The evolutionary conserved family of heat shock proteins (HSP) is responsible for protecting cells against different types
of stress, including oxidative stress. Although the levels of HSPs can be readily measured in blood serum, the levels of HSP70
in patients with different durations of diabetes have not been studied before. We quantified serum HSP70 levels in a healthy
control group (n = 36) and two groups of type 2 diabetic patients, defined as newly diagnosed diabetes (n = 36) and patients with diabetes duration of more than 5 years (n = 37). The clinical characteristics and biochemical parameters were evaluated in the studied population. We found that serum
HSP70 levels were significantly higher in patients with diabetes when compared with controls (p < 0.001) and it was higher in patients with disease for more than 5 years than in newly diagnosed patients (p < 0.001). Serum HSP70 was inversely correlated with fasting blood sugar in patients with diabetes for more than 5 years (r = −0.500, p = 0.002), positively correlated with the history of hypertension in newly diagnosed patients (p < 0.001), and positively correlated with age in patients with diabetes (r = 0.531, p = 0.001). Serum level of HSP70 is significantly higher in patients with diabetes and correlates with the duration of disease.
Higher HSP70 in prolonged diabetes versus newly diagnosed diabetes may be an indicator of metabolic derangement in the course

There is accumulating evidence that the metabolism of several trace elements is altered in diabetes mellitus and that these
nutrients might have specific roles in the pathogenesis and progress of this disease. The aim of present study was to compare
the level of essential trace elements, chromium (Cr), copper (Cu), iron (Fe), manganese (Mn), nickel (Ni), and zinc (Zn) in
biological samples (whole blood, urine, and scalp hair) of patients who have diabetes mellitus type 2 (n = 257), with those of nondiabetic control subjects (n = 166), age ranged (45–75) of both genders. The element concentrations were measured by means of an atomic absorption spectrophotometer
after microwave-induced acid digestion. The validity and accuracy was checked by conventional wet-acid-digestion method and
using certified reference materials. The overall recoveries of all elements were found in the range of (97.60–99.49%) of certified
values. The results of this study showed that the mean values of Zn, Mn, and Cr were significantly reduced in blood and scalp-hair
samples of diabetic patients as compared to control subjects of both genders (p < 0.001). The urinary levels of these elements were found to be higher in the diabetic patients than in the age-matched healthy
controls. In contrast, high mean values of Cu and Fe were detected in scalp hair and blood from patients versus the nondiabetic
subjects, but the differences found in blood samples was not significant (p < 0.05). These results are consistent with those obtained in other studies, confirming that deficiency and efficiency of
some essential trace metals may play a role in the development of diabetes mellitus. 相似文献

Excessive Zn but normal Cu and Mg in the staple food consumed by the people of Chandigarh (Union territory and capital of
Punjab and Haryana States of India) has been considered to be the major risk factor for the prevalence of obesity (33.15%)
and obesity-related diseases in this region. Therefore, in the present investigations, in obesity-related diseases, the status
of these minerals was estimated in their tissues, including hair, nails, and blood serum and urine, and compared with those
of normal subjects. They were grouped as: normal subjects in control Group A, middle-aged diabetics in Group DM, older diabetics in Group DO, and diabetics with osteoarthritis in Group D+ OA, osteoarthritis in Group OA and rheumatoid arthritis in Group RA, respectively.
The results evaluated in the order as: hair Zn, group D+OA>DM>OA>A (control)>RA>DO (p < 0.001); hair Cu, group A (control)>DM>OA>D+OA>DO>RA (p < 0.001); hair Mg, group A (control)>DM>OA>D+OA>RA>DO (p < 0.001, 0.01); hair Mn, group A (control)>RA>OA>D-OA>DM>DO (p < 0.001); nail Zn, group DM>D+OA>OA>A (control)>RA>DO (p < 0.001, 0.05); nail Cu, group A (control)>OA>DM>D+OA>RA>DO (p < 0.001); nail Mg, group A (control)>OA>DM>DO>D+OA >RA (p < 0.001); nail Mn, group A (control) >RA>OA>D+OA>DM>DO (p < 0.01); urine Zn, group DO>DM>D+OA>A (control)>RA>OA (p < 0.01); urine Cu, group RA>D+OA>DO>OA> DM>A (control) (p<0.001); urine Mg, group RA>OA>D+OA>DO>DM>A (control; p < 0.001); urine Mn, group DO>DM>OA>D+OA>RA>A (control; p < 0.001), respectively. The analysis of the mineral status in serum of diabetics further showed their highly significant
rise from lower mean age subgroup to higher mean age subgroup than their control counter parts (p < 0.001, 0.01, and 0.05) with coincident deficiencies of Cu, Mg, and Mn in their tissues. This study would be helpful considering
the status of minerals in these obesity-related diseases depending on the choice of the food consumed to improve the quality

The present study was conducted to investigate the effects of chromium histidinate (CrHis) against experimentally induced
type II diabetes and on chromium (Cr), zinc (Zn), selenium (Se), manganese (Mn), iron (Fe), and copper (Cu) in serum, liver,
and kidney of diabetic rats. The male Wistar rats (n = 60, 8 weeks old) were divided into four groups. Group I received a standard diet (12% of calories as fat); group II were
fed standard diet and received CrHis (110 mcg CrHis/kg body weight per day); group III received a high-fat diet (HFD; 40% of calories as fat) for 2 weeks and then
were injected with streptozotocin (STZ) on day 14 (STZ, 40 mg/kg i.p.; HFD/STZ); group IV were treated as group III (HFD/STZ)
but supplemented with 110 mcg CrHis/kg body weight per day. The mineral concentrations in the serum and tissue were determined
by atomic absorption spectrometry. Compared to the HFD/STZ group, CrHis significantly increased body weight and reduced blood
glucose in diabetic rats (p < 0.001). Concentrations of Cr, Zn, Se, and Mn in serum, liver, and kidney of the diabetic rats were significantly lower
than in the control rats (p < 0.0001). In contrast, higher Fe and Cu levels were found in serum and tissues from diabetic versus the non-diabetic rats
(p < 0.001). Chromium histidinate supplementation increased serum, liver, and kidney concentrations of Cr and Zn both in diabetic
and non-diabetic rats (p < 0.001). Chromium supplementation increased Mn and Se levels in diabetic rats (p < 0.001); however, it decreased Cu levels in STZ-treated group (p < 0.001). Chromium histidinate supplementation did not affect Fe levels in both groups (p > 0.05). The results of the present study conclude that supplementing Cr to the diet of diabetic rats influences serum and
tissue Cr, Zn, Se, Mn, and Cu concentrations. 相似文献

Hypokinesia (HK) induces electrolyte losses in electrolyte-deficient tissue, yet the mechanisms of electrolyte losses in electrolyte-deficient
tissue remain unknown. Mechanisms of electrolyte deposition could be involved. To determine the effect of prolonged HK on
potassium (K+) deposition were measured muscle K+ content and K+ losses. Studies were conducted on 20 physically healthy male volunteers during 30 days pre-experimental period and 364 days
experimental period. Subjects were equally divided into two groups: control subjects (CS) and experimental subjects (ES).
The CS group was run average distances of 9.8 ± 1.7 km day−1 and the ES group was walked average distances of 2.7 ± 0.6 km day−1. Muscle K+ content decreased (p < 0.05) and plasma K+ concentration, and K+ losses in urine and feces increased (p < 0.05) in the ES group compared to their pre-experimental level and the values in their respective CS group. Muscle K+ content, plasma K+ level, and urine and fecal K+ losses did not show any changes in the CS group compared to their pre-experimental values. The conclusion was that K+ losses in K+-deficient muscle of healthy subjects could have been attributable to the less efficient K+ deposition inherently to prolonged HK. 相似文献

The aim of this study was to determine the relationship between serum and cerebrospinal fluid (CSF) magnesium (Mg+2) levels, Glasgow Coma Scores (GCS), and 7-day mortality in acute stroke patients. Patients with acute ischemic or hemorrhagic
stroke arriving within the first 3 h of symptoms were included in the study. The control group consisted of healthy volunteers.
GCS was determined, and blood and CSF samples were taken in order to establish serum and CSF glucose, Mg+2, sodium, potassium, calcium, and chlorine levels. Mortality was recorded at 7 days after admission. CSF Mg+2 in the ischemic infarct group was significantly lower than in the control group (p = 0.006). CSF Mg+2 in the ischemic infarct patients with a GCS ≤ 8 were significantly lower (p = 0.002) than controls and in ischemic infarct patients with a GCS ≥9. In the ischemic stroke patients, CSF Mg+2 and GCS were significantly correlated (r = 55, p = 0.031). CSF Mg+2 levels in ischemic stroke patients who died within 7 days were significantly lower than controls, ischemic stroke patients
who survived, and hemorrhagic stroke patients who died (p = 0.002, p = 0.042, and p = 0.005, respectively). Low CSF Mg+2 levels in patients with acute ischemic stoke at admission predicted a higher 1-week mortality. 相似文献

Ellagic acid (EA) is a natural polyphenolic compound. Although, modulator effects of EA on copper (Cu) and zinc (Zn) levels
in some liver diseases have been reported in experimental animals, its effects in obstructive jaundice (OJ) has not been clarified.
We aimed to evaluate potential effects of EA on Cu and Zn levels in liver and serum of cholestatic rats. Forty Wistar albino
rats were equally divided into four groups. First group was used as controls. Second group received EA (60 mg−1 kg−1 day−1) for 8 days. Third was OJ group, and fourth group was OJ plus EA group. After 8 days, blood and liver samples were obtained.
Higher serum and liver Cu and lower serum and liver Zn levels were found in OJ group (p < 0.05) compared with other groups. However, these differences reached to significant levels for Cu in serum and for Zn in
lever. Higher serum copper levels were decreased, and lower liver Zn levels were increased by EA treatment in cholestatic
rats (p < 0.05). Also, higher Cu/Zn ratio in OJ group was decreased by EA treatment both in liver (p < 0.05) and in serum (p < 0.05). Significantly higher serum bilirubin, alkaline phosphatase, alanine aminotransferase, and aspartate aminotransferase
values were found in OJ and OJ + EA groups compared with the control and EA groups (p < 0.05). In conclusion, result of the current study indicated that ellagic acid has modulator effects on Cu and Zn levels

The purpose of the study was to determine the serum concentration of trace elements of panic disorder patients and to find
out the relationship between trace element levels and nutritional status or socio-economic factors. The study was conducted
among 54 panic disorder patients and 52 healthy volunteers. Patients were recruited from Bangabandhu Sheikh Mujib Medical
University by random sampling. Serum trace element concentrations were determined by flame atomic absorption spectroscopy
(for Mg, Zn, Ca, and Cu) as well as graphite furnace (for Mn). Data were analyzed by independent t test, Pearson’s correlation analysis, regression analysis, and ANOVA. The serum concentration of Mn, Zn, Ca, Cu, and Mg in
panic disorder patients were 0.37 ± 0.30, 0.67 ± 0.20, 99.91 ± 15.15, 0.83 ± 0.23, and 21.14 ± 3.72 mg/L, while those were
0.4163 ± 0.2527, 0.86 ± 0.3, 106.6073 ± 18.6531, 0.8514 ± 0.3646, and 21.37 ± 2.03 mg/L in control subjects, respectively.
The serum concentration of Zn decreased significantly (p = 0.001) in patient group. But the differences of the concentration of Mn, Ca, Cu, and Mg between patient and control group
were not significant (p = 0.522, p = 0.065, p = 0.800, and p = 0.712, respectively). Socio-economic data reveal that most of the patients were very poor and middle aged. Mean BMIs of
the control group (23.74 ± 2.71 kg/m2) and the patient group (22.62 ± 3.74 kg/m2) were within the normal range (18.5–25.0 kg/m2). There was no significant relationship between serum zinc level and BMI of patients (r = 0.038; p = 0.809). So the decreased level of serum zinc in panic disorder patients was not because of other reasons, but rather it
may provide a prognostic tool for the diagnosis and treatment of this disease. 相似文献

A cross-sectional study of 191 adult (>18 years) Bengalee male slum dwellers of Kolkata, India, was undertaken to study the
relationships of family income with body mass index (BMI) and chronic energy deficiency (CED). Results revealed that the mean
height, weight, and BMI of the subjects were 162.2 cm, 54.0 kg, and 20.5 kg/m2, respectively. The overall frequency of CED (BMI < 18.5 kg/m2) was 33.5%. Based on the World Health Organization classification, the prevalence of CED among this population was high (20–39%)
and thus the situation is serious. Overall, monthly family income (MFI) was significantly positively correlated (r = 0.18, p < 0.05) with BMI. Linear regression analyses showed that MFI had significant impact (p < 0.05) on BMI. The percent variation in BMI explained by MFI was 2.6%. Subjects belonging to the lowest family income group
(FIG) had the lowest mean BMI (19.5 kg/m2) and the highest rate of CED (46.6%) while those in the highest FIG had the largest mean BMI (21.4 kg/m2) and lowest rate of CED (23.1%). There was a significant FIG difference (F = 2.965, p < 0.05) in mean BMI. Moreover, there existed FIG differences (χ
2 = 7.54, p < 0.06) in CED rates. In conclusion, this study provided strong evidence that FIG was significantly associated with BMI and
the presence of CED. The rate of CED was high, indicating a serious situation. These findings may have severe public health
implications. It is recommended that immediate nutritional intervention programs be initiated among this population along

The authors, by means of a recently introduced method, evaluated the intraplatelet concentrations of magnesium in 45 normotensive
patients with type II diabetes mellitus, in 45 hypertensive diabetics and in 15 healthy controls. They also evaluated plasma
and erythrocyte concentrations of the cation through direct current plasma spectrometer. Both normotensive and hypertensive
diabetics showed a reduction in plasma, erythrocyte, and platelet concentrations of magnesium compared to controls. On the
contrary, no significant difference was found between hypertensive and normotensive diabetics with regard to plasma and erythrocyte
magnesium, whereas intraplatelet assay of the ion pointed out significantly lower concentrations of magnesium in hypertensive
compared to normotensive patients (56.4±9.0 vs 60.7±10.2 μg/108 cells-p<0.05). The authors believe that intraplatelet assay of magnesium may be the most reliable method for the evaluation of the
cation in hypertensive diabetics, probably because platelets share common features with smooth muscle cells, including the
α-2-adrenoceptor cyclase system and a coupling mechanism concerning the calcium-dependent contraction. 相似文献

The purpose of this research was to use inline real-time near-infrared (NIR) to measure the moisture content of granules manufactured
using a commercial production scale continuous twin-screw granulator fluid-bed dryer milling process. A central composite
response surface statistical design was used to study the effect of inlet air temperature and dew point on granule moisture
content. The NIR moisture content was compared to Karl Fischer (KF) and loss on drying (LOD) moisture determinations. Using
multivariate analysis, the data showed a statistically significant correlation between the conventional methods and NIR. The
R
2 values for predicted moisture content by NIR versus KF and predicted moisture values by NIR versus LOD were 0.94 (p < 0.00001) and 0.85 (p < 0.0002), respectively. The adjusted R
2 for KF versus LOD correlation was 0.85 (p < 0.0001). Analysis of the response surface design data showed that inlet air temperature over a range of 35–55°C had a significant
linear impact on granule moisture content as measured by predicted NIR (adjusted R
2 = 0.84, p < 0.02), KF (adjusted R
2 = 0.91, p < 0.0001), and LOD (adjusted R

The purpose of this study was to compare plasma leptin, plasma zinc, and plasma copper levels and their relationship in trained
female and male judo athletes (n = 10 women; n = 8 men). Blood samples were obtained 24 h after training to measure plasma zinc, copper, and leptin levels. Subjects presented
similar values to age (22 ± 2 years old), body mass index (24 ± 1 kg/m2), plasma zinc (17.2 ± 2 μmol/L), copper (12.5 ± 2 μmol/L), and leptin (5.6 ± 1.3 μg/L). However, height, total body mass,
lean mass, fat mass, and sum of ten-skinfold thickness were higher in male than female. Plasma leptin was associated with
sum of ten skinfolds in male (r = 0.91; p < 0.001) and female athletes (r = 0.84; p < 0.003). Plasma zinc was associated with leptin in males (r = 0.82; p < 0.05) while copper was associated with plasma leptin in females (r = 0.66; p < 0.05). Our results suggest that young judo athletes lost sex-related differences in leptin levels. Plasma zinc, plasma
copper, and energy homeostasis may be involved in regulation of plasma leptin. 相似文献

We investigated the effects of treadmill exercise performed regularly for 6 weeks on the levels of nerve growth factor (NGF),
tyrosine kinase A and p75 receptors, phosphatidylinositol 3-kinase (PI3-K), mitogen-activated protein kinase/extracellular
signal-regulated kinase (Erk) 1,2, cyclic AMP response element-binding protein (CREB), and caspase-3 in the soleus of rats
with streptozotocin (STZ)-induced diabetes. Thirty-two male Sprague–Dawley rats were divided into the following four groups:
(1) normal control group (NCG; n = 8), (2) normal exercise group (NEG; n = 8), (3) diabetes control group (DCG; n = 8), and (4) diabetes exercise group (DEG; n = 8). Diabetes was induced by intraperitoneal injection of STZ (55 mg/kg dissolved in 0.05 M citrate buffer, pH 4.5). Rats
were subjected to treadmill exercise 5 days a week for 6 weeks. The protein level of NGF significantly increased in the NEG
and DEG (p < 0.001), whereas the levels of tyrosine kinase A and p75 receptors significantly increased in the NEG (p < 0.001). The levels of t-PI3-K, p-PI3-K, and p-CREB, and the p-CREB/t-CREB ratio significantly increased in the NEG (p < 0.001, respectively). The p-PI3-K/t-PI3-K ratio significantly increased in the DEG (p < 0.001). The p-Erk1/t-Erk1 ratio significantly increased in the NEG (p < 0.001), whereas the p-Erk2/t-Erk2 ratio significantly decreased in the DCG and DEG (p < 0.001). The caspase-3 level significantly increased in the DCG compared with that in the DEG (p < 0.001). These results suggest that treadmill exercise increases NGF levels and accelerates p-PI3-K activation in order
to suppress apoptotic cell death in the soleus muscle of diabetic rats. 相似文献

The Wellcome Trust Case Control Consortium (WTCCC) genome-wide study found association of PTPN2 with three autoimmune diseases, among them is type 1 diabetes (T1D). This result was confirmed by a follow-up study that
pointed to new independent signals within the region. However, both studies were performed in patients with an early-onset
T1D. We aimed at replicating the previous results and studying the influence of these polymorphisms in the age at T1D debut.
We genotyped 439 T1D Spanish subjects (age at onset, 1 to 65 years) and 861 controls for two PTPN2 single nucleotide polymorphisms (SNPs), rs2542151 and rs478582, and studied the effect of both polymorphisms in age at onset
through stratified and continuous analyses. The frequency of rs2542151*G carriers was significantly higher in the early-onset
group compared with late-onset patients (p = 0.023) and with controls (OR = 1.61 [1.14–2.26]; p = 0.005). No significant differences were found between controls and late-onset patients. The log-rank chi-square test for
the Kaplan–Meier plots (carriers of susceptibility allele vs non carriers) was statistically significant (χ
1df2 = 4.485; p = 0.034), yielding an earlier disease debut for G carriers. The analysis of the SNP rs478582 did not reach statistical significance.
In summary, we replicate the association detected by the WTCCC and propose that the rs2542151*G allele confers risk to an

The purpose of the study was to determine the concentration of trace elements present in scalp hair sample of schizophrenic
patients and to find out the relationship between trace elements level and nutritional status or socioeconomic factors. The
study was conducted among 30 schizophrenic male patients and 30 healthy male volunteers. Patients were recruited from Bangabandhu
Sheikh Mujib Medical University by random sampling. Hair trace element concentrations were determined by flame atomic absorption
spectroscopy and analyzed by independent t test, Pearson’s correlation analysis, regression analysis, and analysis of variance (ANOVA). Mn, Zn, Ca, Cu, and Cd concentrations
of schizophrenic patients were 3.8 ± 2.31 μg/gm, 171.6 ± 59.04 μg/gm, 396.23 ± 157.83 μg/gm, 15.40 ± 5.68 μg/gm, and 1.14 ± 0.89 μg/gm
of hair sample, while those of control subjects were 4.4 ± 2.32 μg/gm, 199.16 ± 27.85 μg/gm, 620.9 ± 181.55 μg/gm, 12.23 ± 4.56 μg/gm,
and 0.47 ± 0.32 μg/gm of hair sample, respectively. The hair concentration of Zn and Ca decreased significantly (p = 0.024; p = 0.000, respectively) and the concentration of Cu and Cd increased significantly (p = 0.021; p = 0.000, respectively) in schizophrenic patients while the concentration of Mn (p = 0.321) remain unchanged. Socioeconomic data reveals that most of the patients were poor, middle-aged and divorced. Mean
body mass indices (BMIs) of the control group (22.26 ± 1.91 kg/m2) and the patient group (20.42 ± 3.16 kg/m2) were within the normal range (18.5−25.0 kg/m2). Pearson’s correlation analysis suggested that only Ca concentration of patients had a significant positive correlation
with the BMI (r = 0.597; p = 0.000) which was further justified from the regression analysis (R
2 = 44%; t = 3.59; p = 0.002) and one-way ANOVA test (F = 3.62; p = 0.015). A significant decrease in the hair concentration of Zn and Ca as well as a significant increase in the hair concentration
of Cu and Cd in schizophrenic patients than that of its control group was observed which may provide prognostic tool for the
diagnosis and treatment of this disease. However, further work with larger population is suggested to examine the exact correlation

The present study was conducted to evaluate the serum selenium levels in first-degree relatives of diabetic patients (FDR)
according to controls. Insulin resistance, serum lipid levels, inflammation markers, and blood pressure were also studied
in these patients. Serum levels of selenium in FDR were significantly lower than control group (74.65 ± 5.9 vs 88.7 ± 8.7 μg/dl,
p < 0.0001). HsCRP, HOMA-IR, insulin, homocysteine levels were significantly higher in FDR according to the control group (1.32 ± 0.9
vs 0.63 ± 0.4 mg/dL, p < 0.0001; 2.07 ± 0.84 vs 1.51 ± 0.69, p < 0.0001; 9.26 ± 3.8 vs 6.8 ± 2.98 μU/MI, p < 0.0001; 15.7 ± 7.4 vs 11.5 ± 5.1 μmol/L, p < 0.0001, respectively). There was significant correlation between selenium levels and hsCRP (r = − 0.450, p < 0.0001). There was also weak significant correlation also between HOMA-IR and selenium levels (r = −0.227, p = 0.003). There was a correlation between systolic blood pressure and BMI (r = 0.365, p < 0.0001). But there was no correlation between selenium levels and blood pressure or other parameters. HsCRP, HOMA-IR, homocysteine
levels in individuals with selenium levels < 80 μg/L (n = 78) was significantly higher than hsCRP HOMA-IR, homocysteine levels in individuals with selenium levels ≥ 80 (n = 91; 1.23 ± 0.98 vs 0.81 ± 0.76 mg/dL, p < 0.003; 1.99 ± 0.88 vs 1.64 ± 0.74, p < 0.005; 15.0 ± 7.6 vs 12.9 ± 5.7 μmol/L, p < 0.049, respectively). Objective Alpha hemoglobin-stabilizing protein (AHSP) inhibits the production of reactive oxygen species in various cells, including
erythrocytes. Reduced AHSP can mean reduced protection from stressors. Our objective was to investigate whether AHSP is involved
in the response to stress in pregnancy.
Study design Placentas were collected from normal term pregnancies (n = 10) and pregnancies complicated by HELLP (n = 10), intrauterine growth restriction (IUGR; n = 10) or fetal death (IUFD; n = 6). AHSP messenger RNA (mRNA) and protein were determined using real time quantitative polymerase chain reaction (PCR)
and Western blot, respectively. All statistical analyses were performed by using the GraphPad Prism Software. Differences
were considered significant at p < 0.05.
Results Placental AHSP mRNA level in HELLP (4.16E10−4 ± 1.77) and IUFD (4.19E10−4 ± 3.37) were significantly decreased compared with controls (28.47E10−4 ± 14.86; p < 0.01), whereas levels in the IUGR group (7.55E10−4 ± 6.4) showed a trend toward being lower but the difference did not reach statistical significance. Western blot analysis
results indicate a no significant increase of ASHP protein in the HELLP syndrome group and a significant decrease in the IUFD
group compared with controls. There was no significant difference between the IUGR and control groups.
Conclusion ASHP mRNA expression in the placenta is decreased in complicated pregnancies, and it may be involved in the pathogenic mechanisms
leading to the adverse pregnancy outcome.

Nickel (Ni) is an environmental pollutant towards which human exposure can be both occupational (mainly through inhalation)
and dietary (through water and food chain-induced bioaccumulation). The aim of this study was to investigate the effects of
short-term Ni-administration (as NiCl2, 13 mg/kg) on the adult rat whole brain total antioxidant status (TAS) and the activities of acetylcholinesterase (AChE),
Na+,K+-ATPase, and Mg2+-ATPase; in addition, the potential effect of the co-administration of the antioxidant L-cysteine (Cys, 7 mg/kg) on the above
parameters was studied. Twenty-eight male Wistar rats were divided into four groups: A (saline-treated control), B (Ni), C
(Cys), and D (Ni and Cys). All rats were treated once daily with intraperitoneal injections of the tested compounds, for 1-week.
Rats were sacrificed by decapitation and the above-mentioned parameters were measured spectrophotometrically. Rats treated
with Ni exhibited a significant reduction in brain TAS (-47%, p < 0.001, BvsA) that was efficiently limited by the co-administration of Cys (-4%, p > 0.05, DvsA; +83%, p < 0.001, DvsB), while Cys (group C) had no effect on TAS. The rat brain AChE activity was found significantly increased by
both Ni (+30%, p < 0.001, BvsA) and Cys (+62%, p < 0.001, CvsA), while it tended to adjust to control levels by the co-administration of Ni and Cys (+13%, p < 0.001, DvsA; −13%, p < 0.001, DvsB). The activity of rat brain Na+,K+-ATPase was significantly decreased by Ni-administration (−49%, p < 0.001, BvsA), while Cys supplementation could not reverse this decrease (-44%, p < 0.001, DvsA). The activity of Mg2+-ATPase was not affected by Ni-administration (−3%, p > 0.05, BvsA), but was significantly reduced when combined with Cys administration (−17%, p < 0.001, DvsA). The above findings suggest that Ni short-term in vivo administration causes a statistically significant decrease
in the rat brain TAS and an increase in AChE activity. Both effects can be, partially or totally, reversed to control levels
by Cys co-administration; Cys could thus be considered (for future applications) as a potential neuroprotective agent against
chronic exposure to Ni. The activity of Na+,K+-ATPase that was inhibited by Ni, could not be reversed by Cys co-administration. The matter requires further investigation
in order to fully elucidate the spectrum of the neurotoxic effects of Ni. 相似文献

This study was designed to understand the cellular mechanisms responsible for defects in the insulin-stimulated signal transduction pathway in a type 2 diabetic animal model. We examined the in vitro PC-1 phosphodiesterase activity and glucose uptake in adipose tissue of streptozotocin (STZ)-induced type 2 diabetic rats. The PC-1 activity was significantly increased in adipose tissue of diabetic rats (0.54 ± 0.08 nmol PNTP hydrolyzed/mg protein/min) compared with controls (0.29 ± 0.05 nmol PNTP hydrolyzed/mg protein/min, p < 0.05). Upon insulin stimulation (100 nM), glucose uptake in the adipose tissue of the controls (4.17 ± 1.28×10−8 μmol/mg/min) was significantly higher than that in the diabetic rats (1.26 ± 0.35×10−8; p < 0.05). These results suggest that elevated PC-1 phosphodiesterase activity and decreased glucose uptake in adipose tissues may be acquired characteristics contributing to the development of type 2 diabetes mellitus. 相似文献

Chromium deficiency is associated with impaired glucose tolerance (IGT) and dyslipidemia. Hence, the objective of the current
study was to investigate chromium status among Saudi men with and without established cardiovascular disease (CVD) and its
relationship to glucose tolerance, lipid profile and other established CVD risk factors. We measured serum and urine chromium
concentrations, fasted lipid profile, plasma glucose, and serum lipid peroxide in 130 Saudi men with an established history
of myocardial infarction and 130 age-matched controls without established CVD. Patients with established CVD had higher serum
triglycerides (p < 0.05) and plasma glucose (p < 0.0001) and lower serum and urinary chromium concentrations (p < 0.0001) than controls. Serum chromium was inversely correlated with plasma glucose among cases and controls (r = −0.189, p < 0.05 and r = −0.354, p < 0.00001, respectively). Plasma glucose (OR 1.127, CI 1.0–1.269, p < 0.05), serum chromium (OR 0.99, CI 0.985–0.995, p < 0.0001), and urinary chromium (OR 0.988, CI 0.981–0.995, p < 0.001) were independently associated with the presence of established coronary disease applying this model. While chromium
metabolism appears to be altered in individuals with CVD, it is unclear whether chromium supplementation would be effective
in CVD prevention among patients with IGT. This would need to be tested in long-term outcome trials. 相似文献
