采用快速制备液相色谱从库拉索芦荟中高效分离制备10-羟基芦荟大黄素苷A和B。以库拉索芦荟药材粉甲醇提取物为原料,采用快速制备液相色谱,EYELA柱(300 mm×20 mm i.d.,20~45μm),甲醇-水为流动相(35∶65,v/v)等度洗脱,流速10 mL/min,检测波长356 nm,对芦荟样品进行分离制备,得到2种化合物单体,经UV、旋光度、HRMS和NMR鉴定,HPLC测定纯度,2个化合物分别为10-羟基芦荟大黄素苷B(98.9%)和10-羟基芦荟大黄素苷A(98.2%)。该方法简便、快速,所得产物纯度较高,可用于对照品的制备和药理毒理活性研究。  相似文献

在对桔梗总皂苷粗分的基础上,利用制备高效液相色谱分离制备桔梗单体皂苷,共分离得到四个化合物,经波谱法分别鉴定为platycodinD(1),platycoside E(2),deapio-platycoside E(3),未知化合物(4,鉴定中),经HPLC分析,纯度均大于98%.对比以前的分离手段,该方法分离制备桔梗单体皂苷工艺简单,效率高,为在桔梗中寻找新的天然活性物质提供了新方法。  相似文献

本实验建立了一种新型的利用高正交性的二维制备型高效液相色谱系统分离强极性动物药多肽的方法。本文以塞隆骨水提取物为研究对象,以亲水性C18AQ制备型高效色谱柱为第一维分离柱,首先在一维分离中将目标混合物分成若干组份;然后以C18MP制备型高效液相色谱柱为第二维色谱分离柱,将第一维分离后得到的组份纯化为单体化合物。本研究最终得到5个塞隆骨单体化合物,化合物纯度均超过98%。经Nano-LCESI-MS/MS鉴定和搜库分析,这些多肽的序列分别为:KTAILVKE、RGAPQDQE、LVGPGAPGR、GFAGD和KPQWHP。此研究方法速度快、效率高且重复性好,可以对类似的研究提供借鉴。  相似文献

采用大孔吸附树脂-制备液相色谱联用技术分离制备杜仲醇。以杜仲叶为原料,采用超声波辅助提取,提取液经过正丁醇萃取、D101大孔吸附树脂柱分离纯化后,再通过反相半制备液相色谱分离,以V(甲醇):V(水)=15∶85为流动相进行洗脱,制备得到杜仲醇单体。采用紫外光谱(UV)、红外光谱(IR)、核磁共振波谱(NMR)及液相色谱-质谱联用(LC-MS)等方法对所得单体进行了结构验证。液相色谱法分析检测表明制备所得杜仲醇纯度达95.12%。  相似文献

本文首次采用高速逆流色谱结合高效液相色谱的方法对桂枝正丁醇相进行分离纯化。首先,以石油醚-乙酸乙酯-甲醇-水(8∶2∶6∶4,v/v)为高速逆流色谱溶剂系统,将桂枝正丁醇萃取相分为两个馏分,然后结合制备高效液相,共分离得到4个高纯度化合物。通过核磁共振波谱鉴定其化学结构,分别为香豆素(1)、反式-邻甲氧基桂皮酸(2)、桂皮酸(3)、反式-桂皮醛(4),这四种化合物纯度经高效液相检测均大于95%。该方法简便、快速、节省溶剂,可以对桂枝正丁醇相进行快速有效的分离纯化,具有较好的实用价值,为桂枝资源的进一步开发应用提供了技术和物质支持。  相似文献

鹅膏菌肽类毒素是蘑菇中毒导致死亡的最主要因素,同时也是生物学研究领域的一种重要工具试剂,但由于其结构相似,分离制备单体化合物比较困难。通过4种大孔吸附树脂对致命鹅膏Amanita exitialis子实体中主要肽类毒素组分的静态吸附与解吸附性能比较,结果表明XAD16的吸附能力最强,同时也有较强的解吸附能力,但XAD16柱层析梯度洗脱时对毒素分离效果差,表明XAD16只具有吸附富集鹅膏肽类毒素的特性但不适于分离。经XAD16柱层析富集的毒素成分,通过葡聚糖凝胶sephadex LH20柱层析分离,结果表明sephadex LH20柱层析对鹅膏肽类毒素具有较好的分离效果,经过2次sephadex LH20柱层析,获得了5个纯度达50%–80%的鹅膏肽类毒素单体化合物,进一步利用半制备高效液相色谱系统(HPLC)纯化,分离得到了7个纯度达95%以上的鹅膏肽类毒素单体,其中5个经质谱分析鉴定为:α‐鹅膏毒肽(α‐amanitin)、β‐鹅膏毒肽(β‐amanitin)、脱氧二羟毒伞素(desoxoviroidin)、羧基三羟鬼笔毒肽(phallisacin)和羧基二羟鬼笔毒肽(phallacidin)。  相似文献

采用硅胶柱层析、凝胶LH-20柱层析和半制备高效液相色谱技术对伯氏致病杆菌SN52的次生代谢产物进行分离纯化,得到6个单体化合物。通过波谱综合解析和文献数据对照方法对所分离到的化合物进行结构鉴定。结果显示,均为吲哚类化合物,其中5个为已知化合物,鉴定为:xenocyloins A(1)、xenocyloins B(2)、xenocyloins C(3)、xenocyloins D(4)、xenocyloins E(5);1个为新化合物,命名为xenocyloins F(6)。  相似文献

不同类型喀斯特植物的荧光特征及抗旱性比较   总被引:1,自引:0,他引:1  
利用叶绿素荧光技术观测了五个不同类型的喀斯特植物翅荚香槐、大盔凤仙、红背叶、牛耳朵和青冈栎在不同作用光强下的光能利用特征,并且对这五种类型植物在PEG诱导水分胁迫下的抗旱性作了比较。结果表明,随着作用光强的增加,这五种植物的光化学反应能力(qP)逐渐降低,非光化学耗散作用(NPQ)明显增加,同时PSⅡ有效光化学效率(Fv′/Fm′)随之下降,导致PSⅡ电子传递量子效率(ΦPSⅡ)也明显下降,但在相同作用光强下这五个类型植物的荧光特征无明显差异。在PEG诱导水分胁迫的实验中,牛耳朵、青冈栎在PEG处理后Fv/Fm变化不明显,而红背叶Fv/Fm值下降程度最大,其次为翅荚香槐、大盔凤仙。由此推测,牛耳朵和青冈栎的耐干旱能力最强,红背叶抗旱能力最弱,翅荚香槐、大盔凤仙居中。  相似文献

The aqueous extract of Verbascum mucronatum Lam. along with its fractions and secondary metabolites were assessed for their antioxidant, acetylcholinesterase (AChE), and butyrylcholinesterase (BChE) inhibitory activities. The antioxidant activity was evaluated by three methods: as 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferrous ion-chelating effect, and ferric-reducing antioxidant power (FRAP) tests. The AChE activity was determined by the Ellman method using an ELISA microplate reader. Phytochemical investigations revealed the presence of four iridoid glucosides, ajugol (1), aucubin (2), lasianthoside I (3) and catalpol (4), two saponins, ilwensisaponin A (5) and C (6), and a phenylethanoid glycoside, verbascoside (7), in Verbascum mucronatum. Their structures were elucidated by spectral techniques. The aqueous extract and fractions including the phenylethanoid glycoside 7 showed DPPH scavenger effect and had the best FRAP. Besides these results, one of the phenylethanoid fractions displayed the highest ferrous ion-chelating effect. While only 7 was found to possess moderate AChE inhibition, the extract, fractions, and all other tested compounds did not inhibit AChE and BChE.  相似文献

In our search for bioactive compounds from the whole plant of Picriafelterrae Lour., three new phenylethanoid glycosides, picfeosides A-C (1-3), along with five known phenylethanoid glycosides,namely wiedemannioside (4), acteoside (5), acteoside isomer (6), cis-acteoside isomer (7), and cis-acteoside (8), were isolated using several chromatographic purification steps, including semipreparation HPLC on RP-18. Two new phenylethanoid glycosides integrifoliosides A (2) and B (3), along with a known phenylethanoid glycoside alyssonoside (1) and a flavone glucoside chrysoeriol-7-O-beta-D-glucopyranoside (4) were isolated from the aerial parts of Phlomis integrifolia. A new nuatigenin-type steroidal saponin, multifidoside (2), was isolated from the aerial parts of Veronica fuhsii and V multifida and its structure was identified as 3-O-([alpha-L-rhamnopyranosyl-(1-->2glu)]-[beta-D-glucopyranosyl-(1-->4rha)-alpha-L-rhamnopyranosyl (1-->4glu)]-beta-D-glucopyranosyl]nuatigenin 26-O-beta-D-glucopyranoside. Normal-phase column chromatography followed by semi-preparative reversed-phase HPLC has been used to isolate, from the rhizomes of Alpinia officinarum, five diarylheptanoids identified as 5-hydroxy-7-(4"-hydroxy-3"-methoxyphenyl)-1-phenyl-3-heptanone, 5-methoxy-7-(4"-hydroxy-3"-methoxyphenyl)-1-phenyl-3-heptanone, 7-(4"-hydroxyphenyl)-1-phenylhept-4-en-3-one, 7-(4"-hydroxy-3"-methoxyphenyl)-1-phenyl-hept-4-en-3-one, 1,7-diphenylhept-4-en-3-one. The levels of these five diarylheptanoids in root material were determined quantitatively by HPLC with UV detection and the assay methods so developed were simple, rapid and accurate. Four of the diarylheptanoids could also be detected by HPLC with electrochemical detection (ECD) in the oxidative mode, and ECD was found to have a higher sensitivity than photodiode array detection.  相似文献

运用硅胶、Sephadex LH-20、ODS柱层析及半制备高效液相色谱技术对一引种虎眼万年青属植物Ornithogalum thyrsoides的化学成分进行研究,从其乙醇提取物的乙酸乙酯部分分离得到7个化合物.通过MS、NMR等波谱技术确定其结构分别为(23S,24S,25S)-23,24-dihydroxyruscogenin- 1-O-α-L-rhamnopyranosyl(1 →2)-α-L-arabinopyranoside(1),(24S,25S)-3β,24-dihydroxyspirost-5 -en- 1β-yl O-α -L-rhamnopyranosyl-(1→2)-α-L-arabinopyranoside(2),β-胡萝卜苷(3)β-谷甾醇(4),山奈酚-4′-O-β-D-吡喃葡萄糖苷(5),千层纸素A(6)和汉黄芩素(7).其中,化合物1、5~7为首次从该属植物中分离得到.体外药理活性结果显示这7个化合物对HCT-8、Bel-7402、BGC-823、A549及A2780实体瘤细胞株无明显细胞毒活性.  相似文献
