Inhibition of chlorophyll synthesis by kinetin in Cucumis cotyledons   总被引:1,自引:0,他引:1  
Kinetin is shown to inhibit the synthesis of chlorophyll inlight while causing expansion of isolated cotyledons of Cucumis.The inhibitory effect is more marked at higher concentrationsof kinetin. Investigations have been made on the changes in the levels ofprotochlorophyll, chlorophyll a and chlorophyll b in relationto the kinetin induced expansion of isolated pumpkin cotyledonsin the presence and absence of chloramphenicol. It has been shown that rise in pigment level keeps pace withexpansion growth of the cotyledons. Kinetin markedly promotes the synthesis of protochlorphyll withoutmuch affecting the rate of its photoreduction to chlorophyll. Chloramphenicol strongly inhibits the development of both chlorophylla and b. The inhibition seems to be due to its interferenceboth with the synthesis of protochlorophyll and its subsequentconversion to chlorophyll. The inhibitory effect of chloramphenicol on the formation ofchlorophyll a is greater than on that of chlorophyll b, suggestingthereby the probability of divergent pathways for the formationof the two chlorophylls. Seeds of Raphanus sativus L. were irradiated with 120 and 240Kr of gamma rays from a 137Cs source. Chlorophyll synthesisand fresh weight increases (an estimate of cell expansion) ofisolated cotyledons proved to be radioresistant processes. Alag phase in chlorophyll synthesis, removable by kinetin (10mg/liter), was observed. Gamma radiation decreased the reactivityto kinetin in the removal of the lag phase, but did not affectthe kinetin-stimulated fresh weight increase both in light anddark. Addition of 8-azaguanine to the incubation media reducedthe kinetin-stimulated fresh weight increase. Kinetin retarded the decrease in chlorophyll content in leafdiscs from 5 species of plants with amphistomatous leaves, wherethe upper surface was exposed to air, but not in Rumex acetosera.When leaf discs were floated so that the lower surface was exposed,the effect of kinetin was less evident. Kinetin also stimulatedtranspiration in leaf discs from Nicotiana tabacum (amphistomatous),but not in leaf discs from Paederia chinensis (hypostomatous).Nor kinetin did retard chlorophyll breakdown in this specieswhen leaf discs were floated so that the stomatal surface wasin contact with the solution. The ineffectiveness of cytokininsin chlorophyll retention in leaf discs from hypostomatous leaveswas not due to reduced uptake of benzylaminopurine-14C. Chlorophyll retention was severely inhibited by coating theleaf surface with vaseline either in the presence or absenceof kinetin. Leaf discs floated on a solution exposed to CO2-lessair retained more chlorophyll than those in normal air. Thereis thus a close relationship between stomatal opening (as measuredby stimulation of transpiration) and chlorophyll retention,as influenced by cytokinins. Pulse-chase experiments indicated that the higher levels ofa-amylase in detached and incubated cotyledons of Vigna mungothan those in cotyledons attached to the embryonic axis weredue to both faster synthesis and slower degradation of the enzymein the detached cotyledons than in the attached cotyledons.Levels of a-amylase in the cotyledons were examined in termsof possible effects of end-products and the effects of exogenouslyapplied plant hormones and growth regulators. Levels of a-amylaseactivity and content were reduced by high concentrations ofglucose and sucrose, and it is suggested that this effect wascaused mostly by osmotic stress and partly by end-product repression.The level of a-amylase was nearly twice that in controls after1 to 10µM GA3 had been applied to the cotyledons. In addition,0.1 mM kinetin, 0.1 mM 2,4-D and 0.1 to 0.S mM naphthaleneaceticacid also increased the level by 34% to 66% as compared to thecontrol. Penicillin stimulated the synthesis of pigments in the cotyledonsof intact embryos and excised cotyledons of mung bean (Phaseolusaureus L.) and enhanced benzyladenine-induced accumulation ofchloroplast pigments. The presence of the embryonic axis duringlight exposure proved to be beneficial for chlorophyll synthesisby the cotyledons whereas its presence in dark germination producedan adverse effect. The possible involvement of nucleic acidand protein synthesis in light-regulated chlorophyll formationis suggested. The stimulating effect on pigment synthesis providedby penicillin in this system seems to involve a maintenanceof nucleic acid and protein synthesis. Isolated cotyledons of Pinus pinaster were cultured under bud-formingconditions. The morphology, histology, lipid content and thechlorophyll pools of the cotyledons were followed during buddevelopment. Comparison with cotyledons cultured continuouslyon hormone-free or hormone-supplemented medium was performed.Three periods were distinguished during bud formation: (i) theinitiation period, characterized by a drastic decrease of triacylglycerols,enhanced by the presence of the hormones; (ii) the differentiationperiod, during which the intense cell divisions and the increaseof the polar lipid and chlorophyll contents appeared stronglycorrelated and (iii) bud development, characterized by stablelevels of both chlorophylls and neutral lipids. Moreover, itappeared that the continuous presence of hormones in the culturemedium inhibited the synthesis of molecular components, in particularchlorophylls and membrane lipids, required for normal cell differentiationand physiology. By manipulation of various growth regulators and physical conditions,plants have been regenerated from excised roots, stem segments,cotyledons, leaves, and callus cultures of red cabbage (Brassicaoleracea var. capitata) grown under in vitro conditions. Shootbuds were induced on isolated root segments (1 cm long) culturedon Murashige and Skoog's medium and the frequency of bud formationwas greatly enhanced by the addition of kinetin (0.5 part 10–6).Callus obtained from the seeds, cotyledons, and hypocotyl segmentscultured on a medium fortified with 2,4-D (1 part 10–6),kinetin (0.1 part 10–6), and coconut milk (10%, v/v) hasbeen repeatedly subcultured. The callus is slow growing, andon transference to a kinetin (2 parts 10–6) and IAA (2parts 10–6) medium underwent morphogenesis to give riseto plants. The significance of the propagation of red cabbageby in vitro culture is pointed out.  相似文献

Accumulation of chlorophyll, development of photosystem I andII activities and contents of chloroplastic components wereinvestigated in greening radish seedlings germinated and grownwith 4-thiouridine (4SU). The development of photosystem I activityprior to that of photosystem II was observed also in the 4SU-culturedgreening radish cotyledons in which chlorophyll accumulationwas inhibited up to 60–80% of that of the control. Photochemicalactivities expressed on a plastid protein basis decreased withthe increase of 4SU in the culture medium. In contrast to ferredoxinand ferredoxin-NADP reductase, which were present in significantamounts in the treated cotyledons, chloroplastic cytochromes(f, b559 and b6 decreased in the plastids from 4SU-culturedcotyledons. These results suggest that 4SU interferes in partwith protein synthesis in plastids and thereby with chloroplastdevelopment. (Received December 4, 1979; )  相似文献

The Effect of Potassium on Cotyledon Expansion Induced by Cytokinins   总被引:3,自引:0,他引:3  
Potassium has been found to enhance greatly the expansion response of cucumber cotyledons to cytokinins. A reduction of the response to kinetin is obtained with increasing age of the cotyledons. The lesser response is associated with lower levels of potassium remaining in the cotyledon. A high level of KCI in the incubation medium offsets the lower potassium content of the tissue and enables a much larger response to the cytokinins. At 40 mM KCI the response to kinetin is 4.2 times greater than in the absence of KCI. Calcium increases the effect of potassium on the response to kinetin. When incubated in 40 mM KCI and 10 mM CaCI2 with 10 mg/I 6-benzylamino-purine, the final weight of the cotyledons is 6.8 times the initial weight after just 4 days. This KCI-CaCI2 combination is also found to promote chlorophyll synthesis in the usual cucumber cotyledon bioassay.  相似文献

Kinetin (6-furfurylaminopurine) treatment resulted in the enhanced formation of total polar lipids, total phospholipids and total glycolipids in the isolated cotyledons of vegetable marrow (Cucurbita pepo L). The content of phosphatidic acid, phosphatidyl glycerol and diphosphatidyl glycerol also rose markedly with kinetin treatment. The increase in the content of DGDG was more pronounced compared to MGDG and even more conspicuous on treatment with kinetin. There was a spectacular rise in the carotenoid and chlorophyll content. The results indicated the accelerated formation of plant cell organelles particularly photosynthetically active chloroplasts from heterotrophic leaf on treatment with kinetin.  相似文献

First successful in vitro synthesis of functional photosynthetic phosphorylating membrane is reported. Etioplasts, highly enriched in cytoplasmic and plastid proteins, isolated from etiolated Cucumber cotyledons pretreated with kinetin and gibberellic acid, and illuminated in a cofactor fortified medium showed commencement of chlorophyll (Chl) synthesis immediately after illumination from exogenous δ-aminolevulinic acid, while photosystem I (PS I) activity commenced 15 min after the onset of illumination. When cotyledons pretreated with kinetin and gibberellic acid were illuminated directly, there was a lag phase of 30 min before the commencement of Chl synthesis and PS I activity developed after 1 h of illumination. In plastids developed both in vivo and in vitro, the electron flow from dichlorophenolindophenol to methyl-viologen was coupled to phosphorylation as observed by an increase in the electron transport rate on the addition of uncouplers. Analysis of polypeptide profiles of the greening plastids in vitro showed the disappearance of many higher molecular weight proteins during greening. Polypeptides of molecular weight 32, 20.5, 19.5 K absent in etioplasts appeared as distinct bands after 4 h of greening in vitro.  相似文献

The changes in chlorophyll-protein complexes (CPs) in cucumbercotyledons during illumination and subsequent dark incubationwere studied by SDS-polyacrylamide gel electrophoresis. Whenetiolated cucumber seedlings were illuminated, chlorophyll wassynthesized and CPs were formed. In the early phase of greening(6 h of illumination), light-harvesting chlorophyll a/b-proteincomplex (LHCP) was the main GP. As the greening proceeded, P700chlorophyll a-protein complex (CP1) accumulated. When 6-h illuminatedseedlings were transferred to darkness, CP1 accumulated concomitantlywith a decrease in LHCP without new chlorophyll synthesis. Thechanges in the amounts of CPs in the dark became smaller withthe progress of greening and were not observed after 72 h ofillumination. These changes were confirmed by examining thechlorophyll/P700 ratio and the low temperature absorption spectrumof cotyledons. These results suggest that in the early phaseof greening, CPs were unstable and their chlorophyll moleculeseasily exchanged with those of other kinds of CPs. The involvement of ethylene in red-light-induced stimulationof chlorophyll (Chl) formation was studied because one of thered-light effects on Chl formation (the lateappearing effect)interacts with the ethylene effect in 3-day-old excised etiolatedcotyledons of cucumber (Cucumis sativus L. cv. Aonagajibai).Ethylene production by etiolated cotyledons of intact seedlingsin the dark is enhanced by a red-light pulse, but the effectdoes not occur in excised cotyledons. Application of ethylenein the dark to 3-day-old intact seedlings has little effecton Chl formation in the cotyledon during subsequent continuousillumination, although ethylene pretreatment of 5-day-old seedlingssignificantly stimulates Chl formation. Removal of endogenousethylene by mercuric perchlorate [Hg(ClO4)2] does not specificallysuppress the red-light action on Chl formation in both attachedand excised cotyledons. Inhibition of ethylene synthesis byaminoethoxyvinylglycine does not affect the red-light effecton Chl formation in excised cotyledons. These facts indicatethat ethylene does not operate as a mediator of red light instimulating Chi formation in either attached or excised cotyledons. When etiolated seedlings of squash (Cucurbita maxima Duch. cv.Houkou-aokawaamaguri) were exposed to light, their cotyledonsbegan to accumulate chlorophyll at a low rate for the firsthour but at a high rate therafter. After 0, 1 and 2 h of illumination,cytokinin levels in cotyledons, hypocotyls and roots were analyzedby ELISA (enzyme-linked immunosorbent assay) and GC-SIM (gaschromatography-selected ion monitoring). Cytokinin levels measuredby ELISA were about 10 times higher than those measured by GC-SIM,suggesting that ELISA was not suitable for the measurementsof cytokinin levels in the extracts from these tissues. Cytokininsfound by GC-SIM were t-RZ (t-ribosyl zeatin), c-RZ, t-Z (t-zeatin),c-Z, RiP (ribosyl isopentenyladenine) and iP (isopoentenyladenine).Levels of biologically active cytokinins (t-RZ and t-Z) didnot show marked changes after illumination. Levels of c-RZ andc-Z did, however, decrease in cotyledons and increase in hypocotylsafter illumination. Promotion of the accumulation of the chlorophyllin detached squash cotyledons by exogenously applied t-Z waspartially inhibited by the addition of c-Z, suggesting thatthe decrease in the endogenous level of c-Z in cotyledons ofintact seedlings after illumination permits the light-inducedformation of chlorophyll. Ethylene enhanced chlorosis and levels of 33-kilodalton cationic peroxidase (33-CPO) in excised cucumber (Cucumis sativus L. cv `Poinsett 76') cotyledons. Compared to other hormones, such as kinetin, indoleacetic acid, gibberellic acid, and abscisic acid, ethylene was the only effective promoter of 33-CPO synthesis. The hypothesis that peroxidase plays a role in chlorophyll degradation was tested by comparing levels of 33-CPO in cotyledons treated with compounds thought to either retard (kinetin, indoleacetic acid and gibberellic acid), or promote (abscisic acid and methyl jasmonate [MJ]) senescence. It was concluded that 33-CPO did not play a role in senescence since no direct correlation between chlorophyll content and 33-CPO was observed. MJ was as effective as ethylene in inducing senescence. However, ethylene did not appear to be involved in the action of MJ. Using immunocytochemistry, 33-CPO was found to be located primarily around starch grains and near the plasmalemma. High levels of 33-CPO were also found in cells destined to be vascular tissue.  相似文献

Endopeptidase (azocaseolytic enzyme) and carboxypeptidase activitiesin cotyledons of germinating Vigna mungo seeds increased until3 days after the onset of imbibition and decreased thereafter.In detached and incubated cotyledons, the endopeptidase activityincreased only a little while the carboxypeptidase activitycontinued increasing even after 3 days of incubation. The activitiesof leucine-aminopeptidase and alanine-aminopeptidase, exceptfor that of one leucine-aminopeptidase isoenzyme relativelyabundantly present in unimbibed dry cotyledons, increased slightlyon the first day and declined during germination. In detachedcotyledons, the activities maintained their initial levels throughoutthe incubation period. When cotyledons were detached from germinatingseedlings on days 2 and 4 then incubated, the endopeptidaseactivity started to decrease just after removal of the axisbut the carboxypeptidase activity increased more markedly thanwhen the axis remained attached. Exogenously supplied GA3, kinetin,IAA, or their combinations, showed no significant effect onthe developmental patterns of the endopeptidase and carboxypeptidaseactivities in cotyledons. These results are discussed in relationto the role of the axis in controlling peptidase formation incotyledons of germinating V. mungo seeds. The content of lead in greening etiolated barley leaves remained the same, regardless the time of incubation of excised leaves in the presence of lead ions (8–24 h). The lead deposits have not been detected within mesophyll cells, but were found in intercellular spaces of mesophyll, in guard cells and in cuticle covering stomata. This suggests that lead may be transported in the leavesvia transpiration stream. Lead reduced the content of chlorophyll, especially chlorophyllb content and the average number of grana, whereas in the presence of kinetin the content of chlorophyll increased. In the combined treatment (lead + kinetin) kinetin diminished the inhibitory effect of lead on the chlorophyll content. The number of chloroplasts in mesophyll cells remained unchanged after lead treatment, whereas kinetin alone or applied together with lead increased the average chloroplasts number. The thylakoids system in chloroplasts of kinetin and kinetin + lead treated plants was similar to that observed in control, although the grana number was smaller. Both lead and kinetin increased the content of condensed chromatin in nuclei.  相似文献

The stay-green mutations cytG and Gd1d2 prevent the normal yellowingduring senescence of soybean (Glycine max) leaves and cotyledons.Because light plays such an important role in regulating morphogenesisand it promotes the formation of chlorophyll (Chl), we determinedthe effect of cytG and Gd1d2 (in a cv. Clark background) onthe development and some light responses of seedlings. AlthoughcytG and Gd1d2 seeds, particularly the cotyledons, are greenwhen mature, 44 and 71 % respectively of this Chl broke downwhen the seeds were germinated in darkness. Chlorophyllidesand phaeophytins were not present in the seeds in significantamounts. cytG and Gd1d2 as well as wild type (cv. Clark) seedlingsdeveloped a full etiolation syndrome (morphology and lack ofChl) in darkness. Light induced rapid Chl accumulation in thedark-grown seedlings with no apparent difference among the threeisolines. A short (8 h) exposure to light induced some Chl inthe cotyledons of dark-grown plants, and 22 h of light producedfour times more. Following return to darkness, the 8-h groupshowed very little breakdown over the next 12 d. After the 22-hgroup was returned to darkness, the wild-type lost Chl steadily,but Gd1d2 and eventually also cytG inhibited this breakdown.In the 22-h group, the Chl a/b ratio decreased in wild typeand cytG indicating preferential breakdown of Chl a relativeto Chl b; however, Gd1d2 prevented this change. cytG and Gd1d2seem to act preferentially on Chl breakdown rather than synthesis.Copyright1995, 1999 Academic Press Glycine max, soybean, chlorophyll, chlorophyll a/b ratio, cotyledons, etiolation, cytG, Gd1d2, mutations, senescence  相似文献
