Multiple mating by queens (polyandry) and the occurrence of multiple queens in the same colony (polygyny) alter patterns of relatedness within societies of eusocial insects. This is predicted to influence kin-selected conflicts over reproduction. We investigated the mating system of a facultatively polygynous UK population of the ant Leptothorax acervorum using up to six microsatellite loci. We estimated mating frequency by genotyping 79 dealate (colony) queens and the contents of their sperm receptacles and by detailed genetic analysis of 11 monogynous (single-queen) and nine polygynous colonies. Results indicated that 95% of queens were singly mated and 5% of queens were doubly mated. The corrected population mean mating frequency was 1.06. Parentage analysis of adults and brood in 17 colonies (10 monogynous, 7 polygynous) showed that female offspring attributable to each of 31 queens were full sisters, confirming that queens typically mate once. Inbreeding coefficients, queen-mate relatedness of zero and the low incidence of diploid males provided evidence that L. acervorum sexuals mate entirely or almost entirely at random. Males mated to queens in the same polygynous colony were not related to one another. Our data also confirmed that polygynous colonies contain queens that are related on average and that their workers had a mixed maternity. We conclude that the mating system of L. acervorum involves queens that mate near nests with unrelated males and then seek readoption by those nests, and queens that mate in mating aggregations away from nests, also with unrelated males.

Queen promiscuity lowers disease within honeybee colonies
Most species of social insects have singly mated queens, but in some species each queen mates with numerous males to create a colony with a genetically diverse worker force. The adaptive significance of polyandry by social insect queens remains an evolutionary puzzle. Using the honeybee (Apis mellifera), we tested the hypothesis that polyandry improves a colony's resistance to disease. We established colonies headed by queens that had been artificially inseminated by either one or 10 drones. Later, we inoculated these colonies with spores of Paenibacillus larvae, the bacterium that causes a highly virulent disease of honeybee larvae (American foulbrood). We found that, on average, colonies headed by multiple-drone inseminated queens had markedly lower disease intensity and higher colony strength at the end of the summer relative to colonies headed by single-drone inseminated queens. These findings support the hypothesis that polyandry by social insect queens is an adaptation to counter disease within their colonies.

Multiple functional queens in a colony (polygyny) and multiple mating by queens (polyandry) in social insects challenge kin selection, because they dilute inclusive fitness benefits from helping. Colonies of the ant Plagiolepis pygmaea brash contain several hundreds of multiply mated queens. Yet, within‐colony relatedness remains unexpectedly high. This stems from low male dispersal, extensive mating among relatives and adoption of young queens in the natal colony. We investigated whether inbreeding results from workers expelling foreign males, and/or from preferential mating between related partners. Our data show that workers actively repel unrelated males entering their colony, and that queens preferentially mate with related males. These results are consistent with inclusive fitness being a driving force for inbreeding: by preventing outbreeding, workers reduce erosion of relatedness within colonies due to polygyny and polyandry. That virgin queens mate preferentially with related males could result from a long history of inbreeding, which is expected to reduce depression in species with regular sibmating.

Kin selection theory predicts conflict between queens and workers in the social insect colony with respect to male production. This conflict arises from the haplodiploid system of sex determination in Hymenoptera that creates relatedness asymmetries in which workers are more closely related to the sons of other workers than to those of the queen. In annual hymenopteran societies that are headed by a single queen, the mating frequency of the queen is the only factor that affects the colony kin structure. Therefore, we examined the mating structure of queens and the parentage of males in a monogynous bumblebee, Bombus ignitus, using DNA microsatellites. In the seven colonies that were studied, B. ignitus queens mated once, thereby leading to the prediction of conflict between the queen and workers regarding male production. In each of the five queen-right colonies, the majority of the males (95%) were produced by the colony’s queen. In contrast, workers produced approximately 47% of all the males in two queenless colonies. These results suggest that male production in B. ignitus is a conflict between queen and workers.

Mating frequency of Vespa analis queens and the genetic relatedness of their workers was analyzed by DNA microsatellite genotyping. Of 20 colonies studied, 18 had a queen inseminated by a single male and two had queens each inseminated by two males. The estimated effective number of matings was 1.05 ± 0.037 (mean ± SE), with 75–85% of the offspring of the two multiply mated queens sired by a single male. The pedigree relatedness between nestmate workers averaged over the 20 colonies was estimated to be 0.74 ± 0.008, almost identical to the predicted value of 0.75 for colonies headed by a singly mated queen. Multiple matrilines; that is, the presence of workers not related to the current queens, were detected in six colonies, suggesting that queen replacement occurred via usurpation of the founding queens in these six colonies. These results demonstrate that the kin structure of V. analis is similar to that reported in other vespid species.

The Australian endemic ant Nothomyrmecia macrops is considered one of the most ‘primitive’ among living ants. We investigated the genetic structure of colonies to determine queen mating frequencies and nestmate relatedness. An average of 18.8 individuals from each of 32 colonies, and sperm extracted from 34 foraging queens, were genotyped using five highly variable microsatellite markers. Queens were typically singly (65%) or doubly mated (30%), but triple mating (5%) also occurred. The mean effective number of male mates for queens was 1.37. No relationship between colony size and queen mate number was found. Nestmate workers were related by b=0.61 ± 0.03, significantly above the threshold under Hamilton’s rule over which, all else being equal, altruistic behaviour persists, but queens and their mates were unrelated. In 25% of the colonies we detected a few workers that could not have been produced by the resident queen, although there was no evidence for worker reproduction. Polyandry is for the first time recorded in a species with very small mature colonies, which is inconsistent with the sperm‐limitation hypothesis for the mediation of polyandry levels. Facultative polyandry is therefore not confined to the highly advanced ant genera, but may have arisen at an early stage in ant social evolution.

Summary: Honey bee queens have been shown to mate with a high number of males, but the evolutionary advantage of this high degree of polyandry is still unclear. Mating data from a number of different Apis species and subspecies are needed to help explain polyandry in honey bees. Pupae of four colonies of Apis mellifera sicula from Sicily were genotyped on three polymorphic microsatellite loci. The genotypes of the queens and fathering drones from these colonies were deduced from the genotypes of the pupae. We found no evidence for polygyny, at least we can exclude more than one functional queen, even super-sister queens, if maternity contributions are equal. The four queens mated with at least 5 to 12 (mean: 9.3 - 3.0 SE) drones. We estimate the error in our determination of the mating frequency that is caused by limited genetic resolution of the marker loci to be less than 1 mating given that Hardy-Weinberg assumptions are satisfied. However, the drones the single queens mated with may be a non-random sample of the whole population, so that detection error may be more severe. The average pedigree relatedness among workers within the colonies was estimated to be 0.341. These results are within the range of those found in other A. mellifera subspecies and Apis species except A. dorsata. We speculate that mating frequency may be positively correlated with drone density.

Recent work has demonstrated considerable benefits of intracolonial genetic diversity for the productivity of honeybee colonies: single-patriline colonies have depressed foraging rates, smaller food stores and slower weight gain relative to multiple-patriline colonies. We explored whether differences in the use of foraging-related communication behaviour (waggle dances and shaking signals) underlie differences in foraging effort of genetically diverse and genetically uniform colonies. We created three pairs of colonies; each pair had one colony headed by a multiply mated queen (inseminated by 15 drones) and one colony headed by a singly mated queen. For each pair, we monitored the production of foraging-related signals over the course of 3 days. Foragers in genetically diverse colonies had substantially more information available to them about food resources than foragers in uniform colonies. On average, in genetically diverse colonies compared with genetically uniform colonies, 36% more waggle dances were identified daily, dancers performed 62% more waggle runs per dance, foragers reported food discoveries that were farther from the nest and 91% more shaking signals were exchanged among workers each morning prior to foraging. Extreme polyandry by honeybee queens enhances the production of worker-worker communication signals that facilitate the swift discovery and exploitation of food resources.

The North American seed-harvester ant Pogonomyrmex (Ephebomyrmex) pima displays a dimorphism that consists of winged (alate) and wingless (intermorph) queens; both types of queens are fully reproductive. Microsatellite allele frequencies and a mitochondrial phylogeny demonstrate (1) alate and intermorph queens represent an intraspecific wing polymorphism, and (2) an absence of assortative mating and inbreeding by males. Surveys at our field site in southcentral Arizona, USA, demonstrated that only one type of queen (intermorph or dealate) occurred in each colony, including those excavated during the season in which reproductive sexuals were present. Colony structure appeared to vary by queen type as most intermorph colonies contained multiple mated queens. Alternatively, dealate queen colonies rarely contained a mated queen. Our inability to find mated dealate queens in these colonies probably resulted from difficulty in excavating the entire colony and reproductive queen, especially given that these colonies were only excavated over one day. A morphometric analysis demonstrated that intermorph queens are intermediate in size to that of workers and alate queens, but that intermorph queens retain all of the specialized anatomical features of alate queens (except for wings). Some colonies had queens that foraged and performed nest maintenance activities, and these queens sometimes accounted for a significant portion of colony foraging trips. Dissections revealed that these queens were uninseminated; some of these queens produced males in the laboratory. In the present study on the bumblebee Bombus terrestris, we investigated the influence of inbreeding on queen fitness by comparing diapause survival and egg-laying success of queens mated with nestmate and non-nestmate males. We then compared the early stage of colonies with or without diploid males and analysed colony characteristics to identify a factor predictive of colony outcome. Diapause survival was no different between queens mated with nestmates and non-nestmates, but in the latter case, egg-laying success was significantly higher. Queens mated with nestmates gave rise to a percentage of diploid male colonies (52.6%) compatible with brother–sister coupling. We obtained 18.6% of colonies with diploid males even from queens mated with non-nestmates, indicating that the colonies of origin were in some way related or homozygous at the sex determination loci. There was no difference in the early growth stage between colonies with or without diploid males, except in the number of workers emerging in the first brood, which was significantly higher in the latter. Among diploid male colonies, the number of workers and the male/worker ratio in the first brood was highly variable and was not a good predictor of subsequent colony growth. Out of 49 colonies with diploid males that reached full development, only 11 had a sufficient size to assume that they could survive in the field or, in a commercial breeding, to be suitable for pollination purposes.
