The aim of the study was to investigate the incorporation of the antibacterial agent, miconazole nitrate into cyclodextrin cavities covalently bonded onto cloth fibers. The cellulosic fabric was grafted with β-cyclodextrin molecules through reaction with monochlorotriaziny β-cyclodextrin (MCT-β-CD). The suitable bonded reaction conditions were found to be MCT-β-CD 60–100 g/L, catalyst Na2CO3 50–60 g/L, the reaction temperature 150–160 °C and the reaction time 5–8 min.

The modified and unmodified fabrics were characterized by UV spectrophotometry. The level of miconazole nitrate entrapped in the fabrics were determined by HPLC and was founded to be much higher (0.458% w/w) for the textile functionalized with MCT-β-CD compared to the unmodified fabric (0.056% w/w). The antibacterial abilities measured by shaker flask method showed that the antibacterial property was markedly enhanced by impregnation with miconazole nitrate of the MCT-β-CD grafted textile. The finished fabric kept the antibacterial abilities more than 70% even after washing 10 times, while the antibacterial activity of the unmodified textile was almost lost.  相似文献

Based on the prodrug concept as well as the combination of two different classes of antimalarial agents, we designed and synthesized two series of ferrocenic antimalarial dual molecules consisting of a ferroquine analogue conjugated with a glutathione reductase inhibitor (or a glutathione depletor) through a cleavable amide bond in order to target two essential pathways in the malarial parasites. The results showed no enhancement of the antimalarial activity of the dual molecules but evidenced a unique mode of action of ferroquine and ferrocenyl analogues distinct of those of chloroquine and nonferrocenic 4-aminoquinoline analogues.  相似文献

After the accident at the Fukushima Dai-ichi Nuclear Power Plant in 2011, high activities of radiocaesium have been reported in wild mushrooms in Japan. Fungi play an important role in the dynamics of radiocaesium in forest ecosystems. We examined the contents of caesium (Cs), rubidium (Rb), and potassium (K) in the mycelium of 15 isolates of ectomycorrhizal (EM) fungi and nine isolates of saprotrophic (SA) fungi in a synthetic medium with either ammonium chloride (NH4Cl) or sodium nitrate (NaNO3), supplemented with 1 ppm caesium chloride and rubidium chloride. The mycelia were harvested after 8 weeks of incubation, and the contents of Cs, Rb, and K were measured by inductively coupled plasma mass spectrometry. The dry weight of the mycelium in the medium with NH4 was significantly higher than that with NO3, although some EM species, Hebeloma, Astraeus, Scleroderma, and Pisolithus, grew well in the medium with NO3. Among SA species, Crucibulum and Cyathus grew in the medium with NO3. The uptakes of Cs, Rb, and K by Suillus, Pisolithus, and Rhizopogon were higher than that in other EM and SA species when they grew on the medium with NH4, while the uptakes of these elements by Astraeus and Scleroderma were higher than those by other species grown on the medium with NO3. The content of Rb was positively correlated with Cs (r = 0.85, p < 0.001) and K (r = 0.51, p < 0.001). The accumulation of Cs, Rb, and K was differently affected by the N source and fungal species.  相似文献

Block copolymers containing stimuli-responsive segments provide important new opportunities for controlling the activity and aggregation properties of protein-polymer conjugates. We have prepared a RAFT block copolymer of a biotin-terminated poly(N-isopropylacrylamide) (PNIPAAm)-b-poly(acrylic acid) (PAA). The number-average molecular weight (M(n)) of the (PNIPAAm)-b-(PAA) copolymer was determined to be 17.4 kDa (M(w)/M(n) = 1.09). The PNIPAAm block had an M(n) of 9.5 kDa and the poly(acrylic acid) (PAA) block had an M(n) of 7.9 kDa. We conjugated this block copolymer to streptavidin (SA) via the terminal biotin on the PNIPAAm block. We found that the usual aggregation and phase separation of PNIPAAm-SA conjugates that follow the thermally induced collapse and dehydration of PNIPAAm (the lower critical solution temperature (LCST) of PNIPAAm is 32 degrees C in water) is prevented through the shielding action of the PAA block. In addition, we show that the cloud point and aggregation properties (as measured by loss in light transmission) of the [(PNIPAAm)-b-(PAA)]-SA conjugate also depended on pH. At pH 7.0 and at temperatures above the LCST, the block copolymer alone was found to form particles of ca. 60 nm in diameter, while the bioconjugate exhibited very little aggregation. At pH 5.5 and 20 degrees C, the copolymer alone was found to form large aggregates (ca. 218 nm), presumably driven by hydrogen bonding between the -COOH groups of PAA with other -COOH groups and also with the -CONH- groups of PNIPAAm. In comparison, the conjugate formed much smaller particles (ca. 27 nm) at these conditions. At pH 4.0, however, large particles were formed from the conjugate both above and below the LCST (ca. 700 and 540 nm, respectively). These results demonstrate that the aggregation properties of the block copolymer-SA conjugate are very different from those of the free block copolymer, and that the outer-oriented hydrophilic block of PAA shields the intermolecular aggregation of the block copolymer-SA bioconjugate at pH values where the -COOH groups of PAA are significantly ionized.  相似文献

The technique of vital staining with a fluorochrome and subsequent flow cytometry has been employed to investigate the DNA content of living sperm. There have been several reports of chromosomal damage caused by staining cells with a bis-benzimidazole dye. Hoechst 33342, either with or without subsequent flow cytometry. Therefore the use of this technique might be expected to affect adversely the fertilising capacity of inseminated sperm, or to result in the production of congenital deformities in the young. Insemination experiments to date have resulted in the production of more than 400 offspring in four species, including 5 successive generations of rabbits, where stained or sorted sperm were used. All progeny were observed to be normal by anatomical criteria. There was no evidence to suggest that sperm treated in this manner were unable to fertilise oocytes in vivo or that embryonic development was affected.  相似文献

Summary A method is described for culturing plants at extremely low nutrient concentrations. Using a Braun infusion pump, a fixed amount of nitrate or ammonium was supplied continuously to plants growing in a culture vessel at a rate limiting the uptake of the plants. At a very low nitrogen concentration an equilibrium was established where uptake rate of the plants is equal to the rate of supply by the infusion pump. The nitrogen concentrations reached appeared to be in the order of 1 μM. The method compared the nitrate uptake byHypochaeris radicata L.ssp.radicata, H. radicata ssp.ericetorum Van Soest andUrtica dioica L. and ammonium uptake byH. radicata ssp.radicata andH. radicata ssp.ericetorum. Plants were cultivated in monocultures or in mixed cultures (two species per culture vessel). For the mixed cultures competition for nitrate (or ammonium) between the species was maintained for long periods. The capacities of the uptake systems of two subspecies ofH. radicata from places different in nitrogen supply and pH were adapted equally well to both low nitrate and low ammonium concentrations. Apparently factors other than nitrogen uptake play a part in the distribution of the subspecies. The capacity of the uptake system ofU. dioica, a nitrophilous species, was lower than that ofH. radicata ssp.radicata, a species from places poorer in nitrogen. This difference is related to the different distribution of the two species in the field. The present results are compared with those of previous experiments where Km and Vmax were measured and the significance of both parameters is discussed.  相似文献

The purpose of this study was to identify bacteria that can perform As(III) oxidation for environmental bioremediation. Two bacterial strains, named JHS3 and JHW3, which can autotrophically oxidize As(III)–As(V) with oxygen as an electron acceptor, were isolated from soil and water samples collected in the vicinity of an arsenic-contaminated site. According to 16S ribosomal RNA sequence analysis, both strains belong to the ?-Proteobacteria class and share 99% sequence identity with previously described strains. JHS3 appears to be a new strain of the Acinetobacter genus, whereas JHW3 is likely to be a novel strain of the Klebsiella genus. Both strains possess the aioA gene encoding an arsenite oxidase and are capable of chemolithoautotrophic growth in the presence of As(III) up to 10 mM as a primary electron donor. Cell growth and As(III) oxidation rate of both strains were significantly enhanced during cultivation under heterotrophic conditions. Under anaerobic conditions, only strain JHW3 oxidized As(III) using nitrate or a solid-state electrode of a bioelectrochemical system as a terminal electron acceptor. Kinetic studies of As(III) oxidation under aerobic condition demonstrated a higher V max and K m from strain JHW3 than strain JHS3. This study indicated the potential application of strain JHW3 for remediation of subsurface environments contaminated with arsenic.  相似文献

Standard alginate-polylysine microcapsules containing isolated rat hepatocytes were prepared. These capsules were intraperitoneally implanted into mice, and retrieved after seven days. Histological sections of the recovered microcapsules showed peritoneal lymphocyte and macrophage infiltration. Additional microscopic observations at various stages of the microencapsulation procedure, and histological observations of control non-implanted microcapsules; illustrate that encapsulated cells became embedded within the microcapsular membrane matrix. The microcapsular membrane at these sites appeared thin and often poorly formed. The cellular infiltration into the implanted microcapsules can occur through holes developed in these thin and poorly formed areas found in the microcapsular membrane. Similar observations were seen in microcapsules prepared with 20 x 10(6) and at a lower cell concentration of 10 x 10(6) suspended cells per millilitre of sodium alginate.  相似文献

Gomori reported that aldehyde fuchsin stained the granules of pancreatic islet beta cells selectively and without need of permanganate pretreatment. Others adopted permanganate oxidation because it makes staining faster though much less selective. All aldehyde fuchsins are not equivalent, being made from "basic fuchsin" whose composition may vary from pure pararosanilin to one of its methylated homologs, rosanilin or a mixture. Mowry et al. have shown that only aldehyde fuchsin made from pararosanilin stained unoxidized pancreatic beta cells (PBC). Aldehyde fuchsins made from methylated homologs of pararosanilin stain PBC cells only after oxidation, which induces basophilia of other cells as well; these are less selective for PBC. Is the staining of PBC by aldehyde fuchsins due to insulin? Others have been unable to stain pure insulin with aldehyde fuchsins except in polyacrylamide gels and only after oxidation with permanganate. They have concluded that insulin contributed to the staining of oxidized but not of unoxidized PBC. This view denies any inherent validity of the more selective staining of unoxidized PBC cells as an indication of their insulin content. We describe here indisputable staining of unoxidized pure insulins by aldehyde fuchsin made with pararosanilin. Dried spots of insulin dissolved in the stain unless fixed beforehand. Spots of dried insulin solution made on various support media and fixed in warm formalin vapor were colored strongly by the stain. Insulin soaked Gelfoam sponges were dried, fixed in formalin vapor and processed into paraffin. In unoxidized paraffin sections, presumed insulin inside gel spaces was stained strongly by aldehyde pararosanilin. Finally, the renal tubules of unoxidized paraffin sections of kidneys from insulin-injected mice fixed in either Bouin's fluid or formalin were loaded with material stained deeply by aldehyde pararosanilin. This material was absent in renal tubules of mice receiving no insulin. The material in the spaces of insulin-soaked gels and in the renal tubules of insulin-injected mice was proven to be insulin by specific immunostaining of duplicate sections. The same material was also stained by aldehyde pararosanilin used after permanganate. So, this dye stains oxidized or unoxidized insulin if fixed adequately.  相似文献

The purpose of this study was to evaluate changes in soccer-specific power endurance of 34 female high school soccer players throughout a season either with or without an intermittent, high-intensity exercise protocol. Thirty-four female high school soccer players were tested prior to the 2000 fall season and again 10 weeks later. The tests included an abridged 45-minute shuttle test (LIST), hydrostatic weighing, vertical jump, 20-m running-start sprint, and 30-second Wingate test. The experimental group (EG; n = 17, age 16.5 +/- 0.9 years) completed a 10-week in-season plyometric, resistive training, and high-intensity anaerobic program. The control group (n = 17, age 16.3 +/- 1.4 years) completed only traditional aerobic soccer conditioning. Statistical significance was set at alpha < 0.05. The experimental group showed significant improvements in the LIST (EG = delta 394 seconds +/- 124 seconds), 20-m sprint (EG = Delta-0.10 seconds +/- 0.10 seconds), increase in fat-free mass (EG = delta 1.14 kg +/- 1.22 kg), and decreases in fat mass (EG = Delta-1.40 kg +/- 1.47 kg) comparing pre- to postseason. This study indicates that a strength and plyometric program improved power endurance and speed over aerobic training only. Soccer-specific power endurance training may improve match performance and decrease fatigue in young female soccer players.  相似文献

Nitrate reductase of spinach (Spinacia oleracea L.) leaves which had been inactivated in vitro by treatment with NADH and cyanide, was reactivated by incubation with oxidant systems and measured as FMNH2-dependent activity. Reactivation was produced with trivalent manganese compounds represented either by manganipyrophosphate or produced by oxidation of Mn2+ ions in the presence of illuminated chloroplasts and compared with reactivation obtained with ferricyanide. Reactivation in the chloroplast system was equivalent to that with ferricyanide when orthophosphate was used but was variable and weak in the presence of pyrophosphate, although manganipyrophosphate was formed, freely. Reactivation by manganipyrophosphate in dark reaction conditions was less effective than with ferricyanide but was not inhibited by the addition of pyrophosphate. Reactivation with illuminated unheated chloroplasts was dependent on added manganese and oxidation of manganese in the presence of pyrophosphate was abolished by boiling the chloroplasts. In the presence of orthophosphate however, boiled, illuminated chloroplasts reactivated the enzyme in the absence of added manganese. Reactivation occurred spontaneously in air, more slowly than with the other oxidants, but to a similar extent to that produced by manganipyrophosphate. The results provide a possible model for physiological reactivation mechanisms.  相似文献

Amblyopia is a developmental abnormality that results in deficits for a wide range of visual tasks, most notably, the reduced ability to see fine details, the loss in contrast sensitivity especially for small objects and the difficulty in seeing objects in clutter (crowding). The primary goal of this study was to evaluate whether crowding can be ameliorated in adults with amblyopia through perceptual learning using a flanked letter identification task that was designed to reduce crowding, and if so, whether the improvements transfer to untrained visual functions: visual acuity, contrast sensitivity and the size of visual span (the amount of information obtained in one fixation). To evaluate whether the improvements following this training task were specific to training with flankers, we also trained another group of adult observers with amblyopia using a single letter identification task that was designed to improve letter contrast sensitivity, not crowding. Following 10,000 trials of training, both groups of observers showed improvements in the respective training task. The improvements generalized to improved visual acuity, letter contrast sensitivity, size of the visual span, and reduced crowding. The magnitude of the improvement for each of these measurements was similar in the two training groups. Perceptual learning regimens aimed at reducing crowding or improving letter contrast sensitivity are both effective in improving visual acuity, contrast sensitivity for near-acuity objects and reducing the crowding effect, and could be useful as a clinical treatment for amblyopia.  相似文献

Treatment of both uninfested and armyworm‐infested maize plants with jasmonic acid (JA) is known to attract the parasitic wasp, Cotesia kariyai Watanabe (Hymenoptera: Braconidae). Here, we show that treatment with a methyl ester of a JA precursor, methyl linolenate (MeLin), also causes maize plants to attract this wasp, yet does not cause elevated levels of endogenous JA. The volatile chemicals emitted from either infested or uninfested maize plants treated with MeLin were qualitatively and quantitatively different from those emitted from JA‐treated plants. Among compounds emitted from MeLin‐treated plants, α‐pinene and menthol attracted wasps in pure form in a two‐choice test using a choice chamber. A mixture of methyl salicylate, α‐copaene, and β‐myrcene also attracted wasps. In contrast, (Z)‐3‐hexenyl acetate was among the main attractants for C. kariyai in JA‐treated plants. These data show that in addition to JA, MeLin also has the potential to increase the host‐finding ability of C. kariyai, but that the composition of attractants they induce differs.  相似文献

Following a study of oxidative tryptophan metabolism to kynurenines, we have now analysed the blood of patients with either Huntington's disease or traumatic brain injury for levels of 5-hydroxytryptamine (5-HT), 5-hydroxyindoleacetic acid (5-HIAA) and melatonin. There were no differences in the baseline levels of these compounds between patients and healthy controls. Tryptophan depletion did not reduce 5-HT levels in either the controls or in the patients with Huntington's disease, but it increased 5-HT levels in patients with brain injury and lowered 5-HIAA in the control and Huntington's disease groups. An oral tryptophan load did not modify 5-HT levels in the patients but increased 5-HT in control subjects. The tryptophan load restored 5-HIAA to baseline levels in controls and patients with brain injury, but not in those with Huntington's disease, in whom 5-HIAA remained significantly depressed. Melatonin levels increased on tryptophan loading in all subjects, with levels in patients with brain injury increasing significantly more than in controls. Baseline levels of neopterin and lipid peroxidation products were higher in patients than in controls. It is concluded that both groups of patients exhibit abnormalities in tryptophan metabolism, which may be related to increased inflammatory status and oxidative stress. Interactions between the kynurenine, 5-HT and melatonin pathways should be considered when interpreting changes of tryptophan metabolism.  相似文献

Flow-through aquifer columns were used to investigate the feasibility of adding sulfate, EDTA–Fe(III) or nitrate to enhance the biodegradation of BTEX and ethanol mixtures. The rapid biodegradation of ethanol near the inlet depleted the influent dissolved oxygen (8 mg l-1), stimulated methanogenesis, and decreased BTEX biodegradation efficiencies from >99% in the absence of ethanol to an average of 32% for benzene, 49% for toluene, 77% for ethylbenzene, and about 30% for xylenes. The addition of sulfate, EDTA–Fe(III) or nitrate suppressed methanogenesis and significantly increased BTEX biodegradation efficiencies. Nevertheless, occasional clogging was experienced by the column augmented with EDTA–Fe(III) due to iron precipitation. Enhanced benzene biodegradation (>70% in all biostimulated columns) is noteworthy because benzene is often recalcitrant under anaerobic conditions. Influent dissolved oxygen apparently played a critical role because no significant benzene biotransformation was observed after oxygen was purged out of the influent media. The addition of anaerobic electron acceptors could enhance BTEX biodegradation not only by facilitating their anaerobic biodegradation but also by accelerating the mineralization of ethanol or other substrates that are labile under anaerobic conditions. This would alleviate the biochemical oxygen demand (BOD) and increase the likelihood that entraining oxygen would be used for the biotransformation of residual BTEX.  相似文献
