Aims

Bacteria possessing ACC deaminase activity reduce the level of stress ethylene conferring resistance and stimulating growth of plants under various biotic and abiotic stresses. The present study aims at isolating efficient ACC deaminase producing PGPR strains from the rhizosphere of rice plants grown in coastal saline soils and quantifying the effect of potent PGPR isolates on rice seed germination and seedling growth under salinity stress and ethylene production from rice seedlings inoculated with ACC deaminase containing PGPR.

Methods

Soils from root region of rice growing in coastal soils of varying salinity were used for isolating ACC deaminase producing bacteria and three bacterial isolates were identified following polyphasic taxonomy. Seed germination, root growth and stress ethylene production in rice seedlings following inoculation with selected PGPR under salt stress were quantified.

Results

Inoculation with selected PGPR isolates had considerable positive impacts on different growth parameters of rice including germination percentage, shoot and root growth and chlorophyll content as compared to uninoculated control. Inoculation with the ACC deaminase producing strains reduced ethylene production under salinity stress.

Conclusions

This study demonstrates the effectiveness of rhizobacteria containing ACC deaminase for enhancing salt tolerance and consequently improving the growth of rice plants under salt-stress conditions.  相似文献

A variety of plants growing on metalliferous soils accumulate metals in their harvestable parts and have the potential to be used for phytoremediation of heavy metal polluted land. There is increasing evidence that rhizosphere bacteria contribute to the metal extraction process, but the mechanisms of this plant–microbe interaction are not yet understood. In this study ten rhizosphere isolates obtained from heavy metal accumulating willows affiliating with Pseudomonas, Janthinobacterium, Serratia, Flavobacterium, Streptomyces and Agromyces were analysed for their effect on plant growth, Zn and Cd uptake. In plate assays Zn, Cd and Pb resistances and the ability of the bacteria to produce indole-3-acetic acid (IAA), 1-amino-cyclopropane-1-carboxylic acid deaminase (ACC deaminase) and siderophores were determined. The isolates showed resistance to high Zn concentrations, indicating an adaptation to high concentrations of mobile Zn in the rhizosphere of Salix caprea. Four siderophore producers, two IAA producers and one strain producing both siderophores and IAA were identified. None of the analysed strains produced ACC deaminase. Metal mobilization by bacterial metabolites was assessed by extracting Zn and Cd from soil with supernatants of liquid cultures. Strain Agromyces AR33 almost doubled Zn and Cd extractability, probably by the relase of Zn and Cd specific ligands. The remaining strains, immobilized both metals. When Salix caprea plantlets were grown in γ-sterilized, Zn/Cd/Pb contaminated soil and inoculated with the Zn resistant isolates, Streptomyces AR17 enhanced Zn and Cd uptake. Agromyces AR33 tendentiously promoted plant growth and thereby increased the total amount of Zn and Cd extracted from soil. The IAA producing strains did not affect plant growth, and the siderophore producers did not enhance Zn and Cd accumulation. Apparently other mechanisms than the production of IAA, ACC deaminase and siderophores were involved in the observed plant–microbe interactions.  相似文献

We report the presence of ACC deaminase in Methylobacterium fujisawaense and its lowering of ethylene levels and promotion of root elongation in canola seedlings under gnotobiotic conditions. To test a part of the previous model proposed for ACC deaminase producing bacteria with Methylobacterium, ACC levels and various enzyme activities were monitored in canola. Lower amounts of ACC were present in the tissues of seeds treated with M. fujisawaense strains than in control seeds treated with MgSO4. Though the increased activities of ACC synthase in the tissue extracts of the treated seedlings might be due to bacterial indole-3-acetic acid, the amount of ACC was reduced due to bacterial ACC deaminase activity. The activities of ACC oxidase, the enzyme catalyzing conversion of ACC to ethylene remained lower in M. fujisawaense treated seedlings. This consequently lowered the ethylene in plants and prevented ethylene inhibition of root elongation. Our results collectively suggest that Methylobacterium commonly found in soils, as well as on the surfaces of leaves, seeds, and in the rhizosphere of a wide variety of plants could be better exploited to promote plant growth.  相似文献

In this study, bacteria were isolated from the rhizosphere and inside the roots and nodules of berseem clover plants grown in the field in Iran. Two hundred isolates were obtained from the rhizosphere (120 isolates), interior roots (57 isolates), and nodules (23 isolates) of clover plants grown in rotation with rice plants. Production of chitinase, pectinase, cellulase, siderophore, salicylic acid, hydrogen cyanide, indole acetic acid (IAA), 1-aminocyclopropane-1-carboxylate (ACC) deaminase, solubilization of phosphate, antifungal activity against various rice plant pathogen fungi, N2 fixation, and colonization assay on rice seedlings by these strains was evaluated and compared (endophytic isolates vs. rhizosphere bacteria). The results showed both the number and the ability of plant growth-promoting (PGP) traits were different between endophytic and rhizosphere isolates. A higher percentage of endophytic isolates were positive for production of IAA, ACC deaminase, and siderophore than rhizosphere isolates. Therefore, it is suggested that clover plant may shape its own associated microbial community and act as filters for endophyte communities, and rhizosphere isolates with different (PGP) traits. We also studied the PGP effect of the most promising endophytic and rhizosphere isolates on rice seedlings. A significant relationship among IAA and ACC deaminase production, the size of root colonization, and plant growth (root elongation) in comparison with siderophore production and phosphate solubilization for the isolates was observed. The best bacterial isolates (one endophytic isolate and one rhizosphere isolate), based on their ability to promote rice growth and colonize rice roots, were identified. Based on 16S rDNA sequence analysis, the endophytic isolate CEN7 and the rhizosphere isolate CEN8 were closely related to Pseudomonas putida and Pseudomonas fluorescens, respectively. It seems that PGP trait production (such as IAA, ACC deaminase) may be required for endophytic and rhizosphere competence as compared to other PGP traits in rice seedlings under constant flooded conditions. The study also shows that the presence of diverse rhizobacteria with effective growth-promoting traits associated with clover plants may be used for sustainable crop management under field conditions.  相似文献

The enzyme 1-aminocyclopropane-1-carboxylate deaminase catalyzes the degradation of 1-aminocyclopropane-1-carboxylic acid (ACC), the immediate precursor of the plant hormone ethylene, into α-ketobutyrate and ammonia. The enzyme has been detected in a limited number of bacteria and plays a significant role in sustaining plant growth and development under biotic and abiotic stress conditions by reducing stress-induced ethylene production in plants. We have screened 32 fluorescent Pseudomonas sp. isolated from rhizosphere and non-rhizosphere soils of different crop production systems for drought tolerance using polyethylene glycol 6000 (PEG 6000). Nine of these isolates were tolerant to a substrate metric potential of ?0.30 MPa (15 % PEG 6000) and therefore considered to be drought-tolerant. All of these drought-tolerant isolates were screened for ACC deaminase activity using ACC as the sole nitrogen source, and one (SorgP4) was found to be positive for ACC, producing 3.71?±?0.025 and 1.42?±?0.039 μM/mg protein/h of α-ketobutyrate under the non-stress and drought stress condition, respectively. The isolate SorgP4 also showed other plant growth-promoting traits, such as indole acetic acid production, phosphate solubilization, siderophore and hydrogen cyanide production. The ACC deaminase gene (acdS) from the isolate SorgP4 was amplified, and the nucleotide sequence alignment of the acdS gene showed significant homology with acdS genes of NCBI Genbank. The 16S rRNA gene sequencing analysis identified the isolate as Pseudomonas fluorescens. Both sequences have been submitted to the NCBI GenBank under the accession numbers JX885767 and KC192771 respectively.  相似文献

Sediment organic matter is a key stressor for submerged macrophyte growth, which negatively impacts the ecological restoration of lakes. Plant growth-promoting rhizobacteria (PGPR) were screened from the rhizosphere of submerged macrophytes and used due to their promoting effect on Vallisneria natans under a high sediment organic matter load. Root exudates were used as the sole carbon source to obtain the root affinity strains. Eight isolates were selected from the 61 isolated strains, based on the P solubilization, IAA production, cytokinins production and ACC deaminase activity. The analysis of the 16S rDNA indicated that one strain was Staphylococcus sp., while the other seven bacterial strains were Bacillus sp. They were all listed in low-risk groups for safety use in agricultural practices. The plant height significantly increased after inoculation with PGPR strains, with the highest rate of increase reaching 96%. This study provides an innovative technique for recovering submerged macrophytes under sediment organic matter stress.  相似文献

Plant growth promoting rhizobacteria affects the overall performance of plants by one or combination of mechanisms. However, little information is available on how ACC deaminase secreting bacteria enhance crop production. The present study aimed at identifying ACC deaminase producing and phosphate solubilizing bacterial strains and to assess their plant growth promoting activities. Additionally, the effect of two ACC deaminase positive bacterial strains Pseudomonas putida and Rhizobium leguminosarum on pea plants was determined to find a novel and compatible bacterial pairing for developing efficient inoculants for enhancing legume production and reducing dependence on chemical fertilizers. The isolated bacterial cultures were characterized biochemically and by 16S rRNA sequence analysis. The plant growth promoting activities was determined using standard microbiological methods. The impact of P. putida and R. leguminosarum, on pea plants was determined both in pots and in field environments. Of the total 40 bacterial strains, strain PSE3 isolated from Mentha arvenss rhizosphere and RP2 strain from pea nodules produced ACC deaminase, solubilized insoluble phosphate, synthesized indole acetic acid, ammonia, cyanogenic compounds, exopolysaccharides and had antifungal activity. The dual inoculation of P. putida strain PSE3 and R. leguminosarum strain RP2 had largest positive effect and markedly increased the growth, symbiotic characteristics, nutrient pool and quantity and quality of pea seeds. The measured parameters were further augmented when inoculated pea plants were grown in soils treated with urea or DAP. A significant variation in the measured parameters of pea plants was observed under both pot and field trials following microbial inoculation but the bacterial cultures did not differ significantly in growth promoting activities. The results suggest that ACC deaminase positive bacterial cultures endowed with multiple potential can be targeted to develop mixed inoculants for enhancing pea production and hence, to reduce dependence on synthetic fertilizers.  相似文献

Aims

The present study was planned to investigate the diversity of 1-aminocyclopropane-1-carboxylate (ACC) deaminase producing bacteria from the rhizosphere of wheat plants and subsequent evaluation of selected PGPR on growth enhancement of wheat seedlings under drought and saline conditions.

Methods

ACC deaminase producing plant growth promoting rhizobacteria (PGPR) were isolated from the rhizosphere of wheat and identified using 16S rRNA gene sequence analysis. Isolates were evaluated for various direct and indirect plant growth promoting (PGP) traits. Plant inoculation experiment was conducted using isolates IG 19 and IG 22 in wheat to assess their plant growth promotion potential under salinity and drought stress.

Results

Thirty-eight ACC deaminase producing PGPR were isolated which belonged to 12 distinct genera and falling into four phyla γ-proteobacteria, β-proteobacteria, Flavobacteria and Firmicutes. Klebsiella sp. was the most abundant genera and followed by Enterobacter sp. The isolates exhibited ACC deaminase activities ranging from 0.106–0.980 μM α- ketobutyrate μg protein?1 h?1. The isolates showed multiple PGP traits such as IAA production, phosphate, zinc, potassium solubilization and siderophore production. Enterobacter cloacae (IG 19) and Citrobacter sp. (IG 22) inoculated wheat seedlings showed notable increases in fresh and dry biomass under non-stress as well as under stressed condition.

Conclusion

To the best of our knowledge this is the first report of presence of ACC deaminase activity and other PGP traits from the genus Citrobacter and Empedobacter. Our finding revealed that the γ-proteobacteria group dominated the wheat rhizosphere. Plant inoculation with PGPR could be a sustainable approach to alleviate abiotic stresses in wheat plants. These native PGPR isolates could be used as potential biofertilizers for sustainable agriculture.

Fluorescent pseudomonads are among the most influencing plant growth-promoting rhizobacteria in plants rhizosphere. In this research work the plant growth-promoting activities of 40 different strains of Pseudomonas fluorescens and Pseudomonas putida, previously isolated from the rhizosphere of wheat (Triticum aestivum L.) and canola (Brassica napus L.) and maintained in the microbial collection of Soil and Water Research Institute, Tehran, Iran, were evaluated. The ability of bacteria to produce auxin and siderophores and utilizing P sources with little solubility was determined. Four strains of Wp1 (P. putida), Cfp10 (Pseudomonas sp.), Wp150 (P. putida), and Wp159 (P. putida) were able to grow in the DF medium with ACC. Thirty percent of bacterial isolates from canola rhizosphere and 33% of bacterial isolates from wheat rhizosphere were able to produce HCN. The results indicate that most of the bacteria, tested in the experiment, have plant growth-promoting activities. This is the first time that such PGPR species are isolated from the Iranian soils. With respect to their great biological capacities they can be used for wheat and canola inoculation in different parts of the world, which is of very important agricultural implications.  相似文献

Piper nigrum is an interesting plant to study the endophytic microbial factors affecting plant growth because of its unique features. Endophytic bacterial isolation from the plant resulted in the isolation of twelve bacterial isolates which were screened for various plant growth promoting properties like phosphate solubilization, ACC deaminase production, siderophore production etc. Interestingly, seven isolates were found to have IAA biosynthetic potential. Bacterial isolates with multiple plant growth promoting properties were studied for their growth promoting effect on Vigna radiata seedlings. This resulted in the identification of Klebsiella sp. (PnB 10) and Enterobacter sp. (PnB 11) as the isolates with excellent growth promoting properties. The results confirm promising applications of the endophytic bacterial isolates obtained in the study and also their possible growth promoting effect in P. nigrum.  相似文献

The ability of streptomycete actinomycetes to promote growth of tomato through the production of 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase was evaluated under gnotobiotic and greenhouse conditions. To achieve this, 64 isolates of Streptomyces spp. obtained from a tomato rhizosphere in the United Arab Emirates were initially selected for their ability to produce ACC deaminase as well as indole-3-acetic acid (IAA) and subsequently for their rhizosphere competence as root colonizers. Of the two selected ACC deaminase-producing isolates showing exceptional rhizosphere competence, S. filipinensis no. 15 produced both ACC deaminase and IAA, whilst S. atrovirens no. 26 did not produce IAA. Under greenhouse conditions, the application of S. filipinensis no. 15 or S. atrovirens no. 26 resulted in the reduction of the endogenous levels of ACC, the immediate precursor of ethylene, in both roots and shoots and increased plant growth. Plant growth promotion was most pronounced in the presence of S. filipinensis no. 15 compared to S. atrovirens no. 26. This relative superiority in performance shows the advantage conferred to S. filipinensis no. 15 due to its ability to produce both IAA and ACC deaminase. In comparison, an ACC deaminase-producing isolate of S. albovinaceus no. 41 which was neither rhizosphere-competent nor capable of producing IAA, failed to promote plant growth compared to S. filipinensis no. 15 or S. atrovirens no. 26 although the growth promotion obtained by S. albovinaceus no. 41 was significant compared to control. The application of S. globosus no. 8, which was not rhizosphere-competent and did not produce detectable levels of ACC deaminase or IAA did not promote plant growth. These results indicate the importance of rhizosphere competence. In conclusion I report the production of ACC deaminase by streptomycete actinomycetes and its ability to enhance plant growth through reduction in the in planta levels of endogenous ACC and the consequent lowering of endogenous ethylene levels in plant tissues.  相似文献

The application of biochar and plant-growth-promoting bacteria (PGPBs) in biocontrol soil-borne pathogens has garnered worldwide interest recently. However, how agricultural replanting disease is alleviated by a combination of biochar and PGPBs treatment (SYBB) remains largely unexplored. In this study, we investigated the beneficial effects of single biochar addition and the combination of biochar and PGPBs on alleviating replanting disease by altering the rhizosphere microbiome and metabolites. Our field experiment showed that the SYBB treatment had a better alleviating effect on replanting disease than the single biochar addition. The study indicated the dominant effect of deterministic processes on the bacterial community and of stochastic processes on the fungal community under biochar and PGPBs treatment. The combination of biochar and PGPBs tended to convert the stochastic processes of fungal community assembly into deterministic processes. We found SYBB treatment increased the abundance of potentially beneficial Pseudomonas, Lysobacter, Gemmatimonadetes and Nitrospira, and decreasing the abundance of potentially pathogenic Fusarium, Talaromyces and Fusarium oxysporum. Moreover, the SYBB treatment increased the abundances of carbohydrates, fatty acids and plant hormones, and decreased the abundances of amino acids in the rhizosphere soil. Co-occurrence network analysis indicated that SYBB treatment increased the connections within the microbial communities and drove the alteration of co-occurrence network among the microbial communities and metabolites, which increased positive correlations in bacteria-metabolite networks and decreased positive correlations in fungi-metabolite networks. Spearman correlation analysis showed the abundances of beneficial Streptomyces, Pseudomonas and Lysobacter were significantly and positively correlated to the metabolites most increased under SYBB treatment. The combination of biochar and PGPBs alleviated replanting disease by mediating the change of rhizosphere soil metabolites, and stimulating the proliferation of indigenous and beneficial soil microbes. The research results are intended to provide the basis for new strategies for green and sustainable remediation of soil-borne pathogens.  相似文献

A collection of 233 rhizobia strains from 30 different sites across Saskatchewan, Canada was assayed for 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity, with 27 of the strains displaying activity. When all 27 strains were characterized based on 16S rRNA gene sequences, it was noted that 26 strains are close to Rhizobium leguminosarum and one strain is close to Rhizobium gallicum. Polymerase chain reaction (PCR) was used to rapidly isolate ACC deaminase structural genes from the above-mentioned 27 strains; 17 of them have 99% identities with the previously characterized ACC deaminase structural gene (acdS) from R. leguminosarum bv. viciae 128C53K, whereas the other ten strains are 84% identical (864~866/1,020 bp) compared to the acdS from strain 128C53K. Southern hybridization showed that each strain has only one ACC deaminase gene. Using inverse PCR, the region upstream of the ACC deaminase structural genes was characterized for all 27 strains, and 17 of these strains were shown to encode a leucine-responsive regulatory protein. The results are discussed in the context of a previously proposed model for the regulation of bacterial ACC deaminase in R. leguminosarum 128C53K. An erratum to this article can be found at  相似文献

Aims: 1‐Aminocyclopropane‐1‐carboxylate (ACC) deaminase activity is an efficient marker for bacteria to promote plant growth by lowering ethylene levels in plants. We aim to develop a method for rapidly screening bacteria containing ACC deaminase, based on a colorimetric ninhydrin assay of ACC. Methods and Results: A reliable colorimetric ninhydrin assay was developed to quantify ACC using heat‐resistant polypropylene chimney‐top 96‐well PCR plates, having the wells evenly heated in boiling water, preventing accidental contamination from boiling water and limiting evaporation. With this method to measure bacterial consumption of ACC, 44 ACC‐utilizing bacterial isolates were rapidly screened out from 311 bacterial isolates that were able to grow on minimal media containing ACC as the sole nitrogen source. The 44 ACC‐utilizing bacterial isolates showed ACC deaminase activities and belonged to the genus Burkholderia, Pseudomonas or Herbaspirillum. Conclusions: Determination of bacterial ACC consumption by the PCR‐plate ninhydrin–ACC assay is a rapid and efficient method for screening bacteria containing ACC deaminase from a large number of bacterial isolates. Significance and Impact of the Study: The PCR‐plate ninhydrin–ACC assay extends the utility of the ninhydrin reaction and enables a rapid screening of bacteria containing ACC deaminase from large numbers of bacterial isolates.  相似文献

In this study we isolated and screened drought tolerant Pseudomonas isolates from arid and semi arid crop production systems of India. Five isolates could tolerate osmotic stress up to −0.73 MPa and possessed multiple PGP properties such as P-solubilization, production of phytohormones (IAA, GA and cytokinin), siderophores, ammonia and HCN however under osmotic stress expression of PGP traits was low compared to non-stressed conditions. The strains were identified as Pseudomonas entomophila, Pseudomonas stutzeri, Pseudomonas putida, Pseudomonas syringae and Pseudomonas monteilli respectively on the basis of 16S rRNA gene sequence analysis. Osmotic stress affected growth pattern of all the isolates as indicated by increased mean generation time. An increase level of intracellular free amino acids, proline, total soluble sugars and exopolysaccharides was observed under osmotic stress suggesting bacterial response to applied stress. Further, strains GAP-P45 and GRFHYTP52 showing higher levels of EPS and osmolytes (amino acids and proline) accumulation under stress as compared to non-stress conditions, also exhibited higher expression of PGP traits under stress indicating a relationship between stress response and expression of PGP traits. We conclude that isolation and screening of indigenous, stress adaptable strains possessing PGP traits can be a method for selection of efficient stress tolerant PGPR strains.  相似文献

Diazotrophic bacteria isolated from the rhizosphere of Chinese cabbage were assessed for other plant growth promoting characteristics viz., production of IAA, ethylene, ACC deaminase, phosphate solubilization, and gnotobiotic root elongation. Their effect on inoculation to Chinese cabbage was also observed under growth chamber conditions. A total of 19 strains that showed higher nitrogenase activity identified by 16S rRNA gene sequence analysis were found to be the members of the genera Pseudomonas and Agrobacterium belonging to α- and γ-Proteobacteria groups. These strains were also efficient in producing IAA and ACC deaminase though they produced low levels of ethylene and no phosphate solubilization. In addition, inoculation of selected diazotrophic bacterial strains significantly increased seedling length, dry weight, and total nitrogen when compared to uninoculated control. The colonization of crop plants by diazotrophic bacteria can be affected by many biotic and abiotic factors, and further studies are oriented towards investigating the factors that could influence the establishment of a selected bacterial community.  相似文献

Out of 62 bacterial isolates obtained from the mangrove Avicennia marina rhizosphere that grows along the Abu Dhabi coast, United Arab Emirates (UAE), an isolate of Pseudoalteromonas maricaloris (Wild type strain) (WT) produced relatively high levels of 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase in vitro. Application of this WT strain under greenhouse conditions to A. marina seedlings significantly (P < 0.05), reduced endogenous levels of ACC in the roots and shoots, and significantly (P < 0.05) increased the levels of in planta endogenous plant growth regulators (PGRs) including indole-3-acetic acid (IAA), indole-3-pyruvic acid (IPYA), putrescine (Put), spermidine (Spd) and spermine (Spm) in roots and shoots compared with control mangrove seedlings. WT application has also significantly (P < 0.05) increased photosynthetic pigment contents, photosynthetic carbon assimilation, plant water use efficiency and promoted mangrove seedlings growth characteristics including increased dry weight and length of roots and shoots, total leaf area and the number of the side branches compared with control mangrove seedlings. In comparison, an ACC deaminase non-producing mutant strain (NPM) failed to reduce endogenous levels of ACC in the roots and shoots and also failed to increase endogenous PGRs and photosynthetic pigments and did not promote seedling growth. Both WT and NPM strains were incapable of producing in vitro detectable levels of IAA, IPYA, Gibberellic acid (GA3), zeatin (Z), Put, Spd and Spm in the culture filtrates. This study demonstrated for the first time the ability of ACC deaminase-producing bacteria to promote mangrove growth under greenhouse conditions. P. maricaloris has potential as biological inoculants to promote the growth of mangrove seedlings in afforestation programs in nutrient impoverished sediments in hyper-saline coastal areas in the UAE.  相似文献
