Effects of hybridization on sesquiterpene lactones in Xanthium strumarium were studied for clues to the relationship of Old and New World populations. In crosses between indigenous Asiatic plants in the “strumarium” morphological complex and various American complexes that produce xanthinin as a major sesquiterpene lactone, the F1 hybrids contained xanthinin and the related compounds, xanthanol, xanthatin, and xanthinosin. In other crosses with various American complexes that produce xanthumin, the stereoisomer of xanthinin, the F1 hybrids contained xanthinin and xanthumin as well as xanthinin-related compounds and their stereoisomers, xanthumanol, deacetoxylxanthumin, and tomentosin. The Asiatic plants of “strumarium” from Hong Kong involved in the crosses produce approximately equal percentages of xanthinin, xanthatin and xanthinosin, but those from India contain only xanthinosin. The putative introduction of the American morphological complex, “chinense,” contains xanthumin as the major component but shows chemical diversity that indicates genes derived from the indigenous “strumarium” complex.  相似文献

Morphologically diverse plants in a wild population of Xanthium strumarium in eastern Texas produced widely variant combinations of sesquiterpene lactones. Some plants showed a preponderance of xanthinin and related compounds, xanthanol and xanthatin, while others a predominance of stereoisomers, xanthumin, xanthumanol and deacetoxylxanthumin. Some plants produced only one sesquiterpene lactone and other plants yielded various mixtures of xanthinin, xanthumin and related compounds, including xanthinosin and/or tomentosin. Experimental hybridization between plants containing primarily xanthinin and plants containing xanthumin, but originating in diverse parts of the world, yielded F1 progenies with xanthanol and/or xanthumanol as the predominant sesquiterpenoid component. The F2 generation showed segregation of the xanthinin, xanthumin and related compounds. The Texas population occurs in an area where the “italicum” (xanthinin-predominants) and “chinense” (xanthumin-predominant) morphological complexes are sympatric. The diverse patterns of sesquiterpene lactones in the Texas population probably resulted from the natural hybridization of morphological complexes that originally were spatially isolated.  相似文献

In Drosophila melanogaster there is one nucleolar organizer (NO) on each X and Y chromosome. Experiments were carried out to compare the ribosomal RNAs derived from the two nucleolar organizers. 32PO4-labelled ribosomal RNA was isolated from two strains of D. melanogaster, one containing only the X chromosome NO, the other containing only the Y chromosome NO. 28 S and 18 S RNA from the two strains were subjected to a variety of “fingerprinting” and sequencing procedures. Fingerprints of 28 S RNA were very different from those of 18 S RNA. Fingerprints of “X” and “Y” 28 S RNA were indistinguishable from each other, as also were fingerprints of “X” and “Y” 18 S RNA. In combined “T1 plus pancreatic” RNAase fingerprints several distinctive products were characterized and quantitated. Identical products were obtained from X and Y RNA, and the molar yields of the products were indistinguishable. Together these findings imply that the rRNA sequences encoded by the X and Y NOs are closely similar and probably identical to each other.Two further findings were of interest in “T1 plus pancreatic” RNAase fingerprints: (1) in 28 S (as well as in 18 S) fingerprints several distinctive products were recovered in approximately unimolar yields. This indicates that 28 S RNA does not consist of two identical half molecules, though it does consist of two non-identical half molecules together with a “5.8 S” fragment. (2) Several methylated components in Drosophila rRNA also occur in rRNA from HeLa cells and yeast. This suggests that certain features of rRNA structure involving methylated nucleotides may be highly conserved in eukaryotic evolution.  相似文献

A growth regulator (G; 4-ethyl-1-hydroxy-4,8,8,10,10 pentamethyl-7,9-dioxo-2,3 dioxyabicyclo (4.4.0) decene-5) from Eucalyptus grandis (Maiden) reduced stomatal conductance and also photosynthetic capacity when fed through the transpiration stream of detached leaves. The concentration of G required for this effect was high (10−4 molar), but the amount of G taken up (dose) was below the level which has previously been found in E. grandis leaves. Similar effects were observed in detached leaves of Xanthium strumarium L. though almost 10 times more G was required. G reduced CO2-dependent O2 evolution from isolated cells of X. strumarium. In spinach (Spinacia oleracea L.) chloroplasts, electron transport through photosystem II was reduced by G. It is proposed that G affects stomatal conductance and photosynthesis by reducing photosystem II activity in both the guard cell chloroplasts and mesophyll cell chloroplasts.  相似文献

Xanthanolides, as the sesquiterpene lactones, are reportedly the major components for the pharmacological properties of X. strumarium L. species. Phytochemical studies indicated that the glandular structures on the surface of plant tissues would form the primary sites for the accumulation of this class of the compounds. As the interface between plants and their natural enemies, glandular trichomes may vary with respect to which of their chemicals are sequestered against different herbivores in different ecologies. However, to date, no data are available on the chemical characterisation of X. strumarium glandular cells. In this study, the trichome secretions of the X. strumarium species originating from nineteen unique areas across eleven provinces in China, were analysed by HPLC, LC-ESI-MS and NMR. For the first time three distinct chemotypes of X. strumarium glandular trichomes were discovered along with the qualitative and quantitative evaluations of their presence of xanthanolides; these were designated glandular cell Types I, II, and III, respectively. The main xanthanolides in Type I cells were 8-epi-xanthatin and xanthumin while no xanthatin was detected. Xanthatin, 8-epi-xanthatin, and xanthumin dominated in Type II cells with comparable levels of each being present. For Type III cells, significantly higher concentrations of 8-epi-xanthatin or xanthinosin (relative to xanthatin) were detected with xanthinosin only being observed in this type. Further research will focus on understanding the ecological and molecular mechanism causing these chemotype differences in X. strumarium glandular structures.  相似文献

Gains of the feedback loops involving intercellular CO2 concentration on one hand, and CO2 assimilation and stomata on the other (= assimilation loop with gain [GA] and conductance loop with gain [Gg]) were determined in detached leaves of Amaranthus powelli S. Wats., Avena sativa L., Gossypium hirsutum L., Xanthium strumarium L., and Zea mays in the absence and presence of 10−5 m (±) abscisic acid (ABA) in the transpiration stream. Determinations were made for an ambient CO2 concentration of 300 microliters per liter. In the absence of ABA, stomata were insensitive to CO2 (Gg between 0.00 and −0.02) in A. sativa, G. hirsutum, and X. strumarium, sensitive in A powelli (Gg = −0.46), and very sensitive in Z. mays (Gg = −3.6). Addition of ABA increased the absolute values of the gain of the conductance loop in A. powelli (Gg = −2.0), G. hirsutum (Gg = −0.31), and X. strumarium (Gg = −1.14). Stomata closed completely in A. sativa. In Z. mays, Gg decreased after application of ABA to a value of −0.86, but stomatal sensitivity to CO2 increased for intercellular CO2 concentrations < 100 microliters per liter. The gain of the assimilation loop increased after application of ABA in all cases, from values between 0.0 (A. powelli) and −0.21 (Z. mays) in the absence of ABA to values between −0.19 (A. powelli) and −0.43 (Z. mays) in the presence of ABA. In none of the species examined did ABA affect the photosynthetic capacity of the leaves.  相似文献

A major quantitative trait locus (QTL) for Fusarium oxysporum Fr. f. sp. niveum race 1 resistance was identified by employing a “selective genotyping” approach together with genotyping-by-sequencing technology to identify QTLs and single nucleotide polymorphisms associated with the resistance among closely related watermelon genotypes.

Abstract

Fusarium wilt is a major disease of watermelon caused by the soil-borne fungus Fusarium oxysporum Schlechtend.:Fr. f. sp. niveum (E.F. Sm.) W.C. Snyder & H.N. Hans (Fon). In this study, a genetic population of 168 F3 families (24 plants in each family) exhibited continuous distribution for Fon race 1 response. Using a “selective genotyping” approach, DNA was isolated from 91 F2 plants whose F3 progeny exhibited the highest resistance (30 F2 plants) versus highest susceptibility (32 F2 plants), or moderate resistance to Fon race 1 (29 F2 plants). Genotyping-by-sequencing (GBS) technology was used on these 91 selected F2 samples to produce 266 single nucleotide polymorphism (SNP) markers, representing the 11 chromosomes of watermelon. A major quantitative trait locus (QTL) associated with resistance to Fon race 1 was identified with a peak logarithm of odds (LOD) of 33.31 and 1-LOD confidence interval from 2.3 to 8.4 cM on chromosome 1 of the watermelon genetic map. This QTL was designated “Fo-1.1” and is positioned in a genomic region where several putative pathogenesis-related or putative disease-resistant gene sequences were identified. Additional independent, but minor QTLs were identified on chromosome 1 (LOD 4.16), chromosome 3 (LOD 4.36), chromosome 4 (LOD 4.52), chromosome 9 (LOD 6.8), and chromosome 10 (LOD 5.03 and 4.26). Following the identification of a major QTL for resistance using the “selective genotyping” approach, all 168 plants of the F 2 population were genotyped using the SNP nearest the peak LOD, confirming the association of this SNP marker with Fon race 1 resistance. The results in this study should be useful for further elucidating the mechanism of resistance to Fusarium wilt and in the development of molecular markers for use in breeding programs of watermelon.  相似文献

Abstract At an applied nitrate concentration of 1 mol m?3, the proportion of xylem sap nitrogen as nitrate was < 15% for Cajanus cajan, Lupinus albus and Trifolium repens, 33% for Pisum sativum and within the range 57–62% for Glycine max, Phaseolus vulgar is, Stellaria media and Xanthium strumarium. At an applied nitrate concentration of 10 mol m~3 the value had increased to 66% for T. repens while at 20 mol m?3 nitrate values had increased to 46, 51 and 49% for C. cajan, L. albus and Pisum sativum, respectively, and 89% and 85% for 5. media and X. strumarium, respectively. Glycine max and Phaseolus vulgaris differed from the other species in that the proportion of their xylem sap nitrogen as nitrate remained constant (~ 60%) as applied nitrate concentration increased from 1 to 20 mol m?3. The proportion of total plant nitrate reductase activity in the shoot of C. cajan, S. media and X. strumarium increased as applied nitrate concentration increased from 1 to 20 mol m?3. Values at the lower and upper concentrations were, respectively, 26 and 72% for C. cajan. 48 and 80% for X. strumarium and 68 and 87% for S. media. The partitioning of nitrate assimilation between root and shoot in these species is discussed.  相似文献

As plants have been shown to accumulate organic compounds from contaminated sediments, there is a potential for long-lasting ecological impact as a result of contaminant accumulation in riparian areas of wetlands, particularly the accumulation of non-biodegradable contaminants such as perfluorooctanoic acid (PFOA). In this study, commonly found riparian wetland plants including reeds, i.e., Xanthium strumarium, Phragmites australis, Schoenoplectus corymbosus, Ruppia maritime; Populus canescens, Polygonum salicifolium, Cyperus congestus; Persicaria amphibian, Ficus carica, Artemisia schmidtiana, Eichhornia crassipes, were studied to determine their susceptibility to PFOA accumulation from PFOA contaminated riparian sediment with a known PFOA concentration, using liquid chromatography/tandem mass spectrometry (LC/MS/MS). The bioconcentration factor (BCF) indicated that the plants affinity to PFOA accumulation was; E. crassipes, > P. salicifolium, > C. congestus, > P. x canescens, > P. amphibian, > F. carica, > A. schmidtiana, > X. strumarium, > P. australis, > R. maritime, > S. corymbosus. The concentration of PFOA in the plants and/or reeds was in the range 11.7 to 38 ng/g, with a BCF range of 0.05 to 0.37. The highest BCF was observed in sediment for which its core water had a high salinity, total organic carbon and a pH which was near neutral. As the studied plants had a higher affinity for PFOA, the resultant effect is that riparian plants such as E. crassipes, X. strumarium, and P. salicifolium, typified by a fibrous rooting system, which grow closer to the water edge, exacerbate the accumulation of PFOA in riparian wetlands.  相似文献

Climatic matching and pre-release performance evaluation were useful predictors of parasitoid establishment in a retrospective analysis of a classical biological control program against Bemisia tabaci biotype “B” in the USA. Laboratory evaluation of 19 imported and two indigenous parasitoid species in quarantine on B. tabaci showed that the Old World Eretmocerus spp, had the highest attack rate. The climate matching program CLIMEX was used to analyze the establishment patterns of five Old World Eretmocerus spp. introduced to the Western USA. The top matches ±10% for the climate of the area of introduction and origin of the introduced parasitoids always included the species that established. The Old World Eretmocerus spp. came from regions characterized by many separate biotypes of B. tabaci other than “B,” but are considered specialists of the B. tabaci complex as compared to the indigenous North American oligophagous Eretmocerus spp. This narrower host range and high attack rate combined with climatic adaptation may account for their establishment in the USA. A set of predictive tools and guidelines were used to select the best candidate for importation and possible release into Australia that has been recently invaded by the “B” biotype. The establishment patterns of the introduced Eretmocerus spp. and a comparison of climates of their respective locations in the USA were compared with the affected area in Australia. The best climatic match was the Lower Rio Grande Valley of Texas suggesting its dominant parasitoid, E. hayati ex. Pakistan be considered as the first candidate for evaluation as a biological control agent.  相似文献

Despite much effort towards resolving the molecular phylogenetic tree for pitvipers, some aspects remain unresolved. In particular, the sister group of the diverse New World radiation has remained impossible to identify with any certainty. In this study, which for the first time includes nuclear intron data from all major groups of Asian pitvipers as well as representatives of the New World radiation, Bayesian inference allows Gloydius to be identified as the most likely sister group to the New World radiation and sheds light on other ambiguous relationships among the Old World pitvipers. The sister group relationship of “Ovophisokinavensis and “Trimeresurusgracilis is confirmed by the addition of nuclear genes, and we hypothesise that they form a sister group to the Gloydius + New World clade, best supported when the phylogenetic signal from gaps is included in the form of a simple-coded matrix.  相似文献

Effects of methanol extracts of Xanthium strumarium on different cancer cell lines and on the mortality rates of Aedes caspius, Culex pipiens (Diptera: Culicidae) were investigated. Among the cell lines tested, the Jurkat cell line was the most sensitive to the methanol extract and ethyl acetate fraction, with reported LC50 values of 50.18 and 48.73 μg/ml respectively. Conversely, methanol extracts were not that toxic to the A549 cell line though the toxicity increased on further purification. The percentage of growth inhibition was dose dependent for the methanol extract and ethyl acetate fraction. The ethyl acetate fraction showed higher toxicity to all cell lines tested when compared to the methanol extract. The results showed that methanol extracts of plant seeds caused 100% mortality of mosquito larvae at a concentration of 1000 μg/ml after 24 h of treatment. The LC50 and LC90 values of X. strumarium were found to be 531.07 and 905.95 μg/ml against Ae. caspius and 502.32 and 867.63 μg/ml against Cx. Pipiens, respectively. From the investigations, it was concluded that the crude extract of X. strumarium showed a weak potential for controlling the larval instars of Ae. caspius and Cx. pipiens. However, on further purification the extract lost the larvicidal activity. The ethyl acetate fraction showed higher toxicity to all cell lines tested when compared to the methanol extract. The ethyl acetate fraction investigated in this study appears to have a weak larvicidal activity but a promising cytotoxic activity. Future studies will include purification and investigation in further detail of the action of X. strumarium on Cancer Cell Lines and mosquitoes.  相似文献

In this preliminary study defense behaviour patterns (fear responses) are described in two closely related, behaviourally different inbred labyrinth fish subspecies and in their F1 generation. The subspecies M. opercularis (characterized briefly by “active escape”) and M. opercularis concolor (characterized by “passive escape”) showed specific differences in the manifestation of certain defense behaviour patterns. In the F1 hybrid generation dominance and overdominance of M. opercularis was found in most defense behaviour patterns. Analysing the frequencies and sequences of movement patterns it could be shown that defensive behaviour is not a random or entirely “plastic” process but that there is sequential linkage between the patterns and they form characteristic clusters. Our results suggest that manifestations of different patterns are under genetic control and presumably, genetic determination of certain patterns is not very complex. Attempts were made to determine whole brain noradrenaline, serotonine and dopamine levels of the two subspecies and a significant difference was found in the noradrenaline content.  相似文献

Thioredoxins (Trxs) are small ubiquitous oxidoreductases that participate in dithiol-disulphide exchange reactions. They are present in all organisms from prokaryotes to higher eukaryotes and involved in a variety of cellular processes in higher plants. The expression pattern of the multigenic family of grape Trxsh was analysed in different tissues by semiquantitative RT-PCR. The grape Trxh genes were approximately expressed in all tissues, but with different expression patterns than each other. The in silico analysis of the promoter regions of grape Trxs h demonstrated the presence of a number of potential cis-acting elements to respond to environmental signals, suggesting that Trxsh may respond to a variety of environmental signals, including dehydration, salinity, heat, cold, heavy metals, light, pathogens, and plant hormones. The grape Trx h genes were also found to be differentially induced under abiotic stress conditions due to the presence of different putative regulatory elements in their promoters. Collectively, the distinct expression patterns and different responses to environmental stress conditions suggest that each isoform of the multigenic family of VvTrxh may have a specific and non-redundant function in grape.  相似文献

Premature flowering reduces the yield and quality of the harvested fleshy taproot in radish. However, there has been little molecular marker research on the radish late-bolting trait. In this study, F2 and F2:3 populations derived from a cross of “Ninengo” (late-bolting) and “Maer” (early-bolting) were analyzed to map late-bolting genes. Five hundred insertion and deletion (InDel) markers were designed according to the whole-genome resequencing data of the two parents. A genetic map was constructed based on the F2 population, and a late-bolting gene was detected in a 1.1-cM region between the markers InDel520 and InDel535 on chromosome R02 that explained the highest (76.4%) phenotypic variance. RsFLC2 was identified as a candidate gene in this region. Notably, “Ninengo” contains a 1627-bp insertion near the 5′ end of the first intron of RsFLC2. Allelic variation analyses in the F2 population further validated that RsFLC2was associated with the late-bolting trait in radish. The expression pattern of RsFLC2 was significantly different between “Ninengo” and “Maer” during vernalization. Vernalization suppressed RsFLC2 expression, and the 1627-bp insertion in the first intron weakened gene repression in “Ninengo” plants, resulting in late-bolting. This study lays a foundation for uncovering the molecular mechanism of late-bolting and marker-assisted selection for breeding late-bolting varieties of radish.  相似文献

Changes in the level of hydrogen peroxide (H2O2) and activity of peroxidases towards phenolic substrates (EC 1.11.1.7) such as pyrogallol (PPX), syringaldazine (SPX) and guaiacol (GPX), and cytosolic ascorbate peroxidase (cAPX, EC 1.11.1.11) in response to infestation of cowpea aphid (Aphis craccivora Koch) were analyzed in soybean (Glycine max (L.) Merr. cv. “Nam Dan”) at the V3 stage (first two trifoliate leaves fully developed, third trifoliate leaf unrolled) for 96 h post-infestation (hpi). Influence of A. craccivora at a varied population size (10, 20 and 30 individuals per each soybean plant) caused a burst of H2O2 generation in the aphid-infested leaves at 12 hpi. Paralleling the H2O2 accumulation, peroxidase activity in all the infested plants remarkably increased and was significantly higher than that observed in controls (uninfested plants). The cascade of enzymes induced was continuously overlapped by the early enhancement of SPX within 6–24 hpi, an expression of cAPX (12–48 hpi) followed by an accumulation of GPX (24–72 hpi) and PPX (24–96 hpi). The differential induction of SPX, GPX, PPX and cAPX resulted in a rapid reduction of H2O2 content in aphid-infested leaves, and the activity of peroxidase was closely correlated with the intensity of A. craccivora infestation around the defined points of time at which the activity of each enzyme reached the maximum level. The increase in activity of peroxidases matched their function as controlling accumulation of H2O2 and detoxifying this reactive oxygen product when soybean plants were challenged with cowpea aphid. Furthermore, peroxidases could directly deter cowpea aphid feeding through other functions such as the anti-nutritive and/or toxicological defenses and/or limiting the penetration of aphid stylets into plant tissues via participating to strengthen and reinforce the cell wall barrier. These results indicated that peroxidases may be some elements of the defense system that increased the resistance of G. max cv. “Nam Dan” to infestation of A. craccivora.  相似文献

Saudi Arabia has no permanent lakes or rivers but has wet meadows, which are desert wetlands temporarily formed due to the seasonal rainfall. This work investigates the environmental variables' impact on the vegetation pattern in the desert-wetland ecosystem in Taif highlands. Forty-one stands were randomly selected representing three main habitats (wet meadows, slopes, and terraces) to study their floristic features and vegetation analysis. A total of 142 species were recorded belonging to 111 genera and 45 families in the desert wetlands of Taif Province. About 64.1% were natural plants, while 25.4% were segetal weeds, 7.7% were aquatic plants, and 2.8% were plants that escaped from cultivation. Therophytes dominated over the other life forms, and mono-regional taxa were the dominant chorotype. Multivariate analysis of the recorded plants produced eight vegetation groups; four of them (A: Potamogeton nodosus-Nasturtium officinale, B: Lemna gibba-Leptochloa fusca, C: Typha domingensis- Xanthium strumarium and D: Conyza stricta- Cyperus longus) represented the wet meadows, while two (E: Acacia gerrardii- Commicarpus plumbagineus and H: Osteospermum vaillantii- Eragrostis Pilosa) for slopes and other two (F: Argemone ochroleuca-Cyperus rotundus and G: Pulicaria undulata- Solanum incanum) for the desert terraces. The vegetation zonation was clear, which started from real aquatic species in the wet meadows passing through mixed vegetation in the slopes and ended with proper xerophytic vegetation in the terraces. Conyza stricta- Cyperus longus community had the highest species diversity, while that of Potamogeton nodosus-Nasturtium officinale had the lowest. The principal component analysis indicated that HCO3, NO3, Mg, Cl, Ca, and pH values were the most effective soil variable. The presence of several segetal weeds suggests the alteration of the natural status of the desert-wetland ecosystem, and thus the conservation of these natural habitats becomes urgent.  相似文献

Associations between individual foods or nutrients and oxidative markers have been reported. Comprehensive measures of food intake may be uniquely informative, given the complexity of oxidative systems and the possibility of antioxidant synergies. We quantified associations over a 20-year history between three food-based dietary patterns (summary measures of whole diet) and a plasma biomarker of lipid peroxidation, F2-isoprostanes, in a cohort of Americans ages 18–30 at year 0 (1985–1986). We assessed diet at years 0, 7, and 20 through a detailed history of past-month food consumption and supplement use and measured plasma F2-isoprostanes at years 15 and 20. We created three dietary patterns: (1) a priori (“a priori diet quality score”) based on hypothesized healthfulness of foods, (2) an empirical pattern reflecting high fruit and vegetable intake (“fruit–veg”), and (3) an empirical pattern reflecting high meat intake (“meat”). We used linear regression to estimate associations between each dietary pattern and plasma F2-isoprostanes cross-sectionally (at year 20, n=2736) and prospectively (year 0/7 average diet and year 15/20 average F2-isoprostanes, n=2718), adjusting for age, sex, race, total energy intake, education, smoking, body mass index, waist circumference, physical activity, and supplement use. In multivariable-adjusted cross-sectional analysis, the a priori diet quality score and the fruit–veg diet pattern were negatively, and the meat pattern was positively, associated with F2-isoprostanes (all p values <0.001). These associations remained statistically significant in prospective analysis. Our findings suggest that long-term adherence to a diet rich in fruits and vegetables and low in red meat may decrease lipid peroxidation.  相似文献
