Summary The intratesticular excurrent duct system of the bull is composed of rete testis, tubuli recti, and the terminal segment of the seminiferous tubules. Each terminal segment is surrounded by a vascular plexus and may be subdivided into a transitional region, middle portion, and terminal plug. The modified supporting cells of the middle portion and the terminal plug no longer display the typical Sertoli-Sertoli junctions seen in the transitional region and the seminiferous tubule proper. In the region of the terminal plug a distinct central lumen is generally not observed: spermatozoa and tubular fluid must pass through an intricate system of communicating clefts between the apices of the closely attached modified supporting cells. Vacuoles in the supranuclear region of the cells in the middle portion indicate strong transepithelial fluid transport. In analogy to the epithelium of rete testis and tubuli recti, the supporting cells of the terminal segment are capable of phagocytosing spermatozoa. The vascular plexus investing the terminal segment serves a dual purpose: it is a regulatory device for fluid and sperm transport, as well as an area of increased diapedesis for white blood cells.Supported by a grant from the Deutsche Forschungsgemeinschaft  相似文献

The DNA of most vertebrate sperm cells is packaged by protamines. The primary structure of mammalian protamine I can be divided into three domains, a central DNA binding domain that is arginine-rich and amino- and carboxyl-terminal domains that are rich in cysteine residues. In native bull sperm chromatin, intramolecular disulfide bonds hold the terminal domains of bull protamine folded back onto the central DNA binding domain, whereas intermolecular disulfide bonds between DNA-bound protamines help stabilize the chromatin of mature mammalian sperm cells. Folded bull protamine was used to condense DNA in vitro under various solution conditions. Using transmission electron microscopy and light scattering, we show that bull protamine forms particles with DNA that are morphologically similar to the subunits of native bull sperm chromatin. In addition, the stability provided by intermolecular disulfide bonds formed between bull protamine molecules within in vitro DNA condensates is comparable with that observed for native bull sperm chromatin. The importance of the bull protamine terminal domains in controlling the bull sperm chromatin morphology is indicated by our observation that DNA condensates formed under identical conditions with a fish protamine, which lacks cysteine-rich terminal domains, do not produce as uniform structures as bull protamine. A model is also presented for the bull protamine.DNA complex in native sperm cell chromatin that provides an explanation for the positions of the cysteine residues in bull protamine that form intermolecular disulfide bonds.  相似文献

Summary The boundary tissue of bovine testicular seminiferous tubules exhibits remarkable regional differences at the level of the seminiferous tubule proper, as compared with its terminal segment. The basal lamina of the seminiferous tubule proper is multilayered and possesses knob-like protrusions. At the level of the terminal segment the basal lamina is highly specialized; in the region of the terminal plug candelabrum-like projections of the tubular basal lamina invade the bases of the modified supporting cells up to a depth of 3.5 m. The adjoining surface of these supporting cells is densely studded with hemidesmosomes. The elongated peritubular cells are arranged in 3–5 concentric layers around the tubulus seminiferus proper but form a loose association at the level of the terminal segment. Where the terminal segment joins the testicular straight tubule, peritubular cells may assemble to constitute a contractile spiral. Elastic tissue is situated mainly subjacent to the tubular basal lamina and to a lesser degree between the peritubular cell layers. A peritubular space lined by endothelium-like cells may surround the seminiferous tubule proper and also the transitional zone of the terminal segment.Supported by a grant from the Deutsche Forschungsgemeinschaft  相似文献

The distribution of carbonic anhydrase (CA) was studied in the testis and epididymis of mature, male rabbits using a cobalt precipitation method. CA was found only in the endothelium of the capillaries in the testis. The epididymal duct was divided into initial, middle and terminal segments. Strong cytoplasmic CA was present in the apical cells in the initial and middle segments. Vacuoles with CA staining in the membranes were found in the principal cells in the middle segment. Intensely stained basal cells were present in the terminal segment. In the last part of the terminal segment and the first of the ductus deferens the basolateral cell membranes were also stained. The function of the enzyme is discussed especially in relation to acidification of the epididymal fluid and facilitation of CO2 diffusion.  相似文献

Distinct morphological regions, initial, middle and terminal segments, were distinguishable histologically; the middle segment was further subdivided into proximal, intermediated and distal parts. PAS-positive, diastase-resistant reaction was detected in the blood vessels, subepithelial tissue and stereocilia of all segments. Acid phosphatase was demonstrated in the epithelial cells with the highest activity being in the proximal part of the middle segment. Non-specific esterase gave a similar reaction but the strongest activity was in the terminal segment. Alkaline phosphatase, adenosine triphosphatase and adenosine monophosphatase were of similar activity in the subepithelial tissue, blood vessels, stereocilia and luminal contents; the strongest reaction occurred in the middle segment. Lactate, succinate, glutamate and glucose-6-phosphate dehydrogenases were examined; LDH was more active than the others particularly in the terminal segment. Some reaction was found in the epithelial cells, subepithelial tissue and luminal contents.  相似文献

Summary Histology and ultrastructure of the connexion of seminiferous and straight testicular tubules were studied in 58 bovine testes of 29 animals ranging from 4 to 52 weeks of postnatal development. In the 4th and 8th week seminiferous tubules are solid. Their non-germinal supporting cells possess spherical nuclei in a basal location and a great amount of granular endoplasmic reticulum. The straight tubules have a narrow lumen and a stratified epithelium rich in intercellular canaliculi. Between 20 and 25 weeks the seminiferous tubules acquire a lumen and develop a terminal segment, the tip of which (terminal plug) protrudes into the cup-shaped modification of the adjacent straight tubule. At 30 weeks the structural differentiation between seminiferous tubule proper and its terminal segment has proceeded: in the former spermatocytes and spermatids make their first appearance, and the supporting cells have transformed to Sertoli cells. In the latter the morphology of the supporting cell preserves a more primitive state. Starting from the 16th week and proceeding through the 30th week and further, the epithelium of the tubulus rectus close to the connexion with the seminiferous tubule becomes monolayered by rearrangement of its cells and advances along the basal lamina into the area of the seminiferous tubule. Those cells of the seminiferous tubule that are cut off from the basal lamina by invading rectus cells degenerate. Between 40 and 52 weeks the adult situation is principally achieved. The terminal segment of the seminiferous tubule is tripartite consisting of transitional region, intermediate portion, and terminal plug. The terminal segment is surrounded by a vascular plexus. The straight testicular tubule adjacent to the terminal segment is modified into a cup region encompassing the terminal plug, followed by a narrow stalk region, which is lined by simple columnar epithelium. The present study deals with the immunohistochemical localization of S-100 protein in the testes of seven mammalian species including rat, cat, dog, pig, sheep, cattle and horse. Significant differences are demonstrated in the cellular distribution and intensity of immunoreaction for the protein. In bull, ram, boar and cat tests S-100 protein was localized in the cytoplasm and nuclei of Sertoli cells. A particularly intense staining was seen in the modified Sertoli cells of the terminal tubular segment. With the exception of the cat and horse S-100 protein immunoreactivity was additionally found in epithelial cells of the straight testicular tubules and in the epithelial cells of the rete testis. Endothelial cells of capillaries, veins and lymphatic vessels are regularly S-100 immunoreactive in ruminants. Leydig cells were found to be strongly positive for S-100 protein in the cat and rat testes and to a lower degree in pig and horse testes. Finally a distinct immunostaining of peritubular cells was restricted to the testis of dogs and rats. The remarkable species-specific variations of immunoreactivity for S-100 protein in different cell types of the testis support the hypothesis that S-100 protein is a multifunctional protein and may have a different function in testicular physiology.  相似文献

Relatively little is known about the morphology and ultrastructure of the Malpighian tubules of spiders (Arachnida: Araneae). Our study represents the first investigation of the Malpighian tubules of a theraphosid spider and is the only study to examine the living Malpighian tubules using confocal laser scanning microscopy. In theraphosid spiders, the Malpighian tubules originate from the stercoral pocket in the posterior portion of the opisthosoma and extend forward toward the prosoma in a dendritic pattern. There are three distinct segments (initial, main, and terminal), all dark brown in appearance. Each segment has distinctive ultrastructural features. Both the terminal and the main segment appear to be composed of at least two cell types with finger-like cytoplasmic protrusions associated with one of these types. The terminal segment, which is most proximal to the stercoral pocket, is the largest in diameter. It is composed of large, cuboidal cells containing many mitochondria and lipid inclusions. The main segment is intermediate in diameter with many mitochondria and secretory vesicles present. The initial segment is relatively thin in comparison to the other segments and is intimately associated with the digestive gland. The cells of the initial segment contain very little cytoplasm, fewer mitochondria, secretory vesicles, and prominent inclusions.  相似文献

Summary The ductus epididymidis of the tammar is lined by an epithelium composed of principal, mitochondria-rich, apical and basal cells, and intraepithelial leucocytes. The epithelium is structurally differentiated into 6 zones referred to as the initial segment, middle segment (3 subdivisions) and terminal segment (2 subdivisions). The occurrence of the initial, middle and terminal segments corresponds quite closely to the anatomical differentiation of the epididymis into a head, body and tail.The initial segment epithelium in the tammar is lower and has shorter and more slender stereocilia than in other mammals which have been described. Otherwise, the structure of the epithelium has similar characteristics in the tammar to that described in other mammals.Spermatozoa begin to develop the capacity for motility within the initial segment, but only show structural signs of maturation in the middle segment. The sperm head rotates through 90 degrees in the proximal subdivision of the middle segment. The cytoplasmic droplet is detached and spermatozoa develop the capacity for motility in the middle subdivision of the middle segment. The cytoplasmic droplets are phagocytosed by the epididymal epithelium of the middle segment. Sperm storage appears to be the main function of the terminal segment.  相似文献

The epididymis of the adult honey possum, Tarsipes rostratus, is enclosed by a heavily pigmented tunica vaginalis and lies with the testis in a prominent prepenile scrotum. It is connected to the testis by a single ductus efferentis and is lined by approximately equal numbers of cuboidal ciliated and principal cells. It is unusual for marsupials in having no well-defined compartments or fibrous septae and in having extensive convolutions of the duct only at the caudal flexure. Three principal functional zones (initial, middle, and terminal segments) were identified in the epididymis, based on epithelial type and ultrastructural evidence of sperm maturation. Luminal diameter increases progressively throughout the tract, and epithelial height variations (from about 2 to 20 microns) are greatest in the terminal segment. The epithelium itself is remarkably low (maximum of 21.6 microns) compared with that seen in the epididymis of other mammals. The thickness of the peritubular smooth muscle coat increases close to the junction of the epididymis and ductus deferens. Sperm concentrations were estimated from counts of sperm nuclei and thus can be no more than approximations. The figures are consistent, however, with a rapid increase in concentration in the initial segment, indicating extensive fluid resorption. Sperm concentrations appear to peak in the distal zone of the terminal segment, although sampling problems and wide variations in count make such a conclusion only tentative. Principal and basal cells are the predominant cell types in the epididymal epithelium. Basal cells are most abundant in the initial and distal middle segment. Principal cells show structural evidence of active exchange with the luminal contents and have abundant apical stereocilia, the structure of which depends on the epididymal zone. Other cell types occur less commonly in the epithelium. Lipid-rich and phagocytic principal cells are restricted to the middle and distal zones of the middle segment, respectively. Clear cells, restricted to the terminal segment, and halo cells were found in very low numbers. As in some other marsupials, principal cells (possibly specialized for this function) selectively remove cytoplasmic droplets and probably other cellular debris from the luminal contents. In Tarsipes, however, this process is not very efficient, and many discarded droplets pass through to the terminal segment where they form large masses of debris associated with aggregates of degenerating spermatozoa.  相似文献

The epididymis of the European mole (Talpa europaea) was studied by light and electron microscopy. In the sexually active animal, spermatozoa mature during their passage through the epididymis and the structure of the cells lining the duct suggests a clear regional division into initial, middle and terminal segments. Numerous intra-epithelial vesicles were present in the distal part of the middle segment of sexually active moles and the lining epithelium in the terminal segment appeared to be secretory. Variation in the sensitivity of different regions of the epididymis to androgens was apparent: the principal cells of the initial segment were morphologically active only during the peak of the breeding season in spring, while the cells of the terminal segment became active earlier and remained so for longer. During sexual regression, many autophagic vacuoles were found in the principal cells, and these became transformed into lipofuscin pigment granules. Cells heavily laden with these granules appeared concurrently in the lining epithelium. It is suggested that such cells may be involved in the regression of principal cells by means of heterophagic activity. A similar situation was also observed, but to a lesser extent, at the beginning of the breeding season. Outside the breeding season, the organelles of the principal cells were poorly developed throughout the epididymis, and lipofuscin pigment granules remained in the principal and basal cells of adults. As luminal fluid moves from the seminiferous tubule and enters the rete testis, its protein concentration declines from approximately 6 mg/ml to 1 mg/ml. It was therefore suggested that protein is either 1) utilized by the spermatozoa, 2) transported across the epithelium of the terminal segment of the seminiferous tubule, the tubuli recti or rete testis, or 3) absorbed and degraded by the epithelium. Horseradish peroxidase (HRP), a protein marker, was microperfused into single seminiferous tubules or perfused directly into the rete. After fixation, the HRP was localized histochemically and the tissue observed under the light- and electron microscope. HRP was taken up via pinocytotic vesicles into the cytoplasm of the Sertoli cells and germ cells but did not permeate extracellularly beyond the tight junctions. Similar results were obtained in the cells lining the terminal segment and the tubuli recti. The rete epithelium showed uptake of HRP into coated and noncoated vesicles, while some cells additionally revealed diffuse cytoplasmic distribution of HRP. The terminal segment, tubuli recti, and rete testis may be important routes by which proteins may leave the testicular fluid either to be degraded or to enter the blood.  相似文献

An anatomical study of the digestive tract of the channel catfish revealed that the oesophageal mucosa was longitudinally folded and that secondary folds were occasionally located on the primary longitudinal folds. The infoldings were more numerous near the stomach. The stratified squamous epithelium covering the folds was made up of a basal layer, large mucous cells and simple squamous cells on the surface. The epithelium on the side of the folds consisted primarily of mucous secreting cells. Taste buds were observed between mucous cells on the apical portion of the oesophageal folds and were more prevalent in the cranial part of the oesophagus. The remaining layers of the oesophagus were: a lamina propria-submucosa, tunica muscularis and adventitia or serosa.
The J-shaped stomach had two regions: a large sac-shaped region containing gastric glands and a smaller, nonglandular pyloric region. The large rugae of the stomach became gradually smaller near the pylorus. There was a well developed pyloric sphincter. The mucosa included a simple columnar epithelium, a lamina propria and adventitia or serosa.
The intestine could be differentiated into a thick ascending segment, a descending segment, a thin convoluted segment and a thicker terminal segment, the rectum. Many mucosal folds containing branched villi characterized the ascending segment of the intestine. The descending and convoluted segments contained fewer folds with shorter and less-branching villi and were smaller in diameter and thinner walled. Descending and convoluted segments were also mildly convoluted and accounted for 80% of the total length of the intestine. An intestinal valve with a sphincter marked the beginning of the rectum. There was an approximately four-fold increase in the thickness of the tunica muscularis of the terminal segment of the intestine.  相似文献

Summary The pars descendens (pars recta) of the proximal tubule in the male rat kidney, consisting of the terminal part of the second proximal segment (P2) and of the third proximal segment (P3), was studied with the electron microscope. A technique of tissue orientation and trimming was used which permitted precise topographic definition of the tubules studied in the electron microscope. The terminal descending part of the P2 showed some minor differences from the convoluted part of this segment, and ultrastructure also changed along the course of the P3. In the beginning of the latter segment numerous, shallow interdigitations were observed between adjacent cells; along the course of the segment they decreased in number or disappeared. In the initial part of the P3 mitochondria were more abundant than in the terminal portion of the segment and at least as numerous as in the straight part of the P2. Also, the dense, acid phosphatase-positive cytoplasmic bodies decreased somewhat in size along the course of the P3. Ferritin-conjugated soybean trypsin inhibitor was used for the ultrastructural localization of acrosin in bull spermatozoa following acrosomal disruption. The ferritin label was observed in the anterior segment of the acrosome in disrupted cells only. Emptied acrosomes were labeled, mostly on the external surface of their outer membrane. Labeling was also found on the material bound to detached acrosomal caps. However, at no time could the ferritin label be found on the inner acrosomal membrane. It is concluded that acrosin activity is not present on the inner acrosomal membrane but is lost from the acrosomal matrix as the acrosomal reaction proceeds.  相似文献

In female Podarcis taurica the kidney collecting tubule always consists entirely of mucous-secreting cells. In males it has a seasonally variable sexual segment and a non variable mucous-secreting segment. In April the sexual segment is composed of columnar cells with cytoplasm rich in ribosomes and Golgi bodies and apical clusters of large vesicles with fibrous contents. The terminal region of the sexual segment also has pillar-shaped cells resembling those of the mucous-secreting segment. By May the accumulation of apical vesicles reaches a maximum, and many cells have apparently extruded their secretion into the lumen. In July all the cells are pillar shaped with dilated endoplasmic reticulum but with few apical vesicles. In September the sexual segment has some cells resembling those of the mucous-secreting segment and others the sexual segment pillar cells in April. It is suggested that during sexual activity in the spring the sexual segment secretes a spermatozoon-nutrient protein but subsequently reverts to mucous secretion. The non variable mucous-secreting regions in both males and females consist of mucous, intermediate, and dark cells. Mucous cells have apical masses of closely packed droplets, whereas dark cells have dense cytoplasm and small, loosely associated apical vesicles. Intermediate cells have some dark cell features but mucous cell apical vesicles. The dark, intermediate, and mucous cells probably represent activity states of a single type. The mucous secretion is interpreted as a protective material which lines the urinary passage and coats the secreted solid urates. Elaborated intercellular spaces in the mucous-secreting regions may indicate a water absorption capacity in urine concentration.  相似文献

Localization and activity of five hydrolases (alkaline phosphatase, adenosine triphosphatase, acid phosphatase, nonspecific esterase and leucylamino-peptidase) were evaluated histochemically in the epididymides of mature dogs. In the ductuli efferentes, cilia and apical parts of the epithelial cells displayed high activity of alkaline phosphatase and adenosine triphosphatase. Strong activity of acid phosphatase, nonspecific esterase and leucylamino-peptidase was present in the basal and supranuclear zones of the epithelium of the ductuli efferentes. Stereocilia of all three segments of the ductus epididymidis showed a high activity of alkaline phosphatase. Positive adenosine triphosphatase reaction was confined to the stereocilia of the initial segment. A complex pattern of acid phosphatase activity was observed in the middle segment. The subdivision of the middle segment in four subsegments was therefore suggested. In the epithelium of the initial segment only a few nonspecific esterase-positive cells were seen. The infranuclear and basal areas of the epithelium in the middle segment and the supranuclear zone of the terminal segment displayed distinct nonspecific esterase activity. The possible contribution of the hydrolases to the function of the epididymis is discussed.  相似文献

Synaptonemal complexes analysis was performed using electron microscopy on surface-spread spermatocytes of a bull heterozygous for the 4;8 Robertsonian translocation. In 19 cells examined, the longest autosomal complex showed kinetochores in a central position whereas the remaining autosomal complexes showed terminal kinetochores. Synapsis in the trivalent appeared complete in all cells, and the trivalents usually showed a CIS configuration. The arm ratio varied from 1.05 to 2.04 with an average of 1.32 +/- 0.43. Out of 47 cells showing X-Y bivalents, 34 showed a small synaptonemal complex at one extremity of the X chromosome, and an unstained gap in the Y chromosome. There was no association between the X-Y bivalent and the trivalent. The absence of association would explain the normal spermatogenesis noted in this bull, in contrast to human and mouse carriers of translocations which show impaired spermatogenesis due to the association between the rearranged chromosomes and the sex vesicle. Further studies involving bulls carrying one or more Robertsonian translocations are needed to determine whether this absence of association is a constant feature in cattle.  相似文献

The highest specific activity of beta-N-acetylglucosaminidase (beta-NAG) was found in the different parts of the epididymis, where the activity seemed to be partly in secretory and partly in non-secretory, tissue-bound form. Epididymal spermatozoa also contained moderate beta-NAG activity. The beta-NAG was separated by chromatofocussing and anion exchange chromatography with HPLC into multiple forms with distinct pI values (8.0-4.0). The cauda epididymidis, ampulla and the seminal vesicles formed the major secretory sources of the high beta-NAG activity in bull seminal plasma. The major secretory forms of beta-NAG in caput and cauda epididymidis showed distinct elution profiles. In the fractionation with gel filtration on Sepharose 6B, the beta-NAG activities derived from bull testis and caput epididymidis had smaller molecular weights than did the secretory enzymes in seminal plasma, seminal vesicle secretion and cauda epididymidis. Maximum activity of all beta-NAG isoenzymes was observed at pH 5.0. They were almost totally inactivated at 60 degrees C and about 75-80% of the activity was lost at 55 degrees C. All the isoenzymes were strongly inhibited by thiol reagents but not with other metal ions and chelating agents. Histochemical studies showed a strong granular (lysosomal) reaction for beta-NAG in basal cells and basal parts of the principal cells in all but the initial segment of the epididymis. An apical (secretory) reaction was prominent in the distal caput and corpus as well as in distal cauda. After the distal caput the luminal sperm mass became increasingly mixed with a beta-NAG-positive material. The epithelial cells of the ampulla and seminal vesicle displayed a moderate apical (secretory) reaction.  相似文献
