We isolated a novel virus strain (YN12246) from Culicoides spp. specimens collected at the China-Laos-Myanmar border in southern Yunnan Province. This virus had a cytopathic effect (CPE) on both insect cells (C6/36) and mammalian cells (BHK-21). Electron microscopy revealed the structure of the virions to be spherical with a diameter of 75 nm. Polyacrylamide gel analysis demonstrated that the viral genome consisted of 10 segments of double-stranded RNA (dsRNA), with a distribution pattern of 3-3-3-1. The coding sequences of 9 genome segments of YN12246 (Seg1, Seg3-Seg10) were obtained by high-throughput sequencing and Sanger sequencing. Comparisons of conserved genome segments 1 and 3 (Seg1 and Seg3), encoding the polymerase-VP1 and sub-core T2 protein, respectively, showed that YN12246 groups with the Culicoides-borne orbiviruses. The highest levels of sequence identity were detected between YN12246 and Tibet orbivirus (TIBOV), indicating that they belong to the same virus species (with amino acid identity of 98.8% and 96.4% for the polymerase and T2 protein, respectively). The data presented here confirm that YN12246 is a member of the TIBOV species, which was first isolated from mosquitoes in 2009. This is the first report of the isolation of TIBOV from Culicoides. 相似文献

A novel compound showing antiproliferative effect was isolated from Streptomyces sp. Its structure was determined based on the interpretation of the NMR spectra, and its conformation was elucidated using molecular modeling and 2D NOESY. A lambda-carrageenan-degrading Pseudoalteromonas bacterium, strain CL19, was isolated from a deep-sea sediment sample. A lambda-carrageenase from the isolate was purified to homogeneity from cultures containing lambda-carrageenan as a carbon source. This is the first report of the isolation of lambda-carrageenase together with the gene sequence for the enzyme. The molecular mass of the purified enzyme was approximately 100 kDa on both SDS-PAGE and gel-filtration chromatography, suggesting that the enzyme is a monomer. The optimal pH and temperature for activity were about 7 and 35 degrees C, respectively. The enzyme had specific activity of 253 U/mg protein. The enzyme required monovalent salts for the activity. Carbohydrates, such as sorbitol, sucrose, trehalose, improved the enzyme stability. The pattern of lambda-carrageenan hydrolysis showed that the enzyme is an endo-type lambda-carrageenase, and the final main product was a tetrasaccharide of the lambda-carrageenan ideal structure with galactose 2,6-disulfate at the reducing end, indicating the enzyme cleaves the beta-1,4 linkages of its backbone structure. Furthermore, the gene (cglA) encoding the enzyme was sequenced. It encoded a mature protein of 103 kDa (917 amino acids). 西双版纳地区稻田CO2排放通量 总被引:4,自引:0,他引:4
采用静态暗箱-气相色谱法对云南西双版纳地区单季稻田CO2排放及氮肥、水热因子对CO2排放的影响进行田间原位观测研究.试验设3个氮肥水平处理:N0(0 kg N hm-2)、N150(150 kg N hm-2)和N300(300 kg N hm-2).结果表明,受一天温度变化的影响,西双版纳地区稻田生态系统呼吸日变化为单峰型,其最大值出现在11:00~13:00之间,最小值出现在凌晨.稻田土壤呼吸呈明显的季节变化趋势,土壤呼吸平均速率为水稻收获后休闲季节>种植前休闲季节>水稻生长季节,差异达到1%显著水平.不同季节影响土壤呼吸的环境因子不同.土壤水分含量低于34%时,土壤呼吸速率与土壤含水量呈正相关,达5%显著水平;地面淹水时,土壤呼吸速率与淹水深度呈1%极显著负相关;水分含量高于38%时,土壤呼吸速率与温度呈极显著指数相关.长期考虑(整个生长季节),氮肥的施用对稻田土壤呼吸和生态系统呼吸无影响;N300处理抑制植株呼吸作用,单位生物量呼吸速率下降.氮肥的施用对土壤呼吸有短期影响,氮肥用量增加,土壤呼吸速率增加.计算得出N0、N150和N300处理年土壤呼吸量分别为6.27、6.31 t C hm-2 a-1和5.89 t C hm-2 a-1;年净固定大气中CO2-C分别为1.41、2.22 t C hm-2 a-1和1 11 t C hm-2 a-1,表明西双版纳稻田生态系统是碳汇. 相似文献

从西双版纳纳板河流域国家级自然保护区的盆距兰Gastrochilus、隔距兰Cleisostoma、毛兰Eria、贝母兰Coelogyne和万代兰Vanda的根、茎、叶分离得到172株非菌根内生真菌,根据ITS1-5.8S-ITS2 rDNA系统发育分析鉴定为盘菌亚门的4纲11目29属,其中炭角菌目、肉座菌目和格孢腔菌目真菌的相对频率最高;Xylaria、Fusarium和Phoma为主要优势属,相对频率分别为23.3%、14.5%和10.5%,它们的组织专化性不同,其中Xylaria在根、茎和叶均有分布,而Fusarium和Phoma都分离自根部。不同组织的内生真菌多样性和种类组成差异明显,根、茎和叶的内生真菌Shannon多样性指数分别为2.4083、1.0312和0.6557,表明根部内生真菌多样性显著高于茎、叶内生真菌多样性,其中根的主要优势内生真菌类群有Fusarium、Xylaria和Phoma,茎的主要优势内生真菌类群有Xylaria、Pestalotiopsis和Colletotrichum,叶的主要优势内生真菌类群有Colletotrichum、Pallidocercospora、Pantospora和Phyllosticta。从附生兰种类来看,盆距兰、隔距兰、毛兰、贝母兰和万代兰内生真菌的Shannon多样性指数分别为2.2689、2.2635、2.0115、1.9197、1.7139,表明盆距兰和隔距兰内生真菌多样性高于毛兰、贝母兰和万代兰。 相似文献

Lowland tropical forests once covered a large fraction of tropical southern China, but currently have an extent of ca 633,800 ha, mostly in Xishuangbanna of southern Yunnan. The Xishuangbanna region has a typical monsoon climate with a mean annual temperature ranging between 15.1°C and 21.7°C, and precipitation between 1200 and 2500 mm. There is a pronounced dry season between November and April with frequent occurrence of heavy fog. Rainfall during the wet season between May and October accounts for over 80 percent of total annual precipitation. Water deposition from fog accounts for over one‐third of total water input during the dry season in the forests, suggesting an important role that fog may play in pushing up the northern limit of tropical rain forest in Southeast Asia. 相似文献

[目的]为了研究2006年从广西病猪肺组织中分离的H1N2亚型猪流感病毒(SIV)A/Swine/Guangxi/13/2006(H1N2)(Sw/Gx/13/06)的遗传学特性和8个基因的来源.[方法]运用RT PCR方法对其全基因进行了克隆并运用分子生物学软件对其基因序列进行了遗传进化分析.[结果]血凝素(HA)、核蛋白(NP)、基质蛋白(M)和非结构蛋白(NS)基因来源于猪古典H1N1亚型流感病毒;神经氨酸酶(NA)和聚合酶蛋白(PB1)基因来源于人的H3N2亚型流感病毒;聚合酶蛋白(PA)和聚合酶蛋白(PB2)基因来自于禽流感病毒.[结论]可见Sw/GX/13/06是一株"人-猪-禽"三源基因重排H1N2亚型SIV且与美国(1999-2001年)和韩国(2002年)分离到该型病毒的有明显的亲缘关系.据我们所知,这是中国首次报道含有禽流感病毒基因片段的重排H1N2 SIV,该病毒是否对养猪业和人类公共卫生健康具有潜在的威胁,有待于进一步研究. 相似文献

A novel alkaloid, aristopyridinone A (1) and a new phenanthrene, aristolamide II (2), were isolated from Aristolochia manshuriensis (Guanmutong) together with eight known phenanthrenes (3-10). All structures were elucidated by spectroscopic methods. Compound 2 showed a selective inhibitory effect on elastase release by human neutrophils in response to fMLP with an IC50 value of 4.11 μg/mL. Compound 7 exhibited significant inhibitory effects on superoxide anion generation and elastase release with IC50 values of 0.12 and 0.20 μg/mL, respectively. 相似文献

The replication and polypeptide synthesis of orbivirus isolate Mill Door/79, a member of the Kemerovo serogroup, were studied. In Vero cells cell-associated virus exceeded cell-free virus by about 2 log10 PFU/ml. Attempts to purify the virus resulted in the demonstration of five polypeptides. Thirteen virus-induced polypeptides and 10 segments of double-stranded RNA were identified in infected cells. Partial proteolysis demonstrated homology between some polypeptides. 相似文献

In renal cell carcinoma (RCC), the level of higher gangliosides is correlated with degree of metastatic potential, and cell lines derived from metastatic deposits of RCC are characterized by high expression of disialogangliosides (Saito, S., Orikasa, S., Ohyama, C., Satoh, M., and Fukushi, Y. (1991) Int. J. Cancer 49, 329-334 and Saito, S., Orikasa, S., Satoh, M., Ohyama, C., Ito, A., and Takahashi, T. (1997) Jpn. J. Cancer Res. (Gann) 88, 652-659). We now report two disialogangliosides, G1 and G2, found in the RCC cell line TOS-1. G1 from TOS-1 cells was characterized as having a novel hybrid structure between ganglio-series (region I as in Structure; same as the terminal structure of ganglioside GM2), and the lacto-series type 1 (region II). The characterization was based on reactivity with various monoclonal antibodies (mAbs) with defined epitope specificity, as well as monosaccharide and fatty acid component analysis, (1)H NMR spectroscopy, and electrospray ionization mass spectrometry of the intact compound. G1 showed strong reactivity with mAb RM2, raised originally against TOS-1 cells, and weak cross-reactivity with anti-GM2 mAb MK-1-8. The antigen is hereby termed GalNAc disialosyl Lc4Cer (IV4GalNAcIV3NeuAcIII6NeuAcLc4; abbreviated GalNAcDSLc4). G2 was identified by 1H NMR and mass spectrometry as having a structure similar to Structure but without the GalNAcbeta1-->4 substitution and showed strong reactivity with mAb FH9 reported previously to be specific for disialosyl lacto-series type 1 (disialosyl Lc(4)) having vicinal alpha2-->3 and alpha2-->6 sialosyl residues, an antigen associated with human colonic cancer. Clinicopathological studies indicate that expression of these disialoganglioside antigens in RCC tissue is correlated with the metastatic potential of RCC. 相似文献

Proteolytic activity was identified in cultures of the marine shipworm bacterium by monitoring of the release of acid-soluble azopeptides from azocasein. Activity was predominantly extracellular (>80%), and its production was coincident with logarithmic cell growth. The protease(s) appeared to be constitutive, since it was present even when the bacterium was grown under nitrogen-fixing conditions. However, activity was stimulated up to 8.6-fold by the addition of complex nitrogen (casein, amino acids) to the growth medium. Maximum activity was observed at 40°C and between pH 6.5 and 9.0. 为调查甘肃省部分地区蚊虫及蚊媒病毒的种类及分布,本课题组2014年在甘肃省酒泉市和陇南市地区共采集蚊虫4 412只.酒泉市共采集蚊虫2属3种2 833只,其中背点伊蚊(Aedes dorsalis)2 665只(94.1%)、刺扰伊蚊(Aedes vexans)150只(5.3%)、淡色库蚊(Culex pipiens pallens)18只(0.6%);陇南市共采集2属2种1 579只蚊虫,其中淡色库蚊1 451只(91.9%)、中华按蚊(Anopheles sinensis)128只(8.1%).经实验室常规处理后,将蚊虫标本分为96批,接种于C6/36、BHK和Vero细胞,进行病毒分离和多种病毒基因特异性扩增检测.结果显示,96批蚊虫标本在3种细胞中均未出现细胞病变效应(cytopathic effect,CPE),在18批蚊虫标本研磨液中检测到辽宁病毒(Liaoning virus,LNV)基因阳性.病毒核苷酸序列的遗传进化分析显示,在甘肃省检测到的辽宁病毒与此前在我国新疆维吾尔自治区发现的辽宁病毒亲缘关系最近. 相似文献

利用景观空间格局定量化分析软件FRAGSTATS和西双版纳勐仑地区1988、2003年两期Landsat影像解译结果,定量分析和比较了两时期景观格局的组成、各类型斑块特征、不同类型斑块间空间分布关系及其动态变化,并进一步分析了其生态效应.结果表明,15年来研究区内以经济林为主的人工景观组分剧烈增加,景观多样性上升;橡胶园等人工种植园地的扩张使得有林地破碎化:平均斑块面积由44km2减至21km2,斑块数由368增至441,同时边缘密度增加;导致其核心区缩小,斑块连接度下降;城镇建设用地的扩展也使得耕地(主要是水田)趋于破碎化.景观中不同斑块的交错分布格局也趋于简单.人工经济林的大面积发展和城市化使该区景观中人为影响显著增强,并产生了生物生境恶化及生态环境质量下降等不良生态效应.未来土地利用规划中应借鉴景观生态学原则,以尽量减少对生态环境的进一步破坏. 相似文献

A new species of Trichoderma, Trichoderma guizhouense, isolated from soil in Guizhou Province, is described based on morphology and phylogenetic analyses. This species exhibits characteristic Trichoderma morphology but is distinct from related species based on characters of phialides and conidia. Two DNA markers, the translation elongation factor 1 alpha (tef1) and RNA polymerase II subunit b (rpb2) were used for phylogenetic analyses. The correlation between morphological and molecular-based clustering demonstrated two studied isolates are a new species. 相似文献
