The changes in amino acid concentrations and transsulfuration enzyme activities in liver were investigated after 4-week fed on 23% casein diet (control group) and 5% casein diet without (protein-calorie malnutrition, PCM group) or with (PCMC group) oral administration of cysteine, 250 mg/kg (twice daily, starting from the fourth week) using rats as an animal model. By supplementation with cysteine in PCM rats (PCMC group), cysteine level was elevated almost close to the control level, and glutathione (GSH), aspartic acid and serine levels were restored greater than the control levels. The measurement of transsulfuration enzyme activities exhibited that gamma-glutamylcysteine ligase (gamma-GCL) activity was up-regulated in rats with protein restriction (PCM group), and cysteine supplementation (PCMC group) down-regulated to the control level. One-week supplementation of cysteine (PCMC group) significantly down-regulated the cysteine sulfinate decarboxylase activity. These results indicate that the availability of sulfur amino acid(s) especially cysteine appears to play a role in determining the flux of cysteine between cysteine catabolism and GSH synthesis.  相似文献

Atomic absorption spectrophotometry was employed in an evaluation of zinc, copper and iron as trace elements in serum and erythrocytes from 56 children with protein-calorie malnutrition at different stages (early kwashiorkor, kwashiorkor and marasma). Iron and zinc concentrations showed a decreasing pattern through the various stages of the P.C.M. Significant changes are present in zinc values. The role of these trace elements in the P.C.M. is shortly discussed.  相似文献

Subperiosteal and medullary cavity diameters of 91 Guatemalan boys hospitalized with a diagnosis of protein-calorie malnutrition show a slight but significant increase in total width but a marked reduction at the endosteal surface, and in cortical area and percent cortical area, indicative of continuing subperiosteal apposition and a dramatic excess of endosteal resorption.  相似文献

The quantitative ultrastructural study of neocortical synaptic junctions has been performed during rehabilitation after protein-caloric deficiency. It has been shown that simple food rehabilitation does not lead to absolute restoration of synaptic junctions. However, food rehabilitation with carnitine restores the ultrastructure of synaptic junctions, with the length of the active zones and the number of spine apparatus cisterns exceeding the control values. Nevertheless, synaptic cleft width and postsynaptic density remain lower than in the control group, both after simple food rehabilitation and food rehabilitation with carnitine.  相似文献

Effects of cysteine on the pharmacokinetics of clarithromycin were investigated after intravenous administration of the drug at a dose of 20 mg/kg to control rats (4-week fed on 23% casein diet) and rats with PCM (protein-calorie malnutrition, 4-week fed on 5% casein diet) and PCMC (PCM treated with 250 mg/kg for oral cysteine twice daily during the fourth week). Clarithromycin has been reported to be metabolized via hepatic microsomal cytochrome P450 (CYP) 3A4 to 14-hydroxyclarithromycin (primary metabolite of clarithromycin) in human subjects. It has also been reported that in rats with PCM, CYP3A23 level decreased to 40-50% of control level, but decreased CYP3A23 level in rats with PCM completely returned to control level by oral cysteine supplementation (rats with PCMC). Human CYP3A4 and rat CYP3A23 proteins have 73% homology. In rats with PCM, the area under the plasma concentration-time curve from time zero to time infinity, AUC (567, 853 and 558 microg min/ml for control rats and rats with PCM and PCMC, respectively) and percentage of clarithromycin remaining after incubation with liver homogenate (69.6, 83.9 and 71.7%) were significantly greater than those in control rats and rats with PCMC. Moreover, in rats with PCM, the total body clearance, CL (35.3, 23.4 and 35.8 ml/min/kg), nonrenal clearance, CL(NR) (21.3, 15.2 and 24.1 ml/min/kg) and maximum velocity for the disappearance of clarithromycin after incubation with hepatic microsomal fraction, V(max) (351, 211 and 372 pmol/min/mg protein) were significantly slower than those in control rats and rats with PCMC. However, above mentioned each parameter was not significantly different between control rats and rats with PCMC. The above data suggested that metabolism of clarithromycin decreased significantly in rats with PCM as compared to control due to significantly decreased level of CYP3A23 in the rats. By cysteine supplementation (rats with PCMC), some pharmacokinetic parameters of clarithromycin (AUC, CL, CL(NR) and V(max)) were restored fully to control levels because CYP3A23 level was completely returned to control level in rats with PCMC.  相似文献

The effects of protein-calorie malnutrition (PCM) on heart structure and function are not completely understood. We studied heart morphometric, functional, and biochemical characteristics in undernourished young Wistar rats. They were submitted to PCM from birth (undernourished group, UG). After 10 wk, left ventricle function was studied using a Langendorff preparation. The results were compared with age-matched rats fed ad libitum (control group, CG). The UG rats achieved 47% of the body weight and 44% of the left ventricular weight (LVW) of the CG. LVW-to-ventricular volume ratio was smaller and myocardial hydroxyproline concentration was higher in the UG. Left ventricular systolic function was not affected by the PCM protocol. The myocardial stiffness constant was greater in the UG, whereas the end-diastolic pressure-volume relationship was not altered. In conclusion, the heart is not spared from the adverse effects of PCM. There is a geometric alteration in the left ventricle with preserved ventricular compliance despite the increased passive myocardial stiffness. The systolic function is preserved.  相似文献

The structure of the anterior pituitary gland of rats with protein-calorie malnutrition was studied at the light and electron microscopic levels. Male Sprague-Dawley rats were fed a low-protein diet from 20 to 50 days of age. The hypophyses from these animals, along with those from age-matched controls, were then removed and fixed for light microscopic immunocytochemical analyses, using anti-rat PRL or anti-ovine LH beta serum, or for routine ultrastructural study. The overall number of mammotrophs and gonadotrophs appeared to be unaffected by the deficiency in dietary protein. However, the nuclei, in both cell types, were significantly smaller in the malnourished rats. Cytoplasmic volume was also apparently less than in control rats, leading to the "crowding" of adjacent cell types. Additionally, in the malnourished rats, the mammotrophs assumed a less polygonal profile, while the gonadotrophs, which were more irregularly shaped in the central zone, rarely displayed the negative image of a Golgi complex. Ultrastructurally, the mammotrophs in the experimental rats had a less extensively developed granular endoplasmic reticulum and Golgi complex. The number of secretary granules was not different; however, their size was markedly less and their shape was round to ovoid, rather than pleomorphic. Two types of gonadotrophs were distinguished, based on the classification of Kurosumi et al. ('76). The type-I cell (former "FSH" cell) had a smaller Golgi complex, less vesicular endoplasmic reticulum and smaller secretory granules. The major alteration in the type-II gonadotroph (former "LH" cell) was the smaller size of the secretory granules. The morphological changes found in this study support our previous physiological observations (Herbert, '80), which demonstrate that prolactin and gonadotrophin secretion are severely impaired in rats suffering from protein-calorie malnutrition.  相似文献

The effect of pre- and postnatal undernutrition, produced according to the method of Chow and Lee (3), on the rate of protein synthesis in the brains of rats 11, 21, 34 and 90 days of age was studied by measuring the incorporation ofl-[14C]valine in vivo andl-[3H]lysine in vitro. Both in vivo and in vitro experiments were performed with high concentration of the precursor to decrease the effects of pool variations and protein degradation. Particular interest was given to the effects of this form of early protein-calorie malnutrition (PCM) on neuronal and glial cells which were isolated from the brains by gradient centrifugation. Brain protein synthesis measured in vivo which showed a peak at 21 days in both animal series, was depressed by PCM at 11 days but stimulated at 34 days of animal age. Small effect was observed in the 90-day-old animals. A similar response as in whole brain was seen for neuronal cells, while glial cells showed a different reaction. Studies of in vitro protein synthesis did not reveal appreciable effects of undernutrition in whole brain. Both neuronal and glial cells showed a moderate but not statistically significant elevation of protein synthesis in animals subjected to early PCM.  相似文献

The effect of early protein-calorie malnutrition (from birth to the age of 55 days) followed by nutritional rehabilitation (from the 58th day) on sexual behaviour was studied in male rats aged about 125 days. The sexual stimulation conditions on the part of the oestrus female were made as optimal as possible and were fully controlled. About half the malnourished males displayed precopulatory and copulatory behaviour, while in the control group these values were almost 100%. When malnourished males copulated, their copulatory performance did not differ very greatly from that of the well-nourished controls. The other experimental males displayed no signs of sexual behaviour during the testing period. Exposure of the males to the scent of a female in the latter's absence greatly stimulated their interest in the odour (sniffing the floor of the experimental box) in both the control and the malnourished animals which afterwards copulated. The results indicate that the sexual behaviour of males subjected to protein-calorie deficiency in early ontogenesis is at the very least delayed, if not completely suppressed.  相似文献

The effect of a low protein-calorie diet (restricted diet) on cellular growth and on RNA metabolism in Wistar rat liver has been studied. Experimentation was carried out over 30 days and the comparisons were made against well-nourished group (10% protein, controls). Liver weight and hepatic proteins dropped significantly in malnourished rats. Both rate of DNA and number of nuclei were unchanged. However, protein/DNA and liver weight/number of nuclei ratios decreased, which led to an atrophy phenomenon. DNase specific activity however, was not modified. Liver RNA content together with RNase activity dropped in deficient rats. Protein synthesis capacity (RNA/protein) did not change. These results suggest that restricted diet leads to a lower hepatocyte size with a decreased rat of RNA turnover.  相似文献

The estimation of free energy differences from computer simulation of macromolecular systems is important for rational strategies for drug design and for protein engineering. As an example of one mutation, we have studied the free energy change resulting from the conversion of a polar group (OH) to an apolar group (CH3) in aqueous solution. We have estimated the effect of various local environments on the magnitude of the free energy difference and find that significant environmental effects are found. The effects of cysteine on the pharmacokinetics and pharmacodynamics of azosemide were investigated after intravenous (10 mg/kg) and oral (20 mg/kg) administration to male Sprague-Dawley rats fed on 23% protein diet (control rats), and 5% protein diet with (rats with PCMC) or without (rats with PCM) oral cysteine (250 mg/kg, twice daily for the fourth week) for 4 weeks. After intravenous administration to rats with PCMC, some pharmacokinetic parameters restored fully or more than the level of control rats; the time-averaged nonrenal clearance (2.70 versus 2.32 ml/min/kg) and apparent volume of distribution at steady state (160 versus 189 ml/kg) were comparable to those in control rats, however, the terminal half-life (34.7 versus 57.2 min) and mean residence time (73.3 versus 99.3 min) were significantly shorter, area under the plasma concentration-time curve from time zero to time infinity (AUC, 1930 versus 2680 microg min/ml) was significantly smaller, and time-averaged renal (2.24 versus 1.21 ml/min/kg) and total body (CL, 4.98 versus 3.65 ml/min/kg) clearances were significantly faster than those in control rats. This could be mainly due to significantly faster renal clearance and at least partly due to increased cytochrome P450 1A2 activity by cysteine supplementation. After intravenous administration to rats with PCMC, the total amount of 8-hr urinary excretion of unchanged azosemide was significantly greater (457 versus 305 microg/g body weight), however, the 8-hr urine output (15.3 versus 31.1 ml/g kidney) was not significantly different between control rats and rats with PCMC. This could be due to the fact that urine output seemed to reach an upper plateau from 10 mg/kg dose of azosemide in rats.  相似文献
