The northern red-legged frog Rana aurora aurora and the California red-legged frog Rana aurora draytonii are traditionally classified together in the same species group. Ten peptides with antimicrobial activity were isolated from norepinephrine-stimulated skin secretions of R. aurora draytonii and purified to near homogeneity. The peptides were identified as belonging to the ranatuerin-2 family (two peptides), brevinin-1 family (four peptides), temporin family (three peptides), and a novel peptide, RV-23 (RIGVLLARLPKLFSLFKLMGKKV) that has limited structural similarity to the bee venom peptide, melittin. This distribution of peptides contrasts with that found previously in skin secretions from R. aurora aurora collected under the same conditions and at the same time of year (one ranatuerin-2 peptide, two brevinin-1 peptides, and one temporin peptide). The variation in amino acid sequences between corresponding R. aurora draytonii and R. aurora aurora peptides is comparable with the variation in sequences of orthologs from other members of the Amerana group of New World ranid frogs (Rana boylii, Rana muscosa, and Rana luteiventris). It is proposed, therefore, that the red-legged frogs should be regarded as separate species (R. aurora and R. draytonii) within the Amerana group rather than conspecific subspecies. The data emphasize that amino acid sequences of antimicrobial peptides in skin secretions may be used to infer taxonomic and phylogenetic relationships between species of ranid frogs.  相似文献

Apoptosis (programmed cell death) occurs during normal development of anurans in organs such as gills, gut, and tail. For example, apoptotic cells have been reported in the luminal epithelium along the length of the digestive tract of both larvae and frogs; however, timing of the peak number of such cells varies in different species. The purpose of the present study was to ascertain whether apoptosis also varies by species during metamorphic restructuring of the skin (as larval epithelium is replaced by adult epidermis). To determine this, cross‐sections of dorsal skin from representative larval stages and frogs of Rana pipiens, R. catesbeiana, and Ceratophrys ornata were incubated with monoclonal antibody against active caspase‐3, one of the main enzymes in the apoptotic cascade. We observed apoptotic cells in the epidermis of the skin of the three species and found that such cells were more numerous in larval stages than in frogs and more abundant in the two ranid species than in C. ornata. The regenerative capacity of limbs was investigated by amputation of limbs at the zeugopodium in postmetamorphic froglets and adults of various sizes in four species of Japanese frogs, all of which showed some regeneration at these ages. In Hyla arborea japonica and Rana brevipoda porosa most young froglets regenerated their limbs well; however, the rate of regeneration decreased with the age of amputation, and the limb became nonregenerative in adults. Limbs of adults in Rana rugosa and R. japonica, on the other hand, exhibited good regeneration. All of the regenerates in the four species were heteromorphic, consisting histologically of well-developed cartilaginous rods surronded by connective tissue and skin. Limited development of muscle was appartment in regenerates of the three ranid species. The relations between body size, innervation of limbs, and regenerative capacity are discussed.  相似文献

A microsatellite library was developed using genomic DNA of the endangered dusky gopher frog, Rana sevosa. Polymerase chain reaction (PCR) primers and conditions are presented for R. sevosa (eight loci) and two sister taxa — other gopher frogs, Rana capito (seven loci) and crawfish frogs, Rana areolata (three loci). Polymorphism of each microsatellite locus was evaluated for each species. All loci have moderate to high genetic variation in terms of allelic richness (four to 10 alleles per locus), observed heterozygosity (0.595–0.946), and expected heterozygosity (0.531–0.856).  相似文献

Non-native fish and bullfrogs (Lithobates catesbeianus) are frequently cited as contributing to the decline of ranid frogs in the western United States, so we hypothesized that non-native species, habitat, or a combination of these relate to the probability of local extinction for northern red-legged frogs (Rana aurora) in Oregon, USA. We also hypothesized that the probability of colonization relates to land use, wetland size, or riparian forest. In a 5-yr study, we found no support for an effect of non-native species on northern red-legged frogs. Instead, probability of local extinction decreased with the extent of emergent vegetation and riparian forest. This finding suggests that managers consider the role of habitat when confronting non-native species problems. © 2010 The Wildlife Society.  相似文献

Recent phylogenetic studies of Southeast Asian megophryid Leptobrachium, while clarifying (1) distinct specific status of three Philippine populations and (2) high genetic diversities within Bornean Leptobrachium montanum, posed two questions, (1) relationships and divergence histories of two Philippine species and Bornean Leptobrachium gunungense, and (2) possible discordance between phylogenetically and morphologically defined lineages. In order to solve these questions, and especially reviewing current taxonomy of Bornean species, we estimated the phylogenetic relationships of endemic Bornean species together with their putative relatives from Philippines and Sumatra, using 2451 bp sequences of the 12S rRNA, tRNA(val), and 16S rRNA of mitochondrial DNA genes. With respect to Leptobrachium hasseltii and Leptobrachium chapaense, lineages from Borneo, Philippines, and Sumatra formed a monophyletic group with Leptobrachium lumadorum from Mindanao as the basal clade, while two other Philippine species from Palawan and Mindoro formed a clade and nested in Bornean lineages. Sister species relationship of the two Philippine species and L. gunungense is not supported, rejecting the hypothesis of Philippine origin of L. gunungense. Phylogeny does not conform to morphological variation, and the topotypic L. montanum and one lineage of Leptobrachium abbotti are genetically very close despite their clear difference in ventral color pattern. Furthermore, each of these species forms a paraphyletic group and contains several lineages, each of which is a candidate of good species. These results clearly indicate that current taxonomy of Bornean species based on several morphological characteristics requires complete revision. Detailed studies on adult and larval morphology, as well as acoustic characteristics, are necessary to evaluate the taxonomic status of all lineages recovered.  相似文献

Based on partial sequences of the 12S and 16S ribosomal RNA genes, we estimated phylogenetic relationships among brown frogs of the Rana temporaria group from China. From the phylogenetic trees obtained, we propose to include Rana zhengi in the brown frogs. Monophyly of the brown frogs was not unambiguously supported, but four well-supported clades (A, B, C, and D) always emerged, although relationships among them remained unresolved. Clade A contained brown frogs with 24 chromosomes and was split into two distinct subclades (Subclade A-1: R. chensinensis and R. huanrenensis; Subclade A-2: R. dybowskii). Polytomous relationships among populations of R. chensinensis and R. huanrenensis suggested the necessity of further taxonomic assessment. Rana kunyuensis proved to be the sister group to R. amurensis, and these two species formed Clade B. Clade C was composed of R. omeimontis and R. chaochiaoensis, and Clade D included R. sauteri, which has been placed in other ranid genera. These relationships did not change after adding published data, and monophyly of Subclade A-1, A-2, and other East Asian brown frogs with 24 chromosomes (R. pirica and R. ornativentris) was ascertained, though their relationships were unresolved. Clade C, together with R. japonica and R. longicrus, also formed a monophyletic group. Brown frogs related to Clades A and C were estimated to have dispersed from continental Asia to adjacent regions through multiple events.  相似文献

The Chinese bamboo leaf odorous frog (Rana (Odorrana) versabilis) and the North American pickerel frog (Rana palustris) occupy different ecological niches on two different continents with no overlap in geographical distribution. R. palustris skin secretions contain a formidable array of antimicrobial peptides including homologs of brevinin-1, esculentin-1, esculentin-2, ranatuerin-2, a temporin and a family of peptides considered of unique structural attributes when isolated, palustrins 1-3. Here we describe the structures of mature peptides and precursors of eight putative antimicrobial peptides from the skin secretion of the Chinese bamboo leaf odorous frog (Rana (Odorrana) versabilis). Each peptide represents a structural homolog of respective peptide families isolated from R. palustris, including two peptides identical in primary structure to palustrin 1c and palustrin 3b. Additionally, two peptides were found to be structural homologs of ranatuerin 2B and ranatuerin 2P from the closely-related North American species, Rana berlandieri (the Rio Grande leopard frog) and Rana pipiens (the Northern leopard frog), respectively. Both palustrins and ranatuerins have hitherto been considered unique to North American ranid frogs. The use of primary structures of amphibian skin antimicrobial peptides is thus questionable as a taxonomic device or alternatively, the micro-evolution and/or ancestry of ranid frogs is more highly complex than previously thought.  相似文献

Temporal characteristics of advertisement calls of water frogs of 10 localities in Turkey, Syria, Israel and Egypt were compared with those of Rana ridibunda in Kazakhstan, Armenia and Greece (Thrace) as reference populations. These study sites include the type localities of R. ridibunda , R. r. caralitana , R. esculenta var. bedriagae und R. levantina . The temperature-dependent as well as the temperature-independent call parameters clearly revealed two species. R. ridibunda is represented by the three reference populations and, in addition, a population in central Turkey. The other populations (in Turkey, Syria, Israel and Egypt) represent R. bedriagae . R. bedriagae is the oldest available name for water frogs of these regions and was given priority over R. r. caralitana and R. levantina .  相似文献

Aim We examine the genetic diversity within the lizard genus Gekko in the Philippine islands to understand the role of geography and geological history in shaping species diversity in this group. We test multiple biogeographical hypotheses of species relationships, including the recently proposed Palawan Ark Hypothesis. Location Southeast Asia and the Philippines. Methods Samples of all island endemic and widespread Philippine Gekko species were collected and sequenced for one mitochondrial gene (NADH dehydrogenase subunit 2) and one nuclear gene (phosducin). We used maximum likelihood and Bayesian phylogenetic methods to derive the phylogeny. Divergence time analyses were used to estimate the time tree of Philippine Gekko in order to test biogeographical predictions of species relationships. The phylogenetic trees from the posterior distribution of the Bayesian analyses were used for testing biogeographical hypotheses. Haplotype networks were created for the widespread species Gekko mindorensis to explore genetic variation within recently divergent clades. Results Both maximum likelihood and Bayesian phylogenetic analyses indicated that Philippine Gekko species are a diverse clade with a long history in the archipelago. Ancestral range reconstruction and divergence time analyses suggest a Palawan microcontinental origin for this clade, coinciding with Palawan’s separation from Asia beginning 30 Ma, with subsequent diversification in the oceanic Philippine islands. The widespread species G. mindorensis and G. monarchus diversified in the late Miocene/early Pliocene and are potentially complexes of numerous undescribed species. Main conclusions The view of the Philippine islands as a ‘fringing archipelago’ does not explain the pattern of species diversity in the genus Gekko. Philippine Gekko species have diversified within the archipelago over millions of years of isolation, forming a large diverse group of endemic species. Furthermore, the Philippine radiation of gekkonid lizards demonstrates biogeographical patterns most consistent with stochastic colonization followed by in situ diversification. Our results reveal the need to consider deeper time geological processes and their potential role in the evolution of some Philippine terrestrial organisms.  相似文献

Suggestions that the extinct Vegas Valley leopard frog (Rana fisheri = Lithobates fisheri) may have been synonymous with one of several declining species have complicated recovery planning for imperiled leopard frogs in southwestern United States. To address this concern, we reconstructed the phylogenetic position of R. fisheri from mitochondrial and nuclear sequence data obtained from century-old museum specimens. Analyses incorporating representative North American Rana species placed archival specimens within the clade comprising federally Threatened Chiricahua leopard frogs (Rana chiricahuensis = Lithobates chiricahuensis). Further analysis of Chiricahua leopard frogs recovered two diagnosable lineages. One lineage is composed of R. fisheri specimens and R. chiricahuensis near the Mogollon Rim in central Arizona, while the other encompasses R. chiricahuensis populations to the south and east. These findings ascribe R. chiricahuensis populations from the northwestern most portion of its range to a resurrected R. fisheri, demonstrating how phylogenetic placement of archival specimens can inform recovery and conservation plans, especially those that call for translocation, re-introduction, or population augmentation of imperiled species.  相似文献

The diurnal behaviour of frogs was recorded quantitatively in a habitatlike experimental environment by a new method of automatic registration via thermocouples. Three species with different habitat preferences during their summer activity period were chosen: the terrestrial common frog Rana temporaria LINNÈ and the semiaquatic water frog Rana lessonae CAMERANO and Rana ridibunda PALLAS. The activity, the location and the skin temperatures of these frogs were recorded continuously in four different temperature ranges (8.1° C–31.0° C) within the temperature span of the summer activity period. The thermal requirements and behavioural adaptations to the habitat of each species were analysed. The experimental results coincided with field observations and showed some details not yet known. The northern species R. temporaria exhibited a distinctive behavioural thermoregulation including cooling behaviour at high temperatures and warming behaviour at low temperatures during the day. In all temperatures tested these frogs remained on land changing their preferred location from the open area to the hiding places. The main activity period shifted from night to day with decreasing temperatures. The more southern species R. lessonae preferred higher temperatures than the other species showing basking behaviour during the day independent of the ambient temperature. With decreasing temperatures the preferred location changed from the shore to the water and the activity during the night almost disappeared. The closely related R. ridibunda tolerated high temperatures too, but only rarely basked. This species, however, reacted contrarily to decreasing temperatures: it changed its preference from the water to the land. This behavioural differenciation between the water frog species may be of importance for the actual habitat preference of these central european water frogs.  相似文献

Canadian northern leopard frogs (Rana pipiens) and bullfrogs (Rana catesbeiana) were acclimated to 3 degrees C and submerged in anoxic (0-5 mmHg) and normoxic (Po(2) approximately 158 mmHg) water. Periodic measurements of blood Po(2), Pco(2), and pH were made on samples taken anaerobically from subsets of each species. Blood plasma was analyzed for [Na(+)], [K(+)], [Cl(-)], [lactate], [glucose], total calcium, total magnesium, and osmolality. Blood hematocrit was determined, and plasma bicarbonate concentration was calculated. Both species died within 4 d of anoxic submergence. Anoxia intolerance would rule out hibernation in mud, which is anoxic. Both species survived long periods of normoxic submergence (R. pipiens, 125 d; R. catesbeiana, 150 d) with minimal changes in acid-base and ionic status. We conclude that ranid frogs require a hibernaculum where the water has a high enough Po(2) to drive cutaneous diffusion, allowing the frogs to extract enough O(2) to maintain aerobic metabolism, but that an ability to tolerate anoxia for several days may still be ecologically meaningful.  相似文献

The ontogeny of ventral pigmentation of two species of leopard frog, Rana pipiens and R. chiricahuensis, was examined by light microscopy and transmission electron microscopy to reveal how the unusual melanistic ventral pigmentation of R. chiricahuensis is achieved at the cellular level. Ventral skin of R. pipiens is always white. Ventral skin of adult R. chiricahuensis is white when frogs are background-adapted to a white substrate, but ventral skin becomes nearly as dark colored as the dorsal skin when frogs darken in response to a black background. Skin samples from tadpoles of both species, newly metamorphosed frogs, and adult frogs were analyzed for chromatophore composition and distribution. Ventral skin of R. pipiens larvae, newly metamorphosed frogs, and adults and of R. chiricahuensis larvae was white due to abundant iridophores and no melanophores. Melanophore density in the ventral integument of R. chiricahuensis was 9.1 ± 2.8/mm2 in newly metamorphosed frogs and 87.0 ± 4.8/mm2 in adult frogs. A new host and geographic locality record is reported for tetrathyridia of Mesocestoides sp. in two species of ranid frogs (Rana berlandieri and R. pipiens) from Texas and New York, respectively. Tetrathyridia were found encapsulated in liver and mesenteries of the hosts. Morphological examination and experimental inoculation of these tetrathyridia into mice demonstrated the absence of capacity for asexual proliferation. Overall prevalence of infection was low in anurans from Arkansas, Texas and New York, but intensities can be generally high. In addition, a summary of frogs and toads from North America reported as hosts of tetrathyridia of Mesocestoides sp. is presented.  相似文献

Hybrid zones are either distributed along clines or in a mosaic of patches. This distribution may depend upon variation in taxon habitat use. Habitat use and distribution of diverse taxa of water frogs (Rana ridibunda, R. lessonae, R. perezi, R. kl. grafi and R. kl. esculenta) in France are analysed to determine whether water frog complexes conform to the mosaic or clinal model. Biogeographical scenarios may be invoked in order to explain the distribution of water frogs. However, the distribution of R. perezi and R. kl. grafi, being restricted to regions characterized by Mediterranean or Oceanic climatic conditions, suggests that these frogs do not endure cold winters. R. ridibunda is widespread in Southern France and its distribution suggests multiple introductions. It is concluded that water frogs conform to the mosaic zone model rather than to the tension zone model because: (i) taxa exhibited differences in habitat use, (ii) pure parental species were documented and (iii) hybrids are not unfit relative to parental species.  相似文献

The aim of this study was to demonstrate in the adrenocortical and renal tissues of two species of frog, Rana italica and Rana esculenta, the presence and distribution of five neuropeptides: atrial natriuretic peptide (ANP), Leu-enkephalin (Leu-ENK), neuropeptide Y (NPY), substance P (SP) and vasoactive intestinal peptide (VIP).In anurans, the adrenal medulla is the site for the synthesis, storage and secretion of not only catecholamines but also various peptides. These peptides should not be regarded only as neurotransmitters or modulators for the secretion of catecholamines, but also as hormonal substances that induce systemic effects.All the peptides studied (ANP, Leu-ENK, NPY, SP and VIP) are present in both organs. However, different patterns of expression were observed for some of the peptides in two frogs.Immunopositivity to ANP was found in small clusters of chromaffin cells in both frogs whereas a clear strong positivity was present only in Rana esculenta kidney. Large clusters of chromaffin cells were immunoreactive to Leu-ENK in Rana italica but there were approximately 25% fewer compared to the positive cells present in Rana esculenta. Epithelial cells of renal tubules showed strong immunopositivity to Leu-ENK in Rana esculenta but not in Rana italica. A large number of adrenal cells (70–80%) were immunoreactive to NPY in Rana italica, while in Rana esculenta this peptide was localized in small clusters of chromaffin cells. Both frogs showed many NPY-positive cells in kidney. Many chromaffin cells were found positive to SP and VIP. A strong positivity was also observed in kidney in both frogs. These observations suggest a possible role of these peptides in the control of the physiological functions of adrenal glands and kidney of the two species of frogs studied.  相似文献

Phylogenetic relationships within the Euro-Mediterranean stock of the Rana temporaria species group (R. temporaria, R. iberica, R. cameranoi, R. macrocnemis, R. holtzi, R. dalmatina, R. graeca, R. latastei) were studied by (1) morphometric analysis, (2) osteological study and (3) electrophoresis. The three data sets were analysed independently and phenograms and cladograms examined for congruent patterns. The tree resulting from electrophoretic analysis is the most clearly resolved (and being also compatible with the other two) is assumed to represent the most likely phylogeny of the Mediterranean brown frogs. Patterns indicated by electrophoretic data matched the molecular clock-hypothesis. This suggests that the first split of the main stock of Mediterranean brown frogs separated ancestors of Rana temporaria and R. iberica from the others during the Late Pleistocene. A second split occurred separating the ancestors of what are currently the other big-bodied and the small-bodied brown frogs. The relationships between and within the three main stocks are discussed in the context of biogeographical and geological data.  相似文献

测定了中国蛙科动物24种和蟾蜍科的中华大蟾蜍线粒体12S rRNA基因长约400bp片段的序列。采用邻接法,对其数据的系统发生分析表明24种可分成3个支系:第一个支系包括棘腹蛙、合江棘蛙、棘胸蛙、大头蛙、虎纹蛙、倭蛙、泽陆蛙共7种,其中3种棘蛙组成一个单系;第二个支系由3种湍蛙,即戴云湍蛙、华南湍蛙和武夷湍蛙组成,应属于湍蛙亚科;第三支系包含余下的14种蛙,其中5种林蛙和4种臭蛙分别组成一个单系;2种侧褶蛙先聚合后,再与弹琴水蛙聚合,2种粗上先聚合在一起,再与由林蛙组成的单系群聚合。第三支系首先与第二支系相聚,然后它们与第一支系形成姐妹群,即由蛙亚科的7个物种组成的第一支系,不与蛙亚科另14个物种组成的第三支系直接聚合,表明第一支系的物种可能不属于蛙亚科。此外,分子证据提示把陆蛙属、大头蛙属、虎纹蛙属、棘棘属、臭蛙属、粗皮蛙属和侧褶蛙属从原来的蛙属分出有其合理性。、  相似文献

Comparative landscape genetics has uncovered high levels of variability in which landscape factors affect connectivity among species and regions. However, the relative importance of species traits versus environmental variation for predicting landscape patterns of connectivity is unresolved. We provide evidence from a landscape genetics study of two sister taxa of frogs, the Oregon spotted frog (Rana pretiosa) and the Columbia spotted frog (Rana luteiventris) in Oregon and Idaho, USA. Rana pretiosa is relatively more dependent on moisture for dispersal than R. luteiventris, so if species traits influence connectivity, we predicted that connectivity among R. pretiosa populations would be more positively associated with moisture than R. luteiventris. However, if environmental differences are important drivers of gene flow, we predicted that connectivity would be more positively related to moisture in arid regions. We tested these predictions using eight microsatellite loci and gravity models in two R. pretiosa regions and four R. luteiventris regions (= 1,168 frogs). In R. pretiosa, but not R. luteiventris, connectivity was positively related to mean annual precipitation, supporting our first prediction. In contrast, connectivity was not more positively related to moisture in more arid regions. Various temperature metrics were important predictors for both species and in all regions, but the directionality of their effects varied. Therefore, the pattern of variation in drivers of connectivity was consistent with predictions based on species traits rather than on environmental variation.  相似文献
