Disease epidemics have caused extensive damage to tropical coral reefs and to the reef-building corals themselves, yet nothing is known about the abilities of the coral host to resist disease infection. Understanding the potential for natural disease resistance in corals is critically important, especially in the Caribbean where the two ecologically dominant shallow-water corals, Acropora cervicornis and A. palmata, have suffered an unprecedented mass die-off due to White Band Disease (WBD), and are now listed as threatened under the US Threatened Species Act and as critically endangered under the IUCN Red List criteria. Here we examine the potential for natural resistance to WBD in the staghorn coral Acropora cervicornis by combining microsatellite genotype information with in situ transmission assays and field monitoring of WBD on tagged genotypes. We show that six percent of staghorn coral genotypes (3 out of 49) are resistant to WBD. This natural resistance to WBD in staghorn corals represents the first evidence of host disease resistance in scleractinian corals and demonstrates that staghorn corals have an innate ability to resist WBD infection. These resistant staghorn coral genotypes may explain why pockets of Acropora have been able to survive the WBD epidemic. Understanding disease resistance in these corals may be the critical link to restoring populations of these once dominant corals throughout their range.

Winter conditions in aquatic habitats of the temperate zone markedly differ from those present in warmer seasons, nevertheless, relatively scarce information is available on planktonic microbial composition, as sites are not easily accessible and it was supposed traditionally that microbial activity is low during this cold period. Since microorganisms could have great impact on the ecosystem even during winter, we explored various sites in the Eastern Carpathians regarding the abundance and taxonomic composition of planktonic microorganisms. Although many of the studied environments were extreme habitats, planktonic microbial communities were abundant and mostly diverse with the presence of previously unidentified taxa.  相似文献

Coastal dunes worldwide harbor threatened habitats characterized by high diversity in terms of plant communities. In Italy, recent assessments have highlighted the insufficient state of conservation of these habitats as defined by the EU Habitats Directive. The effects of predicted climate change could have dramatic consequences for coastal environments in the near future. An assessment of the efficacy of protection measures under climate change is thus a priority. Here, we have developed environmental envelope models for the most widespread dune habitats in Italy, following two complementary approaches: an “indirect” plant-species-based one and a simple “direct” one. We analyzed how habitats distribution will be altered under the effects of two climate change scenarios and evaluated if the current Italian network of protected areas will be effective in the future after distribution shifts. While modeling dune habitats with the “direct” approach was unsatisfactory, “indirect” models had a good predictive performance, highlighting the importance of using species’ responses to climate change for modeling these habitats. The results showed that habitats closer to the sea may even increase their geographical distribution in the near future. The transition dune habitat is projected to remain stable, although mobile and fixed dune habitats are projected to lose most of their actual geographical distribution, the latter being more sensitive to climate change effects. Gap analysis highlighted that the habitats’ distribution is currently adequately covered by protected areas, achieving the conservation target. However, according to predictions, protection level for mobile and fixed dune habitats is predicted to drop drastically under the climate change scenarios which we examined. Our results provide useful insights for setting management priorities and better addressing conservation efforts to preserve these threatened habitats in future.  相似文献

Tropical reefs are dynamic ecosystems that host diverse coral assemblages with different life-history strategies. Here, we quantified how juvenile (<50 mm) coral demographics influenced benthic coral structure in reef flat and reef slope habitats on the southern Great Barrier Reef, Australia. Permanent plots and settlement tiles were monitored every six months for three years in each habitat. These environments exhibited profound differences: the reef slope was characterised by 95% less macroalgal cover, and twice the amount of available settlement substrata and rates of coral settlement than the reef flat. Consequently, post-settlement coral survival in the reef slope was substantially higher than that of the reef flat, and resulted in a rapid increase in coral cover from 7 to 31% in 2.5 years. In contrast, coral cover on the reef flat remained low (~10%), whereas macroalgal cover increased from 23 to 45%. A positive stock-recruitment relationship was found in brooding corals in both habitats; however, brooding corals were not directly responsible for the observed changes in coral cover. Rather, the rapid increase on the reef slope resulted from high abundances of broadcast spawning Acropora recruits. Incorporating our results into transition matrix models demonstrated that most corals escape mortality once they exceed 50 mm, but for smaller corals mortality in brooders was double those of spawners (i.e. acroporids and massive corals). For corals on the reef flat, sensitivity analysis demonstrated that growth and mortality of larger juveniles (21–50 mm) highly influenced population dynamics; whereas the recruitment, growth and mortality of smaller corals (<20 mm) had the highest influence on reef slope population dynamics. Our results provide insight into the population dynamics and recovery trajectories in disparate reef habitats, and highlight the importance of acroporid recruitment in driving rapid increases in coral cover following large-scale perturbation in reef slope environments.  相似文献

Recurring seasonal patterns of microbial distribution and abundance in three third-order temperate streams within the southeast Pennsylvania Piedmont were observed over 4 years. Populations associated with streambed sediments and rocks (epilithon) were identified using terminal restriction length polymorphism (tRFLP) and sequencing of 16S rRNA genes selectively amplified with primers for the bacterial domain. Analyses of the relative magnitudes of tRFLP peak areas by using nonmetric multidimensional scaling resolved clear seasonal trends in epilithic and sediment populations. Oscillations between two dominant groups of epilithic genotypes, explaining 86% of the seasonal variation in the data set, were correlated with temperature and dissolved organic carbon. Sequences affiliated with epilithic phototrophs (cyanobacteria and diatom chloroplasts), a Rhodoferax sp., and a Bacillus species clustered in the summer, whereas sequences most closely related to “Betaproteobacteria” (putative Burkholderia sp.), and a putative cyanobacterium clustered in the fall/spring. The sediment genotypes also clustered into two groups, and these explained 85% of seasonal variation but correlated only with temperature. A summer tRFLP pattern was characterized by prevalence of “Betaproteobacteria,” “Gammaproteobacteria,” and a Bacillus sp., whereas the winter/spring pattern was characterized by phylotypes most closely related to “Firmicutes,” “Gammaproteobacteria,” and “Nitrospirae.” A close association between these headwater streams and their watersheds was suggested by the recovery of sequences related to microbial populations provisionally attributed to not only freshwaters but also terrestrial habitats.  相似文献

Cryptoendolithic habitats in the Canadian high Arctic are associated with a variety of microbial community assemblages, including cyanobacteria, algae, and fungi. These habitats were analyzed for the presence of metal ions by sequential extraction and evaluated for relationships between these and the various microorganisms found at each site using multivariate statistical methods. Cyanobacteria-dominated communities exist under higher pH conditions with elevated concentrations of calcium and magnesium, whereas communities dominated by fungi and algae are characterized by lower pH conditions and higher concentrations of iron, aluminum, and silicon in the overlying surfaces. These results suggest that the activity of the dominant microorganisms controls the pH of the surrounding environment, which in turn dictates rates of weathering or the possibility for surface crust formation, both ultimately deciding the structure of microbial diversity for each cryptoendolithic habitat.  相似文献

Black band disease (BBD) is a pathogenic consortium of microorganisms that primarily affects massive framework-building scleractinian corals on reefs worldwide. There has been considerable debate concerning the microbial community composition of BBD. The aim of this study was to utilize microbial profiling to assess overall patterns of variation in the BBD bacterial community with respect to geographic location, host coral species, time, and nutrient regime. Length heterogeneity polymerase chain reaction (LH-PCR) was employed to differentiate BBD communities based on the natural variation in the sequence lengths within hypervariable domains of the 16S rRNA gene. Analysis of LH-PCR profiles of 97 BBD samples using multivariate ordination methods and analysis of similarity revealed significant clustering with respect to geographic region when comparing BBD sampled from reefs near Lee Stocking Island in the Bahamas’ Exuma Chain, the Northern Florida Keys (NFK), and St. John in the US Virgin Islands. There was much variability in BBD community composition on a regional basis, between sites in the NFK, and in terms of coral host species. The observed differences among BBD microbial community profiles were driven primarily by variation in relative abundance of 313–316-bp amplicons, which correspond to cyanobacteria and α-proteobacteria. The results obtained in this study support previous reports of intrinsic variability and complexity of the BBD microbial community but also suggest that this variability has biogeographic patterns.  相似文献

Mucus, i.e., particulate and dissolved organic matter (POM, DOM) released by corals, acts as an important energy carrier in tropical ecosystems, but little is known on its ecological role in temperate environments. This study assessed POM and DOM production by the temperate coral Cladocora caespitosa under different environmental conditions. The subsequent enzymatic degradation, growth of prokaryotes and virus-like particles (VLPs) as well as changes in the structure of the prokaryotic communities were also monitored. C. caespitosa produced an important quantity of mucus, which varied according to the environmental conditions (from 37.8 to 67.75 nmol carbon h-1 cm-2), but remained higher or comparable to productions observed in tropical corals. It has an important nutritional value, as highlighted by the high content in dissolved nitrogen (50% to 90% of the organic matter released). Organic matter was rapidly degraded by prokaryotes’ enzymatic activities, and due to its nitrogen content, aminopeptidase activity was 500 fold higher than the α-glucosidase activity. Prokaryotes, as well as VLPs, presented a rapid growth in the mucus, with prokaryote production rates as high as 0.31 μg h-1 L-1. Changes in bacterial and archaeal communities were observed in the ageing mucus and between mucus and the water column, suggesting a clear impact of mucus on microorganism diversity. Overall, our results show that the organic matter released by temperate corals, such as C. caespitosa, which can form reef structures in the Mediterranean Sea, stimulates microbial activity and thereby functions as a significant carbon and nitrogen supplier to the microbial loop.  相似文献

Alkaloids, which are generally basic N‐containing compounds, have been found in a variety of natural sources. Recently, the interest in alkaloids from corals increased significantly due to their remarkable bioactivities. This review deals with the chemical structures and biological activities of alkaloids in corals. The literature has been covered up to June 2011, and a total of 102 alkaloids from the 51 publications are discussed and reviewed. Some of these compounds showed various biological properties, such as cytotoxic, antibacterial, insecticidal, antifouling, and other activities.  相似文献

Increasing loss of biodiversity in agricultural landscapes is often debated in the bioenergy context, especially with respect to non-traditional crops that can be grown for energy production in the future. As promising renewable energy source and additional landscape element, the potential role of short rotation coppice (SRC) plantations to biodiversity is of great interest. We studied plant species richness in eight landscapes (225 km2) containing willow and poplar SRC plantations (1,600 m2) in Sweden and Germany, and the related SRC α-diversity to species richness in the landscapes (γ-diversity). Using matrix variables, spatial analyses of SRC plantations and landscapes were performed to explain the contribution of SRC α-diversity to γ-diversity. In accordance with the mosaic concept, multiple regression analyses revealed number of habitat types as a significant predictor for species richness: the higher the habitat type number, the higher the γ-diversity and the lower the proportion of SRC plantation α-diversity to γ-diversity. SRC plantation α-diversity was 6.9 % (±1.7 % SD) of species richness on the landscape scale. The contribution of SRC plantations increased with decreasing γ-diversity. SRC plantations were dominated more by species adapted to frequent disturbances and anthropo-zoogenic impacts than surrounding landscapes. We conclude that by providing habitats for plants with different requirements, SRC α-diversity has a significant share on γ-diversity in rural areas and can promote diversity in landscapes with low habitat heterogeneity and low species pools. However, plant diversity enrichment is mainly due to additional species typically present in disturbed and anthropogenic environments.  相似文献

To further explore possible avenues for accessing microbial biodiversity for drug discovery from natural products, we constructed and screened a 5,000-clone “shotgun” environmental DNA library by using an Escherichia coli-Streptomyces lividans shuttle cosmid vector and DNA inserts from microbes derived directly (without cultivation) from soil. The library was analyzed by several means to assess diversity, genetic content, and expression of heterologous genes in both expression hosts. We found that the phylogenetic content of the DNA library was extremely diverse, representing mostly microorganisms that have not been described previously. The library was screened by PCR for sequences similar to parts of type I polyketide synthase genes and tested for the expression of new molecules by screening of live colonies and cell extracts. The results revealed new polyketide synthase genes in at least eight clones. In addition, at least five additional clones were confirmed by high-pressure liquid chromatography analysis and/or biological activity to produce heterologous molecules. These data reinforce the idea that exploiting previously unknown or uncultivated microorganisms for the discovery of novel natural products has potential value and, most importantly, suggest a strategy for developing this technology into a realistic and effective drug discovery tool.  相似文献

In this paper, I review data on the magnitude and extent of reef coral bleaching events and consider modern hypotheses on the mechanisms of this natural phenomenon and experimental data lying at their basis. Four possible mechanisms of color loss by hermatypic corals have been confirmed experimentally: bacterial infection, change of zooxanthellae type in the polyps to improve the heat resistance of the photosynthetic function of coral to elevated seawater temperature, intoxication of zooxanthellae by animal metabolic wastes at high temperature and light levels, and thermal and photodestruction of the animal and algal cells. The heating effect of photosynthetic active radiation on the zooxanthellar cells in coral polyps was verified theoretically. The calculations showed that in the natural environment, the additional light-induced heating of zooxanthellae is not above 0.01°C and that it cannot cause disruption of the animal and zooxanthellae symbiosis.  相似文献

Black band disease (BBD) of corals is a complex polymicrobial disease considered to be a threat to coral reef health, as it can lead to mortality of massive reef-building corals. The BBD community is dominated by gliding, filamentous cyanobacteria with a highly diverse population of heterotrophic bacteria. Microbial interactions such as quorum sensing (QS) and antimicrobial production may be involved in BBD disease pathogenesis. In this study, BBD (whole community) samples, as well as 199 bacterial isolates from BBD, the surface mucopolysaccharide layer (SML) of apparently healthy corals, and SML of apparently healthy areas of BBD-infected corals were screened for the production of acyl homoserine lactones (AHLs) and for autoinducer-2 (AI-2) activity using three bacterial reporter strains. AHLs were detected in all BBD (intact community) samples tested and in cultures of 5.5% of BBD bacterial isolates. Over half of a subset (153) of the isolates were positive for AI-2 activity. AHL-producing isolates were further analyzed using LC-MS/MS to determine AHL chemical structure and the concentration of (S)-4,5-dihydroxy-2,3-pentanedione (DPD), the biosynthetic precursor of AI-2. C6-HSL was the most common AHL variant detected, followed by 3OC4-HSL. In addition to QS assays, 342 growth challenges were conducted among a subset of the isolates, with 27% of isolates eliciting growth inhibition and 2% growth stimulation. 24% of BBD isolates elicited growth inhibition as compared to 26% and 32% of the bacteria from the two SML sources. With one exception, only isolates that exhibited AI-2 activity or produced DPD inhibited growth of test strains. These findings demonstrate for the first time that AHLs are present in an active coral disease. It is possible that AI-2 production among BBD and coral SML bacteria may structure the microbial communities of both a polymicrobial infection and the healthy coral microbiome.  相似文献
