signal intensified upon cold stress. The localization of zeatin O-glycosyltransferase in Z. mays root tips was directly comparable to the distribution of the zeatin O-glycosides. The enzyme was detected in the nucleus, cytoplasm, and closely associated with the plasma membrane and in the cell wall of Z. mays root cells. Southern analysis suggested that more than one gene in Z. mays that were homologous to zeatin O-glycosyltransferase in P. vulgaris. Zeatin O-glycosyltransferase may be involved in modulation of cytokinins under cold stress.  相似文献

Reclaimed mine soils of the Lusatian mining district are characterised by small-scale heterogeneous distribution of lignite fragments of varying size embedded in a matrix of Tertiary and Quaternary sandy material. Despite amelioration with basic fly ashes, ongoing pyrite oxidation and the subsequent acidification generate a high physical and chemical heterogeneity within the substrate, which could negatively affect root proliferation. We hypothesised that this limitation for the root system may be compensated for by intensive exploration of the porous lignite fragments by roots and/or mycorrhizal hyphae to access water and nutrients stored in these fragments. To test this hypothesis, we compared growth, shoot nutrient content, and root distribution of mycorrhizal and non-mycorrhizal Pinus sylvestris L. seedlings in lignite-containing and lignite-free sandy substrate. Rhizotrons used for this experiment were filled with a sandy matrix with 6–9 evenly distributed spots of lignite fragments. Treatments included different levels of water and nutrient availability. After 8 months of growth, root tip vitality as well as growth and shoot nutrient concentration of the plants was higher for treatments with lignite spots in the sandy substrate than for sandy substrate without such amendments. Compared to the non-mycorrhizal plants, the seedlings inoculated with Paxillus involutus (Batsch) Fr. had a higher root dry mass, an increased number of root tips and a higher root length. These results confirm our hypothesis that the lignite fragments are an important nutrient and water reservoir for plants in these mine soils and they indicate that mycorrhizal colonisation may allow an intensive exploration of porous lignite fragments by mycorrhizal hyphae.  相似文献

Development of tobacco root systems was characterized under controlled environmental conditions by use of morphometric root analysis. According to the classification scheme of this system, roots terminating in apical meristems are defined as first-order roots. Elements of second-order roots begin where two first-order roots merge, and so forth. Growth of root systems was similar for susceptible and resistant tobacco cultivars in nonautoclaved and autoclaved soils. During 15 days of growth subsequent to transplanting of 2-week-old plants, relative multiplication and extension rates of first-order and second-order roots were constant. Apparent unit extension rates of first-order and second-order root elements increased through 15 days of root system growth. Classification of tobacco root systems by the morphometric scheme provided a useful means of partitioning susceptibility of tissues to infection byPhytophthora parasitica var.nicotianae. Zoospores applied at the tips of first-order roots were most successful in causing infections; 73.3% of the roots inoculated with 16 zoospores per root tip became infected. Percentages of infections after inoculation of first-order root tissues 2 cm behind root tips or after inoculation of second-order roots were 10 and 4.3%, respectively.Florida Agricultural Experiment Station, Journal Series Paper 8106.  相似文献
