Changes in intracellular activities of H+, K+, Na+, and Cl ions were recorded with ion-selective microelectrodes during progesterone-induced maturation of full-grown oocytes of the urodele amphibians Ambystoma mexicanum and Pleurodeles waltlii. The membrane potential (Em) and electrical resistance (Rm) were also determined. During the first hours after initiation of maturation, the oocytes slowly depolarized and Rm gradually increased. By the end of maturation of Pleurodeles oocytes Em had stabilized at about −10 mV and Rm had increased from 410 to 1760 kΩ. The same initial pattern was observed for Ambystoma, but in most oocytes a rapid transition occurred at about the time of germinal vesicle breakdown (GVBD): Em spontaneously shifted from about −15 to about +30 mV; simultaneously Rm dropped from 1230 down to 100 kΩ (i.e., less than the initial 270 kΩ resistance). The internal K+ activity did not show any important variation during maturation of Ambystoma and Pleurodeles oocytes. Na+ activity increased slightly at the onset of GVBD in Ambystoma; a further marked increase of Na+, accompanied by an increase in Cl activity, was observed as soon as Em shifted to a positive value. In Pleurodeles sodium activity was also more elevated in matured than in immature oocytes. The average pH of Ambystoma immature oocytes was 7.48 ± 0.05 (external pH 7.5). A transient alkalinization to 7.64 ± 0.04 took place during the first 4–6 hr postprogesterone. Cytoplasmic pH was restored to 7.50 ± 0.07 between 10 and 12 hr postprogesterone, before the onset of GVBD and the shift of Em. The difference between the measured oocyte pH and the calculated equilibrium pH decreases during the course of maturation, due partly to the depolarization of Em.  相似文献

Uptake of 14C-labelled amino acids into single oocytes was determined using 3H-labelled choline to correct for extracellular space. Cycloleucine, a non-metabolisable amino acid sharing an entry mechanism with methionine and phenylalanine, was transported in accord with Michaelis-Menten kinetics. At extracellular levels below 8 mM, cycloleucine was concentrated within the oocyte. The proportion of sheep oocytes having a functional amino acid transport system (i.e. cycloleucine flux > 1 nmole cm−2 h−1) was highest in pre-ovulatory follicles (97%), and lowest in atretic follicles (59%). Amino acid fluxes in functional germinal vesicle oocytes were similar at all stages of development studied. An increase in Vmax but not Km during meiotic maturation resulted in a doubling of amino acid uptake in metaphase II oocytes. These increased fluxes were under gonadotropic regulation and were independent of nuclear maturation. Amino acid uptake by mouse oocytes was approximately half that measured in sheep oocytes.  相似文献

Synopsis New Zealand snapper, Pagrus auratus, were captured by trawling from NE New Zealand over two successive spawning seasons, and examined for acute temporal changes in gonad condition. Fish with oocytes completing final oocyte maturation predominated during the morning, with a peak in ovulated fish occurring just after midday. Afternoon catches were dominated by fish in which the most advanced oocytes had yet to begin final maturation. This suggests that ovulation is synchronised to occur soon after midday, and the high proportion (up to 100% of the catch) of fish with particular gonad stages captured at any one time indicates that daily spawning involves most of the population. Diurnal changes in oocyte diameter support a daily spawning rhythm, with numbers of large hydrated oocytes peaking in the late morning, followed by the disappearance of these oocyte stages in the afternoon. Snapper captured alive by longlining were returned to the laboratory to examine the relationship between ovulation and probable time of spawning. Unovulated fish generally ovulated close to midday on the day of capture (morning captures), or the following day (afternoon captures). The viability of ovulated eggs (proportion undergoing division after fertilization) decreased markedly after oviduct residence times of over 8 hours. This suggests that natural spawning occurs before the late evening. The results of this study are consistent with anecdotal evidence suggesting that spawning occurs every day during the late afternoon or early evening, and is similar to the reproductive patterns displayed by a number of closely related sparids. Summary The responses of the kinetic properties of malate dehydrogenase to environmental temperature variability were compared for two populations of Potentilla glandulosa (Rosaceae). The two populations are native to regions of contrasting climates, with the inland population experiencing a high level of temperature variability during growth and the coastal populaton a low level of temperature variability. The substrate binding ability, as measured by apparent K m of both populations was relatively insensitive to assay temperature (Q 10<2.0) over the range of temperatures likely to be encountered during growth. The breadth of this thermal optimum was different for the two populations with the K m of the inland plants exhibiting relative temperature insensitivity over a much wider range of temperatures than the K m of the coastal plants. There was no difference between the two populations in the thermal stability of MDH activity.  相似文献

The influx of K+(86Rb+) into intact roots of rye (Secale cereale L. cv. Rheidal) exposed to a differential temperature (DT) between the root (8° C) and shoot (20° C) is initially reduced compared with warm-grown (WG) controls with both shoot and root maintained at 20° C. Over a period of 3 d, however, K+-influx rates into DT plants are restored to levels similar to or greater than those of the WG controls, the absolute rates of K+ influx being strongly dependent upon the shoot/root ratio. Acclimation in DT plants results in a reduction of K+ influx into the apical (0–2 cm) region of the seminal root which is associated with a compensatory increase in K+ influx into the more mature, basal regions of the root. Values of V max and apparent K m for K+ influx into DT plants were similar to those for WG plants at assay temperatures of 8° C and 20° C except for an increase in the apparent K m at 8° C. The influx of K+ from solutions containing 0.6 mol·m-3 K+ into both WG and DT plants was found to be linearly related to assay temperature over the range 2–27° C, and the temperature sensitivity of K+ influx to be dependent upon shoot/root ratio. At high shoot/root ratios, the ratio of K+ influx at 20° C:K+ influx at 8° C for WG plants approached a minimum value of 1.9 whereas that for DT plants approached unity indicating that K+ influx into DT plants has a large temperature-insensitive component. Additionally, when plants were grown in solutions of low potassium concentration, K+ influx into DT plants was consistently greater than that into WG plants, in spite of having a greater root potassium concentration ([K+]int). This result indicates some change in the regulation of K+ influx by [K+]int in plants exposed to low root temperatures. We suggest that K+ influx into rye seedlings exposed to low root temperatures is regulated by the increased demand placed on the root system by a proportionally larger shoot and that the acclimation of K+ influx to low temperatures may be the result of an increased hydraulic conductivity of the root system.Abbreviations DT differential temperature pretreatment - [K+]int root potassium concentration - [K+]ext potassium concentration of nutrient medium - WG warm-grown pretreatment  相似文献

Full-grown ovarian oocytes removed from non-hormone-treated Rana pipiens females exhibit a low level of protein synthesis, the rate of which is dependent upon the ionic environment. The highest rates of protein synthesis in these oocytes are obtained in media containing either a divalent cation (Ca++ or Mg++) or high levels of K+. The dependence of protein synthesis on ionic environment persists through about the first 18-24 hours of maturation (at 18°C). Normal maturation of oocytes in vitro also has specific ionic requirements for the first 24 hours. In this case, the process requires high ionic strength (T/2 = 1.0-1.2) and divalent cations. The kinetics of K+ exchange suggest that K+ exists in the ovarian oocyte in two compartments; one in equilibrium with the exogenous medium and freely exchangeable; the other in equilibrium with the exogenous medium and freely exchangeable; the other in equilibrium with the first internal compartment and only very slowly exchangeable. The slowly exchangeable (bound) compartment contains about 95% of all endogenous K+. In hormone stimulated oocytes, the kinetics of K+ exchange are essentially the same. Oocyte adaptation to ionic environment is discussed as a possible regulatory mechanism during maturation.  相似文献

The upper thermal tolerance of brook trout Salvelinus fontinalis was estimated using critical thermal maxima (CTmax) experiments on fish acclimated to temperatures that span the species' thermal range (5–25°C). The CTmax increased with acclimation temperature but plateaued in fish acclimated to 20, 23 and 25°C. Plasma lactate was highest, and the hepato-somatic index (IH) was lowest at 23 and 25°C, which suggests additional metabolic costs at those acclimation temperatures. The results suggest that there is a sub-lethal threshold between 20 and 23°C, beyond which the fish experience reduced physiological performance.  相似文献

Many populations of shortnose sturgeon, Acipenser brevirostrum, in the southeastern United States continue to suffer from poor juvenile recruitment. High summer water temperatures, which may be exacerbated by anthropogenic activities, are thought to affect recruitment by limiting available summer habitat. However, information regarding temperature thresholds of shortnose sturgeon is limited. In this study, the thermal maximum method and a heating rate of 0.1°C min−1 was used to determine critical and lethal thermal maxima for young-of-the-year (YOY) shortnose sturgeon acclimated to temperatures of 19.5 and 24.1°C. Fish used in the experiment were 0.6 to 35.0 g in weight and 64 to 140 days post hatch (dph) in age. Critical thermal maxima were 33.7°C (±0.3) and 35.1°C (±0.2) for fish acclimated to 19.5 and 24.1°C, respectively. Critical thermal maxima significantly increased with an increase in acclimation temperature (p < 0.0001). Lethal thermal maxima were 34.8°C (±0.1) and 36.1°C (±0.1) for fish acclimated to 19.5 and 24.1°C, respectively. Lethal thermal maxima were significantly affected by acclimation temperature, the log10 (fish weight), and the interaction between log10(fish weight) and acclimation temperature (p < 0.0001). Thermal maxima were used to estimate upper limits of safe temperature, thermal preferences, and optimal growth temperatures of YOY shortnose sturgeon. Upper limits of safe temperature were similar to previous temperature tolerance information and indicate that summer temperatures in southeastern rivers may be lethal to YOY shortnose sturgeon if suitable thermal refuge cannot be found.  相似文献

We tested the effect of growing conditions during micropropagation on the fast kinetics of chlorophyll (Chl) fluorescence of Gardenia jasminoides Ellis plantlets during a 4-week acclimation to ex vitro. We studied whether photoautotrophic growing in vitro produced plantlets with less photoinhibition impairment during acclimation. Of the growing conditions stimulating photoautotrophy in vitro, only loose tube caps had a positive effect, whereas low sucrose or sucrose-free content in the medium and high PPFD showed a negative effect. Thus, plantlets cultured with 3 % (m/v) of sucrose were subsequently less photoinhibited throughout acclimation than those cultured with low sucrose (0.5 %) or sucrose-free media. Moreover, at the end of acclimation the former plantlets showed Fv/Fm and Fv/F0 ratios typical of unstressed ex vitro plants as well as a higher Chl content and ratio of Chls to carotenoids. Plantlets cultured at a photosynthetic photon fluence density (PPFD) of 50 µmol m–2 s–1 also showed a better performance at the end of acclimation than those cultured at a higher (110 µmol m–2 s–1) PPFD. Thus except in the case of loose-tube closure, gardenia plantlets cultured in vitro under conventional sucrose concentration and PPFD are the least photoinhibited during acclimation. Nevertheless, significant interactions between the in vitro growing factors were observed at the end of acclimation.  相似文献

The dynamics and regulation of oogenesis in single and multiple-spawner cyprinid fish showing group-synchronous oocyte development, was investigated in three species from the River Meuse (Belgium): the roach Rutilus rutilus as a single spawner, and the bleak Alburnus alburnus and the white bream Blicca bjoerkna as multiple spawners. This paper compares the seasonal profiles of sex steroids (oestradiol-17β, testosterone and 17,20β-dihydroxy-4-pregnen-3-one) and plasma alkali-labile protein phosphorus. Different patterns of plasma oestradiol-17β (E2) and plasma protein phosphorus (PPP) have been observed not only between the single and the multiple spawner fish, but also among the two multiple spawners. In roach, two increases of E2 levels were observed. The first occurred in September after a short gonadal quiescent period, and coincided with the increase of the PPP at the onset of exogenous vitellogenesis. The second took place in spring before the spawning season. The low PPP recorded during that period probably reflected its rapid incorporation by the oocytes. In both multiple spawners, highest values of PPP were recorded just before the spawning season. In white bream, the PPP declined progressively once the differentiation of exogenous vitellogenic oocytes was completed before the onset of the spawning season. In bleak, PPP levels remained high throughout the spawning season and corresponded to a sustained oocyte recruitment during the whole of this period. Regardless of the pattern of oocyte growth recruitment, the E2 concentrations were high in both multiple spawner species during the breeding season. In the three species, testosterone levels remained low regardless of the maturation stage (ranging from 0.6 to 13.4ng ml?1). Except for relatively high concentrations of 17,20βP in roach during final maturation and postspawning stages (20 and 28 ng ml?1, respectively), low levels of this steroid were measured in these cyprinids, and especially in the multiple spawner fish. The role of this progestogen as the maturation inducing Steroid is discussed.  相似文献

Histology of gonads of Oreochromis karongae was undertaken to study internal cell characteristics during maturation. This study was necessitated by low spawning output of the fish species. Several oocyte stages, ranging from primary forms to vitellogenesis, suggest that the maturation was generally succesfully attained in the fish ponds. Pre‐vitellogenesis oocytes (oogonia to perinuclear stage) and more advanced vitellogenesis (primary vesicle to tertiary yolk vesicle) oocyte stages were all found in the same gonads. However, there were some discontinuities observed during stages 3 and 4, suggesting selective maturation. Failure of gonads to mature normally is attributed to an ecological crunch that was in a previous study associated with environmental factors. Atretic oocytes were also recorded in the same gonads, a sign that some oocytes failed to mature normally. This indicates insufficient stimuli for normal gonad development. Several stages of spermatogenesis (spermatocytes, spermatid and spermatozoa) were also found in the same gonads. Selective recrudescence was more pronounced in O. karongae because generally less oocytes attained final maturation stages compared to Oreochromis niloticus and other tilapias. This could be the main reason for low natural breeding that has been observed in both wild and captive stocks, and led to the abandonment of its use in aquaculture. This study corroborates findings of previous studies that depended solely on external gonad characteristics. Histology provides conclusive evidence from internal cell characteristics that other techniques are unable to show.  相似文献

The mullet Mugil liza occurs along the Atlantic coast of South America from Venezuela to Argentina, but 95% of the commercial catch is collected from south Brazil between São Paulo and Argentina. Mugil liza is a single spawner with oocyte development occurring synchronously in two groups. Spawning happens in marine areas and occurs after migration. The reproductive migration occurs from Argentina (38° S) to the southern Brazilian states (24–26° S) from April to July, with peak spawning in June between northern Santa Catarina and Paraná. The presence of hyaline oocytes was associated with high salinity and sea surface temperatures of 19–21° C, and followed the seasonal northward displacement of these oceanographic conditions. The average size at first maturity (Lm) for both sexes was 408·3 mm total length, LT. Males (Lm = 400·1) matured earlier than females (Lm = 421·9 mm). Fecundity ranged from 818 992 to 2 869 767 oocytes (mean = 1 624 551) in fish that were between 426 and 660 mm LT.  相似文献

Mouse oocytes develop sensitivity to inositol 1,4,5-trisphosphate (IP3) during oocyte maturation. We recently reported that a change in the organization of the endoplasmic reticulum (ER) during oocyte maturation may contribute to this enhanced sensitivity (Mehlmannet al.,1995,Dev. Biol.170, 607–615). Here, we investigated whether there is an increase in the number of available IP3receptors after maturation and whether there is a redistribution of IP3receptors similar to the redistribution of the ER that occurs during maturation. Western blot analysis of the IP3receptor in oocytes and eggs demonstrated a 1.8-fold increase in immunoreactive mass of the IP3receptor following oocyte maturation. Microinjection of the function-blocking monoclonal antibody 18A10 inhibited IP3-induced Ca2+release in a concentration-dependent manner in both eggs and oocytes. More antibody was required to inhibit Ca2+release to the same extent in eggs compared to oocytes when both were injected with the same concentration of IP3, suggesting that eggs contain a greater number of functional IP3receptors. Immunolocalization of the IP3receptor revealed that receptors were present in large clusters, 1–2 μm in diameter, in the cortex of the mature egg except in a ring-shaped band of cortex adjacent to the meiotic spindle. In contrast, receptor clusters were located around the entire cortex of the immature oocyte and were much smaller (<1 μm); larger patches were sometimes seen, but they did not display the same spherical organization as those in eggs. These results suggest that the number of cortical IP3receptors increases during mouse oocyte maturation and that this increase may contribute to enhanced Ca2+release at fertilization.  相似文献

Photoinhibition of photosynthesis was induced in attached leaves of kiwifruit grown in natural light not exceeding a photon flux density (PFD) of 300 mol·m-2·s-1, by exposing them to a PFD of 1500 mol·m-2·s-1. The temperature was held constant, between 5 and 35° C, during the exposure to high light. The kinetics of photoinhibition were measured by chlorophyll fluorescence at 77K and the photon yield of photosynthetic O2 evolution. Photoinhibition occurred at all temperatures but was greatest at low temperatures. Photoinhibition followed pseudo first-order kinetics, as determined by the variable fluorescence (F v) and photon yield, with the long-term steady-state of photoinhibition strongly dependent on temperature wheareas the observed rate constant was only weakly temperature-dependent. Temperature had little effect on the decrease in the maximum fluorescence (F m) but the increase in the instantaneous fluorescence (F o) was significantly affected by low temperatures in particular. These changes in fluorescence indicate that kiwifruit leaves have some capacity to dissipate excessive excitation energy by increasing the rate constant for non-radiative (thermal) energy dissipation although temperature apparently had little effect on this. Direct photoinhibitory damage to the photosystem II reaction centres was evident by the increases in F o and extreme, irreversible damage occurred at the lower temperatures. This indicates that kiwifruit leaves were most susceptible to photoinhibition at low temperatures because direct damage to the reaction centres was greatest at these temperatures. Serotonin (5-HT) was found to inhibit steroid (17α,20β-dihydroxy-4-pregnen-3-one; 17,20βP)-induced resumption of oocyte meiosis (oocyte maturation) in vitro in the teleost Fundulus heteroclitus. Serotonin inhibited both follicle-enclosed and denuded oocytes, which indicates the presence of oocyte-associated 5-HT sensitive sites. The response of oocytes to 5-HT was characterized pharmacologically, i.e., the capacity of serotonergic agonists and antagonists to mimic or block the 5-HT inhibition of the steroid-induced oocyte maturation was assessed by the changes in the percentage of oocyte germinal vesicle breakdown (GVBD). Dose-response curves for each compound were drawn and compared. The rank order of potency among the agonists was: 5-HT > 5-methoxytryptamine > tryptamine = 5,6-diHT = 5-carboxidotryptamine > 5,7-diHT = 5-methoxy-dimethyltryptamine > α-methyl-5-HT > 2-methyl-5-HT. Incubation of ovarian follicles with high doses of some antagonists (mianserin and metergoline) induced oocyte GVBD, although this effect was associated with high levels of oocyte atresia during GVBD or shortly after maturation. Consequently, doses of the antagonist too low to induce GVBD were tested for their ability to block the 5-HT inhibitory action; the rank order of potency was: MDL-72222 = metoclopramide > metergoline > propanolol > ketanserin. Dopamine, acetylcholine, epinephrine, and norepinephrine could also inhibit 17,20βP-induced GVBD, although at doses much higher than those of 5-HT; melatonin and histamine had no effect on oocyte maturation. These results suggest that specific receptors mediate the inhibitory action of 5-HT on the steroid-triggered meiosis resumption. The pharmacological profile of these 5-HT receptors is different from those of any known mammalian 5-HT receptor, although they showed some similarities to the 5-HT1A, 5-HT2, and 5-HT3 receptors, as well as to 5-HT receptors on oocytes of some bivalve molluscs. Summary When apparent Michaelis constants (K m's) for pyruvate of M4-lactate dehydrogenases from differently thermally adapted vertebrates are measured at the species' normal cell temperatures, a marked degree of conservation inK m is observed, but only when the pH of the assay medium is varied in the manner in which intracellular pH varies with temperature in most animals (Fig. 2).K m measurements performed at a constant pH do not yield this high degree of interspecific conservation inK m (Figs. 2 and 3).The temperature dependence of intracellular pH preserves the charge states of imidazoles of protein histidines during temperature transitions. Thus under intracellular conditions the ionization state of the active site histidine of LDH will be independent of temperature, reducing the temperature dependence of pyruvate binding. This effect appears important in the contexts of short-term temperature variation experienced by an individual ectotherm and of long-term, evolutionary temperature changes important in speciation processes.These findings emphasize the importance of utilizing biologically realistic pH values in enzyme studies if major adaptive trends are to be observed.  相似文献

We have prepared polyclonal antibodies againstXenopus20S proteasomes. The antibodies cross-react with several proteins that are common to 20S and 26S proteasomes and with at least two proteins that are unique to 26S proteasomes. The antibodies were used to analyze changes in the components of proteasomes during oocyte maturation and early development ofXenopus laevis.A novel protein with a molecular weight of 48 kDa, p48, was clearly detected in immature oocytes, but was found at very low levels in mature oocytes and ovulated eggs. p48 was reduced to low levels during oocyte maturation, after maturation-promoting factor was activated. The amount of p48 in eggs remained low during early embryonic development, but increased again after the midblastula transition. These results show that at least one component of 26S proteasomes changes during oocyte maturation and early development and suggest that alterations in proteasome function may be important for the regulation of developmental events, such as the rapid cell cycles, of the early embryo.  相似文献

Currently, it is believed that toad oocyte maturation is dependent on the physiological conditions of winter hibernation. Previous antibody-blocking experiments have demonstrated that toad ubiquitin carboxyl-terminal hydrolase L1 (tUCHL1) is necessary for germinal vesicle breakdown during toad oocyte maturation. In this paper, we first supply evidence that tUCHL1 is highly evolutionarily conserved. Then, we exclude protein availability and ubiquitin carboxyl-terminal hydrolase enzyme activity as factors in the response of oocytes to winter hibernation. In the context of MPF (maturation promoting factor) controlling oocyte maturation and to further understand the role of UCHL1 in oocyte maturation, we performed adsorption and co-immunoprecipitation experiments using toad oocyte protein extracts and determined that tUCHL1 is associated with MPF in toad oocytes. Recombinant tUCHL1 absorbed p34cdc2, a component of MPF, in obviously larger quantities from mature oocytes than from immature oocytes, and p13suc1 was isolated from tUCHL1 with a dependence on the ATP regeneration system, suggesting that still other functions may be involved in their association that require phosphorylation. In oocytes from hibernation-interrupted toads, the p34cdc2 protein level was significantly lower than in oocytes from toads in artificial hibernation, providing an explanation for the different quantities isolated by recombinant tUCHL1 pull-down and, more importantly, identifying a mechanism involved in the toad oocyte’s dependence on a low environmental temperature during winter hibernation. Therefore, in toads, tUCHL1 binds p34cdc2 and plays a role in oocyte maturation. However, neither tUCHL1 nor cyclin B1 respond to low temperatures to facilitate oocyte maturation competence during winter hibernation.  相似文献

Summary Phospholipase A2 (PLA2) was extracted from liver microsomal membranes of both 5 and 20°C-acclimated rainbow trout (Salmo gairdneri), using the non-ionic detergent, Triton X-100. Further purification was achieved by precipitation with 35–65% ammonium sulfate followed by gel filtration chromatography in the presence of 0.1% Triton X-100 on Sephadex G-200. These procedures resulted in a 30-fold purification and the removal of all traces of phospholipid from the enzyme of both warm-and cold-acclimated trout. Column elution profiles were similar for both acclimation groups, yielding a molecular weight estimate for the trout liver enzyme of 73,000. Comparisons of activity levels and kinetic parameters of PLA2 from warm-and cold-acclimated fish, indicated that compensation for temperature at nonsaturating substrate concentrations was an attribute of both the particulate (microsomal) enzyme and the lipid-free protein. Cold acclimation resulted in higher activity belowV max due primarily to decreased apparentK m values. These adaptations to temperature could not be attributed to the interaction of the enzyme with the membrane lipids, but were due to qualitative changes in the enzyme that resulted from acclimation. Other adaptive qualities of PLA2, such as reducedK m in response to acute decreases in temperature in warm-acclimated fish, were only apparent in particulate preparations, and thus were a function of the protein-lipid complex. Striped Bass naturally inhabit a wide range of temperatures, yet little is known about the processes that control their acute and chronic temperature limits. The objective of this study was to determine the effect of temperature acclimation on acute thermal maxima and physiology of juvenile Striped Bass. Juvenile fish were acclimated to 15, 25 or 30 °C for 4 weeks, then split into two sampling groups: post-acclimation and post-critical thermal maximum trials. We found that fish survived in all acclimation temperatures with little change to underlying hematology, and that critical thermal maximum (CTmax) increased with increasing acclimation temperature. At CTmax, fish acclimated to 30 °C had elevated plasma cortisol, lactate and potassium levels. These results suggest that, while 30 °C is likely to be outside their thermal optima, Striped Bass can survive at high temperatures. This ability to cope with warm temperatures may provide an advantage with increasing global temperatures.
