In this study, we used sequences of two mitochondrial genes, cytochrome c oxidase I (COI) and 16S rRNA, and one nuclear gene, 28S rRNA, to test the monophyly of the sea star genus Echinaster, and understand the phylogenetic relationships among species and subgenera within this genus. Phylogenetic analyses based on Bayesian inference and maximum likelihood methods revealed three clades with high values of genetic divergence among them (K2P distances for COI over 23%). One of the clades grouped all Echinaster (Othilia) species, and the other two clades included Echinaster (non‐Othilia) species and Henricia species, respectively. Although the relationships among Henricia, Othilia, and Echinaster could not be completely clarified, the Othilia clade was a well‐supported group with shared diagnostic morphological characters. Moreover, the approximately unbiased test applied to the phylogenetic reconstruction rejected the hypothesis of the genus Echinaster as a monophyletic group. According to these results, we suggest the revalidation of Othilia as a genus instead of a subgenus within Echinaster. Our study clarifies important points about the phylogenetic relationships among species of Echinaster. Other important systematic questions about the taxonomic classification of Echinaster and Henricia still remain open, but this molecular study provides bases for future research on the topic.  相似文献

Based on morphological data and analysis of mitochondrial cytochrome b gene and nuclear (S7 intron 1) DNA sequences, the phylogenetic relationships of all Pollimyrus species known from the Omo‐Turkana enclosed basin and Nile system below the Murchison Falls were solved. A mormyrid “Pollimyruspetherici is distantly related to all other studied Pollimyrus species and clusters together with Cyphomyrus species forming with the later a monophyletic group. Moreover, the West African (but not the Congo River) populations of Cyphomyrus psittacus, the type species of the genus, seem to be conspecific to C. petherici. That is, the range of the genus Cyphomyrus is extended toward the Nile and Omo‐Turkana basins. This genus belongs to the large clade widely distributed in sub‐Saharian Africa and characterized by the presence of a chin appendage. Significance of this character for mormyrid phylogeny is discussed. Two distinct lineages of Pollimyrus occurring sympatrically in the White Nile tributaries and previously reported as the light and dark forms of Pollimyrus isidori together with five other congeneric species studied form a monophyletic group. The light form apparently represents P. isidori distributed in the Nile system downstream of the Murchison Falls and West Africa; the dark‐colored form (designated as Pollimyrus “D”) represents a distinct phylogenetic lineage inhabiting both the Omo‐Turkana and the White Nile basin. Morphological and ecological data suggest that this form may be conspecific to East African Pollimyrus nigricans or most probably represents a new species.  相似文献

The large and diverse genus Salix L. is of particular interest for decades of biological research. However, despite the morphological plasticity, the reconstruction of phylogenetic relationships was so far hampered by the lack of informative molecular markers. Infrageneric classification based on morphology separates dwarf shrubs (subg. Chamaetia) and taller shrubs (subg. Vetrix), while previous phylogenetic studies placed species of these two subgenera just in one largely unresolved clade. Here we want to test the utility of genomic RAD sequencing markers for resolving relationships at different levels of divergence in Salix. Based on a sampling of 15 European species representing 13 sections of the two subgenera, we used five different RAD sequencing datasets generated by Ipyrad to conduct phylogenetic analyses. Additionally we reconstructed the evolution of growth form and analyzed the genetic composition of the whole clade. The results showed fully resolved trees in both ML and BI analysis with high statistical support. The two subgenera Chamaetia and Vetrix were recognized as nonmonophyletic, which suggests that they should be merged. Within the Vetrix/Chamaetia clade, a division into three major subclades could be observed. All species were confirmed to be monophyletic. Based on our data, arctic‐alpine dwarf shrubs evolved four times independently. The structure analysis showed five mainly uniform genetic clusters which are congruent in sister relationships observed in the phylogenies. Our study confirmed RAD sequencing as a useful genomic tool for the reconstruction of relationships on different taxonomic levels in the genus Salix.  相似文献

The Northern Goshawk Accipiter gentilis is a medium‐sized bird of prey inhabiting boreal and temperate forests. It has a Holarctic distribution with 10 recognized subspecies. Traditionally, it has been placed within the Accipiter [gentilis] superspecies, together with Henst's Goshawk A. henstii, the Black Sparrowhawk A. melanoleucus, and Meyer's Goshawk A. meyerianus. While those four taxa are geographically separated from each other, hence referred to as allospecies, their phylogenetic relationships are still unresolved. In the present study, we performed phylogenetic analyses on the Accipiter [gentilis] superspecies, including all recognized subspecies of all four allospecies, using partial sequences of two marker loci of the mitochondrial genome, the control region and the cytochrome b gene. We found a deep split within A. gentilis into two monophyletic groups, a Nearctic clade (three subspecies) and a Palearctic clade (seven subspecies). The Palearctic clade is closely related to A. meyerianus, and together these two were more closely related to the other Old World taxa A. henstii and A. melanoleucus, which in turn were reciprocally monophyletic sister species. As a consequence, A. gentilis as usually conceived (including all Holarctic subspecies) was non‐monophyletic. We found a strong genetic homogeneity within Palearctic A. gentilis despite the fact that it comprises seven subspecies distributed from the Atlantic coast in Western Europe to Eastern Siberia. Relationships between the four clades could not be resolved unambiguously. Our results, if confirmed by more integrative data, would imply a taxonomic revision of Nearctic A. gentilis into a separate allospecies, Accipiter [gentilis] atricapillus.  相似文献

We conducted phylogenetic analyses based on complete mitochondrial cytochrome b gene sequences among southern and central Mexican cyprinid species, included in the genera Notropis and Hybopsis. In addition 15 northern species of the genera Notropis and Hybopsis were included in the analyses in order to place the Mexican species into a larger phylogenetic framework. The phylogenetic relationships supported the existence of five major clades: (1) including species of the subgenus Alburnops of the genus Notropis plus N. shumardi; (2) species of the subgenus Notropis; (3) species of the genus Hybopsis; (4) species of the N. texanus + N. volucellus species group of the genus Notropis; (5) Mexican endemic species of the genus Notropis plus the genus Yuriria. Previous phylogenetic inferences based on morphological characters resolved the Mexican minnows analysed as N. sallaei, N. calientis, N. boucardi and Y. alta, non‐monophyletic. According to our cytochrome b evidence all Mexican minnows of the genera Notropis and Yuriria formed a monophyletic group with respect to the northern species of the genera Notropis and Hybopsis. Within the Mexican clade, three well‐supported clades were identified: the first included the closely related species N. moralesi and N. boucardi, which occur in three independent drainages of south Mexico; the second consisted of two different lineages, N. imeldae and an undescribed species of Notropis, inhabiting two independent drainages of south Mexico; the third comprised two central Mexican Notropis species (N. calientis and N. sallaei) and the Y. alta populations. Based on this study and pending a more extensive taxonomic revision of the genus Notropis, we adopt the conservative criterion of considering all Notropis species from southern and central Mexico examined, including Y. alta, as belonging to the genus Notropis. © 2003 The Linnean Society of London, Biological Journal of the Linnean Society, 2003, 80 , 323–337.  相似文献

The taxonomic validity of the genus Hydropuntia Montagne (1843) (including Polycavernosa) within the Gracilariaceae (Gracilariales, Rhodophyta) is controversial. Morphological characters that define species of Hydropuntia are said to be variable and to overlap with those of Gracilaria. Here we present a global phylogenetic study of the family based on a Bayesian analysis of a large rbcL DNA sequence dataset indicating that the genus Hydropuntia forms a well supported monophyletic clade within the family, and recognize Hydropuntia as a genus distinct from Gracilaria. We also conducted smaller phylogenetic analyses in which thirty four Hydropuntia rbcL sequences resulted in two major clades within the genus, comprising a Caribbean clade and an Indo‐Pacific clade. Diagnostic reproductive stages that separate these two clades will be illustrated.  相似文献

基于60个形态学性状,对中国广义报春花科(Primulaceae s.l.)紫金牛属(Ardisia)90个分类群的系统发育关系运用分支分析方法进行了分析。采用最简约性分析得到了100个同等简约树。50%多数规则一致树的分支结构与以前建立的紫金牛亚属划分系统基本一致。外类群酸藤子属、铁仔属、密花树属聚在分支树的最基部,紫金牛属为一单系类群。形态分支树的一致性指数和保持性指数和各分支内部支持率均较低,只在种与变种或亚种之间获得较高的支持率。高木亚属、腋序亚属、短序亚属、顶序亚属处于分支树较为基部的位置,推测这四个亚属的类群在紫金牛属中较为原始;圆齿亚属和锯齿亚属共同组成一大支,二者亲缘关系紧密,推测这两个亚属为该属中最为进化的类群。结合形态学对属内系统发育关系进行了讨论和推测了一些性状的演化趋势,以期为分类修订提供依据。  相似文献

Based on multilocus phylogenetic analyses (18S, 28S, EF1‐α, SRP54, HSP70, CO1, 10 860 nt aligned), we show that the house dust mite subfamily Guatemalichinae is nested within non‐onychalgine pyroglyphid mites and forms the sister group to the genus Sturnophagoides (bootstrap support 100, posterior probability 1.0). Because high bootstrap support values may be misleading in the presence of incongruence, we evaluate robustness of the Guatemalichinae+Sturnophagoides clade using: (1) internode certainty indices to estimate the frequency of conflicting bipartitions in maximum‐likelihood bootstrap trees, (ii) consensus networks to investigate conflict among different loci; and (iii) statistical hypothesis testing based on information theory, both multi‐scale and regular bootstrap. Results suggest that this grouping is very well supported given the data. The molecular analyses were integrated with detailed morphological study using scanning electron and light microscopy. We suggest that the subfamilial status of Guatemalichinae should be reconsidered, and this lineage should be placed within the subfamily Dermatophagoidinae. The latter subfamily is currently accepted in the literature as a monophyletic group but was here inferred as paraphyletic and was not supported by any morphological synapomorphy. The paraphyly involved the most species‐rich and medically important genus, Dermatophagoides. Our findings suggest the need for a comprehensive revision of the higher‐level relationships of pyroglyphid house dust mites using both DNA sequences and morphology coupled with a broad taxonomic sampling.  相似文献

Ranunculaceae are a nearly cosmopolitan plant family with the highest diversity in northern temperate regions and with relatively few representatives in the tropics. As a result of their position among the early diverging eudicots and their horticultural value, the family is of great phylogenetic and taxonomic interest. Despite this, many genera remain poorly sampled in phylogenetic studies and taxonomic problems persist. In this study, we aim to clarify the infrageneric relationships of Clematis by greatly improving taxon sampling and including most of the relevant subgeneric and sectional types in a simultaneous dynamic optimization of phenotypic and molecular data. We also investigate how well the available data support the hypothesis of phylogenetic relationships in the family. At the family level, all five currently accepted subfamilies are resolved as monophyletic. Our analyses strongly imply that Anemone s.l. is a grade with respect to the Anemoclema Clematis clade. This questions the recent sinking of well‐established genera, including Hepatica, Knowltonia and Pulsatilla, into Anemone. In Clematis, 12 clades conceptually matching the proposed sectional division of the genus were found. The taxonomic composition of these clades often disagrees with previous classifications. Phylogenetic relationships between the section‐level clades remain highly unstable and poorly supported and, although some patterns are emerging, none of the proposed subgenera is in evidence. The traditionally recognized and horticulturally significant section Viorna is both nomenclaturally invalid and phylogenetically unsupported. Several other commonly used sections are likewise unjustified. Our results provide a phylogenetic background for a natural section‐level classification of Clematis.  相似文献

The broadly defined genus Chaetophora consisted of species with minute, uniseriate branching filaments enveloped in soft or firm mucilage forming macroscopic growths that are spherical, hemispherical, and tubercular or arbuscular, growing epiphytically on freshwater aquatic plants and other submerged surfaces in standing or fast‐flowing water. Recent molecular analyses clearly showed that this genus was polyphyletic. In this study, eight strains of Chaetophora and three strains of Stigeoclonium were identified and successfully cultured. In combination with the morphological data, a concatenated data set of four markers (18S + 5.8S + ITS2+ partial 28S rDNA) was also used to determine their taxonomic relationships and phylogenetic positions. The molecular analysis resolved the broadly defined Chaetophora to at least two genera. Species with a globose thallus of genus Chaetophora formed a separate monophyletic clade, which clearly separated from, a type of lobe‐form Chaetophora species. Therefore, we propose to erect a new genus, Chaetophoropsis, which includes all globose species of the Chaetophora. Chaetophoropsis aershanensis was determined to be a new species, based on its special characteristic of profuse long rhizoids. Stigeoclonium polyrhizum, as the closest relative to Chaetophoropsis, revealed its distant relationships to other species of Stigeoclonium. A globose thallus with a thick, soft mucilage matrix, and special rhizoidal branches lent further support to the placement of S. polyrhizum in the genus Chaetophoropsis and had the closest relationship to C. aershanensis. Taxonomic diversity was proven by distinctive morphological differences and by phylogenetic divergence in the broadly defined Chaetophora identified herein.  相似文献

We provide a taxonomic review of the extinct testudinid Testudo catalaunica, based on published and unpublished material from several Miocene (late Aragonian and early Vallesian) sites of the Vallès‐Penedès Basin (north‐east Iberian Peninsula). We show that Testudo catalaunica irregularis is a junior subjective synonym of T. catalaunica, and further provide an emended diagnosis of the latter based on newly reported material. Contrary to some recent suggestions, this emended diagnosis discounts an alternative attribution of T. catalaunica to Paleotestudo. The latter is merely recognized as a subgenus of Testudo, based on a cladistic analysis that assessed the phylogenetic position of all extant and most extinct species of Testudo currently recognized as valid (including T. catalaunica). Our phylogenetic analysis (which recovers the molecular phylogeny of extant Testudo s.l.) supports a taxonomic scheme in which the three extant subgenera of Testudo are represented in the fossil record. Testudo s.s. is retrieved as the sister taxon of Testudo (Agrionemys) + [Testudo (Paleotestudo) + Testudo (Chersine)]. The extinct Testudo (Paleotestudo) is therefore the sister taxon of the Testudo (Chersine) clade. The latter subgenus reveals as the most diverse clade of Testudo s.l. in the fossil record, with T. catalaunica Testudo steinheimensis constituting a subclade distinct from that including Testudo hermanni.  相似文献

Proctonotidae and Madrellidae are families that belong to the suborder Cladobranchia. Historically, both have been the subjects of taxonomic confusion. Thus, Proctonotidae Gray, 1853, was subsequently named as Zephyrinidae Iredale and O'Donoghue, 1923 and Janolidae Pruvot‐Fol, 1933, but currently both are considered as synonyms of Proctonotidae. On the other hand, Alder and Hancock (1864) erected the genus Madrella in Proctonotidae. Here, we completed a detailed morphological and molecular study of four apparently undescribed species of Madrellidae and Proctonotidae from the Indo‐Pacific. We performed a maximum likelihood and Bayesian inference phylogenetic analyses using two mitochondrial and one nuclear genes to improve the understanding of the families. Prompted by our results, Janolidae is removed from synonymy with Proctonotidae. Within Janolidae, there are two well‐supported clades. One includes species with smooth cerata that are found in the Atlantic and eastern Pacific Oceans. The taxa in this clade include the type species of Antiopella and several other species. We resurrect Antiopella as the valid name for this clade. The sister clade to Antiopella includes a variety of taxa with species that have been traditionally included in Janolus Bergh, 1884 and Bonisa Gosliner, 1981. Further systematic revision requires more comprehensive taxon sampling. The new species discovered have clear morphological differences and strong molecular support. They include Madrella amphora Pola and Gosliner sp. nov. , Janolus tricellariodes Pola and Gosliner sp. nov. , Janolus flavoanulatus Pola and Gosliner sp. nov., and Janolus incrustans Pola and Gosliner sp. nov.  相似文献

(2010) Phylogenetic relationships of the Chinese Labeoninae (Teleostei, Cypriniformes) derived from two nuclear and three mitochondrial genes. —Zoologica Scripta, 39, 559–571. The majority of genera within the Labeoninae occur in the South of China and the phylogeny of the subfamily Labeoninae has been a controversial topic over the years. The early and more recent phylogenetic results based on morphology are not in agreement, and some of the molecular analyses contradict those based on morphology. However, none of the previous studies has included an extensive sampling of Labeoninae genera. In this study, partial sequences of two nuclear (exon 3 of recombination activating protein 1 and rhodopsin) and three mitochondrial genes (cytochrome b, cytochrome oxidase subunit I and 16S ribosomal RNA) from 39 ingroup taxa and 11 outgroup taxa were used analysed to provide a hypothesis of phylogenetic relationships within the Chinese Labeoninae. The results supported the monophyly of Labeoninae and refuted the subdivision within Labeoninae based on the oromandibular morphology. It also further confirmed that the presence of a disc on lower lip arose through convergent evolution. Labeo was the basal clade and Osteochilus was closely related to Cirrhinus, forming the second basal clade. The monophyly of Garra and Bangana was refuted in this study, and they were both subdivided into two lineages. One lineage of Garra had closer relationships with Crossocheilus and Akrokolioplax, and the other lineage had closer relationships with Placocheilus. One lineage of Bangana represented the species with broadly interrupted postlabial groove; these species represented the true Bangana. The other lineage of Bangana included the species with continuous postlabial groove, and these species should be assigned to a new genus. Discogobio, as currently conceived, was paraphyletic, but it rendered monophyletic with the inclusion of Discocheilus. These results indicated that Discocheilus should be synonymised with Discogobio. Hongshuia and Sinocrossocheilus, respectively, formed independent lineages related to a lineage consisting of Pseudocrossocheilus plus seven other nominal genera. The relationships received weak support, but the repeatability of the clades could form the basis of recognising only the three genera mentioned above.  相似文献

The taxonomic history of the diatom genus Amphora is one of a broad early morphological concept resulting in the inclusion of a diversity of taxa, followed by an extended period of revision and refinement. The introduction of molecular systematics has increased the pace of revision and has largely resolved the relationships between the major lineages, indicating homoplasy in the evolution of amphoroid symmetry. Within the two largest monophyletic lineages, the genus Halamphora and the now taxonomically refined genus Amphora, the intrageneric morphological and ecological relationships have yet to be explored within a phylogenetic framework. Critical among this is whether the range of morphological features exhibited within these diverse genera are reflective of evolutionary groupings or, as with many previously studied amphoroid features, are nonhomologous when examined phylogenetically. Presented here is a four‐marker molecular phylogeny that includes 31 taxa from the genus Amphora and 77 taxa from the genus Halamphora collected from fresh, brackish, and salt waters from coastal and inland habitats of the United States and Japan. These phylogenies illustrate complex patterns in the evolution of frustule morphology and ecology within the genera and the implications of this on the taxonomy, classification, and organization of the genera are discussed.  相似文献

Hartmannellid amoebae are an unnatural assemblage of amoeboid organisms that are morphologically difficult to discern from one another. In molecular phylogenetic trees of the nuclear‐encoded small subunit rDNA, they occupy at least five lineages within Tubulinea, a well‐supported clade in Amoebozoa. The polyphyletic nature of the hartmannellids has led to many taxonomic problems, in particular paraphyletic genera. Recent taxonomic revisions have alleviated some of the problems. However, the genus Saccamoeba is paraphyletic and is still in need of revision as it currently occupies two distinct lineages. Here, we report a new clade on the tree of Tubulinea, which we infer represents a novel genus that we name Ptolemeba n. gen. This genus subsumes a clade of hartmannellid amoebae that were previously considered in the genus Saccamoeba, but whose mitochondrial morphology is distinct from Saccamoeba. In accordance with previous research, we formalize the clade as distinct from Saccamoeba. Transmission electron microscopy of our isolates illustrate that both molecularly discrete species can be further differentiated by their unique mitochondrial cristal morphology.  相似文献

The present molecular systematic and phylogeographic analysis is based on sequences of cytochrome c oxidase subunit 1 (cox1) (mtDNA) and 28S ribosomal DNA and includes 59 isolates of cestodes of the genus Anoplocephaloides Baer, 1923 s. s. (Cyclophyllidea, Anoplocephalidae) from arvicoline rodents (lemmings and voles) in the Holarctic region. The emphasis is on Anoplocephaloides lemmi (Rausch 1952) parasitizing Lemmus trimucronatus and Lemmus sibiricus in the northern parts of North America and Arctic coast of Siberia, and Anoplocephaloides kontrimavichusi (Rausch 1976) parasitizing Synaptomys borealis in Alaska and British Columbia. The cox1 data, 28S data and their concatenated data all suggest that A. lemmi and A. kontrimavichusi are both non‐monophyletic, each consisting of two separate, well‐defined clades, that is independent species. As an example, the sister group of the clade 1 of A. lemmi, evidently representing the ‘type clade’ of this species, is the clade 1 of A. kontrimavichusi. For A. kontrimavichusi, it is not known which one is the type clade. There is also fairly strong evidence for the non‐monophyly of Anoplocephaloides dentata (Galli‐Valerio, 1905)‐like species, although an earlier phylogeny suggested that this multispecies assemblage may be monophyletic. The results suggest a deep phylogenetic codivergence of Lemmus spp. and A. lemmi, primarily separating the two largely allopatric host and parasite species at the Kolyma River in east Siberia. There are also two allopatric sublineages within each main clade/species of A. lemmi and Lemmus, but the present distributions of the sublineages within the eastern L. trimucronatus and clade 1 of A. lemmi are not concordant. This discrepancy may be most parsimoniously explained by an extensive westward distributional shift of the easternmost parasite subclade. The results further suggest that the clade 1 of A. kontrimavichusi has diverged through a host shift from the precursor of L. trimucronatus to S. borealis.  相似文献

The lizards of the Egernia group of Australia and Melanesia include some of the most distinctive members of the family Scincidae in morphology (including giant size, spinose scalation), ecology and behaviour. Social behaviour, including long‐term recognition of individuals and kin, mate fidelity and home site fidelity, is amongst the most complex known in squamate reptiles and is the subject of an expanding number of studies. Lack of a sound phylogeny for the Egernia group has limited our ability to understand the evolution and patterns of variation in social behaviour within this group, and evidence for the monophyly of the largest genus, Egernia (64% of the species), has been lacking. We present data derived from nucleotide sequences that establish a phylogenetic framework for the Egernia group. We used two mitochondrial sequences, the protein‐encoding ND4 gene and a ribosomal gene, 12s rRNA, and two nuclear sequences, the protein‐encoding c‐mos, and non‐encoding intron 7 of β‐fibrinogen. Our phylogenetic analyses show that Corucia of the Solomon Islands is the sister group of the rest of the Egernia group. The genus Egernia is paraphyletic, including four well‐supported monophyletic units, one of which is the sister lineage of the Tiliqua lineage (Tiliqua plus Cyclodomorphus).
