Modulation of gene network activity allows cells to respond to changes in environmental conditions. For example, the galactose utilization network in Saccharomyces cerevisiae is activated by the presence of galactose but repressed by glucose. If both sugars are present, the yeast will first metabolize glucose, depleting it from the extracellular environment. Upon depletion of glucose, the genes encoding galactose metabolic proteins will activate. Here, we show that the rate at which glucose levels are depleted determines the timing and variability of galactose gene activation. Paradoxically, we find that Gal1p, an enzyme needed for galactose metabolism, accumulates more quickly if glucose is depleted slowly rather than taken away quickly. Furthermore, the variability of induction times in individual cells depends non-monotonically on the rate of glucose depletion and exhibits a minimum at intermediate depletion rates. Our mathematical modeling suggests that the dynamics of the metabolic transition from glucose to galactose are responsible for the variability in galactose gene activation. These findings demonstrate that environmental dynamics can determine the phenotypic outcome at both the single-cell and population levels.  相似文献

The R-R intervals were measured in 25 female volunteers aged 21–34 years after exposure to lower body negative pressure (LBNP). A number of advantages of information entropy (h) as an index of cardiovascular performance were revealed. With h used as a controlled variable, it was possible to ensure individual targeted changes in the functional state (FS) during LBNP. It was found that an individual exposure to LBNP caused regular changes in h and FS. As heart-rate variability (HRV) is under evaluation in clinical applications, the authors sought to better define the interdependent impact of age, maximal exercise, and diurnal variation under physiologic conditions. The authors evaluated the diurnal changes in HRV 24-h pre- and post-maximal aerobic exercise testing to exhaustion in young (19–25 yrs, n?=?12) and middle-aged (40–55 yrs, n?=?12) adults. Subjects wore a portable 5-lead electrocardiogram holter for 48?h (24?h prior to and following a maximal aerobic capacity test). Time-, frequency-, time-frequency-, and scale-invariant-domain measures of HRV were computed from RR-interval data analyzed using a 5-min window size and a 2.5-min step size, resulting in a different set of outputs every 2.5?min. Results were averaged (mean?±?SE) over four prespecified time periods during the morning, afternoon, evening, and night on Day 1 and Day 2. Diurnal changes in HRV in young and middle-aged adults were compared using a two-way, repeated-measures analysis of variance (ANOVA). Young adults demonstrated higher HRV compared to middle-aged adults during periods of wakefulness and sleep prior to maximal exercise stress testing (i.e., high-frequency power during Day 1: young adults: morning 1862?±?496?ms2, afternoon 1797?±?384?ms2, evening 1908?±?431?ms2, and night 3202?±?728?ms2; middle-aged adults: morning 341?±?53?ms2, afternoon 405?±?68?ms2, evening 469?±?80?ms2, and night 836?±?136?ms2) (p < .05). Exercise resulted in reductions in HRV such that multiple measures of HRV were not significantly different between age groups during the afternoon and evening periods. All measures of HRV demonstrated between-group differences overnight on Day 2 (p < .05). Young adults are associated with higher baseline HRV during the daytime. Sleep increases variability equally and proportionally to daytime variability. Given the higher baseline awake HRV and equal rise in HRV during sleep, the change in HRV from sleep to morning with exercise is greater in younger subjects. 上个世纪60年代,Kimura提出的“中性进化”假说使经典的达尔文自然选择学说遭遇了前所未有的挑战。但新近的研究表明:在DNA水平,越来越多的证据支持“自然选择”的进化理论。这些研究成果得益于近年来大量群体和基因组DNA数据的积累,以及理论群体遗传学的发展。在DNA水平检测选择作用是否存在的方法包括两大类:种内多态性检验和种间差异度检验。前者以Tajima(1989)提出的D检验为代表,后者大都基于“中性条件下,种内与种间进化速率一致”的原理。这些方法以中性假说作为零假设,结合统计检验方法分析DNA数据,被称为“中性检验”。这些方法对于解决一些有关进化的基础理论问题和人类遗传学及生物信息学的深入研究都具有重要意义。本文介绍几个应用广泛的检测方法,以使国内的读者了解它们的基本思路和操作方法。  相似文献

A comparative analysis of five teleostean genomes, namely zebrafish, medaka, three-spine stickleback, fugu and pufferfish was performed with the aim to highlight the nature of the forces driving both length and base composition of introns (i.e., bpi and GCi). An inter-genome approach using orthologous intronic sequences was carried out, analyzing independently both variables in pairwise comparisons. An average length shortening of introns was observed at increasing average GCi values. The result was not affected by masking transposable and repetitive elements harbored in the intronic sequences. The routine metabolic rate (mass specific temperature-corrected using the Boltzmann''s factor) was measured for each species. A significant correlation held between average differences of metabolic rate, length and GC content, while environmental temperature of fish habitat was not correlated with bpi and GCi. Analyzing the concomitant effect of both variables, i.e., bpi and GCi, at increasing genomic GC content, a decrease of bpi and an increase of GCi was observed for the significant majority of the intronic sequences (from ∼40% to ∼90%, in each pairwise comparison). The opposite event, concomitant increase of bpi and decrease of GCi, was counter selected (from <1% to ∼10%, in each pairwise comparison). The results further support the hypothesis that the metabolic rate plays a key role in shaping genome architecture and evolution of vertebrate genomes.  相似文献

Coping with pressure and anxiety is an ineluctable demand of sports performance. Heart rate variability (HRV) Biofeedback (BFB) shall be used as a tool for self regulating physiological responses resulting in improved psycho physiological interactions. For further analysis, the present study has been designed to examine the relationship between anxiety and performance and also effectiveness of biofeedback protocol to create stress-eliciting situation in basketball players. Thirty basketball players of university level and above (both male and female) aged 18–28 years, who scored a minimum of 20 in state trait anxiety inventory, were randomly divided into three equal groups- Experimental (Biofeedback) group, Placebo group and Control (No Treatment) group. The BFB group received HRV BFB training for 10 consecutive days for 20 min that included breathing at individual’s resonant frequency through a pacing stimulus; Placebo group was shown motivational video clips for 10 consecutive days for 10 min, whereas No Treatment Control group was not given any intervention. Two way repeated measure ANOVA was applied to analyze the differences within and between the groups. Anxiety, coping self-efficacy, heart rate variability, respiration rate, and performance (dribbling, passing and shooting) at session 1, 10 and 1 month follow up were statistically significant in each group along with interaction of group and time (p < 0.001). Also, all the measures showed statistically significant inter group difference (p < 0.05). The findings are harmonious with existing data on HRV BFB as a strategy for dealing with anxiety. The Placebo group showed improvement in self efficacy and performance post training. The Control group showed no change in any variable except performance. The results of the study support the idea that HRV BFB lowers the anxiety and thus there seems to be a potential association between HRV BFB and performance optimization.  相似文献

目的:了解心率变异性(heart rate variability,HRV)在不稳定型心绞痛(unstable angina pectoris,UAP)发病中的作用及其用于病情严重程度判断的价值;了解美托洛尔对UAP患者HRV的影响。方法:选择年龄、性别相匹配的UAP患者、SAP患者和健康人各60例做动态心电图检查,经Holter软件分析进行HRV时域指标的比较。UAP组动态心电图检查后加用美托洛尔25-50 mg/d(分两次口服),20天后重复动态心电图检查。结果:1UAP组与正常组比较各项指标均降低(P0.01)。2正常人HRV具有昼夜节律变化的特点,UAP组HRV昼夜节律变化性降低。3UAP病人服用美托洛尔后,心肌缺血、心律失常得到改善。结论:1.UAP患者心脏自主神经活性主要是副交感神经活性受损,交感神经活性相对占优势。2.无合并症的UAP病人HRV呈昼夜节律性降低。3.美托洛尔可改善UAP患者HRV。  相似文献

A method is presented for the study of the entrance of in vitro stimulated cells into their first poststimulation S phase. PHA-stimulated lymphocytes were incubated continuously with 14C-TdR. This isotope was then removed at different intervals and the cells were incubated for 8 h in medium containing 3H-TdR. Cells which had incorporated 3H-TdR but not 14C-TdR were considered to have entered their first post-stimulation S phase during the 8 h incubation with 3H-TdR. These cells were identified by double-layer autoradiography.The majority of PHA-stimulated lymphocytes entered their first period of DNA synthesis between 48 and 72 h after the addition of PHA. However, the variability was pronounced, some cells entering their first S phase at about 24 h and others some 100 h later. Cells entering their first S phase accounted for a considerable part of the population of cells in DNA synthesis still as late as 72 h after the addition of PHA.Calculation of the total number of cells that entered their first S phase during the 6 day culture period showed that DNA synthesis was initiated in some 40 % of the cells of the initial population.  相似文献

The phylogeny of the grey mullets is considered problematic both at the intra- and interfamily level. Such a difficulty arises from the highly homogeneous morphology displayed by this group of fish and, consequently, from the paucity of the key morphological characters suitable to address their phylogeny and evolution. In the present work, we have approached the phylogenetic and evolutionary relationships of seven species of Mugilidae, six of which from the Mediterranean Sea, on the basis of the DNA sequences of two mitochondrial genes (cytochromeband 12S rRNA). Despite the morphological homogeneity exhibited by the taxa considered, the two species of the genusMugil(M. cephalusandM. curema) showed a remarkable genetic divergence compared to all the other members of the family. The relative rate test revealed a significantly higher rate of evolution along theMugillineage.  相似文献

Slow breathing is widely applied to improve symptoms of hyperarousal, but it is unknown whether its beneficial effects relate to the reduction in respiration rate per se, or, to a lower inhalation/exhalation (i/e) ratio. The present study examined the effects of four ventilatory patterns on heart rate variability and self-reported dimensions of relaxation. Thirty participants were instructed to breathe at 6 or 12 breaths/min, and with an i/e ratio of 0.42 or 2.33. Participants reported increased relaxation, stress reduction, mindfulness and positive energy when breathing with the low compared to the high i/e ratio. A lower compared to a higher respiration rate was associated only with an increased score on positive energy. A low i/e ratio was also associated with more power in the high frequency component of heart rate variability, but only for the slow breathing pattern. Our results show that i/e ratio is an important modulator for the autonomic and subjective effects of instructed ventilatory patterns.  相似文献

A method of analysis of heart rate variability based on the graph theory principle was suggested. The main parameters of the heart rate graph structure were determined and analyzed using models of harmonic oscillations, white noise, and various functional tests (including controllable respiration and mental load). The efficiency of the use of parameters of the heart rate graph for diagnosing some functional states was considered. A correlation of the parameters of the heart rate graph structure with the frequency characteristics of heart rate variability was studied. A general model of changes in the heart rate graph structure parameters at different levels of mental activity was constructed in terms of entropy changes.  相似文献

Aim was to elucidate autonomic responses to dynamic and static (isometric) exercise of the lower limbs eliciting the same moderate heart rate (HR) response. Method: 23 males performed two kinds of voluntary exercise in a supine position at similar heart rates: static exercise (SE) of the lower limbs (static leg press) and dynamic exercise (DE) of the lower limbs (cycling). Subjective effort, systolic (SBP) and diastolic blood pressure (DBP), mean arterial pressure (MAP), rate pressure product (RPP) and the time between consecutive heart beats (RR-intervals) were measured. Time-domain (SDNN, RMSSD), frequency-domain (power in the low and high frequency band (LFP, HFP)) and geometric measures (SD1, SD2) as well as non-linear measures of regularity (approximate entropy (ApEn), sample entropy (SampEn) and correlation dimension D2) were calculated. Results: Although HR was similar during both exercise conditions (88±10 bpm), subjective effort, SBP, DBP, MAP and RPP were significantly enhanced during SE. HRV indicators representing overall variability (SDNN, SD 2) and vagal modulated variability (RMSSD, HFP, SD 1) were increased. LFP, thought to be modulated by both autonomic branches, tended to be higher during SE. ApEn and SampEn were decreased whereas D2 was enhanced during SE. It can be concluded that autonomic control processes during SE and DE were qualitatively different despite similar heart rate levels. The differences were reflected by blood pressure and HRV indices. HRV-measures indicated a stronger vagal cardiac activity during SE, while blood pressure response indicated a stronger sympathetic efferent activity to the vessels. The elevated vagal cardiac activity during SE might be a response mechanism, compensating a possible co-activation of sympathetic cardiac efferents, as HR and LF/HF was similar and LFP tended to be higher. However, this conclusion must be drawn cautiously as there is no HRV-marker reflecting “pure” sympathetic cardiac activity.  相似文献

Introns of four X-linked genes (Hprt, Plp, Glra2, and Amg) were sequenced to provide an estimate of nucleotide diversity at nuclear genes within the house mouse and to test the neutral prediction that the ratio of intraspecific polymorphism to interspecific divergence is the same for different loci. Hprt and Plp lie in a region of the X chromosome that experiences relatively low recombination rates, while Glra2 and Amg lie near the telomere of the X chromosome, a region that experiences higher recombination rates. A total of 6022 bases were sequenced in each of 10 Mus domesticus and one M. caroli. Average nucleotide diversity (π) for introns within M. domesticus was quite low (π = 0.078%). However, there was substantial variation in the level of heterozygosity among loci. The two telomeric loci, Glra2 and Amg, had higher ratios of polymorphism to divergence than the two loci experiencing lower recombination rates. These results are consistent with the hypothesis that heterozygosity is reduced in regions with lower rates of recombination, although sampling of additional genes is needed to establish whether there is a general correlation between heterozygosity and recombination rate as in Drosophila melanogaster.  相似文献

SYNOPSIS. Models of energy budgets often allocate energy betweenthe broad categories of maintenance, growth, and reproduction.An important component of maintenance costs is standard metabolicrate, which is partially determined by body temperature. Therelationship between thermoregulation, standard metabolism,and egg production was investigated in adult females of theacridid grasshopper Xanthippus corallipes from six populationsalong an elevation gradient. Individuals thermoregulate andmaintain stable afternoon body temperatures which correlatenegatively with elevation. Standard metabolic rates were calculatedat each population's afternoon body temperature. These rateswere then used to investigate the effect of variation in standardmetabolic rate on egg production. Because of the pervasive effectof mass on standard metabolic rate and egg number, the variableswere log-transformed. The effect of mass was removed by regressionanalyses, and residuals were calculated. An analysis of theresiduals revealed that intermediate values of field standardmetabolic rate correlate with high egg production, suggestingstabilizing selection on standard metabolic rate.  相似文献

The paper presents allele frequencies at 15 STR loci (D3S1358, vWA, FGA, TH01, TPOX, CSFIPO, D5S818, D13S317, D7S820, D16S539, D2Sl338, D8S1179, D21S1l, D18S51, D19S433), used in forensic medicine, in Russian sample (n = 176) representing population of the European part of the Russian Federation. The combined power of discrimination (PD) and the combined power of exclusion (PE) for the 15 STR loci were 0.999 999 999 999 999 986 and 0.999 999 331 310 171 000, respectively. The data obtained for allele and genotype frequencies conformed to Hardy-Weinberg expectations. According to the presented data, loci D2S1338, D18S51, D21Sll and FGA are the most informative markers for Russians. The data obtained may be used as reference database for forensic medicine laboratories in Russian Federation.  相似文献

Influence of light and dark aerobic conditions and of various metabolic poisons [3-(3,4-dichlorophenyl)1,1-dimethylurea (DCMU), 2,4-dinitrophenol (2,4-DNP), carbonyl cyanide-m-chlorophenylhydrazone (m-CCCP), α,α′bis(hexafluoroacetonyl) acetone (1799), N,N′-dicyclohexylcarbodiimide (DCCD), sodium azide, fatty acids] on the active phosphate uptake and on the ATP level has been investigated. Cells in the dark have the same level of ATP as in the light though the phosphate transport is decreased. Similar findings are observed with DCMU. Other poisons used inhibit phosphate uptake and decrease ATP level. However their effect is more drastic on phosphate transport than on the ATP level. There is no good correlation between the ATP level and phosphate transport.  相似文献

Testing the Metabolic Scaling Theory of tree growth   总被引:1,自引:0,他引:1  
1.  Metabolic Scaling Theory (MST) predicts a 'universal scaling law' of tree growth. Proponents claim that MST has strong empirical support: the size-dependent growth curves of 40 out of 45 species in a Costa Rican forest have scaling exponents indistinguishable from the MST prediction.
2.  Here, we show that the Costa Rican study has been misinterpreted. Using Standardized Major Axis (SMA) line-fitting to estimate scaling exponents, we find that four out of five species represented by more than 100 stems have scaling exponents that deviate significantly from the MST prediction. On the other hand, sample sizes were too small to make strong inferences in the cases of 33 species represented by fewer than 50 stems.
3.  Recently, it has been argued that MST is useful for predicting average scaling exponents, even if individual species do not conform to the theory. We find that the mean scaling exponent of the Costa Rican trees is greater than predicted (across-species mean  =  0.44), and hypothesize that scaling exponents in natural forests will generally be greater than predicted, because the theory fails to model asymmetric competition for light.
4.   Synthesis . We highlight shortcomings in the interpretation of data used in support of a key MST prediction. We recommend that future research into biological scaling should compare the merits of alternative models rather than focusing attention on tests of a single theory.  相似文献

综述了小型哺乳动物持续能量收支限制的研究概况和进展。最大持续能量收支在决定物种的地理分布、生存适应、繁殖成功等方面都具有重要意义,但在许多条件下受到限制。食物的丰富度,或者动物自身的摄食、消化和吸收能力似乎不是主要限制因素。持续能量收支可能被外周组织和器官消耗能量的能力限制,即"外周限制"假说;或者机体的散热能力所限制,即"热耗散限制"假说。动物也可能通过衡量季节性繁殖投资的价值,实现最大繁殖输出,即"季节性投资"假说。尽管这些假说得到了一些研究的证实,但仍未阐明持续能量收支限制的机理。本文对相关研究的发展方向进行了展望。  相似文献
