The primary aim of our study was to determine the influence of taking chromium plus carnitine on insulin resistance, with a secondary objective of evaluating the influences on lipid profiles and weight loss in overweight subjects with polycystic ovary syndrome (PCOS). In a 12-week randomized, double-blind, placebo-controlled clinical trial, 54 overweight women were randomly assigned to receive either supplements (200 μg/day chromium picolinate plus 1000 mg/day carnitine) or placebo (27/each group). Chromium and carnitine co-supplementation decreased weight (− 3.6 ± 1.8 vs. − 1.0 ± 0.7 kg, P < 0.001), BMI (− 1.3 ± 0.7 vs. − 0.3 ± 0.3 kg/m2, P < 0.001), fasting plasma glucose (FPG) (− 5.1 ± 6.0 vs. − 1.1 ± 4.9 mg/dL, P = 0.01), insulin (− 2.0 ± 1.4 vs. − 0.2 ± 1.2 μIU/mL, P < 0.001), insulin resistance (− 0.5 ± 0.4 vs. − 0.04 ± 0.3, P < 0.001), triglycerides (− 18.0 ± 25.2 vs. + 5.5 ± 14.4 mg/dL, P < 0.001), total (− 17.0 ± 20.3 vs. + 3.6 ± 12.0 mg/dL, P < 0.001), and LDL cholesterol (− 13.3 ± 19.2 vs. + 1.4 ± 13.3 mg/dL, P = 0.002), and elevated insulin sensitivity (+ 0.007 ± 0.005 vs. + 0.002 ± 0.005, P < 0.001). In addition, co-supplementation upregulated peroxisome proliferator-activated receptor gamma (P = 0.02) and low-density lipoprotein receptor expression (P = 0.02). Overall, chromium and carnitine co-supplementation for 12 weeks to overweight women with PCOS had beneficial effects on body weight, glycemic control, lipid profiles except HDL cholesterol levels, and gene expression of PPAR-γ and LDLR. The effect of orally administered l-carnitine on biochemical parameters was examined in lactating Tuj-ewes. Ewes were orally given 500 mg of l-carnitine daily for 3 weeks. To evaluate the changes on selected blood indicators (total protein, albumin, glucose, triglyceride, cholesterol, urea, aspartate amino transferase, alanine amino transferase, lipase, triiodothyronine and thyroxine), blood samples were collected at the beginning of the study, and at the end of 1st, 2nd and 3rd week of study. Oral administration of supplemental carnitine significantly decreased serum triglyceride (P < 0.05), glucose (P < 0.05), cholesterol (P < 0.05) and triiodothyronine (P < 0.05) concentrations. In addition, serum thyroxine (P < 0.001) and albumin (P < 0.01) concentrations were significantly elevated as a result of oral carnitine treatment. These results suggest that supplemental l-carnitine improves selected biochemical indicators in Tuj-ewes.  相似文献

The effects of retinoic acid (RA) (50 μg/100 g body wt. per day) on hepativ heme oxygenase activty, δ-aminolevulinate synthase (ALAS) activity and on cytochrome P-450 content were determined in thyroidectomized rats treated with T3 (10 μg/100 g body wt. per day) or diluent. RA, when administered for 3 days, failed to influence significantly the activity of either heme oxygenase or ALAS, however, the retinoid depleted hepatic cytochrome P-450 content by 17% (P < 0.01) and microsomal heme content by 47% (P < 0.001). T3 administration enhanced heme oxygenase activity by 72% (P < 0.001) and ALAS activity by 251% (P < 0.001) above levels in diluent treated controls and depleted cytochrome P-450 levels by 55% (P < 0.001) and heme levels by 75% (P < 0.001). When RA and T3 were administered together, the retinoid markedly enhanced the T3 stimulation of heme oxygenase activity; 173% above controls (P < 0.001), and 61% above T3 alone (P < 0.001). However, RA failed to influence the effect of T3 on ALAS activity or cytochrome P-450 depletion. The results indicate that RA can influence the levels of hepatic cytochrome P-450 and can modulate the stimulation of heme oxygenase activity by thyroid hormone in vivo.  相似文献

Resistant starch (RS) has been suggested to prolong satiety in adult pigs. The present study investigated RS-induced changes in behaviour, satiety-related hormones and metabolites in catheterized growing pigs to explore possible underlying mechanisms for RS-induced satiety. In a cross-over design with two 14-day periods, 10 pigs (initial BW: 58 kg) were assigned to two treatments comprising diets containing either 35% pregelatinized starch (PS) or 34% retrograded starch (RS). Diets were isoenergetic on gross energy. Pigs were fed at 2.8× maintenance. Postprandial plasma response of satiety-related hormones and metabolites was measured at the end of each period using frequent blood sampling. Faecal and urinary energy losses were measured at the end of each period. Behaviour was scored 24 h from video recordings using scan sampling. Energy digestibility and metabolizability were ~6% lower in RS compared with PS diet (P<0.001), and metabolizable energy (ME) intake was ~3% lower in RS-fed than in PS-fed pigs (P<0.001). RS-fed pigs showed less feeder-directed (P=0.001) and drinking (P=0.10) behaviours than PS-fed pigs throughout the day. Postprandial peripheral short-chain fatty acid (SCFA) levels were higher in RS-fed than in PS-fed pigs (P<0.001). Postprandial glucose and insulin responses were lower in RS-fed than in PS-fed pigs (P<0.001). Triglyceride levels were higher in RS-fed than in PS-fed pigs (P<0.01), and non-esterified fatty acid levels did not differ between diets (P=0.90). Glucagon-like peptide-1 (GLP-1) levels were lower in RS-fed than in PS-fed pigs (P<0.001), and peptide tyrosine tyrosine (PYY) levels did not differ between diets (P=0.90). Blood serotonin levels were lower (P<0.001), whereas monoamine oxidase activity (P<0.05) and tryptophan (P<0.01) levels were higher in RS-fed than in PS-fed pigs. Despite a lower ME intake, RS seemed to prolong satiety, based on behavioural observations. Possible underlying mechanisms for RS-induced satiety include increased 24 h plasma SCFA levels, and decreased postprandial glucose and insulin responses. GLP-1 and PYY seemed not to play a role in RS-induced satiety. Low blood serotonin levels in RS-fed pigs suggested a difference in intestinal serotonin release between treatments. Increased postprandial plasma triglyceride levels corresponded with increased SCFA levels, but it is unclear whether triglycerides may have signalled satiety in RS-fed pigs.  相似文献

A total of 48 sub-adult hares and adult reproducing farmed hares were used to characterize carcass and meat traits according to the age and gender of animals. With respect to carcass traits, when age increased, the carcass weight significantly increased (2022 to 3391 g; P<0.001), but dressing out percentages did not change. The dissectible fat (1.3% to 2.2%; P<0.05) and Longissimus lumborum (LL) proportions (13.5% to 14.5%; P<0.001) and muscle-to-bone ratio of hind legs (5.11 to 6.23; P<0.001) increased, whereas the hind leg proportions decreased (37.3% to 36.3%; P=0.01). As for the meat quality, the pH of hind leg (5.74 to 5.83; P<0.001) and LL (5.53 to 5.69; P<0.001) increased with age, while the L* index decreased in both cuts (42.9 to 34.4 in hind leg; 45.1 to 40.3 in LL; P<0.001). The redness index increased at the hind leg (4.07 to 5.76; P<0.001), while it decreased at LL (3.03 to 1.46; P<0.001). In the case of the hind leg, meat thawing losses decreased (1.58% to 1.02%), and shear force increased (2.97 to 4.02 kg/g). In the case of LL, thawing losses decreased (8.79% to 4.91%; P<0.001) in the adult hares compared with the sub-adult ones. Meat water and protein contents decreased in the hind leg and LL of the adult hares compared with the sub-adult ones, whereas ether extract increased in a restricted range in LL only (0.92% to 1.11%; P<0.001). In the case of the hind leg, the rate of the saturated fatty acids (SFA) decreased (41.0% to 26.7%), and the rate of the polyunsaturated fatty acids (PUFA) increased (34.0% to 45.3%) (P<0.001). In the case of LL, SFA (38.6% to 42.9%) and monounsaturated fatty acids (19.4% to 27.2%) increased, whereas PUFA decreased (42.0% to 30.1%) when the age increased (P<0.001). Gender affected only the slaughter results and carcass traits. In conclusion, farmed hares have favourable slaughter results (high dressing percentage), carcass traits (high hind legs and loins rates), and meat nutritional value (high-protein, low-fat meat). This fact would offer additional commercial opportunities, in addition to restocking, to hare farmers.  相似文献

A total of 160 Duroc×(Landrace×Large White) pigs, 50% barrows and 50% gilts, of 28.3±4.52 kg of BW were used to study the effect of lysine (Lys) restriction in the finisher period, on growth performances and serum and carcass and meat quality traits. The grower diet (from 30 to 90 kg BW) was slightly Lys-restricted (7.8 g standardised ileal digestible (SID) Lys/kg) in accordance with results from a previous trial. During the finisher period (90 to 130 kg BW), four experimental diets with decreasing SID Lys contents (6.3, 5.6, 4.2 and 3.2 g/kg) were tested. Each of the eight treatments (two sexes×four levels of Lys) was replicated five times. Each replicate was a pen with four pigs allocated together. When animals achieved 129±2.59 kg were slaughtered and carcass and meat characteristics were evaluated. No significant interaction sex×diet was found. During the finisher period, barrows grew faster (P<0.001) and ate more feed (P<0.001) but tended to be less efficient (P=0.055) than gilts. The Lys restriction affected linearly (P<0.001) all productive performance traits; daily BW gain and feed intake decreased and feed conversion ratio increased. Also, the concentration of serum urea at slaughter tended to be higher in barrows than in gilts (P=0.065) and was reduced quadratically by the restriction of Lys in feed (P<0.001). Carcasses from barrows had higher backfat thickness (P<0.01) and lower weight of main trimmed lean cuts (ham+shoulder+loin; P<0.05) than those from gilts. The Lys restriction during the finisher period decreased carcass yield (quadratic; P<0.001) and the weight of major cuts (linear; P<0.001). Sex and diet had limited effect on meat characteristics; the Lys restriction decreased quadratically the proportion of protein (P<0.01) and increased linearly the intramuscular fat (IMF) content (P<0.001). We can conclude that dietary Lys restriction during finisher period in pigs impaired growth performances and was not successful to increase the carcass fat deposition, although it could have positive effects on IMF proportion of pork.  相似文献

Leptin is believed to link metabolic status to reproductive processes. The aim of the present study was to investigate the effect of exogenous leptin on the secretory activity of GnRH/LH system in acutely undernourished prepubertal, female lambs. Merino lambs were randomly divided into four groups, two standard-fed and two fasted for 72 h. One standard and one fasted groups were infused intracerebroventricularly (i.c.v.) with the vehicle; the remaining standard and fasted groups were infused with leptin (25 μg/120 μl/h). Leptin was administered in series of four 1-h infusions at 30-min intervals for 3 consecutive days from 08:30 to 14:00 h. Blood samples were collected on day 0 (before infusions) and on day 3 every 10 min over a 6-h period. Immediately after the experiment, the sheep were slaughtered and brains fixed in situ. Hypothalamic and pituitary tissues were prepared for further immunohistochemical and hybridization in situ analysis. In fasted sheep, increased GnRH levels in the median eminence (P < 0.001) and LHβ levels in the pituitary cells (P < 0.001) plus decreased LHβ mRNA and LH pulsatility in blood plasma were observed (P < 0.05). In leptin-infused fasted sheep, GnRH levels in the median eminence decreased (P < 0.001), LHβ mRNA hybridization signal increased, LHβ levels decreased in the pituitary cells (P < 0.001) and LH pulsatility increased (P < 0.05) in the blood plasma. These results indicate that, in prepubertal sheep, the GnRH/LH axis is sensitive to the fasting signal, that influence of which can be reversed by leptin. Leptin cancels out the suppressing effect of fasting on LH secretion by augmentation of GnRH.  相似文献

Selenium (Se) supplements have been used to control Kashin–Beck disease (KBD) for decades, but the effect of diet without Se supplements is unclear because the prevalence of KBD has decreased. This matched cohort study was undertaken to determine dietary factors affecting selenium nutrition status of children living in KBD areas and the effects of Se supplements in preventing KBD. A total of 593 children aged 5–12 years were randomly selected during the high prevalence period of KBD from 1992 to 1995. Children in one village received Se supplemented (Se+) salt and were matched with three children in 16 other villages who did not receive Se supplemented (Se?) salt. A questionnaire and determinations of occipital hair Se to reflect body Se status were obtained at baseline (April 1992), at 6 months (October 1992), and yearly each April through 1995. Hair Se content in the Se+ group was significantly higher than in the Se? group (P?<?0.001) at all time-points and was significantly related to the incidence of suspected KBD symptoms (P?=?0.018). Four dietary factors significantly affected hair Se contents. Se levels were increased by consumption of Se+ salt (P?<?0.001) and eating meat/egg often (P?=?0.019) or occasionally (P?=?0.001). Se levels were decreased by consumption of grain mildewed at harvest or in storage (P?<?0.001 for each) and drinking ditch, river, or cellar water (P?<?0.001; P?=?0.002; P?<?0.001, respectively). These results show that Se+ salt had a significant effect in maintaining the Se nutrition status of children in this cohort study but that dietary factors in those without Se supplements contributed as well.  相似文献

Alteration in energy metabolism of postmenopausal women might be related to the reduction of dehydroepiandrosterone sulfate (DHEAS). DHEA and DHEAS decline with age, leveling at their nadir near menopause. DHEA and DHEAS modulate fatty acid metabolism by regulating carnitine acyltransferases and CoA. The purpose of this study was to determine whether dietary supplementation with DHEAS would also increase tissue l-carnitine levels, carnitine acetyltransferase (CAT) activity and mitochondrial respiration in oophorectomized rats. Plasma l-carnitine levels rose following oophorectomy in all groups (P<0.0001). Supplementation with DHEAS was not associated with further elevation of plasma l-carnitine levels, but with increased hepatic total and free l-carnitine (P=0.021 and P<0.0001, respectively) and cardiac total l-carnitine concentrations (P=0.045). In addition, DHEAS supplementation increased both hepatic and cardiac CAT activities (P<0.0001 and P=0.05 respectively). CAT activity positively correlated with the total and free carnitine levels in both liver and heart (r=0.764, r=0.785 and r=0.700, r=0.519, respectively). Liver mitochondrial respiratory control ratio, ADP:O ratio and oxygen uptake were similar in both control and supplemented groups. These results demonstrate that in oophorectomized rats, dietary DHEAS supplementation increases the liver and heart l-carnitine levels and CAT activities. In conclusion, DHEAS may modulate l-carnitine level and CAT activity in estrogen deficient rats. The potential role of DHEAS in the regulation of fatty acid oxidation in postmenopausal women is worthy of investigation.  相似文献

The aim of this study was to investigate the effects of different levels of cadmium supplementation (0, 5, 10, 20, 40 and 80 mg/kg) in the diet on performance, egg quality, tibia biomechanical properties and eggshell and bone mineral contents in laying quails. In this 10-week trial, a total of 96 laying quails, aged 21 weeks, were randomly distributed among six experimental groups. Each experimental group contained four replicates of four birds each. The performance parameters were adversely affected quadratically when cadmium was added to the diets in the concentrations of 20 mg/kg and above (P<0.01). The specific gravity and eggshell weight were maximal with the addition of 20 mg/kg cadmium to the diet. The biomechanical properties of the tibia were negatively affected by cadmium supplementation in quails (P<0.05). The eggshell boron content decreased linearly (P<0.001) with cadmium supplementation to the diet. The cadmium content in bone increased when cadmium was added to the diets (P<0.001). The bone boron concentration decreased as dietary cadmium supplementation was increased (P<0.001).  相似文献

In a previous study, carnitine supplementation to piglets during the suckling period resulted in an increased total muscle fibre number at weaning in piglets of low birth weight. The objective of the present study was to investigate whether this effect is maintained until market age and whether this would attenuate the negative consequences of low birth weight on carcass and meat quality. Using a split-plot design with litter as block, sex as whole plot and treatment as subplot, the effects of early-postnatal l-carnitine supplementation on female and castrated male piglets of low birth weight were investigated on a total of 56 German Landrace piglets from 14 litters. From days 7 to 27 of age piglets were orally supplemented once daily with 400 mg of l-carnitine dissolved in 1 ml of water or received an equal volume of water without carnitine. From weaning (day 28) until slaughter (day 166 of age) all pigs were fed standard diets. At weaning, carnitine-supplemented piglets had a twofold increased concentration of free carnitine (P < 0.001) and a lower concentration of non-esterified fatty acids (P < 0.05) in blood plasma indicating that carnitine became bioavailable and increased fatty acid utilization during the period of supplementation. Growth performance was not influenced by treatment in any growth period. Dual-energy X-ray absorptiometry revealed no differences in body composition between groups in weeks 12, 16 and 20 of age. LW at slaughter, carcass weight, measures of meat yield and fat accretion, as well as body composition by chemical analyses and dissection of primal cuts did not differ between treatments. No differences between control and carnitine-treated pigs in total fibre number (P = 0.85) and fibre cross-sectional area (P = 0.68) in m. semitendinosus (ST) measured at slaughter could be observed. The carnitine group tended to exhibit a smaller proportion of slow-twitch oxidative fibres (P = 0.08), a greater proportion of fast-twitch glycolytic fibres (P = 0.11), and increased specific lactate dehydrogenase activity (P = 0.09) in ST indicating a more glycolytic muscle metabolism. Compared with the controls, a lower pH24 value was observed (P = 0.05) in ST muscle of carnitine-supplemented pigs, which – in castrates only – was associated with an increased drip loss (P < 0.01). Meat quality traits in m. longissimus were not influenced by treatment. In conclusion, our hypothesis that early-postnatal carnitine supplementation to piglets of low birth weight permanently increases myofibre number and improves later carcass and meat quality could not be confirmed by this experiment.  相似文献

This study investigated the effects of rider weight in the BW ratio (BWR) range common for Icelandic horses (20% to 35%), on stride parameters in tölt in Icelandic horses. The kinematics of eight experienced Icelandic school horses were measured during an incremental exercise test using a high-speed camera (300 frames/s). Each horse performed five phases (642 m each) in tölt at a BWR between rider (including saddle) and horse starting at 20% (BWR20) and increasing to 25% (BWR25), 30% (BWR30), 35% (BWR35) and finally 20% (BWR20b) was repeated. One professional rider rode all horses and weight (lead) was added to saddle and rider as needed. For each phase, eight strides at speed of 5.5 m/s were analyzed for stride duration, stride frequency, stride length, duty factor (DF), lateral advanced placement, lateral advanced liftoff, unipedal support (UPS), bipedal support (BPS) and height of front leg action. Stride length became shorter (Y=2.73−0.004x; P<0.01) and more frequent (Y=2.56+0.002x; P<0.001) with added weight. Duty factor and BPS increased with increased BWR (P<0.001), whereas UPS decreased (P<0.001). Lateral advanced timing of limb placement and liftoff and height of front leg action were not affected by BWR (P>0.05). In conclusion, increased BWR decreased stride length and increased DF proportionally to the same extent in all limbs, whereas BPS increased at the expense of decreased UPS. These changes can be expected to decrease tölt quality when subjectively evaluated according to the breeding goals for the Icelandic horse. However, beat, symmetry and height of front leg lifting were not affected by BWR.  相似文献

We analyzed the time course of changes in muscle activity of the prime mover and synergist muscles during a sustained brake-pulling action and investigated the relationship between muscle activity and braking force fluctuation (FF). Thirty-two participants performed a continuous fatiguing protocol (CFP) at 30% of maximal voluntary contraction (MVC) until failure. Surface electromyography was used to analyze root mean square (RMS) values in the flexor digitorum superficialis (FD), flexor carpi radialis (FC), extensor digitorum communis (ED), extensor carpi radialis (EC), brachioradialis (BR), biceps brachii (BB), and triceps brachii (TB). The FF and RMS in all muscles increased progressively (P<0.01) during the CFP, with sharp increments at time limit particularly in FD and FC (P<0.001). The RMS of the FD and FC were comparable to the baseline MVC values at time limit, in comparison to the other muscles that did not reach such levels of activity (P<0.003). The three flexor/extensor ratios used to measure coactivation levels decreased significantly (P<0.001). In contrast to RMS, MVC was still depressed at the minute 10 of recovery. The results suggest that the time limit was mainly constrained by fatigue-related mechanisms of the FD and FC but not by those of other synergist and antagonist muscles.  相似文献

Four silages were harvested at approximately one-week intervals from the same timothymeadow fescue sward. Advanced maturity of the herbage was evidenced by increased neutral detergent fibre [409, 497, 579 and 623 g in 1 kg dry matter (DM)] and decreased nitrogen (N; 29.9, using four ruminally and duodenally cannulated young cattle in a 4 × 4 Latin square experiment. On DM basis (g kg−1), the diet comprised grass silage (700), rolled barley (240) and rapeseed meal (60) and it was given at a rate of 70 g DM (kg live weight)−0.75 per day.Organic matter digestibility decreased in a curvilinear manner (PLINEAR (L) < 0.001, PCUBIC (C) < 0.01) the values being 0.821, 0.816, 0.758 and 0.747 for the diets based on the four silages in the order of harvest date. Rumen pH increased linearly (PL < 0.05) and ammonia N concentration decreased curvilinearly (PL < 0.01, PC < 0.05) as the grass matured. The molar proportion of acetate in the rumen VFA increased (PL < 0.001) and the proportion of butyrate decreased (PL < 0.001) with increased grass maturity. The silage harvest date did not affect the proportion of propionate. The changes in rumen fermentation pattern were associated with a decrease (PL < 0.05) in rumen protozoal number with increasing maturity of grass.N intake decreased significantly (PL < 0.001, PC < 0.01) with the maturity of grass from 167.5 to 118.0 g per day, but duodenal non-ammonia N decreased only from 111.3 to 97.3 g per day indicating greater N losses from the rumen with early-cut silages. The efficiency of microbial protein synthesis in the rumen was not affected by the maturity of grass ensiled. Apparent digestibility of N decreased (PL < 0.001, PC < 0.01) and the degradability of N in the rumen decreased (PL < 0.05) as the grass matured.  相似文献

Serum levels of osteocalcin (OC) have been found to be a specific biochemical parameter of bone formation. We measured serum levels of osteocalcin, parathyroid hormone (PTH) and 25-hydroxyvitamin D (25(OH)D) in 49 patients with liver cirrhosis, who are known to have an increased prevalence of metabolic bone disease, and a matched control group (n = 35). Serum levels of OC were significantly decreased in the patients with liver cirrhosis when compared to control subjects (P < 0.001). Serum levels of 25(OH)D were decreased (P < 0.001), whereas no statistical difference was found between the serum levels of PTH in the patients with liver cirrhosis and those of the controls. In a subgroup of 23 patients with cirrhosis of the liver and 34 control subjects, the bone mineral content (BMC) of the non-dominant forearm was determined by single photon absorptiometry. BMC was significantly lower in the patient with liver cirrhosis than the control subjects (P < 0.04). Our data demonstrate vitamin D deficiency, decreased bone formation and a decreased BMC in patients with liver cirrhosis.  相似文献

The mechanism of action of leptin in pregnant breeding sows, in which hyperphagia is managed through dietary strategies, is yet to be clarified. The aim of this study was to monitor leptin concentrations and their interactions with lactogenic hormones in Large White×Landrace breeding multiparous sows (n=15). All sows showed a normal body condition (mean body condition score: 2.96). Blood samples were collected the day after weaning the litters, at insemination, every 15 days up to day 45 of pregnancy and every 7 days from day 46 to farrowing. At delivery, the placenta was collected for the analysis of leptin and leptin receptor expressions. Plasma leptin levels increased from the end of mid gestation (day 72) and remained high until farrowing (P<0.05). As expected, plasma prolactin (PRL), low during most of pregnancy, increased during the 2 weeks before farrowing (P<0.05), whereas progesterone levels reached plateau at 30 days of gestation and decreased at farrowing (P<0.05). Cortisol levels peaked close to farrowing (P<0.05). Leptin was expressed in the placenta, where the receptor expression analysis showed the presence of the short form but not of the long form. A positive correlation was found between leptin and PRL concentrations during mid (r=0.430; P<0.001) and late (r=0.687; P<0.001) pregnancy, and with progesterone in early pregnancy (r=0.462; P<0.05). During late gestation, a positive correlation was observed between leptin and cortisol (r=0.585; P<0.001). Our results suggested that, in restrictively fed pregnant sows, the leptin levels increased from the end of mid pregnancy to delivery, confirming the presence of leptin resistance. We showed a correlation between leptin and lactogenic hormones during different stages of pregnancy in sows. Lactogenic hormones show pregnancy-specific changes in their secretion and all may become involved in modulating leptin signal.  相似文献

The objectives of this study were to assess the relationship between blood lactate variation measured at the plant, and pork quality variation on a large sample size and under commercial preslaughter handling conditions. A total of 600 pigs were randomly chosen on arrival at a commercial slaughter plant and blood samples taken from the ear vein at unloading (UN), after lairage (LA), in the restrainer (RE; before stunning) and at exsanguination (EX) were analysed for lactate content using a Lactate Scout Analyzer (LSA). In order to have a large range of measures, pigs were distributed into two groups; one kept in lairage overnight (G1) and the other for 2 to 3 h (G2) before slaughter. Meat quality was assessed in the Longissimus thoracis (LT), Semimembranosus (SM) and Adductor (AD) muscles by measuring the pH 30 min postmortem (pH1) and at 24 h postmortem (pHu), the colour and the drip loss. Blood lactate levels did not differ between G1 and G2 (P>0.05). A reduced muscle lactate and glucose contents (P=0.02 and P=0.004, respectively) resulting in a lower (P<0.001) glycolytic potential (GP) was observed in the LT muscle of G1 pigs when compared with G2 loins. In the LT muscle of G1 pigs, the lower GP resulted in an increased pHu (r=−0.67; P<0.001), decreased drip loss (r=0.57; P<0.001) and darker colour (r=0.50; P<0.001) compared with G2. In both G1 and G2 pigs, the lower GP was correlated to higher pHu value in the SM and AD muscles (r=−0.73; P<0.001). The greatest correlation was observed in G2 between blood lactate levels at LA and pHu value of the SM and AD muscles (r=0.46 and r=0.44, respectively; P<0.001 for both muscles). The second greatest correlation was found between blood lactate levels at EX and pH1 value in the SM muscle in both groups (r=−0.37 and r=−0.41, respectively; P<0.001 for both groups). Based on the results of this study, it appears that blood lactate levels, as measured by the LSA, reliably reflect the physiological response of pigs to perimortem stress and may help explain the variation in pork quality.  相似文献

According to the Regulation No. 1831/2003 of the European Parliament and European Union Council, the use of antibiotics as a dietary supplements has been prohibited. It seems that the administration of prebiotics, instead of antibiotics, into the pig's diet, may regulate the intestinal microbiota and has a long-term health-related impact on the host. Inulin-type fructans can stimulate mineral absorption from the gut. Additionally, it may regulate energy metabolism and activate enzymatic mechanisms preventing oxidative stress. The goal of the present study was to estimate the influence of dietary supplementation with dried chicory root or native chicory inulin on 1) liver histology; 2) liver and kidney lipid metabolism indices, activity of selected enzymes, concentration of macro- and micronutrients and heavy metals; 3) blood plasma, liver and kidney oxidative stress biomarkers and 4) blood plasma water–electrolyte homeostasis indices in growing pigs. The nutritional study was conducted on 24 piglets assigned to 3 dietary groups (n = 8): control (C) fed a basal diet and two experimental groups receiving basal diet supplemented with 2% of inulin (IN) either 4% of dried chicory root (CR). The animals were fed with a group-specific diets for 40 days and then subjected to euthanasia. Subsequently, blood, liver and kidney samples were harvested for further processing. In the control and experimental groups, no apparent morphological abnormalities in the liver tissues were seen. The percent of periodic acid Schiff positive glycogen liver cells was significantly lower in the CR group as compared to C and IN groups (P < 0.001). Chicory root supplementation improved blood plasma prooxidative-antioxidative balance – PAB (P < 0.001) and liver PAB (P < 0.01) and thiobarbituric acid reactive substances – thiobarbituric acid-reactive substances (P < 0.05). Feeding the CR diet increased calcium (P < 0.001) and potassium (P < 0.05) and decreased cadmium (P ≥ 0.05) content in the liver when compared to the C group. Administration of the CR and IN diets increased selenium (Se) and sodium concentrations, whereas decreased zinc content both in the liver (P < 0.01; P < 0.05 and P < 0.05, respectively) and in the kidney (P < 0.01; P < 0.001 and P < 0.001, respectively) of pigs. Additionally, a higher concentration of lead (P < 0.05) was observed in the kidney of pigs fed the CR diet. In conclusion, both dietary supplements had a potential to significantly improve the Se status and oxidoreductive homeostasis in growing pigs.  相似文献

Cardiovascular diseases are the main reason of high mortality among hemodialysis patients. Decreased serum selenium levels may have a role in accelerated atherosclerosis in this patient group. The hypothesis of this study was to show a correlation between decreased serum selenium levels and coronary flow reserve as an indicator of endothelial dysfunction and atherosclerosis in HD patients. Seventy-one chronic hemodialysis patients and age 65 and sex-matched healthy controls were included in the study. Plasma selenium levels were measured by spectrophotometry, and coronary flow reserve was assessed by transthoracic Doppler echocardiography. Serum selenium levels (34.16?±?6.15 ng/ml vs. 52.4?±?5.51 ng/ml, P?<?0.001) and coronary flow reserve values (1.73?±?0.11 vs. 2.32?±?0.28, P?<?0.001) were significantly lower in hemodialysis patients compared with controls, respectively. There was a significant positive correlation between coronary flow reserve and serum levels of selenium (r?=?0.676, P?<?0.001). A linear regression analysis showed that serum levels of selenium were independently and positively correlated with coronary flow reserve (regression coefficient?=?0.650, P?<?0.05). This study was the first to show a positive and independent correlation between decreased selenium levels and diminished coronary flow reserve as an indicator of endothelial dysfunction and atherosclerosis in hemodialysis patients. Our data suggest that decreased serum selenium levels may facilitate the development of endothelial dysfunction and disruption of coronary flow reserve which occur before the development of overt atherosclerosis.  相似文献

The protective effects of dietary pyrroloquinoline quinone disodium (PQQ.Na2) supplementation against oxidized sunflower oil-induced oxidative stress and liver injury in laying hens were examined. Three hundred and sixty 53-week-old Hy-Line Gray laying hens were randomly allocated into one of the five dietary treatments. The treatments included: (1) a diet containing 2% fresh sunflower oil; (2) a diet containing 2% thermally oxidized sunflower oil; (3) an oxidized sunflower oil diet with 100 mg/kg of added vitamin E; (4) an oxidized sunflower oil diet with 0.08 mg/kg of PQQ.Na2; and (5) an oxidized sunflower oil diet with 0.12 mg/kg of PQQ.Na2. Birds fed the oxidized sunflower oil diet showed a lower feed intake compared to birds fed the fresh oil diet or oxidized oil diet supplemented with vitamin E (P=0.009). Exposure to oxidized sunflower oil increased plasma malondialdehyde (P<0.001), hepatic reactive oxygen species (P<0.05) and carbonyl group levels (P<0.001), but decreased plasma glutathione levels (P=0.006) in laying hens. These unfavorable changes induced by the oxidized sunflower oil diet were modulated by dietary vitamin E or PQQ.Na2 supplementation to levels comparable to the fresh oil group. Dietary supplementation with PQQ.Na2 or vitamin E increased the activities of total superoxide dismutase and glutathione peroxidase in plasma and the liver, when compared with the oxidized sunflower oil group (P<0.05). PQQ.Na2 or vitamin E diminished the oxidized sunflower oil diet induced elevation of liver weight (P=0.026), liver to BW ratio (P=0.001) and plasma activities of alanine aminotransferase (P=0.001) and aspartate aminotransferase (P<0.001) and maintained these indices at the similar levels to the fresh oil diet. Furthermore, oxidized sunflower oil increased hepatic DNA tail length (P<0.05) and tail moment (P<0.05) compared with the fresh oil group. Dietary supplementation of PQQ.Na2 or vitamin E decreased the oxidized oil diet induced DNA tail length and tail moment to the basal levels in fresh oil diet. These results indicate that PQQ.Na2 is a potential antioxidant and is as effective against oxidized oil-related liver injury in laying hens as vitamin E. The protective effects of PQQ.Na2 against liver damage induced by oxidized oil may be partially due to its role in the scavenging of free radicals, inhibiting of lipid peroxidation and enhancing of antioxidant defense systems.  相似文献
