Polyacrylamide gel electrophoresis was used to resolve allozymes in the cosmopolitan blood-feeding stable fly,Stomoxys calcitrans (L.). Nineteen of 38 loci were polymorphic (53%). Mean heterozygosities among all loci and among only polymorphic loci were 0.096 and 0.182, respectively. These gene diversity measures are about half those among other muscid Diptera. Variation in gene frequencies was examined in 10 natural stable fly populations from Iowa and Minnesota. Gene frequencies were homogeneous at five of eight loci among six populations in 1990 and eight of eight loci among four populations in 1992. Wright'sF statistics showed no significant departure from random mating among stable flies. It was concluded that gene flow compensates for any local differentiation in stable fly populations.  相似文献

Patterns of genetic variation within and between populations of five species of mycophagous Drosophila were examined by gel electrophoresis of several polymorphic loci. Populations of the five species could not be shown to be subdivided into sympatric host-adapted races. Statistically significant, but small, between-host differences in gene frequencies were observed at three of 15 loci. Mean gene frequencies at all loci were similar in New York and Tennessee, and, with one exception, relatively little genetic differentiation was observed among study sites within those two regions. Gene frequencies generally were stable over several years of collecting as well. The unpredictable nature of the fungal hosts may preclude the site fidelity and continuity of diversifying selection necessary for adaptive divergence of populations.  相似文献

Summary Allelic and genotypic frequencies were sampled from a single age class of the common house fly, Musca domestica L., at five farms on six dates from July 6 to October 12, 1982. Allozymes at six loci were resolved with vertical polyacrylamide gel electrophoresis. No consistent departures from random mating were detected. No consistent linkage disequilibrium was observed. Allele frequencies at the farms changed in independent and unpredictable ways. Gene frequencies at the five farms were initially divergent, converged in midsummer, and then progressively diverged. The divergence occured in mid-August when fly populations were large. Variation in gene frequencies at adjacent farms accounted for a large proportion of the variance in allele frequencies among all farms. Morphological and genetic variation were studied in Plantago cordata Lam., an imperiled aquatic plant. The existence of distinct seasonal morphs was confirmed by numerical morphological analyses of specimens from 53 populations. Plants from two North Carolina populations possessed leaves that were significantly shorter, narrower, and fewer-veined than populations in other portions of the species range. Genetic variation within and among ten populations in seven states/provinces (Illinois, Missouri, New York, North Carolina, Ohio, Ontario, and Wisconsin) was examined electrophoretically. The species is an apparent allopolyploid with fixed heterozygosity observed at 67% of the loci surveyed. Electrophoretic variation was mostly partitioned among populations. The genetic identity among populations was high except for North Carolina populations which differed at 27% of the loci surveyed.  相似文献

In species that demonstrate female choice, geographically distinct populations can vary in their signal‐response behaviors as a result of environmental differences or genetic drift. Observing whether or not females discriminate against males from allopatric populations can establish such signal deviations. Here we compare mating success within and between populations of the spotted cucumber beetle (Diabrotica undecimpunctata howardi) collected from Delaware, Tennessee, Missouri, and New Mexico, USA. A no‐choice cross‐mating experiment was employed to measure female preference for sympatric and allopatric males. While only two of the populations (Tennessee and Missouri) demonstrated statistically significant female preference for sympatric males, this trend was observed in all populations tested. Further, we show that (i) males from Tennessee, Missouri, and New Mexico differ in their scent, (ii) females may use population‐specific scents to discriminate among males, and (iii) females whose antennae have been surgically removed are unable to recognize acceptable mates. New Mexico males, which were never accepted by either Tennessee or Missouri females, became acceptable mates when crowded with Tennessee or Missouri males prior to copulation. We infer that male odor may be an important factor in determining cucumber beetle mating success.  相似文献

Vertical polyacrylamide gel electrophoresis was used to separate enzyme proteins at 73 putative loci in natural house fly populations sampled in central Iowa. Thirty-nine of the loci were polymorphic (53%). The mean effective number of alleles per polymorphic locus was 1.93 and 1.47 alleles among 68 scored loci. Observed and expected heterozygosities at 34 house fly loci were 0.1628 and 0.1834, respectively. No statistically significant differentiation was detected among nine central Iowa fly populations in 1989 or among nine Iowa and three Minnesota populations in 1990. The frequency of insecticide-resistance alleles for two genes (Vssc1 and CYP6D1) was studied in field collected populations of house flies from two different climates. While the frequency of these resistance alleles in flies at dairies from four states has recently been reported, there is no information on the relative change of these allele frequencies over time. House flies were collected during the 2003-2004 season from New York and Florida before the first application of permethrin, during the middle of the field season, after the final application, and again the following spring (following months without permethrin use). Bioassay results indicated that homozygous susceptible and extremely resistant flies were rare, while moderately and highly resistant individuals were relatively common at all times in both states. The frequency of resistance alleles at the New York dairy rose during the season and declined over the winter, suggesting an overwintering fitness cost associated with these alleles. The super-kdr allele was detected for the first time in North America at the end of 2003. In Florida the frequency of the resistance alleles did not increase during the spray season or decrease during the winter, suggesting there is substantial immigration of susceptible alleles to the Florida dairy and no overwintering fitness cost associated with resistance alleles in this climate. Resistance to permethrin correlated well with the frequency of the Vssc1 and CYP6D1 resistance alleles in flies from New York, but not as well in the population from Florida. This suggests there may be a new resistance mechanism or allele evolving in Florida.  相似文献

Genetic variation and spatial genetic structure in balsam fir (Abies balsamea) were examined in two isolated populations in Iowa and Minnesota thought to be paleorefugia and in two ecologically central populations in old-growth forests of Upper Michigan. Overall levels of genetic variability at 22 allozyme loci were lower than that found in most conifer species (H(o) values ranged from 0.005 in the isolated populations to 0.025 in the central populations). The mean F(IS) value (0.154) was larger than usually found in conifers and suggests moderate levels of inbreeding. The mean F(ST), an estimate of genetic diversity among populations, was 3.7% of the total diversity, a value lower than the mean for conifers. Nm, the number of migrants per generation, was 6.5, suggesting either some gene flow among populations or a lack of genetic differentiation. Spatial autocorrelation analysis revealed a moderately patchy structure, with gene flow distances of 30-70 m in the central populations and at least 10 m in the isolated populations. The future of the ecologically central populations depends on maintenance of an intact forest mosaic. The low genetic variability in the small, isolated populations suggests that habitat fragmentation has led to a reduction in evolutionary potential and that the future viability of these populations will likely require active management in the face of global climate change.  相似文献

Vertical polyacrylamide gel electrophoresis was used to resolve enzymes at 50 putative loci in Iowa face flies, a recently arrived, colonizing, Palearctic species. Sixty-two percent of the 50 loci were polymorphic. The mean number of alleles per polymorphic locus was 3.2 +/- 1.38; the mean among all loci was 2.38 +/- 1.62 alleles. The effective number of alleles among 50 loci was 1.4 +/- 0.62. Mean observed and expected heterozygosities for the 50 face fly loci were 0.167 +/- 0.037 and 0.186 +/- 0.031, respectively. Comparison of the electrophoretic data for Musca autumnalis and for M. domestica L. showed similar high levels of gene diversity. A survey of gene diversity at 12 loci among six geographically independent laboratory colonies demonstrated statistically significant genetic differentiation that was probably due to drift after colonization.  相似文献

Persistent seed banks may provide information on historical changes in the genetic composition of populations. We used stratified sampling of the soil seed bank of Astragalus bibullatus (Pyne's ground plum) to assess levels of temporal variation in population genetic structure and to infer historical changes in the levels of inbreeding and relative gene flow. This species has an extremely limited distribution in the Central Basin of Tennessee, where it is found in open areas and along the edges of cedar glades. Protein electrophoresis was conducted on seedlings grown from seeds that had been recovered from three successive 1 cm thick layers of soil sampled from six sites. Analyses of seven polymorphic allozyme loci indicated that there were substantial levels of genetic differentiation among soil layers and sites. Higher levels of genetic diversity were found in seed than in vegetative populations that had been sampled in a previous study. Seed populations from the uppermost soil layer had higher heterozygote deficiencies, displayed higher levels of differentiation among sites, and had higher private allele frequencies than seed populations from the lower two layers. The change in heterozygosity and distribution of genetic variation among sites for the youngest soil layer is consistent with a pattern of increased selfing, sib mating, and decreased gene flow among populations. These changes in inbreeding and relative levels of gene flow are corroborated by information on historical land use practices in the region and support the hypothesis that loss of appropriate habitat has led to smaller population sizes and a more fragmented distribution of this cedar glade endemic.  相似文献

Allozyme and mitochondrial gene diversities were estimated in house flies, Musca domestica L. (Diptera: Muscidae), sampled in Iowa, USA; Berkshire, England; and Kudang, The Gambia. Comparison of genomic allele frequencies among the three populations indicated small differences between the English and American samples but very large distances between English or American and the African. The FST statistic was 0.65 +/- 0.09 for allozymes. Pairwise FST was 0.14 between the English and the American samples; FST was 0.65 between the African population and the English and American. Mitochondrial variation in the same flies was assessed by SSCP methods which revealed nine haplotypes, none of which were shared in common. FST was 0.637 for the mitochondrial haplotypes. The research indicates greatly restricted gene flow between Africa and the temperate regions.  相似文献

The prevalence of developing Thelazia nematodes in face flies (Musca autumnalis) was studied for 7 yr at a beef farm in central Iowa. Juvenile nematodes were not found among flies in reproductive diapause in autumn, nor among nulliparous, overwintered flies in spring, but only among actively reproducing insects. Thus Thelazia probably do not overwinter in face flies. A mean prevalence of 2.0% infected was recorded among flies in 7 fly breeding seasons. No heterogeneity in Thelazia prevalence was detected within fly breeding seasons. The frequency distribution of Thelazia among face flies by year of occurrence was homogeneous, with a mean of 2.75 larval nematodes per infected fly. Thelazia gulosa and T. skrjabini were recovered in necropsy from the eyes of bovines from central Iowa. Prevalence among fat cattle 18-27 mo old was 15%; among cows 3-15 yr old, prevalence was only 3%. Thelazia skrjabini was found in 29 eyes and T. gulosa in 4 eyes. Infections were randomly distributed among the eyes of subject cattle. The mean worm burden was 2.3 T. skrjabini and 12 T. gulosa per infected eye. Thelaziasis is clearly enzootic in Iowa.  相似文献

The emu (Dromaius novaehollandiae) occupies most regions of the Australian continent and in recent times has been farmed for meat, oil, and leather. Very little is known about the genetic structure of natural or farmed populations of these birds. We report a preliminary study of genetic variation in emus undertaken by typing birds from five farms and two natural populations at five polymorphic microsatellite loci. Genetic diversity was high for all populations and there was little evidence of inbreeding, with most populations conforming to Hardy-Weinberg equilibrium for most loci. Significant heterozygote deficiencies at one locus in a number of populations were detected and may indicate the presence of null alleles. Comparisons of allele frequencies showed little evidence of genetic differentiation either among farmed populations or between farmed and natural populations.  相似文献

We examined morphological variation among populations of the stream-dwelling waterstrider, Aquarius remigis. A previous analysis of allozyme variation along a transect from southern Quebec to southern North Carolina revealed two distinct 'types' of A. remigis, north and south of a transition zone in southern New York and northern Pennsylvania. To assess the concordance of morphological and allozyme differentiation, adults were sampled from 15 populations along the original transect, as well as 11 additional populations in New York, Connecticut and New Jersey. Eight measurements of leg and body size were taken of preserved individuals. Univariate and multivariate analyses (multiple regression, PCA, discriminant analysis) reveal significant differentiation between northern and southern populations with a transition zone between latitudes 39oand 42onorth. Strong clines within each region and a clear transition between regions support previous suggestions that there are two distinct 'types' of this species in eastern North America. The transition zone, in morphology coincides with that revealed by the allozyme variation, but extends further south. The covariance of allozyme frequency and morphometric traits suggests that differentiation has occurred through vicariance and secondary contact, and that in this system, cladistic and adaptive differentiation are coupled.  相似文献

Chromosome studies of wild D. melanogaster populations from Missouri, Mississippi, Louisiana and Texas uncovered 58 inversions. Six were common and cosmopolitan; 52 were new, rare and generally endemic. In one of two Missouri populations tested, structurally heterozygous females carried significantly more sperm at capture than did the homozygotes. In both populations comparisons of wild sperms with the females carrying them indicated significant positive assortative mating and an excess production of homozygotes among the F1 progeny. Wild females structurally heterozygous in up to three major autosomal arms showed no associated nondisjunctional egg lethality; those heterozygous in all four arms produced from 0% to 24% dead eggs, suggesting the presence of intrapopulational gene modifiers of meiosis. Texas populations supported on windfall citrus fruit showed a slight but significant difference in inversion frequencies between flies breeding on oranges and those breeding on grapefruit. Within these populations inversions were not distributed at random among individuals; rather there was an observed excess of individuals carrying intermediate numbers, and a deficiency of those carrying very few or very many inversions. While there was no significant linkage disequilibrium associated with this central tendency, there was a significant interchromosomal interaction: flies carrying inversions in chromosome 2 tended not to carry them in chromosome 3, and vice versa.  相似文献

BACKGROUND AND AIMS: Oncidium hookeri is a neotropical species of epiphytic Orchidaceae found in the Brazilian Atlantic rainforest at the top of the Mantiqueira Range of mountains. The genetic variation of O. hookeri was studied to assess the distribution of genetic variability within and among six populations localized in Atlantic rainforest remnants. Gene flow among populations and the occurrence of recent bottlenecks were investigated in order to infer the degree of isolation of these populations. METHODS: Thirteen polymorphic loci were used for allozyme electrophoresis. The data were analysed by means of standard statistical approaches, to estimate gene diversity and the genetic structure of the populations. KEY RESULTS: The mean gene diversity and allelic richness were H(e) = 0.099 and A = 1.75, respectively. F-statistics revealed high heterozygote deficiencies in all populations (F(IS) = 0.43-0.82). Several rare alleles were found in all the populations, and three populations presented private alleles. Low genetic differentiation among O. hookeri populations was detected (F(ST) = 0.029); natural selection may be involved in PGM locus differentiation among populations. The genetic differentiation between paired populations was low, bearing no correlation with geographic distance (Mantel test: r = -0.34, P = 0.72). Only two populations showed signs of recent bottlenecks. CONCLUSIONS: The heterozygote deficiency found seems to be caused by pollinator behaviour; the low frequencies of several alleles of different loci can be maintained due to clonal propagation. Despite the stochastic nature of the wind-dispersal of seeds to long distances, this process may promote an effective gene flow among populations, thus avoiding genetic differentiation.  相似文献

The effect of temperature ranging from 5-30 degrees C on in vitro vegetative growth and conidial germination of isolates of the entomophthoralean fungus Furia gastropachae was investigated. Eleven isolates were used for growth studies; two from Maryland, six from New York, and three from Ontario. A subset of four isolates, one each from Maryland and New York and two from Ontario, were used in conidial germination experiments. Growth and germination were significantly associated with temperature for all isolates, occurring throughout the range 5-30 degrees C, though both processes were inhibited to varying degrees at upper and lower extremes. Temperature optima for growth ranged from 20 to 27 degrees C, and for germination from 20 to 25 degrees C. Although significant variability was observed among isolates in growth at temperatures above 13 degrees C, temperature optima were not significantly different among isolates, and variability did not appear to relate to the geoclimatic origins of the isolates. In contrast, germination responses to temperature did appear to be related to geographic origin. Furia gastropachae isolates from New York and Maryland germinated more slowly at 10 degrees C than did Ontario isolates, although the percentage of conidia ultimately germinating at each temperature was the same for all isolates. The New York and Maryland isolates performed much better at 30 degrees C, with significantly greater overall germination and secondary conidial discharge, than the Ontario isolates. Compared with other isolates at 30 degrees C, Ontario isolates were the least active, often failing to successfully discharge any secondary conidia.  相似文献

Genetic variation among 14 populations of Plutella xylostella (Linnaeus) from USA (Geneva, New York), Brazil (Brasilia), Japan (Okayama), The Philippines (Caragan de Oyo), Uzbekistan (Tashkent), France (Montpellier), Benin (Cotonou), South Africa (Johannesburg), Réunion Island (Montvert), and five localities in Australia (Adelaide, Brisbane, Mareeba, Melbourne, Sydney) were assessed by analysis of allozyme frequencies at seven polymorphic loci. Most of the populations were not in Hardy-Weinberg equilibrium and had a deficit in heterozygotes. The global differentiation among populations was estimated by the fixation index (Fst) at 0.103 for the 14 populations and at 0.047 when populations from Australia and Japan, which differed most and had a strong genetic structure, were excluded from the analysis. By contrast, the populations from Benin (West Africa) and Brazil (South America) were very similar to each other. Genetic differentiation among the populations was not correlated with geographical distance.  相似文献

Variation in immune responses is an important part of life history variation. When correlations between multiple immune measures are reported, studies report different patterns. I tested whether the correlation between levels of immune response was consistent across a species range. The ability of tree swallows Tachycineta bicolor to simultaneously produce immune responses to both a humoral immune response and T-cell mediated local inflammation to PHA was tested at three sites that span the breeding range. Females in Tennessee maintained stronger PHA responses than did females in either New York or Alaska. In New York and Alaska, individuals that produced strong PHA responses produced low levels of antibodies to a humoral challenge of sheep red blood cells (SRBC). However, in Tennessee, individuals that showed strong local PHA inflammation also mounted strong responses to SRBC. Increasing daily daytime temperatures led to increased PHA response, but there were no differences in the effect of temperature among sites. These results indicate spatial and/or temporal variation occurs in the ability to produce multiple immune responses simultaneously; this pattern suggests important geographic (or temporal) differences in factors driving investment in immune activity. In addition, these results suggest that studies extrapolating results across populations should be careful to consider geographic variation in immune activity.  相似文献

Conservation genetics is a powerful tool to assess the population structure of species and provides a framework for informing management of freshwater ecosystems. As lotic habitats become fragmented, the need to assess gene flow for species of conservation management becomes a priority. The eastern hellbender (Cryptobranchus alleganiensis alleganiensis) is a large, fully aquatic paedamorphic salamander. Many populations are experiencing declines throughout their geographic range, yet the genetic ramifications of these declines are currently unknown. To this end, we examined levels of genetic variation and genetic structure at both range-wide and drainage (hierarchical) scales. We collected 1,203 individuals from 77 rivers throughout nine states from June 2007 to August 2011. Levels of genetic diversity were relatively high among all sampling locations. We detected significant genetic structure across populations (Fst values ranged from 0.001 between rivers within a single watershed to 0.218 between states). We identified two genetically differentiated groups at the range-wide scale: 1) the Ohio River drainage and 2) the Tennessee River drainage. An analysis of molecular variance (AMOVA) based on landscape-scale sampling of basins within the Tennessee River drainage revealed the majority of genetic variation (∼94–98%) occurs within rivers. Eastern hellbenders show a strong pattern of isolation by stream distance (IBSD) at the drainage level. Understanding levels of genetic variation and differentiation at multiple spatial and biological scales will enable natural resource managers to make more informed decisions and plan effective conservation strategies for cryptic, lotic species.  相似文献
