Solenopsis invicta virus 2 (SINV-2) is the second virus identified from the fire ant, S. invicta Buren. SINV-2 is unique among positive-strand RNA viruses from insects by possessing four cistrons in a monopartite genome. Fire ant colonies testing positive for SINV-2 by RT-PCR did not exhibit any discernable symptoms. RT-PCR-based surveys for SINV-2 among 688 fire ant mounds in Alachua County, Florida, sampled during the period January, 2006 through December, 2007 showed that the prevalence of SINV-2 among nests ranged from 1.6% to 16.4%. Unlike S. invicta virus 1, no seasonal-associated prevalence was observed with regard to SINV-2 infection among fire ant colonies. No social form specificity was evident; SINV-2 was found in both monogyne and polygyne S. invicta ants. Real-time quantitative PCR experiments showed that SINV-2 genome equivalents per individual ant ranged from 1.9 × 107 in. pupae to 4.3 × 1011 in. inseminated queens. The SINV-2 infection was detected in all ant stages examined (eggs, larvae, pupae, workers, and queens). Tissue tropism studies indicated that the alimentary canal (specifically the midgut) is most likely the susceptible tissue. SINV-2 was successfully transmitted to uninfected S. invicta ants by feeding a partially purified homogenate of SINV-2-infected ants. The SINV-2 transmission rate ranged from 30% to 80%, and both positive (genomic) and negative (replicative) SINV-2 RNA strands accumulated in recipient ants over the course of the experiment. Concurrent infections of Solenopsis invicta colonies with S. invicta virus 1 (SINV-1), SINV-2, and SINV-3 has been reported. However, whether individual ants were capable of supporting multiple virus infections simultaneously was not known, nor whether the social form of the colony (polygyne or monogyne) had an influence on the occurrence of multiple infection rates in individual ants. S. invicta field populations were sampled sequentially to establish whether multiple virus infections co-occurred in individual worker ants. In addition, the intra-colony virus infection rates were compared in monogyne and polygyne field colonies to determine whether social form played a role in the viral infection prevalence. All combinations of virus infection (SINV-1, SINV-2, or SINV-3 alone, SINV-1 & SINV-2, SINV-1 & SINV-3, SINV-2 & SINV-3, and SINV-1, SINV-2 & SINV-3) were detected in individual worker ants as well as queens in the field. Thus, individual S. invicta ants can be infected simultaneously with all combinations of the S. invicta viruses. Colony social form did have an influence on the intra-colony prevalence of multiple S. invicta virus infections. Polygyne colonies exhibited significantly greater intra- and inter-colony single and multiple virus infections compared with monogyne colonies.  相似文献

Kneallhazia solenopsae is a pathogenic microsporidium that infects the fire ants Solenopsis invicta and Solenopsis richteri in South America and the USA. In this study, we analyzed the prevalence and molecular diversity of K. solenopsae in fire ants from North and South America. We report the first empirical evidence of K. solenopsae infections in the tropical fire ant, Solenopsis geminata, and S. geminata × Solenopsis xyloni hybrids, revealing an expanded host range for this microsporidium. We also analyzed the molecular diversity at the 16S ribosomal RNA gene in K. solenopsae from the ant hosts S.invicta, S. richteri, S. geminata and S. geminata × S. xyloni hybrids from North America, Argentina and Brazil. We found 22 16S haplotypes. One of these haplotypes (WD_1) appears to be widely distributed, and is found in S. invicta from the USA and S. geminata from southern Mexico. Phylogenetic analyses of 16S sequences revealed that K. solenopsae haplotypes fall into one of two major clades that are differentiated by 2-3%. In some cases, multiple K. solenopsae haplotypes per colony were found, suggesting either an incomplete homogenization among gene copies within the 16S gene cluster or multiple K. solenopsae variants simultaneously infecting host colonies.  相似文献

The morphology and number of ommatidia of the compound eyes of Solenopsis invicta, Solenopsis richteri and their hybrid are investigated. Comparisons are made of the morphology and number of ommatidia in S. invicta, S. richteri and their hybrid. Major workers have significantly more facets than minor workers, male reproductive more than female reproductive, and the reproductives more than workers. Workers and reproductives have significantly more ommatidia in S. invicta than in S. richteri and significantly fewer in the hybrid than in either species. The major workers of each species have significantly wider facets than the minor workers, and workers have significantly wider facets than reproductives, which showed no difference in their facet diameter. There are no significant differences when the facet diameter is compared between the members of the castes of different species. These morphological and numerical dissimilarities are discussed in connection with known behavioral differences in the castes.  相似文献

A series of oligonucleotide primer pairs covering the entire genome of Solenopsis invicta virus 1 (SINV-1) were used to probe the genome of its host, S. invicta, for integrated fragments of the viral genome. All of the oligonucleotide primer sets yielded amplicons of anticipated size from cDNA created from an RNA template from SINV-1. However, no corresponding amplification was observed when genomic DNA (from 32 colonies of S. invicta) was used as template for the PCR amplifications. Host DNA integrity was verified by amplification of an ant-specific gene, SiGSTS1. The representation of fire ant colonies included both social forms, monogyne and polygyne, and those infected and uninfected with SINV-1. Furthermore, no amplification was observed from genomic DNA from ant samples collected from Argentina or the US. Thus, it appears that SINV-1 genome integration, or a portion therein, has not likely occurred within the S. invicta host genome.  相似文献

Quantitative real-time PCR (QPCR) was used to quantify the genome of Solenopsis invicta virus-2 (SINV-2) from infected individual ants of S. invicta. Strand-specific cDNA synthesis oligonucleotide primers and RNase digestion after cDNA synthesis allowed quantification of plus (genomic) and minus (replicative) strands of the SINV-2 genome. Both strands were detected in adult workers and larval fire ants indicating that the virus was replicating within the ant. The differences between the genomic to replicative strand ranged from 199-fold in larvae to 479-fold in workers with an average ratio of 339:1.  相似文献

Loss of natural enemies during colonization is a prominent hypothesis explaining enhanced performance of invasive species in introduced areas. Numerous studies have tested this enemy release hypothesis in a wide range of taxa but few studies have focused on invasive ants. We conducted extensive surveys for the presence of six microbes in recently established populations (California, Australia, New Zealand, Taiwan, and China) of the invasive fire ant Solenopsis invicta. These microbes include Wolbachia, two microsporidia (Kneallhazia solenopsae and Vairimorpha invictae) and three RNA viruses (SINV-1, -2 and -3), all of which previously have been reported in native South American populations of S. invicta. These surveys showed that the total number of enemy species is lower in the recently invaded areas compared with both South American and US populations. Only two microbes were found in any of these recently invaded areas: SINV-1 was detected in all surveyed populations except Australia and New Zealand, and SINV-2 was detected in California and Taiwan only. These results support the general prediction that invasive species lose many of their natural enemies during invasion. Further, the conspicuous absence of some of these microbes in these areas may result from strong selection against founders due to fitness costs associated with harboring detrimental infections rather than the alternative hypothesis that they simply were absent among the original founders. While the successful invasion of S. invicta in these recently invaded areas may be explained partly by the absence of natural enemies, other factors likely have been important as well.  相似文献

The rather unspecific fire ant brood pheromone assay of Glancey et al. (1970) could not be duplicated, probably because they confounded a food response with a brood response. A more specific assay for the brood recognition signal of the red imported fire ant, Solenopsis invicta Buren is described. Worker response in the assay was unaffected by altering social conditions in the colonies but demanded the presence of animal tissue in the ants' diet in order to be consistent. Evidence is presented for a nonvolatile contact brood pheromone that is distributed evenly over the pre-adult cuticle and whose potency is abruptly reduced with the shedding of the pupal skin at eclosion. The signal is completely lost within 72 hr of death.  相似文献

The PBAN/Pyrokinin peptide family is a major neuropeptide family characterized with a common FXPRLamide in the C‐termini. These peptides are ubiquitously distributed in the Insecta and are involved in many essential endocrinal functions, e.g., pheromone production. Previous work demonstrated the localization of PBAN in the fire ant central nervous system, and identified a new family of PBAN from the red imported fire ant, Solenopsis invicta. In this study, we identified five more PBAN/Pyrokinin genes from S. geminata, S. richteri, S. pergandii, S. carolinensis, and a hybrid of S. invicta and S. richteri. The gene sequences were used to determine the phylogenetic relationships of these species and hybrid, which compared well to the morphologically defined fire ant subgroup complexes. The putative PBAN and other peptides were determined from the amino acid sequences of the PBAN/pyrokinin genes. We summarized all known insect PBAN family neuropeptides, and for the first time constructed a phylogenetic tree based on the full amino acid sequences translated from representative PBAN cDNAs. The PBAN/pyrokinin gene is well conserved in Insecta and probably extends into the Arthropod phylum; however, translated pre‐propeptides may vary and functional diversity may be retained, lost, or modified during the evolutionary process. Abstract 1. The imported fire ants, Solenopsis invicta Buren (red), S. richteri Forel (black), and their hybrids (S. invicta × S. richteri) are sympatric congeners with overlapping but shifting spatial distributions in northern Alabama and Mississippi, United States. 2. The abilities of workers of the three ant forms (or species) to recognise non‐nestmate conspecifics and heterospecifics were compared in separate aggression bioassays using three group sizes of competitor numbers (battles): 1‐1, 5‐1, and 5‐5 resident–intruder battles. 3. Workers of all three forms showed significant aggression toward non‐nestmate conspecifics and heterospecifics. The results of the intra‐specific aggression bioassays showed that S. invicta had the lowest aggression threshold, whereas S. richteri was the least aggressive. Survival rates 1 day after intra‐specific encounters were higher for S. richteri than for S. invicta or hybrids, consistent with its lower aggressiveness. 4. In inter‐specific interactions, S. invicta workers showed the greatest aggression and were more aggressive towards S. richteri than to hybrids. Furthermore, survival 1 day after inter‐specific encounters was highest for S. invicta workers, whether they were the intruders or residents. The hybrid form was intermediate between both parental forms in aggression and post‐aggression survival. 5. The live 1‐1 and 5‐5 battles produced similar and consistent results, but differences were amplified in the 5‐5 battles, suggesting an effect of group size on aggression. These results may explain the observed shifts in the spatial distributions of the three forms and range restriction of S. richteri in southern United States.  相似文献

The red imported fire ant, Solenopsis invicta Buren, is an invasive pest from South America that currently occupies much of the south‐eastern USA. Global warming is likely to allow range expansion of many invasive species, including S. invicta. We used a dynamic, ecophysiological model of fire ant colony growth coupled with models simulating climate change to predict the potential range expansion of S. invicta in the eastern USA over the next century. The climate change scenario predicted by the Vegetation–Ecosystem Modelling and Analysis Project (VEMAP) was used in our analyses. Our predictions indicate that the habitable area for S. invicta may increase by c. 5% over the next 40–50 years (a northward expansion of 33 ± 35 km). As the pace of global warming is expected to quicken in the latter half of the century, however, the habitable area for S. invicta in 2100 is predicted to be > 21% greater than it currently is (a northward expansion of 133 ± 68 km). Because the black imported fire ant, S. richteri Forel, occupies higher latitudes than S. invicta, the overall area of the eastern USA infested with invasive Solenopsis species could be greater than that estimated here.  相似文献

Division of labour in Solenopsis invicta follows a familiar pattern: younger, smaller ants tend toward brood care while older, larger ants tend toward foraging. However, long-term observations of marked individuals reveal that length of nursing and foraging ‘careers’ and the age of transition between these activities vary considerably between and within size groups, and are related to length of life. Experiments with entire colonies show that larger ants are more likely than smaller ants to forage for insect prey. There are two main worker castes, ‘nurses’ and ‘foragers’, whose members span a wide age-size range, and a large ‘reserve’ subcaste, heterogeneous in age, size, and behaviour: reserves may nurse, forage, store liquid food, or relay food from nurses to foragers. The proportion of ants engaged in foraging decreases with colony size because many ants in large colonies are not exposed to recruitment signals.  相似文献

The specificity of the trail pheromones of four Solenopsis species was determined using natural trails. Dufour's gland extracts, and purified fractions from Dufour's gland extracts collected after gas-chromatographic separation. S. richteri and S. invicta possess species-specific major trail pheromones, while S. geminata and S. xyloni appear to have a common trail pheromone. Preliminary chemical characterization of the main trail pheromone of S. richteri indicates a M.W. of 218 and empirical formula of C16H26. The trail pheromone system of S. richteri consists of a blend of compounds and this phenomenon may also occur in the other species. The lowest concentration of their trail pheromone that workers of S. richteri could detect was about 10 fg per cm. The significance of blends of pheromones being utilized to generate chemical trails is discussed.  相似文献

Gram-negative, facultatively anaerobic bacteria were isolated from symptomatic oak tissue in the UK and USA. Partial gyrB sequencing placed ten strains in the genus Brenneria, with B. goodwinii as the closest phylogenetic relative. The strains were investigated further using a polyphasic approach including MLSA (based on partial gyrB, rpoB, infB and atpD gene sequences), 16S rRNA gene sequencing, DNA–DNA relatedness studies and both phenotypic and chemotaxonomic assays. The MLSA and 16S rRNA gene analyses separated the strains into two groups based on origin, suggesting that they belong to Brenneria as two novel species. However, the DNA–DNA relatedness values revealed a closer relationship between the groups and indicated that they should belong to the same species. As the two groups of strains from the UK and USA can be differentiated from each other phenotypically and by ERIC PCR fingerprints, it is proposed to classify them as novel subspecies of a novel Brenneria species. The name Brenneria roseae sp. nov. (FRB 222T = LMG 27714T = NCPPB 4581T) is proposed, with Brenneria roseae subsp. roseae ssp. nov. (FRB 222T = LMG 27714T = NCPPB 4581T) for the strains from the UK and Brenneria roseae subsp. americana ssp. nov. (FRB 223T = LMG 27715T = NCPPB 4582T) for the strains from the USA.  相似文献

Specimens of seven fire ant species collected from their native ranges in Argentina were studied by protein electrophoresis and morphological analysis. Concordance between the genetic and morphological character sets is strong (96% agreement on identifications), suggesting that recognition of reproductively isolated populations and partitioning of intra- and interspecific variation can in most cases be achieved using appropriate characters of either type in this taxonomically difficult group. Genetic differentiation between native (Argentina) and introduced (USA) conspecific populations of two species, Solenopsis invicta and S. richteri, is rather typical of the differentiation existing between conspecific populations found within either country. Furthermore, there appears to have been little reduction of variability (heterozygosity) at enzyme loci following colonization by either species of the United States, although some rare alleles have been lost in the introduced populations. Hybridization is rare between S. invicta and S. richteri where their native ranges overlap in central Argentina, in contrast to the extensive hybridization of these species in the United States, suggesting that prezygotic barriers to gene flow have been compromised in introduced populations. Phylogenetic analysis of the seven species indicates that S. invicta and S. richteri are relatively distantly related within the S. saevissima complex. Given that hybrids between these species in the United States suffer little apparent loss of fitness, genomic incompatibilities generally may be insufficient to create effective postzygotic barriers to interspecific gene flow in this group of ants.  相似文献

Using radiolabelled triglycerides and fatty acids we have shown that these 2 lipids are absorbed into the haemolymph of the imported fire ant, Solenopsis invicta Buren from the post-pharyngeal gland. The post-pharyngeal glands attain their greatest weight and contain the highest hexane extractable lipid in establishing queens which have just reared the first minum brood. The lipid content of the digestive system is greatest in queens initiating a mating flight with the majority of the lipid contained in the crop. During colony establishment by the queen the lipid content of the crop moves forward, some is fed to the developing larvae and some moves into the post-pharyngeal gland. The hexane extractable lipid from the postpharyngeal gland of newly mated queens consists of hydrocarbons, sterols. tri-, di-, and monoglycerides and free fatty acids with a trace of wax esters.Excision of the post-pharyngeal glands from mated established queens causes no noticeable change in their behaviour or the behaviour they elicit from the remainder of the colony. However, post-pharyngeal glandectomized females lost weight and died in ca. 2 months although their crop and midgut contained food. The post-pharyngeal glands therefore appear to function in much the same way as a gastric caecum.  相似文献

【目的】2013年10月云南省元谋县首次发现红火蚁。掌握红火蚁蚁巢各品级发生动态,能为当地红火蚁的防控提供技术支撑。【方法】2015年1—12月,在昆明宜良县对红火蚁蚁巢取样、分离,红火蚁取样、称重,统计红火蚁各品级的头数。【结果】地上蚁巢红火蚁蚁群数量发生高峰期在8—9月,其次是3—4月。蚁后数量比例高峰在3—4月;有翅雌蚁比例高峰在4月,其次是7月;有翅雄蚁比例高峰的在5月,其次是9月;生殖蚁幼虫和蛹比例高峰的3、4和12月。地上蚁巢工蚁、兵蚁、职能蚁幼虫和蛹数量发生的高峰期在8—9月,发生小高峰期在4月。【结论】昆明红火蚁蚁群数量动态有2个峰值,第一个峰值为8—9月,以职能蚁防治为主;第二个蜂值3—4月,以生殖蚁和职能蚁防治为主。该结果为昆明红火蚁防控技术提供了数据支撑。  相似文献

Summary In the fire ant,Solenopsis invicta, some winged virgin queens are known to shed their wings (dealate) upon removal of the mated mother queen. These virgin queens then develop their ovaries and begin to lay eggs, thereby foregoing the option of leaving on mating flights and attempting to found their own colonies. Such a response of virgin queens to queenlessness has not been reported for other ants. In order to determine if virgin queens of some other fire ants (subgenusSolenopsis) would respond in the same way, experiments were conducted onS. richteri, hybridS. invicta/richteri andS. geminata, a member of a species complex different from that of the other taxa. Just as inS. invicta, virgin queens ofS. richteri and the hybrid dealated and began to lay eggs within days of the removal of the queen. In addition, workers executed many of the reproductively active virgin queens, a phenomenon also found inS. invicta. In contrast, virgin queens ofS. geminata did not dealate or quickly begin to lay eggs upon separation from the queen. Reasons for the variability in the response of virgin queens of the different species may be 1) higher probability of reproductive success for unmated dealated queens compared to normal claustral founding inS. invicta andS. richteri linked to relatively frequent loss of the mother queen; or 2) phylogenetic constraint.  相似文献
