Studies on ant communities in agroecosystems have contributed to the knowledge of the effect of agricultural activities on biological communities. The aim of this study is to explain the effect of soil use on ant communities. We tested the hypothesis that there was a decrease in ant species richness and a change in the species composition at habitats with more intense soil use. We collected ants using sardine baits, subterranean traps and direct sampling at four habitats with different soil use (secundary forest, Acacia forestry, initial stage of succession and mixed crops). The ant species richness did not decrease with intensity of soil use. In successional habitat the species numbers collected using sardine baits and subterranean traps were significantly different. Species composition of communities had a pronounced variation, with the epigaeic and hypogaeic ant faunas of the habitat with high intense soil use (mixed crops) had low similarity with ant communities of the three other habitats. The predator species were restricted to habitats with low intensity of soil use. Then, species composition could better reflect the functional changes on ant communities than species richness. Our data can help to choose the component of ant community that better reflect the response of biodiversity to agricultural impacts. 相似文献

Maternally inherited endosymbionts of arthropods are one of the most abundant and diverse group of bacteria. These bacterial endosymbionts also show extensive horizontal transfer to taxonomically unrelated hosts and widespread recombination in their genomes. Such horizontal transfers can be enhanced when different arthropod hosts come in contact like in an ecological community. Higher rates of horizontal transfer can also increase the probability of recombination between endosymbionts, as they now share the same host cytoplasm. However, reports of community‐wide endosymbiont data are rare as most studies choose few host taxa and specific ecological interactions among the hosts. To better understand endosymbiont spread within host populations, we investigated the incidence, diversity, extent of horizontal transfer, and recombination of three endosymbionts (Wolbachia, Cardinium, and Arsenophonus) in a specific soil arthropod community. Wolbachia strains were characterized with MLST genes whereas 16S rRNA gene was used for Cardinium and Arsenophonus. Among 3,509 individual host arthropods, belonging to 390 morphospecies, 12.05% were infected with Wolbachia, 2.82% with Cardinium and 2.05% with Arsenophonus. Phylogenetic incongruence between host and endosymbiont indicated extensive horizontal transfer of endosymbionts within this community. Three cases of recombination between Wolbachia supergroups and eight incidences of within‐supergroup recombination were also found. Statistical tests of similarity indicated supergroup A Wolbachia and Cardinium show a pattern consistent with extensive horizontal transfer within the community but not for supergroup B Wolbachia and Arsenophonus. We highlight the importance of extensive community‐wide studies for a better understanding of the spread of endosymbionts across global arthropod communities. 相似文献

During a survey of microfungi on the subantarctic island of South Georgia, large numbers of phylloplane yeasts were isolated in late spring from leaves of a tussock grass. The dominant yeast was identified asCandida saké, this being the first record for the Antarctic region. Isolates in liquid culture had a temperature optimum for growth of 20–25°C. It was capable of assimilation of a range of simple carbohydrates, similar to those found in leachates from new leaves of the tussock grass. The seasonal decline of yeasts on the phylloplane is discussed in terms of the availability of leachate and the growth of filamentous microfungi on new leaves. 相似文献

Question: The quantification of functional traits in natural communities can be difficult (e.g. root traits, RGR). Can functional traits measured on pot grown plants be reliably applied to natural communities? Alternatively, can below‐ground plant traits be predicted from above‐ground traits? Location: Southeastern Australia. Methods: We compared 17 shoot, root and whole‐plant morphological traits measured on 14 plant species in a native grassland community to those measured under two different pot conditions: unfertilised and fertilised. Results: The majority of trait values for pot grown plants differed to plants in the field, however, species ranking remained consistent for most leaf traits between the field and the two pot growing conditions. In contrast, species ranking was not consistent for most whole plant traits when comparing field plants to fertilised pot grown plants, providing a caution against the tendency to grow plants in controlled conditions at ‘optimal’ (high) resource levels. Moderate to strong correlations were found between below‐ground and above‐ground plant traits, including between root dry matter content and leaf dry matter content, and between specific root area and specific leaf area. Conclusions: The utility of pot grown plants to quantify traits for field plants is highly dependent on the selection of the growing conditions in the controlled environment. The consistency we observed between above‐ground and below‐ground trait strategies suggests that below‐ground traits may be predictable based on above‐ground traits, reducing the need to quantify root traits on cultured plants. 相似文献

The influence of mature trees on colonization of Douglas-fir (Pseudotsuga menziesii) seedlings by ectomycorrhizal fungi (EMF) is not well understood. Here, the EMF communities of seedlings planted near and far from trees are compared with each other, with EMF of seedlings potted in field soils and with EMF of mature trees. Seedlings were planted within 6 m, or beyond 16 m, from residual Douglas-fir trees in recently harvested green-tree retention units in Washington State, USA, or potted in soils gathered from near each residual tree. Mature tree roots were sampled by partly excavating the root system. The EMF communities were assessed by polymerase chain reaction-restriction fragment length polymorphism and sequence analysis of ribosomal RNA genes. Seedlings near trees had higher species richness and diversity of EMF communities compared with seedlings far from trees. The EMF communities of seedlings near trees were more similar to those of mature trees, while seedlings far from trees were more similar to glasshouse seedlings. By enhancing the EMF diversity of seedlings, residual trees may maintain or accelerate the re-establishment of mycorrhizal communities associated with mature forests. 相似文献

We have used differential scanning calorimetry to determine the effect of low concentrations (C = 0-2 M) of the osmolyte sarcosine on the Gibbs energy changes (deltaG) for the unfolding of hen-egg-white lysozyme, ribonuclease A, and ubiquitin, under the same buffer and pH conditions. We have also computed this effect on the basis of the additivity assumption and using published values of the transfer Gibbs energies for the amino acid side chains and the peptide backbone unit. The values thus predicted for the slope delta deltaG/deltaC agree with the experimental ones, but only if the unfolded state is assumed to be compact (that is, if the accessibility to solvent of the unfolded state is modeled using segments excised from native structures). The additivity-based calculations predict similar delta deltaG/deltaC values for the three proteins studied. We point out that, to the extent that this approximate constancy of delta deltaG/deltaC holds, osmolyte-induced increases in denaturation temperature will be larger for proteins with low unfolding enthalpy (small proteins that bury a large proportion of apolar surface). We have studied the effect of ethylene on the localization of the basic isoforms of glucan endo-1,3--glucosidase (-1,3-glucanase, EC 3.2.1.39) and endo-chitinase (chitinase, EC 3.2.1.14) in leaves of Nicotiana tabacum L. cv. Havana 425. Comparisons of the enzyme contents of the lower epidermis of the leaf, leaf expiants with the lower epidermis removed, and intercellular wash fluid indicate that both enzymes are localized inside epidermal cells of untreated leaves. Ethylene treatment (20 l·l-1, 4d) induced a marked -10- to 30-fold-coordinated accumulation of the enzymes. This was due primarily to induction of the basic isoforms inside chlorenchyma cells of the leaf interior. The localization of basic -1,3-glucanase was confirmed by immunofluorescence histochemistry and immunogold cytochemistry. Immunolabelling was confined to electron-dense bodies of the cell vacuole. No extracellular immunolabelling was detected in control or ethylene-treated leaves. We conclude that ethylene changes the cell-type-specific distribution but not the intracellular compartmentation of the two enzymes. These results support the generalization that basic isoforms of chitinase and -1,3-glucanase are intracellular whereas the acidic isoforms are secreted into the extracellular space.Abbreviations IgG

Oceanic islands are renowned for their unique flora and high levels of endemism. Native island plants, however, are imperilled by non-native species that can become invasive by outcompeting natives. The threat of native island assemblages generally increases with isolation and the number of endemics featured, but also with human-associated disturbance and land use. Based on this, the Canary Island native plant systems should be highly threatened by invasives, similar to other oceanic islands globally. However, Canarian native plant systems are only weakly infiltrated and are rarely directly threatened by invasive plants. Further, highly disturbed areas, usually among the first colonized by invasives on islands, are recolonized here by natives. Based on this, we postulate four hypotheses (climatic filter, well-preservation status, human legacy and permanent colonization) for explaining this unusual behaviour of plant systems on the Canary Islands, providing an opportunity to understand the drivers and processes behind invasion into plant communities on islands. 相似文献

The effect of ethylene and cytokinin on guanosine 5′-triphosphate binding and protein phosphorylation in leaves of Arabidopsis thaliana 总被引:2,自引:0,他引:2
Binding of [α-32P]guanosine 5′-triphosphate ([α-32P]GTP) has been demonstrated in a Triton X-100-solubilised membrane fraction from leaves of Arabidopsis thaliana (L.) Heynh. Binding was stimulated by 1 h pre-treatment of leaves with ethylene and this effect was antagonised by the inclusion
of N6-benzyladenine in the medium used for homogenisation. The ethylene-insensitive mutants eti5 and etr showed contrasting responses. In eti5 the constitutive level of GTP binding was higher than in the wild type whereas in etr the level was much lower. Neither ethylene nor cytokinin affected GTP binding in the mutants. The GTP-binding activity was
localised in two bands at 22 and 25 kDa, both of which were immunoprecipitated by anti-pan-Ras antibodies, indicating that
the activity is due to small GTP-binding proteins. In a similar membrane fraction, ethylene was shown to increase protein
phosphorylation and benzyladenine antagonised this effect. In eti5 the constitutive level of protein phosphorylation was higher than in the wild type, but benzyladenine increased activity
substantially while ethylene was without effect. In etr, protein phosphorylation was lower than in the wild type, ethylene was without effect, but cytokinin increased activity.
A protein of Mr 17 kDa was detected on gels using antibodies to nucleoside diphosphate kinase. Phosphorylation of this protein was upregulated
by ethylene but nucleoside diphosphate kinase activity was unaffected. The results are compared with the effect of the two
hormones on the senescence of detached leaves and discussed in relation to pathways proposed for ethylene signal transduction.

Impact of habitat diversification on arthropod communities: A study in the fields of Chinese cabbage, Brassica chinensis 总被引:1,自引:0,他引:1

Field trials were carried out from June to August in 2004 at Wuyishan (Wuyi Mountains), Fujian province, China, to determine the effects of habitat diversification on arthropod communities. Two Chinese cabbage, Brassica chinensis, field 1 (F1) and field 2 (F2) surrounded by diverse vegetable cultivars were selected, while a monoculture of Chinese cabbage served as the control field (CK). The results showed that: (i) when comparing insect abundance of each order between different habitats, significantly higher numbers of lepidopterous insects (39.76% from the each order) and lower densities of Hymenoptera (19.82%) were found in CK than in F1 and F2; (ii) compared with CK, F1 and F2 had a lower percentage of species richness and an abundance of herbivorous insects, but increased richness, abundance and biodiversity of predatory insects; (iii) no differences were observed in neutral insects' guild between different fields; and (iv) the dominant species for each guild depends on the habitat types and sampling dates. This study suggests that intercropping could conceivably be used in these habitats to increase the population of natural predators, thus achieving desirable and ecologically friendly results in vegetable fields. 相似文献

Are sucrosyl-oligosaccharides synthesized in mesophyll protoplasts of mature leaves of Cucumis melo?
Biosynthesis of sucrosyl-oligosaccharides (raffinose, stachyose) was traced in source leaves of Cucumis melo after 14C-photoassimilation. The main carbon compound exported was 14C-labeled stachyose. No oligosaccharide synthesis was detected in young, importing leaves. Mesophyll protoplasts, isolated from mature leaves which had previously photosynthesized 14CO2, did not contain 14C-oligosaccharides but contained [14C]-sucrose and 14C-hexoses. Isolated minor-vein-enriched fractions from the same leaves, however, showed nearly 30% of the 14C of the neutral fraction to be in oligosaccharides. Isolated, viable mesophyll protoplasts incubated with NaH14CO3 also failed to incorporate radioactivity into oligosaccharides, although sucrose and galactinol synthesis was unimpaired. Galactinolsynthase activity in leaf extracts and in mesophyll protoplasts was 16.8 mol·h-1·mg-1 protein and 13.8 mol·h-1·mg-1 protein, respectively. Galactosyltransferase (EC 2.4.1.67), which synthesizes stachyose from raffinose and galactinol, had an activity of 50 nmol·h-1·mg-1 protein in leaf extracts and was also present in the minor-vein-enriched fraction, but could not be detected in mesophyll protoplast lysates. The results indicate that mesophyll cells may not be the site of stachyose synthesis although precursor compounds like sucrose and galactinol are synthesized there.Abbreviation HPLC

Small β-hairpin peptides have been widely used as models for the folding of β-sheets. But how applicable is the folding of such models to β-structure in larger proteins with conventional hydrophobic cores? Here we present multiple unfolding simulations of three such proteins that contain the WW domain double hairpin β-sheet motif: cold shock protein A (CspA), cold shock protein B (CspB) and glucose permease IIA domain. We compare the behavior of the free motif in solution and in the context of proteins of different size and architecture. Both Csp proteins lost contacts between the double-hairpin motif and the protein core as the first step of unfolding and proceeded to unfold with loss of the third β-strand, similar to the isolated WW domain. The glucose permease IIA domain is a larger protein and the contacts between the motif and the core were not lost as quickly. Instead the unfolding pathway of glucose permease IIA followed a different pathway with β1 pulling away from the sheet first. Interestingly, when the double hairpin motif was excised from the glucose permease IIA domain and simulated in isolation in water it unfolded by the same pathway as the WW domain, indicating that it is tertiary interactions with the protein that alter the motif’s unfolding not a sequence dependent effect on its intrinsic unfolding behavior. With respect to the unfolding of the hairpins, there was no consistent order to the loss of hydrogen bonds between the β-strands in the hairpins in any of the systems. Our results show that while the folding behavior of the isolated WW domain is generally consistent with the double hairpin motif’s behavior in the cold shock proteins, it is not the case for the glucose permease IIA domain. So, one must be cautious in extrapolating findings from model systems to larger more complicated proteins where tertiary interactions can overwhelm intrinsic behavior. 相似文献

Benthic component of an ecosystem is considered in ecological status assessment of the key European Directives. Most of the metrics proposed for the benthic quality assessment are biodiversity based. Their robustness and applicability are widely discussed in many recent studies. However an impact of invasive alien species on biotic indices and environmental quality assessments has been largely overlooked by researchers so far. In the current study we assessed Benthic Quality Index (BQI) in a coastal ecosystem, highly affected by the invasive zebra mussel Dreissena polymorpha. Zebra mussel is able of modifying benthic habitats and enhancing local biodiversity. In the analyzed ecosystem it affected benthic species richness, abundance and community structure. As a result the calculated BQI values were significantly higher in the presence of zebra mussel with evident outliers in samples with particularly high zebra mussel abundances. Therefore we found that BQI determined in our study was artificially elevated providing false signal of the ecological status improvement. Based on the results presented, we suggested data correction framework that has been tested on the current dataset and proved to be effective minimizing zebra mussel impact on BQI assessment. Our experience could be applied for other coastal ecosystems invaded by the zebra mussel or any other aquatic invasive species with resembling biological traits and bioinvasion impacts. 相似文献

Tiny extra weights (brass nuts) were placed or removed from the thorax of ‘nectar’-collecting workers of the honeybee (Apis mellifera L.) while they foraged for sugar solution in a patch of artificial flowers. The weights could be added singly, so as to increase the rate at which weight was gained during nectar-collecting independently of crop volume. Extra weights could also be removed so as to keep the bee at a more or less constant total weight while collecting more and more nectar. I investigated whether departure for the hive, before a full crop is obtained (as repeatedly reported in the literature) is due to the effect of an increasing weight of the load. The results did not suggest straightforward effects of an increasing load weight (or crop volume). Rather, it is likely that bees take into account their cumulative metabolic expenditures while exploiting the patch, in the present case irrespective of the temporal pattern of carrying extra loads. 相似文献

A hallmark of Alzheimer’s disease is production of amyloid β peptides resulting from aberrant cleavage of the amyloid precursor protein. Amyloid β assembles into fibrils under physiological conditions, through formation of neurotoxic intermediate oligomers. Tachykinin peptides are known to affect amyloid β neurotoxicity in cells. To understand the mechanism of this effect, we studied how tachykinins affect Aβ(1–40) aggregation in vitro. Fibrils grown in the presence of tachykinins exhibited reduced thioflavin T (ThT) fluorescence, while their morphology, observed in transmission electron microscopy (TEM), did not alter. Cross linking studies revealed that the distribution of low molecular weight species was not affected by tachykinins. Our results suggest that there may be a specific interaction between tachykinins and Aβ(1–40) that allows them to co-assemble. This effect may explain the reduction of Aβ(1–40) neurotoxicity in cells treated with tachykinins. 相似文献
